Autoregulation of Acetylcholine Release and Micro-Pharmacodynamic Mechanisms at Neuromuscular Junction: Selective Acetylcholinesterase Inhibitors for Therapy of Myasthenic Syndromes.

Neuromuscular junctions (NMJs) are directly involved into such indispensable to life processes as respiration and locomotion. However, motor nerve forms only one synaptic contact at each muscle fiber. This unique configuration requires specific properties and constrains to be effective. The very high density of acetylcholine receptors (AChRs) of muscle type in synaptic cleft and an excess of acetylcholine (ACh) released under physiological conditions make this synapse extremely reliable. Nevertheless, under pathological conditions such as myasthenia gravis and congenital myasthenic syndromes, the safety factor can be markedly reduced. Drugs used for short-term symptomatic therapy of these pathological states, cause partial inhibition of cholinesterases (ChEs). These enzymes catalyze the hydrolysis of ACh, thus terminate its action on AChRs. Extension of the lifetime of ACh molecules compensates muscular AChRs abnormalities and, consequently, rescues muscle contractions. In this mini review, we will first outline the functional organization of the NMJ, and then, consider the concept of the safety factor and how it may be changed. This will be followed by a look at autoregulation of ACh release that influences the safety factor of NMJs. Finally, we will consider the morphological features of NMJs as a putative reserve to increase effectiveness of pathological muscle weakness therapy by ChEs inhibitors due to opportunity to use micro-pharmacodynamic mechanisms.

CLASP2-dependent microtubule capture at the neuromuscular junction membrane requires LL5β and actin for focal delivery of acetylcholine receptor vesicles.

A hallmark of the neuromuscular junction (NMJ) is the high density of acetylcholine receptors (AChRs) in the postsynaptic muscle membrane. The postsynaptic apparatus of the NMJ is organized by agrin secreted from motor neurons. The mechanisms that underlie the focal delivery of AChRs to the adult NMJ are not yet understood in detail. We previously showed that microtubule (MT) capture by the plus end-tracking protein CLASP2 regulates AChR density at agrin-induced AChR clusters in cultured myotubes via PI3 kinase acting through GSK3β. Here we show that knockdown of the CLASP2-interaction partner LL5β by RNAi and forced expression of a CLASP2 fragment blocking the CLASP2/LL5β interaction inhibit microtubule capture. The same treatments impair focal vesicle delivery to the clusters. Consistent with these findings, knockdown of LL5β at the NMJ in vivo reduces the density and insertion of AChRs into the postsynaptic membrane. MT capture and focal vesicle delivery to agrin-induced AChR clusters are also inhibited by microtubule- and actin-depolymerizing drugs, invoking both cytoskeletal systems in MT capture and in the fusion of AChR vesicles with the cluster membrane. Combined our data identify a transport system, organized by agrin through PI3 kinase, GSK3β, CLASP2, and LL5β, for precise delivery of AChR vesicles from the subsynaptic nuclei to the overlying synaptic membrane.

Synergistic effects of neuregulin and agrin on muscle acetylcholine receptor expression

The proper function of neuromuscular junctions requires an extremely high density of acetylcholine receptors (AChRs) that may be achieved from neuron-derived factors including agrin and neuregulin. Here, we show that neuregulin-1 and agrin co-localize at neuromuscular junctions in vivo and form complexes when co-transfected into COS-7 cells. When these COS-7 cells are cultured with myotubes, synergistic effects are observed for AChR clustering, membrane insertion of new AChRs, and induction of AChR mRNA. Even a muscle form of agrin that lacks intrinsic clustering activities by itself, significantly enhances neuregulin-induced clustering and insertion of AChRs. While the heparin-binding (A) domain of agrin is required for agrin localization in the extracellular matrix adjacent to AChR clusters, the heparan sulfate-containing domain of agrin is needed for the synergistic effects and co-localization with neuregulin-1. These results suggest that matrix interactions between exogenously supplied agrin and neuregulin-1 on the muscle surface provide a localized source of signaling factors needed to produce high densities of AChRs at neuromuscular junctions.

