Effects of photobiomodulation at various irradiances on normal and dihydrotestosterone-treated human hair dermal papilla cells in vitro.

Androgenetic alopecia (AGA) is the most common type of hair loss caused by dihydrotestosterone (DHT) binding to androgen receptors in dermal papilla cells (DPCs). Photobiomodulation (PBM) is a promising treatment for AGA but suffers from inconsistent outcomes and inconsistent effective light parameters. This study investigated the impact of red light at various irradiances on normal and DHT-treated DPCs. Our results suggested that red light at 8 mW/cm2 was most effective in promoting DPCs growth. Furthermore, a range of irradiances from 2 to 64 mW/cm2 modulated key signaling pathways, including Wnt, FGF, and TGF, in normal and DHT-treated DPCs. Interestingly, 8 mW/cm2 had a greater impact on these pathways in DHT-treated DPCs and altered the Shh pathway, suggesting that the effect of PBM varies with the cellular environment. This study highlights specific factors that influence PBM effectiveness and provides insight into the need for personalized PBM treatment approaches.

A Bio‐Functional Wnt3a Gradient Microarray for Cell Niche Studies

AbstractThe Wingless/Integrated (Wnt) signaling is a central player in regulating multiple cellular events ranging from developmental process, adult homeostasis to tumorigenesis. In native microenvironments, Wnt signals often function through the concentration gradients between the secreting site and the targeting site. Hence, presenting Wnt molecules in a concentration gradient in vitro in a controllable manner recapitulates the native Wnt niche. Nevertheless, most in vitro Wnt niche models, where the ligands are homogeneously immobilized on material surface, fail in reconstituting the concentration gradient of Wnt as that in vivo. Herein, using a previously established Multiphoton Microfabrication and Micropatterning (MMM) technology, a concentration gradient of Wnt3a is immobilized on a protein substrate via neutravidin‐biotin interactions, in a quantitatively and spatially controllable manner. Such Wnt3a gradient is bioactive as it triggers a dose‐ and time‐dependent upregulation of β‐catenin and cell proliferation, in model mouse L cells. Most importantly, human hair dermal papilla cells (HHDPCs) cultured in a cell niche with Wnt3a gradient proliferate in a Wnt3a‐gradient‐dependent manner and exhibit a Wnt3a‐gradient‐dependent hair‐inductive phenotype including Wnt/β‐catenin signaling activation and alkaline phosphatase (ALP) expression. This work contributes to the future development of a hair follicle inductivity biochip for screening of drugs and therapeutics on hair regeneration.

HX109 Herbal Extract Promotes Hair Growth by Regulating Cell Cycle in Hair Follicle Dermal Papilla Cells

Purpose: This study aimed to determine the promoting effects of HX109 on hair growth.Methods: We prepared HX109 by extracting Taraxacum officinale, Cuscuta australis, and Nelumbo nucifera with 25% ethanol. The hair growth-promoting effects were measured in C57BL/6J mice which were orally administered HX109 (30, 100, and 300 mg/kg) with dorsal hair depilation. Western blotting was used to examine the protein expression in skin tissues (cyclin family, bax, and bcl-2). Moreover, the WST-1 assay was used to assess the cell viability of human hair dermal papilla cells (HFDPC). Flow cytometry was used to examine the change in the HFDPC cell cycle proportion.Results: In HX109-treated C57BL/6J mice, hair growth was increased in a dose-dependent manner compared with the vehicle-treated group. Specifically, 300 mg/kg of HX109 treated group showed significantly enhanced hair regrowth (p<0.01). Furthermore, HX109 significantly increased cyclin B1 expression and decreased the bax/bcl-2 ratio in the back skin of mice. HX109 also increased cell viability in HFDPCs in a dose-dependent manner by stimulating the S-G2/M phase transition.Conclusion: The mixed herbal extract HX109 has a hair growth-promoting effect by increasing HFDPC viability through regulation of the S-G2/M phase transition.

Effects of Baicalin on Alopecia and the Associated Mechanism.

