Holographic fiducial marks: A system for precise alignment of x-ray optics

This Letter covers the design and implementation of a generalizable system for the precise alignment of X-ray gratings. Next-generation high-energy grating-based Differential Phase Contrast (gDPC) X-ray imaging systems require precise alignment of the X-ray gratings as low as 1 mrad in rotation and 0.5 mm in translation. In this work, we designed holographic fiducial marks, consisting of four reflective Fresnel zone plates, each placed in a separate quadrant of the mark. When illuminated with a collimated laser beam, each mark creates a predefined pattern of four points, which changes quantitatively for any misalignment in each of the three translational and three rotational degrees of freedom. We fabricated the designed fiducial marks using photolithography and etching processes. The experimental system is implemented using a HeNe laser and an optical imaging system, which includes a beam expander, a plate beam splitter, and a CMOS camera, suitable for aligning practical gratings in gDPC X-ray imaging systems. Our experimental results demonstrate the rotational precision capabilities of the reported alignment system down to 0.42 mrad around the optical axis and 0.03 mrad around the axes perpendicular to the optical axis. The translational precision of 83.64 μm along the optical axis and 1.22 μm along the axes perpendicular to the optical axis is also demonstrated.

Evaluation of differential phase contrast cone beam CT imaging system.

Grating-based differential phase contrast (DPC) imaging enables the use of a hospital-grade X-ray tube, but compromises the image quality due to insufficiently coherent illumination. In this research, a bench-top DPC cone beam CT (DPC-CBCT) was systematically evaluated and compared with the traditional attenuation-based CBCT in terms of contrast to noise ratio, noise property, and contrast resolution through phantom studies. In order to evaluate DPC-CBCT for soft tissue imaging, breast specimen and small animal studies were carried out. Phantom studies indicate that phase image has lower-frequency noise, higher CNR, and improved contrast resolution. However, phase image quality was degraded in soft tissue imaging due to coherence loss caused by small-angle scattering. Hence dark-field imaging was introduced to quantitatively investigate small-angle scattering caused by an object. Experimental results indicate that inhomogeneous objects affect phase contrast imaging, phase image is more sensitive to noise, and its performance is material dependent. Dark-field imaging could also be used to locate and reduce phase image noise and artifact caused by small-angle scattering.

Interior Tomography from Differential Phase Contrast Data via Hilbert Transform Based on Spline Functions.

X-ray phase contrast imaging is an important mode due to its sensitivity to subtle features of soft biological tissues. Grating-based differential phase contrast (DPC) imaging is one of the most promising phase imaging techniques because it works with a normal x-ray tube of a large focal spot at a high flux rate. However, a main obstacle before this paradigm shift is the fabrication of large-area gratings of a small period and a high aspect ratio. Imaging large objects with a size-limited grating results in data truncation which is a new type of the interior problem. While the interior problem was solved for conventional x-ray CT through analytic extension, compressed sensing and iterative reconstruction, the difficulty for interior reconstruction from DPC data lies in that the implementation of the system matrix requires the differential operation on the detector array, which is often inaccurate and unstable in the case of noisy data. Here, we propose an iterative method based on spline functions. The differential data are first back-projected to the image space. Then, a system matrix is calculated whose components are the Hilbert transforms of the spline bases. The system matrix takes the whole image as an input and outputs the back-projected interior data. Prior information normally assumed for compressed sensing is enforced to iteratively solve this inverse problem. Our results demonstrate that the proposed algorithm can successfully reconstruct an interior region of interest (ROI) from the differential phase data through the ROI.

A novel crystal-analyzer phase retrieval algorithm and its noise property.

A description of the rocking curve in diffraction enhanced imaging (DEI) is presented in terms of the angular signal response function and a simple multi-information retrieval algorithm based on the cosine function fitting. A comprehensive analysis of noise properties of DEI is also given considering the noise transfer characteristic of the X-ray source. The validation has been performed with synchrotron radiation experimental data and Monte Carlo simulations based on the Geant4 toolkit combined with the refractive process of X-rays, which show good agreement with each other. Moreover, results indicate that the signal-to-noise ratios of the refraction and scattering images are about one order of magnitude better than that of the absorption image at the edges of low-Z samples. The noise penalty is drastically reduced with the increasing photon flux and visibility. Finally, this work demonstrates that the analytical method can build an interesting connection between DEI and GDPCI (grating-based differential phase contrast imaging) and is widely suitable for a variety of measurement noise in the angular signal response imaging prototype. The analysis significantly contributes to the understanding of noise characteristics of DEI images and may allow improvements to the signal-to-noise ratio in biomedical and material science imaging.

