Synergistic Antifungal Activity of Green Synthesized Zinc Oxide Nanoparticles and Fungicide Against Rhizoctonia solani Causing Rice Sheath Blight Disease.

The biosynthesis of metal oxide nanoparticles using leaf extract of medicinal plants is a promising substitute for the traditional chemical method. This work aimed to synthesize zinc oxide nanoparticles using a green approach from local "Dholkolmi" (Ipomoea carnea) leaf extract which is a medicinal plant growing outside the roads of different regions of Bangladesh. The biosynthesized zinc oxide nanoparticles (ZnONPs) were characterized using ultraviolet-visible spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction, particle size analyzer, zeta-potential, scanning electron microscopy-energy dispersive spectroscopy, field emission scanning electron microscopy, and transmission electron microscopy. The results of UV-visible spectrophotometers observed an absorption peak at 373 nm wavelength, which confirmed the synthesis of ZnONPs in the solution. ZnONP sizes determined by XRD, DLS, and TEM are approximately ~37 nm, 105.61 nm, and 19.66 nm, respectively. ZnONPs were present because of the strong oxygen and zinc signals in the EDX profile. Additionally, this research assessed the antifungal activity of the biosynthesized ZnONPs and as well as folicur-incorporated ZnONPs against Rhizoctonia solani by the poison bait technique. According to the result of this study, ZnONPs synthesized from Ipomoea carnea leaf extract showed no promising result against Rhizoctonia solani mycelial growth reduction. But folicur-incorporated ZnONPs revealed a significant finding with a maximum 100% inhibition of mycelial growth at 1:1 and 3:1 ratio of ZnONPs with folicur fungicide under in vitro conditions. In the net house experiment, folicur-incorporated ZnONPs at a 1:1 ratio of ZnONPs with folicur showed considerable disease inhibition (26.96% RLH) as compared to disease control (52.83% RLH). In the case of rainfed transplanted Aus (March-June), the highest percentage of RLH was recorded in disease control (64.61%), and the lowest RLH was found in folicur (24.79%) followed by a 1:1 ratio of ZnONPs with folicur (32.10%) in field condition. On the other hand, the highest percentage of RLH was recorded in disease control (65.31%) and the lowest RLH was found in folicur (18.14%) followed by a 1:1 ratio of ZnONPs with folicur (21.39%) in rainfed transplanted Aman (July-November) season. The findings of the in vitro and in vivo studies provided evidence that ZnONPs and folicur had a strong synergistic antifungal impact and may be employed as a possible rice sheath blight disease management agent.

Photocatalytic degradation of toxic basic blue 41 and reactive orange M2R dyes using green synthesized zinc oxide nanoparticles

Zinc oxide nanoparticles (Cm-ZnO NPs) were synthesized to eliminate organic dye pollutants from textile dye effluents. The present study reports Cucumis maderaspatanus L. leaves utilized as a green reducing/stabilizing agent for Cm-ZnO NPs synthesis. The nanoparticles underwent thorough analysis using techniques such as XRD, UV–vis, TEM, and FT-IR. Photocatalytic degradation of toxic basic blue 41 (BB41) and reactive orange M2R (ROM2R) dyes using Cm-ZnO NPs, showed a degradation rate of 97.69 % and 94.30 % respectively under optimized conditions. The kinetics of photocatalytic degradation of BB41 and ROM2R dyes followed first-order kinetics. The scavenger studies were conducted to identify the photogenerated reactive species involved in dye degradation. The singlet oxygen (1O2) and hydroxyl radicals (●OH) are mainly responsible for the degradation of BB41 and ROM2R dye respectively. LC-MS analysis was used to identify the photocatalytic degraded products, and ECOSAR software assessed their toxicity. The recyclability was performed up to four cycles and the stability of Cm-ZnO NPs was confirmed by XRD analysis. Thus, the as-synthesized Cm-ZnO NPs are suitable for industrial textile wastewater treatment.

Exogenously applied nano-zinc oxide mitigates cadmium stress in Zea mays L. through modulation of physiochemical activities and nutrients homeostasis

