Bioactive Metabolites from the Fruiting Body and Mycelia of Newly-Isolated Oyster Mushroom and Their Effect on Smooth Muscle Contractile Activity.

Higher basidiomycetes are recognized as functional foods due to their bioactive compound content, which exerts various beneficial effects on human health, and which have been used as sources for the development of natural medicines and nutraceuticals for centuries. The aim of this study was to evaluate and compare the biological potential of basidiocarp and mycelial biomass produced by submerged cultivation of a new regionally isolated oyster mushroom. The strain was identified with a high percentage of confidence (99.30%) as Pleurotus ostreatus and was deposited in the GenBank under accession number MW 996755. The β-glucan content in the basidiocarp and the obtained mycelial biomass was 31.66% and 12.04%, respectively. Three mycelial biomass and basidiocarp extracts were prepared, and the highest total polyphenol content (5.68 ± 0.15 mg GAE/g DW and 3.20 ± 0.04 mg GAE/g DW) was found in the water extract for both the fruiting body and the mycelium biomass. The in vitro antioxidant activity of the extracts was investigated, and it was determined that the water extracts exhibited the most potent radical scavenging activity. The potential ability of this new fungal isolate to affect the contractile activity (CA) of dissected smooth muscle preparations (SMP) was examined for the first time. It was found that oyster mushrooms likely exhibit indirect contractile effects on the gastric smooth muscle (SM) cells.

Use of concentrated fruit juice extracts to improve the antioxidant properties in a soft drink formulation

Three juices obtained respectively from pomegranate, prickly pear and orange fruit were separately concentrated using an innovative technique, that allow to obtain a final product with high content of bioactive compounds and elevate antioxidant activity. The concentrated juices were employed in the formulation of a soft drink to improve its bioactive properties. Despite a low amount of each concentrated juice was used, the experimental soft drink obtained evidenced a high content of total polyphenols. The antioxidant activity detected was similar to that of tea commonly considered a healthy beverage with strong radical scavenging activity. The sensory analysis of the soft drink showed a consumers' liking score potentially promising for market acceptance. In addition, the experimental drink produced was evaluated for its stability by simulating domestic storage and from the results obtained is possible to assess that a good antioxidant activity was maintained even after 5 days of conservation. Therefore, the present work, stimulates the use of high-quality concentrated juices as food ingredient in order to improve the functional properties of soft drinks.

The effects of extraction conditions on the antioxidant activities, total polyphenol and monomer anthocyanin contents of six edible fruits growing wild in Hungary

Although wild fruits are significantly underutilized in most countries, they could be good sources of valuable bioactive compounds with antioxidant properties. Therefore the present study focused on the study of a conventional extraction technique (maceration with shaking; MACS) to extract natural antioxidants and anthocyanin colorants from six edible wild-growing fruits (European crab apple, bilberry, yellow-, red-, and purple-skinned greengage, and quince). One-factor-at-a-time (OFAT) methodology was chosen to investigate the influences of three different parameters (solvent type, extraction time and solvent acidity) on the total polyphenol contents (TPCs), total monomeric anthocyanin (TMA) contents, and antioxidant capacities, specifically ferric reducing power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl radical-scavenging activity (DPPH). After optimization, the recorded TPCs and antioxidant activities proved to be significantly higher for all analyzed fruits when compared to differing extraction conditions. For European crab apple and purple-skinned greengage, the best extraction conditions were a ratio of 80:20 (v/v) EtOH–H2O, 1% (v/v) of HCOOH, and an extraction time of 90 min. In the case of red-skinned greengage, the extraction parameters were the same as the above except for the acid concentration (0.5%; v/v) used. For quince, the optimized conditions required a 50:50 (v/v) EtOH–H2O mixture, an extraction time of 90 min, and 0.5% (v/v) HCOOH concentration. The best conditions for the extraction of bilberry and yellow-skinned greengage were an EtOH–H2O combination of 50:50 (v/v), extraction time of 60 min, and HCOOH concentration of 0.5% (v/v). The highest TPC and antioxidant activity were observed in quince (281–510 mg GAE/100g and 109–395 mg AAE/100g) whereas the lowest were measured in European crab apple (55.9–70.0 mg GAE/100g and 20.1–43.2 mg AAE/100g). Bilberry exhibited the highest TMA content (346 mg CGE/100g). Overall, our results showed that these wild fruits could be a good source of natural antioxidants for the local residents.

