Rhizosphere bacteria from the Bolivian highlands improve drought tolerance in quinoa (Chenopodium quinoa Willd.).

Drought is one of the most destructive abiotic factors for agricultural production, causing considerable yield losses. Quinoa (Chenopodium quinoa Willd.) is cultivated worldwide in different environmental conditions due to its nutritional characteristics and ability to grow in harsh environments. This study aims to select drought stress tolerant rhizosphere bacteria from the Bolivian altiplano to evaluate their quinoa growth-promoting capacity, including in vitro germination, seedling growth under drought stress in greenhouse conditions and field studies. Rhizosphere soil from the southern highlands of Bolivia was collected to isolate 164 drought-stress tolerant bacteria. From these, 28 strains were shown to produce indole acetic acid, and/or to possess nitrogen-fixing or phosphate solubilizing capacity under in vitro conditions. Furthermore, all strains were evaluated for improvement of in vitro quinoa seed germination. Based on these properties, nine bacterial strains were formulated in three different matrixes and evaluated for quinoa seedling growth promotion during drought stress in a 3-month greenhouse experiment. Three strains were shown to significantly (P<0.05) increase root length of the quinoa seedlings. One strain was selected and shown to significantly (P<0.05) increase leaf number in a field trial under semi-arid conditions in the southern altiplano in Bolivia. DNA sequencing and phylogenetic analyses of the 16S locus putatively identified the three strains with growth-promoting potential under drought stress as members of the genera Bacillus, Pseudomonas, and Serratia. Microorganisms from the arid Bolivian altiplano constitute a potential biological source of bioinoculants to improve quinoa productivity and provide sustainable mitigation of climate change effects.

NATIVE MYCORRHIZAE FROM ETHIOPIA IMPROVE TREE GROWTH AND SEEDLINGS SURVIVAL CONTRIBUTING TO THE GREEN LEGACY PROGRAM

&lt;p&gt;&lt;strong&gt;Background&lt;/strong&gt;: A rapid production of tree seedlings in nurseries with a high survival rate after transplanting is important to respond to the current demand for programs of restoration of arid environments by forestation. The low level of seedlings’ survival and establishment, caused by low moisture and nutrient content of soils, has been a bottleneck to reaching the target of the forest national restoration in Ethiopia of last years. It is suggested that, inoculation with root-associated plant growth promoting microorganisms could help to ameliorate this scenario and also respond to the Ethiopia's Green Legacy Program. &lt;strong&gt;Objective&lt;/strong&gt;: To assess the potential of inoculation with arbuscular mycorrhizal fungi (AMF) native to Ethiopia to improve the survival and growth of trees that could be used in afforestation programs in Ethiopia. &lt;strong&gt;Methodology&lt;/strong&gt;: The study was carried out in three stages: (1) soil samples associated with roots of selected acacia species (T&lt;sub&gt;1&lt;/sub&gt;-AMF of&lt;em&gt; A. abyssinica&lt;/em&gt;, T&lt;sub&gt;2&lt;/sub&gt;-AMF of&lt;em&gt; A. seyal&lt;/em&gt;, T&lt;sub&gt;3&lt;/sub&gt;-AMF&lt;em&gt; &lt;/em&gt;of &lt;em&gt;A. tortilis &lt;/em&gt;and&lt;em&gt; &lt;/em&gt;T&lt;sub&gt;4&lt;/sub&gt;-Control) were collected of highland and lowland areas from Ethiopia, (2) Sorghum (&lt;a href="https://www.researchgate.net/publication/342429051_Evaluation_of_sorghum_Sorghum_bicolor_L_Moench_variety_performance_in_the_lowlands_area_of_wag_lasta_north_eastern_Ethiopia"&gt;&lt;em&gt;Sorghum bicolor&lt;/em&gt; (L.), provided by the Melkasa Agricultural Research Center-(MARC) served as a trap plant for &lt;/a&gt;the AMF consortium multiplication and (3) plant growth promotion by AMF was assessed throughout inoculations of seedlings of&lt;em&gt; &lt;/em&gt;&lt;em&gt;Delonix regia&lt;/em&gt; (Hook.) Raf.,&lt;em&gt; Sesbania grandiflora &lt;/em&gt;(L.)&lt;em&gt;, Cassia fistula &lt;/em&gt;L&lt;em&gt;., &lt;/em&gt;and&lt;em&gt; Azadirachta indica &lt;/em&gt;A. Juss., trees&lt;em&gt;. &lt;/em&gt;&lt;strong&gt;Results&lt;/strong&gt;: All inoculated seedlings showed significantly greater responses in all growth and mycorrhizal parameters over the non-inoculated trees. Consortium T&lt;sub&gt;2&lt;/sub&gt;-AMF associated to roots of&lt;em&gt; A. seyal &lt;/em&gt;from lowlands of Batu, showed significantly greater responses in all plant growth and mycorrhizal parameters over the AMF inoculums associated to other tree species evaluated. Significant and positive correlations were found between mycorrhizae and plant-growth parameters&lt;em&gt;. &lt;/em&gt;&lt;strong&gt;Implications:&lt;/strong&gt; Our results suggest that inoculation with native arbuscular mycorrhizal fungi indigenous from Ethiopia has the potential to significantly enhance survival and growth rates of tree seedlings. This could thereby advance national reforestation goals and addressing challenges in seedling establishment in arid environments. &lt;strong&gt;Conclusion&lt;/strong&gt;: The potential for growth promotion and establishment of tree seedlings evidenced, implies that further efforts should be directed towards the in-mass production of AMF-based inoculants, particularly associated with &lt;em&gt;A. seyal&lt;/em&gt; roots.&lt;/p&gt;

