Autochthonous Fermentation as a Means to Improve the Bioaccessibility and Antioxidant Activity of Proteins and Phenolic Compounds of Yellow Pea Flour.

This study focused on evaluating the potential of the natural fermentation of pea flour to improve the release of antioxidant compounds. Preliminary fermentations of 36.4% w/w flour dispersions were performed in tubes under different conditions (24 and 48 h, 30 and 37 °C). Finally, fermented flours (FFs) were obtained in a bioreactor under two conditions: 1: 36.4% w/w, 24 h, 30 °C (FF1); 2: 14.3% w/w, 24 h, 37 °C (FF2). The pH values decreased to 4.4-4.7, with a predominance of lactic acid bacteria. As in the fermentations in tubes, an increment in the proteolysis degree (TNBS method) (greater for FF2), polypeptide aggregation and a decrease in their solubility, an increase in <2 kDa peptides, and an increase in the Oxygen Radical Absorption Capacity (ORAC) potency of PBS-soluble fractions after fermentation were demonstrated. Also, fermentation increased the proteolysis degree after simulated gastrointestinal digestion (SGID, COST-INFOGEST) with respect to the non-fermented flour digests, with some differences in the molecular composition of the different digests. ORAC and Hydroxyl Radical Averting Capacity (HORAC) potencies increased in all cases. The digest of FF2 (FF2D) presented the greater ORAC value, with higher activities for >4 kDa, as well as for some fractions in the ranges 2-0.3 kDa and <0.10 kDa. Fermentation also increased the 60%-ethanol-extracted phenolic compounds, mainly flavonoids, and the ORAC activity. After SGID, the flavan-3-ols disappeared, but some phenolic acids increased with respect to the flour. Fermentation in condition 2 was considered the most appropriate to obtain a functional antioxidant ingredient.

Physicochemical, Antioxidant, and Antimicrobial Properties of Three Medicinal Plants from the Western Part of the Rhodope Mountains, Bulgaria.

The present study examined the physicochemical, antioxidant, and antimicrobial properties of three medicinal plants: thyme (Thymus callieri Borbás ex Velen), cotton thistle (Onopordum acanthium L.), and hawthorn fruit (Crataegus monogyna Jacq.) from the Western Rhodope Mountains, Bulgaria. The first stage determined the physicochemical characteristics (moisture, ash, carbohydrates, proteins, and vitamin C) of the three herbs. The second stage investigated four types of extracts (aqueous, oil, methanolic, and ethanolic) of each herb and evaluated their total phenolic content, the presence of phenolic compounds (flavonoids and phenolic acids), their antioxidant activity, and antimicrobial properties. Thyme was characterised by the highest ash, protein, and vitamin C content (6.62%, 11.30%, and 571 mg/100 g, respectively). Hawthorn fruit showed the highest moisture and carbohydrate content (8.50% and 4.20%, respectively). The 70% ethanolic extracts of the three herbs exhibited the highest levels of phenolic compounds and, consequently, pronounced antioxidant activity, compared to the other three types of extracts. The aqueous, oil, methanolic, and ethanolic thyme extracts demonstrated the highest total phenolic content-TPC (27.20 mg GAE/g, 8.20 mg GAE/g, 31.70 mg GAE/g, and 310.00 mg GAE/g, respectively), compared to the extracts of the other two plants. These results were consistent with the highest antioxidant activity of the thyme extracts determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, the oxygen radical absorbance capacity (ORAC) assay, and the hydroxyl radical averting capacity (HORAC) assay (except for the oil extract examined using the DPPH method). The results from the high-performance liquid chromatography (HPLC) analysis revealed that the flavonoid quercetin-3-ß-glucoside had the highest concentration in thyme (374.5 mg/100 g), while myricetin dominated in the cotton thistle (152.3 mg/100 g). The phenolic acid content analysis showed prevalence of rosmaric acid in the thyme (995 mg/100 g), whereas chlorogenic acid was detected in the highest concentration in the cotton thistle and hawthorn fruit (324 mg/100 g and 27.7 mg/100 g, respectively). The aqueous, methanolic, and ethanolic extracts showed moderate to high antibacterial potential but limited antifungal activity. None of the oil extracts inhibited the test microorganisms used in the study.

