Synthetic Antimicrobial Peptides. V. Histidine-containing Antifungal Peptides with a “Linear” Type of Amphipathicity

A number of histidine-containing synthetic antifungal peptides with a “linear” type of amphipathicity (SAMP LTA) (F2Hx, H10F2, H10, where x = 7, 10, 13 and 16) have been synthesized and studied. Biological screening of such histidine-containing peptides for their antifungal and hemolytic activity was carried out. It has been shown that the presented histidine-containing SAMP LTAs are capable of effectively inhibiting the growth of opportunistic fungi Candida albicans and have low hemolytic activity in most cases not exceeding 10% even at their relatively high concentration of 400 μM in a medium containing erythrocytes. The antifungal activity of the studied peptides increases with increasing histidine residues in their composition, reaching the maximum value for the histidine-containing peptide F2H16 (MIC50 = 1.0 µM). It has been shown that as the chain length of peptides increases, their hemolytic toxicity also increases. In terms of therapeutic significance, the optimal peptides in the presented series of peptides were F2H10 and F2H13, which have higher selectivity than the short or longer peptides F2H7 or F2H16. The therapeutic index (TI) for these peptides was 233, 247, 79 and 60, respectively. It has been shown that histidine-containing derivatives of peptides with phenylalanine residues at the N-terminus of the peptide (F2H10) are less effective compared to similar peptides (H10F2) containing phenylalanine residues at the C-terminus. Among all the studied peptides, the most active was the H10 peptide (MIC50 = 0.7 µM), which does not contain phenylalanine residues, which in its antifungal activity is not only more effective than all other histidine-containing peptides, including the F2H16 peptide with 16 histidine residues, but also 4-5 times more effective than the antifungal peptide P113 (MIC50 = 3.4 µM), a short active fragment of natural histatin 5, well known in the literature. Due to its relatively low hemolytic and high antifungal activity, the presented histidine-containing SAMP LTAs have relatively high TI values, more than 60. Among all the studied peptides, peptides H10 and P113 have minimal, almost zero, hemolytic activity. However, due to its higher antifungal activity, the selectivity of peptide H10 (TI 1400) exceeds that of peptide P113 (TI 340) by more than 4 times. Thus, peptide H10, due to its high antifungal activity, low hemolytic toxicity and, accordingly, high therapeutic significance, can be used as a promising antifungal peptide drug.

Active droplets through enzyme-free, dynamic phosphorylation.

Life continuously transduces energy to perform critical functions using energy stored in reactive molecules like ATP or NADH. ATP dynamically phosphorylates active sites on proteins and thereby regulates their function. Inspired by such machinery, regulating supramolecular functions using energy stored in reactive molecules has gained traction. Enzyme-free, synthetic systems that use dynamic phosphorylation to regulate supramolecular processes have not yet been reported, to our knowledge. Here, we show an enzyme-free reaction cycle that consumes the phosphorylating agent monoamidophosphate by transiently phosphorylating histidine and histidine-containing peptides. The phosphorylated species are labile and deactivate through hydrolysis. The cycle exhibits versatility and tunability, allowing for the dynamic phosphorylation of multiple precursors with a tunable half-life. Notably, we show the resulting phosphorylated products can regulate the peptide's phase separation, leading to active droplets that require the continuous conversion of fuel to sustain. The reaction cycle will be valuable as a model for biological phosphorylation but can also offer insights into protocell formation.

Fabrication of 3D aligned porous nanocellulose-based aerogel for enrichment of the histidine-containing polypeptides