Comparative Metagenomic Profiling of Seed-Borne Microbiomes in a Landrace and a Hybrid Maize Variety

The plant seed-borne microbiome comprises microorganisms vertically inherited from the mother plant. This microbiome is often linked to early-life protection and seedling growth promotion. Herein, we compare the seed-borne bacteriomes of a commercial hybrid (Santa Helena) and a landrace maize variety (Sol da Manhã). The landrace variety displays a more diverse seed-borne microbiome, featuring a variety of taxa across samples with an average Shannon’s diversity index of 1.12 compared to 0.45 in the hybrid variety. The landrace variety also showed a greater alpha diversity of 165.8, in contrast to 144.1 in the hybrid. Although both microbiomes lack a functional nitrogen fixation apparatus, we found a remarkably distinct presence of genes associated with phytohormone production and phosphate solubilization, particularly in the landrace variety. In addition, we recovered 18 metagenome-assembled genomes (MAGs), including four from potentially novel species. Collectively, our results allow for a better understanding of the contrasting diversity between maize varieties. The higher potential for phytohormone production in landraces, the absence of nif genes in both varieties, and the identification of core microbiome taxa offer valuable insights into how microbial communities impact plant health and development. This knowledge could pave the way for more sustainable and innovative agricultural practices in crop management.

Effect of silver nanochitosan on control of seed-borne pathogens and maintaining seed quality of wheat

Seeds, considered as the foundation of agriculture, are invaded by a broad spectrum of seed-borne pathogens. The current study aimed to control seed-borne fungal pathogens of wheat, Aspergillus flavus and A. niger, by using Ag+ nanochitosan (Ag-NC) for nano-priming of seeds and enhancing seed quality. Nanochitosan (NC) and Ag-NC were synthesized using the gelation method and characterized by UV–vis spectrophotometry, FESEM, EDXS, and HRTEM. NC and Ag-NC showed irregular surface topography with an average particle size of 275 and 325 nm, respectively. Antifungal activity of both the nanoparticles at 0.1, 0.2, 0.3, 0.4, and 0.5 mg/mL revealed that Ag-NC at 0.5 mg/mL has completely terminated the mycelial growth of both pathogens. Malonaldehyde content increased to 77.77% in A. flavus and 82.66% in A. niger when exposed to 0.5 mg/mL Ag-NC. High-intensity fluorescence due to oxidative stress was observed in Ag-NC-treated pathogens. Ultra-structural changes in Ag-NC treated pathogenic spores under SEM displayed pronounced membrane damages. Wheat seeds were nano-primed with NC and Ag-NC at 0.5 mg/mL, and fungal load was examined to evaluate the mitigation of pathogenic stress and its effect on seedling growth promotion activity. Ag-NC priming reduced the fungal load and allowed successful seed germination. Ag-NC priming increased the albumin, gliadin, gluten, and glutenin content along with total phenol, reducing sugar and starch levels. Ag-NC priming increased the overall protein levels traced through SDS-PAGE. Seed priming with Ag-NC promotes seed germination, mean germination time, stress tolerance index, vigour, etc. NC and Ag-NC at 0.5 mg/mL showed no cytotoxic effect on the Human Embryonic Kidney (HEK293) cell line that ensures the nanoparticles are non-toxic. Thus, the synthesized nanoparticles exhibit a dual role in antifungal activity and plant growth promotion.