The increasing levels of cadmium (Cd) pollution in agricultural soil reduces plant growth and yield. This study aims to determine the impact of green synthesized zinc oxide nanoparticles (ZnO-NPs) on the physiochemical activities, nutrition, growth, and yield of Zea mays L. under Cd stress conditions. For this purpose, ZnO-NPs (450 ppm and 600 ppm) synthesized from Syzygium aromaticum were applied through foliar spray to Z. mays and also used as seed priming agents. A significant decline in plant height (35.24%), biomass production (43.86%), mineral content, gas exchange attributes, and yield (37.62%) was observed in Cd-spiked plants compared to the control. While, 450 ppm ZnO-NPs primed seed increased plant height (18.46%), total chlorophyll (80.07%), improved ascorbic acid (25.10%), DPPH activity (26.66%), and soil mineral uptake (Mg+2 (38.86%), K+ (27.83%), and Zn+2 (43.68%) as compared to plants only spiked with Cd. On the contrary, the foliar-applied 450 ppm ZnO-NPs increased plant height (8.22%), total chlorophyll content (73.59%), ascorbic acid (21.39%), and DPPH activity (17.61%) and yield parameters; cob diameter (19.45%), and kernels numbers 6.35% enhanced compared to plants that were spiked only with Cd. The findings of the current study pave the way for safer and more cost-effective crop production in Cd-stressed soils by using green synthesized NPs and provide deep insights into the underlying mechanisms of NPs treatment at the molecular level to provide compelling evidence for the use of NPs in improving plant growth and yield.

Spectroscopic investigations of green synthesized zinc oxide nanoparticles (ZnO NPs): antioxidant and antibacterial activity

In the recent decade, zinc oxide nanoparticles (ZnO NPs) have been widely explored owing to their versatile properties and prodigious demands in the drug delivery, medical, energy storage, cosmetics, and the healthcare sectors. Therefore, the current work opts for an environmentally benign method to prepare ZnO NPs. The leaf extract of Calendula officinalis L. acts as a reducing agent for the metal ions; therefore, in the current research, ZnO NPs were prepared via green route by using Calendula officinalis leaf extract. Furthermore, the ZnO NPs were analysed with different spectroscopic techniques to confirm the structure and stability of nanomaterials. The prepared ZnO NPs were characterized by XRD, FE-SEM, FT-IR and UV–Vis studies. Also, the antioxidant and antimicrobial properties of the synthesized ZnO NPs were investigated. The XRD result of synthesized ZnO NPs showed the crystalline size 28.23 nm with wurtzite hexagonal structure along with the most intense peak (101). Following preliminary confirmations of the intended ZnO NPs, both big and small agglomerated forms were observed in the FE-SEM, which is often used to determine their exterior assembly. Further, the results of Fourier transform infrared spectroscopy (FT-IR) indicated the formation of pure ZnO NPs with an absorption peak of the Zn–O bond between 4000 cm−1 and 500 cm−1 and no discernible peak in the monitoring range. The UV–Vis spectrum of the green synthesized ZnO NPs were revealed two prominent absorption peaks at 355 nm and 370 nm with energy band gap of 2.986 eV. Using the 1, 1-di phenyl-2-picrylhydrazyl (DPPH) test, the antioxidant activity of the described ZnO NPs was assessed. It demonstrated how, ZnO NPs significantly increased their antioxidant activity by scavenging 1, 1-di phenyl-2-picrylhydrazyl (DPPH) free radicals. It could be seen that synthesis of the naturally occurring plant product ZnO NPs have been acting as an alternate of chemical antioxidant. The antimicrobial analysis was also performed with the help of disk diffusion method where three multi-drug resistant human pathogens namely Staphylococcus aureus, Klebsiella pneumoniae and E.coli were used. The Zone of Inhibition diameter values are 35.2 mm ± 0.9, 23.6 mm ± 0.1 and 13.5 mm ± 0.1, respectively, which showed that the ZnO NPs was highly effective against S. aureus. Thus, the green synthesis method of ZnO NPs using leaf extract of Calendula officinalis is evidence that it is superior and environmentally friendly method for the preparation of ZnO NPs and hence it can be utilized in various nano-medicine approaches.

Enhanced healing of burn wounds by multifunctional alginate-chitosan hydrogel enclosing silymarin and zinc ‎oxide ‎nanoparticles

Multifunctional wound dressings have been applied for burn injuries to avoid complications and promote tissue regeneration. In the present study, we fabricated a natural alginate-chitosan hydrogel comprising silymarin and green-synthesized zinc ‎oxide ‎nanoparticles (ZnO NPs). Then, the physicochemical attributes of ZnO NPs and loaded hydrogels were analyzed. Afterward, wound healing efficacy was evaluated in a rat model of full-thickness dermal burn wounds. The findings indicated that ZnO NPs were synthesized via reduction with phytochemicals from Elettaria cardamomum seeds extract. The microscopic images exhibited fairly spherical ZnO NPs (35-45nm), and elemental analysis verified the relevant composition. The hydrogel, containing silymarin and biosynthesized ZnO NPs, displayed a uniform appearance, smooth surfaces, and a porous structure. Moreover, infrared spectroscopy ‎identified functional groups, confirming the successful loading without adverse interactions. The obtained hydrogel exhibited great water absorption, high porosity, sustainable degradation for several days, and enhanced antioxidant capability of the combined loaded component. In vivo studies revealed faster and superior wound healing, achieving nearly complete closure by day 21. Histopathology confirmed improved cell growth, tissue regeneration, collagen deposition, and neovascularization. It is believed that this multifunctional hydrogel-based wound dressing can be applied for effective burn wound treatment.

