SBL-JP-0004: A promising dual inhibitor of JAK2 and PI3KCD against gastric cancer.

Gastric cancer (GC) remains a global health burden and is often characterized by heterogeneous molecular profiles and resistance to conventional therapies. The phosphoinositide 3-kinase and PI3K and Janus kinase (JAK) signal transducer and activator of transcription (JAK-STAT) pathways play pivotal roles in GC progression, making them attractive targets for therapeutic interventions. This study applied a computational and molecular dynamics simulation approach to identify and characterize SBL-JP-0004 as a potential dual inhibitor of JAK2 and PI3KCD kinases. KATOIII and SNU-5 GC cells were used for in vitro evaluation. SBL-JP-0004 exhibited a robust binding affinity for JAK2 and PI3KCD kinases, as evidenced by molecular docking scores and molecular dynamics simulations. Binding interactions and Gibbs binding free energy estimates confirmed stable and favorable interactions with target proteins. SBL-JP-0004 displayed an half-maximal inhibitory concentration (IC50) value of 118.9 nM against JAK2 kinase and 200.9 nM against PI3KCD enzymes. SBL-JP-0004 exhibited potent inhibition of cell proliferation in KATOIII and SNU-5 cells, with half-maximal growth inhibitory concentration (GI50) values of 250.8 and 516.3 nM, respectively. A significant elevation in the early phase apoptosis (28.53% in KATOIII cells and 26.85% in SNU-5 cells) and late phase apoptosis (17.37% in KATOIII cells and 10.05% in SNU-5 cells) were observed with SBL-JP-0004 treatment compared to 2.1% and 2.83% in their respective controls. The results highlight SBL-JP-0004 as a promising dual inhibitor targeting JAK2 and PI3KCD kinases for treating GC and warrant further preclinical and clinical investigations to validate its utility in clinical settings.

Divalent Organo‐Bimetallic Chelates of Poly‐Dentate O,N,O‐dihydrazone Ligand as Effective Agents for Antitumor and Antimicrobial Assays, Interacting Modes With DNA

ABSTRACTSubstituted hydrazones and dihydrazones, as polydenatate ligand, reported high coordination chemical behavior towards numerous transition metals of different oxidation states for alternated applicable interest. Therefore, with a facile condensed reaction of the salicylaldehyde with tartric dihydrazide, a chelated tartrato‐dihydrazone ligand (H2Ltz) was formed. The coordinated features of H2Ltz versus two divalent metal ions of Cu (II) and Ni (II) ions were assumed to form two new bimetallic‐organic framework complexes, assigning triagonal bipyramidal and tetrahedral geometries (CuLtz and NiLtz, respectively). Their structural elucidating was determined within the analytical tools spectroscopically. The effectiveness of the growth inhibition for H2Ltz, CuLtz, and NiLtz against six named bacterial and fungal series, and three human cancer cell lines were examined discovering the reactive role of two central M (II) ions in CuLtz and NiLtz, respectively. The binding character of H2Ltz, CuLtz, and NiLtz, with calf thymus DNA, that is, DNA, was estimated based on the variation in viscometric/spectrophotometric features. The two bimetallic‐chelates represented more distinguished inhibitive attitudes than the regarded one of H2Ltz assigning to the inhibitive zones (mm) and the half‐inhibited concentration (IC50, μM) of growth inhibition of the studied microbes and human tumor cells, respectively. The examination of the binding action of H2Ltz, CuLtz, and CuLtz with DNA was approved according to the enhanced viscosity and the electronic spectral changes. Also, binding constants (14.09, 15.09, and 16.12 107 mol−1 dm3), Gibb's free energy (−42.06, −45.23, and −46.41 kJ mol−1), and the chromism (mainly hypo modes) are assigned to estimate the binding modes. Rewardingly, the bimetallic‐chelates MLtz displayed modified binding action more than that of H2Ltz against DNA regarding their hydrophobicity/lipophilicity.

Nanocochleate-based delivery of quercetin with enhanced therapeutic potential: formulation, cytotoxicity and pharmacokinetics study

BackgroundQuercetin has well-proven anticancer potential through various mechanisms, but its applications in drug delivery are limited due to poor biopharmaceutical properties. The present study was aimed to formulate quercetin nanocochleates (QNC) in order to overcome these limitations. QNC formulation was fabricated by a trapping method, optimized and evaluated for various formulation aspects, in vitro cytotoxicity and pharmacokinetic parameters.ResultDeveloped nanocochleates possess particle size and encapsulation efficiency of 205.6 ± 2.55 nm and 76.36 ± 0.88%, respectively. In vitro cytotoxicity study performed using MCF-7 cell lines revealed the comparative efficiency of QNC over pure quercetin. Total growth inhibition concentration (TGI) for pure quercetin was 96.73 μg/ml, while for QNC it was 83.29 μg/ml. Pharmacokinetic study results showed improvement in Cmax and AUC after QNC formulation with increased Tmax showing sustained release.ConclusionOverall, the developed QNC formulation markedly improved cytotoxic potential and biopharmaceutical aspects.