The effects of roasting on antioxidant and anticancer activities of fermented Hass avocado seeds

Avocado by-products present a waste issue for cold-pressed avocado oil processors in New Zealand. The avocado seed contains many extractable compounds that are beneficial to health. This work aims to evaluate the effects of roasting fermented avocado seed to produce a food ingredient with beneficial antioxidant and anticancer properties. Avocado seeds were subjected to natural fermentation and inoculated fermentation with either Lactobacillus plantarum or kefir. The fermented samples were evaluated in terms of total plate count values of lactic acid bacteria acetic acid bacteria and yeasts. The number of microorganisms increased significantly (p < 0.001) over the 7 days of fermentation for all samples. Fermentation with L. plantarum resulted in significantly (p < 0.05) higher total phenolic content (TPC) and antioxidant activities compared to kefir and naturally fermented samples. The fermented avocado seeds were further subjected to roasting to yield a shelf-stable dried powder. Roasted samples fermented by L. plantarum had significantly (p < 0.001) higher total polyphenolic content and antioxidant capacity (CUPRAC and Ferric Reducing Antioxidant Power assays) compared to kefir and naturally fermented samples. The avocado seed powder showed the best inhibition effect on Hep G2 followed by the MDA-MB-231 and MCF-7 cancer cell lines. The roasting conditions for optimal antioxidant and anticancer activities were determined to be at 127°C for 24.7 min. This study demonstrated that fermentation of avocado seeds in combination with roasting yielded a powder with good antioxidant and anticancer activities, which can potentially be incorporated into food for added health benefits.

Deep eutectic solvent-based ultrasound-assisted extraction of polyphenols from Cosmos sulphureus

In this study, deep eutectic solvents-based ultrasound-assisted extraction (DES-UAE) was done for extraction of polyphenols from Cosmos sulphureus. Among seven kinds of DESs prepared, choline chloride-lactic acid (1:2) was considered as a good candidate for polyphenols extraction due to superior extraction performance. Subsequently, process parameters for DES-UAE were optimized by response surface methodology. The highest total polyphenolic content (TPC) was achieved at 47.5 ℃ temperature, 32.6% water content and 4.0 W/mL ultrasonic energy density. This was 21% higher than that acquired with conventional ethanol-based UAE. Furthermore, Pearson’s correlation coefficient revealed high correlation between TPC and antioxidant activity of extracts (r = 0.85). Moreover, HPLC analysis indicated that the substitution of DES for ethanol in UAE can effectively improve the individual concentrations in extracts of interested polyphenols (chlorogenic acid, caftaric acid, rutin and kaempferol) in the range of from 10.7% to 43.5%. In conclusion, this study implies that DES-UAE is a viable, sustainable, efficient and integrated technique to obtain polyphenols from Cosmos sulphureus. DES-UAE is instrumental for exploring the feasibility of commercial scale extraction of polyphenols.

Potato Slices Drying: Pretreatment Affects the Three-Dimensional Appearance and Quality Attributes

In the current study, the effects of steam blanching, saline immersion, and ultrasound pretreatment on the drying time, three-dimensional (3D) appearance, quality characteristics, and microstructure of potato slices were investigated. All the pretreatment methods enhanced the drying kinetics relative to the untreated potato slices. The 3D appearance was evaluated by reconstructed 3D images, shrinkage, and curling degree. The reconstructed images could well reproduce the appearance changes in the potato slices during drying. All the three pretreatment methods reduced the shrinkage during the drying process relative to the untreated potatoes. The curling degree was evaluated by the height standard deviation (HSD) of the material surface. The results showed that saline immersion inhibited the curling of the potato slices during the drying process, while ultrasound aggravated the curling of the potato slices. The potatoes treated by blanching obtained a lower total color difference (ΔE), higher total polyphenol content, and antioxidant capacity compared with the samples treated with saline immersion and ultrasound pretreatments. The observation of the microstructure by scanning electron microscope (SEM) verified the effects of the pretreatments on the drying time and appearance deformation. Therefore, it is of great significance to regulate the 3D appearance and quality characteristics of agricultural products during the drying process by an appropriate pretreatment.

