Synthesis and characterization of multinuclear ruthenium clusters assembled by terminal alkyne alcohols and ortho-carborane diselenolate ligands

Two novel half-sandwich trinuclear ruthenium complexes bridged by ortho-carborane-1,2-diselenolato ligands (p-cymene)Ru3(Se2C2B10H10)2(Se2C2B10H9)[HCCH(cyclo-C6H10)OCCH(cyclo-C6H10)] (2) and (p-cymene)Ru3(Se2C2B10H10)2(Se2C2B10H9)[HCCH(CH3)2COCCHC(CH3)2] (3) have been prepared by the reactions of ortho-carborane-1,2-diselenolate ligands and [(p-cymene)RuCl2]2 with CHC(cyclo-C6H10)(OH) and CHCC(OH)(CH3)2, respectively. The complexes have been fully characterized by 1H, 13C, and 11B NMR, mass and IR spectroscopy as well as by element analyzes. The molecular structures of two complexes have been determined by single-crystal X-ray diffraction analyzes.

Targeting Multiresistant Gram-Positive Bacteria by Ruthenium, Osmium, Iridium and Rhodium Half-Sandwich Type Complexes With Bidentate Monosaccharide Ligands.

Bacterial resistance to antibiotics is an ever-growing problem in heathcare. We have previously identified a set of osmium(II), ruthenium(II), iridium(III) and rhodium(III) half-sandwich type complexes with bidentate monosaccharide ligands possessing cytostatic properties against carcinoma, lymphoma and sarcoma cells with low micromolar or submicromolar IC50 values. Importantly, these complexes were not active on primary, non-transformed cells. These complexes have now been assessed as to their antimicrobial properties and found to be potent inhibitors of the growth of reference strains of Staphylococcus aureus and Enterococcus faecalis (Gram-positive species), though the compounds proved inactive on reference strains of Pseudomonas aerugonisa, Escherichia coli, Candida albicans, Candida auris and Acinetobacter baumannii (Gram-negative species and fungi). Furthermore, clinical isolates of Staphylococcus aureus and Enterococcus sp. (both multiresistant and susceptible strains) were also susceptible to the organometallic complexes in this study with similar MIC values as the reference strains. Taken together, we identified a set of osmium(II), ruthenium(II), iridium(III) and rhodium(III) half-sandwich type antineoplastic organometallic complexes which also have antimicrobial activity among Gram-positive bacteria. These compounds represent a novel class of antimicrobial agents that are not detoxified by multiresistant bacteria suggesting a potential to be used to combat multiresistant infections.

Enantiopure Cyclometalated Rh(III) and Ir(III) Complexes Displaying Rigid Configuration at Metal Center: Design, Structures, Chiroptical Properties and Role of the Iodide Ligand

Enantiopure N-heterocyclic carbene half-sandwich metal complexes of the general formula [Cp*M(C^C:)I] (M = Rh, Ir; C^C: = NI-NHC; NI-H = Naphthalimide; NHC = N-heterocyclic carbene) are reported. The rhodium compound was obtained as a single isomer displaying six membered metallacycle and was resolved on chiral column chromatography to the corresponding enantiomers (S)-[Cp*Rh(C^C:)I] (S)-2 and (R)-[Cp*Rh(C^C:)I] (R)-2. The iridium congener, however, furnishes a pair of regioisomers, which were resolved into (S)-[Cp*Ir(C^C:)I] (S)-3 and (R)-[Cp*Ir(C^C:)I] (R)-3 and (S)-[Cp*Ir(C^C:)I] (S)-4 and (R)-[Cp*Ir(C^C:)I] (R)-4. These regioisomers differ from each other, only by the size of the metallacycle; five-membered for 3 and six-membered for 4. The molecular structures of (S)-2 and (S)-4 are reported. Moreover, the chiroptical properties of these compounds are presented and discussed. These compounds display exceptional stable configurations at the metal center in solution with enantiomerization barrier ΔG≠ up to 124 kJ/mol. This is because the nature of the naphthalimide-NHC clamp ligand and the iodide ligand contribute to their configuration’s robustness. In contrast to related complexes reported in the literature, which are often labile in solution.