Enhanced photocatalytic activity of green synthesized zinc oxide nanoparticles using low-cost plant extracts

Developing stable and highly efficient metal oxide photocatalysts remains a significant challenge in managing organic pollutants. In this study, zinc oxide nanoparticles (ZnO NPs) were successfully synthesized using various plant extracts, pomegranate (P.M), beetroot roots (B.S), and seder, along with a chemical process. The produced ZnO NPs were characterized using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), ultraviolet–visible spectroscopy (UV–Vis), Field Emission Scanning Electron Microscope (FESEM), High-Resolution Transmission Electron Microscopy (HRTEM), and Surface Area. For all prepared samples, the results indicated that the composition of the plant extract affects several characteristics of the produced particles, such as their photocatalytic properties, energy bandgap (Eg), particle size, and the ratio of the two intensity (0 0 2) and (1 0 0) crystalline planes. The particle size of the produced NPs varies between 20 and 30 nm. To examine NPs' photocatalytic activity in the presence of UV light, Methyl Orange (MO) was utilized. The Eg of ZnO synthesized by the chemical method was 3.16 e. V, whereas it was 2.84, 2.63, and 2.59 for P.M, Seder, and B.S extracts, respectively. The most effective ZnO NPs, synthesized using Beetroots, exhibited a degradation efficiency of 87 ± 0.5% with a kinetic rate constant of 0.007 min−1. The ratio of the two intensity (0 0 2) and (1 0 0) crystalline planes was also examined to determine a specific orientation in (0 0 2) that is linked to the production of oxygen vacancies in ZnO, which enhances their photocatalytic efficiency. Furthermore, the increase in photocatalytic effectiveness can be attributed to the improved light absorption by the inter-band gap states and effective charge transfer.

Acrylamide-targeting renal miR-21a-5p/Fibrotic and miR122-5p/ inflammatory signaling pathways and the role of a green approach for nano-zinc detected via in silico and in vivo approaches.

Introduction: Any disruption in renal function can have cascading effects on overall health. Understanding how a heat-born toxicant like acrylamide (ACR) affects kidney tissue is vital for realizing its broader implications for systemic health. Methods: This study investigated the ACR-induced renal damage mechanisms, particularly focusing on the regulating role of miR-21a-5p/fibrotic and miR-122-5p/inflammatory signaling pathways via targeting Timp-3 and TP53 proteins in an In silico preliminary study. Besides, renal function assessment, oxidative status, protein profile, and the expression of renal biomarkers (Timp-1, Keap-1, Kim-1, P53, TNF-α, Bax, and Caspase3) were assessed in a 60-day experiment. The examination was additionally extended to explore the potential protective effects of green-synthesized zinc oxide nanoparticles (ZNO-MONPs). A four-group experiment including control, ZNO-MONPs (10mg/kg b.wt.), ACR (20mg/kg b.wt.), and ZNO-MONPs + ACR was established encompassing biochemical, histological, and molecular levels. The study further investigated the protein-binding ability of ZNO and MONPs to inactivate caspase-3, Keap-1, Kim-1, and TNFRS-1A. Results: ZNO-MONPs significantly reduced ACR-induced renal tissue damage as evidenced by increased serum creatinine, uric acid, albumin, and oxidative stress markers. ACR-induced oxidative stress, apoptosis, and inflammationare revealed by biochemical tests, gene expression, and the presence of apoptotic nuclei microscopically. Also, molecular docking revealed binding affinity between ACR-BCL-2 and glutathione-synthetase, elucidating the potential mechanisms through which ACR induces renal damage. Notably, ZNO-MONPs revealed a protective potential against ACR-induced damage. Zn levels in the renal tissues of ACR-exposed rats were significantly restored in those treated with ACR + ZNO-MONPs. In conclusion, this study establishes the efficacy of ZNO-MONPs in mitigating ACR-induced disturbances in renal tissue functions, oxidative stress, inflammation, and apoptosis. The findings shed light on the potential renoprotective activity of green-synthesized nanomaterials, offering insights into novel therapeutic approaches for countering ACR-induced renal damage.

Green-synthesized CuO and ZnO nanoparticles derived from Calotropis gigantea (Apple of Sodom): enhancing plant growth, efficient dye removal, and potent antibacterial applications.