Control efficacy of Bt-Cry1Ab maize (event DBN9936) against Ostrinia furnacalis (Guenée) in Sichuan Province, China.

The Asian corn borer (ACB), Ostrinia furnacalis (Guenée), is a major pest restricting maize production in Asia. The Chinese government has approved the commercial planting of Bt-Cry1Ab maize (event DBN9936), but its control potential against the ACB in southern regions remains unclear. This study evaluated the sensitivity of ACB to Cry1Ab protein expressed in Bt-Cry1Ab maize and determined the control efficacy of Bt-Cry1Ab maize against the ACB in Sichuan Province, a major maize-producing region in southern China, based on pilot planting in the field, and larval feeding bioassays in the field and laboratory. The Cry1Ab protein contents in different tissues of Bt-Cry1Ab maize ranged from 31.20-88.27 μg g-1. The range of median lethal concentrations (LC50) and median growth inhibitory concentration (GIC50) values of Cry1Ab protein expressed in Bt-Cry1Ab maize against ACB larvae were 0.036-0.109 μg mL-1 and 0.002-0.018 μg mL-1, respectively. The first and fourth instar ACB larvae were unable survive feeding on different tissues of Bt-Cry1Ab maize plants. Field experiments conducted from 2023 to 2024 indicated that the number of ACB larvae per 100 plants, plant damage rate, leaf damage rate, male ear damage rate, female ear damage rate, and stalk damage rate in the Bt-Cry1Ab maize fields were reduced by 95.36% ± 2.17%, 83.98% ± 1.73%, 89.45% ± 1.50%, 100.00% ± 0.00%, 69.79% ± 4.88% and 100.00% ± 0.00%, respectively, compared to conventional maize fields. The ACB population in Sichuan Province, China is sensitive to Cry1Ab expressed in Bt-Cry1Ab maize (event DBN993). Planting Bt-Cry1Ab maize efficiently reduces the population of ACB larvae and the percentage of damaged maize plants, and has great application potential in the integrated pest management of the ACB in Sichuan Province, China. © 2024 Society of Chemical Industry.

Antifungal effect of Crescentia cujete fruit extracts on clinical strains of Candida albicans

Candidiasis is a common fungal infection globally, ranging from superficial to severe invasive forms that can be life-threatening. This study aimed to evaluate the antifungal effects of ethanol and n-hexane extracts from Crescentia cujete fruit against Candida albicans strains. Crescentia cujete fruit pulp was dried and extracted using Soxhlet with ethanol and n-hexane. Two reference strains of C. albicans (ATCC 10231 and 90028) and 12 clinical isolates from vaginal smears were tested under various conditions: without extract, with serial dilutions of ethanol or n-hexane extracts, and with ethanol and n-hexane at the same concentrations. Growth was measured using MicroELISA equipment. Results showed that n-hexane and ethanol extracts significantly reduced the growth of 9/12 and 4/12 clinical strains, respectively, and also affected the ATCC 90028 and ATCC 10231 strains. A direct correlation was observed between growth inhibition and extract concentration, with n-hexane exhibiting a stronger correlation (Spearman's Rho=0.609, p<0.05) compared to ethanol (Spearman's Rho=0.387, p<0.05). This study is the first to demonstrate the antifungal effects of Crescentia cujete fruit extracts on clinical Candida albicans strains, suggesting that a low-polarity compound in the fruit inhibits Candida albicans, including the ATCC 1031 strain resistant to common antifungals.

Unraveling the complexities of antifungal susceptibility testing in Candida spp.: Insights from design of experiments

Our study delved into the intricate dynamics of antifungal susceptibility testing for Candida spp., employing a Design of Experiments approach. We systematically investigated the influence of pH, temperature, inoculum size, and glucose concentration on both growth patterns and inhibitory concentrations of Candida spp. Our findings underscore the nuanced interplay between these factors, revealing significant impacts on susceptibility outcomes. Notably, even minor adjustments in these parameters yielded substantial variations in growth and inhibitory concentrations, underscoring the critical importance of meticulous control over growth conditions in antifungal susceptibility testing protocols. Each Candida isolates exhibited unique susceptibility profiles, necessitating tailored culture conditions for accurate testing. Our study sheds light on the variability inherent in Candida spp. growth patterns and emphasizes the need for standardized protocols to ensure consistency across laboratories. By leveraging the design of experiments, our research provides a systematic framework for unraveling the complexities of antifungal susceptibility testing, offering valuable insights for optimizing testing protocols and informing clinical decision-making in antifungal treatment. These findings represent a significant step towards enhancing the efficacy and reliability of antifungal susceptibility testing in clinical practice.