Theoretical study of photovoltaic performances of Ru, Rh and Ir half sandwich complexes containing N,N chelating ligands in Dye-Sensitized Solar Cells (DSSCs). DFT and TD-DFT investigation

Owing to their low cost and improved energy efficiency, the dye-sensitized solar cells (DSSC) share significant priority in the toolbox of solar cell researchers. Herein, nine mononuclear Ru, Rh and Ir half sandwich complexes used in dye sensitized solar cells containing the phenyl hydrazone functionality and N,N chelating sites have been studied. The structural, electronic and optical properties, namely the FMO energy gap (Eg), chemical hardness (η), softness (S), electronegativity (χ), Natural bond orbital (NBO) charge distribution, open circuit voltage (Voc), molecular electrostatic potential (MEP), light harvesting efficiency (LHE) and driving force of regeneration (ΔGreg), the transition energy and the wavelengths (λ), were calculated at DFT/B3LYP/SDD/6-31+G(d,p) level of theory. A comparative study of the wavelength λ computed at B3LYP/SDD/6-31+G(d,p) level with those at CAM-B3LYP/SDD/6-31+G(d,p) and WB97XD/SDD/6-31+G(d,p) level of theory in gas phase and acetonitrile has also been performed. The predicted results were in conformity with the experiments.

Synthesis and Reactivity of Cyclic Oxonium Derivatives of nido-Carborane: A Review

Nucleophilic ring-opening reactions of cyclic oxonium derivatives of anionic boron hydrides are a convenient method of their modification which opens practically unlimited prospects for their incorporation into various macro- and biomolecules. This contribution provides an overview of the synthesis and reactivity of cyclic oxonium derivatives of nido-carborane as well as half-sandwich complexes based on it.

A Solid Support-Based Synthetic Strategy for the Site-Selective Functionalization of Peptides with Organometallic Half-Sandwich Moieties.

The number of donor atoms available on peptides that can competitively coordinate to metal centers renders the site-selective generation of advanced metal-peptide conjugates in high purity a challenging venture. Herein, we present a transmetalation-based synthetic approach on solid support in which an imidazolium pro-ligand can be used to selectively anchor a range of transition metal half-sandwich complexes onto peptides in the presence of multiple coordinative motifs. Amenable to solid support, a range of N-terminus and/or lysine conjugated metal-peptide conjugates were obtained in high purity after cleavage from the resin. The metalated peptides were evaluated for their anticancer properties against human cancer cell lines. While no cytotoxic activity was observed, this platform has the potential to i) provide a pathway to site-selective peptide labelling, ii) be explored as a biorthogonal handle and/or iii) generate a new strategy for ligand design in transition metal catalysts.

Visible-Light-Driven, Iridium-Catalyzed Hydrogen Atom Transfer: Mechanistic Studies, Identification of Intermediates, and Catalyst Improvements.

The harvesting of visible light is a powerful strategy for the synthesis of weak chemical bonds involving hydrogen that are below the thermodynamic threshold for spontaneous H2 evolution. Piano-stool iridium hydride complexes are effective for the blue-light-driven hydrogenation of organic substrates and contra-thermodynamic dearomative isomerization. In this work, a combination of spectroscopic measurements, isotopic labeling, structure–reactivity relationships, and computational studies has been used to explore the mechanism of these stoichiometric and catalytic reactions. Photophysical measurements on the iridium hydride catalysts demonstrated the generation of long-lived excited states with principally metal-to-ligand charge transfer (MLCT) character. Transient absorption spectroscopic studies with a representative substrate, anthracene revealed a diffusion-controlled dynamic quenching of the MLCT state. The triplet state of anthracene was detected immediately after the quenching events, suggesting that triplet–triplet energy transfer initiated the photocatalytic process. The key role of triplet anthracene on the post-energy transfer step was further demonstrated by employing photocatalytic hydrogenation with a triplet photosensitizer and a HAT agent, hydroquinone. DFT calculations support a concerted hydrogen atom transfer mechanism in lieu of stepwise electron/proton or proton/electron transfer pathways. Kinetic monitoring of the deactivation channel established an inverse kinetic isotope effect, supporting reversible C(sp2)–H reductive coupling followed by rate-limiting ligand dissociation. Mechanistic insights enabled design of a piano-stool iridium hydride catalyst with a rationally modified supporting ligand that exhibited improved photostability under blue light irradiation. The complex also provided improved catalytic performance toward photoinduced hydrogenation with H2 and contra-thermodynamic isomerization.

N-Heterocyclic Carbene-Phosphinidenide Complexes as Hydroboration Catalysts.