Nanoparticles, owing to their unique physicochemical properties, have garnered significant attention in various scientific disciplines, including materials science, chemistry, biology, and environmental engineering. In recent years, the synthesis of metal oxide nanoparticles, such as NiO, Fe2O3, ZnO, SnO2, and CuO via green routes, has gained attraction due to their diverse applications in fields ranging from catalysis and electronics to medicine and environmental remediation. This study focuses on the green synthesis of copper oxide (CuO) and zinc oxide (ZnO) nanoparticles using Calotropis gigantea (Apple of Sodom) leaf extract as a reducing agent and stabilizer, with zinc nitrate (ZnNO3.6H2O) and copper nitrate (CuNO3.3H2O) as precursors. The hexagonal phase of ZnO and monoclinic plan structure of CuO with high crystallinity was confirmed by XRD and elemental composition by EDX analysis. With the help of an SEM image, particle size measured for CuO and ZnO using ImageJ software was found to be 56.08nm and 46.49nm, respectively. This study investigates the efficacy of nanoparticles in wastewater treatment, particularly focusing on methylene blue dye decolorization using the statistical processing of response surface methodology (RSM) using the Box-Behnken method. Additionally, it explores the impact of synthesized nanoparticles on seed growth enhancement, using Vigna radiata (green gram) seeds immersed in various doses of nanoparticles (0, 0.5, 1, 1.5, 2mg/30mL). Furthermore, the antibacterial activity of the nanoparticles against both gram-positive and gram-negative bacteria is evaluated. The results confirm the effectiveness of the materials for methylene blue dye removal, achieving 80.53% with CuO and 78.25% with ZnO. Significant seed growth was observed with a low nanoparticle dosage of 1.5mg/30mL, resulting in the highest seedling vigour index and germination percentage. This reduces the need for fertilizers and lessens environmental impact.

Assessing anticancer, antidiabetic, and antioxidant capacities in green-synthesized zinc oxide nanoparticles and solvent-based plant extracts

Cancer and diabetes represent significant challenges in the field of biomedicine, with major and global impacts on public health. Acacia nilotica, commonly called 'gum arabic tree,' is recognized for its unique biomedical properties. The current study aimed to investigate the pharmacological potential of A. nilotica-based zinc-oxide nanoparticles (ZnO-NPs) in comparison to the ethanol and methanol-based extracts against cancer, diabetes, and oxidative stress. Green synthesis of ZnO-NPs was performed using barks of Acacia nilotica. Different techniques for the characterization of ZnO-NPs, including UV–Visible spectroscopy, Scanning Electron Microscopy, Fourier Transmission Infrared (FT-IR) spectroscopy, and X-ray Diffraction (XRD), were utilized. The morphological analysis of ZnO-NPs revealed that the fine NPs have mean particle sizes of 15 ± 1.5 nm. For the solvent based-extraction, leaves and barks were utilized and dissolved into ethanol and methanol for further processing. The MTT assay revealed that the optimum concentration of ZnO-NPs to inhibit the proliferation of liver cancer cell line HepG2 was 100 μg/mL where 67.0 % inhibition was observed; and both ethanol- and methanol-based extracts showed optimum inhibition at 100 μg/mL. The DPPH assay further demonstrated that 250 μg/mL of ZnO-NPs and 1000 μg/mL of both ethanol- and methanol-based extracts, as the optimum concentration for antioxidant activity (with 73.1 %, 68.9 % and 68.2 % inhibition respectively). The α-Glucosidase inhibition assay revealed that 250 μg/mL of ZnO-NPs and 10 μg/mL of both ethanol- and methanol-based extracts as the optimum concentration for antidiabetic activity (with 95 %, 93.7 % and 93.4 % inhibition respectively). The study provided interesting insights into the efficacy and reliability of ZnO-NPs for potential pharmacological application. Further research should be focused on examining specific pathways and the safety of ZnO-NPs in comparison to solvent-based extracts.

Green route to synthesize zinc oxide nanoparticles (ZnO[sbnd]NPs) using leaf extracts of Merremia quinquefolia (L.) Hallier f. and their potential applications

