Effect of Lentilactobacillus buchneri on Chemical and Microbial Compositions of Herba Leonuri (Leonurus japonicus Houtt.)-Contained Alfalfa Silage

Lactic acid bacteria (LAB) inoculants are commonly used in silage production, yet their effects on silage containing antimicrobial components, such as those found in Leonurus japonicus, remain less explored. Herein, the harvested alfalfa were thoroughly mixed with dried Leonurus japonicus Houtt. (LJH) at a ratio of 9:1 on a fresh weight basis and treated without (CK) or with a lactic acid bacterial inoculant (L; Lentilactobacillus buchneri). The mixtures were stored under anaerobic conditions in vacuum-sealed polyethylene bags for 30 days at ambient temperature. The L-treated silage exhibited high levels of water-soluble carbohydrates (4.98% dry matter (DM)) and acid detergent fiber (27.88% DM). Compared to that of treatment CK, treatment with L increased the acetic acid content of the silage, as a result of increased (p < 0.05) bacterial dominance and decreased (p < 0.05) bacterial richness indices (e.g., Pielou’s E, Shannon, and Simpson) in the pre-storage period. However, these changes gradually reduced as the storage length increased. Treatment L reshaped the bacterial community structure of silage, by increasing the prevalence of Lactobacillus and reducing relative abundances of Enterococcus and Weissella. However, the principal coordinate and Bray–Curtis index analyses illustrated that samples from the L-treated silages exhibited similarities to the CK samples post-fermentation. Overall, the effect of LJH on LAB was only observed in the later stages of fermentation, which did not sufficiently change the silage quality. Hence, using LJH in silage is vital for clean livestock production without compromising the function of LAB when mixed with alfalfa silage.

The Investigation of the Fermentative, Chemical and Microbial Effects of Grape and Tangerine Pomace Added to High Moisture Alfalfa Silage

This study investigated the impact of grape pulp (GRP) and tangerine pulp (TNP) supplementation (10% each) on alfalfa silage quality. The control silage received no additives. After a two-month fermentation period, silage samples were analyzed for chemical composition, fermentation quality, microflora, and organic acids. According to our results, both silage additives effectively lowered pH levels and isobutyric acid, while increased Fleig score and lactic acid levels compared to control group. However, GRP supplementation significantly increased the levels of crude protein (CP), yeast-mold colony count, and butyric acid levels. Conversely, TNP supplementation resulted in higher levels of water-soluble carbohydrates (WSC), valeric acid and lower acetic acid levels in the alfalfa silage samples. In conclusion, both GRP and TNP supplements have distinct effects on the chemical composition, silage quality, microflora, and organic acid profiles of alfalfa silage. These findings provide valuable insights into optimizing alfalfa silage production and its utilization in animal nutrition. Further research could explore optimal inclusion rates and potential synergistic effects with other additives to enhance silage quality.

Effect of co-inoculation of Lactobacillus plantarum and Lentilactobacillus buchneri on aerobic stability and microbial community composition of perennial sorghum silage

In the present study, the aerobic stability of perennial sorghum (Sorghum bicolor × Sorghum propinquum) silage following exposure to air was evaluated, which was pretreated with either Lactiplantibacillus plantarum (L group) or Lactiplantibacillus plantarum + Lentilactobacillus buchneri (M group). Samples from the M group had lower dry matter loss rates and higher acetic acid content after exposure to air. Furthermore, samples from the M group showed high levels of water-soluble carbohydrates (2.79–6.55% in dry matter) and crude protein (3.82–9.02% in dry matter) after exposure to air. Moreover, M pretreatment led to the inhibition of the growth of Debaryomyces hansenii whereas it promoted the growth of Levilactobacillus and Clavispora sp. Overall, M pretreatment of perennial sorghum during storage hindered aerobic deterioration and promoted the formation of total fermentation products that act as precursors for biogas production.