The aim of the present study was to explore the potential pharmacological mechanism of baicalin by combining network pharmacology prediction and the experimental verification of alopecia. Networks of baicalin-associated targets and alopecia-related genes were constructed using the STRING database. Potential targets and pathways associated with the therapeutic efficacy of baicalin were identified via enrichment analysis using Cytoscape and the database for annotation, visualization and integrated discovery (Metascape). The back hair of C57BL/6J mice was removed with depilatory cream to verify the therapeutic effect of baicalin. Human hair dermal papilla cells (HHDPCs) were used to explore the mechanism of action of baicalin. Network pharmacology analysis revealed that the potential targets of baicalin mainly include protein serine/threonine kinase, Src protein, epidermal growth factor receptor, and insulin-like growth factor 1 (IGF1), which were indicated to mediate neutrophil degranulation and regulation of cell-cell adhesion, vesicle lumen, cytoplasmic vesicle, membrane raft, and endopeptidase activity. Multiple pathways were identified, such as proteoglycans in cancer, PI3K/AKT, and forkhead box O signaling pathways. Following baicalin treatment for the experimental mice, the coverage, length, and weight of the hair increased in a baicalin dose-dependent manner. Moreover, the histological evaluation showed that the number of hair follicles increased after baicalin treatment and melanin formation were pronounced. In addition, baicalin induced an increase in the phosphorylated p-AKT, p-glycogen synthase kinase-3β, p-PI3K, TGF-β1, and vascular endothelial growth factor levels. Furthermore, the activation levels of key protein p-AKT were increased. Baicalin induced the proliferation of HHDPCs in vitro and significantly upregulated p-AKT, IGF1, and alkaline phosphatase. In conclusion, the present study revealed that the pharmacological mechanisms of baicalin in alopecia therapy were associated with the proliferation of DPCs, the activation of the AKT pathway, and the transmission of downstream signals, indicating that baicalin is a potential drug candidate for the clinical treatment of hair loss.

The effects of centipedegrass extract on hair growth via promotion of anagen inductive activity.

To investigate the CGE on hair growth and to explore the mechanism that is involved in the acceleration of anagen induction, we investigated the effects of CGE studied on cell proliferation and molecular mechanism in human hair dermal papilla cells (hDPCs) and keratinocytes (HaCaT cells). Additionally, hair growth evaluation was carried out following topical treatment of the dorsal skin of telogen C57BL/6 mice with CGE for 14 days. As result, CGE increased cell viability and ALP activity in hDPCs. Moreover, CGE increased the expression of catenin beta 1 (CTNNB1), ALP, sex-determining region Y-box 2 (SOX2), insulin-like growth factor 1 (IGF1), and vascular endothelial growth factor A (VEGFA) genes in hDPCs. CGE increased the expression of proteins such as ALP, β-catenin, and phosphorylation of glycogen synthase kinase 3β (pGSK3β), and protein kinase B (pAKT) in hDPCs. Furthermore, CGE induced the proliferation of HaCaT cells and up-regulated AKT-ERK-GSKβ-β-catenin signaling in HaCaT cells. Additionally, the anagen induction effects of CGE were confirmed on the telogen-anagen transition mice model. these findings demonstrated that CGE promoted the entering the growth phase of hair follicle via activation of β-catenin signaling pathways in vivo. Thus, this study suggests that CGE might be a potential therapeutic reagent for hair growth.

Hair Growth-Promoting Mechanisms of Red Ginseng Extract through Stimulating Dermal Papilla Cell Proliferation and Enhancing Skin Health.

This study aimed to investigate the underlying mechanisms of red ginseng extract (RGE) on regulating hair growth and hair follicle development. Results from in vitro studies showed that RGE treatment simultaneously enhanced viability and inhibited apoptosis in human hair dermal papilla cells. Moreover, RGE administration promoted telogen-to-anagen transition, prolonged anagen in hair follicular cycling, and increased the size of hair follicles and skin thickness in a C57BL/6 mouse model. Furthermore, RGE treatment significantly upregulated the expression of β-catenin, phospho-glycogen synthase kinase 3β, cyclin D1, cyclin E, and Bcl-2, phospho-extracellular signal-regulated protein kinase, and phospho-Akt, which are associated with promoting hair growth. In addition, RGE enhanced skin health by activation of antiox-idant defense systems. Our data demonstrates that hair regenerative mechanisms of RGE may be mediated by stimulating dermal papilla cell proliferation and enhancing skin functions.

Application of Ethanol Extracts From Alnus sibirica Fisch. ex Turcz in Hair Growth Promotion.

