Progressive increase of brain gray matter volume in individuals with regular soccer training

The study aimed to investigate alterations in gray matter volume in individuals undergoing regular soccer training, using high-resolution structural data, while also examining the temporal precedence of such structural alterations. Both voxel-based morphometry and source-based morphometry (SBM) methods were employed to analyze volumetric changes in gray matter between the soccer and control groups. Additionally, a causal network of structural covariance (CaSCN) was built using granger causality analysis on brain structural data ordering by training duration. Significant increases in gray matter volume were observed in the cerebellum in the soccer group. Additionally, the results of the SBM analysis revealed significant increases in gray matter volume in the calcarine and thalamus of the soccer group. The analysis of CaSCN demonstrated that the thalamus had a prominent influence on other brain regions in the soccer group, while the calcarine served as a transitional node, and the cerebellum acted as a prominent node that could be easily influenced by other brain regions. In conclusion, our study identified widely affected regions with increased gray matter volume in individuals with regular soccer training. Furthermore, a temporal precedence relationship among these regions was observed.

Higher Regional Gray Matter Volume and White Matter Integrity in Individuals With Central Neuropathic Pain After Spinal Cord Injury.

Spinal cord injury (SCI) is a debilitating neurological condition that often leads to central neuropathic pain (CNP). As the fundamental mechanism of CNP is not fully established, its management is one of the most challenging problems among people with SCI. To shed more light on CNP mechanisms, the aim of this cross-sectional study was to compare the brain structure between individuals with SCI and CNP and those without CNP by examining the gray matter (GM) volume and the white matter (WM) integrity. Fifty-two individuals with SCI-28 with CNP and 24 without CNP-underwent a magnetic resonance imaging (MRI) session, including a T1-weighted scan for voxel-based morphometry, and a diffusion-weighted imaging (DWI) scan for WM integrity analysis, as measured by fractional anisotropy (FA) and mean diffusivity (MD). We found significantly higher GM volume in individuals with CNP compared with pain-free individuals in the right superior (p < 0.0014) and middle temporal gyri (p < 0.0001). Moreover, individuals with CNP exhibited higher WM integrity in the splenium of the corpus callosum (p < 0.0001) and in the posterior cingulum (p < 0.0001), compared with pain-free individuals. The results suggest that the existence of CNP following SCI is associated with GM and WM structural abnormalities in regions involved in pain intensification and spread, and which may reflect maladaptive neural plasticity in CNP.

Effects of Mindfulness-Based Interventions on Gray Matter Volume in Patients with Opioid Dependence

Introduction: Recently, several mindfulness-based programs showed promising clinical effects in the treatment of psychiatric disorders including substance use disorders. However, very little is known about the effects of mindfulness-based interventions (MBIs) on brain structure in such patients. Methods: This study aimed to detect changes in gray matter volume (GMV) in opioid-dependent patients receiving MBI during their first month of treatment. Thirty patients were assigned to either 3 weeks of MBI (n = 16) or treatment as usual (TAU, n = 14) and were investigated using structural magnetic resonance imaging before and after treatment. Longitudinal pipeline of the Computational Anatomy Toolbox for SPM (CAT12) was used to detect significant treatment-related changes over time. The identified GMV changes following treatment were related to clinically relevant measures such as impulsivity, distress tolerance, and mindfulness. Results: After treatment, increased mindfulness scores were found in individuals receiving MBI compared to TAU. In the MBI group, there were also significant differences with respect to distress tolerance and impulsivity. Effects on mindfulness, distress tolerance, and impulsivity were also found in the TAU group. Longitudinal within-group analysis revealed increased left anterior insula GMV in individuals receiving MBI. Anterior insula volume increase was associated with decreased impulsivity levels. In the TAU group, significant GMV changes were found in the right lingual gyrus and right entorhinal cortex. Discussion/Conclusion: MBI can yield significant clinical effects during early abstinence from opioid dependence. MBI is particularly associated with increased insula GMV, supporting an important role of this region in the context of MBI-induced neural changes.

Association of body mass index and its classifications with gray matter volume in individuals with a wide range of body mass index group: A whole-brain magnetic resonance imaging study.

