Analysis types and quantification methods applied in UHPLC-MS metabolomics research: a tutorial.

Different types of analytical methods, with different characteristics, are applied in metabolomics and lipidomics research and include untargeted, targeted and semi-targeted methods. Ultra High Performance Liquid Chromatography-Mass Spectrometry is one of the most frequently applied measurement instruments in metabolomics because of its ability to detect a large number of water-soluble and lipid metabolites over a wide range of concentrations in short analysis times. Methods applied for the detection and quantification of metabolites differ and can either report a (normalised) peak area or an absolute concentration. In this tutorial we aim to (1) define similarities and differences between different analytical approaches applied in metabolomics and (2) define how amounts or absolute concentrations of endogenous metabolites can be determined together with the advantages and limitations of each approach in relation to the accuracy and precision when concentrations are reported. The pre-analysis knowledge of metabolites to be targeted, the requirement for (normalised) peak responses or absolute concentrations to be reported and the number of metabolites to be reported define whether an untargeted, targeted or semi-targeted method is applied. Fully untargeted methods can only provide (normalised) peak responses and fold changes which can be reported even when the structural identity of the metabolite is not known. Targeted methods, where the analytes are known prior to the analysis, can also report fold changes. Semi-targeted methods apply a mix of characteristics of both untargeted and targeted assays. For the reporting of absolute concentrations of metabolites, the analytes are not only predefined but optimized analytical methods should be developed and validated for each analyte so that the accuracy and precision of concentration data collected for biological samples can be reported as fit for purpose and bereviewed by the scientific community.

Studies on Nigerian cashew nut shell liquid: Greening-up extraction process, chemical composition and cost effectiveness

Extractions of cashew nut shell liquid (CNSL) have always widely been carried out with non-environmentally friendly solvents. Hence, greener solvents are desired as alternative to currently used solvents. Herein, batch extractions of Nigerian CNSL with greener solvents: ethyl acetate and cyclohexane mixture in different ratios were investigated in hot and cold system and results compared with conventional solvent (petroleum ether). Compositional studies of the extracted CNSL and its isolated components were done with Fourier-Transformed Infrared (FTIR), Nuclear Magnetic Resonance (NMR) spectroscopy and Ultra High-Performance Liquid Chromatography-Mass Spectrometry (UHPLC-MS). Cost effectiveness analysis for the extraction solvents and methods was also investigated. It was revealed that ethyl acetate and ethyl acetate-cyclohexane mixture at cold and hot conditions recorded higher CNSL yields than previous reports even at a lower liquid-to-solid ratio. Chemical composition and component quantity extracted did not significantly change under hot or cold system. The present composition of the Nigerian CNSL contains unusually higher amount of anarcardic acid (79 %) and cardanol (14 %) than ever reported. Additionally, degree of unsaturation in the alkyl chain of the CNSL is in the order triene>monoene>diene in the anarcardic acid and cardanol components of the liquid. Cost effectiveness analysis at a kilogramme scale revealed that the order of greenness of extraction route based on solvent type, energy requirement and condition of extraction is ethyl acetate-solvent cold system> ethyl acetate-hot system> ethyl acetate-cyclohexane-system> cyclohexane system > pet. ether system.

Prostacyclin Synthase Deficiency Leads to Exacerbation or Occurrence of Endothelium-Dependent Contraction and Causes Cardiovascular Disorders Mainly via the Non-TxA2 Prostanoids/TP Axis.