Secondary Metabolites of Halobacillus sp.: Antimicrobial and Antioxidant Activity, Biological Compatibility, and Gas Chromatography-Mass Spectrometry (GC-MS) Analysis.

Background The rise of infectious diseases and the emergence of resistant pathogens pose significant challenges to human health. In response to this global threat, researchers are exploring novel sources of bioactive compounds for effective antimicrobial therapies. One avenue of investigation is the study of halophilic bacteria and their secondary metabolites. These bacteria thrive under extreme conditions and produce valuable bioactive metabolites, which have the potential for therapeutic applications. Methods In this study, the potent bacterial cultures obtained from the Payanur salt pan, Tamil Nadu, were analyzed for the antimicrobial activity of their metabolites. The secondary metabolites were obtained from the halophilic bacteria by culturing the bacteria in 8% NaCl. The resultant secondary metabolites produced were extracted using ethyl acetate and their antimicrobial property was studied using the well diffusion method. The minimum inhibitory concentration (MIC) of these metabolites against five clinical pathogens, namely, Staphylococcus aureus, Escherichia coli, Enterococcus faecalis, Pseudomonas aeruginosa, and Candida albicans was determined. Their antioxidant property was studied using the DPPH(2,2-diphenyl-1-picrylhydrazyl) method and biological compatibility was determined by hemolytic assay of the secondary metabolites. Results The potent halophilic bacteria isolated from salt pan bacteria were phenotypically and genotypically identified as Halobacillus sp.The secondary metabolites extracted from these bacteria yielded 110 mg of crude metabolites. The antimicrobial activity of crude metabolites shows a moderate zone of inhibition of 14 mm for P. aeruginosa, 13 mm for E. coli and C. albicans, and 11 mm for S. aureus. The minimum inhibitory concentration was 128 µg/mL for E. coli, P. aeruginosa, and C. albicans,which was found to bethe best growth inhibition concentration. The DPPH scavenging activity shows a higher activity till the concentration of 64 µg/mL.The hemolytic activity of 25% is obtained at 128 µg/mLand below 64µg/mL, there is no hemolytic activity. The gas chromatography-mass spectrometry (GC-MS) analysis of the secondary metabolites shows the presence of 17 compounds. Among them, there were fourmajor compounds: (i) cyclo(L-prolyl-L-valine) (probability of 95.63%), (ii) pyrrolo[1,2-a]pyrazine- 1,4-dione,hexahydro-3-(2-methylpropl) (probability of 94.45%), (iii) 2,5-piperazinedione,3,6-bis(2-methylpropyl)(probability of 71.94%) and (iv) pyrrolo[1,2-a]pyrazine-1,4-dione,hexahydro-3-(phenylmethyl)(probability of 88.01%). Conclusion In conclusion, the isolated bacterium is confirmed to be Halobacillus sp.and the secondary metabolites produced by this bacterium could be the potential source for the development of novel antimicrobial and antioxidant compounds that are highly biologically compatible. Further research mayhelpto developnovel compounds in the pharmaceutical industry.

Resistant risk and resistance mechanism of florylpicoxamid in Colletotrichum gloeosporioides isolated from Chinese walnut

Colletotrichum gloeosporioides is the causal pathogen for the devastating walnuts anthracnose. A novel quinone inside inhibitor (QiI) fungicide florylpicoxamid has strong inhibitory efficacy against C. gloeosporioides. This study looked into the resistance risk and mechanism of C. gloeosporioides to florylpicoxamid. The basal level sensitivity of C. gloeosporioides isolates (n = 102) to florylpicoxamid was established with an average 50% mycelial growth inhibition concentration (EC50) value of 0.069 ± 0.035 μg/mL. Six stable florylpicoxamid-resistant mutants with resistance factors of >1000 were produced. The fitness of every mutant was much lower than that of their parental isolates. In general, the resistance risk of C. gloeosporioides to florylpicoxamid would be moderate. Molecular docking results revealed that the amino acid substitutions A37V, and S207L in CgCytb lead to a reduction in the binding affinity between florylpicoxamid and CgCytb, indicating that these two mutations (S207L and A37V in CgCytb) indeed confer florylpicoxamid resistance in C. gloeosporioides. These findings offer a fresh viewpoint on the mechanism underlying QiI fungicide resistance and could support the prudent application of florylpicoxamid in the future to combat walnut anthracnose.