Influence of phenotypic variation of Paenibacillus polymyxa E681 on growth promotion in cucumbers.

The goal of the current study is to better understand how bacteria may adapt to survive under adverse environmental conditions by altering and improving their phenotypes. In this study, we report the consequences of phenotypic variation in Paenibacillus polymyxa E681 (E681), a plant growth-promoting rhizobacterium (PGPR), isolated from winter barley root that has a variety of advantageous effects on crop plants. In our previous study, two different types of bacterial cells in E681 were distinguished. We used the term F-type for the variant that doesn't produce endospores and B-type for the endospore-producing wild type. Under the circumstances of our experiment, the cucumber rhizosphere soil and the surface of the seeds produced phenotypic variance. On tryptic soy agar (TSA) plates, the B-type spontaneously converted into the F-type, but the reverse was not reversible. Intriguingly, the plant growth promotion test displayed that cucumber seedlings treated with F-type cells had characteristics resembling those of the untreated control. Whereas, growth promotion of cucumber seedlings treated with B-type depends on temperature conditions. In particular, an increased growth promotion was observed at a low temperature of 20°C. The phenotypic change from B-type to F-type did not occur at 20°C for 6 days in the growth curve analysis of E681, but it did occur on the fourth and second days at 30 and 37°C, respectively. Therefore, before using PGPR strains as a bacterial inoculant for sustainable agriculture, it is imperative to resolve phenotypic variance in these strains.

Influence of peat-derived humic acid on the growth of agarwood seedlings

Humic acid (HA) is widely used in agriculture as a biostimulant to enhance plant growth and nutrient uptake. Studies on the early growth promotion of agarwood seedlings using plant biostimulants have not been explored. Therefore, the objective of this study was to extract and characterize peat-derived HA and to determine the optimum rate that would exert biostimulant action on the early growth of agarwood seedlings at the nursery stage. HA was extracted from peat soil using a modified rapid alkaline hydrolysis method. Produced peat humic acid (PHA) was compared with commercial humic acid (CHA) for its chemical and spectroscopic characteristics. One-month-old agarwood seedlings were treated with PHA at different rates (0, 20, 40, 80, 160 and 320 mg kg−1) before the commencement of the experiment. The study was conducted for four months. The C/N ratio of both PHA and CHA were 25.85 and 26.93, respectively. Scanning electron microscope (SEM) and fourier-transform infrared spectroscopy (FTIR) spectra observation reveals that PHA and CHA have similar surface morphology and functional groups respectively. Results revealed that agarwood seedlings treated with 80 mg kg−1 of PHA significantly increased the plant height, stem diameter, the number of leaves, leaf area, dry matter weight, N and P concentrations in plant tissues, and N, P and K uptakes. Based on the findings, incorporation of PHA at the rate of 80 mg kg−1 with planting medium could be used as an optimum rate to enhance the growth of agarwood seedlings in the nursery stage.

