Comparative immunochemical studies with antisera to sheep prolactin and bovine growth hormone on anterior pituitaries of ox, sheep, rats and mice

Rabbit antisera to sheep prolactin and bovine growth hormone were used in the indirect fluorescent antibody technique on cryostat sections of anterior pituitaries of sheep, ox, rats and mice. It is demonstrated that in sheep and ox prolactin and growth hormone are manufactured by different acidophilic pituitary cells. Though the antisera do not precipitate the analogous hormones of rats and mice in gel diffusion tests, evidence is given for the specificity of the cross-reaction of the antisera with the analogous murine hormones in situ as found with the fluorescent antibody technique.

EFFECT OF HEAT ON THE IMMUNOLOGICAL PROPERTIES OF HUMAN, BOVINE AND OVINE GROWTH HORMONE.

ABSTRACT Human, bovine and sheep (ovine) growth hormone (HGH, BGH and SGH) were heated in solution at temperatures between 60 and 100 °C. The electrophoretic mobility and immunological properties, such as precipitation reactions in agar gel and haemagglutination with antiserum to untreated hormone, were studied at different degrees of heating. It was found that heat progressively reduced the immunological properties of the growth hormone; however, human growth hormone was more resistant to heat treatment than the bovine and sheep growth hormone. HGH retained precipitation properties when heated at 100° C up to 30 minutes, and reacted in the haemagglutination test when heated at 100° C for less than 60 minutes. BGH and SGH clotted at 100° C. The precipitation reaction with antiserum to BGH disappeared when BGH or SGH was heated at 70° C for more than 10 minutes. Only a weak haemagglutination reaction was retained when BGH or SGH was heated at 80° C for 15 minutes.

The preparation of sheep growth hormone.

SUMMARY Growth hormone has been prepared from sheep pituitary glands by chromatography of a simple buffer extract on DEAE-cellulose. The preparation appears to be free of other anterior pituitary hormones but shows two main components when analysed by starch gel electrophoresis. These components appear similar to those present in standard preparations of ox growth hormone. Sheep growth hormone prepared by this method is not significantly less active than purified ox growth hormone when compared by the tibial-epiphysial cartilage response in hypophysectomized rats.

Immunological estimation of sheep growth hormone.

SUMMARY An electrophoretically homogeneous component obtained by starch gel electrophoresis of sheep growth hormone (GH) has been used to prepare antiserum in rabbits. By means of a haemagglutination—inhibition reaction, this antiserum was used to assay GH both in sheep pituitary extracts and in sheep sera. The values for the GH content of a number of pituitary extracts obtained by both immunological and biological assay methods were in good agreement. GH levels in sheep serum were found to range between 38 and 600 μg./100 ml.

Preparation of Sheep Growth Hormone

EXISTING methods for the preparation of sheep growth hormone by low-temperature ethanol fractionation1 or salt fractionation, combined with ion exchange chromatography2, are lengthy and involve a large number of individual steps. The same criticism applies to existing methods for preparation of ox growth hormone3,4.

METABOLIC EFFECTS OF HUMAN GROWTH HORMONE AND GROWTH HORMONE OF OTHER SPECIES IN MAN

ABSTRACT The metabolic effects of human growth hormone were investigated under varied clinical conditions in 15 balance studies on 10 subjects. Nitrogen retention occurred uniformly, the amount varying from 2 to 4 Gm. daily. Potassium and phosphorus were likewise retained, but in over half the studies, in amounts greater than those calculated from the nitrogen retention. This suggested the deposition of parenchymal tissue rather than muscle tissue. Hypercalciuria and sodium chloride retention often accompanied the administration of human growth hormone. Growth hormone was effective in the hypothyroid subject. Neither increased nor decreased adrenal function altered the anabolic effect. Sheep and whale growth hormone were metabolically inactive in single trials. Nine studies with chymotrypsinized beef growth hormone likewise failed to indicate anabolic activity.