Dual-targeting class I HDAC inhibitor and ATM activator, SP-1-303, preferentially inhibits estrogen receptor positive breast cancer cell growth.

Dual-targeting chromatin regulation and DNA damage repair signaling presents a promising avenue for cancer therapy. Applying rational drug design, we synthesized a potent dual-targeting small molecule, SP-1-303. Here, we report SP-1-303 as a class I isoform selective histone deacetylase (HDAC) inhibitor and an activator of the ataxia-telangiectasia mutated protein (ATM). In vitro enzymatic assays demonstrated selective inhibition of HDAC1 and HDAC3. Cellular growth inhibition studies show that SP-1-303 differentially inhibits growth of estrogen receptor positive breast cancer (ER+ BC) cells with effective growth inhibition concentrations (EC50) for MCF-7 and T47D cells ranging from 0.32 to 0.34 μM, compared to 1.2-2.5 μM for triple negative breast cancer cells, and ~12 μM for normal breast epithelial cells. Western analysis reveals that SP-1-303 decreases estrogen receptor alpha (ER-α) expression and increases p53 protein expression, while inducing the phosphorylation of ATM and its substrates, BRCA1 and p53, in a time-dependent manner in ER+ BC cells. Pharmacokinetic evaluation demonstrates an area under the curve (AUC) of 5227.55 ng/ml × h with an elimination half-life of 1.26 h following intravenous administration in a rat model. Collectively, SP-1-303 emerges as a novel second generation class I (HDAC1 and HDAC3) selective HDAC inhibitor, and ATM activator, capable of modulating ER expression, and inhibiting growth of ER+ BC cells. Combined targeting of class I HDACs and ATM by SP-1-303 offers a promising therapeutic approach for treating ER+ breast cancers and supports further preclinical evaluation.

In vitro Development of Two Alternaria solani Strains, Causal Agent of Alternariose in Tomato (Lycopersicon esculentum) under the Influence of Thevetia peruviana Seeds Extracts

Alternaria solani is a fungus that causes yield losses of up to 80 % in tomato production in field. Synthetic fungicides are the most widely used for its controbl, but have harmful consequences. The objective of this work was to test in vitro the antifungal potential of Thevetia peruviana seed extracts against two A. solani isolates. Aqueous, methanol, ethyl acetate and acetone extracts, at concentrations 12.5, 25, 50 and 100 µL/mL were used. Two synthetic fungicides Maneb (5.33 µg/mL) and Dimethomorph + Clorothalonil (3.75 µg/mL) and control (0 µL/mL) were also tested on two A. solani isolates (Mbal and Foum). The investigation was repeated three times. Phytochemical screening, mycelial growth, spore germination and minimum inhibitory concentrations (MIC50 and MIC90) were determined. The results showed that T. peruviana extracts are rich in many families of bioactive compounds such as alkaloids, phenolic compounds and sugars. All extracts tested show high inhibition of mycelial growth (100%) and spore germination (100%) of the two strains at highest concentration (100 µL/mL). Acetone extract at a concentration of 50 µL/mL inhibited mycelial growth by 88.45 and 86.55% and spore germination by 88.33 and 80.33%, respectively for the Mbal and Foum isolates. The lowest MIC50 (16.63 µL/mL) and MIC90 (54.6 µL/mL) were obtained with the acetone extract on the Mbal isolate while the highest MIC50 (27.5 µL/mL) and MIC90 (61.7 µL/mL) were observed with ethyl acetate on the Foum isolate These extracts can therefore be used in the biological control against Alternariose in tomato.

Rosa × damascena Herrm. From Azaran region, Kashan: rich in saturated and unsaturated fatty acids with inhibitory effect against Proteus mirabilis