The reactions of the N-heterocyclic carbene-phosphinidene adducts (NHC)PSiMe3 and (NHC)PH with the dinuclear ruthenium and osmium complexes [(η6-p-cymene)MCl2]2 (M = Ru, Os) afforded the half-sandwich complexes [(η6-p-cymene){(NHC)P}MCl] and [(η6-p-cymene){(NHC)PH}MCl2] with two- and three-legged piano-stool geometries, respectively (NHC = IDipp, IMes; IDipp = 1,3-bis(2,6-diisopropylphenyl)imidazolin-2-ylidene; IMes = 1,3-bis(2,4,6-trimethylphenyl)imidazolin-2-ylidene). The complexes were initially tested as precatalysts for the hydroboration of benzonitrile, and the most active species, the ruthenium complex [(η6-p-cymene){(IMes)P}RuCl], was further used for the efficient hydroboration of a wide range (ca. 50 substrates) of nitriles, carboxylic esters, and carboxamides in neat pinacolborane (HBpin) under comparatively mild reaction conditions (60-80 °C, 3-5 mol % catalyst loading). Preliminary mechanistic and kinetic studies are reported, and stoichiometric reactions with HBpin indicate the initial formation of the monohydride complex [(η6-p-cymene){(IMes)P}RuH] as the putative catalytically active species.

DNA binding, antitubercular, antibacterial and anticancer studies of newly designed piano-stool ruthenium(II) complexes.

The chemotherapeutic potential of ruthenium(II) complexes has recently attracted researchers' interest as antibacterial and anticancer agents. In this study, two novel half-sandwich imine-based Ru complexes ([Ru(p-cymene)Cl(L-1)][PF6] (Ru-1) and [Ru(p-cymene)Cl(L-2)][PF6] (Ru-2)) were reported for their deoxyribonucleic acid (DNA) binding and antitubercular, antibacterial, and anticancer activities. The molecular structure of Ru-2 was obtained by single-crystal X-ray crystallography. DNA interaction studies were conducted by UV-Vis absorbance and fluorescence spectral titration which gave rise to DNA binding constants (Kb) of 1.32 × 106 and 1.82 × 106 for Ru-1 and Ru-2, respectively and the Stern-Volmer binding constant (KSV) values for Ru-1 and Ru-2 were 1.7763 × 104 M-1 and 7.6 × 103 M-1, respectively. The in vitro antitubercular activity was evaluated against Mycobacterium tuberculosis H37Ra. The antibacterial potential of both the Ru-complexes was examined against Gram-negative (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive (Staphylococcus aureus and Bacillus subtilis) bacteria. The half-maximal inhibitory concentration (IC50) values for the antitubercular activity of Ru-1 and Ru-2 were 4.87 ± 1.32 μM and 5.78 ± 0.54 μM, respectively. A cytotoxic study of these complexes was performed against the human breast cancer cell line (MCF-7) and the human embryonic kidney cell line (HEK293) (normal cells). The study revealed meaningful activity of the Ru-1 complex against (cancer) MCF-7 cells, while the viability of HEK293 (normal) cells in the presence of Ru-2 was higher as compared to a reference drug 5FU. We suggest that these kinds of Ru-complexes could have potential for application in metallopharmaceuticals.

N-Alkylation of organonitrogen compounds catalyzed by methylene-linked bis-NHC half-sandwich ruthenium complexes.

An efficient ruthenium-catalyzed N-alkylation of amines, amides and sulfonamides has been developed employing novel pentamethylcyclopentadienylruthenium(II) complexes bearing the methylene linked bis(NHC) ligand bis(3-methylimidazol-2-ylidene)methane. The acetonitrile complex 2 has proven particularly effective with a broad range of substrates with low catalyst loading (0.1-2.5 mol%) and high functional group tolerance under mild conditions. A total of 52 N-alkylated organonitrogen compounds including biologically relevant scaffolds were synthesized from (hetero)aromatic and aliphatic amines, amides and sulfonamides using alcohols or diols as alkylating agents in up to 99% isolated yield, even on gram-scale reactions. In the case of sulfonamides, it is the first example of N-alkylation employing a transition-metal complex bearing NHC ligands.

Synthesis, physicochemical characterization and antiproliferative activity of phosphino Ru(II) and Ir(III) complexes.