Biological control of Aspergillus flavus infection and growth promotion of peanut seedlings by Lactiplantibacillus plantarum and Levilactobacillus brevis

BackgroundAspergillus spp. infection might induce negative effects on peanut seeds, including decreased germination rates and suppressed seedling vigor. Furthermore, A. flavus can secret aflatoxin, regarding food safety and human health. The prolonged use of fungicides for treating mold infections has raised concerns regarding the emergence of fungicide-resistant strains, environmental pollution, and adverse effects on human health. The usage of lactic acid bacteria, including Lactiplantibacillus plantarum and Levilactobacillus brevis for the management of plant diseases, has garnered increasing attention in recent years as a viable alternative to chemical-based therapies. This study aimed to investigate the efficacy of LABs in pre-treating peanut seeds as a biological solution against A. flavus infection before cultivation.ResultsLactiplantibacillus plantarum and Levilactobacillus brevis have demonstrated the ability to suppress A. flavus in vitro. In the in vivo investigation, pre-treatment of peanut seeds with cell-free supernatant derived from L. plantarum (LP-CFS) and L. brevis (LB-CFS) significantly reduced A. flavus infection levels. The conidial count decreased from 8.63 log conidia/g in the untreated group to 5.35 log conidia/g with LP-CFS treatment and 4.59 log conidia/g with LB-CFS treatment. Additionally, A. flavus infection reduced the germination rate of peanut seeds to only 20.4% compared to 63.6% in the control group. In comparison, pre-treatment with LP-CFS and LB-CFS increased the germination rate to 75.6% and 76.8%, respectively, and further improved the vigor index in A. flavus-infected peanut seeds.ConclusionThe present findings indicated that bioactive compounds derived from L. plantarum and L. brevis emerge as promising candidates for treating peanut seeds, effectively protecting them against A. flavus infection. Moreover, these compounds facilitate the growth of seedlings, which could be a potential alternative to chemical fungicides, and contribute to sustainable agricultural development.

Sustainable seedling pots: Development and characterisation of banana waste and natural fibre-reinforced composites for horticultural applications

Plastic pots used in horticultural nurseries generate substantial waste, causing environmental pollution. This study aimed to develop biodegradable composites from banana pseudo-stem reinforced with agricultural residues like pineapple leaves, taro and water hyacinth as eco-friendly substitutes. The aim of this study is to develop optimised banana biocomposite formulations with suitable reinforcements that balance mechanical durability, biodegradation, and seedling growth promotion properties to serve as viable eco-friendly alternatives to plastic seedling pots. This study was carried out by fabricating banana fibre mats through pulping, drying and hot pressing. Composite sheets were reinforced with 50% pineapple, taro or water hyacinth fibres. The mechanical properties (tensile, yield strength, elongation, bursting strength), hydrophilicity (contact angle, water absorption), biodegradability (soil burial test), and seedling growth promotion were evaluated through appropriate testing methods. The results show that banana-taro composites exhibited suitable tensile strength (25MPa), elongation (27%), water uptake (41%) and 82% biodegradation in 60days. It was observed that biodegradable seedling trays fabricated from banana-taro composite showed 95% tomato seed germination and a 125cm plant height increase in 30days, superior to plastic trays. The finding shows that the study demonstrates the potential of banana-taro biocomposites as alternatives to plastic nursery pots, enabling healthy seedling growth while eliminating plastic waste pollution through biodegradation.

Comparative Effect of Seed Coating and Biopriming of Bacillus aryabhattai Z-48 on Seedling Growth, Growth Promotion, and Suppression of Fusarium Wilt Disease of Tomato Plants.

Beneficial plant microbes can enhance the growth and quality of field crops. However, the benefits of microbes using cheap and efficient inoculation methods are still uncommon. Seed coating with biocontrol agents can reduce the amount of inocula along with having the potential for large-scale application. Hence, in this research work, the comparative potential of tomato seed coating and biopriming with Bacillus aryabhattai Z-48, harboring multiple plant-beneficial traits, to suppress Fusarium wilt disease along with its beneficial effect on seedling and plant growth promotion was analyzed. Among two bacterial strains, B. aryabhattai Z-48 was able to antagonize the mycelial growth of Fusarium oxysporum f.sp. lycopersici in vitro and its application as a seed coating superiorly benefited seedling traits like the germination percentage, vigor index, and seedling growth index along with a reduced germination time. The seed coating with B. aryabhattai Z-48 resulted in significant increases in the shoot length, root length, dry biomass, and total chlorophyll contents when compared with the bioprimed seeds with the same bacterial strain and non-inoculated control plants. The seed coating with B. aryabhattai Z-48 significantly reduced the disease index (>60%) compared with the pathogen control during pot trials. Additionally, the seed coating with B. aryabhattai Z-48 resulted in a significantly higher production of total phenolics, peroxidase, polyphenol oxidase, and phenylalanine ammonia lyase enzyme in tomato plants. The GC/MS-based non-targeted metabolic profiling indicated that the seed coating with B. aryabhattai Z-48 could cause large-scale metabolite perturbations in sugars, sugar alcohols, amino acids, and organic acids to increase the fitness of tomato plants against biotic stress. Our study indicates that a tomato seed coating with B. aryabhattai Z-48 can improve tomato growth and suppress Fusarium wilt disease effectively under conventional agricultural systems.