BackgroundOne of the most widely used medicinal plants in Iranian traditional medicine, Rosa × damascena Herrm. (mohammadi flower) that the people of Kashan use as a sedative and to treat nervous diseases and constipation. In this research, the yield, chemical composition and antimicrobial activity of the essential oil of this plant were evaluated for the first time from Azaran region, Kashan.MethodsThe essential oil was extracted by means of hydrodistillation (Clevenger), and its chemical compounds were identified and determined by GC/MS. The antimicrobial activity of the essential oil was determined by the diffusion method in agar, the minimum growth inhibitory concentration (MIC) and the minimum concentration capable of killing bacterial/fungal microorganisms (MBC/MFC).ResultsThe results showed that the yield of essential oil was 0.1586 ± 0.0331% (w/w). Based on the results of the chemical composition analysis of R. x damascena essential oil, 19 different compounds (98.96%) were identified. The dominant and main components of the essential oil were oleic acid (48.08%), palmitic acid (15.44%), stearic acid (10.17%), citronellol (7.37%) and nonadecane (3.70%). Based on the results of diffusion in agar, the highest zone of inhibition against Candida albicans (ATCC 10231) was ~ 9.5 mm. The strongest inhibitory activity of R. x damascena essential oil against Gram-negative Proteus mirabilis (ATCC 43071) was with the diameter of the inhibition zone (~ 9 mm), which was equal to the strength of rifampin (~ 9 mm).ConclusionTherefore, this essential oil is a promising natural option rich in fatty acids, which can be a potential for the production of natural antimicrobials against infectious diseases, especially urinary tract infections.

Integrative omics analysis reveals insights into small colony variants of Staphylococcus aureus induced by sulfamethoxazole-trimethoprim

BackgroundLong-term treatment with trimethoprim-sulfamethoxazole (SXT) can lead to the formation of small-colony variants (SCVs) of Staphylococcus aureus. However, the mechanism behind SCVs formation remains poorly understood. In this study, we explored the phenotype and omics-based characterization of S. aureus SCVs induced by SXT and shed light on the potential causes of SCV formation.MethodsStable SCVs were obtained by continuously treating S. aureus isolates using 12/238 µg/ml of SXT, characterized by growth kinetics, antibiotic susceptibility testing, and auxotrophism test. Subsequently, a pair of representative strains (SCV and its parental strain) were selected for genomic, transcriptomic and metabolomic analysis.ResultsThree stable S. aureus SCVs were successfully screened and proven to be homologous to their corresponding parental strains. Phenotypic tests showed that all SCVs were non-classical mechanisms associated with impaired utilization of menadione, heme and thymine, and exhibited slower growth and higher antibiotic minimum inhibitory concentrations (MICs), compared to their corresponding parental strains. Genomic data revealed 15 missense mutations in 13 genes in the representative SCV, which were involved in adhesion, intramolecular phosphate transfer on ribose, transport pathways, and phage-encoded proteins. The combination analysis of transcriptome and metabolome identified 35 overlapping pathways possible associated with the phenotype switching of S. aureus. These pathways mainly included changes in metabolism, such as purine metabolism, pyruvate metabolism, amino acid metabolism, and ABC transporters, which could play a crucial role in promoting SCVs development by affecting nucleic acid synthesis and energy metabolism in bacteria.ConclusionThis study provides profound insights into the causes of S. aureus SCV formation induced by SXT. The findings may offer valuable clues for developing new strategies to combat S. aureus SCV infections.

Anlotinib inhibits growth of human esophageal cancer TE-1 cells by negative regulating PI3K/Akt signaling pathway

Anlotinib is effective in treatment of many kinds of malignant cancer, but its antineoplastic effects on esophageal cancer remains unclear. This study aims to investigate its impact on esophageal cancer and the underlying mechanisms. Anlotiniband 5-fluorouracil + cisplatin (5-FU + DDP) was administered separately to human esophageal cancer TE- 1 cells tumor xenograft mouse models every 3 days. Tumor size and body weight were measured before each treatment and at the end of the experiment. In vitro studies were conducted using TE- 1 cells to examine the effects of Anlotinib. Cell viability, migration, proliferation, apoptosis, cell cycle, their regulatory proteins and the transcriptomic changes were analyzed. Anlotinib reduced tumor size, tumor weight, and the ratio of tumor weight to body weight in vivo. It decreased the viability of TE- 1 cells, with a 50% growth-inhibitory concentration of 9.454 μM for 24 h, induced apoptosis, and arrested TE- 1 cell cycle in the S phase. It inhibited migration and proliferation while negatively regulating the PI3K/Akt signaling pathway. Enhanced expressions of P21, Bax, and lowered expressions of cyclin A1, cyclin B1, CDK1, PI3K, Akt, p-Akt, and Bcl-2 were observed after Anlotinib treatment. Anlotinib exhibits antineoplastic activity against human esophageal cancer TE- 1 cells by negatively regulating the PI3K/Akt signaling pathway, consequently altering the expressions of proteins related to proliferation, apoptosis, and the cell cycle.