A Study on Mastectomy Samples to Evaluate Breast Imaging Quality and Potential Clinical Relevance of Differential Phase Contrast Mammography

Differential phase contrast and scattering-based x-ray mammography has the potential to provide additional and complementary clinically relevant information compared with absorption-based mammography. The purpose of our study was to provide a first statistical evaluation of the imaging capabilities of the new technique compared with digital absorption mammography. We investigated non-fixed mastectomy samples of 33 patients with invasive breast cancer, using grating-based differential phase contrast mammography (mammoDPC) with a conventional, low-brilliance x-ray tube. We simultaneously recorded absorption, differential phase contrast, and small-angle scattering signals that were combined into novel high-frequency-enhanced images with a dedicated image fusion algorithm. Six international, expert breast radiologists evaluated clinical digital and experimental mammograms in a 2-part blinded, prospective independent reader study. The results were statistically analyzed in terms of image quality and clinical relevance. The results of the comparison of mammoDPC with clinical digital mammography revealed the general quality of the images to be significantly superior (P < 0.001); sharpness, lesion delineation, as well as the general visibility of calcifications to be significantly more assessable (P < 0.001); and delineation of anatomic components of the specimens (surface structures) to be significantly sharper (P < 0.001). Spiculations were significantly better identified, and the overall clinically relevant information provided by mammoDPC was judged to be superior (P < 0.001). Our results demonstrate that complementary information provided by phase and scattering enhanced mammograms obtained with the mammoDPC approach deliver images of generally superior quality. This technique has the potential to improve radiological breast diagnostics.

Grating-based phase contrast tomosynthesis imaging: proof-of-concept experimental studies.

This paper concerns the feasibility of x-ray differential phase contrast (DPC) tomosynthesis imaging using a grating-based DPC benchtop experimental system, which is equipped with a commercial digital flat-panel detector and a medical-grade rotating-anode x-ray tube. An extensive system characterization was performed to quantify its imaging performance. The major components of the benchtop system include a diagnostic x-ray tube with a 1.0 mm nominal focal spot size, a flat-panel detector with 96 μm pixel pitch, a sample stage that rotates within a limited angular span of ± 30°, and a Talbot-Lau interferometer with three x-ray gratings. A total of 21 projection views acquired with 3° increments were used to reconstruct three sets of tomosynthetic image volumes, including the conventional absorption contrast tomosynthesis image volume (AC-tomo) reconstructed using the filtered-backprojection (FBP) algorithm with the ramp kernel, the phase contrast tomosynthesis image volume (PC-tomo) reconstructed using FBP with a Hilbert kernel, and the differential phase contrast tomosynthesis image volume (DPC-tomo) reconstructed using the shift-and-add algorithm. Three inhouse physical phantoms containing tissue-surrogate materials were used to characterize the signal linearity, the signal difference-to-noise ratio (SDNR), the three-dimensional noise power spectrum (3D NPS), and the through-plane artifact spread function (ASF). While DPC-tomo highlights edges and interfaces in the image object, PC-tomo removes the differential nature of the DPC projection data and its pixel values are linearly related to the decrement of the real part of the x-ray refractive index. The SDNR values of polyoxymethylene in water and polystyrene in oil are 1.5 and 1.0, respectively, in AC-tomo, and the values were improved to 3.0 and 2.0, respectively, in PC-tomo. PC-tomo and AC-tomo demonstrate equivalent ASF, but their noise characteristics quantified by the 3D NPS were found to be different due to the difference in the tomosynthesis image reconstruction algorithms. It is feasible to simultaneously generate x-ray differential phase contrast, phase contrast, and absorption contrast tomosynthesis images using a grating-based data acquisition setup. The method shows promise in improving the visibility of several low-density materials and therefore merits further investigation.

Preliminary comparison of grating-based and in-line phase contrast X-ray imaging with synchrotron radiation for mouse kidney at TOMCAT

Phase contrast imaging has been demonstrated to be advantageous in revealing detailed structures inside biological specimens without contrast agents. Grating-based differential phase contrast (DPC) and in-line phase contrast (ILPC) X-ray imaging are the two modalities frequently used at the beamline of TOmographic Microscopy and Coherent rAdiology experimenTs (TOMCAT) at the Swiss Light Source (SLS). In this paper, we preliminarily compared the abilities of two types of phase contrast imaging in distinguishing micro structures in mouse kidneys. The 3D reconstructions showed that the microstructures in kidney, such as micro vessels and renal tubules, were displayed clearly with both imaging modalities. The two techniques may be viewed as complementary. For larger features with very small density variations DPC is the desirable method. In cases where dose and time limits may prohibit the multiple steps required for DPC, and when the focus is on finer features, the ILPC method may be considered as a more viable alternative. Moreover, high resolution ILPC images are comparable with histological results.