Antifungal and Plant-Growth Promotion Effects of Bacillus velezensis When Applied to Coastal to Pine (Pinus thunbergii Parl.) Seedlings

Fungal diseases such as root rot and leaf blight cause substantial losses in coastal pine (Pinus thunbergii Parl.) seedling production, which hinders afforestation/forest restoration programs. We isolated and identified Fusarium oxysporum and Alternaria alternata as the causal agents of root rot and needle blight diseases and investigated the biocontrol efficacy against the fungal pathogens and growth promotion of coastal pine seedlings using Bacillus velezensis CE 100. The bacterium produced the hydrolytic enzymes chitinase, β-1,3-glucanase, and protease enzymes, and the crude enzyme fraction of the biocontrol strain caused the deformation of the fungal cell wall and antagonized F. oxysporum and A. alternata, causing respective inhibition of spore germination by 91.0% and 85.9% and mycelial growth by 58.3% and 54.3%, at a concentration of 1000 µL/mL. Consequently, the bacterial treatment improved the survival rate of seedlings 1.9 times relative to the control group. The bacterium secreted indole-acetic acid (IAA) phytohormone and enhanced root growth and absorption of nutrients, which notably enhanced the biomass production of coastal pine seedlings. Therefore, these results provide evidence that B. velezensis CE 100 is an effective antifungal and growth-promoting bacterium that can facilitate the production of high-quality coastal pine seedlings for the restoration and establishment of coastal forests.

Photosynthetically active radiation is required for seedling growth promotion by volcanic dacitic tuff breccia (Azomite).

A plant's growth and development are shaped by its genome and the capacity to negotiate its environment for access to light, water, and nutrients. There is a vital need to understand the interactions between the plant, its physical environment, and the fertilizers used in agriculture. In this study, a commercially available volcanic ash fertilizer, Azomite®, characterized as dacitic (rhyolitic) tuff breccia, was tested for its effect on promoting early seedling vigor. Early growth and photomorphogenesis processes are well studied in Arabidopsis. Seedling assays under different light conditions were used to dissect the underlying mechanisms involved. These assays are well established and can be translated to agriculturally important crop plants. The volcanic ash fertilizer was tested at different concentrations on seedlings grown on basic media lacking sucrose either in continuous darkness (Dc), continuous Red (Rc), Far-Red (FRc), or White Light (WLc). Micronutrients in the volcanic ash significantly increased seedling growth under Rc and WLc, but not under Dc and FRc, indicating that photosynthetically active radiation was required for the observed growth increase. Furthermore, red-light photoreceptor mutant, phyB-9, lacked the growth response, and higher amount of fertilizer reduced growth in all conditions tested. These data suggest that light triggers the ability of the seedling to utilize micronutrients in volcanic ash in a dose-dependent manner. The methods described here can be used to establish mechanisms of activity of various nutrient inputs and, coupled with whole-genome expression profiling, can lead to better insights into optimizing nutrient field applications to improve crop production.

The Small Auxin-Up RNA SAUR10 Is Involved in the Promotion of Seedling Growth in Rice.