Modulation of bacterial membranes and cellular macromolecules by dimethyl sulfoxide: A dose-dependent study providing novel insights

Using Escherichia coli as a model, this manuscript delves into the intricate interactions between dimethyl sulfoxide (DMSO) and membranes, cellular macromolecules, and the effects on various aspects of bacterial physiology. Given DMSO's wide-ranging use as a solvent in microbiology, we investigate the impacts of both non-growth inhibitory (1.0 % and 2.5 % v/v) and slightly growth-inhibitory (5.0 % v/v) concentrations of DMSO. The results demonstrate that DMSO causes alterations in bacterial membrane potential, influences the electrochemical characteristics of the cell surface, and exerts substantial effects on the composition and structure of cellular biomolecules. Genome-wide gene expression data from DMSO-treated E. coli was used to further investigate and bolster the results. The findings of this study provide valuable insights into the complex relationship between DMSO and biological systems, with potential implications in drug delivery and cellular manipulation. However, it is essential to exercise caution when utilizing DMSO to enhance the solubility and delivery of bioactive compounds, as even at low concentrations, DMSO exerts non-inert effects on cellular macromolecules and processes.

Abstract CT167: Trial in progress: Phase 1 dose escalation study of T-1101, a first-in-class oral Hec1/Nek2 inhibitor, in patients with advanced refractory solid tumors

Abstract Background: T-1101, a novel small molecule, is the first-in-class oral agent that specifically disrupts the interaction between two commonly overexpressed, critical mitotic regulators, Hec1 and Nek2, within cancer cells. Such disruption leads to abnormal mitosis followed by apoptosis of cancer cells and provides a potential therapeutic strategy for cancer treatment. T-1101, with growth inhibition concentrations (IC50) ranging from14 to 74 nM across various human cancer cell lines, exhibited potent anti-liver and breastcancer activities in vivo, and highly active (IC50 7-19 nM) in MDR (multiple drugresistance) expressing cell lines. The effective inhibitory dose of T-1101 was determined to be between 10 and 25 mg/kg twice a day in xenograft animal models. Moreover, T-1101 has synergistic activity when combined with other anticancer drugs such as doxorubicin, topotecan and paclitaxel etc. Clinically, oral powder for constitution (OPC) form was first used in the initial study. To improve better patient compliance, the capsule (CAP) form is then deployed in current T-1101’s phase 1 study. Methods: Two T-1101 oral formulations (OPC and CAP) have been designed for the first-in-human, multi-center, open-label, dose-escalation studies to evaluate the safety, tolerability and establish the Recommended Phase 2 Dose (RP2D) in subjects with advanced refractory solid tumors. The secondary objectives are to assess the pharmacokinetics (PK) and therapeutic response after receiving treatment. DLT was determined within the first treatment cycle. Patients showing clinical benefit after first 2cycles of T-1101may enter extension cohort to continue the treatment. In OPC 3+3 escalation study (NCT03195764, NCT03349073), T-1101 was administered orally once daily, days 1 to 14 every 21 days per cycle with 8 planned dose levels at dose of 9, 18, 36, 54, 72, 90, 108 to 126 mg/m2. The OPC study was terminated at 36 mg/m2 before the primary endpoint (determination of maximum tolerated dose; MTD) was reached and subsequent dosing level will be evaluated with the CAP formulation. Based on the 52% of relative oral bioavailability (%F) of CAP versus OPC in preclinical Beagle dogs’ PK study, the starting dose of CAP form of T-1101 was determined at 100 mg daily forconsecutive 28-day per cycle for 2 cycles with dose levels of 100, 200, 225, 250, 300 and 350 mg/day. The accelerated titration and Bayesian Optimal Interval (BOIN) designs are used for the T-1101 capsule dose escalation study (NCT04685473), in which one subject will be enrolled per dose level until one subject have DLT or two subjects experience ≥ Grade 2 drug-related AE during Cycle 1 at any dose level, then the dose escalation will follow the BOIN design. Cohorts 1 (100 mg/day) 2 (200 mg/day) and 3 (225 mg/day) have been completed without DLT. As of November 2023, the trial remains open to enrollment. Next-generation microbiological testing in intraabdominal infections with PCR technology