Effect of drying methods on yield, physicochemical properties, and total polyphenol content of chamomile extract powder.

Chamomile (Matricaria chamomilla L.) is a traditional medicinal plant used to treat hay fever, inflammation, muscle spasms, menstrual disorders, insomnia ulcers, wounds, gastrointestinal disorders, rheumatic pain, and hemorrhoids. Dried chamomile flowers have a longer shelf life and the dried extract in form of powder offers much flexibility for new therapeutic formulations as it could be used as a replacement for liquid extract and serve as a shelf-stable ingredient in new applications. This study aims to determine the effect of drying methods, i.e., convection oven-drying at 45 °C, freeze-drying at -50°C, and spray-drying at 140°C at 10.5 and 12ml/min, respectively) on powder yield, physicochemical properties (moisture content, water activity, and color attributes), and total polyphenol content of chamomile extract powder. Our findings showed that spray-drying conducted at 140°C, 12ml/min resulted in the lowest yield of powder (16.67%) compared to convection oven-drying (90.17%) and freeze-drying (83.24%). Decreasing the feed flow rate to 10.5ml/min during spraying caused an increase in powder yield to 26.99%. The moisture content of spray-dried chamomile extract powder obtained at 140°C, 10.5ml/min was higher (11.00%) compared to that of convection oven-dried (8.50%) and freeze-dried (7.50%). Both convection oven-dried and freeze-dried chamomile extract powder displayed no significant difference (p > 0.05) in moisture content. The higher feed flow rate (12ml/min) in spray-drying also led to an increase in the moisture content of chamomile extract powder to 12.00%. The higher residual moisture found in the spray-dried samples resulted in partial agglomeration of particles. In terms of water activity, freeze-dried chamomile extract powder was found to have the highest water activity (0.63) compared to that of convection oven-dried (0.52), spray-dried at 140°C, 10.5ml/min (0.57), and spray-dried at 140°C, 12ml/min (0.58). Spray-dried and freeze-dried chamomile extract powder with high moisture content and water activity could be highly susceptible to microbial growth. In terms of color attributes, higher drying temperature in spray-drying led to darker, redder, and more yellowish chamomile extract powder that could be caused by heat-induced Maillard reaction and caramelization. Since lower drying temperature was used in both convection oven-drying and freeze-drying, both convection oven-dried (56.94mg GAE/g powder) and freeze-dried chamomile extract powder (55.98mg GAE/g powder) were found to have higher total polyphenol content compared to those of spray-dried (42.79-46.79mg GAE/g powder). The present findings allow us to understand the effect of drying methods on the properties of chamomile extract powder and provide a better drying option to dry chamomile extract. Due to higher powder yield with ideal powder properties such as low moisture content and water activity, desirable color, and high total polyphenol content obtained from convection oven-drying, convection oven-drying was a better option than freeze-drying and spray-drying for drying chamomile extract.

Optimization of bioactives extraction from grape marc via a medium scale ambient temperature system and stability study.

A scalable procedure with minimum energy requirements, MSAT (Medium Scale Ambient Temperature), in combination with solvents generally recognized as safe (GRAS), has been optimized to obtain polyphenolic extracts from white grape (Vitis vinifera) marc. The solvents considered were propylene glycol (Pg), ethanol (Et), and ethyl lactate (Lc), as well as their respective hydro-organic mixtures. In a first approach, the operating parameters were optimized through a response surface matrix: extraction solvent volume (range 10-150 mL), marc mass (range 20-200 g) and marc/dispersant mass ratio (range 0.5-2 g⋅g-1), using the total polyphenol content (TPC) and the antioxidant activity (AA) of the extracts as response parameters. The highest TPC (5,918 mgGAE⋅L-1) and AA (44 mmolTE⋅L-1) values were obtained using 200 g marc and 100 mL solvent. Regarding the type of solvent, a better response was reached with Lc > Et > Pg > H2O obtaining a polyphenol concentration of 252 mg⋅L-1 for the hydro-organic isovolumetric ratio of ethyl lactate. In addition, the stability of the extracts was studied for 62 days. The effect of factors such as temperature, light exposure, and oxidative reactivity was evaluated. The bioactivity indices showed no changes with the storage conditions of the extracts in the first month of analysis, after which 75% of the antioxidant activity as the concentration of the polyphenolic profile (204 mg⋅L-1) remains. The absence of reactive oxygen and the cooling of the extract (4°C) were the most determining factors (p < 0.05) in modulating the stability of the total polyphenolic profile.

