Assessment of Quality Control parameters under Accelerated Stability Conditions for Manasamitra Vatakam; A Herbo Mineral Formulation

Accelerated Stability Studies predicts the stability data and significantly explains the drug decision process. In the present study, Manasamitra Vatakam, a Herbo mineral formulation was subjected to stability conditions at 40°C ± 2°C /75%± 5% RH for 06 months. Control and stability samples were determined for parameters like Weight variation, Moisture content, Marker content determination, and Disintegration test, Dissolution study with different mediums, Friability and Hardness test. The phytomarker analysis showed decrease assay value for β-sitosterol, Vitexin, Kampferol and Withaferin. The dissolution studies were performed for the major biomarker Berberine in 0.1N HCl, pH 4.5 acetate and pH 6.8 phosphate buffer in which pH 6.8 buffer showed maximum release of 66.82% at 04 hours. The other Quality control parameters showed minimal difference under acceptance criteria. The study explains the need of stability analysis in polyherbal formulations and developed method can be used for routine analytical studies in various formulation containing berberine. THERAPEUTIC EFFICACY OF ASHTABHAIRAVA MATHIRAI IN FEVER MANAGEMENT: A REVIEW

Ashtabhairava mathirai is an herbomineral formulation used in the Siddha system. Its application in treating fever has been described in the Siddha Vaidhiya Thirattu textbook, Hence, a review of the ingredients is essential. This review aims to critically assess the available literature on the indication of Ashtabhairava mathirai in treating fever, its therapeutic potential, and its mechanism of action. A comprehensive literature search was conducted across multiple databases, including Siddha literature. The ingredients of Ashtabhairava mathirai exhibit various pharmacological activities, such, as antipyretic, anti-inflammatory, antimicrobial, and antioxidant activity. The study concluded that the multiple actions of ingredients of Ashtabhairava mathirai which is used to treat the condition, fever. These findings support the traditional therapeutic value of Ashtabhairava mathirai in treating 64 types of fever.

UNRAVELLING THE WISDOM OF KHARALIYA RASAYANA: A REVIEW

From Ancient Practices to Modern Science: A Review on the Synthesis and Characterization of Ayurvedic Bhasmas

Ayurvedic Bhasma, an ancient form of Indian traditional medicine, represents a unique class of herbo-mineral formulations prepared through a meticulous process involving calcination. This review provides a comprehensive synthesis and characterization of Ayurvedic Bhasmas, focusing on its preparation methods, physicochemical properties, and characterization. The synthesis of Bhasma involves a series of Shodhana (purification), Marana (incineration), and Bhavana (soaking and drying) processes, each contributing to the transformation of raw minerals and herbs into bioavailable nanoparticles with enhanced efficacy and reduced toxicity. The review delves into the intricate processes involved in the production of various bhasmas, prepared in our laboratory as well as reports by other researchers. A CLINICAL STUDY TO EVALUATE THE EFFECTIVENESS OF GANDHAKA RASAYANA IN THE MANAGEMENT OF GRAHANI ROGA

Background & Objective: Grahani Roga is considered a Tridoshatmaka disorder of the digestive system, directly related to the status of Agni and diet habits, which occurs due to the vitiation of Agni (Mandagni) and Doshas. Gandhaka Rasayana is a standard Herbo-mineral formulation of Ayurveda, used extensively in treating various skin, wound and gastrointestinal disorders. It was believed to be significant in Amapachana, Agnideepana, Rasayana, Raktaprasadana, Kledaghna, and Krimihara activity. In this study, we evaluated the effect of Gandhaka Rasayana in the management of Grahani Roga. Materials & Methods: The trial drug Gandhaka Rasayana was administered 500 mg twice a day to 90 selected patients of Grahani Roga. After 45 days of trial, the effectiveness of Gandhaka Rasayana was statistically evaluated based on various parameters, such as Subjective, Clinical, and Objective improvement. Results and Conclusion: Gandhaka Rasayana was found statistically highly significant in the Clinical Symptoms of Grahani Roga, with 38.39% average improvement; it also had a significant effect on Agni Bala and Deha Bala, with 48.81% and 39.09% average improvement, respectively. Hence, the overall average effect on parameters of clinical improvement after the therapeutic trial of Gandhaka Rasayana in 90 patients of Grahani Roga was 42.09%. Thus, Gandhaka Rasayana is a very potent drug and can be used effectively in the management of Grahani Roga.