Herein, we present the synthesis of new complexes based on ruthenium(II) (Ru(η6-p-cymene)Cl2PPh2CH2OH (RuPOH) and Ru(η6-p-cymene)Cl2P(p-OCH3Ph)2CH2OH (RuMPOH)) and iridium(III) (Ir(η5-Cp*)Cl2P(p-OCH3Ph)2CH2OH (IrMPOH) and Ir(η5-Cp*)Cl2PPh2CH2OH (IrPOH)) containing phosphine ligands with/without methoxy motifs on phenyl rings (P(p-OCH3Ph)2CH2OH (MPOH) and PPh2CH2OH (POH)). The complexes were characterized by mass spectrometry, NMR spectroscopy (1D: 1H, 13C{1H}, and 31P{1H} and 2D: HMQC, HMBC, and COSY NMR) and elemental analysis. All the complexes were structurally identified by single-crystal X-ray diffraction analysis. The Ru(II) and Ir(III) complexes have a typical piano-stool geometry with an η6-coordinated arene (RuII complexes) or η5-coordinated (IrIII compounds) and three additional sites of ligation occupied by two chloride ligands and the phosphine ligand. Oxidation of NADH to NAD+ with high efficiency was catalyzed by complexes containing P(p-OCH3Ph)2CH2OH (IrMPOH and RuMPOH). The catalytic property might have important future applications in biological and medical fields like production of reactive oxygen species (ROS). Furthermore, the redox activity of the complexes was confirmed by cyclic voltamperometry. Biochemical assays demonstrated the ability of Ir(III) and Ru(II) complexes to induce significant cytotoxicity in various cancer cell lines. Furthermore, we found that RuPOH and RuMPOH selectively inhibit the proliferation of skin cancer cells (WM266-4; IC50, after 24 h: av. 48.3 μM; after 72 h: av. 10.2 μM) while Ir(III) complexes were found to be moderate against prostate cancer cells (DU145).

Rare-earth-metal half-sandwich complexes incorporating methyl, methylidene, and hydrido ligands.

Complexes Cp*3Ln3(μ2-CH3)3(μ3-CH2)(μ3-H)(thf)3 (Ln = Y, Dy; Cp* = C5Me5) are formed via elimination of CH4 and C2H4 from the respective trinuclear polymethyl complexes [Cp*Ln(μ2-CH3)2]3, involving the methyl/methylidene complex Cp*3Ln3(μ2-CH3)3(μ3-CH2)(μ3-CH3)(thf)2 as an intermediate. Compound Cp*3Y3(μ2-CH3)3(μ3-CH2)(μ3-H)(thf)3 displays "Schrock-type" nucleophilic carbene reactivity, converting ketones into the corresponding terminal alkenes. Treatment of Cp*3Y3(μ2-CH3)3(μ3-CH2)(μ3-H)(thf)3 with [D4]methanol proved accessibility of all small-group functionalities as indicated by the simultaneous formation of deuterated HD and deuterated methanes CH3D and CH2D2.

The synthesis and initial observations regarding the reactivity of borole-based half-sandwich complexes with apical divalent group 14 elements germanium and tin are described. The 2,5-disilylborole substitution pattern allows their access via salt metathesis of the respective borole dianions.

Stability of a half-sandwich Os(II) complex with indomethacin-functionalized ligand in the presence of carboxypeptidase A.

In the presence of carboxypeptidase, the hydrolytically stable complex [Os(η6-pcym)(L2)Cl]PF6 (2) partially released the bioactive substituent indomethacin, bound through the amide bond to the chelating 2-(1,3,4-thiadiazol-2-yl)pyridine-based moiety of L2. Stability in the presence of other relevant biomolecules (GSH, NADH, GMP) and cancer cell viability were also studied.