Redistribution of presynaptic proteins during alpha-latrotoxin-induced release of neurotransmitter and membrane retrieval at the frog neuromuscular junction.

Calcium-dependent exocytosis at the nerve terminal involves the synaptic core (SNARE) complex composed of the t-SNAREs syntaxin 1 and synaptosome-associated protein of 25 kDa (SNAP-25), and the v-SNARE vesicle-associated membrane protein (VAMP/synaptobrevin), a stable heterotrimer which can associate with the putative calcium sensor protein, synaptotagmin. The distribution of these proteins at the frog neuromuscular junction was examined by immunofluorescent staining and confocal microscopy following exocytosis induced by alpha-latrotoxin. Experiments were performed under conditions in which synaptic vesicle recycling was either maintained in balance with exocytosis, or completely blocked, or during recovery from block of endocytosis. When endocytosis was maintained, protein distribution was essentially identical to that of unstimulated nerve terminals, in which syntaxin 1 and SNAP-25 are localized to the presynaptic active zones coincident with the postsynaptic folds that contain a high density of acetylcholine receptors (AChRs). Block of endocytosis led to complete incorporation of vesicle proteins into the plasmalemma, and t-SNARE distribution was no longer restricted to active zones. Five minutes after the onset of recovery, both synaptic vesicle proteins and t-SNARE proteins were concentrated into small spots, in a similar pattern to that obtained following endocytosis of the vital styryl dye FM1-43. These findings are consistent with a model in which following sustained exocytosis, t-SNARE trafficking involves internalization and transit via a vesicular compartment before recycling to the presynaptic plasma membrane.

Formation of functional synaptic connections between cultured cortical neurons from agrin-deficient mice

Numerous studies suggest that the extracellular matrix protein agrin directs the formation of the postsynaptic apparatus at the neuromuscular junction (NMJ). Strong support for this hypothesis comes from the observation that the high density of acetylcholine receptors (AChR) normally present at the neuromuscular junction fails to form in muscle of embryonic agrin mutant mice. Agrin is expressed by many populations of neurons in the central nervous system (CNS), suggesting that this molecule may also play a role in neuron-neuron synapse formation. To test this hypothesis, we examined synapse formation between cultured cortical neurons isolated from agrin-deficient mouse embryos. Our data show that glutamate receptors accumulate at synaptic sites on agrin-deficient neurons. Moreover, electrophysiological analysis demonstrates that functional glutamatergic and gamma-aminobutyric acid (GABA)ergic synapses form between mutant neurons. The frequency and amplitude of miniature postsynaptic glutamatergic and GABAergic currents are similar in mutant and age-matched wild-type neurons during the first 3 weeks in culture. These results demonstrate that neuron-specific agrin is not required for formation and early development of functional synaptic contacts between CNS neurons, and suggest that mechanisms of interneuronal synaptogenesis are distinct from those regulating synapse formation at the neuromuscular junction.

Distribution of components of the SNARE complex in relation to transmitter release sites at the frog neuromuscular junction.

At the frog neuromuscular junction, neurotransmitter release sites are regularly spaced at 1 micron intervals along the nerve terminal, directly facing postsynaptic folds which contain a high density of acetylcholine receptors. Immunostaining and laser confocal scanning microscopy were used to compare the distribution of presynaptic proteins implicated in exocytosis with that of fluorescent alpha-bungarotoxin. Syntaxin, synaptosome-associated 25 kDa protein and calcium channels were located predominantly at release sites. Synaptobrevin (vesicle-associated membrane protein) was distributed in the cytoplasm of the nerve terminal, presumably in the packets of microvesicles associated with each active zone. N-Ethylmaleimide-sensitive fusion protein (NSF) and soluble NSF attachment proteins (alpha beta SNAP) displayed a diffuse distribution throughout the terminal cytoplasm and also colocalized in distinct concentrated zones adjacent to the presynaptic membrane.