The high-throughput immobilized metal affinity chromatography (IMAC) material is urgently required to improve the efficiency of separation and purification of tagged peptides in related biomedical applications. Herein, the amino-thiourea-modified nanocellulose (S-TOCN) -polyvinyl alcohol (PVA) composite aerogel with aligned pores was prepared by a directional freeze-casting technique and then used to immobilize metal ions (M2+) for the selective separation and purification of histidine-containing peptides from protein hydrolysates. The obtained S-TOCN-PVA materials with an oriented porous structure exhibited excellent mechanical properties, good adsorption performance, and reusability under optimal conditions. The adsorption capacity of the S-TOCN-PVA aerogel for metal ions was as follows: Cu2+>Ni2+>Zn2+>Co2+, and the higher metal ion chelating amount of the S-TOCN-PVA composite aerogel for Cu2+ and Ni2+ reached 87.21 mg/g at pH=7 and 83.66 mg/g at pH=6 in the 2 h incubation period, respectively. The enrichment effects of Cu2+/S-TOCN-PVA and Ni2+/S-TOCN-PVA aerogels with the selectivity of His-containing peptides from apricot kernel protein hydrolysates were up to 3.99 folds and 4.81 folds, respectively, for one cycle. Furthermore, after 5cycles, the Ni2+/S-TOCN-PVA aerogels remained at 87.5% of their initial adsorption capacity (848 mg/g), and the concentration increased by 39.05-fold. Molecular docking simulations show that the oxygen of the carboxyl group (-C-O and -CO) and the hydrogen of the imidazole group on the His molecule are mainly bonded to the hydrogen of the amino groups and the oxygen of the carboxyl group (C-O-C) in the structure of M2+/S-TOCN-PVA aerogels. The fabricated metal ion-immobilized 3D-aligned porous nanocellulose-based aerogels are promising IMAC materials for efficient separation and purification of the histidine-containing polypeptides from complicated biological samples.

Recent mass-spectrometry based analyses of the "ADP-ribosylome" have identified histidine as ADP-ribose acceptor site. In order to study this modification, a fully synthetic strategy towards α-configured N(τ)- and N(π)-ADP-ribosylated histidine-containing peptides has been developed. Ribofuranosylated histidine building blocks were obtained via Mukaiyama-type glycosylation and the building blocks were integrated into an ADP-ribosylome derived peptide sequence using fluorenylmethyloxycarbonyl (Fmoc)-based solid-phase peptide synthesis. On-resin installation of the ADP moiety was achieved using phosphoramidite chemistry, and global deprotection provided the desired ADP-ribosylated oligopeptides. The stability under various chemical conditions and resistance against (ADP-ribosyl) hydrolase-mediated degradation has been investigated to reveal that the constructs are stable under various chemical conditions and non-degradable by any of the known ADP-ribosylhydrolases.

Decolorization of porcine hemoglobin hydrolysates: The role of peptide characteristics and pH values.

The unpleasant color caused by heme limits the utilization of hemoglobin as a food ingredient. Enzymatic hydrolysis has been used to decolorize hemoglobin, but the underlying mechanisms are poorly understood. The aim of this study was to investigate the decolorization efficiency of porcine hemoglobin using different enzymes and final pH values, and to elucidate their influence on decolorization. Based on higher yields and better decolorization, hemoglobin hydrolysates produced by papain, bromelain, savinase, and protease A were further studied. Compared to hydrolysates by savinase and protease A, a higher proportion of histidine-containing peptides was responsible for better decolorization by papain and bromelain. For all hydrolysates, a moderate reduction in pH to 4.0-5.0 facilitated decolorization of the hydrolysates. Similar peptide profiles of hydrolysates from the same enzyme treatment reflected that pH mainly affected the precipitation of the heme-containing fraction through heme-heme interaction rather than heme-peptide interaction. Overall, this study sheds light on the use of enzymatic hydrolysis to remove the heme group from hemoglobin. PRACTICAL APPLICATION: Slaughterhouses produce tons of protein-rich blood each year. Due to the presence of the heme group in hemoglobin, blood has a dark red color and metallic taste, making it generally unacceptable for consumers. This study provided information on the decolorization of porcine hemoglobin by removing the heme fraction, which should facilitate the utilization of decolored hemoglobin hydrolysates as nutritional food ingredients.

Chloroacetamide-Modified Nucleotide and RNA for Bioconjugations and Cross-Linking with RNA-Binding Proteins.

Reactive RNA probes are useful for studying and identifying RNA-binding proteins. To that end, we designed and synthesized chloroacetamide-linked 7-deaza-ATP which was a good substrate for T7 RNA polymerase in in vitro transcription assay to synthesize reactive RNA probes bearing one or several reactive modifications. Modified RNA probes reacted with thiol-containing molecules as well as with cysteine- or histidine-containing peptides to form stable covalent products. They also reacted selectively with RNA-binding proteins to form cross-linked conjugates in high conversions thanks to proximity effect. Our modified nucleotide and RNA probes are promising tools for applications in RNA (bio)conjugations or RNA proteomics.