Effects of bacterial inoculants and an enzyme on the fermentation quality and aerobic stability of ensiled whole-crop sweet sorghum

A study was conducted to evaluate the effects of bacterial inoculation and cellulase on the fermentation quality of ensiled whole-crop sweet sorghum (WCSS, Sorghum bicolor L. Moench). The WCSS (323 g dry matter (DM)/kg, 251 g water soluble carbohydrates (WSC)/kg DM, 43 g crude protein (CP)/kg DM and 439 g neutral detergent fibre (NDF)/kg DM) was ensiled with i) no additive (control); ii) Lactobacillus buchneri (LB); iii) Lactobacillus plantarum (LP); and iv) LB+E, a combination of LB and enzyme. These treatments were ensiled in 1 L anaerobic jars for 25 days. The jars were opened on days 3, 7 and 15 to determine pH, while those of day 25 were sampled to determine nutrient composition, fermentation characteristics and aerobic stability. Inoculation reduced pH, butyric acid and ammonia-N and increased lactic acid content in sweet sorghum silage compared with the control. The aerobic stability of WCSS was improved with LB, while it was reduced with the homofermentative LP treatment compared with the control. The LB+E reduced the fibre, but increased residual WSC of silage. The aerobic stability of LB+E silage was lower than LB treated silage. Using enzymes to increase the WSC content of crops that already have high levels of WSC may result in reduced aerobic stability of silage. Further work is needed to evaluate these effects on silage produced on farm scale and on animal production performance.

Linked gene networks involved in nitrogen and carbon metabolism and levels of water-soluble carbohydrate accumulation in wheat stems

High levels of water-soluble carbohydrates (WSC) provide an important source of stored assimilate for grain filling in wheat. To better understand the interaction between carbohydrate metabolism and other metabolic processes associated with the WSC trait, a genome-wide expression analysis was performed using eight field-grown lines from the high and low phenotypic tails of a wheat population segregating for WSC and the Affymetrix wheat genome array. The 259 differentially expressed probe sets could be assigned to 26 functional category bins, as defined using MapMan software. There were major differences in the categories to which the differentially expressed probe sets were assigned; for example, probe sets upregulated in high relative to low WSC lines were assigned to category bins such as amino acid metabolism, protein degradation and transport and to be involved in starch synthesis-related processes (carbohydrate metabolism bin), whereas downregulated probe sets were assigned to cell wall-related bins, amino acid synthesis and stress and were involved in sucrose breakdown. Using the set of differentially expressed genes as input, chemical-protein network analyses demonstrated a linkage between starch and N metabolism via pyridoxal phosphate. Twelve C and N metabolism-related genes were selected for analysis of their expression response to varying N and water treatments in the field in the four high and four low WSC progeny lines; the two nitrogen/amino acid metabolism genes demonstrated a consistent negative association between their level of expression and level of WSC. Our results suggest that the assimilation of nitrogen into amino acids is an important factor that influences the levels of WSC in the stems of field-grown wheat.

Improvement of Fermentation Quality of Rice Straw Silage by Application of a Bacterial Inoculant and Glucose

The improvement of the fermentation quality of rice straw silage by application of lactic acid bacteria (LAB) and glucose was investigated in this study. Sixteen rice varieties were harvested at the maturity stage and the rice straw was ensiled with LAB inoculant (1×10 5 cfu/g of fresh weight) and glucose (2% of fresh weight). Inoculation with LAB improved the fermentation as reflected in reductions in pH, acetic acid (by 3.7 to 78.3%), butyric acid (by -6.0 to 100.0%) and ammonia nitrogen (by 1.0 to 71.7%) concentrations, and increases in lactic acid (by 43.9 to 282.9%) and crude protein concentrations compared with the control. Application of LAB plus glucose was more effective in improving fermentation quality than LAB alone. The variety of rice straw which contained relatively high levels of water soluble carbohydrates (WSC) tended to obtain better fermentation quality. The results suggested that LAB application and selection of rice varieties whose straw contained high levels of WSC were effective in improvement of the fermentation quality of rice straw silage.

