A turning point in my current research project was when the reaction intermediate was successfully isolated and characterized. The most challenging part of my job is seeking a glimmer of hope from failed attempts and negative results. My favorite thing about my lab group is group trips and group meetings. I advise my students to always be patient and curious. I'm always in a good mood when I go into my lab. A good day at work starts with a cup of hot tea. If I had one year of paid leave I would travel around the world. When I was a kid I wanted to be a racing driver. My favorite molecule is bis(benzene)chromium, a sandwich compound. Given that an efficient conversion of aryl fluorides into phenols remains to be demonstrated, we started to investigate the feasibility by applying η6-coordination catalysis. In this Angewandte paper, we demonstrate a solution to prepare phenols from aryl fluorides by using a Rh catalyst. We have encountered two significant challenges: one is merging arene exchange and nucleophilic aromatic hydroxylation in one catalytic cycle; the other is tracing the elusive reaction intermediate. AKR1B10 expression in benign prostatic hyperplasia and its related mechanism.

The aim of the present study was to investigate the expression of aldo-keto reductase family 1 member B10 (AKR1B10) in benign prostatic hyperplasia (BPH) and its related mechanism. In total, 142 BPH patients admitted from March 2017 to March 2019 at the First Hospital of Hunan University of Chinese Medicine and 140 healthy people undergoing physical examination were selected as the research subjects. The clinical value of AKR1B10 in BPH was analyzed. Twenty clean SD rats were selected, and 10 were selected to establish the prostate hyperplasia model, while the remaining 10 were set as the control group. Ten days after the model was established, AKR1B10 and NF-κB expression in prostate tissues of rats in both groups was detected by PCR and immunohistochemistry. The primary cells in prostate hyperplasia were cultured, and then they were transfected with AKR1B10 to observe the changes of cell biological behavior. AKR1B10 and NF-κB mRNA significantly increased in peripheral blood of BPH patients and prostate tissue of BPH model rats (P<0.001), and AKR1B10 had good diagnostic value for BPH (P<0.001). In addition, it was positively correlated with PSA, EGF, IL-6 and TNF-α (P<0.001). After transfection with AKR1B10-inhibitor, it was revealed that the proliferation of prostate hyperplasia cells decreased, while the apoptosis of prostate hyperplasia cells increased and the NF-κB protein expression decreased (P<0.001). Collectively, high expression of AKR1B10 in BPH promoted the proliferation of prostate cells and reduced their apoptosis, and the mechanism may be through regulation of NF-κB.

We investigate the entanglement dynamics of two distinguishable two-level systems (TLSs) characterized by energy difference δ located inside a rectangular hollow metallic waveguide of transverse dimensions a and b. The effects of energy difference δ and the inter-TLS distance on the time evolution of the concurrence of the TLSs are examined in the single excitation subspace when the energy separation of the TLS is far away from the cutoff frequencies of the transverse mode.

It is highly delighted to introduce the 2021 7th International Conference on Manufacturing Technology and Applied Materials (ICAMMT 2021), which held on March 26-28, 2021 in Sanya, China. ICAMMT 2021 is to bring together innovative academics and industrial experts in the field of Manufacturing Technology and Applied Materials to a common forum. The primary goal of the conference is to promote research and developmental activities in manufacturing technology and engineering materials, and another goal is to promote scientific information interchange between researchers, developers, engineers, students, and practitioners working all around the world. The conference will be held every year to make it an ideal platform for people to share views and experiences in Manufacturing Technology, Intelligent Manufacturing and Applied Materials and related areas.About 170 participants from academic, high-education institutes and other organizations took part in the conference. The conference model was divided into two sessions, including oral presentations and keynote speeches. In the first part, some scholars, whose submissions were selected as the excellent papers, were given 15 minutes to perform their oral presentations one by one. Then in the second part, keynote speakers were each allocated 30-45 minutes to hold their speeches. We were very honored to have Prof. Ruliang Zhang, from Shandong University of Science and Technology as our committee chair.In the keynote presentation part, we invited five professors as our keynote speakers. Our finale keynote speaker, Prof. Kaichen XU, from School of Mechanical Engineering, Zhejiang University. He devoted in researching Hybrid Advanced Manufacturing of Multifunctional Flexible Sensors & System Integration, Laser Micro/Nano-Processing and other related area.The proceedings present a selection of high-quality papers submitted to the conference by researchers from universities, research institutes, and industry. All papers were subjected to peer-review by conference committee members and international reviewers. The papers were selected based on their quality and their relevance to the conference. A Long-Lasting Public Emotional Trauma after the Recovery of COVID-19: A Full-Year Tracking of Online Public Sentiment