PurposeIntraabdominal infections (IAI) are increasing worldwide and are a major contributor to morbidity and mortality. Among IAI, the number of multi-drug resistant organisms (MDRO) is increasing globally. We tested the Unyvero A50® for intraabdominal infections, compared the detected microorganisms and antibiotic resistance, and compared the results with those of routine microbiology.MethodsWe prospectively compared samples obtained from surgical patients using PCR-based Unyvero IAI cartridges against routine microbiology for the detection of microorganisms. Additionally, we identified clinical parameters that correlated with the microbiological findings. Data were analyzed using the t-test and Mann–Whitney U test.ResultsSixty-two samples were analyzed. The PCR system identified more microorganisms, mostly Bacteroides species, Escherichia coli, and Enterococcus spp. For bacterial resistance, the PCR system results were fully concordant with those of routine microbiology, resulting in a sensitivity, specificity, and positive and negative predictive value (PPV, NPV) of 100%. The sensitivity, specificity, PPV, and NPV for the detection of microorganisms were 74%, 58%, 60%, and 72%, respectively. CRP levels were significantly higher in patients with detectable microorganisms. We identified more microorganisms and bacterial resistance in hospital-acquired intra-abdominal infections by using the PCR system.DiscussionIAI warrants early identification of the microorganisms involved and their resistance to allow for adequate antibiotic therapy. PCR systems enable physicians to rapidly adjust their antibiotic treatment. Conventional microbiological culture and testing remain essential for determining the minimal growth inhibition concentrations for antibiotic therapy.

Abstract 3256: Novel SOX2-targeting bioPROTACs for the treatment of cancers

Abstract Targeted protein degradation (TPD) technology has been developed showing the therapeutic potential to modulate historically ‘undruggable’ disease-associated proteins. Currently, proteolysis targeting chimeras (PROTACs) using small-molecule binders represent the TPD field, and PROTAC-induced proximity between target proteins and E3 ligases enables specific degradation of target proteins. However, substantial amounts of clinically relevant cancer targets such as transcription factors lack active sites or allosteric binding pockets for small molecules which challenges the development of classical PROTACs for these targets. SOX2 is one of the pluripotency transcription factors and has been well-known for its crucial role in tumor development through promoting cancer cell survival, metastasis, stemness, and drug resistance. Numerous studies have also reported SOX2 amplification and overexpression in various tumor tissues. Thus, SOX2 has been validated as an attractive anticancer target, but directly targeting SOX2 has been proved to be difficult since SOX2 is an “undruggable” transcription factor. EPD Biotherapeutics develops EPDegTM, a next-generation engineered protein degrader platform in order to overcome the limitations of small molecule-based PROTAC. Here we report a series of SOX2-targeting bioPROTACs, being composed of a nanobody against SOX2 with high affinity and several ubiquitin E3 ligases with deletion of natural substrate binding domain to allow highly selective degradation of SOX2 protein. To evaluate the in vitro efficacy, SOX2 bioPROTACs were generated by mRNA IVT with CleanCap ® and modified uridine. First, robust degradation of SOX2 with a half-maximal degradation concentration (DC50) of the picomolar range was observed with the treatment of SOX2 bioPROTACs in multiple SOX2-overexpressing cancer cell lines. The degradation was ubiquitin- and proteosome-dependent manner, and neither nanobody nor ubiquitin E3 ligase alone affected SOX2 protein level. Then, SOX2 bioPROTACs led to significant cancer cell growth inhibition caused by apoptotic cell death with a half-maximal growth inhibition concentration (GI50) of the picomolar range. Notably, chemo-resistant cancer cell lines expressing SOX2 protein in high levels were sensitized to chemotherapy in combination with our SOX2 bioPROTACs. Furthermore, i.v. injection of one of our lead candidates formulated with LNP significantly inhibited tumor growth in the A549 xenograft model, proving the in vivo anti-tumor efficacy of SOX2 degraders for the first time in the world. Taken together, EPD’s SOX2 bioPROTACs show potent in vitro and in vivo anti-tumor efficacy as first-in-class SOX2 degraders and provide new insights for the therapeutic potential of transcription factor-degrading strategies. Toxicity of commercial and pure forms of three nonsteroidal anti-inflammatory drugs in Xenopus laevis embryos before and after ozonation

In this study, the toxic and teratogenic effects of three commercial drugs and their active ingredients on Xenopus laevis embryos before and after ozonation were evaluated using the Frog Embryos Teratogenesis Assay–Xenopus (FETAX). First, the median lethal concentration (LC50) and, if data were available, the median effective concentration, teratogenic index and minimum growth inhibitory concentration were determined for each drug substance without ozonation. Then, the active substance amounts of three selected nominal concentrations (LC50/2, LC50, and LC50×2) of each test substance before ozonation were measured by HPLC analysis and the toxicity of these substances was evaluated after 2, 3, 4, and 5 h of ozonation. In addition, degradation products that may occur during ozonation were evaluated by LC-MS analysis. The 96-h LC50s of Dolphin–diflunisal, Dichloron–diclofenac sodium, and Apranax–naproxen drug–active substance pairs were determined to be 22.3 and 11.1, 25.7 and 18.7, and 47.8 mg active substance/L and 45.3 mg/L, respectively. According to the FETAX test results, the Dolphin–diflunisal drug–active ingredient pair did not cause growth retardation in exposed embryos. Dichloron–diclofenac sodium and Apranax–naproxen drug–active ingredient pairs were both teratogenic and growth inhibitory. In the second stage of the study, in which the effectiveness of ozonation in eliminating the toxic effects of drugs is evaluated, it is seen that ozonation is partially successful in eliminating the toxic effects of Dolphin–diflunisal and Dichloron–diclofenac sodium pairs, but insufficient for eliminating the effects of the Apranax–naproxen pair.