PH-Sensitive Targeting of Tumors with Chemotherapy-Laden Nanoparticles: Progress and Challenges.

Accumulating chemotherapeutic drugs such as doxorubicin within a tumor while limiting the drug dose to normal tissues is a central goal of drug delivery with nanoparticles. Liposomal products such as Doxil® represent one of the marked successes of nanoparticle-based strategies. To replicate this success for cancer treatment, many approaches with nanoparticles are being explored in order to direct and release chemotherapeutic agents to achieve higher accumulation in tumors. A promising approach has been stimulus-based therapy, such as the release of chemotherapeutic agents from the nanoparticles in the acidic environments of the tumor matrix or the tumor endosomes. Upon reaching the acidic environments of the tumor, the particles, which are made up of pH-dependent polymers, become charged and release the entrapped chemotherapy agents. This review discusses recent advances in and prospects for pH-dependent histidine-based nanoparticles that deliver chemotherapeutic agents to tumors. The strategies used by investigators include an array of histidine-containing peptides and polymers which form micelles, mixed micelles, nanovesicles, polyplexes, and coat particles. To date, several promising histidine-based nanoparticles have been demonstrated to produce marked inhibition of tumor growth, but challenges remain for successful outcomes in clinical trials. The lessons learned from these histidine-containing particles will provide insight in the development of improved pH-dependent polymeric delivery systems for chemotherapy.

On the Copper-Promoted Backbone Arylation of Histidine-Containing Peptides Using Triarylbismuthines

AbstractWe report herein our detailed investigation on the histidine-directed backbone arylation of histidine-containing peptides using triarylbismuth reagents. The reaction proceeds on the backbone NH of the amino acid that precedes the histidine, the so-called n–1 position. The protocol is applicable to dipeptides where the histidine is located at the C-terminus and to tripeptides where the histidine occupies the central position. The transformation is promoted by copper(II) acetate in the presence of phenanthroline (Phen) and diisopropylethylamine in dichloromethane at 50 °C under oxygen. An excellent scope was observed for the triarylbismuthines. In all cases, the imidazole ring of the histidine is protected with a trityl group to prevent the arylation of the side chain. An ATCUN-like model is proposed to explain the observed results.

Design and Synthesis of Porphyrin-Nitrilotriacetic Acid Dyads with Potential Applications in Peptide Labeling through Metallochelate Coupling.

The need to detect and monitor biomolecules, especially within cells, has led to the emerging growth of fluorescent probes. One of the most commonly used labeling techniques for this purpose is reversible metallochelate coupling via a nitrilotriacetic acid (NTA) moiety. In this study, we focus on the synthesis and characterization of three new porphyrin–NTA dyads, TPP-Lys-NTA, TPP-CC-Lys-NTA, and Py3P-Lys-NTA composed of a porphyrin derivative covalently connected with a modified nitrilotriacetic acid chelate ligand (NTA), for possible metallochelate coupling with Ni2+ ions and histidine sequences. Emission spectroscopy studies revealed that all of the probes are able to coordinate with Ni2+ ions and consequently can be applied as fluorophores in protein/peptide labeling applications. Using two different histidine-containing peptides as His6-tag mimic, we demonstrated that the porphyrin–NTA hybrids are able to coordinate efficiently with the peptides through the metallochelate coupling process. Moving one step forward, we examined the ability of these porphyrin–peptide complexes to penetrate and accumulate in cancer cells, exploring the potential utilization of our system as anticancer agents.

Supramolecular assemblies of histidine-containing peptides with switchable hydrolase and peroxidase activities through Cu(II) binding and co-assembling.