CRITICAL REVIEW OF PASHANBHINNA RASA ON ASHMARI WITH SPECIAL REFERENCE TO UROLITHIASIS

Ayurveda, an ancient system of medicine, addresses numerous disorders related to the Mutravaha Srotas (uri-nary system), such as Mutrakricchra (dysuria), Mutraghata (obstructive uropathy), and Mutrashmari (urinary calculi). Among these, Mutrashmari, corresponding to urolithiasis in modern medicine, is one of the most com-mon and distressing conditions, characterised by the formation of stones within the urinary tract. Sushruta iden-tifies two primary mechanisms of stone formation: urine stagnation and supersaturation and crystallisation of urinary crystalloids. Urolithiasis affects approximately 12% of the global population, with a high recurrence rate, especially in males. This review aims to provide a comprehensive analysis of Pashanbhinna Rasa, a herbo-mineral formulation from the Bhaishajya Ratnavali, Ashmari Prakaran, assessing its efficacy in treating Ash-mari (urolithiasis)—the study endeavours to bridge the knowledge gap in contemporary literature regarding this formulation. A thorough literature review focused on references to Pashanbhinna Rasa in classical Ayurvedic texts and modern scientific studies. The review highlights Pashanbhinna Rasa's multifaceted approach to man-aging urolithiasis. The herbo-mineral components of Pashanbhinna Rasa are believed to exert a synergistic ef-fect, potentially dissolving stones, preventing their formation, and providing symptomatic relief. This critical review elucidates the potential of Pashanbhinna Rasa as an effective treatment for urolithiasis. While traditional Ayurvedic formulations offer promising therapeutic benefits, further clinical studies are necessary to validate their efficacy scientifically. Establishing Pashanbhinna Rasa as a standardised treatment could integrate Ayur-vedic wisdom with modern medical practice, offering a holistic approach to managing urolithiasis.

Role of Shwasakuthar Rasa and Other Herbomineral Formulations with Yoga in Migraine

Ayurveda is an ancient traditional system of medicine to treat illness and encourage well being by using natural whole body healing techniques. Migraine is a primary headache disorder which is characterized by episodic recurrent attacks. As it affects one billion individuals each year across the worldwide has proven to one of the most common neurological disorders with high prevalence and morbidity among the younger age groups. Migraine can be classified on the basis of presence of Aura which shows prodromal symptoms like irritability, behavioural changes, etc and associated symptoms of photophobia, phonophobia and vomiting that usually lasts for 4 to 72 hours. In Ayurveda, migraine is clinically correlated with Ardhavabhedaka which is described as Tridoshapradhana by Sushruta and Vatakaphapradhana by Vagabhata. The management of migraine is done routinely by reducing the symptoms and the episodic recurrent attacks with Ayurvedic approach as modern medicine cause gastric irritation and drug dependency which further triggers the attack. The present case of 35 years old female diagnosed with migraine has shown symptoms of headache involving left side of head associated with nausea, vomiting and sensitivity to light and sound. The patient was treated with Nasya Karma which act as a gateway to head as said by Acharya Charaka and with oral Ayurvedic herbomineral formulation Shamana Chikitsa was done that alleviates the symptoms of Ardhavabhedaka. A significant relief from all symptoms was observed within a period of 15 days, demonstrating the Ayurveda as the best way to improve the overall well being of mind, body and spirit.

Management of Bronchial Asthma Through Herbo-Mineral Drugs

Ayurveda is one of alternative way of treating illness, prevention and management of respiratory disorders. Nowadays respiratory disorders are the major health issue resulting primary cause of death and disability in the world. Asthma is a common respiratory problem increasing globally and affecting approximately 339 million people worldwide and 17.23 million people with an overall prevalence of 2.05% in India. Bronchial asthma is a heterogeneous disease, usually characterized by chronic inflammatory disease of the airway. It is defined by chronic respiratory problem characterized by hyper-responsiveness, inflammation and narrowing of airway resulting in cardinal symptoms cough, chest tightness, dyspnea and wheeze. Tamak swasa closely resembles to bronchial asthma. Swasa roga is major health problem affecting the Pranavaha srotas. According to Acharya Charak tamak swasa is Vata-Kaphaja Vyadhi which is originated from Pithasthan (Amashaya). The treatment of this disease is still unsatisfactory in modern medicine as the use of corticosteroids have many side effects and may lead to medicine dependency. Ayurveda is proven to be a boon for management of asthmatic patients (Tamak swasa). Our Ayurveda classics have explained a lot about signs and symptoms of asthma that has a direct correlation with Tamak swasa. So this is the need of the time that a direct, safe and holistic approach should be established to treat the disease with specific herbomineral formulations as mentioned in our Ayurveda classics.