Alnus sibirica Fisch. ex Turcz (ASFT), belonging to the family of Betulaceae, grows naturally in Asia, Europe, and America. The aims of this study are determining the efficacy of various biomarkers related to hair loss, evaluated by extracting the branch with 60% alcohol, and purely separating diarylheptanoid oregonin, an indicator and active substance, from 60% alcohol extract of the tree. To determine the preventive effects on hair loss, we investigated the anti-oxidative and anti-apoptotic effects on hydrogen peroxide-induced cytotoxicity on human hair dermal papilla cells using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and Western blotting analysis for proving of apoptosis-related marker alteration, respectively. Moreover, we examined the ameliorative effects of 60% alcohol extract of the tree and oregonin against changes of oxidative stress-induced cytokine and testosterone-induced dihydrotestosterone production as crucial pathways of the hair loss mechanism. These results suggest that 60% alcohol extract of the tree and oregonin were available as novel natural materials for maintaining hair health in mammals.

MoS2 Nanoclusters-based biomaterials for disease- impaired wound therapy

Disease-impaired wound healing is one of the most challenging issues clinically, which requires the migration, proliferation and differentiation of multiple types of skin cells, and overcomes the disease-associated problems. To achieve this goal, we set out to firstly synthesize MoS2 (MS) nanoclusters and then to incorporate them into sodium alginate (SA) hydrogel to prepare SA-MS hydrogel as a kind of MoS2 nanoclusters-based biomaterial. The prepared SA-MS hydrogel demonstrates porous microstructure and uniform distribution of Mo elements. The released Mo4+ ions from SA-MS hydrogel exhibit distinct effects on the promotion of the migration and proliferation of human dermal fibroblasts cells (HDFs), human umbilical vein endothelial cells (HUVECs) and human hair dermal papilla cells (HhDPCs) and significantly up-regulated the expression of specific genes in HUVECs and HhDPCs in vitro. SA-MS hydrogel has significant effect on promoting angiogenesis around the wound bed and regeneration of hair follicles of diabetes-impaired wound in C57BL/6 mice diabetic model in vivo by the released Mo4+ ions. Besides, SA-MS hydrogel successfully suppressed skin cancer and realized the skin cancer-impaired wound healing of melanoma-bearing nude mice by the excellent photothermal performance of SA-MS hydrogel in combination of the stimulatory effect of the released Mo4+ ions. Therefore, the prepared SA-MS hydrogel may be one of the promising candidates for disease-impaired wound healing and provides a broad prospect for the development of skin regeneration in the future.The prepared SA-MS hydrogel releases Mo4+ ions to promote the migration and proliferation of HDFs, HVUECs, and HhDPCs. Moreover, the released Mo4+ ions have a significantly promoted role for the diabetes-impaired wound healing and combine with the photothermal performance of the SA-MS hydrogel to affect the healing of skin cancer impaired wound. Eventually, achieve the role of wound healing by promoting angiogenesis and hair follicle regeneration at the wound bed. Therefore, SA-MS hydrogels have an excellent ability of promoting healing for disease-impaired wound.

Blackcurrant Extract with Phytoestrogen Activity Alleviates Hair Loss in Ovariectomized Rats.

Ancocyanin-rich blackcurrant extract (BCE) has phytoestrogen activity; however, its effect on hair follicles is unknown. Additionally, hair loss is known to occur during menopause in women owing to decreased estrogen secretion. This study examined whether BCE alleviated female pattern hair loss using a rat model. RNA was extracted and analyzed using a microarray and ingenuity pathway analysis. A quantitative polymerase chain reaction revealed that 1 μg/mL BCE altered many genes downstream of beta-estradiol in human hair dermal papilla cells. Additionally, the expression of the hair follicle stem cell marker keratin 19 was greatly enhanced. In a menopause model, ovariectomized rats were fed a diet containing 3% BCE for three months. An analysis of the number of hair shafts revealed that BCE increased the number of hairs by 0.5 hairs/follicular unit. Moreover, immunostaining revealed that the expression of Ki67 also increased by 19%. Furthermore, fluorescent immunostaining showed that the expression of other stem cell markers, including keratin 15, CD34, and keratin 19, was induced in rat hair follicular cells. In conclusion, these findings suggest that BCE has phytoestrogen activity in hair follicles and contributes to the alleviation of hair loss in a menopausal model in rats.

Establishment of an in vitro organoid model of dermal papilla of human hair follicle.