Effect of different tea manufacturing methods on the antioxidant activity, functional components, and aroma compounds of Ocimumgratissimum

Ocimum gratissimum is a herbaceous medicinal plant known for its high antioxidant activity. In the present study, we investigated the effect of different tea manufacturing processes in retaining the biochemical properties of O. gratissimum and determined which of the processes yields a high-quality tea. We determined the color tone, total polyphenol content (TPC), odor, 2-diphenyl-1-picrylhydrazyl (DPPH) values, and hydrophilic oxygen radical absorbance capacity (H-ORAC) of treated samples. Steaming time affected the color tone, TPC, DPPH value, chicoric acid content, and odor, while the drying temperature and fermentation treatment affected all the parameters. Furthermore, caryophyllene, germacrene-D, and eugenol were identified as the main aroma compounds in O. gratissimum. Our findings indicate that the drying temperature and fermentation treatment are the major factors affecting the biochemical properties and acceptance quality of O. gratissimum tea. Among the different treatments used, machine-drying without steaming treatment was the best on odor. Freeze-drying, which is more costly, was the best on the TPC and antioxidant activities; however, it was found that MD, which is relatively cheap compared to freeze-drying treatment, maintained high TPC, DPPH, H-ORAC values, and chicoric acid content by steaming for 2 min before drying.

Bread as a Valuable Raw Material in Craft Ale Beer Brewing.

One of the ingredients used for brewing is barley, which should be malted; it is considered the most polluting agricultural input. On the other hand, food wastage is today a widespread problem that causes significant environmental damage and also generates large economic losses worldwide. One of the most wasted food products is bread; it is estimated that hundreds of tons of bread are wasted every day worldwide. In this study, the brewing of ale beers with bread was carried out. For this purpose, up to 50% of the malt weight was replaced by different types of bread: wheat bread, whole wheat bread, rye bread, and corn bread. A physicochemical and sensory comparison was made with 100% malt ale beer. All beers brewed with bread had an alcoholic strength similar to that of the control beer, except the corn beer. Beers brewed with whole grain bread showed a higher antioxidant capacity and a higher total polyphenol content. The sensory analysis presented different profiles depending on the type of bread; in general, the addition of bread created a greater olfactory intensity in nose. Thus, it was found that it is possible to brew beer with bread substituting up to 50% of the malt. In addition, it was also shown that the beer brewed with whole wheat bread had similar characteristics to the control beer, even improving some beneficial health properties, representing a great advantage for the brewing industry all over the world.

Variation in Physical-Chemical Parameters and Phenolic Compounds in Fruits of Four Calafate Clones

Calafate (Berberis microphylla G. Forst) is an evergreen shrub with blue berries that grows naturally in Patagonia, in South America. It has beneficial nutraceutical characteristics for human health. The objective of the research was to evaluate the effect of different harvest dates of calafate clones in the south-central zone of Chile on the polyphenolic content, antioxidant capacity, quality parameters and fruit yield. To meet this objective, during three consecutive years, four wild calafate clones located in the town of San Ignacio, Chile, were selected. Where a harvest period was established from 110 to 140 days after full bloom (DAFBs), each of the harvests carried out were used for the following measurements: antioxidant capacity, determination of anthocyanin content, concentration of polyphenols, phenolic compounds, soluble solids, total titratable acidity, pH, fruit yield and quality. Among the main results, it can be highlighted that clone 2 was the one that obtained the highest concentration of soluble solids, with 38.0 °Brix at 140 DAFBs. Together, it was the one that obtained the highest content of total polyphenols and concentration of anthocyanins, with 1121 g GAE kg−1 fw and 714 g cy-3-glu 100 g−1 fw, respectively.