AimTo examine the association of body mass index (BMI) [kg/m2] and its classifications (underweight [BMI < 18.5], normal [18.5 ≤ BMI < 25], overweight [25 ≤ BMI < 30], and obese [BMI ≥ 30]) with brain structure in individuals with a wide range of BMI group.Materials and methodsThe participants included 382 right-handed individuals (mean age: 46.9 ± 14.3 years, 142 men and 240 women). The intelligence quotient was assessed using the Japanese Adult Reading Test. Voxel-based morphometry (VBM) and diffusion tensor imaging (DTI) were performed to analyze the association of BMI and its classifications with gray and white matter structures, respectively.ResultsAccording to VBM, BMI was significantly and negatively correlated with the bilateral cerebellum exterior volumes. In group comparisons, the right cerebellum exterior volume was significantly lower in the overweight or obese group than in the underweight or normal group, while the bilateral cuneus and calcarine cortex, left cuneus, and left precuneus volume was significantly lower in the underweight group than in the non-underweight group. Sex-related stratification analyses for VBM revealed that BMI was significantly and negatively correlated with the bilateral cerebellum exterior volumes only in women. In group comparisons, the left cerebellum exterior volume was significantly lower in obese women than in non-obese women. The left thalamus proper and the right cerebellum exterior volumes were significantly lower in overweight or obese group than in underweight or normal group in men and women, respectively. The bilateral cuneus and calcarine cortex, left cuneus and carcarine cortex, and bilateral cuneus volume was significantly lower in underweight men than in non-underweight men. In contrast, there were no notable findings on DTI.ConclusionOur results suggest association of continuous BMI, being overweight or obese, and being underweight with decreased gray matter volume in individuals with a wide range of BMI group. Furthermore, sex-related differences are seen in the association of BMI and its classifications with regional gray matter volume reductions. Abnormally high or low BMIs may have a negative influence on regional gray matter volumes.

Gray Matter Brain Alterations in Type 1 Diabetes - Findings Based on Detailed Phenotyping of Neuropathy Status.

This study investigated brain structure in patients of type 1 diabetes with diabetic peripheral neuropathy (DPN) and type 1 diabetes with neuropathic pain and the associations to clinical, peripheral, and cognitive measurements. Sixty individuals with type 1 diabetes and 20 healthy controls were included in the study. Nineteen individuals with type 1 diabetes and neuropathic pain, 19 with type 1 diabetes and DPN, 18 with type 1 diabetes without DPN, and 20 healthy controls were included in the brain analyses. We utilized structural brain magnetic resonance imaging to investigate total and regional gray matter volume. Significant lower gray matter volume was found in type 1 diabetes with neuropathic pain and in type 1 diabetes without DPN compared to healthy controls (p=0.024 and p=0.019, respectively). Lower insula volume was observed in all three diabetes groups (all p≤0.050). Thalamus and hippocampus volume was lower in type 1 diabetes with neuropathic pain, cerebellum volume was lower in type 1 diabetes with DPN, and somatosensory cortex volume was lower in type 1 diabetes without DPN (all p≤0.018). Attenuated memory was associated with lower gray matter volume in type 1 diabetes with DPN. No associations were found between gray matter volume and clinical/peripheral measurements. We demonstrated lower gray matter volume in individuals with type 1 diabetes regardless of the presence of DPN and neuropathic pain. Hence, central gray matter alteration was not associated with peripheral alterations.

Longitudinal Changes in Resting-State Functional Connectivity and Gray Matter Volume Are Associated with Conversion to Hearing Impairment in Older Adults.