A new divalent organoeuropium(II) fluoride and serendipitous discovery of an alkoxide complex from pentaphenylcyclopentadiene precursors

From the redox-transmetallation protolysis (RTP) reaction of europium metal, Hg(C6F5)2 and pentaphenylcyclopentadiene, we isolated and crystallographically characterised small amounts of the first divalent europium fluoride half-sandwich complex [Eu(C5Ph5)(μ-F)(thf)2]2 (1). Subsequently, a rational synthesis of this complex from in situ formed [EuF2(thf)n] and [Eu(C5Ph5)2] was carried out. In addition, the new divalent Eu alkoxide complex [Eu(OC5Ph5*)2(thf)4] (2) (OC5Ph5* = 2,3,4,5,5 pentaphenylcyclopenta-1,3-dienolate) was identified by X-ray diffraction analysis, in which an intriguing phenyl group migration in the cyclopentadiene ligand occurred. This complex was shown to be derived from small impurities of 1,2,3,4,5-pentaphenylcyclopenta-1,3-dienol (C5Ph5OH) in the C5Ph5H starting material and was then synthesised on a larger scale. Density functional theory calculations provided evidence for the facile phenyl group migration observed in the cyclopentadienolate ring.

PM-IRRAS and DFT investigation of the surface orientation of new Ir piano-stool complexes attached to Au(111).

Surface immobilization of organometallic catalysts is a promising approach to developing new catalytic systems that combine molecular catalysts with heterogenous surfaces to probe surface mechanisms. The orientation of the catalyst relative to the surface is one important parameter that must be considered in such hybrid systems. In this work, we synthesize three new sulfide-modified Ir piano-stool complexes with sulfide-modified bipyridine and phenylpyridine ligands for the attachment to Au(111) surfaces. Self-assembled monolayers made from (Cp*Ir(2,2'-bipyridine-4-sulfide)Cl)2[Cl]2 (C1m) and [Cp*Ir(2-phenylpyridine-4-sulfide)Cl]2 (C2m) were characterized by combining polarization modulation infrared reflection absorption spectroscopy (PM-IRRAS) with DFT calculations of the minimum energy orientations of the complexes on the surface. We find that the bipyridine and phenylpyridine ligands are oriented at between 73-77° relative to the surface normal, irrespective of the orientation of the other ligands. Additionally, DFT and PM-IRRAS support that there is no orientation preference for C1m and C2m, with both orientations present on the surface.

Rhodium and Iridium Complexes of Anionic Thione and Selone Ligands Derived from Anionic N-Heterocyclic Carbenes.

The lithium salts of anionic N‐heterocyclic thiones and selones [{(WCA‐IDipp)E}Li(toluene)] (1: E=S; 2: E=Se; WCA=B(C6F5)3, IDipp=1,3‐bis(2,6‐diisopropylphenyl)imidazolin‐2‐ylidene), which contain a weakly coordinating anionic (WCA) borate moiety in the imidazole backbone were reacted with Me3SiCl, to furnish the silylated adducts (WCA‐IDipp)ESiMe3 (3: E=S; 4: E=Se). The reaction of the latter with [(η 5‐C5Me5)MCl2]2 (M=Rh, Ir) afforded the rhodium(III) and iridium(III) half‐sandwich complexes [{(WCA‐IDipp)E}MCl(η 5‐C5Me5)] (5–8). The direct reaction of the lithium salts 1 and 2 with a half or a full equivalent of [M(COD)Cl]2 (M=Rh, Ir) afforded the monometallic complexes [{(WCA‐IDipp)E}M(COD)] (9–12) or the bimetallic complexes [μ 2‐{(WCA‐IDipp)E}M2(COD)2(μ 2‐Cl)] (13–16), respectively. The bonding situation in these complexes has been investigated by means of density functional theory (DFT) calculations, revealing thiolate or selenolate ligand character with negligible metal‐chalcogen π‐interaction.

Theoretical investigation of mechanism and ligand effects on half-sandwich iridium complexes for direct reductive amination

The reaction mechanisms of direct reductive amination catalyzed by half-sandwich Cp*Ir complexes and the possible side reactions — transfer hydrogenation of ketones and N-formylation of amines have been explored by density functional theory (DFT). The reaction involves two steps: (1) the hydridoiridium formation as a rate-determining step, and (2) the hydride transfer to obtain amines. The relationships between the structure and activity of Cp*Ir complexes were analyzed by natural population analysis (NPA), Bader's atoms in molecules theory (AIM) and frontier molecular orbital (FMO). In general, the picolinamidato ligands are more favorable than the sulfonamidato ones due to the electronic effect. The catalyst with the phenyl ring bearing functional groups on the N-amide exerts the positive effect on the catalytic activity. Furthermore, the electron-donating group on the pyridine ring provides better performance owing to the higher NPA charge on the Ir atom, the lower electron density at the Ir-Cl bond critical point, and the destabilization of the highest occupied molecular orbital (HOMO).