Phenolic Profile, Antioxidant and Enzyme Inhibitory Activities of Leaves from Two Cassia and Two Senna Species.

Several species within the genera Cassia or Senna have a treasure of traditional medicines worldwide and can be a promising source of bioactive molecules. The objective of the present study was to evaluate the phenolic content and antioxidant and enzyme inhibition activities of leaf methanolic extracts of C. fistula L., C. grandis L., S. alexandrina Mill., and S. italica Mill. The two Cassia spp. contained higher total polyphenolic content (42.23–49.75 mg GAE/g) than the two Senna spp., and C. fistula had significantly (p ˂ 0.05) the highest concentration. On the other hand, the Senna spp. showed higher total flavonoid content (41.47–59.24 mg rutin equivalent per g of extract) than that found in the two Cassia spp., and S. alexandrina significantly (p ˂ 0.05) accumulated the highest amount. HPLC–MS/MS analysis of 38 selected bioactive compounds showed that the majority of compounds were identified in the four species, but with sharp variations in their concentrations. C. fistula was dominated by epicatechin (8928.75 µg/g), C. grandis by kaempferol-3-glucoside (47,360.04 µg/g), while rutin was the major compound in S. italica (17,285.02 µg/g) and S. alexandrina (6381.85). The methanolic extracts of the two Cassia species exerted significantly (p ˂ 0.05) higher antiradical activity, metal reducing capacity, and total antioxidant activity than that recorded from the two Senna species’ methanolic extracts, and C. fistula displayed significantly (p ˂ 0.05) the highest values. C. grandis significantly (p ˂ 0.05) exhibited the highest metal chelating power. The results of the enzyme inhibition activity showed that the four species possessed anti-AChE activity, and the highest value, but not significantly (p ≥ 0.05) different from those obtained by the two Cassia spp., was exerted by S. alexandrina. The Cassia spp. exhibited significantly (p ˂ 0.05) higher anti-BChE and anti-Tyr properties than the Senna spp., and C. grandise revealed significantly (p ˂ 0.05) the highest values. C. grandise revealed significantly (p ˂ 0.05) the highest α- amylase inhibition, while the four species had more or less the same effect against the α-glucosidase enzyme. Multivariate analysis and in silico studies showed that many of the identified phenols may play key roles as antioxidant and enzyme inhibitory properties. Thus, these Cassia and Senna species could be a promising source of natural bioactive agents with beneficial effects for human health.

The effect of malting on phenolic compounds and radical scavenging activity in grains and breakfast cereals.

Breakfast cereals are popular grain foods and sources of polyphenols. Malting alters polyphenol content and activity; however, effects are varied. The total polyphenol content (TPC), radical scavenging activity (RSA), and polyphenol profile were analyzed in unmalted and malted grains (wheat, barley, and sorghum) and breakfast cereals (wheat, barley) by Folin Ciocalteu Reagent (FCR), % inhibition of the free radical 2,2-diphenyl-1-picryl-hydrazyl, and high performance liquid chromatography. Higher TPC was observed in all malted grains and breakfast cereals compared with unmalted samples (p<0.05). Higher RSA was also observed in all malted samples compared to unmalted samples (p<0.05) except for wheat grain to malted wheat grain. In this study, malting induced additional polyphenols and antioxidant activity in grains and cereal products. Malted grain breakfast cereals may be practical sources of polyphenol antioxidants. PRACTICAL APPLICATION: This study utilized malting in a unique way to investigate potential health benefits of polyphenols and antioxidant activity in grains (wheat, barley, and sorghum) and ready-to-eat breakfast cereals (wheat and barley). This study found that grains and breakfast cereals are important sources of antioxidant polyphenols, and these were significantly increased in malted varieties. Understanding this is important as grains and breakfast cereals are widely consumed staple foods. Consuming healthier grain products may be a practical strategy in reducing the risk of noncommunicable diseases such as colorectal cancer and type-2 diabetes, where wholegrain consumption may be important in prevention.