Phytochemical Composition, Antioxidant and Antimicrobial Activity of the Balkan Endemic Micromeria frivaldszkyana (Degen) Velen. (Lamiaceae)

The current study investigates the content of sugars, organic acids, phenolic acids and flavonoids, as well as antioxidant and antimicrobial activity of Balkan-endemic Micromeria frivaldszkyana. Glucose was the most abundant sugar in the plant (2.77%), followed by fructose (1.18%) and galactose (0.82%). Eight organic acids were detected with quinic acid being in the highest content—556.3 mg/100 g DW. From the individual phenolic acids, rosmarinic acid was found in the most significant amounts (2040.1 ± 1.97 mg/100 g) and hesperidin was the major representative of flavonoids with content 131.2 ± 5.6 mg/100 g DW. The antioxidant activity of the plant was studied by six methods: 2,2′-diphenylpicrylhydrazyl (DPPH)—286.4 ± 10.43 mM TE/g, 2,2′azinobis (3)-ethylbenzthiazoline-6-sulfonic acid (ABTS)—358.4 ± 10.4 mM TE/g, ferric reducing antioxidant power (FRAP)—388.0 ± 32.4 mM TE/g, cupric reducing antioxidant capacity (CUPRAC)—905.6 ± 19.2 mM TE/g, Oxygen Radical Absorbance Capacity (ORAC)—3250.5 ± 208.1 µmol TE/g and Hydroxyl Radical Averting Capacity (HORAC)—306.1 ± 23.5 µmol GAE/g. In vitro antimicrobial activity against nine microorganism was evaluated but the extract displayed antimicrobial activity only against Listeria monocytogenes ATCC 19111 with inhibition zone diameter 9 mm and minimal inhibitory concentration (MIC) 10 mg/mL.

Yellow pea flour and protein isolate as sources of antioxidant peptides after simulated gastrointestinal digestion

AbstractAlthough peas are widely consumed legumes throughout the world, the bioactivity of the peptides released by the gastrointestinal digestion has not been sufficiently studied so far. The objective of the present work was to evaluate the potential of flours and protein isolates obtained from two varieties of yellow peas as sources of antioxidant peptides. Flours and protein isolates were prepared and submitted to a simulated gastrointestinal digestion. Protein hydrolysis degree (TNBS method) and protein solubility (in phosphate buffer saline, pH = 7.4) values were independent on the starting material. Antioxidant activity measured by oxygen radical absorbance capacity (ORAC) and hydroxyl radical averting capacity (HORAC) showed no differences between varieties. A lower activity was registered for protein isolates with respect to flours in the case of HORAC, which could be associated with a loss of molecules with molecular masses lower than 43 kDa in the protein isolates. A significant increase in activities was evidenced by both methods after gastrointestinal digestion, except in the case of HORAC activity of flours. Digested from protein isolates presented a greater ratio of molecules smaller than 1.4 kDa and a lower ratio of those larger than 6.5 kDa with respect to digested flours, according to electrophoresis and gel filtration chromatography studies. Results suggested that the presence of other components or/and the initial state of proteins would affect proteolytic attack of digestive enzymes. Both, pea flours and protein isolates, present interesting potential as antioxidants food ingredients.

Synthesis, Cytotoxicity and Antioxidant Activity of New Analogs of RC-121 Synthetic Derivatives of Somatostatin.