The current investigation focus on the facile, cost-effective, ecologically sustainable and green synthesized zinc oxide nanoparticles (ZnO NPs) from the Merremia quinquefolia leaf aqueous extract and tests their biomedical application. The ultraviolet (UV) spectrometry absorption peak at 373 nm validated the synthesis of zinc oxide nanoparticles. The X-ray diffraction (XRD) pattern indicates the crystalline nature of the material, and Fourier transform infrared (FTIR) analysis reveals its functional groups. Morphological studies in the scanning electron microscopy (SEM) and transmission electron microscope (TEM) analysis revealed rod-like shapes for ZnO NPs with sizes in the range of 43–70 nm. Energy-dispersive X-ray spectroscopy (EDX) confirmed the presence of zinc, oxygen and organic molecules. Additionally, the vibrational peak at 437 and 480 cm−1 in the Raman spectra indicated it. By using the disc diffusion method, the zinc oxide nanoparticles that were made more effective at killing gram-positive pathogens, with Staphylococcus aureus having the largest zone of inhibition (14 ± 0.23). Assays for DPPH and ABTS radical scavenging showed higher antioxidant activity. It had a strongest effect on the anti-inflammatory activity of the IC50 values for albumin denaturation (92.78 μg/mL) and human red blood cells (84.59 μg/mL). Furthermore, the IC50 value of 41.81 μg/mL for zinc oxide nanoparticles was good for stopping the development of breast cancer. These findings indicate that green synthesized zinc oxide nanoparticles will make great biocompatible, antibacterial, anti-inflammatory and anticancer drugs in the future.

Supercritical CO2 directional-assisted green synthesis of ZnO nanoparticles obtained from Euphorbia stricta L. for functional applications

The green synthesis method is increasingly favored due to its economic and environmental benefits. Metal oxides, such as zinc oxide (ZnO) nanoparticles produced via green synthesis, exhibit biocompatible and stable properties. In this study, the Euphorbia stricta plant was extracted using the supercritical CO2 extraction method. Subsequently, ZnO nanoparticles were synthesized using the Euphorbia stricta extract through a microwave-assisted hydrothermal method. The synthesized ZnO was characterized using X-ray diffraction (XRD), Ultraviolet–visible spectroscopy (UV–Vis), and Fourier-transform infrared spectroscopy (FT-IR). Euphorbia stricta L. extract was also characterized by FT-IR. To evaluate the photocatalytic activities against methylene blue, ZnO/polyacrylonitrile (PAN) membranes were fabricated via electrospinning for comparative analysis. The degradation efficiencies of ZnO and ZnO/PAN nanofibers were 76.37 % and 66.14 % over 180 min, respectively. Furthermore, the green synthesized ZnO nanoparticles demonstrated significant antibacterial properties against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. These bacteria are of critical importance in medical and environmental contexts: cause of skin infections, pneumonia and bloodstream infections; foodborne illnesses and urinary tract infection and hospital-acquired infections. The findings suggest that ZnO nanoparticles synthesized from Euphorbia stricta extract have substantial potential for application in antibacterial treatments and photocatalysis, offering a promising approach to addressing challenges posed by these pathogenic bacteria.

Topical gel formulations as potential dermal delivery carriers for green-synthesized zinc oxide nanoparticles.

This study aimed to incorporate green-synthesized zinc oxide nanoparticles (ZnO NPs), functionalized with polyethylene glycol (PEG) and linked to doxorubicin (DOX), into various topical gel formulations (hydrogel, oleogel, and bigel) to enhance their dermal delivery. The ZnO NPs were produced using the aqueous extract of the root hair of Phoenix dactylifera. The optimized green-synthesized ZnO NPs, PEGylated and conjugated to DOX, demonstrated a particle size below 100nm, low polydispersity index, and zeta potential between - 11 and - 19 mV. The UV-Vis spectroscopy analysis confirmed characteristic absorption peaks at 351 and 545nm for ZnO and DOX, respectively. The transmission electron microscope (TEM) images revealed well-dispersed spherical nanoparticles without aggregation. Additionally, ZnO NPs-loaded gels exhibited uniformity, cohesion, no phase separation, pseudoplastic flow, and viscoelastic properties. The in vitro release studies showed that DOX-PEG-ZnO NPs hydrogel released 99.5% of DOX after 5h of starting the release. Moreover, the penetration of DOX-PEG-ZnO NPs through excised rat skin was visualized by TEM. In conclusion, the hydrogel formulation containing green-synthesized DOX-PEG-ZnO NPs holds great promise for dermal administration in skin cancer treatment. Furthermore, the release rate and skin penetration of DOX from gels were varied based on the type of gel matrix and corroborated with their corresponding rheological properties.

Bacteriophages and Green Synthesized Zinc Oxide Nanoparticles in Combination Are Efficient against Biofilm Formation of Pseudomonas aeruginosa.