Ethnopharmacological Survey, Mineral and Chemical Content, In Vitro Antioxidant, and Antibacterial Activities of Aqueous and Organic Extracts of Chamaerops humilis L. var. argentea Andre Leaves.

Introduction The present study is carried out for the first time on Chamaerops humilis L. var. argentea Andre from the region of Taza using an ethnopharmacological survey, an experimental study of the mineralogical and chemical compositions, and evaluations of the antioxidant and antibacterial activities. Methods After conducting the ethnopharmacological survey, a mineralogical and phytochemical study involving the preparation of aqueous and organic extracts was done. Essential oils were also extracted by hydrodistillation. Subsequently, qualitative and quantitative chemical analyses were performed. In vitro evaluation of antioxidant activities was performed by five tests (H2O2, DPPH, ABTS, FRAP, and RP) and antibacterial activities by the disc method and determination of MIC and MBC. A principal component analysis (PCA) was performed to visualize the different correlations. Results The different parts of the plant are used for the treatment of digestive disorders, cardiovascular diseases, and diabetes. In addition, the leaves are rich in mineral compounds, catechic tannins, flavonoids, and sterols. However, they have some traces of essential oils. The quantitative analysis revealed that the ethanolic macerated had a higher content of total polyphenols (100.27 ± 1.95 mg EAG/g E) and catechic tannins (52.11 ± 1.02 mg EC/g E). This extract had a strong antioxidant capacity (H2O2 (37.34 ± 0.55%), DPPH (IC50 = 31.18 ± 0.66 μg/ml), ABTS (108.28 ± 1.29 mg E AA/g E), FRAP (148.85 ± 0.43 mg E T/g E), and RP (10.86 ± 0.01 mg E AA/g E). The same extract had a bactericidal effect against Staphylococcus aureus. Principal component analysis (PCA) showed that antioxidant activity was highly correlated with the chemical composition of C. humilis leaves; a high correlation was recorded between the total polyphenol content and ABTS (r = 0.9779), FRAP (r = 0.9644), DPPH (r = 0.9418), and PR (r = 0.9271) tests. In addition, cathectic tannins were highly correlated with the tests of DPPH (r = 0.9753) and ABTS (r = 0.8843). Flavonoids were similarly correlated with DPPH (r = 0.8897) and ABTS (r = 0.7599) tests. Conclusion These results could justify the traditional use of the leaves of Chamaerops humilis in the region of Taza for the treatment of some diseases.

We investigated the antioxidant and anti-inflammatory activities in the leaf, stem, and flower of Centella asiatica (CA). CA is widely used in the medical and cosmetic industries. The leaf contained the highest total polyphenol content (236.88 mg%) and antioxidant activity when compared to the stem and flower. The total triterpenoid content was 60.65 mg/g, 54.20 mg/g, and 12.50 mg/g (leaf, flower, and stem, respectively), indicating the triterpenoids in the stem were approximately 4 to 5 times lower than the CA leaf or flower. The cell viability evaluation using RAW 264.7 and HaCaT cells revealed no significant levels of cytotoxicity (≤ 100 μg/mL). The anti-inflammatory activity assessed using RAW 264.7 cells revealed the flower had higher activity than the leaf and stem, with NO (88.3 % – inhibitory rate), TNF-α (64.8 %), IL-6 (65.7 %), and MCP-1 (28.9 %) inhibitory activity. An analysis of the inflammatory inhibition activity on the skin using HaCaT cells revealed the flower possessed a potent activity level of IL-6 (72.0 %) and IL-8 (89.4 %) inhibition. Finally, the flower and leaf had potent physiological effects and functional components. We expect that the leaf and flowers can be used as functional materials, such as antioxidant and anti-inflammatory activities.