Potassium inward rectifier and acetylcholine receptor channels in embryonic Xenopus muscle cells in culture.

Embryonic muscle cells of the frog Xenopus laevis were isolated and grown in culture and single-channel recordings of potassium inward rectifier and acetylcholine (ACh) receptor currents were obtained from cell-attached membrane patches. Two classes of inward rectifier channels, which differed in conductance, were apparent. With 140 mM potassium chloride in the electrode, one channel class had a conductance of 28.8 +/- 3.4 pS (n = 21), and, much more infrequently, a smaller channel class with a conductance of 8.6 +/- 3.6 pS (n = 7) was recorded. Both channel classes had relatively long mean channel open times, which decreased with membrane hyperpolarization. The probability of finding a patch of membrane with an inward rectifier channel was high (66%) and many membrane patches contained more than one inward rectifier channel. The open state probability (with no applied potential) was high for both inward rectifier channel classes so that 70% of the time there was a channel open. Seventy-three percent of the membrane patches with ACh receptor channels (n = 11) also had at least one inward rectifier channel present when the patch electrode contained 0.1 mu M ACh. Inward rectifier channels were also found at 71% of the sites of high ACh receptor density (n = 14), which were identified with rhodamine-conjugated alpha-bungarotoxin. The results indicate that the density of inward rectifier channels in this embryonic skeletal muscle membrane was relatively high and includes sites of membrane that have synaptic specializations.

Acetylcholine receptor-associated 43K protein contains covalently bound myristate.

Torpedo electroplaque and vertebrate neuromuscular junctions contain high levels of a nonactin, 43,000-Mr peripheral membrane protein referred to as the 43K protein. 43K protein is associated with the cytoplasmic face of postsynaptic membranes at areas of high acetylcholine receptor density and has been implicated in the establishment and/or maintenance of these receptor clusters. Cloning of cDNAs encoding Torpedo 43K protein revealed that its amino terminus contains a consensus sequence sufficient for the covalent attachment of the rare fatty acid myristate. To examine whether 43K protein is, in fact, myristoylated, mouse muscle BC3H1 cells were metabolically labeled with either [35S]cysteine or [3H]myristate and immunoprecipitated with a monospecific antiserum raised against isolated Torpedo 43K protein. In cells incubated with either precursor, a single labeled species was specifically recovered that comigrated on SDS-PAGE with 43K protein purified from Torpedo electric organ. Approximately 95% of the 3H labeled material released from [3H]myristate-43K protein by acid methanolysis was extractable in organic solvents and eluted from a C18 reverse-phase HPLC column exclusively at the position of the methyl myristate internal standard. Thus, 43K protein contains authentic myristic acid rather than an amino or fatty acid metabolite of [3H]myristate. Myristate appears to be added to 43K protein cotranslationally and cannot be released from it by prolonged incubation in SDS, 2-mercaptoethanol, or hydroxylamine (pH 7.0 or 10.0), characteristics consistent with amino terminal myristoylation. Covalently linked myristate may be responsible for the high affinity of purified 43K protein for lipid bilayers despite the absence of a notably hydrophobic amino acid sequence.

Acetylcholine receptor-aggregating proteins are associated with the extracellular matrix of many tissues in Torpedo.