Bacteriophages (phages) are viruses that infect the bacteria within which their reproduction cycle takes place, a process that ends in the lysis and death of the bacterial cell. Some phages are also able to destroy bacterial biofilms. Due to increased antibiotics resistance, Pseudomonas aeruginosa, another biofilm-forming pathogen, is a problem in many parts of the world. Zinc oxide (ZnO) and other metal nanoparticles (NPs) are biologically active and also possess anti-biofilm properties. ZnO-NPs were prepared by the green synthesis method using orange peels. The vibrational peaks of the ZnO-NPs were analyzed using FTIR analysis, and their size and morphological properties were determined using scanning electron microscopy (SEM). The ability of the ZnO-NPs to reduce or eliminate P. aeruginosa biofilm alone or in combination with phages PB10 and PA19 was investigated. The P. aeruginosa cells were effectively killed in the preformed 48 h biofilms during a 24 h incubation with the ZnO-NP-phage combination, in comparison with the control or ZnO-NPs alone. The treatments on growing biofilms were most efficient in the final stages of biofilm development. All five treatment groups showed a significant biofilm reduction compared to the control group (p < 0.0001) at 48 h of incubation. The influence of the ZnO-NPs and phages on the quorum sensing system of P. aeruginosa was monitored by quantitative real-time PCR (qRT-PCR) of the autoinducer biosynthesis gene lasI. While the ZnO-NPs repressed the lasI gene transcription, the phages slightly activated it at 24 and 48 h of incubation. Also, the effect of the ZnO-NPs and phage PA19 on the viability of HFF2 cells was investigated and the results showed that the combination of NPs with PA19 reduced the toxic effect of ZnO-NPs and also stimulated the growth in normal cells.

Green-synthesized zinc oxide nanoparticles by Enterobacter sp.: unveiling characterization, antimicrobial potency, and alleviation of copper stress in Vicia faba (L.) plants

BackgroundThe biosynthesis of zinc oxide nanoparticles (ZnO NPs) using Enterobacter sp. and the evaluation of their antimicrobial and copper stress (Cu+ 2)-reducing capabilities in Vicia faba (L.) plants. The green-synthesized ZnO NPs were validated using X-ray powder diffraction (XRD); Fourier transformed infrared (FTIR), Ultraviolet-Visible spectroscopy (UV-Vis), Transmission electron microscope (TEM) and scanning electron microscopy (SEM) techniques. ZnO NPs could serve as an improved bactericidal agent for various biological applications. as well as these nanoparticles used in alleviating the hazardous effects of copper stress on the morphological and physiological traits of 21-day-old Vicia faba (L.) plants.ResultsThe results revealed that different concentrations of ZnO NPs (250, 500, or 1000 mg L-1) significantly alleviated the toxic effects of copper stress (100 mM CuSO4) and increased the growth parameters, photosynthetic efficiency (Fv/Fm), and pigments (Chlorophyll a and b) contents in Cu-stressed Vicia faba (L.) seedlings. Furthermore, applying high concentration of ZnO NPs (1000 mg L-1) was the best dose in maintaining the levels of antioxidant enzymes (CAT, SOD, and POX), total soluble carbohydrates, total soluble proteins, phenolic and flavonoid in all Cu-stressed Vicia faba (L.) seedlings. Additionally, contents of Malondialdehyde (MDA) and hydrogen peroxide (H2O2) were significantly suppressed in response to high concentrations of ZnO NPs (1000 mg L-1) in all Cu-stressed Vicia faba (L.) seedlings. Also, it demonstrates strong antibacterial action (0.9 mg/ml) against various pathogenic microorganisms.ConclusionsThe ZnO NPs produced in this study demonstrated the potential to enhance plant detoxification and tolerance mechanisms, enabling plants to better cope with environmental stress. Furthermore, these nanoparticles could serve as an improved bactericidal agent for various biological applications.

Green synthesis of zinc oxide nanoparticles from Sida acuta leaf extract for antibacterial and antioxidant applications, and catalytic degradation of dye through the use of convolutional neural network

This study synthesized zinc oxide nanoparticles (ZnO NPs) using a novel green approach, with Sida acuta leaf extract as a capping and reducing agent to initiate nucleation and structure formation. The innovation of this study lies in demonstrating the originality of utilizing zinc oxide nanoparticles for antibacterial action, antioxidant potential, and catalytic degradation of Congo red dye. This unique approach harnesses eco-friendly methods to initiate nucleation and structure formation. The synthesized nanoparticles' structure and conformation were characterized using UV–vis (λmax = 280 nm), X-ray, atomic force microscopy, SEM, HR-TEM and FTIR. The antibacterial activity of the Nps was tested against Pseudomonas sp, Klebsiella sp, Staphylococcus aureus, and E. coli, demonstrating efficacy. The nanoparticles exhibited unique properties, with a crystallite size of 20 nm (XRD), a surface roughness of 2.5 nm (AFM), and a specific surface area of 60 m2/g (SEM). A Convolutional Neural Network (CNN) was effectively employed to accurately classify and analyze microscopic images of green-synthesized zinc oxide nanoparticles. This research revealed their exceptional antioxidant potential, with an average DPPH scavenging rate of 80% at a concentration of 0.05 mg/mL. Additionally, zeta potential measurements indicated a stable net negative surface charge of approximately −12.2 mV. These quantitative findings highlight the promising applications of green-synthesized ZnO NPs in healthcare, materials science, and environmental remediation. The ZnO nanoparticles exhibited catalytic capabilities for dye degradation, and the degradation rate was determined using UV spectroscopy. Key findings of the study encompass the green synthesis of versatile zinc oxide nanoparticles, demonstrating potent antibacterial action, antioxidant capabilities, and catalytic dye degradation potential. These nanoparticles offer multifaceted solutions with minimal environmental impact, addressing challenges in various fields, from healthcare to environmental remediation.