Agronomic evaluation of different lettuce (Lactuca sativa L.) varieties under unheated plastic tunnel

Among the leafy vegetables, lettuce (Lactuca sativa L.) is the most widely grown species in the world. Wide ranges of physical properties of the products are used to assess the degree of maturity. In this study, different lettuce varieties were grown under an unheated plastic tunnel in springtime. We evaluated the agronomic properties of different lettuce varieties, evaluated the condition of the plants, and determined the bioactive substances. Larger head weight can be achieved by the ‘King of May’ butterhead (259.31 g) and ‘Great Lakes 659’crisphead (220.40 g) genotypes. A very strong correlation (r = 0.995) was observed between the lettuce head index, and leaf index and both had a strong positive correlation (r = 0.828 and 0.760) with NDVI. The highest SPAD values were measured for cos lettuce ‘Romaine lettuce’ (44.01) and iceberg lettuce ‘Great Lakes 659’ (42.71). However, these genotypes showed the highest (9.52%; 8.74%) dry matter content, too. The red leaf variety 'Lollo Rosso' had the highest total polyphenol content (181.53 GAE/100 g FW). Among the evaluated properties, iceberg lettuce showed favorable morphology, plant condition, and good dry matter content. In addition, between the loose-leaf lettuces, the red leaf lollo type was outstanding with bioactive content.

Maqui (Aristotelia chilensis (Mol.) Stuntz): A Natural Antioxidant to Improve Quality of Meat Patties.

Aristotelia chilensis is an endemic shrub of the South Pacific with high concentrations of bioactive compounds in its leaves and, therefore, it is highly valued. The effect of Aristotelia chilensis leaf powders (maqui leaf powders; Ma) on the quality and shelf life of beef patties during 7 days of storage was investigated. Five beef patties treatments were prepared: (1) Control without antioxidants (CT); (2) Beef patties with synthetic antioxidants plus color (250 mg/kg) (PL); (3) Beef patties with 500 ppm of maqui leaf powders (Ma500); (4) Beef patties with 1000 ppm of maqui leaf powders (Ma1000); and (5) Beef patties with 2000 ppm of maqui leaf powders (Ma2000). The quality of the beef patties was evaluated on day 0 and day 7 of storage by physicochemical analysis (moisture, ash and lipid content, color, pH, fatty acid profile and lipid oxidation) and organoleptic analysis. The addition of maqui leaf powders did not produce changes in the proximate composition of the beef patties. The pH for all treatments showed a range of 5.50–5.75 and significant differences (p < 0.05) were observed at the beginning and end of storage. The pH of the control beef patties increased during storage while the pH of the beef patties with synthetic and natural antioxidants decreased. Redness (a*) was the color indicator that was mostly affected by the inclusion of 1000 ppm and 2000 ppm powders. High lipid oxidation was observed in control samples on the seventh day of storage due to the high percentage of fat used in the formulation and the absence of any antioxidant. However, the Ma500, Ma1000, and Ma2000 treatments presented the lowest lipid oxidation rates (42.05%, 40.29%, and 43.14%, respectively) in comparison with the synthetic antioxidant (52.23%). This lipid inhibition is related to the strong antioxidant activity (29.75 µg/mL IC50 DPPH) of the maqui leaf powder due to its high content of total polyphenols (148.76 mg GAE/g), mainly characterized by having great amounts of hydroxybenzoic acids (82.5 mg GAE/g), flavonoids (7.1 mg QE/g), and hydroxycinnamic acids (3.7 mg CAE/g). Although minimal variations were observed in some individual fatty acids, and despite the trend to decrease MUFA and increase SFA with the maqui leaf powder addition, these differences were minimal and, according to the nutritional indices results, without any influence on the nutritional quality of the beef patties. The organoleptic analysis showed that the addition of maqui leaf powders did not affect the general acceptability of the new formulations. This study reports for the first time the substitution of synthetic antioxidants with Aristotelia chilensis leaves extract. Based on the results, it can be concluded that this ingredient can be used as an alternative for the production of raw meat products with clean labels.

Bioactivity of Biomass and Crude Exopolysaccharides Obtained by Controlled Submerged Cultivation of Medicinal Mushroom Trametes versicolor.