Prostaglandin I2 synthesized by endothelial COX (cyclooxygenase) evokes potent vasodilation in some blood vessels but is paradoxically responsible for endothelium-dependent constriction (EDC) in others. Prostaglandin I2 production and EDC may be enhanced in diseases such as hypertension. However, how PGIS (prostaglandin I2 synthase) deficiency affects EDC and how this is implicated in the consequent cardiovascular pathologies remain largely unknown. Experiments were performed with wild-type, Pgis knockout (Pgis-/-) and Pgis/thromboxane-prostanoid receptor gene (Tp) double knockout (Pgis-/-Tp-/-) mice and Pgis-/- mice transplanted with unfractionated wild-type or Cox-1-/- bone marrow cells, as well as human umbilical arteries. COX-derived prostanoids were measured by high-performance liquid chromatography-mass spectrometry. Vasomotor responses of distinct types of arteries were assessed by isometric force measurement. Parameters of hypertension, vascular remodeling, and cardiac hypertrophy in mice at different ages were monitored. PGF2α, PGE2, and a trace amount of PGD2, but not thromboxane A2 (TxA2), were produced in response to acetylcholine in Pgis-/- or PGIS-inhibited arteries. PGIS deficiency resulted in exacerbation or occurrence of EDC ex vivo and in vivo. Endothelium-dependent hyperpolarization was unchanged, but phosphorylation levels of eNOS (endothelial nitric oxide synthase) at Ser1177 and Thr495 were altered and NO production and the NO-dependent relaxation evoked by acetylcholine were remarkably reduced in Pgis-/- aortas. Pgis-/- mice developed high blood pressure and vascular remodeling at 16 to 17 weeks and subsequently cardiac hypertrophy at 24 to 26 weeks. Meanwhile, blood pressure and cardiac parameters remained normal at 8 to 10 weeks. Additional ablation of TP (TxA2 receptor) not only restrained EDC and the downregulation of NO signaling in Pgis-/- mice but also ameliorated the cardiovascular abnormalities. Stimulation of Pgis-/- vessels with acetylcholine in the presence of platelets led to increased TxA2 generation. COX-1 disruption in bone marrow-derived cells failed to affect the development of high blood pressure and vascular remodeling in Pgis-/- mice though it largely suppressed the increase of plasma TxB2 (TxA2 metabolite) level. Our study demonstrates that the non-TxA2 prostanoids/TP axis plays an essential role in mediating the augmentation of EDC and cardiovascular disorders when PGIS is deficient, suggesting TP as a promising therapeutic target in diseases associated with PGIS insufficiency.

ZIF-67/melamine-derived cobalt encapsulated in N-doped CNTs for periodate activation toward tetracycline degradation

Advance oxidation processes (AOPs) based on periodate (PI) offer new methods for removing pollutants, but the activation of PI by carbon nanotubes and its composites has been rarely reported. In this study, N-doped carbon nanotubes encapsulated cobalt (Co@NCNTs) was synthesized through melamine assisted ZIF–67 pyrolysis, and it was used to activate PI for the tetracycline (TC) degradation. Effects of experimental parameters including catalyst dosage, periodate concentration, initial pH, and temperature were investigated. The results show that this system exhibited excellent activation performance under wide pH (4–10) with a low reaction activation energy (Ea = 8.874 kJ/mol). Radical quenching experiments manifested that iodate radical (IO3) played a main role in the TC elimination, followed by superoxide radical (O2−), singlet oxygen (1O2), and hydroxyl radicals (OH). Additionally, it is found that IO4− is not converted into toxic iodine species (I2, HOI) but nontoxic iodate (IO3−), indicating that no secondary pollution occurred. Finally, possible TC degradation pathways were proposed based on the results of high-performance liquid chromatography mass spectrometry (HPLC-MS) analysis. This article provides a novel approach for the activation PI to remove pollutants and a theoretical support for practical applications.

Occurrence, removal and risk assessment of chemicals of emerging concern in selected rivers and wastewater treatment plants in western Kenya

Water resources play a crucial role in sustaining life on earth yet chemicals of emerging concern (CECs) arising from extensive human applications are an increasing threat towards their existence. In this study, we examined the occurrence, removal and potential risk of CECs found in rivers and wastewater treatment plants (WWTPs) in western Kenya. Samples were prepared by solid-phase extraction and analysed using high performance liquid chromatography-mass spectrometry with a target list of 785 compounds. Out of these, 333 and 352 (influent 322, effluent 265) compounds were quantified in rivers and wastewater respectively, with pharmaceuticals, industrial compounds, and pesticides being frequently detected in both rivers and WWTPs. Compounds with highest concentrations included saccharin (9.9 μg/L), metformin (7.5 μg/L), and oxypurinol (6.5 μg/L) in rivers whereas caffeine (280 μg/L), deoxycholic acid (179 μg/L), 2-oxindole (10.9 μg/L) and ibuprofen (8.1 μg/L) were found at high concentrations in WWTPs. Based on the types of crops grown, samples from maize growing regions recorded the highest number of pesticides (75) which coincided with the spraying season. The WWTP showed the capacity to eliminate some compounds although the removal efficiencies varied greatly with 204 compounds exhibiting an average removal efficiency exceeding 50 %. Based on the risk assessment, crustaceans had the highest potential risk for toxicity with toxic unit (TU) values up to 5.4 driven primarily by diazinon and dichlorvos followed by algae (TU up to 0.07) and fish (TU up to 0.01) in rivers. A similar trend was observed in WWTP with diazinon (TU up to 5.5), diuron (TU up to 0.07) and carbendazim (TU up to 0.006) driving the risk for crustaceans, algae and fish respectively. These findings highlight the significance of surface water and WWTPs as sources and sinks of CECs in the environment translating to potential risks on aquatic organisms and humans.