Small auxin-up-regulated RNAs (SAURs) are genes rapidly activated in response to auxin hormones, significantly affecting plant growth and development. However, there is limited information available about the specific functions of SAURs in rice due to the presence of extensive redundant genes. In this study, we found that OsSAUR10 contains a conserved downstream element in its 3' untranslated region that causes its transcripts to be unstable, ultimately leading to the immediate degradation of the mRNA in rice. In our investigation, we discovered that OsSAUR10 is located in the plasma membrane, and its expression is regulated in a tissue-specific, developmental, and hormone-dependent manner. Additionally, we created ossaur10 mutants using the CRISPR/Cas9 method, which resulted in various developmental defects such as dwarfism, narrow internodes, reduced tillers, and lower yield. Moreover, histological observation comparing wild-type and two ossaur10 mutants revealed that OsSAUR10 was responsible for cell elongation. However, overexpression of OsSAUR10 resulted in similar phenotypes to the wild-type. Our research also indicated that OsSAUR10 plays a role in regulating the expression of two groups of genes involved in auxin biosynthesis (OsYUCCAs) and auxin polar transport (OsPINs) in rice. Thus, our findings suggest that OsSAUR10 acts as a positive plant growth regulator by contributing to auxin biosynthesis and polar transport.

Effect of Steeping Duration and Inoculum Concentration on Growth Promotion of Seedlings of Food Crops Primed in Cultures of Bacillus spp from Rhizosphere

Aims: Seed priming represents a viable, low-cost approach to improving germination and plant growth. This study aimed to assess the effects of steeping duration and inoculum concentration of seed crops on germinability enhancement of seed primed in cultures of Bacillus species from rhizospheres. The seeds used were cowpea (Vigna unguiculata), soya bean (Glycine max), sorghum (Sorghum bicolor), sesame (Sesamum indicum), and okra (Abelmoschus esculentus). Methods: A total of five Bacillus species (four species of B. cereus and one species of B. thuringiensis) were used for priming the seeds. For the effect of steeping duration on germinability, viable surface-sterilized seeds were primed in growth broth cultures of the respective isolates. Every one hour, for a five-hour duration, a known number of seeds were withdrawn from the cultures and sown. In the case of initial inoculum, seeds were steeped in different dilutions of the bacterial cultures at the optimal steeping duration obtained in the first study before planting. At the expiration of planting duration, final germination percentage, germination time, germination index, and seedling vigor index of the respective seeds were estimated. Results: The results highlight the importance of steeping duration for seeds such as cowpea and soybean, and the effect of inoculum concentration was less drastic than that of steeping duration. Conlcusion: Further field studies need to be carried out to validate these results, using results here as baseline data.

Bacillus Strains with Catalase Enzyme Improve the Physiology and Growth of Rice (Oryza sativa L.)

Catalase can decompose hydrogen peroxide in plants under controlled and stressed conditions. In a stressed environment, an increase in hydrogen peroxide production makes catalase activity a major defense system for plant growth. The current study was conducted to evaluate the catalase activity of the pre-isolated and identified bacterial strains Bacillus aryabhattai (AN30), Bacillus megaterium (AN24), Bacillus megaterium (AN31) and Bacillus megaterium (AN35) and their potential for rice seedling growth promotion. These strains were characterized for quantitative catalase, urease, siderophore and exopolysaccharide production using LB media. Subsequently, the effectiveness of these strains was checked by quantifying the catalase activity in the rhizosphere, roots and shoots of rice seedlings. The secretion of organic and phenolic compounds produced by the tested strains in liquid culture was also investigated. Plant growth parameters were also studied in a growth room trial. Our results showed that the strain AN24 showed the maximum catalase activity (1.36 mol cm−1), urease activity (1.35 mol cm−1) and exopolysaccharide (4.20 µg mL−1) and siderophore (2.32%) production in LB media. All tested strains showed significantly higher catalase activity in soil compared to the control. Among sole applications, strain AN24 showed better results; however, the consortium application of strains AN24 + AN30 + AN35 + AN31 showed the maximum improvement in dry biomass, shoot and root length, and increase in catalase activity of rice seedlings. The results showed that a consortium of these Bacillus strains with catalase activity has greater potential to enhance the antioxidant defense system and growth promotion of rice seedlings. However, further experimentation under natural conditions is required before using these strains as potential bioinoculants for improving rice growth and yield.