Ayurvedic treatment protocol in the management of pancreatitis: A nonrandomized observational study

Recurrent Acute/Chronic Pancreatitis (RA/CP) is a progressively debilitating disease with rising incidences in recent years. The limitations of conventional treatment, along with the psychological fear and financial burden associated with it, compel the patients to explore alternative options. In India, where traditional medicines are recognized as treatment options, a North India-based ayurvedic clinic has been treating RA/CP patients using an ayurvedic Herbo-Mineral Formulation (HMF) with a balanced diet and regulated lifestyle. The HMF is prepared using processed mercury, copper, and sulfur following the principles of Rasashastra. The HMF has demonstrated pancreatitis preventive properties in rat models and passed acute, subacute, and chronic toxicity assessments. This retrospective study enrolled 1750 well-diagnosed cases of RA/CP from January 1997 to July 2023. About 67% of the enrolled patients were nonalcoholics, 81% were nontobacco users, and 93% had no family history of the disease. The age group of 19–45 years represented the highest proportion of patients, with a male predominance (5:1). Nine hundred and sixteen patients with RA/CP completed 1-year ayurvedic intervention using HMF, without pancreatic enzymes. The ayurvedic treatment resulted in a significant 93% reduction in the frequency of pancreatitis attacks and a 97% decrease in emergency hospitalizations. The HMF has shown no adverse effects or toxicity in the treated patients. About 1.7% of patients experienced mortality during the treatment or follow-up period due to various reasons. The ayurvedic treatment protocol demonstrated sustainable effects, with the longest remission exceeding 26 years, and has brought a significant reduction in frequency and intensity of RA/CP attacks with an overall improvement in quality of life, warranting further randomized clinical trials to establish strong proof of efficacy.

Role of Rasayana in communicable diseases with special reference to Covid-19: A Review

Communicable diseases continue to be prevalent and pose serious problems for public health. Recently, COVID 19 emerged out as one of the communicable diseases which was declared as a pandemic. COVID-19 pandemic has left its imprints on various health systems globally and caused immense social and economic disruptions. The scientific community across the globe is expecting potential leads from traditional systems of healthcare. Rasayana (rejuvenation therapy) is one of the comprehensive disciplines of Ayurveda which includes various approaches like use of herbs, food articles, herbo-mineral formulations, dietary and lifestyle changes. It strengthens the body’s immune system by improving health of body tissues and promoting a balanced state of body. Rasayana essentially imparts nutrition to the body at all levels from macro to micro-cellular level. Rasayana therapy replenishes the vital fluids of the body and it enhances the Ojas (vital force of life) resulting in increased energy and balanced mental and emotional health. Rasayana therapy is particularly helpful in management of such disorders where immunity and infections are playing central role like COVID-19. The therapy is helpful to increase the immunity of a person and hence protects from communicable diseases like COVID-19. As COVID 19 is still active, so Rasayana therapy can play a vital role to combat with the disease.

Pharmaceutical Analysis of a Herbo-mineral Formulation-Pippalyadi Agad from Yogaratnakar

The development of pharmaceuticals brought a revolution in human health. For the therapeutic efficacy of any drug, it should be authentic, and free from impurities. These pharmaceuticals may develop impurities at various stages of their development, transportation, and storage which makes them risky to be administered thus they must be detected and quantitated. Hence, a detailed pharmaceutical study of the drug is needed, including its Authentication and Standardization. Various medicinal formulations in Ayurveda need to be analysed with modern techniques. Pippalyadi Agad is one of them. Hence, the present study,“Pharmacological analysis of a herbo-mineral formulation- Pippalyadi Agad from Yogaratnakar” was selected. Methodology- The raw drugs were procured from an authentic source. Authentication and Standardization of raw drugs was conducted. The powder of raw drugs was mixed together to make the formulation. Then Physiochemical analysis was done and results were noted. Observation and Results- Organoleptic parameters of the raw drugs and the study drug were according to the standards. The results of Standardization comply with the standards given in API. This work can provide the correct identification and Standardization of the crude drug. These findings will be helpful in establishing the Standardization of Pippalyadi Agad.