Antimicrobial and anti-inflammatory efficiency of green synthesized zinc oxide nanoparticles using Polianthes tuberosa flower concentrate

Aim: In the present study, zinc oxide nanoparticles (ZnO NPs) were synthesized using Polianthes tuberosa flower concentrate. Antimicrobial and anti-inflammatory efficiency of the green synthesized ZnO NPs were analysed under in vitro conditions. Methodology: Nanoparticles formation was confirmed by UV-visible spectroscopy. Size, shape and morphology of the green synthesized ZnO NPs was determined by SEM coupled with EDAX and AFM. Molecular and elemental compositions of the nanoparticle were determined by XRD and FT-IR analyses. Results: X-ray diffraction analysis depicted the hexagonal wurtzite structure of ZnO NPs with a particle size of 80 nm. The FT-IR analysis illustrated the functional groups responsible for the encapsulation and stabilization of ZnO NPs. At micromolar concentration, ZnO NPs was found to inhibit the growth of selected skin pathogens and suppress biofilm formation in Pseudomonas aeruginosa. Interpretation: P. tuberosa flower concentrate and green synthesized ZnO Nps inhibited the growth of bacterial and fungal pathogen in a dose-dependent manner. Biofilm inhibitory efficiency and anti-inflammatory activity of the zinc oxide nanoparticle illustrated that it can be used for treating skin infections. Cytotoxicity of green synthesized zinc oxide nanoparticles using Musa acuminata on Vero cells

Zinc oxide nanoparticles (ZnO NPs) have become a highly regarded substance in various industries especially biologically synthesized ZnO NPs due to their adherence to the principles of green chemistry. However, concerns have been raised regarding the potential cytotoxic effects of ZnO NPs on biological systems. This study aimed to investigate and compare the cytotoxicity of ZnO NPs that were synthesized through chemical (C–ZnO NPs) and green approach using Musa acuminata leaf aqueous extract (Ma-ZnO NPs) on Vero cells.Characterization of ZnO NPs through Uv–Vis, FESEM, EDX, XRD, FTIR and XPS confirmed the successful synthesis of C- and Ma-ZnO NPs. MTT and ROS assays revealed that C- and Ma-ZnO NPs induced a concentration- and time-dependent cytotoxic effect on Vero cells. Remarkably, Ma-ZnO NPs showed significantly higher cell viability compared to C–ZnO NPs. The corelation of ROS and vell viability suggest that elevated ROS levels can lead to cell damage and even cell death. Flow cytometry analysis indicated that Ma-ZnO NPs exposed cells had more viable cells and a smaller cell population in the late and early apoptotic stage. Furthermore, more cells were arrested in the G1 phase upon exposure to C–ZnO NPs, which is associated with oxidative stress and DNA damage caused by ROS generation, proving its higher cytotoxicity than Ma-ZnO NPs. Similarly, time-dependent cytotoxicity and morphological alterations were observed in C- and Ma-ZnO NPs treated cells, indicating cellular damage. Furthermore, fluorescence microscopy also demonstrated a time-dependent increase in ROS formation in cells exposed to C- and Ma-ZnO NPs.In conclusion, the findings suggest that green ZnO NPs possess a favourable biocompatibility profile, exhibiting reduced cytotoxicity compared to chemically synthesized ZnO NPs on Vero cells. These results emphasize the potential of green synthesis methods for the development of safer and environmentally friendly ZnO NPs.

