A Study on HPTLC Quantification for Quality Control of Myricetin a Nutraceutical from Different Plant Parts of Myrica esculenta Linn

The present study was performed to report the quantification of myricetin present in different extracts (i.e. ethanol, methanol, petroleum ether, chloroform and aqueous extract) of root, leaf, bark, stem and seed of Myrica esculenta using TLC densitometric method. Densitometric scanning was performed at 366 nm for the determination and quantitation of standard compounds. The compact spots were achieved at Rf 0.67 corresponding to myricetin. The content of myricetin present in different solvent extracts of M. esculenta was 0.2159 ± 0.002% w/w, 0.3139 ± 0.003% w/w, 0.0184 ± 0.004% w/w, 0.0139 ± 0.003% w/w and 0.2120 ± 0.005% w/w in ethanolic, methanolic, petroleum ether, chloroform and aqueous extract of M. esculenta stem, stem, bark, leaf and leaf plant parts, respectively. The highest myricetin content was found in the order of methanolic stem extract>ethanolic stem extract>aqueous leaf extract>petroleum ether bark extract>chloroform leaf extract of M. esculenta. The developed HPTLC method was a precise, simple, convenient and accurate for screening of large number of active compounds present in various solvent extracts. Thus, the study concluded that the developed HPTLC method for quantification of myricetin can be used for a routine quality check and analysis of any formulation and composition containing M. esculenta.

For the first time a simple, sensitive, rapid, precise and accurate HPTLC method has been established for quantification of belleric acid in Terminalia bellerica (TB) fruit pericarp and its formulations. Silica gel plates were used with toluene-ethyl acetate-methanol-formic acid 3:3:1.4:0.2 (v/v) as mobile phase, and densitometric evaluation was performed at 205 nm. The method was validated for precision, repeatability, and accuracy. Average recovery of belleric acid at three levels (98.67, 100.2, and 100.90%) showed the reproducibility of the method was excellent. The calibration plot was linear in the range 250–1250 ng per band. The method could be easily adapted for high-throughput screening and for quality control of TB and its formulations.

Summary In this paper we describe a simple, precise, and accurate HPTLC method for quantification of lupeol in the bark of Mimosoups elengi (ME). The bark was extracted with hot methanol in a Soxhlet apparatus. Chromatographic separation of the extract was performed on aluminum foil plates coated with silica gel 60 F254 as the stationary phase. The mobile phase was toluene-ethyl acetate-formic acid 12:2:1 (υ/υ). Densitometric evaluation of the separated zones was performed at 220 nm. The lupeol was satisfactorily resolved at RF 0.64 ± 0.02. The accuracy and reliability of the method were assessed by evaluation of linearity (1000–4000 ng per band), precision (method and instrumental precision, as RSD, 1.06 and 1.03%, respectively), accuracy (97.28 ± 0.89), and specificity in accordance with ICH guidelines.

Summary This paper describes a new, simple, precise, and accurate HPTLC method for quantification of (−)-epicatechin in the leaves of Cassia fistula. The leaves were separately extracted with methanol and water by both maceration and hot extraction (Soxhlet apparatus). Chromatographic separation of the drug was performed on aluminium foil silica gel 60 F254 plates with toluene-ethyl acetate-formic acid-methanol 20:12:4:4 (v/v) as mobile phase. Densitometric evaluation of the separated zone was performed at 280 nm. Epicatechin in the extract was satisfactorily resolved with RF 0.22 ± 0.02. The accuracy and reliability of the method were assessed by evaluation of linearity (200–800 ng per band), precision (method precision RSD 1.42% and instrumental precision RSD 1.12%), accuracy (98.12%), and specificity in accordance with ICH guidelines.

Swertiamarin is a secoiridoid glucoside present in members of the Gentianaceae family, including Swertia chirata (Wall) Clarke, S. japonica Makino, S. angustifolia Buch.-Ham.ex D. Don, and Enicostemma littorale Blume. It has antidepressant and anticholinergic activity and can thus be used as a biomarker. We have developed a simple HPTLC method for quantification of swertiamarin which can be used for analysis of plant materials and formulations to determine swertiamarin content. The method was validated for precision, repeatability, and accuracy and found to be precise; intra-and inter-day relative standard deviations ( RSD ) were in the ranges 0.68 to 0.85 and 0.71 to 1.03, respectively, for two different concentrations. Instrumental precision and repeatability of the method were 0.95 and 0.69 (% CV ), respectively.