Clinical evaluation of therapeutic efficacy of a Siddha herbo mineral formulation “Kumaara Veeriya Kaantha Chenduram’’ with “Nellikkai Legiyam” (internal) in the treatment of Iron Deficiency Anemia

Abstract: Background: Kumaara veeriya kaantha chenduram (KVKC) is a siddha herbomineral formulation indicated for paandunoi in Siddha literature (1).Paandu noi is a disease characterized by changes in the body's natural color andpallor found in the skin, nails and conjunctiva. Objectives: To determine the efficacy of the KVKC, the open labed clinical study was conducted in National Institute of Siddha OPD. Materials and Methods: The 30 patients with Iron deficiency anaemiawere selected and treated with KVKC orally with the dose of 260 milli gram twice in a day with Nellikailegiyam6 gram continuously for 45 days. Laboratory investigations for hemoglobin was done on 1st day and every 15 days once of the study. Results: Clinical assessment was done, based on the before and after treatment scores which was compared statistically and recorded.Regarding Hemoglobin level, 24(80%) patients showed increase of 1 to 3 grams from base level, 6(20%) patients showed increase of 0.1 to 0.9 grams from base level.Regarding Serum Ferritin, out of 30 patients, 9 (30%) patients showed 0.1-3.5 ng/ml increase from its base level, 21 (70%) patients showed 3.6-7.0 ng/ml increase from its base level.From the Statistical studies, the Mean ± Standard deviation for Haemoglobin before treatment was 9.7 ± 0.93 and after treatment was 11.18 ± 1.08 which was statistically highly significant (p<0.0001).And Serum ferritin before treatment was 9.27 ± 3.31, after treatment was 13.5 ± 3.49 which was statistically significant (p<0.0001).

In vitro cytotoxicity and apoptosis inducing effect of Vallarai Kirutham with Rasa Parpam in HeLa Cell Lines

Cancer is an uncontrolled proliferation of cells that can affect nearby tissues as well as distant organs. Cervical cancer is predicted to be among the four most prevalent cancers in women overall in 2020, with 342,000 mortality and 604,000 new cases. Around 6-29% of all cancers in women in India are caused by cervical cancer. Women who have had several sexual partners, recurrent abortions, or vaginal deliveries—all of which cause repetitive stress to the cervix are more likely to develop cervical cancer. Radiation, chemotherapy, and surgery are being used to treat cancer. All of these treatments come with side effects, which can range in severity and have a serious psychological impact on patients. The Siddha system has a number of accessible herbo mineral formulations for treating cervical cancer. According to Siddha literature (Agasthiyar Vaidhya Rathina Churukkam, 360), the treatment of cervical cancer (yoni puttru) is advised for the Siddha medicine Vallarai Kirutham with Rasa parpam. The goal of this research was to find out this formulation's potential anti-cancer properties using MTT assays on HeLa cell lines. These findings of the current study suggest that the MTT technique's lowest reading for cell viability was 1.26±0.009% at a concentration of 100 µl/ml. This was followed by concentrations of 75 µl/ml, 50µl/ml and 25µl/ml which showed 7.91±0.004%, 23.91 ±0.01% and 37.17± 0.01% similarly, 10 µl/ml shows 52.09±0.01%.The matching IC50 value was discovered to be 12.44% and AO/EB dual staining was used to examine apoptotic activity. The outcomes of in-vitro experiments using the HeLa cell line. Herbo-Mineral Medicine, Lithom Exhibits Anti-Nephrolithiasis Activity in Rat Model of Hyperoxaluria by Attenuating Calcium Oxalate Crystal Formation and Oxidative Stress.