Background: As the outbreak of COVID-19 has caused widespread panic in the general public, it is essential to understand how public sentiment has changed.Methods: In the current study, we continuously tracked online public sentiment through the year 2020 by analyzing emotion reflected on 64,723,242 posts across China’s largest social media platform Sina Weibo.Findings: We found that the pandemic not only affected online public sentiment in the initial outbreak but also induced long-term negative effects even in the aftermath of the successful recovery from the pandemic. The long-term negative effect was not due to the pandemic’s severity either locally or globally, or even the post-pandemic economic recession. Rather, we speculate that the long-term negative effects may have resulted from the prolonged emotional trauma associated with the COVID-19 outbreak.Interpretation: COVID-19 induces long-term negative effects of public sentiments even in the aftermath of the successful recovery from the pandemic. Male inheritance of X-linked liver glycogenosis from an undiagnosed maternal grandfather in a Chinese pedigree: a report of two cases

Abstract Hepatic phosphorylase kinase (PhK) plays an important role in glycogen metabolism by activating phosphorylase. Patients with PhK deficiency may get glycogen storage disease (GSD) type-IXa, an X-linked liver glycogenosis disease. To inform genetic counseling in a family with two affected GSD brothers, we performed a genetic analysis. The GSD in the older brother was confirmed by histological examination of a liver biopsy, which showed glycogen accumulation in liver cells. A liver biopsy was not available from the younger brother. The two patients and their parents were analyzed by whole exome sequencing. A pathogenic mutation in a gene encoding a regulatory subunit of PhK, PHKA2 located on chromosome Xp22, was identified as c.G3373A (p.E1125K) and confirmed by Sanger sequencing. The proband's maternal grandparents and the brothers and sisters of the proband's maternal grandfather were physically examined and genetically tested by Sanger sequencing. Pedigree analysis showed that the mother was a carrier and that the two patients inherited the mutation from their undiagnosed maternal grandfather. Moreover, among the maternal grandfather and four granduncles, three of them possessed the same mutation and four suffered from fatty liver. This is the first report of this mutation causing X-linked liver glycogenosis in a Chinese family and shows that GSD IXa is a mild form of glycogenosis in terms of clinical symptoms, indicating that GSD may be undiagnosed or underestimated. Nevertheless, to provide appropriate intervention and genetic counseling, early identification of the genetic cause is imperative. This study was approved by the Ethics Committee of First Affiliated Hospital, Hunan University of Chinese Medicine (approval No. HN-LL-ZFKY-2018-001-01) on January 12, 2018.

Optimal reactive power dispatch: a bibliometric analysis

Optimal reactive power dispatch (ORPD) plays a significant role in the control and smooth operation of the power system through the enhancement of the network’s reliability, security, and economic aspects. This paper presents a bibliometric and visual analysis of ORPD-related research articles extracted from the Web of Science (WoS) database from its inception to October 29, 2019. A total of 263 articles drawn from 166 journals, published between 1989 and 2019, were retrieved and analysed using Excel, HistCite, and VOSviewer visualisation software. The total number of citations for the 263 articles ranges from 0 to 297. The top three journals with the most significant number of ORPD publications were the International Journal of Electrical Power and Energy Systems, Applied Soft Computing, and two journals qualified for the third place, IEEE Transaction on Power Systems and IET Generation Transmission and Distribution. The most active researcher is Provas Kumar Roy, with nine (9) articles from Kalyani Government Engineering College. The most trending/cited researcher is Yi Jia Cao, with 129 Total Local Citation Scores from Hunan University, Changsha. In terms of contribution by countries, India, China, Iran, and the United States were the most significant contributors with 27.8%, 20.9%, 11.8%, and 8% of the total articles, respectively. The top 5 most frequently used substantive keywords to identify the trending topic and research direction were Particle Swarm Optimisation, Genetic Algorithm, Gravitational Search Algorithm, Linear Programming, Evolutional Algorithm, and Hybrid Algorithm. This study provides a detailed outline and reveals the future research directions for both experienced and novice ORPD researchers to identify research topics, questions, and collaboration partners.