Based on the structure of RC-121 (D-Phe-c (Cys-Tyr-D-Trp-Lys-Val-Cys)-Thr-NH2, - synthetic derivatives of somatostatin), some analogs were synthesized and tested for in vitro cytotoxic and antioxidant activity. The new analogs were modifyed at position 5 with Dap (diaminopropanoic acid), Dab (diaminobutanoic acid) and Orn and at position 6 with the unnatural amino acids Tle (t-leucine). The in vitro cytotoxic effects of the substances were investigated against a panel of human tumor cell lines HT-29 (Human Colorectal Cancer Cell Line), MDA-MB-23 (Human Breast Cancer Cell Line), Hep G-2 (Human Hepatocellular Carcinoma Cell Line) and HeLa (cervical cancer cell line). The antioxidant capacities were tested by ORAC (Oxygen Radical Antioxidant Capacity) and HORAC (Hydroxyl Radical Averting Capacity) methods. All substances expressed significantly higher antioxidant capacity by comparison with galic acid and Trolox. All substances showed considerable antioxidant capacity as well. Compound 2T (D-Phe-c(Cys-Tyr-DTrp- Dap-Tle-Cys)-Thr-NH2)had the highest antioxidant effect. The compound 4T (D-Phe-c(Cys-Tyr-D-Trp- Orn-Tle-Cys)-Thr-NH2) displayed antiproliferative effect on HeLa cells with IC50 30 µM. The peptide analog 3T (D-Phe-c(Cys-Tyr-D-Trp-Lys-Tle-Cys)-Thr-NH2) exerted the most pronounced inhibition on the cell vitality up to 53%, 56% and 65% resp. against MDA-MB-23, Hep G-2, HeLa in the higher tested concentration. The somatostatin analogs showed moderate influence on the vitality of different tumor cells and could be used in changing their pathology.

The study investigated the antioxidant potential as well as the inhibitory potential of the seed extracts of Picralima nitida on &alpha;-amylase and &alpha;-glucosidase enzymes. The methanolic, aqueous and coconut water extracts were obtained using 70% methanol, distilled water and coconut water, respectively. Antioxidant properties were studied in vitro using DPPH (1, 1-diphenyl-2-pricrylhydrazyl) radical scavenging, Ferric Reducing Antioxidant Power (FRAP), Total Antioxidant Capacity (TAC), Hydroxyl Radical Averting Capacity (HORAC), Total Phenol Content (TPC), and Total Flavonoid Content (TFC) assays. Different concentrations (0.1 - 0.5 mg/ml) of the extracts were subjected to &alpha;-amylase and &alpha;-glucosidase inhibitory assays using acarbose as standard. Absorbance was measured at 540 (for &alpha;-amylase) and 405 nm (&alpha;-glucosidase). The percentage of &alpha;-amylase and &alpha;-glucosidase inhibitory activity of the extracts and their IC50 values were determined. The seed extracts of P. nitida showed significant antioxidant potential. The inhibitory activity of the extracts on &alpha;-amylase and &alpha;-glucosidas compared favourably with acarbose with the methanolic extract possessing the highest inhibitory activity. The methanolic extract also possessed the highest antioxidant capacity with the lowest IC50 value among the extracts. The results indicate that the seed extracts of P. nitida possess significant antioxidant properties and are effective inhibitors of &alpha;-amylase and &alpha;-glucosidase enzymes. Chemical Composition and Antioxidant Activity of Chaenomeles Maulei Fruit Juice

Summary Chaenomeles maulei is the name of the cultivar of Chaenomeles japonica, which was introduced in Europe. The present study aimed to investigate the chemical composition and the antioxidant activity of Chaenomeles maulei fruit juice (CMFJ). The ingredients of the juice sample were measured by spectrophotometric, high-performance liquid chromatography (HPLC) and gravimetric methods. The antioxidant activity was measured by the oxygen radical absorbance capacity (ORAC) and hydroxyl radical averting capacity (HORAC) assays. The results showed that CMFJ was extremely rich in polyphenolic substances, amongst which the highest was the concentration of procyanidin oligomers, followed by phenolic acids (vanillic &gt; caffeic &gt; chlorogenic &gt; neochlorogenic &gt; p-coumaric &gt; ellagic &gt; ferulic &gt; 4-dihydroxy-benzoic) and flavonoids (epicatechin &gt; catechin &gt; quercetin-3-β-glucoside &gt; quercetin &gt; rutin &gt; naringin &gt; kaempferol &gt; myricetin). The sour taste of the juice and its low pH were due to the high content of organic acids (malic &gt; quinic &gt; citric &gt; shikimic &gt; ascorbic &gt; oxalic). The carbohydrates were presented by glucose &gt; fructose &gt; galactose &gt; xylose &gt; rhamnose &gt; arabinose. The antioxidant activity measured by ORAC and HORAC was very high. It could be attributed to the high content of polyphenolics in the juice.