Modulating enzyme activities or functionalities is one of the primary features of biological systems, which is, however, a great challenge for artificial enzyme systems. In this work, we designed and synthesized a series of self-assembling peptides from histidine and other amino acids (Asp, Ser, Lys or Arg), which exist in the active site of natural enzymes. These peptides could undergo a conformational transition from random coils to β-sheet structures under physiological conditions and formed self-assembled nanotubes with obvious hydrolase activities. After incorporation of transition metal ions such as Cu2+, these peptides could coordinate with Cu2+ ions, switch molecular conformations, and self-assemble into hybrid nanomaterials with altered morphologies and peroxidase-like activities. This work illustrates a facile approach for constructing artificial enzymes from self-assembling peptides with histidine residues whose catalytic functions could be modulated by incorporation of Cu2+ ions.

Identifying Protein-Drug Interactions in Cell Lysates Using Histidine Hydrogen Deuterium Exchange.

Identifying the targets of a drug is critical to understand the mechanism of action and predicts possible side effects. The conventional approach is capturing interacting proteins by affinity purification. However, it requires drugs to be immobilized to a solid support or derivatized with chemical moieties used for pulling down interacting proteins. Such covalent modifications to drugs may mask a critical recognition site for or alter the binding affinity to their targets. To overcome the drawback, several methods that do not require covalent modifications to drugs have been developed. These methods identify targets by detecting proteins whose thermodynamic stability is enhanced in the presence of drugs. Although the utility of these methods has been demonstrated, the difficulty in identifying low abundant targets is the common problem of these methods. We have developed a new target identification method that increases the likelihood of identifying low abundant targets. The method uses histidine-hydrogen deuterium exchange (His-HDX) as a readout technique to probe the changes in protein stability induced by drugs. The workflow involves incubating cell lysates in various concentrations of a protein denaturant in the presence and absence of a drug in D2O followed by digestion of the proteins, enrichment of His-containing peptides, and analysis of the enriched His-peptides by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The developed method was successfully applied to identify the interaction between endogenously expressed MAPK14 and its inhibitor in HEK293 cell lysates. The implementation of selective enrichment of histidine-containing peptides in the workflow was a key that enabled identifying the MAPK14-inhibitor interaction.

Modified histidine containing amphipathic ultrashort antifungal peptide, His[2-p-(n-butyl)phenyl]-Trp-Arg-OMe exhibits potent anticryptococcal activity.

In pursuit of ultrashort peptide-based antifungals, a new structural class, His(2-aryl)-Trp-Arg is reported. Structural changes were investigated on His-Trp-Arg scaffold to demonstrate the impact of charge and lipophilic character on the biological activity. The presence and size of the aryl moiety on imidazole of histidine modulated overall amphiphilic character, and biological activity. Peptides exhibited IC50 of 0.37-9.66μg/mL against C.neoformans. Peptide 14f [His(2-p-(n-butyl)phenyl)-Trp-Arg-OMe] exhibited two-fold potency (IC50=0.37μg/mL, MIC=0.63μg/mL) related to amphotericin B, without any cytotoxic effects up to 10μg/mL. Peptide 14f act by nuclear fragmentation, membranes permeabilization, disruption and pore formations in the microbial cells as determined by the mechanistic studies employing Trp-quenching, CLSM, SEM, and HR-TEM. The amalgamation of short sequence, presence of appropriate aryl group on l-histidine, potent anticryptococcal activity, no cytotoxicity, and detailed mechanistic studies directed to the identification of 14f as a new antifungal structural lead.

Key role of histidine residues orientation in affinity binding of model pentapeptides with Ni2+ ions: A theoretical supported experimental study

Understanding the metal ion-protein interactions is crucial in transport, accumulation, and excretion of metals. Furthermore, it seems to be highly important to investigate the role of such complexes in many diseases, such as cancer and neurodegenerative disorders. Unexpectedly, such studies in the systems comprising peptides and Ni2+ ions are still scarce and should be given more importance – particularly considering the toxicity and carcinogenicity of nickel compounds on the living organisms.We report herein the results of a study on the interactions of Ni2+ ions and four pentapeptide analogs: EYHHQ (p1), EHYHQ (p2), HEYHQ (p3), and HEYQH (p4). Potentiometric titration was used to study acid-base properties as well as binding properties of the investigated peptides. Furthermore, stoichiometry of the peptide complexes with Ni2+ ions was evaluated by steady-state fluorescence spectroscopy and potentiometric titration method. Additionally, based on the density functional theory (DFT) calculations, we propose the most likely structures of the complexes of the peptides p1-p4 with Ni2+ ions. It was found that all the peptides form 1:1 and 1:2 thermodynamically stable complexes with Ni2+ cation. In particular, we observed that the interaction between the metal ion and histidine-containing peptide sequences does not depend only on histidines splicing, but also on their orientation in respect to other amino acids in the sequence. Also, the closer proximity of His residue in these short peptides does not increase complex stability, due to stacking interaction between the neighboring imidazole rings.