Hearing loss was recently identified as a modifiable risk factor for dementia although the potential mechanisms explaining this relationship are unknown. The current study examined longitudinal change in resting-state fMRI functional connectivity and gray matter volume in individuals who developed a hearing impairment compared to those whose hearing remained normal. This study included 440 participants from the UK Biobank: 163 who had normal hearing at baseline and impaired hearing at follow-up (i.e., converters, mean age = 63.11±6.33, 53% female) and 277 who had normal hearing at baseline and maintained normal hearing at follow-up (i.e., non-converters, age = 63.31±5.50, 50% female). Functional connectivity was computed between a priori selected auditory seed regions (left and right Heschl's gyrus and cytoarchitectonic subregions Te1.0, Te1.1, and Te1.2) and select higher-order cognitive brain networks. Gray matter volume within these same regions was also obtained. Converters had increased connectivity from left Heschl's gyrus to left anterior insula and from right Heschl's gyrus to right anterior insula, and decreased connectivity between right Heschl's gyrus and right hippocampus, compared to non-converters. Converters also had reduced gray matter volume in left hippocampus and left lateral visual cortex compared to non-converters. These findings suggest that conversion to a hearing impairment is associated with altered brain functional connectivity and gray matter volume in the attention, memory, and visual processing regions that were examined in this study.

Increased frontal gray matter volume in individuals with prodromal psychosis.

BackgroundBrain anatomical deficits associated with cognitive dysfunction have been reported in patients with schizophrenia. However, it remains unknown whether such anatomical deficits exist in individuals with prodromal psychosis. The present study is designed to investigate anatomical deficits in prodromal individuals and their associations with clinical/cognitive features.MethodsSeventy‐four prodromal individuals and seventy‐six healthy controls were scanned using structural magnetic resonance imaging. Support vector machines were applied to test whether anatomical deficits might be used to discriminate prodromal individuals from healthy controls.ResultsProdromal individuals showed significantly increased gray matter volume (GMV) in the right inferior frontal gyrus (IFG) and right rectus gyrus relative to healthy controls. No correlations were observed between increased GMV and clinical/cognitive characteristics. The combination of increased GMV in the right rectus gyrus and right IFG showed a sensitivity of 74.32%, a specificity of 67.11%, and an accuracy of 70.67% in differentiating prodromal individuals from healthy controls.ConclusionOur results provide evidence of increased frontal GMV in prodromal individuals. A combination of GMV values in the two frontal brain areas may serve as potential markers to discriminate prodromal individuals from healthy controls. The results thus highlight the importance of the frontal regions in the pathophysiology of psychosis.

Relationships between gray matter volume and reading ability in typically developing children, adolescents, and young adults

Reading is explicitly taught and foreshadows academic and vocational success. Studies comparing typical readers to those with developmental dyslexia have identified anatomical brain differences in bilateral temporo-parietal cortex, left temporo-occipital cortex, and bilateral cerebellum. Yet, it is unclear whether linear relationships exist between these brain structures and single real word reading ability in the general population. If dyslexia represents the lower end of the normal continuum, then relationships between gray matter volume (GMV) and reading ability would exist for all reading levels. Our study examined this question using voxel-based morphometry in a large sample (n = 404) of typically developing participants aged 6–22 derived from the NIH normative database. We tested correlations between individual GMV and single word reading and found none. After dividing this sample into groups based on age and on sex, we only found results in the group aged 15–22: positive correlations between GMV in left fusiform gyrus and reading, driven by females; and in right superior temporal gyrus in males. Multiple regressions also yielded no results, demonstrating that there is no general linear relationship between GMV and single real word reading ability. This provides an important context by which to interpret findings of GMV differences in dyslexia.

Increased Frontal Gray Matter Volume in Individuals with Ultra-High Risk for Psychosis

Background: Anatomical brain deficits have been reported in patients with schizophrenia. However, whether anatomical brain deficits exist in individuals with ultra-high risk (UHR) for psychosis remains unknown. The present study is designed to investigate structural changes in UHR individuals and their associations with clinical/cognitive features. Methods: Seventy-four UHR individuals and seventy-six healthy controls were scanned using structural MRI. Receiver operating characteristic curves and support vector machines were applied to test whether abnormal brain regions may be used to discriminate UHR individuals from healthy controls. Findings: UHR individuals showed significantly increased gray matter volume (GMV) in the right inferior frontal gyrus (IFG) and the right rectal gyrus relative to the healthy controls. No significant correlations were observed between abnormal GMV and clinical characteristics. ROC analyses showed that the under curve area of the GMV values in the right rectal gyrus was 0·717 with sensitivity and that specificity values were 66·22% and 72·37%.The combination of increased GMV in the right rectal gyrus and right IFG showed a sensitivity of 74·32%, a specificity of 67·11%, and an accuracy of 70·67% in differentiating UHR individuals from healthy controls. Interpretation: Our results provide evidence of increased frontal GMV in UHR individuals. The GMV values in the right rectal gyrus might serve as potential biomarkers to discriminate UHR individuals from controls at an individual level. Moreover, the combination of GMV in the two frontal brain areas may be a potential image marker to differentiate UHR from healthy controls. The results thus highlight the importance of the frontal regions in the pathophysiology of psychosis. Reduction of Interhemispheric Functional Brain Connectivity in Early Blindness: A Resting-State fMRI Study.