Evaluation of the antimicrobial activity and chickpea growth stimulation effects of green synthesized zinc oxide nanoparticles

Green synthesized nanoparticles are believed to alleviate the detrimental effects of chemical fertilizers and the multidrug resistance of various microbial strains. This study evaluates the use of bush tea-mediated zinc oxide nanoparticles (ZnO NPs) for chickpea fertilization and assesses their cytotoxicity, antioxidant and antibacterial activity. The study demonstrated that the application of ZnO NPs improved the number of leaves, nodules, biomass and yield of chickpea cultivars under glasshouse conditions. The authors suggest that biosynthesized ZnO NPs are a good plant growth stimulant. The control with no nanoparticles application, had higher values of photosynthetic rate and water use efficiency in the Desi genotype whereas for the Kabuli genotype, 25 mg/L showed higher values of stomatal conductance and transpiration efficiency, while 15 mg/L gave the highest water use efficiency and photosynthetic rate. Furthermore, the inhibitory activities of bacteria were reported in which E. coli and S. aureus were the most sensitive bacterial strains. The ZnO NPs also quenched free radicals that demonstrate antioxidant activity. Thus, they can be beneficial to both plants and humans as they reduce oxidative stress that can cause diseases. Furthermore, toxicity to cells was evaluated and ZnO NPs were less toxic to the studied cells with IC50 values of 16.17 µg/mL and 21.30 µg/mL against lung cancer and embryonic cell lines. Thus, the use of ZnO nanoparticles with antimicrobial activity could help fight against several diseases attacking humans and plants.

تأثير دقائق اوكسيد الزنك النانوية المصنعة بايولوجيًا على النمط الظاهري والجيني للغشاء الحيوي لبكتريا Proteus mirabilis

تصنف بكتريا Proteus mirabilis كثالث مسبب لالتهاب المجاري البولية المصاحب لتركيب أنابيب القسطرة البولية. تعد الاغشية الحيوية من عوامل الضراوة اللتي تستعملها البكتريا لمقاومة المضادات الحيوية وفي ضل تطور مقاومة البكتريا للمضادات الحيوية, صار لزامًا على العلماء والباحيثن في مجال الاحياء المجهرية أيجاد بدائل ذات كفائة عالية قادرة على اختراق جدار الخلية البكتيرية وبالتالي قتلها.في السنوات الاخيرة, اكتسبت دقائق النانو المصنعة بايولوجيا والصديقة للبيئة أهتمامًا كبيرًا من قبل الباحثين لأمتلاكها خصائص مميزة كصغر حجمها المتناهي واشكالها المختلفة يمَكنهم من استعمالها كمواد مضادة للبكتيريا في مختلف المجلات الطبية والصحية. تهدف هذه الدراسة الى التحري عن قدرة دقائق اوكسيد النحاس النانوية المصنعة بايولوجيا على تثبيط تكوين الغشاء الحيوي المنتج من قبل بكتيريا Proteus mirabilis. أنتجت هذه الدقائق بأستعمال راشح بكتيري يحتوي على الانزيمات المختزلة لملح النحاس واستخدمت عدة تقنيات لتحديد الصفات الفيزيائية المظهرية لدقائق النانو المنتجة حيث استخدمت تقنية XRD لتأكيد التركيب خماسي الشكل لهذه الدقائق , بينما استعملت تقنية TEM لتحديد قطر الجزيئات المنتجة بمعدل 84.45 نانومتر. اما FESEM فقد تم استخدامه لتحديد الصفات الخارجية لسطح جزيئات النانو المنتجة , تم تأكيد النتائج بأستخدام تقنية AFM للكشف عن توزيع و وعورة جزيئات النانو.اما UV فقد سجلت ااقصى امتصاصية على الطول الموجي nm287 .تم التحري في هذه الدراسة عن فعالية دقائق اوكسيد النحاس النانوية في منع تكون الغشاء الحيوي حيث أستعمل مستحلب دقائق اوكسيد النحاس النانوي بالتركيز تحت القاتل 32 μg/ml . سجلت نتائج هذه الدراسة انخفاضا ملحوضَا في انتاج الغشاء الحيوي للبكتريا المنتجه له حيث سجلت العزلات ذات الانتاجية العالية للغشاء الحيوي ضعفأ في الانتاج بعد معاملتها بالتركيز المستعمل من دقائق اوكسيد النحاس النانوية. جاء تأثير هذه الدقائق على كل من النمط الظاهري والتعبير الجيني لجين LuxS المسؤول عن تكوين الغشاء الحيوي بأستخدام تقنية rtPCRحيث سجلت الدراسة انخفاضا في التعبير الجيني لهذا الجين بعد معاملة سلالات البكتريا قيد الدراسة بدقائق اوكسيد النحاس النانوية مقارنتاً بالعزلات غير المعاملة. من خلال نتائج هذه التجربة يمكن التوصل الى امكانية اعتماد دقائق اوكسيد الزنك النانوية كعامل مضاد للبكتيريا عن استخدامه بالتركيز المناسب.