Calcium oxalate monohydrate (COM) forms the most common type of kidney stones observed in clinics, elevated levels of urinary oxalate being the principal risk factor for such an etiology. The objective of the present study was to evaluate the anti-nephrolithiatic effect of herbo-mineral formulation, Lithom. The in vitro biochemical synthesis of COM crystals in the presence of Lithom was performed and observations were made by microscopy and Scanning Electron Microscope (SEM) based analysis for the detection of crystal size and morphology. The phytochemical composition of Lithom was evaluated by Ultra-High-Performance Liquid Chromatography (UHPLC). The in vivo model of Ethylene glycol-induced hyperoxaluria in Sprague-Dawley rats was used for the evaluation of Lithom. The animals were randomly allocated to 5 different groups namely Normal control, Disease control (ethylene glycol (EG), 0.75%, 28 days), Allopurinol (50 mg/kg, q.d.), Lithom (43 mg/kg, b.i.d.), and Lithom (129 mg/kg, b.i.d.). Analysis of crystalluria, oxalate, and citrate levels, oxidative stress parameters (malondialdehyde (MDA), catalase, myeloperoxidase (MPO)), and histopathology by hematoxylin and eosin (H&E) and Von Kossa staining was performed for evaluation of Lithom. The presence of Lithom during COM crystals synthesis significantly reduced the average crystal area, feret's diameter, and area-perimeter ratio, in a dose-dependent manner. SEM analysis revealed that COM crystals synthesized in the presence of 100 and 300 μg/mL of Lithom exhibited a veritable morphological transition from irregular polygons with sharp edges to smoothened smaller cuboid polygons. UHPLC analysis of Lithom revealed the presence of Trigonelline, Bergenin, Xanthosine, Adenosine, Bohoervinone B, Vanillic acid, and Ellagic acid as key phytoconstituents. In EG-induced SD rats, the Lithom-treated group showed a decrease in elevated urinary oxalate levels, oxidative stress, and renal inflammation. Von Kossa staining of kidney tissue also exhibited a marked reduction in crystal depositions in Lithom-treated groups. Taken together, Lithom could be a potential clinical-therapeutic alternative for management of nephrolithiasis.

Development of standard manufacturing process of Tryushanadya Lauha – An organo-metalic preparation

Tryushanadya Lauha (TL) is one of the herbo-mineral formulations in many Ayurvedic texts. Tryushanadya Lauha consists Loha Bhasma and Tryushana, which includes Pippali (Piper longum Linn), Maricha (Piper nigrum Linn), and Shunti (Zingiber officinale Roscoe), Cavya (Piper chaba Hunter), Citraka (Plumbago zeylanica Linn), Bakuchi (Psoralea Corylifolia Linn), and Lavana (salt), which includes Saindhava (Sodium chloride), Aubhida (sodium carbonate), Vida (Ammonium chloride), and Sauvarchala (Sodium sulphate). This study is an effort to develop the standard operating process for manufacturing of Loha Bhasma and Tryushanadya Lauha. As per the reference of Rasatarangini, Loha Bhasma (incinerated ash of iron) was prepared in three batches. The processing of Loha Bhasma (ash of iron) was performed by adopting, Shodhana (purification), a special heating process and Marana (incineration). For the process of Levigation decoction of Triphala was used. Puta (heating process) was given in Electric Muffle Furnace at a temperature of 500 0C. The percentage of loss was 49.9% after purification. During Loha Bhasma (incinerated ash of iron) preparation 14.7%loss and 85.3% gain were observed. This Loha Bhasma was used for the preparation of TL. During TL preparation, 0.6% loss was observed & 99.3% was obtained. This study will give the direction for the standard manufacturing process of Loha Bhasma (incinerated ash of iron) and Tryushanadya Lauha.

A Review on Probable Mode of Action of Gandhaka Rasayana - An Ayurvedic Herbo-Mineral Formulation with Multifaceted Action

As per ancient Ayurvedic scholars for getting a desirable outcome of any medication, it should be precisely analysed before prescribing to the patient. There are numbers of classical formulations successfully practiced by Ayurvedic physicians for treating various ailments but in maximum cases there is lack of data regarding their details mechanism of action. Such one multidrug classical formulation is Gandhaka rasayana which is widely practiced Ayurvedic formulation. Gandhaka rasayana have been used in Ayurvedic Rasashastra to treat various types of diseases related to Tvak, Mandhagni, various Srotas. It is a time tested formulation used by Ayurvedic practitioners for correcting Kushta, Indralupta, Vatarakta, Palitya, Dushta vrana, Arumsika etc. Modern scientific world needs explanation and logic behind the mode of action of Gandhaka rasayana with respective to each particular disease. This review is an attempt to explain multiple mode of action of Gandhaka rasayana in various diseases on analysing properties of ingredients. Analysis of mode of action gives deep insight about use of formulation with a better understanding.