Human hair dermal papilla (DP) cells are specialized mesenchymal cells that play a pivotal role in hair regeneration and hair cycle activation. The current study aimed to first develop three-dimensional (3D) DP spheroids (DPS)with or without a silk-gelatin (SG) microenvironment, which showed enhanced DP-specific gene expression, resulting in enhanced extracellular matrix (ECM) production compared with a monolayer culture. We tested the feasibility of using this DPS model for drug screening by using minoxidil, which is a standard drug for androgenic alopecia. Minoxidil-treated DPS showed enhanced expression of growth factors and ECM proteins. Further, an attempt has been made to establish an in vitro 3D organoid model consisting of DPS encapsulated by SG hydrogel and hair follicle (HF) keratinocytes and stem cells. This HF organoid model showed the importance of structural features, cell-cell interaction, and hypoxia akin to in vivo HF. The study helped to elucidate the molecular mechanisms to stimulate cell proliferation, cell viability, and elevated expression of HF markers as well as epithelial-mesenchymal crosstalks, demonstrating high relevance to human HF biology. This simple in vitro DP organoid model system has the potential to provide significant insights into the underlying mechanisms of HF morphogenesis, distinct molecular signals relevant to different stages of the hair cycle, and hence can be used for controlled evaluation of the efficacy of new drug molecules.

Effect of Cinnamomum osmophloeum Kanehira Leaf Aqueous Extract on Dermal Papilla Cell Proliferation and Hair Growth

In this study, we explored the effect of the water extract of Cinnamomum osmophloeum Kanehira (COK) leaves on hair growth by in vitro and in vivo assays. Using an in vitro 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, it was found that the proliferation of rat vibrissae and human hair dermal papilla cells (hDPCs) was significantly enhanced by the COK leaf extract treatment. As determined by quantitative real-time polymerase chain reaction (RT-PCR), the messenger RNA (mRNA) levels of some hair growth–related factors including vascular endothelial growth factor, keratinocyte growth factor (KGF), and transforming growth factor-β2 were found to be higher in the cultured hDPCs exposed to COK leaf extract than those in the untreated control group. In the hair-depilated C57BL/6 mouse model, the stimulation of hair growth was demonstrated in the group of COK leaf extract treatment. Both photographical and histological observations revealed the promotion of the anagen phase in the hair growth cycle by the COK leaf extract in the C57BL/6 mice. Finally, the ultra performance liquid chromatography (UPLC) showed that the COK extract contained mostly cinnamic aldehyde and a small amount of cinnamic acid. The results suggest that the COK leaf extract may find use for the treatment of hair loss.

Wheat Sprout Extract Stimulates Cell Proliferation via Phosphorylation of ERK and Akt in Human Dermal Papilla Cells

We investigated the effect of Wheat Sprout MeOH extract (WSM) on cell proliferation, and analyzed the molecular mechanism in human hair dermal papilla cells (HHDPCs). After exposure to WSM in HHDPCs, cell proliferation was increased compared with minoxidil-treated cells. Treatment with WSM led to phosphorylation of extracellular signal-related kinase (ERK) and Akt. Moreover, WSM-mediated increase of cell proliferation was attenuated by PD98059, ERK inhibitor, and LY294002, Akt inhibitor. These results suggest that WSM induced cell proliferation in HHDPCs through activation of ERK and Akt.

In this study, we investigated the effect of Puerariae Radix ethanol extracts (EPR). The effect of the EPR on proliferation of human hair dermal papilla cells(HHDPCs) by MTT assay and observed Expression of mechanisms that regulate cell proliferation extracellular signal-regulated kinase(ERK) and Akt by western blot. The results showed EPR increased the proliferation of HHDPCs and up-regulation phosphorylation of ERK and Akt. ERK and Akt increased by EPR inhibited phosphorylation by PD98059 (ERK inhibitor) and LY294002 (Akt inhibitor), and cell proliferation was also inhibited. These results suggested EPR increases the proliferation of HHDPCs through phosphorylation of ERK and Akt, and therefore is a beneficial effect for the alopecia treatment.

Cryptomphalus aspersa Mollusc Egg Extract Promotes Regenerative Effects in Human Dermal Papilla Stem Cells.

The aim of this study was to test, by an in vitro approach, whether a natural extract derived from eggs of the mollusc Cryptomphalus aspersa (e-CAF) that seems to present regenerative properties, can enhance the mobilization of human hair dermal papilla cells (HHDPCs) and play a role on tissue repair and regeneration. We have tested HHDPCs proliferation by the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium-bromide (MTT) assay; cell migration by using a wound healing assay, as well as the modulation of the expression of cytoskeletal (F-actin and vimentin) and cell adhesion to the extracellular matrix (ECM) (vinculin and P-FAK) proteins. We also explored whether e-CAF could lead HHDPCs to keratinocytes and/or fibroblasts by evaluating the expression of specific markers. We have compared these e-CAF effects with those induced by TGFβ1, implicated in regulation of cell proliferation and migration. e-CAF promotes proliferation and migration of HHDPCs cells in a time- and dose-dependent manner; it also increases the migratory behavior and the expression of adhesion molecules. These results support the fact that e-CAF could play a role on skin regeneration and be used for the prevention or repair of damaged tissue, either due to external causes or as a result of cutaneous aging.