Objective The purpose of this study was to investigate the resting-state interhemispheric functional connectivity in early blindness by using voxel-mirrored homotopic connectivity (VMHC). Materials and Methods Sixteen early blind patients (EB group) and sixteen age- and gender-matched sighted control volunteers (SC group) were recruited in this study. We used VMHC to identify brain areas with significant differences in functional connectivity between different groups and used voxel-based morphometry (VBM) to calculate the individual gray matter volume (GMV). Results VMHC analysis showed a significantly lower connectivity in primary visual cortex, visual association cortex, and somatosensory association cortex in EB group compared to sighted controls. Additionally, VBM analysis revealed that GMV was reduced in the left lateral calcarine cortices in EB group compared to sighted controls, while it was increased in the left lateral middle occipital gyri. Statistical analysis showed the duration of blindness negatively correlated with VMHC in the bilateral middle frontal gyri, middle temporal gyri, and inferior temporal gyri. Conclusions Our findings help elucidate the pathophysiological mechanisms of EB. The interhemispheric functional connectivity was impaired in EB patients. Additionally, the middle frontal gyri, middle temporal gyri, and inferior temporal gyri may be potential target regions for rehabilitation.

Gray Matter Increase in Motor Cortex in Pediatric ADHD: A Voxel-Based Morphometry Study

Objective: Several studies report that ADHD is associated with reduced gray matter (GM), whereas others report no differences in GM volume between ADHD patients and controls, and some even report more GM volume in individuals with ADHD. These conflicting findings suggest that reduced GM is not a universal finding in ADHD, and that more research is needed to delineate with greater accuracy the range of GM alterations. Method: The present study aimed to identify GM alterations in ADHD using pediatric templates. 19 drug-naïve ADHD patients and 18 controls, all aged 7 to 14 years, were scanned using magnetic resonance imaging. Results: Relative to the controls, the ADHD patients had more GM, predominantly in the precentral and supplementary motor areas. Moreover, there were positive correlations between GM volume in these areas and ADHD scale scores. Conclusion: The clinical and pathophysiological significance of increased GM in the motor areas remains to be elucidated by additional research.

Changes of individual BrainAGE during the course of the menstrual cycle

Brain morphology varies during the course of the menstrual cycle, with increases in individual gray matter volume at the time of ovulation. This study implemented our previously presented BrainAGE framework to analyze short-term neuroanatomical changes in healthy young women due to hormonal changes during the menstrual cycle. The BrainAGE approach determines the complex multidimensional aging pattern within the whole brain by applying established kernel regression methods to anatomical brain MRIs. The “Brain Age Gap Estimation” (i.e., BrainAGE) score is then calculated as the difference between chronological age and estimated brain age. Eight women (21–31years) completed three to four MRI scans during their menstrual cycle (i.e., at (t1) menses, (t2) time of ovulation, (t3) midluteal phase, (t4) next menses). Serum levels of estradiol and progesterone were evaluated at each scanning session.Individual BrainAGE scores significantly differed during the course of the menstrual cycle (p<0.05), with a significant decrease of −1.3years at ovulation (p<0.05). Moreover, higher estradiol levels significantly correlated with lower BrainAGE scores (r=−0.42, p<0.05). In future, the BrainAGE approach may serve as a sensitive as well as easily implementable tool to further explore the short-term and maybe long-term effects of hormones on brain plasticity and its modulating effects in lifestyle-related diseases and dementia.