Physicochemical characterization, Phytochemical analysis and Anti-microbial activity of Elathy mathirai

Elathy Mathirai is a Siddha herbomineral formulation indicated to cure sori(itching), sirangu(Scabies), suram(fever), vaayu, maandham(gastro intestinal disorder) . The aim of the study is to standardize Elathy Mathirai to ensure its quality, purity and safety through its physicochemical, chromatographic, phytochemical, heavy metal, pesticide residue and aflatoxins analysis and the secondary objective is to evaluate its antimicrobial property. This study determines the physicochemical paramaters that are loss on drying, total ash, acid insoluble ash, water soluble extractive, alcohol soluble extractive and pH as 5.667% , 5%, 0.014 %, 14.93 %, 8.3% and 5.6 respectively denotes purity and quality of the drug. From the study results it is ensured that the test drug Elathy mathirai is free from pesticide toxicity and aflatoxins. Qualitative preliminary phytochemical analysis shows the presence of alkaloids, Flavanoids,steroids, triterpenoids, saponins, coumarins, phenol, tannin, sugar and betacyanin. HPTLC finger printing analysis reveals the presence of seven prominent peaks Rf value ranges from 0.04 to 0.39 corresponds to the presence of seven components present within it. It was observed from the results of In vitro anti microbial assay that the formulation EM reveals significant level of activity against bacteria Klebsiella pneumonia and observed moderate activity against Staphylococcal aureus, Pseudomonas aeruginosa and posess consistent anti fungal activity against Candida albicans and there is no significant effect on Aspergillus niger. The obtained results shows that the EM complies with the regulatory standard and also possess significant anti microbial property against the tested microbes.

Ayurvedic Management of Pancreatitis

Pancreatitis is the life threatening metabolic pathology of the gastrointestinal system. It progressively damages the pancreas and result in the loss of its functions. The prevalence rate of pancreatitis in India is 7.9 per 10, 0000. For male and female respectively 8.6 and 8 in India. With overall 10 year and 20 year survival rates are estimated to be about 70% to 45% respectively. In the contemporary science the treatment protocol is limited to painkillers, intravenous fluids and pancreatic enzymes. Despite of advance techniques in the allopathic science the prognosis of the disease is unclear. Ayurvedic formulations of mineral complex including a diet and moderate rest have shown excellent result. In this case report a 40 year old male patient a known case of chronic pancreatitis with recurrent abdominal pain which radiates to back, nausea, vomiting, tenderness, anorexia, along with elevated serum amylase level was managed effectively with Ayurvedic medications by the Department of PG Study in Shalyatantra OPD Dr. BRKR Government Ayurvedic Medical College and Hospital, Hyderabad. In Ayurveda this condition can be correlated Agnashayashoth which comes under Pittodara. And it was successfully treated with the medications like Shankha vati, Kravyadi ras, Arogyavardgani vati. Within 4 months of treatment patient become normal. There was no pain abdomen, nausea, vomiting, bloating of abdomen. And serum amylase came to its normal level. This shows that Ayurvedic herbo mineral formulations showing promising result in the management of pancreatitis.

Analytical Evaluation of Abhraka Bhasma Samples after Marana, Amritikarana and Lohitikarana

Rasaushadhies or herbo-mineral formulations undergo rigorous processes to convert into nano size. The particle size does matters a lot in terms of absorption, assimilation and proper excretion from the body. The pharmaceutical procedures should be strictly adhered so as to ensure safety and efficacy. Abhraka bhasma is a popularly used formulation in Ayurvedic practice. Abhraka or black mica is a bad conductor of heat. So the process of Bhasmikarana requires high quantum of heat in order to render into Bhasma form. Amritikarana is a specific process included in the preparation of Abhraka bhasma. The current study evaluates the essentiality of this process in Bhasmikarana. A detailed analytical study is been carried out to find the differences in the samples before and after Amritikarana. The present study reveals that the particle size has reduced much and the iron content was at maximum concentration in the samples after the process of Amritikarana and Lohitikarana.