Dietary Response of Chimpanzees and Cercopithecines to Seasonal Variation in Fruit Abundance. II. Macronutrients

In a continuation of our study of dietary differentiation among frugivorous primates with simple stomachs, we present the first comparison of differences in dietary macronutrient content between chimpanzees and cercopithecine monkeys. Previously we have shown that chimpanzee and monkey diets differ markedly in plant part and species content. We now examine whether this diet diversity is reflected in markedly different dietary macronutrient levels or the different feeding strategies yield the same macronutrient levels in their diets. For each primate group we calculated the total weighted mean dietary content of 4 macronutrients: crude lipid (lipid), crude protein (CP), water-soluble carbohydrates (WSC), and total nonstructural carbohydrates (TNC). We also calculated 4 fiber fractions: neutral-detergent fiber (NDF), which includes the subfractions hemicellulose (HC), cellulose (Cs), and sulfuric acid lignin (Ls). The HC and Cs are potentially fermentable fibers and would contribute to the energy provided by plant food, depending on the hind gut fermenting capacity of the individual primate species. The chimpanzee diet contained higher levels of WSC and TNC because during times of fruit abundance the chimpanzees took special advantage of ripe fruit, while the monkeys did not. The monkey diets contained higher levels of CP because the monkeys consumed a constant amount of leaf throughout the year. All four primate species consumed diets with similar NDF levels. However, the chimpanzees also took advantage of periods of ripe fruit abundance to decrease their Ls levels and to increase their HC levels. Conversely, the monkey diets maintained constant levels of the different fiber fractions thoughout the year. Nevertheless, despite these differences, the diets of the 4 frugivores were surprisingly similar, considering the substantial differences in body size. We conclude that the chimpanzee diet is of higher quality, particularly of lower fiber content, than expected on the basis of their body size.

The effects of sorbates on the ensilage of chopped whole‐plant maize and lucerne

AbstractThe incorporation of (a) sorbic acid (0.18 and 0.90 g kg−1, fresh weight basis) and potassium sorbate (0.90 g kg−1) with chopped lucerne, and (b) sorbic acid (0.90 g kg−1) and potassium sorbate (0.90 g kg−1) with chopped maize (whole‐plant) at the time of ensiling, led to a reduction in the surface spoilage associated with the ensilage of these two forages. Use of sorbic acid (0.90 g kg−1) and potassium sorbate (0.90 g kg−1) led to (a) a reduction of volatile nitrogen (VN), higher levels of residual water‐soluble carbohydrates (WSC) in lucerne silage and more aerobically stable lucerne silage and (b) a reduction in weight loss, population of yeasts and moulds, and VN, higher levels of WSC in maize silage and more aerobically stable maize silage when compared to the untreated forages or forages treated with sorbic acid and potssium sorbate at 0.045 and 0.18 g kg−1 (fresh weight basis), respectively.

Beef production from formic acid-treated and wilted silages

SUMMARY1. Herbage was ensiled, fresh at 17% DM or wilted to 32% DM, with and without the addition of formic acid (19·2 kg/tonne DM). The silages were fed individually ad libitum to 36 fifteen-month-old British Friesian steers of about 280 kg initial live weight (nine animals per treatment).2. Wilting and the application of formic acid both inhibited fermentation, resulting in silages with higher levels of water-soluble carbohydrates, lower concentrations of volatile nitrogen and lower organic acid contents.3. There were no significant dry matter × formic acid interactions in dry-matter intake or daily live-weight gain.4. Treatment with formic acid did not enhance significantly either dry-matter intake or daily live-weight gain. Furthermore, it had little effect on digestibility, nitrogen retention or metabolizable energy (ME) concentration.5. Wilting before ensiling increased dry-matter intake from 5·0 to 8·3 kg/head per day with consequent marked increases in ME intake, nitrogen retention and live-weight gain. Wilting, however, had little effect on digestibility, ME concentration, or the efficiency with which dietary nitrogen intake was retained.