Direct conjugation of fluorescent quantum dots with E. coli via surface-displayed histidine-containing peptides

Biocompatible approaches to labeling bacteria with fluorescent nanoparticles are essential in order to create living bacterial bioconjugates for imaging, biosensors, medicine, and other applications. Herein we report the direct conjugation of carboxyl quantum dots (QDs) with E. coli outer membrane via surface-displayed binding peptides. The histidine-containing peptide H6G9 was displayed at the N-terminus of membrane-embedded enhanced circularly permuted outer membrane protein X (eCPX) scaffold, which was expressed upon chemical induction. The presence of the binding peptide creates an environment distinct from the negatively charged E. coli surface and provides strong binding affinity to carboxyl quantum dots (QDs). Transmission electron microscopy (TEM) analysis of E. coli-QD bioconjugates revealed high loading densities of these QDs immobilized on the cell surface, even when adding a very low concentration (10 μg/mL) of QDs in order to reduce the cell exposure. These hybrid cells strongly fluoresce with each of the distinct colors of loaded QDs with different emission wavelengths, which can be easily visualized by fluorescence microscopy or differentiated using flow cytometry. Importantly, the E. coli–QD bioconjugates were highly viable and maintained the ability to grow and divide. This study demonstrates a simple, direct, and highly efficient method for labelling bacteria with QDs, without significantly compromising the vitality of the cells.

Chemical Depletion of Histidine-Containing Peptides Allows Identification of More Low-Abundance Methylation Sites from Proteome Samples.

Protein methylation, especially that occurs on arginine and lysine residues, is one of the most important post-translational modifications involved in various cellular processes including RNA splicing, DNA repair, and so forth. Systematic analysis of protein methylation would facilitate the understanding of its regulatory mechanisms. Strong cation chromatography has been used to globally analyze arginine/lysine methylation at the proteome scale with good performance. However, the co-enriched histidine-containing peptides severely interfere with the detection of low-abundance methylpeptides. Here, we developed a novel chemical strategy which enabled almost complete depletion of histidine-containing peptides in the protein digest, thereby resulting in the identification of more low-abundance arginine/lysine methylpeptides. Totally, 333 arginine and lysine methylation forms from 207 proteins were identified in this study. Overall, the number of methylation identifications increased about 50% by using our new method. Preparation of Highly Selective Sorbents Composed of Peptides and Silica Using Novel Molecular Imprinting Technology for Target Metal Ions

A novel molecular imprinting technology was developed to prepare highly selective sorbents, composed of adhesive peptides and microporous silica particles, for the adsorption of target metal ions from aqueous environments. Using solid-phase peptide-synthesis, tyrosine-, lysine-, and histidine-containing peptides were tailored to contain tyrosine-, lysine- and histidine-rich regions. The tyrosine residues of the peptide were converted to 3,4-dihydroxy-L-phenylalanine (DOPA) residues to give the peptides adhesive properties mimicking the adhesion of mussel-foot proteins to wet surfaces. The resulting DOPA-peptides were incubated with target metal ions Ni(II), Cu(II), Co(II), or Zn(II), and subsequently with microporous silica particles, at pH 8.0 and 25°C. Almost all the DOPA-peptides, with the metal ions, strongly attached to the particles within 24 h and were not detached from them upon washing with an aqueous solution of pH 1–12. However, the metal ions were easily released from the particles at a pH below 7.0, implying that imprinted cavities from the target metal ions, created on the peptide-immobilized silica particles, remained after the removal of the template metal ions. The metal adsorption characteristics of the resulting particles, which were termed metal-imprinted peptide–silica particles (metal-iPSPs), were investigated. The target metal ions were adsorbed to metal-iPSPs at a pH of 8.0. The adsorption kinetics fit a pseudo-second-order model well. The adsorption isotherm was well approximated using the Langmuir model, indicating that the metal-iPSP surface was homogeneous, adsorption sites were equivalent, and the coverage formed a monolayer. The metal-iPSPs exhibited a significant and high selectivity for the imprinted target metal ions in the presence of an equimolar quantity of competitive metal ions.