Pharmacokinetics of anti-Mycobacterium avium-intracellulare disease drugs in silkworms

Pulmonary Mycobacterium avium-intracellulare complex (MAC) disease is a typical non-tuberculous mycobacterial infection. The incidence of pulmonary MAC is increasing worldwide. This study aimed to clarify the pharmacokinetic parameters of anti-pulmonary MAC disease drugs in silkworms. The pharmacokinetic parameters investigated included maximum concentration, area under the concentration–time curve, total clearance, and volume of distribution at steady-state. In addition, protein-binding rates, fat body transferability, and drug-drug interactions were examined. Antibiotic concentrations were measured using a validated high-performance liquid chromatography-mass spectrometry method. Among the antibiotics investigated, amikacin was not eliminated from silkworms during the 48-h observation period. In contrast, dose-proportional pharmacokinetics were observed in silkworms for all antibiotics tested, except for amikacin. Protein-binding rates in hemolymph for clarithromycin, azithromycin, rifampicin, ethambutol, and amikacin were 39.6 ± 3.0%, 39.5 ± 4.3%, 76.3 ± 3.2%, 20.9 ± 4.2%, and 73.1 ± 4.7%, respectively (mean ± standard deviation). The distribution of antibiotics in the fat bodies of silkworms was related to drug lipophilicity. No drug-drug interactions were observed in the silkworms. The pharmacokinetics of these drugs in silkworms differed significantly from those in humans. Therefore, while it is challenging to predict the pharmacokinetics of these drugs in humans based on silkworm data, the silkworm infection model has facilitated a comprehensive assessment of the relationship between antibiotic exposure and efficacy.

Solvothermal preparation of CuO/g-C3N4 and photocatalytic degradation of ciprofloxacin

Composite materials CuO/g-C3N4 (CuOCN) were prepared using a solvothermal method to investigate their degradation effect on ciprofloxacin (CIP). The materials were characterized through X-ray diffraction analysis (XRD), Fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), UV-visible diffuse reflectance spectrophotometry (DRS), and X-ray photoelectron spectroscopy (XPS). The toxicity of degradation products of CIP was assessed through antibacterial experiments, and the degradation mechanism of CIP was investigated using quenching agent capture and high-performance liquid chromatography-mass spectrometry (HPLC-MS). The results of XRD and FTIR indicate that the CuOCN composite has been successfully synthesized. The photocatalytic experiment shows that adding 15% (w/w) of Cu3(BTC)2(H2O)3 in melamine (CuOCN2) has a relatively good degradation effect on CIP and can be reused multiple times. The TEM results indicate that both CuO and g-C3N4 have a layered structure and are attached together. HRTEM analysis reveals that the two components in CuOCN2 are arranged in a layered structure, with CuO displaying two crystal planes. Energy Dispersive Spectrometer (EDS) analysis reveals that the proportion of CuO in the three composite materials is 7.86% (w/w), 15.93% (w/w), and 84.83% (w/w), respectively. DRS detection reveals that the greater the amount of CuO added, the broader the visible light absorption range of the composite material, and the narrower the bandgap width. The photocurrent and photoresistance indicate that CuOCN2 exhibits a good photoelectric effect. XPS analysis shows that the CuOCN2 composite material possesses both the chemical composition and structure of CuO and g-C3N4. During the catalytic degradation process of CIP, four active substances, h+, e−, ·O2 −, and ·OH, were produced, leading to a reduction in the toxicity of the degradation products. Through HPLC-MS analysis, it was found that CIP undergoes reactions such as defluorination, ring opening of piperazine, and quinolone during degradation, resulting in reduced toxicity. This study provides an experimental basis for the degradation of CIP by CuOCN2.

Visible-Light Photocatalytic Degradation Efficiency of Tetracycline and Rhodamine B Using a Double Z-Scheme Heterojunction Catalyst of UiO-66-NH2/BiOCl/Bi2S3.