The synaptic basal lamina, a component of extracellular matrix (ECM) in the synaptic cleft at the neuromuscular junction, directs the formation of new postsynaptic specializations, including the aggregation of acetylcholine receptors (AChRs), during muscle regeneration in adult animals. Although the molecular basis of this phenomenon is unknown, it is mimicked by AChR-aggregating proteins in ECM-enriched fractions from muscle and the synapse-rich electric organ of the ray Torpedo californica. Molecules immunologically similar to these proteins are concentrated in the synaptic basal lamina at neuromuscular junctions of the ray and frog. Here we demonstrate that immunologically, chemically, and functionally similar AChR-aggregating proteins are also associated with the ECM of several other tissues in Torpedo. Monoclonal antibodies against the AChR-aggregating proteins from electric organ intensely stained neuromuscular junctions and the ventral surfaces of electrocytes, structures with a high density of AChRs. However, they also labeled many other structures which have basal laminae, including the extrajunctional perimeters of skeletal muscle fibers, smooth and cardiac muscle cells, Schwann cell sheaths in peripheral nerves, walls of some blood vessels, and epithelial basement membranes in the gut, skin, and heart. Some structures with basal laminae did not stain with the antibodies; e.g., the dorsal surfaces of electrocytes. Bands of similar molecular weight were detected by the antibodies on Western blots of extracts of ECM-enriched fractions from electric organ and several other tissues. Proteins from all tissues examined, enriched from these extracts by affinity chromatography with the monoclonal antibodies, aggregated AChRs on cultured myotubes. Thus, similar AChR-aggregating proteins are associated with the extracellular matrix of many Torpedo tissues. The broad distribution of these proteins suggests they have functions in addition to AChR aggregation.

Development of the electromotor system of Torpedo marmorata: distribution of extracellular matrix and cytoskeletal components during acetylcholine receptor focalization.

A combination of direct fluorescence and indirect immunofluorescence microscopy has been used to compare the distribution of the acetylcholine receptor with the distribution of major cytoskeletal and extracellular matrix components during electrocyte differentiation in the electric organs of Torpedo marmorata. Laminin, fibronectin and extracellular matrix proteoglycan are always more extensively distributed around the differentiating cell than the acetylcholine receptor-rich patch that forms on the ventral surface of the cell. The distribution of acetylcholinesterase within the ventral surface of the differentiating electrocyte closely resembles the distribution of the acetylcholine receptor. Areas of apparently high acetylcholine receptor density within the ventrally forming acetylcholine receptor-rich patch are always areas of apparently high extracellular matrix proteoglycan density but are not always areas of high laminin or fibronectin density. Desmin levels appear to increase at the onset of differentiation and desmin initially accumulates in the ventral pole of each myotube as it begins to form an electrocyte. During differentiation F-actin-positive filament bundles are observed that extend from the nuclei down to the ventrally forming acetylcholine receptor-rich patch. Most filament bundles terminate in the acetylcholine receptor-rich region of the cell membrane. Electron-microscopic autoradiography suggests that the filament bundles attach to the membrane at sites where small acetylcholine receptor clusters are found. The results of this study suggest that, out of the four extracellular matrix components studied, only the distribution of acetylcholinesterase (which may be both matrix- and membrane-bound at this stage) closely parallels that of the acetylcholine receptor, and that F-actin filament bundles terminate in a region of the cell that is becoming an area of high acetylcholine receptor density.

Association of beta-cytoplasmic actin with high concentrations of acetylcholine receptor (AChR) in normal and anti-AChR-treated primary rat muscle cultures.

Previous immunocytochemical studies in which an antibody specific for mammalian cytoplasmic actin was used showed that a high concentration of cytoplasmic actin exists at neuromuscular junctions of rat muscle fibers such that the distribution of actin corresponded exactly to that of the acetylcholine receptors. Although clusters of acetylcholine receptors also are present in noninnervated rat and chick muscle cells grown in vitro, neither the mechanism for the formation and maintenance of these clusters nor the relationship of these clusters to the high density of acetylcholine receptors at the neuromuscular junction in vivo are known. In the present study, a relationship between beta-cytoplasmic actin and acetylcholine receptors in vitro has been demonstrated immunocytochemically using an antibody specific for the beta-form of cytoplasmic actin. Networks of cytoplasmic actin-containing filaments were found in discrete regions of the myotube membrane that also contained high concentrations of acetylcholine receptors; such high concentrations of acetylcholine receptors have been described in regions of membrane-substrate contact. Moreover, when primary rat myotubes were exposed to human myasthenic serum, gross morphological changes, accompanied by an apparent rearrangement of the cytoplasmic actin-containing cytoskeleton, were produced. Although whether the distribution of cytoplasmic actin-containing structures was influenced by the organization of acetylcholine receptor or vice versa cannot be determined from these studies, these findings suggest that in primary rat muscle cells grown in vitro, acetylcholine receptors and beta-cytoplasmic actin-containing structures may be somehow connected.