Assessing the functional diversity of rhizobacteria from cacao by partitioning root and shoot biomasses.

Plant-microbe interactions are critical for the sustainability of agricultural production. In this study, our aims were to characterize the genetic and functional diversity of the culturable bacterial community associated with the cacao rhizosphere and access their potential for growth promotion of cacao seedling. Culture-dependent and molecular methods were used to characterize the population densities and diversity of bacterial communities from soil and cacao plants at two locations and two plant ages. A total of 63 strains were identified through hsp60 sequencing. Pseudomonas and Enterobacter were the most abundant genera in association with the cacao rhizosphere, whereas Bacillus was more numerous in soil. Parameters of seedling growth promotion were evaluated 60days after inoculation of seeds, with partition of the assessments into root and shoot weight. Each isolate showed beneficial, neutral or deleterious effects on plant growth, depending on the isolate and on the parts of plant assessed. Interestingly, although an apparent overall decrease in total biomass of seedlings (roots + shoots dry matters) was observed for the majority of isolates (89%), 94% of all isolates, in fact, revealed an increase in plant roots/shoots dry biomass ratio. Despite that part of the isolates (35%) appeared to significantly decrease plant height, and that 65% did not influence plant height (neutral effect), 18 had significantly increased root dry biomass; nevertheless, seven of these root growth-increasing isolates simultaneously decreased shoots-related growth parameters. The results of this study evidentiated the functional diversity of culturable cacao rhizobacteria and how the partitioning of roots and shoots in the assessment of plant growth parameters could reveal the biotechnological potential of these isolates for promoting growth of clones for rehabilitation of commercial cacao plantations. New insight on tomato seed priming with Anabaena minutissima phycobiliproteins in relation to Rhizoctonia solani root rot resistance and seedling growth promotion

AbstractCyanobacteria phycobiliproteins (PBPs) are already exploited in the food industries and for biotechnological applications but not in the agricultural field. Different concentrations (0.6 – 4.8 mg/mL) of Anabaena minutissima PBPs were applied to tomato seed to study their priming effect against the soil-borne fungal pathogen Rhizoctonia solani and in promoting plant growth. PBPs increased seedling emergence and vigour, showed activity against root rot disease (67%), and enhanced plant dry weight, length, and height. Generally, no dose effect has been observed except for dry weight (55% at 4.8 mg/mL). Seed treatment primed seeds and seedlings by leading to the activation of defence responses raising phenol (26% in hypocotyls) and flavonoid (26 and 45% in hypocotyls and epicotyls, respectively) contents and chitinase (4-fold at 2.4 and 4.8 mg/mL in hypocotyls) and β-1,3-D-glucanase (up to about 2-fold at all doses in epicotyls) activities. Micro-Attenuated Total Reflection Fourier Transform Infrared revealed changes in functional groups of primed seeds, hypocotyls and exudates released into the agar because of treatment. Protein extract from PBP-primed seedlings inhibited mycelial growth (67% for epicotyl proteins) and caused morphological alterations in hyphae. This research emphasizes the potential priming role of PBPs applied by seed treatment against soil-borne pathogens.

Bacillus-Loaded Biochar as Soil Amendment for Improved Germination of Maize Seeds.

Biochar is considered one of the most promising long-term solutions for soil quality improvement, representing an ideal environment for microorganisms' immobilization. Hence there is a possibility to design microbial products formulated using biochar as a solid carrier. The present study was aimed at development and characterization of Bacillus-loaded biochar to be applied as a soil amendment. The producing microorganism Bacillus sp. BioSol021 was evaluated in terms of plant growth promotion traits, indicating significant potential for production of hydrolytic enzymes, indole acetic acid (IAA) and surfactin and positive tests for ammonia and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase production. Soybean biochar was characterised in terms of physicochemical properties to evaluate its suitability for agricultural applications. The experimental plan for Bacillus sp. BioSol021 immobilisation to biochar included variation of biochar concentration in cultivation broth and adhesion time, while the soil amendment effectiveness was evaluated during maize germination. The best results in terms of maize seed germination and seedling growth promotion were achieved by applying 5% of biochar during the 48 h immobilisation procedure. Germination percentage, root and shoot length and seed vigour index were significantly improved when using Bacillus-biochar soil amendment compared to separate treatments including biochar and Bacillus sp. BioSol021 cultivation broth. The results indicated the synergistic effect of producing microorganism and biochar on maize seed germination and seedling growth promotion, pointing out the promising potential of this proposed multi-beneficial solution for application in agricultural practices.