BiOCl is a promising photocatalyst, but due to its weak visible light absorption capacity and low photogenerated electron-hole pair separation rate, its practical application is limited to a certain extent. In this study, a novel double Z-scheme heterojunction UiO-66-NH2/BiOCl/Bi2S3 catalyst was constructed to broaden the visible light response range and promote high photogenerated hole-electron separation of BiOCl. Its photocatalytic performance is evaluated by dissociating tetracycline (TC) and rhodamine B (RhB) in visible light. The optimal proportion of UiO-66-NH2/BiOCl/Bi2S3 hybrids exhibits the best degradation efficiency of visible light illumination (∼93% in 120 min for TC and ∼98% in 60 min for RhB). The synergistic effect of a large visible light response range and the Z-scheme charge transfer mechanism ensure the excellent visible photocatalytic activity of UiO-66-NH2/BiOCl/Bi2S3. It is proven that h+ and •O2- are the main active substances in the photocatalysis process by active substance capture experiments and electron spin resonance tests. The intermediates and degradation processes are analyzed by high-performance liquid chromatography-mass spectrometry. This study proves that the new UiO-66-NH2/BiOCl/Bi2S3 photocatalyst has great application potential in the field of water pollution degradation and will provide a new idea for the optimization of BiOCl.

Enhanced solar light-driven photocatalysis of norfloxacin using Fe-doped TiO2: RSM optimization, DFT simulations, and toxicity study.

This study investigates the photocatalytic degradation of norfloxacin (NFX) utilizing Fe-doped TiO2 nanocomposite under natural sunlight. TiO2-based photocatalysts were synthesized using chemical precipitation varying Fe-dopant concentration and characterized in detail. Theoretical modelling, centred on density functional theory (DFT), elucidated that Fe ions within the TiO2 lattice are effectively confined, thereby narrowing the wide band gap of TiO2. The findings strongly support that Fe3+ ions augmented the photocatalytic activity of TiO2 by facilitating an intermediate interfacial route for electron and hole transfer, particularly up to an optimal dopant concentration of 1.5M%. Subsequently, a three-level Box-Behnken design (BBD) was developed to determine the initial pH, optimal catalyst concentration, and drug dosage. High-performance liquid chromatography-mass spectrometry (HPLC-MS) was employed to identify reaction intermediates, thereby establishing a potential degradation pathway. Notably, sustained recyclability was achieved, with 82% degradation efficiency maintained over five cycles. Additionally, the toxicity of degradation intermediates was evaluated through bacterial and phytotoxicity tests, affirming the environmental safety of treated water. In vitro toxicity of the nanomaterial was also examined, emphasizing its environmental implications. Scavenger experiments revealed that hole and hydroxyl radicals were the primary active species in Fe-TiO2-based photocatalysis. Furthermore, the antibacterial potential of the synthesized catalyst was assessed using Escherichia coli and Staphylococcus aureus to observe their respective antibacterial responses.

Effects of Renal Denervation on Gap Junction in High-Pacing-Induced Heart Failure Dogs.

Gap junction remodeling is an important cause of ventricular arrhythmia in heart failure. However, it remains unclear whether renal denervation (RDN) regulates gap junction remodeling in heart failure. To explore the effect of RDN on gap junction remodeling in dogs with high-pacing-induced heart failure. Fifteen dogs were randomly divided into control (n = 5), heart failure (HF) (n = 5), and RDN+HF (n = 5) group. A high-pacing-induced-heart failure model was established using rapid right ventricular pacing for 4 weeks. The RDN+HF group underwent surgical and chemical ablation of both renal arteries before 4 weeks rapid right ventricular pacing. After 4 weeks, echocardiography, High-Performance Liquid Chromatography-Mass Spectrometry test for norepinephrine and epinephrine, and pathological analysis were performed in the above 3 groups. Further, immunohistochemical staining was used to detect tyrosine hydroxylase, ChaT, connexin 43 (Cx43), and connexin 40 (Cx40). Connexin 43 and Cx40 expression was detected by western blotting. Transmission electron microscopy was used to observe the gap junction. Compared to the control group, myocardial fibrosis and sympathetic hyperactivity were observed in the HF group. Immunohistochemical staining and western blotting showed that Cx40 expression and Cx43 expression was significantly reduced in the HF group. Compared with the HF group, the RDN+HF group showed reduced sympathetic hyperactivity, Cx40 expression, Cx40/Cx43 ratio, and increased Cx43 expression. Renal denervation alleviates gap junction remodeling in high-pacing-induced heart failure dogs.