Protective Effect of a (Poly)phenol-Rich Extract Derived from Sweet Cherries Culls against Oxidative Cell Damage.

Oxidative stress is one of the key phenomena behind the most common types of chronic diseases. Therefore, the modulation of oxidative stress is an interesting target for acting either through prevention or as a therapeutic approach. In this work, a Portuguese variety of cherry (Saco Cherry) was processed in order to obtain a potent in vitro antioxidant phenolic-rich extract (Ch-PRE), which was further explored to evaluate its potential application as nutraceutical agent against cellular oxidative stress damage. Ch-PRE was mainly composed of anthocyanins, particularly cyanidin-3-rutinoside, cyanidin-3-glucoside, peonidin-3-glucoside and neochlorogenic acid, and exhibited a potent chemical antioxidant activity expressed by its oxygen radical absorbance capacity (ORAC) and hydroxyl radical averting capacity (HORAC) values. Ch-PRE also displayed effective intracellular radical scavenging properties in intestinal epithelial and neuronal cells challenged with oxidative stress but showed a different order of effectiveness regarding the modulation of endogenous antioxidant system. Ch-PRE could be an attractive candidate to formulate an agent for the prevention of oxidative stress-induced disorders such as intestinal inflammation disorders or with an appropriated delivery system for neurodegenerative diseases.

Assessment of oxidant/antioxidant status in saliva of cell phone users

Hazardous health effects resulting from exposure to radiofrequency electromagnetic radiation (RF-EMR) emitted from cell phones have been reported in the literature. However, the cellular and molecular targets of RF-EMR are still controversial. The aim of this study was to examine the oxidant/antioxidant status in saliva of cell phone users. Saliva samples collected before using a cell phone as well as at the end of 15 and 30 min calls were tested for two commonly used oxidative stress biomarkers: malondialdehyde (MDA) and 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-Oxo-dG). The 8-oxo-dG levels were determined by enzyme-linked immunosorbent (ELISA) competitive assay, while the MDA levels were measured using the OxiSelect MDA adduct ELISA Kit. The antioxidant capacity of the saliva was evaluated using the oxygen radical absorption capacity (ORAC) and the hydroxyl radical averting capacity (HORAC) assays according to the manufacture instructions. The mean 8-oxo-dG and the Bradford protein concentrations (ng/ml and mg/ml, respectively) peaked at 15 min. The levels of HORAC, ORAC and MDA progressively increased with time and reached maximum at 30 min. However, there was no significant effect of talking time on the levels of 8-OxodG and MDA. Similarly, there was no statistically significant effect of talking time on the oxygen and hydroxyl radicals averting capacities, (ORAC) and (HORAC), respectively. These findings suggest that there is no relationship between exposure to radio frequency radiation (RFR) and changes in the salivary oxidant/antioxidant profile.

Antioxidant Capacity of Pea (Pisum sativum L. cv. Ran) Plants Determined by Different Analytical Methods

The change in the antioxidant capacity (AOC) of pea plants during vegetative growth was followed by means of hydroxyl radical averting capacity (HORAC), oxygen radical absorbance capacity (ORAC) and Folin–Ciocalteau (F–C) assays. During the studied period the pea plants developed all leaf stages before flower formation; root and shoot biomasses enhanced, which was accompanied by an increase of AOC. Highest AOC was localized in the green organs such as leaves, especially those in the growing apical part; stem and root AOC were lower. AOC increased during the organ growth; it was expressed in different magnitude by the three assays. Most likely the assays reflected the antioxidant properties of different compounds. The contribution of potential non-enzymatic antioxidants in the applied assays was discussed.