Isolation and Characterization of Plant Growth promoting Rhizobacteria from Bamboo Rhizosphere and Their Role in Plant Growth Promotion

Plant growth-promoting rhizobacteria (PGPR) are a group of root-associated bacteria that intimately interact with the plant roots and consequently enhance growth by extemporising nutrient retrieval or phytohormone production. We isolated and screened indigenous phosphate solubilising and auxinproducing PGPR from bamboo rhizosphere. 66 soil samples from bamboo (Bambusa nutans subsp. cupulata, B. balcooa and B. tulda) rhizospheres were collected from Dhanusha, Mahottari and Sarlahi districts, Nepal. 120 isolates of PGPR were obtained by serial dilution method in (PVK) agar and Luria Bertani agar. 92 out of 120 isolates of PGPR with the ability to solubilise phosphate were selected based on the halo colony ratio in PVK agar medium and auxin production in Luria Bertani agar. Among them, six isolates having high phosphate solubilising index and high production capacity of indole-3-acetic acid were further screened. Biochemical analysis revealed that these isolates belonged to the genus Pseudomonas. Phosphate solubilising index and indole-3- acetic acid production by six isolates ranged from 4.19±0.8 to 7.65±1.3, and IAA production ranged from 72.93±0.2 to 82.48±0.9µg/ml respectively. These isolates significantly increased shoot length (13.26±0.56cm), shoot fresh weight (16.26±1.02mg), shoot dry weight (10.56±0.09mg), root length (4.9±0.5cm), root fresh weight (7.56±1.05mg), root dry weight (3.21±0.01mg), and chlorophyll ‘a’ and chlrophyll‘b’ and carotenoid (2.16±0.01mg/g, 1.19±0.06mg/g and 0.92±0.01mg/g respectively) of B. nutans subsp. cupulata seedlings. This study suggests that PGPR isolated from bamboo rhizosphere demonstrated outstanding contribution to the growth promotion of seedlings of B. nutans subsp. cupulata as compared to negative control.

Orchid mycorrhiza: Isolation, culture, characterization and application

• This review states the isolation, culture, characterization and application of OMF. • Major bottlenecks of traditional identification of OMF and its elucidation are discussed. • States the application OMF for bio-hardening and growth promotion of seedlings. • Provides insights for mass cultivation and conservation of orchids. In nature all orchids develop a mutualistic association with specialized group of fungi called orchid mycorrhiza. In natural environment, orchids are exclusively reliant on mycorrhizal fungi for seed germination, establishment, growth and development. In the early developmental stages, all orchids are mycoheterotrophic, and in most cases the association subsists throughout their lifespan. The significance of orchid mycorrhizal fungi (OMF) for seed germination, nourishment, growth promotion, and survival has been established more than a century ago and tremendous advancements were made in recent decades. Till date, understanding the mode of interaction between orchids and mycorrhizal fungi and determining the architecture of interaction networks are major challenges in ecology and evolutionary biology. The development of molecular techniques has empowered us to unscramble the fungal diversity comprised in the symbiosis and has facilitated us to overcome the difficulties associated with the conventional identification of OMF and orchid-fungal interactions. In addition, the application of modern biotechnological approaches resolved many controversial issues of OMF. Recently, OMF are used for biological hardening and growth promotion of in vitro raised seedlings, early flowering and quality improvement of flowers, and diseases and pests management. The aim of this communication is to update and summarize orchid mycorrhizal research including isolation, culture, and characterization of OMF, understanding the mechanisms of orchid-fungus symbiosis, and application of OMF in seed germination, growth promotion, and survival of asymbiotically raised seedlings and conservation of orchids. This appraisal will convey valuable insights to orchidologists engaged in biology, propagation, and conservation of orchids.