Association of 1-bromopropane exposure with asthma prevalence: A Korean National Health and Nutritional Examination Survey (2020–2021)-based study1

Exposure to 1-bromopropane (1-BP) is an emerging environmental and health concern due to its increasing environmental prevalence. Although the health effects of 1-BP exposure have been under-recognized, current evidence suggests the possibility of adverse pulmonary health effects due to 1-BP exposure. However, the association between 1-BP exposure and asthma prevalence remains unclear. Thus, we aimed to examine the association between 1-BP exposure and asthma prevalence in the general population. Using nationally representative data, we explored the potential impacts of indoor air quality (IAQ)-related behavioral factors on the level of 1-BP exposure. This study included 1,506 adults from the 2020-2021 Korea National Health and Nutrition Examination Survey. The prevalence of asthma was based on self-reported physician-diagnosed asthma. Urinary N-acetyl-S-(n-propyl)-L-cysteine (BPMA) levels were measured as a biomarker of 1-BP exposure, using high-performance liquid chromatography-mass spectrometry. Multiple logistic regression models were performed to investigate the associations between urinary BPMA metabolite and asthma prevalence after adjusting for potential confounders. Log-linear multiple regression models were used to examine the association between IAQ-related behavior and urinary BPMA concentration. Forty-seven individuals with asthma and 1,459 without asthma were included. Individuals in the highest quartile of urinary BPMA concentration had a 2.9 times higher risk of asthma than those in the lowest quartile (odds ratio [OR]: 2.85, 95% confidence interval [CI]: 1.02-7.98). The combination of natural and mechanical ventilation was associated with a reduced urinary BPMA concentration. Our findings suggest that 1-BP exposure is associated with the prevalence of asthma in adults and revealed higher urinary levels of BPMA in our study population compared to those in other countries. Given the emerging importance of IAQ, actively managing and modifying behavioral patterns to reduce 1-BP exposure in indoor environments could substantially attenuate the risk of asthma-related to 1-BP exposure.

Unraveling the independent role of METTL3 in m6A modification and tumor progression in esophageal squamous cell carcinoma

METTL3 and METTL14 are traditionally posited to assemble the m6A methyltransferase complex in a stoichiometric 1:1 ratio, modulating mRNA fate via m6A modifications. Nevertheless, recent investigations reveal inconsistent expression levels and prognostic significance of METTL3 and METTL14 across various tumor types, challenging their consistent functional engagement in neoplastic contexts. A pan-cancer analysis leveraging The Cancer Genome Atlas (TCGA) data has identified pronounced disparities in the expression patterns, functional roles, and correlations with tumor burden between METTL3 and METTL14, particularly in esophageal squamous cell carcinoma (ESCC). Knockdown experiments of METTL3 in EC109 cells markedly suppress cell proliferation both in vitro and in vivo, whereas METTL14 knockdown shows a comparatively muted effect on proliferation and does not significantly alter METTL3 protein levels. mRNA sequencing indicates that METTL3 singularly governs the expression of 1615 genes, with only 776 genes co-regulated with METTL14. Additionally, immunofluorescence co-localization studies suggest discrepancies in cellular localization between METTL3 and METTL14. High-performance liquid chromatography–mass spectrometry (HPLC–MS) analyses demonstrate that METTL3 uniquely associates with the Nop56p-linked pre-rRNA complex and mRNA splicing machinery, independent of METTL14. Preliminary bioinformatics and multi-omics investigations reveal that METTL3’s autonomous role in modulating tumor cell proliferation and its involvement in mRNA splicing are potentially pivotal molecular mechanisms. Our study lays both experimental and theoretical groundwork for a deeper understanding of the m6A methyltransferase complex and the development of targeted tumor therapies focusing on METTL3.