REVIEW ON DIFFERENT METHODS TO ASSESS THE ANTIOXIDANT ACTIVITY OF SOME COMMON PLANTS OF INDIAN TRADITIONAL MEDICINE

A lot of medicinal plants, traditionally used for thousands of years, are present in a group of herbal preparations of the Indian traditional health care system (Ayurveda) named Rasayana proposed for their interesting antioxidant activities. Among the medicinal plants used in ayurvedic Rasayana for their therapeutic action, some of these have been throughly investigated. In the present paper seven plants (Emblica officinalis L., Curcuma longa L., Mangifera indica L., Momordica charantia L., Santalum album L., Swertia chirata Buch-Ham, Withania somnifera (L.) Dunal) are viewed for their historical, etymological, morphological, phytochemical and pharmacological aspects. The plants described contain antioxidant principles, that can explain and justify their use in traditional medicine in the past as well as the present. In order to identify the plants with antioxidant activity in Ayurveda, a formulation of some rasayanas with well defined antioxidant properties has been examinated. For this purpose, we have considered Sharma's work on the preparation MAK4, MAK5, MA631, MA 471, MA Raja's Cup, MA Student Rasayana, MA Ladies Rasayana. The different methods to assess the antioxidant total peroxyl radical-trapping parameter (TRAP), oxygen radical absorbance capacity (ORAC) and hydroxyl radical averting capacity (HORAC) methods. ORAC, TRAP and HORAC values well correlated with polyphenol content. A good correlation was found also between the methods for measuring antioxidant capacity. Nevertheless, ORAC has been found to be the most sensitive method to measure chain-breaking antioxidant activity. Although we have found a good correlation between TRAP, ORAC and HORAC, using more than one antioxidant assay is recommended for more detailed understanding the principles of antioxidant properties of samples. Antioxidant activity of the giant jellyfish Nemopilema�nomurai measured by the oxygen radical absorbance capacity and hydroxyl radical averting capacity methods

The giant jellyfish Nemopilema nomurai (reaching sizes of up to 2 m diameter and 150 kg), which forms dense blooms, has caused extensive damage to fisheries by overloading trawl nets, while its toxic nematocysts cause dermatological symptoms. Giant jellyfish are currently discarded on the grounds of pest control. However, the giant jellyfish is considered to be edible and is part of Chinese cuisine. Therefore, we investigated whether any benefits for human health may be derived from consumption of the jellyfish in order to formulate medicated diets. Antioxidant activity of Nemopilema nomurai was measured using the oxygen radical absorbance capacity (ORAC) and hydroxyl radical averting capacity (HORAC) methods. Based on the results, the ORAC value of the giant jellyfish freeze-dried sample was 541 µmol trolox equivalent (TE)/100 g and the HORAC value was 3,687 µmol gallic acid equivalent (GAE)/100 g. On the other hand, the IC50 value of hydroxyl radical scavenging activity measured by using the electron spin resonance method was 3.3%. In conclusion, the results suggest that the freeze-dried powder of the giant jellyfish Nemopilema nomurai is a potentially beneficial food for humans.

Studies on the Benefits for Human Health on Fermented Seafood and the Development of Freshness Maintenance Technique of Fish

I and my students have developed some fish sauces made from puffer, whale and sea urchin gonad as the local specialties of Shimonoseki city in west Japan. These sauces were commercialized. Antioxidant actives of these sauces were measured by using chemiluminescence, ESR (electron spin resonance), ORAC (oxygen radical absorbance capacity) and HORAC (hydroxyl radical averting capacity) methods. As the results, these sauces indicated the high antioxidant activities. Furthermore, these sauces were researched by the next-generation food functional estimation method, i.e., the new simultaneous measurement method using cellular signal transduction mechanisms. This method means the simultaneous measurement of chemiluminescence and fluorescence by LED light indicating superoxide anion radical generation and intracellular calcium ion concentration, respectively, of neutrophil-like HL60 culture cells. It was found that some sauces indicated the anti-inflammatory activity in addition to antioxidant activity. On the other hand, I and co-researchers investigated the supply of high quality frozen raw fish by a quick freezing method using alcohol brine. The whole body of Japanese horse mackerel was used. Alcohol brine freezing at -25°C was compared with air freezing at -20°C. Quality of fish meat was analyzed using K value as an indicator of freshness by HPLC, pH, TBARS value as an indicator of lipid oxidation, remaining ethanol by GC/MS and sensory test. As the results, it was indicated that the fish meat frozen quickly by alcohol brine method was superior to that by air freezing.