Evidence that coated vesicles transport acetylcholine receptors to the surface membrane of chick myotubes.

Coated vesicles are present in the myoplasm of embryonic chick myotubes grown in vitro. They are most numerous beneath regions of the surface membrane that contain a high density of acetylcholine receptors (AChR). Prolonged exposure of myotubes to saline extract of chick brain increases the number of intracellular AChR and the number of coated vesicles. This suggests that coated vesicles contain AChR, and this hypothesis was tested with horseradish peroxidase-alpha-bungarotoxin (HRP-alpha BTX) conjugates. The conjugates enter saponin-permeabilized cells and, as judged by the inhibition of [125I] alpha BTX binding, they label the entire intracellular AChR pool. Approximately 50% of the coated vesicles contained HRP-alpha BTX reaction product. In addition, reaction product was detected in Golgi cisternae and along membranes that bound a subsurface tubulovesicular network. The majority of labeled vesicles are probably involved in exocytosis rather than endocytosis of AChR because very few coated vesicles were labeled when HRP-alpha BTX was added to the medium bathing intact cells. Moreover, inhibition of protein synthesis with puromycin resulted in a large decrease in the number of labeled vesicles. These results suggest that a subpopulation of coated vesicles ferry newly synthesized AChR to the cell surface.

Association of laminin and other basement membrane components with regions of high acetylcholine receptor density on cultured myotubes

The distribution of immunoreactivity to basement membrane components in cultures of rat skeletal myotubes was compared to acetylcholine receptor distribution by fluorescence microscopy. Laminin occurred in patches on the myotube surface, and most laminin patches coincided or overlapped with acetylcholine receptor aggregates. Almost all receptor aggregates coincided with laminin patches. Most of the heparan sulfate proteoglycan and fibronectin was associated with non-muscle cells, but some patches coincided with receptor aggregates on myotubes. In cultures treated with l-ascorbate, collagen types IV and V covered much of the myotube surface and receptor aggregates often coincided with intense collagen patches. When receptor aggregation was induced by treatment of cultures with soluble neural factors, the newly formed receptor aggregates coincided with laminin patches.The results suggest that each of the basement membrane components studied has a distinct distribution pattern in primary rat muscle cell cultures. The striking association of laminin immunoreactivity with receptor aggregates, together with the enhancement of receptor aggregation by laminin reported previously (Vogel et al., J. Neurosci. 3, 1058–1068, 1983) suggests that laminin could have a role in the organization of acetylcholine receptors on developing muscle fibers.

Increased adhesiveness at sites of high acetylcholine receptor density on embryonic amphibian muscle cells cultured without nerve

In culture, myotomal muscle cells from Xenopus laevis embryos develop discrete patches of high acetylcholine receptor (AChR) density. To examine the relative adhesiveness of these sites, muscle cells having AChR patches on their lower surface (apposed to the culture dish) were identified and were then treated with dibucaine or potassium-Ringer in order to cause the cells to round up. More than 90% of these cells remained attached at an AChR patch after rounding up, and this was the case even when the cells had a single patch on their lower surface. When the cells were torn away from the culture dish by mechanical agitation, small cellular fragments still remained firmly attached to the dish and many of these fragments contained an AChR patch. It is concluded that AChR patches on the lower surface of the cell are often located at sites of increased adhesiveness to the culture dish. The findings are discussed in terms of the formation and maintenance of AChR patches.