Mycoremediation of Acetaminophen by the Fungi of Dothideomycetes sp. Isolate

Resources like air, water, and land are found to be contaminated by a broad variety of pharmaceutical contaminants. The release of industrial effluents, hospital wastewater, and untreated domestic water, which contains medications, colours, pharmaceutical blisters, and heavy metals that are ecotoxic even at concentrations of nanograms to micrograms, is a significant cause of water pollution. Mycoremediation is an operative approach is to detoxify these contaminants. The fungal strain was isolated from pharma industrial wastewater. Identification of the fungi was founded on the morphological and 18s rRNA sequencingtechnique. Isolate shared 99.82 % similarity with Zasmidium cellare and Cladosporium iridis strain CBS 138.40, suggesting the new species of fungi. The strain has received Gen bank accession number of OR081620.1. The fungal strain was found to degrade 85% of acetaminophen (1000 ppm) in 10 days when grown in mineral salt medium with acetaminophen as only carbon source. Glucose was found to enhance sporulation potential but not the degradative potential of the fungi. Identification of biodegradative metabolite was done by using thin layer chromatography, high performance liquid chromatography and liquid chromatography-mass spectroscopy. Mycodegradative metabolite of 4 Aminophenol was identified as acetaminophen.

Applications of high performance liquid chromatography-mass spectrometry in proteomics

Proteomics profiling plays an important role in biomedical studies. Proteomics studies are much more complicated than genome research, mainly because of the complexity and diversity of proteomic samples. High performance liquid chromatography-mass spectrometry (HPLC-MS) is a fundamental tool in proteomics research owing to its high speed, resolution, and sensitivity. Proteomics research targets from the peptides and individual proteins to larger protein complexes, the molecular weight of which gradually increases, leading to sustained increases in structural and compositional complexity and alterations in molecular properties. Therefore, the selection of various separation strategies and stationary-phase parameters is crucial when dealing with the different targets in proteomics research for in-depth proteomics analysis. This article provides an overview of commonly used chromatographic-separation strategies in the laboratory, including reversed-phase liquid chromatography (RPLC), hydrophilic interaction liquid chromatography (HILIC), hydrophobic interaction chromatography (HIC), ion-exchange chromatography (IEC), and size-exclusion chromatography (SEC), as well as their applications and selectivity in the context of various biomacromolecules. At present, no single chromatographic or electrophoretic technology features the peak capacity required to resolve such complex mixtures into individual components. Multidimensional liquid chromatography (MDLC), which combines different orthogonal separation modes with MS, plays an important role in proteomics research. In the MDLC strategy, IEC, together with RPLC, remains the most widely used separation mode in proteomics analysis; other chromatographic methods are also frequently used for peptide/protein fractionation. MDLC technologies and their applications in a variety of proteomics analyses have undergone great development. Two strategies in MDLC separation systems are mainly used in proteomics profiling: the "bottom-up" approach and the "top-down" approach. The "shotgun" method is a typical "bottom-up" strategy that is based on the RPLC or MDLC separation of whole-protein-sample digests coupled with MS; it is an excellent technique for identifying a large number of proteins. "Top-down" analysis is based on the separation of intact proteins and provides their detailed molecular information; thus, this technique may be advantageous for analyzing the post-translational modifications (PTMs) of proteins. In this paper, the "bottom-up" "top-down" and protein-protein interaction (PPI) analyses of proteome samples are briefly reviewed. The diverse combinations of different chromatographic modes used to set up MDLC systems are described, and compatibility issues between mobile phases and analytes, between mobile phases and MS, and between mobile phases in different separation modes in multidimensional chromatography are analyzed. Novel developments in MDLC techniques, such as high-abundance protein depletion and chromatography arrays, are further discussed. In this review, the solutions proposed by researchers when encountering compatibility issues are emphasized. Moreover, the applications of HPLC-MS combined with various sample pretreatment methods in the study of exosomal and single-cell proteomics are examined. During exosome isolation, the combined use of ultracentrifugation and SEC can yield exosomes of higher purity. The use of SEC with ultra-large-pore-size packing materials (200 nm) enables the isolation of exosomal subgroups, and proteomics studies have revealed significant differences in protein composition and function between these subgroups. In the field of single-cell proteomics, researchers have addressed challenges related to reducing sample processing volumes, preventing sample loss, and avoiding contamination during sample preparation. Innovative methods and improvements, such as the utilization of capillaries for sample processing and microchips as platforms to minimize the contact area of the droplets, have been proposed. The integration of these techniques with HPLC-MS shows some progress. In summary, this article focuses on the recent advances in HPLC-MS technology for proteomics analysis and provides a comprehensive reference for future research in the field of proteomics.