Solid-phase extraction of berries’ anthocyanins and evaluation of their antioxidative properties

Solid-phase extraction (SPE) was used to obtain anthocyanin-rich extracts from five berry species: chokeberry, elderberry, black currant, blackberry and blueberry. During SPE more than 94.4% of the sugars and more than 88.5% of the acids present in the crude extracts were separated. The SPE resulted in 90–95.6% anthocyanins recovery. The antioxidative properties of the anthocyanin-rich extracts were tested by measuring their oxygen radical absorption capacity (ORAC), hydroxyl radical averting capacity (HORAC), total peroxyl radical trapping antioxidant parameter (TRAP), scavenging of nitric oxide and inhibition of lipid peroxidation. Elderberry extract revealed the highest ORAC value of 5783 μmol TE/g. Chokeberry extract was the most potent inhibitor of lipid peroxidation and had the highest TRAP value of 4051 μmol TE/g. Blueberry extract had the highest HORAC result – 1293 μmol GAE/g and was the most powerful scavenger of NO. The high antioxidant activity according to all antioxidant assays revealed opportunities to apply these preparations as antioxidants.

Quantification of polyphenols and ergothioneine in cultivated mushrooms and correlation to total antioxidant capacity

Agaricus bisporus, Lentinula edodes, Pleurotus ostreatus, Pleurotus eryngii and Grifola frondosa were analyzed for antioxidant capacity, as measured by oxygen radical absorbance capacity (lipophilic and hydrophilic) (ORAC total), hydroxyl radical averting capacity (HORAC), peroxynitrite radical averting capacity (NORAC), superoxide radical averting capacity (SORAC) assays, Folin-Ciocalteu reagent and ergothioneine (ERG) content. ORAC totalvalues ranged from 39 to 138 μmol of Trolox equivalents (TE)/g dry weight (dw). HORAC values ranged from 3.0 to 13.6 μmol of caffeic acid equivalents/g dw. NORAC values ranged from 2.0 to 9.0 μmol TE/g dw. SORAC values ranged from 0.37 to 2.6 kunit superoxide dismutate equivalents/g dw. Polyphenols ranged from 4.2 to 10.6 mg gallic acid equivalents/g dw. A. bisporus mushrooms, especially portabellas, had higher antioxidant capacity relative to the specialty mushrooms tested. ERG ranged from 0.21–2.6 mg/g dw with L. edodes, P. ostreatus, G. frondosa containing a statistically significant greater amount compared to A. bisporus. A good correlation was found between ORAC total and polyphenols ( R 2 = 0.86).

Antioxidant Capacity and Other Bioactivities of the Freeze-Dried Amazonian Palm Berry,Euterpe oleraceaeMart. (Acai)

The fruit of Euterpe oleraceae, commonly known as acai, has been demonstrated to exhibit significantly high antioxidant capacity in vitro, especially for superoxide and peroxyl scavenging, and, therefore, may have possible health benefits. In this study, the antioxidant capacities of freeze-dried acai fruit pulp/skin powder (OptiAcai) were evaluated by different assays with various free radical sources. It was found to have exceptional activity against superoxide in the superoxide scavenging (SOD) assay, the highest of any food reported to date against the peroxyl radical as measured by the oxygen radical absorbance capacity assay with fluorescein as the fluorescent probe (ORACFL), and mild activity against both the peroxynitrite and hydroxyl radical by the peroxynitrite averting capacity (NORAC) and hydroxyl radical averting capacity (HORAC) assays, respectively. The SOD of acai was 1614 units/g, an extremely high scavenging capacity for O2*-, by far the highest of any fruit or vegetable tested to date. Total phenolics were also tested as comparison. In the total antioxidant (TAO) assay, antioxidants in acai were differentiated into "slow-acting" and "fast-acting" components. An assay measuring inhibition of reactive oxygen species (ROS) formation in freshly purified human neutrophils showed that antioxidants in acai are able to enter human cells in a fully functional form and to perform an oxygen quenching function at very low doses.