Postnatal maturation of nerve-muscle junctions in hindlimb muscles of the mouse

The morphological maturation of nerve-muscle junctions in hindlimb muscles of the mouse was studied during the first month after birth. At birth the adult population of muscle fibers and motor axons is nearly complete and the latter are about to become myelinated in their extramuscular region. Each neonatal muscle fiber is innervated within a well-defined junctional zone by numerous fine processes of several motor axons. This zone is occupied roughly uniformly by a high density of acetylcholine receptors (AChR, labeled with a fluorescent conjugate of α-bungarotoxin). Polyneuronal innervation of individual muscle fibers is lost during the second week after birth. Following the elimination of redundant synaptic contacts the surviving preterminal axons grow and become myelinated and their terminals expand within the junctional zone which begins to grow more rapidly. The distribution of AChR changes markedly during the third week after birth. Spots of low AChR density appear which enlarge to form the nonsynaptic regions of the mature junctional zone as the receptors become closely associated with the maturing branches of the motor axon terminals. The subsequent increase in area of the junctional zone closely parallels that of the muscle fiber as a whole.

Localization of cholinesterase at sites of high acetylcholine receptor density on embryonic amphibian muscle cells cultured without nerve

Myotomal muscle cells, derived from Xenopus embryos and grown in culture without nerve, develop discrete sites of cholinesterase (ChE) activity on their surface. The spatial relationship of these ChE patches to surface patches of acetylcholine receptors (AChRs) has been examined in the present study by a combination of ChE histochemistry and fluorescent staining of the receptors. ChE patches and AChR patches developed as early as the 1st day in culture and exhibited a high incidence of spatial overlap. The frequency of overlap varied with the age of the culture and ranged from 50 to 98% for patches on the lower cell surface (facing the floor of the culture dish) and from 28 to 79% for patches on the upper cell surface. The high incidence of overlap cannot be explained on the basis of a random distribution of patches since both types of patch occupied less than 3% of the cell surface. ChE and AChR patches also developed when cultures were grown in a serum-free medium as well as when cultures were prepared from young embryos in which muscle innervation had not yet begun. At some patches, the surface membrane was invaginated and at these invaginations, there was also a high incidence of overlap between the ChE and AChR stains. It is concluded that the mechanisms involved in the localization of AChRs and ChE on the surface of Xenopus myotomal muscle cells tend to be closely linked and operate even in the absence of innervation, previous contact by nerve, or electrical and contractile activity. Considered together with previous ultrastructural observations, the present findings suggest that these cells develop elaborate postsynaptic-like specializations even in the absence of neural factors.

Neuromuscular junctions and alpha-bungarotoxin-binding sites in denervated and contralateral cat skeletal muscles.

1. The distributions of cholinesterase (ChE) activity and acetylcholine (ACh) receptors were studied in normal and denervated cat hind-limb fast-twitch skeletal muscles and in muscles contralateral to denervated muscles. 2. On normal muscle fibres almost all receptors were confined to the immediately post-junctional membrane, although a perijunctional gradient of increased ACh receptor density was found on both fast- and slow-twitch fibres. After denervation, the extrajunctional ACh receptor density increased greatly and remained high for at least 10 months. Ectopic regions staining for ChE activity and having a high density of ACh receptors appeared in denervated muscle. The number of junctional ACh receptors decreased slowly after denervation, with a half-time of about 140 days. 3. Fast-twitch muscles contralateral to denervated muscles also showed changes, including an increase in junctional size and a small but significant increase in extra-junctional ACh receptor density.