Ag-bridged Z-scheme AgI/NU-1000 heterojunction for enhanced photocatalytic degradation of sulfamethazine: Pathways, mechanism insight, and DFT calculations

Constructing heterojunction structures is a crucial approach for improving carrier separation efficiency and expanding the visible-light absorption range of photocatalysts. Herein, a series of Ag-bridged Z-scheme (AgI/NU-1000) heterojunction photocatalysts were successfully prepared using a simple in-situ precipitation method. The photodegradation efficiency of the AgI/NU-1000 samples was assessed by degrading sulfamethazine under visible-light conditions. The optimized AgI/NU-10000.35 (AN-2) photocatalyst exhibited exceptional photodegradation activity, achieving a degradation rate of 89.68 % within 40 min. The toxicity test results proved that the solution degraded by AN-2 exhibited no detrimental effects on biological growth, thereby demonstrating its potential practical application value. Electrochemical and photochemical tests demonstrated that the creation of a Z-scheme heterojunction was favorable for the separation of photogenerated carriers. Density functional theory (DFT) calculations and X-ray photoelectron spectroscopy were used to clarify the electron migration direction within the AgI/NU-1000 heterojunction. Possible degradation pathways and mediate products were investigated using high-performance liquid chromatography–mass spectrometry and DFT calculations. This work demonstrates the potential application of AgI/NU-1000 composite materials in environmental treatment.

MrgX2-targeted ligand screening from Artemisia capillaris Thunb. extract and receptor-ligand interaction analysis based on MrgX2-HALO-tag/CMC

Artemisia capillaris Thunb. (A. capillaris) is a well-known traditional Chinese herbal medicine with a wide range of pharmacological effects, such as soothing the liver and gallbladder, heat clearance, and detoxifying. Hence, its extract is commonly added to various traditional Chinese medicine formulas. Traditional Chinese medicine injection (TCMI) is a mature pharmaceutical dosage form developed using TCM theory combined with modern science and technology. Notably, allergic reactions, especially pseudo‑allergic reactions (PARs), greatly limited the use of these injections. Therefore, screening pseudo‑allergic components in A. capillaris extract is clinically significant. In the present study, we proposed a two-dimensional screening and identification system based on mas-related G protein-coupled receptor X2-HALO-tag/cell membrane chromatography (MrgX2-HALO-tag/CMC) high performance liquid chromatography mass spectrometry (HPLC-MS); seven potential active components were screened from 75 % ethanol extract of A. capillaris: NCA, CA, CCA, 1,3-diCQA, ICA-B, ICA-A, and ICA-C. The receptor-ligand interactions between these seven compounds and MrgX2 protein were analyzed using frontal analysis and molecular docking technology. Furthermore, a mast cell degranulation-related assay was used to assess the pseudo‑allergic activity of these compounds. The screened compounds can serve as ligands of MrgX2, and this study provides a research basis for pseudo‑allergic reactions caused by TCMIs containing A. capillaris.

Comparative study for purity assessment of recombinant human growth hormone using mass balance approach and amino acid-based isotope dilution mass spectrometry

Growth hormone plays an unreplaceable role in regulating human growth and promoting protein synthesis, etc. For this reason, accurate quantification of its purity has become a focus of attention in the pharmaceutical industry and medical research. Herein, the purity assessment of human growth hormone (hGH) was established on the basis of mass balance approach and amino acid (AA)-based isotope dilution mass spectrometry (IDMS). In the first method, hGH purity was quantified by the determination of all the impurities in a hGH study material. In the second method, hGH purity was assessed by quantifying hydrolyzed AAs, e.g. leucine (Leu), phenylalanine (Phe), and proline (Pro). To accurately quantify these AAs, the corresponding isotope-labeled AAs were used as internal standards using high-performance liquid chromatography- mass spectrometry (HPLC-MS). The results of the two methods for determining the purity of hGH were consistent, with the mass balance method giving a result of (0.352 ± 0.010) g/g and IDMS a result of (0.363 ± 0.028) g/g. The work in this study could give a benefit for protein quantification and provides a reference for the development of hGH certified reference materials.