Enrichment of histidine containing peptides by on-line immobilised metal affinity solid-phase extraction capillary electrophoresis-mass spectrometry

The analysis and detection of targeted peptides has received due attention in many fields of proteomics research, such as discovery of biomarkers. In this regard, capillary electrophoresis-mass spectrometry (CE-MS) is widely used, however, direct analysis of low-abundance peptides such as histidine-containing peptides (His-peptides) in complex matrixes by CE-MS remains an analytical challenge. In the present study, an immobilised metal affinity solid-phase extraction containing Ni(II) coupled on-line to capillary electrophoresis-mass spectrometry (IMA-SPE-CE-MS) method has been developed to selectively enrich His-peptides from protein tryptic digests and enhance sensitivity. The method was optimised with α-casein and validated with other standard proteins (β-casein and κ-casein). Later, it was applied to an Escherichia Coli (E. Coli) whole cell lysate. IMA-SPE-CE-MS was very selective and allowed an enrichment factor up to 100-fold. The on-line enrichment and separation method coupled to MS detection is straightforward and advantageous over off-line pretreatment methods in terms of simplicity, cost-effectiveness and throughput.

Activation Effects of Carnosine- and Histidine-Containing Dipeptides on Human Carbonic Anhydrases: A Comprehensive Study.

l-Carnosine (β-Ala-l-His) and several other histidine-containing peptides, including two N-methylated forms on the imidazole ring (l-anserine and l-balenine), two derivatives modified on the carboxyl function (carcinine and l-carnosinamide), two analogues differing in the length of the N-terminal residue (l-homocarnosine and Gly-l-His) and the N-acetyl derivatives, were investigated as activators of four isoforms of the metalloenzyme carbonic anhydrase (CA, EC 4.2.1.1). The four human isoforms hCA I, II, VA and IX were activated in the low to high micromolar range, with a rather complex structure activity relationship. A performed computational study allowed us to rationalize these results and to propose a binding mode of these activators within the enzyme active site. Similarly to other CA activators, the here studied peptides could find relevant pharmacological applications such as in the management of CA deficiencies, for therapy memory and enhancing cognition or for artificial tissues engineering.

Tuning the Aggregation of GHG by Changing Sample Concentration and pH

Self-assembly of peptides is an important issue explored in biomedical, biophysical, and biomaterial research. Small peptides have been heavily researched due to their inherent biodegradability. Low molecular weight di- and tripeptides with aromatic residues and terminal groups have been found to self-assemble into gels. However, despite their aromatic character, short histidine-containing peptides were predicted to exhibit a low aggregation propensity. Contrary to this expectation, we discovered that unblocked GHG forms a hydrogel upon deprotonation of the imidazole side chain. Concomitantly, its apparent pK decreases due to a gelation induced stabilization of its deprotonated state. The apparent pK decreases further with increasing concentration. The sub-millimeter fibril length differs significantly from those of other hydrogel-forming peptides, which are generally on the sub-micrometer scale. We utilized rheology, circular dichroism, and vibrational spectroscopy to study the gelation process and the thermal stability of the gel. Rheology measurements yielded G’ values on the order of 105 kPa. As we increase the pH past the initial point of gelation, the rate of formation and the final G’ value increase. The enhanced circular dichroism of the amide I’ band is diagnostic of long and stable fibrils. The circular dichroism signal indicates anisotropy of the sample. Corresponding IR band profiles indicate a complex mixture of β-sheet and amorphous aggregate structures. Our results show that histidine can be used as a switch for the formation of large supramolecular structures even in very short peptides and that the properties of the gel can be tuned by altering the concentration and pH of the sample.