Identification of regions of high acetylcholine receptor density in tannic acid-fixed postsynaptic membranes from electric tissue

Several distinct membrane specializations in electric tissue of the torpedine ray, Narcine brasiliensis, are rendered visible in thin section electron microscopy by the use of tannic acid during sample preparation. The acetylcholine receptor-containing regions of the postsynaptic membrane have a morphology shared by no other membrane in the tissue: a thick outer lamina, previously ascribed by others to the receptor protein itself, and a thick (∼8nm) inner lamina which is precisely co-extensive with the thick outer lamina. both of these features are found in biochemical preparations of the postsynaptic membrane upon fixation in the presence of tannic acid. Other membrane specializations having thick outer laminae can be distinguished from that due to the receptor. Thus, receptor-containing regions in isolated membrane fragments can be unambiguously identified by inspection. This provides an important practical tool for the study of these membranes. In addition, the data presented here and elsewhere suggest a model for the receptor-containing membrane in which material, presumably protein, is roughly symmetrically distributed on the two sides of the membrane.

Localization of acetylcholine receptors and synaptic ultrastructure at nerve-muscle contacts in culture: dependence on nerve type.

In cultures of xenopus myotomal muscle cells and spinal cord (SC) some of the nerve-muscle contacts exhibit a high density of acetylcholine receptors (AchRs [Anderson et al., 1977, J. Physiol. (Lond.). 268:731- 756,757-773]) and synaptic ultrastructure (Weldon and Cohen, 1979, J. Neurocytol. 8:239-259). We have examined whether similarly specialized contacts are established when the muscle cells are cultured with explants of xenopus dorsal root ganglia (DRG) or sympathetic ganglia (SG). The outgrowth from the ganglionic explants contained neuronal and non- neuronal cell processes. Although both types of processes approached within 100 A of muscle cells, synaptic ultrastructure was rarely observed at these contacts. Because patches of postsynaptic ultrastructure also develop on noncontacted muscle cells, the very few examples of contacts with such specializations probably occurred by chance. AChRs were stained with fluroscent alpha-bungarotoxin. More than 70 percent of the SC-contacted muscle cells exhibited a high receptor density along the path of contact. The corresponding values for DRG- and SG- contacted muscle cells were 10 and 6 percent. Similar values were obtained when the ganlionic and SC explants were cultured together in the same chamber. The few examples of high receptor density at ganglionic-muscle contacts resembled the characteristic receptor patches of noncontacted muscle cells rather than the narrow bands of high receptor density seen at SC-muscle contacts. In addition, more than 90 percent of these ganglionic- contacted muscle cells had receptor patches elsewhere, compared to less than 40 percent for the SC-contacted muscle cells. These findings indicate that the SC neurites possess a specific property which is important for the establishment of synaptically specialized contacts with muscle and that this property is lacking in the DRG and SG neurites.

Development of the neuromuscular junction in the chick embryo: The number, distribution, and stability of acetylcholine receptors

The number, distribution, and stability of skeletal muscle acetylcholine receptors during development of the neuromuscular junction in the chick embryo were studied. The distribution and turnover of receptors labeled with 125I-labeled α-bungarotoxin were determined by quantitative autoradiography on individual teased muscle fibers. Each posterior latissimus dorsi muscle fiber, which in the adult is singly innervated, has a high density of acetylcholine receptors at a single spot from embryonic Day 10 through hatching. The spots stain more intensely than elsewhere for acetylcholinesterase and are assumed to be end plates. The receptors at these spots are presumed to be junctional receptors. The junctional receptor density remains constant at 10 4/μm 2 from embryonic Day 14 through adult life, although the area of the junction increases 40-fold. In contrast, the extrajunctional receptor density drops precipitously from 250/μm 2 on Day 14 to only 10/μm 2 on Day 19. This decrease in extrajunctional receptor density can be prevented by chronic paralysis with curare. The rate of autoradiographic grain loss from junctional and extrajunctional regions after a pulse injection of 125I-labeled α-bungarotoxin indicates that both classes of embryonic receptors turn over at the same rate ( t 1 2 ⋍ 30 hr ).