Poster #S59 DECREASED GREY MATTER VOLUME AS THE EXPRESSION OF DIFFERENTIAL SENSITIVITY TO ENVIRONMENTAL RISK EXPOSURE IN PSYCHOTIC DISORDER

Background: Cannabis use, childhood trauma and urban upbringing are important risk factors for psychotic disorder. Differential sensitivity to these environmental exposures may be expressed as structural brain alterations. The aim of this study was to examine whether cannabis use, childhood trauma and urban upbringing are associated with alterations in gray matter volume (GMV) and whether this is dependent on different genetic risk levels for psychotic disorder. Methods: T1-weighted MRI scans were acquired from 89 patients with a psychotic disorder, 98 healthy siblings of patients with psychotic disorder and 87 controls. Freesurfer software was used to measure GMV. Cannabis use was assessed with the Composite International Diagnostic Interview and a urine test. Childhood trauma was measured with the Childhood Trauma Questionnaire Short Form. The developmental urbanicity exposure comprised 5 levels, reflecting the average population density between birth and the 15th birthday. Multilevel random regression analyses were used to examine the association between group and environment (as well as their interaction) on the one hand and GMV as the dependent variable on the other. The three-way interaction group x environment x sex was also investigated. Results: There were significant main effects of group (B=−10.44, p=0.04) and cannabis (B=−8.18, p<0.05) on GMV, which was not the case for childhood trauma and urbanicity. Both the two-way interaction between group and cannabis (χ2=5.31, p=0.07) and between group and childhood trauma (χ2=10.32, p=0.01) in the model of GMV were significant. Stratified analyses showed that cannabis use and childhood trauma were associated with lower GMV in the patient group (cannabis use: B=−14.89, p=0.03; childhood trauma: B linear trend = −8.58, p=0.04). Although three way interactions (group x cannabis or childhood trauma x sex) were inconclusive, environmental effects on GMV were found in male patients (cannabis: B=−24.86, p<0.01; childhood trauma: B linear trend = −12.16, p=0.02) and not in female patients. The two-way interaction between group and developmental urbanicity was not significant (χ2=1.79, p=0.41). Discussion: The findings suggest that exposure to cannabis and childhood trauma, but not urban upbringing, may impact global GMV in individuals at the highest genetic risk level for psychotic disorder. Thus, patients may be more sensitive to cannabis and childhood trauma, expressed as a reduction in GMV, than individuals at higher than average (siblings) or average (controls) genetic risk. This increased cerebral sensitivity to cannabis and trauma may be confined to male patients.

A two-level multimodality imaging Bayesian network approach for classification of partial epilepsy: preliminary data.

Quantitative neuroimaging analyses have demonstrated gray and white matter abnormalities in group comparisons of different types of non-lesional partial epilepsy. It is unknown to what degree these type-specific patterns exist in individual patients and if they could be exploited for diagnostic purposes. In this study, a two-level multi-modality imaging Bayesian network approach is proposed that uses information about individual gray matter volume loss and white matter integrity to classify non-lesional temporal lobe epilepsy with (TLE-MTS) and without (TLE-no) mesial-temporal sclerosis and frontal lobe epilepsy (FLE). 25 controls, 19 TLE-MTS, 22 TLE-no and 14 FLE were studied on a 4T MRI and T1 weighted structural and DTI images acquired. Spatially normalized gray matter (GM) and fractional anisotropy (FA) abnormality maps (binary maps with voxels 1 SD below control mean) were calculated for each subject. At the first level, each group's abnormality maps were compared with those from all the other groups using Graphical-Model-based Morphometric Analysis (GAMMA). GAMMA uses a Bayesian network and a Markov random field based contextual clustering method to produce maps of voxels that provide the maximal distinction between two groups and calculates a probability distribution and a group assignment based on this information. The information was then combined in a second level Bayesian network and the probability of each subject to belong to one of the three epilepsy types calculated. The specificities of the two level Bayesian network to distinguish between the three patient groups were 0.87 for TLE-MTS and TLE-no and 0.86 for FLE, the corresponding sensitivities were 0.84 for TLE-MTS, 0.72 for TLE-no and 0.64 for FLE. The two-level multi-modality Bayesian network approach was able to distinguish between the three epilepsy types with a reasonably high accuracy even though the majority of the images were completely normal on visual inspection.