Study on high-speed stability of automobile based on aerodynamic lift feedback

For a long time, the impact of aerodynamic lift on the stability of high-speed vehicles has been studied only from the perspective of the aerodynamic force, and there is a lack of quantitative assessment of the vehicle body. In this paper, a model of multi-body dynamics of the vehicle was constructed by using an in-use vehicle model as the research object. Accuracy was verified using computational fluid dynamics (CFD) and compared with HD-2 (the second wind tunnel of Hunan University) wind tunnel test data. The stability of the vehicle under the condition of high-speed driving was analysed from two aspects of body acceleration and lateral displacement. The results show that the aerodynamic lift at high speed will reduce the maximum lateral acceleration and the maximum yaw acceleration of the vehicle body in the obstacle avoidance condition; and reduce the response speed of the vehicle body to driver's control. The anti-floating front bumper can effectively reduce aerodynamic lift and improve the high-speed driving stability of the vehicles.

Hippocampal insulin resistance and the Sirtuin 1 signaling pathway in diabetes-induced cognitive dysfunction.

In the peripheral nervous system, the activation of Sirtuin 1 can improve insulin resistance; however, the role played by Sirtuin 1 in the central nervous system remains unknown. In this study, rat models of diabetes mellitus were generated by a single injection of streptozotocin. At 8 weeks after streptozotocin injection, the Morris water maze test and western blot assays confirmed that the diabetic model rats had learning and memory deficits, insulin resistance, and Sirtuin 1 expression could be detected in the hippocampus. Insulin and the insulin receptor inhibitor S961 were intranasally administered to investigate the regulatory effects of insulin signaling on Sirtuin 1. The results showed that insulin administration improved the impaired cognitive function of diabetic model rats and increased the expression levels of phosphorylated insulin receptor, phosphorylated insulin receptor substrate 1, and Sirtuin 1 in the hippocampus. Conversely, S961 administration resulted in more severe cognitive dysfunction and reduced the expression levels of phosphorylated insulin receptor, phosphorylated insulin receptor substrate 1, and Sirtuin 1. The Sirtuin 1 activator SRT2104 and the inhibitor Sirtinol were injected into the lateral ventricle, which revealed that the activation of Sirtuin 1 increased the expression levels of target of rapamycin complex 1, phosphorylated cAMP-response element-binding protein, and brain-derived neurotrophic factor. Hippocampal dendritic length and spine density also increased in response to Sirtuin 1 activation. In contrast, Sirtinol decreased the expression levels of target of rapamycin complex 1, phosphorylated cAMP-response element-binding protein, and brain-derived neurotrophic factor and damaged the dendritic structure. These findings suggest that the Sirtuin 1 signaling pathway plays an important role in the development of insulin resistance-related cognitive deficits in diabetic rats. Effects of Astragaloside Ⅳ on exosome secretion and its microRNA-126 expression in human endothelial progenitor cells