Changes in flavour- and taste-presenting substances in Hypsizygus marmoreus mushroom packaged in polypropylene nanofilm

Hypsizygus marmoreus mushroom is known for its pleasant flavour and delicious taste. However, its flavour quality deteriorates rapidly during postharvest storage. In the present work, we explored the dynamic changes in flavour-presenting substances and taste components in H. marmoreus packaged in polyethylene nanofilm (0.05 mm) during low temperature storage (12 days). We used headspace solid-phase microextraction combined with gas chromatography-mass spectrometry to analyse the volatile flavour components; high performance liquid chromatography and liquid chromatography-mass spectrometry to analyse the non-volatile flavour (taste-presenting) substances (e.g., soluble sugars, taste nucleotides, free amino acids, and organic acids); and the relative odour activity value (ROAV) and taste active value (TAV) methods to determine the flavour substances in fresh and stored H. marmoreus. Results revealed that n-octanal, 1-octene-3-one, and 1-octene-3-ol were the characteristic volatile flavour substances, and that trans-2-octenal was the key substance affecting the formation of volatile flavours in fresh H. marmoreus. Trans,trans-2,4-nonadienal, and 3-octanone were the key volatile substances that resulted in flavour deterioration. Among the non-volatile flavour substances, trehalose, 5'-GMP, glutamic acid, alanine, and arginine were the key taste-presenting substances. In addition, oxalic acid and lysine were the key taste substances after H. marmoreus deterioration. These results provided a theoretical basis for rapid quality detection, flavour identification, and shelf-life prediction of H. marmoreus.

Characterization of HphA: The First Enzyme in the Homologation Pathway of l-Phenylalanine and l-Tyrosine.

Homologation of amino acids is the insertion or deletion of a methylene group to their side chain, which is a relatively uncommon chemical transformation observed in peptide natural product (NP) structure. Homologated amino acids can potentially make the NP more stable in a biological system, but its biosynthesis is yet to be understood. This study biochemically characterized the first of three unexplored enzymes in the homologation pathway of l-phenylalanine and l-tyrosine. Previously proposed reactions catalyzed by HphA were confirmed by reversed-phase high-performance liquid chromatography and tandem mass spectrometry analysis. The substrate profile and kinetic parameters showed high selectivity for the natural substrates and their close analogs. The comparability of HphA to homologous enzymes in primary metabolic pathways, 2-isopropylmate synthase and homocitrate synthase which are involved in l-leucine and l-lysine biosynthesis, respectively, was validated by bioinformatical and site-directed mutagenesis studies. The knowledge obtained from this study has deepened the understanding of the homologation of amino acids, which can lead to future combinatorial biosynthesis and metabolic engineering studies.

Probiotics relieve perioperative postoperative cognitive dysfunction induced by cardiopulmonary bypass through the kynurenine metabolic pathway

Postoperative cognitive dysfunction (POCD) has become the popular critical post-operative consequences, especially cardiopulmonary bypass surgery, leading to an increased risk of mortality. However, no therapeutic effect about POCD. Probiotics are beneficial bacteria living in the gut and help to reduce the risk of POCD. However, the detailed mechanism is still not entirely known. Therefore, our research aims to uncover the effect and mechanism of probiotics in relieving POCD and to figure out the possible relationship between kynurenine metabolic pathway. 36 rats were grouped into three groups: sham operated group (S group, n = 12), Cardiopulmonary bypass group (CPB group, n = 12), and probiotics+CPB (P group, n = 12). After CPB model preparation, water maze test and Garcia score scale was performed to identify the neurological function. Immunofluorescence and Hematoxylin and eosin staining has been used for hippocampal neurons detection. Brain injury related proteins, oxidative stress factors, and inflammatory factors were detected using enzyme-linked immunosorbent assays (ELISA). Neuronal apoptosis was detected by TdT-mediated dUTP nick end-labeling (TUNEL) staining and western blot. High-performance liquid chromatography/mass spectrometry (HPLC/MS) was performed to detect the key factors of the kynurenine metabolic pathway. Our results demonstrated that probiotics improved neurological function of post-CPB rats. The administration of probiotics ameliorated memory and learning in spatial terms CPB rats (P < 0.05). Hematoxylin and eosin (H&E) staining data, S‐100β and neuron-specific enolase (NSE) data convinced that probiotics agonists reduced brain damage in CPB rats (P < 0.05). Moreover, probiotics regulated inflammatory factors, meanwhile attenuated hippocampal neuronal apoptosis. Probiotics alleviated POCD in rats with CPB through regulation of kynurenine metabolic signaling pathway.