The aim of this study is to characterize the bioactivity of mycelial biomass and crude exopolysaccharides (EPS) produced by Trametes versicolor NBIMCC 8939 and to reveal its nutraceutical potential. The EPS (1.58 g/L) were isolated from a culture broth. The macrofungal biomass was rich in protein, insoluble dietary fibers and glucans. The amino acid composition of the biomass was analyzed and 18 amino acids were detected. Three mycelial biomass extracts were prepared and the highest total polyphenol content (16.11 ± 0.14 mg GAE/g DW) and the total flavonoid content (5.15 ± 0.03 mg QE/g DW) were found in the water extract. The results indicated that the obtained EPS were heteropolysaccharides with glucose as the main building monosaccharide and minor amounts of mannose, xylose, galactose, fucose and glucuronic acid. Fourier Transform Infrared Spectroscopy (FTIR) confirmed the complex structure of the crude EPS. Five probiotic lactic acid bacteria strains were used for the determination of the prebiotic effect of the crude EPS. The anti-inflammatory potential was tested in vitro using cell line HT-29. The significant decrease of IL-1 and IL-8 and increase of TGF-beta expression revealed anti-inflammatory potential of the crude exopolysaccharides from T. versicolor.

Nutritional Value and Antioxidant Activity of Fresh Pumpkin Flowers (Cucurbita sp.) Grown in Poland

Pumpkin flowers, in their composition, contain many bioactive ingredients that have a beneficial effect on the human body. The aim of the research was to evaluate the antioxidant activity and chemical composition of flowers of various species and varieties of pumpkins: Amazonka, Ambar, Atlantic Giant, Bambino (Cucurbita maxima L.), Butternut, Muscade de Provence, Rouge vif d’Etampes (Cucurbita moschata Duch.), and Miranda (Cucurbia pepo L.). The flowers came from flowering pumpkin shoots, grown in Poland (Krakow). The total polyphenols, carotenoids, total sugar contents, antioxidant activity, and fatty acid composition were determined. The content of dry matter, protein, ash, fat, and selected minerals were also determined. Pumpkin flowers of the Atlantic Giant variety were characterized by the highest content of total polyphenols and sugars and antioxidant activity. They also showed the highest percentage of n-myristic acid (C14:0) and docosanoic acid (C22:0). The energy value of fresh pumpkin flowers, of all varieties, was low and averaged 22 kcal/100 g. Fresh pumpkin flowers are a significant source of iron, covering 60–80% of the EAR standard for adults (Atlantic Giant and Bambino varieties).

Biological Activities of Grape Seed By-Products and Their Potential Use as Natural Sources of Food Additives in the Production of Balady Bread.

The biological function of bioactive compounds found in plant by-products has triggered expanded interest in recent years. This study aims to produce balady bread enriched with dietary fiber, mineral, and phenolic compounds by the addition of grape seeds powder (GSP) at different levels (5%, 10%, and 15% as a partial substitute for wheat flour). The results show that balady bread (Bb) and grape seed powder have ash contents of about 1.97% and 3.04%, lipid contents of 3.22% and 17.15%, protein contents of 11.16% and 12.10%, fiber contents of 1.06% and 44.90%, and carbohydrates contents of 56.52% and 29%, respectively. Moreover, grape seed powder contains a higher level of iron and zinc about 30.02 and 9.43 mg/kg than the Bb control sample which contains about 8.19 and 7.25 mg/kg respectively. The findings revealed that balady bread fortified with grape seed powder contains a high amount of total polyphenols content (TPC), total flavonoid content (TF), and antioxidant capacity. The farinograph test results showed that increasing the GSP concentration in the flour above 10% reduced dough development, stability, and farinograph quality number. The addition of GSP to wheat flour accelerated the dough’s water absorption and mixing tolerance. Grape seed incorporation levels up to 10% (w/w) had no negative effect on dough rheological performance. The sensory evaluation of bread showed that samples that were enriched with grape seeds powder at up to 10% had good quality. Based on these findings, it is recommended to replace up to 10% GSP in the manufacturing of fortified balady bread with satisfactory physical and sensory characteristics and high TPC and antioxidant activity.