Objective: To investigate the effects of Astragaloside Ⅳ on the secretion of exosomes in human endothelial progenitor cells (EPCs) and the expression of microRNA (miRNA)-126 in exosomes. Methods: The umbilical cord blood from one healthy full-term newborn from the Department of Obstetrics and Gynecology of the First Affiliated Hospital of Hunan University of Traditional Chinese Medicine in 2019 was harvested for isolating mononuclear cells by density gradient centrifugation and cultured for 7 days. Morphological observation was performed during this period. Cells of the third passage were collected for identification by CD31 immunomagnetic bead sorting and double fluorescence staining. According to the random number table, the identified EPCs were divided into Astragaloside Ⅳ group and phosphate buffer solution (PBS) group. The cells in Astragaloside Ⅳ group were cultured with Astragaloside Ⅳ in final mass concentration of 100 mg/L for 24 hours, and the cells in PBS group were cultured with the same volume of PBS for 24 hours. After culture, the exosomes from the cell culture supernatant of the two groups were collected, and the expressions of characteristic markers of exosomes CD9, CD63, and CD81 were detected by Western blotting, the morphology of EPC exosomes (EPC-Exos) was observed under transmission electron microscope, and the particle size of EPC-Exos was detected by nanoparticle tracking analysis technique. The concentration of EPC-Exos was determined by dioctyl butyric acid method (the sample number was 3), and the expressions of miRNA-126-3p and miRNA-126-5p related to angiogenesis in EPC-Exos were determined by reverse transcription polymerase chain reaction (the sample number was 3). Data were statistically analyzed with independent sample t test. Results: (1) On the 4th day of culture, the cells began to adhere to the wall, and the multi-forms such as circle, fusiform, and strip appeared at the same time. On the 7th day of culture, the edge of the cells was clear and arranged like a paving stone, the central cells were round, and the surrounding cells were fusiform. (2) CD31 immunomagnetic beads sorting method identification showed that the membrane was stained with green fluorescence and the nucleus was stained with blue fluorescence. Double fluorescence staining method showed that the cells were orange-yellow. The cells were identified as EPCs. (3) After 24 hours of culture, the expressions of CD9, CD63, and CD81 in EPC-Exos were all positive, confirming that EPC-Exos were extracted successfully in this experiment. (4) After 24 hours of culture, the EPC-Exos of the two groups showed round membrane vesicles, and there was no significant difference in morphology. (5) After 24 hours of culture, the particle size of 98.7% EPC-Exos in Astragaloside Ⅳ group was 84.7 to 143.1 nm, and that of 98.0% EPC-Exos in PBS group was 88.7 to 123.5 nm. (6) After 24 hours of culture, the mass concentration of EPC-Exos in Astragaloside Ⅳ group was (310±5) μg/mL, which was significantly higher than (257±5) μg/mL in PBS group, t=13.369, P<0.01. (7) After 24 hours of culture, there were more miRNA-126-3p (t=16.062, P<0.01) and miRNA-126-5p (t=3.252, P<0.05) in EPC-Exos of Astragaloside Ⅳ group than in PBS group. Conclusions: Astragaloside Ⅳ can improve the function of human EPC secretory exosomes, and the secreted exosomes are loaded with miRNA-126.

Abnormal Glu/mGluR2/3/PI3K pathway in the hippocampal neurovascular unit leads to diabetes-related depression.

Our previous studies have shown that glutamate and hippocampal neuron apoptosis are key signals and direct factors associated with diabetes-related depression, and structural and functional damage to the hippocampal neurovascular unit has been associated with diabetes-related depression. However, the underlying mechanism remains unclear. We hypothesized that diabetes-related depression might be associated with the glutamate (Glu)/metabotropic glutamate receptor2/3 (mGluR2/3)/phosphoinositide 3-kinase (PI3K) pathway, activated by glucocorticoid receptors in the hippocampal neurovascular unit. To test this hypothesis, rat hippocampal neurovascular unit models, containing hippocampal neurons, astrocytes, and brain microvascular endothelial cells, were treated with 150 mM glucose and 200 µM corticosterone, to induce diabetes-related depression. Our results showed that under conditions of diabetes complicated by depression, hippocampal neurovascular units were damaged, leading to decreased barrier function; elevated Glu levels; upregulated glucocorticoid receptor, vesicular glutamate transporter 3 (VGLUT-3), and metabotropic glutamate receptor 2/3 (mGluR2/3) expression; downregulated excitatory amino acid transporter 1 (EAAT-1) expression; and alteration of the balance of key proteins associated with the extracellular signal-regulated kinase (ERK)/glial cell-derived neurotrophic factor (GDNF)/PI3K signaling pathway. Moreover, the viability of neurons was dramatically reduced in the model of diabetes-related depression, and neuronal apoptosis, and caspase-3 and caspase-9 expression levels, were increased. Our results suggest that the Glu/mGluR2/3/PI3K pathway, induced by glucocorticoid receptor activation in the hippocampal neurovascular unit, may be associated with diabetes-related depression.