Identification of key genes induced by platelet-rich plasma in human dermal papilla cells using bioinformatics methods.

Dermal papilla cells (DPCs) are located at the base of hair follicles, and are known to induce hair follicle regeneration. Platelet-rich plasma (PRP) functions in hair follicle regeneration. To investigate the influence of PRP on DPCs, the present study analyzed RNA-seq data of human hair dermal papilla cells (HHDPCs) that were treated or untreated by PRP. The data included in the RNA-seq were from two normal and two treated HHDPC samples. Following identification by Cuffdiff software, differentially expressed genes (DEGs) underwent enrichment analyses, and protein-protein interaction networks were constructed using Cytoscape software. Additionally, transcription factor (TF)-DEG and TF-long non-coding RNA (lncRNA) regulatory networks were constructed. A total of 178 differentially expressed lncRNA were screened, 365 were upregulated and 142 were downregulated. Notably, upregulated cyclin dependent kinase 1 (CDK1) (degree=76), polo-like kinase 1 (PLK1) (degree=65), cell division cycle 20 (degree=50), cyclin B1 (degree=49), aurora kinase B (degree=47), cyclin dependent kinase 2 (degree=46) and downregulated v-myc avian myelocytomatosis viral oncogene homolog (MYC) (degree=12) had higher degrees in networks. In addition, CCAAT/enhancer binding protein β, E2F transcription factor 1 (E2F1), early growth response 1 and MYC may be key TFs for their target genes, and were enriched in pathways associated with the cell cycle. They may also be involved in cell proliferation via various interactions with other genes, for example CDK1-PLK1 and E2F1→CDK1. These dysregulated genes induced by PRP may affect proliferation of HHDPCs.

This study determined the effect of 30% ethanol extract of plant mixture (EPM: Glycyrrhiza uralensis, Angelica gigas, Acorus calamus, Cnidium officinale, Panax ginseng, Camellia sinensis, Salvia miltiorrhiza, Zanthoxylum schinifolium, Carthamus tinctorius, Prunus persica and Scrophularia buergeriana) on promotion of hair growth in human hair dermal papilla cells and C57BL/6J mice. EPM significantly increased the proliferation of human hair dermal papilla cells in a dose- and time- dependent manner (p < 0.05 and p < 0.01). EPM also enhanced mRNA and protein levels of growth factors such as IGF-1, VEGF, KGF and HGF. Moreover, photographical and histological observation showed that application of EPM resulted in the early onset and prolongation of the anagen phase of the hair cycle. In addition, the EPM was revealed to possess potent inhibitory effect on the 5α-reductase activity. Taken together, these results suggest that EPM has hair growth promoting potential and can be used for hair growing products. Identification of a new plant extract for androgenic alopecia treatment using a non-radioactive human hair dermal papilla cell-based assay.

BackgroundAndrogenic alopecia (AGA) is a major type of human scalp hair loss, which is caused by two androgens: testosterone (T) and 5α-dihydrotestosterone (5α-DHT). Both androgens bind to the androgen receptor (AR) and induce androgen-sensitive genes within the human hair dermal papilla cells (HHDPCs), but 5α-DHT exhibits much higher binding affinity and potency than T does in inducing the involved androgen-sensitive genes. Changes in the induction of androgen-sensitive genes during AGA are caused by the over-production of 5α-DHT by the 5α-reductase (5α-R) enzyme; therefore, one possible method to treat AGA is to inhibit this enzymatic reaction.MethodsRT-PCR was used to identify the presence of the 5α-R and AR within HHDPCs. A newly developed AGA-relevant HHDPC-based assay combined with non-radioactive thin layer chromatography (TLC) detection was used for screening crude plant extracts for the identification of new 5α-R inhibitors.ResultsHHDPCs expressed both 5α-R type 1 isoform of the enzyme (5α-R1) and AR in all of the passages used in this study. Among the thirty tested extracts, Avicennia marina (AM) displayed the highest inhibitory activity at the final concentration of 10 μg/ml, as the production of 5α-DHT decreased by 52 % (IC50 = 9.21 ± 0.38 μg/ml).Conclusions Avicennia marina (AM) was identified as a potential candidate for the treatment of AGA based on its 5α-R1-inhibitory activity.