Why do lactic acid bacteria thrive in chain elongation microbiomes?

Efficient waste management is necessary to transition towards a more sustainable society. An emerging trend is to use mixed culture biotechnology to produce chemicals from organic waste. Insights into the metabolic interactions between community members and their growth characterization are needed to mediate knowledge-driven bioprocess development and optimization. Here, a granular sludge bioprocess for the production of caproic acid through sugar-based chain elongation metabolism was established. Lactic acid and chain-elongating bacteria were identified as the two main functional guilds in the granular community. The growth features of the main community representatives (isolate Limosilactobacillus musocae G03 for lactic acid bacteria and type strain Caproiciproducens lactatifermentans for chain-elongating bacteria) were characterized. The measured growth rates of lactic acid bacteria (0.051 ± 0.005h-1) were two times higher than those of chain-elongating bacteria (0.026 ± 0.004h-1), while the biomass yields of lactic acid bacteria (0.120 ± 0.005g biomass/g glucose) were two times lower than that of chain-elongating bacteria (0.239 ± 0.007g biomass/g glucose). This points towards differential growth strategies, with lactic acid bacteria resembling that of a r-strategist and chain-elongating bacteria resembling that of a K-strategist. Furthermore, the half-saturation constant of glucose for L. mucosae was determined to be 0.35 ± 0.05g/L of glucose. A linear trend of caproic acid inhibition on the growth of L. mucosae was observed, and the growth inhibitory caproic acid concentration was predicted to be 13.6 ± 0.5g/L, which is the highest reported so far. The pre-adjustment of L. mucosae to 4g/L of caproic acid did not improve the overall resistance to it, but did restore the growth rates at low caproic acid concentrations (1-4g/L) to the baseline values (i.e., growth rate at 0g/L of caproic acid). High resistance to caproic acid enables lactic acid bacteria to persist and thrive in the systems intended for caproic acid production. Here, insights into the growth of two main functional guilds of sugar-based chain elongation systems are provided which allows for a better understanding of their interactions and promotes future bioprocess design and optimization.

A novel defensin-like peptide C-13326 possesses protective effect against multidrug-resistant Aeromonas schubertii in hybrid snakehead (Channa maculate ♀ × Channa argus ♂).

The purpose of this study was to investigate whether a defensin-like antimicrobial peptide (C-13326 peptide) identified in Hermetia illucens could possess protective effect against multidrug-resistant Aeromonas schubertii in hybrid snakehead (Channa maculate ♀ × Channa argus ♂). The cDNA of C-13326 peptide comprised 243 nucleotides encoding 80 amino acids, with six conserved cysteine residues and the classical CSαβ structure. The recombinant expression plasmid pPIC9K-C-13326 was constructed and transformed into GS115 Pichia pastoris, and the C-13326 peptide was expressed by induction with 1% methanol. The crude extract of C-13326 peptide was precipitated by ammonium sulfate, assayed by Braford method, detected by tricine-SDS-PAGE, evaluated by BandScan software and identified by liquid chromatography-mass spectrometry. The C-13326 peptide was shown to have inhibitory activity against the growth of multidrug-resistant A. schubertii DM210910 by using the minimum growth inhibitory concentration and Oxford cup method. In addition, scanning electron microscopy analysis suggested that C-13326 peptide inhibited the growth of A. schubertii DM210910 by damaging the bacterial cell membrane. To explore the role of peptide C-13326 invivo, hybrid snakehead was fed with peptide C-13326 as feed additives for 7 days. The results revealed that C-13326 peptide could significantly down-regulate the expression levels of IL-1β, IL-8, IL-12 and TNF-α (p < .05), and significantly improved the survival rate of hybrid snakehead after challenging with A. schubertii DM210910. Therefore, the C-13326 peptide is a promising antimicrobial agent for A. schubertii treatment in aquaculture.