Variability of (functional) hemodynamics as measured with simultaneous fNIRS and fMRI during intertemporal choice

Neural processing inferred from hemodynamic responses measured with functional near infrared spectroscopy (fNIRS) may be confounded with individual anatomical or systemic physiological sources of variance. This may hamper the validity of fNIRS signal interpretations and associations between individual traits and brain activation, such as the link between impulsivity-related personality traits and decreased prefrontal cognitive control during reward-based decision making. Hemodynamic responses elicited by an intertemporal choice reward task in 20 healthy subjects were investigated for multimodal correlations of simultaneous fNIRS-fMRI and for an impact of anatomy and scalp fMRI signal fluctuations on fNIRS signals. Moreover, correlations of prefrontal activation with trait "sensitivity to reward" (SR) were investigated for differences between methods. While showing substantial individual variability, temporal fNIRS-fMRI correlations increased with the activation, which both methods consistently detected within right inferior/middle frontal gyrus. Here, up to 41% of fNIRS channel activation variance was explained by individual gray matter volume simulated to be reached by near-infrared light, and up to 20% by scalp-cortex distance. Extracranial fMRI and fNIRS time series showed significant temporal correlations in the temple region. SR was negatively correlated with fMRI but not fNIRS activation elicited by immediate rewards of choice within right inferior/middle frontal gyrus. Higher SR increased the correlation between extracranial fMRI and fNIRS signals and decreased fNIRS-fMRI correlations. Task-related fNIRS signals might be impacted by regionally and individually weighted sources of anatomical and systemic physiological error variance. Trait-activation correlations might be affected or biased by systemic physiological responses, which should be accounted for in future fNIRS studies of interindividual differences.

Morphometric gray matter differences of the medial frontal cortex influence the social Simon effect

Interacting with others plays a fundamental role in human life. Although several brain regions have recently been associated with complex cognitive control processes, surprisingly little is known about the structural correlates underlying cognitive control processes involved in social interactions. In the present study we used gray matter voxel-based morphometry (VBM) to investigate structural brain correlates of individual performance differences in a social Simon task. Here, two people share a Simon task, which requires each participant to respond to only one of two possible stimuli, rendering the paradigm a go-nogo task, so that a Simon effect - known as the Social Simon Effect (SSE) - is observable across both participants. Using a whole brain approach, we found that inter-individual differences in the SSE are negatively correlated with gray matter (GM) volume of the medial frontal cortex (MFC). The present data indicate that individuals with larger MFC GM volume were those with better conflict resolution in a social Simon task and vice versa. This brain-behavior relationship between cognitive control processes and individual GM volume differences might help to improve our understanding of social interactions in joint task performance.

Substance use and regional gray matter volume in individuals at high risk of psychosis

Individuals with an at risk mental state (ARMS) are at greatly increased risk of developing a psychotic illness. Risk of transition to psychosis is associated with regionally reduced cortical gray matter volume. There has been considerable interest in the interaction between psychosis risk and substance use. In this study we investigate the relationship between alcohol, cannabis and nicotine use with gray matter volume in ARMS subjects and healthy volunteers. Twenty seven ARMS subjects and 27 healthy volunteers took part in the study. All subjects underwent volumetric MRI imaging. The relationship between regional gray matter volume and cannabis use, smoking, and alcohol use in controls and ARMS subjects was analysed using voxel-based morphometry. In any region where a significant relationship with drug was present, data were analysed to determine if there was any group difference in this relationship. Alcohol intake was inversely correlated with gray matter volume in cerebellum, cannabis intake was use was inversely correlated with gray matter volume in prefrontal cortex and tobacco intake was inversely correlated with gray matter volume in left temporal cortex. There were no significant interactions by group in any region. There is no evidence to support the hypothesis of increased susceptibility to harmful effects of drugs and alcohol on regional gray matter in ARMS subjects. However, alcohol, tobacco and cannabis at low to moderate intake may be associated with lower gray matter in both ARMS subjects and healthy volunteers—possibly representing low-level cortical damage or change in neural plasticity.