Implication of microRNA regulation in para-phenylenediamine-induced cell death and senescence in normal human hair dermal papilla cells.

Para-phenylenediamine (PPD) is a major component of hair coloring and black henna products. Although it has been largely demonstrated that PPD induces allergic reactions and increases the risk of tumors in the kidney, liver, thyroid gland and urinary bladder, the effect on dermal papilla cells remains to be elucidated. Therefore, the current study evaluated the effects of PPD on growth, cell death and senescence using cell-based assays and microRNA (miRNA) microarray in normal human hair dermal papilla cells (nHHDPCs). Cell viability and cell cycle analyses demonstrated that PPD exhibited a significant cytotoxic effect on nHHDPCs through inducing cell death and G2 phase cell cycle arrest in a dose-dependent manner. It was additionally observed that treatment of nHHDPCs with PPD induced cellular senescence by promoting cellular oxidative stress. In addition, the results of the current study indicated that these PPD-mediated effects were involved in the alteration of miRNA expression profiles. Treatment of nHHDPCs with PPD altered the expression levels of 74 miRNAs by ≥2-fold (16 upregulated and 58 downregulated miRNAs). Further bioinformatics analysis determined that these identified miRNA target genes were likely to be involved in cell growth, cell cycle arrest, cell death, senescence and the induction of oxidative stress. In conclusion, the observations of the current study suggested that PPD was able to induce several cytotoxic effects through alteration of miRNA expression levels in nHHDPCs.

Arctiin blocks hydrogen peroxide-induced senescence and cell death though microRNA expression changes in human dermal papilla cells.

BackgroundAccumulating evidence indicates that reactive oxygen species (ROS) are an important etiological factor for the induction of dermal papilla cell senescence and hair loss, which is also known alopecia. Arctiin is an active lignin isolated from Arctium lappa and has anti-inflammation, anti-microbial, and anti-carcinogenic effects. In the present study, we found that arctiin exerts anti-oxidative effects on human hair dermal papilla cells (HHDPCs).ResultsTo better understand the mechanism, we analyzed the level of hydrogen peroxide (H2O2)-induced cytotoxicity, cell death, ROS production and senescence after arctiin pretreatment of HHDPCs. The results showed that arctiin pretreatment significantly inhibited the H2O2-induced reduction in cell viability. Moreover, H2O2-induced sub-G1 phase accumulation and G2 cell cycle arrest were also downregulated by arctiin pretreatment. Interestingly, the increase in intracellular ROS mediated by H2O2 was drastically decreased in HHDPCs cultured in the presence of arctiin. This effect was confirmed by senescence associated-beta galactosidase (SA-β-gal) assay results; we found that arctiin pretreatment impaired H2O2-induced senescence in HHDPCs. Using microRNA (miRNA) microarray and bioinformatic analysis, we showed that this anti-oxidative effect of arctiin in HHDPCs was related with mitogen-activated protein kinase (MAPK) and Wnt signaling pathways.ConclusionsTaken together, our data suggest that arctiin has a protective effect on ROS-induced cell dysfunction in HHDPCs and may therefore be useful for alopecia prevention and treatment strategies.

Avicequinone C isolated from Avicennia marina exhibits 5α-reductase-type 1 inhibitory activity using an androgenic alopecia relevant cell-based assay system.

Avicennia marina (AM) exhibits various biological activities and has been traditionally used in Egypt to cure skin diseases. In this study, the methanolic heartwood extract of AM was evaluated for inhibitory activity against 5α-reductase (5α-R) [E.C.1.3.99.5], the enzyme responsible for the over-production of 5α-dihydrotestosterone (5α-DHT) causing androgenic alopecia (AGA). An AGA-relevant cell-based assay was developed using human hair dermal papilla cells (HHDPCs), the main regulator of hair growth and the only cells within the hair follicle that are the direct site of 5α-DHT action, combined with a non-radioactive thin layer chromatography (TLC) detection technique. The results revealed that AM is a potent 5α-R type 1 (5α-R1) inhibitor, reducing the 5α-DHT production by 52% at the final concentration of 10 µg/mL. Activity-guided fractionation has led to the identification of avicequinone C, a furanonaphthaquinone, as a 5α-R1 inhibitor with an IC50 of 9.94 ± 0.33 µg/mL or 38.8 ± 1.29 µM. This paper is the first to report anti-androgenic activity through 5α-R1 inhibition of AM and avicequinone C.