Fundamental relationship between the noise properties of grating-based differential phase contrast CT and absorption CT: Theoretical framework using a cascaded system model and experimental validation

Using a grating interferometer, a conventional x-ray cone beam computed tomography (CT) data acquisition system can be used to simultaneously generate both conventional absorption CT (ACT) and differential phase contrast CT (DPC-CT) images from a single data acquisition. Since the two CT images were extracted from the same set of x-ray projections, it is expected that intrinsic relationships exist between the noise properties of the two contrast mechanisms. The purpose of this paper is to investigate these relationships. First, a theoretical framework was developed using a cascaded system model analysis to investigate the relationship between the noise power spectra (NPS) of DPC-CT and ACT. Based on the derived analytical expressions of the NPS, the relationship between the spatial-frequency-dependent noise equivalent quanta (NEQ) of DPC-CT and ACT was derived. From these fundamental relationships, the NPS and NEQ of the DPC-CT system can be derived from the corresponding ACT system or vice versa. To validate these theoretical relationships, a benchtop cone beam DPC-CT/ACT system was used to experimentally measure the modulation transfer function (MTF) and NPS of both DPC-CT and ACT. The measured three-dimensional (3D) MTF and NPS were then combined to generate the corresponding 3D NEQ. Two fundamental relationships have been theoretically derived and experimentally validated for the NPS and NEQ of DPC-CT and ACT: (1) the 3D NPS of DPC-CT is quantitatively related to the corresponding 3D NPS of ACT by an inplane-only spatial-frequency-dependent factor 1∕f (2), the ratio of window functions applied to DPC-CT and ACT, and a numerical factor C(g) determined by the geometry and efficiency of the grating interferometer. Note that the frequency-dependent factor is independent of the frequency component f(z) perpendicular to the axial plane. (2) The 3D NEQ of DPC-CT is related to the corresponding 3D NEQ of ACT by an f (2) scaling factor and numerical factors that depend on both the attenuation and refraction properties of the image object, as well as C(g) and the MTF of the grating interferometer. The performance of a DPC-CT system is intrinsically related to the corresponding ACT system. As long as the NPS and NEQ of an ACT system is known, the corresponding NPS and NEQ of the DPC-CT system can be readily estimated using additional characteristics of the grating interferometer.

Imaging brain amyloid deposition using grating-based differential phase contrast tomography

One of the core pathological features of Alzheimer's disease (AD) is the accumulation of amyloid plaques in the brain. Current efforts of medical imaging research aim at visualizing amyloid plaques in living patients in order to evaluate the progression of the pathology, but also to facilitate the diagnosis of AD at the prodromal stage. In this study, we evaluated the capabilities of a new experimental imaging setup to image amyloid plaques in the brain of a transgenic mouse model of Alzheimer's disease. This imaging setup relies on a grating interferometer at a synchrotron X-ray source to measure the differential phase contrast between brain tissue and amyloid plaques. It provides high-resolution images with a large field of view, making it possible to scan an entire mouse brain. Here, we showed that this setup yields sufficient contrast to detect amyloid plaques and to quantify automatically several important structural parameters, such as their size and their regional density in 3D, on the scale of a whole mouse brain. Whilst future developments are required to apply this technique in vivo, this grating-based setup already gives the possibility to perform powerful studies aiming at quantifying the amyloid pathology in mouse models of AD and might accelerate the evaluation of anti-amyloid compounds. In addition, this technique may also facilitate the development of other amyloid imaging methods such as positron emission tomography (PET) by providing convenient high-resolution 3D data of the plaque distribution for multimodal comparison.

WE-G-211-08: Performance Study of Regularized Phase Retrieval from X-Ray Differential Phase Contrast Projection Image

Purpose: X-ray differential phase contrast (DPC) projection imaging has shown promise for superior image contrast. In the field of medical imaging, however, its differential nature makes the projection image difficult to use for direct diagnosis. In this study, a phase retrieval method is introduced to generate phase images, which are compared with absorption images to demonstrate the benefits of phase contrast projection imaging. Methods: The method involves the minimization of a cost function that includes two terms: a quadratic norm that enforces consistency between the phase image and the DPC image, and a nonconvex norm of the phase imageˈs gradient in the direction perpendicular to the stripes. A weighting parameter α is applied to the second term to tailor the content of stripes. This cost function is minimized to solve for the phase image. A grating-based DPC system was used to acquire the DPC and absorption images of a cylindrical phantom. The DPC, absorption, directly-integrated phase and regularized phase images were compared. To evaluate the image quality, simulated spheres of different contrasts and diameters were added to the experimental data. CNR of the beads in the integrated image was used as the figure of merit to optimize the regularization. Results: The retrieved phase image provides complementary information of the phantom that is otherwise lost in the direct-integrated images because of the presence of stripe noise. Regularized integration improved image quality in terms of CNR and the content of stripe noise. The optimal regularization parameter can be determined from the peaks of the CNR-α curves and is dependent on the objectˈs contrast. Conclusions: Regularized integration allows for artifact-free phase retrieval from DPC images and a fair comparison between phase contrast and absorption images. DPC imagingˈs potential application in medical imaging is highlighted by the complementary information provided by the phase image.

Noise analysis of grating-based x-ray differential phase contrast imaging

The sensitivity of x-ray radiographic images, meaning the minimal detectable change in the thickness or in the index of refraction of a sample, is directly related to the uncertainty of the measurement method. In the following work, we report on the recent development of quantitative descriptions for the stochastic error of grating-based differential phase contrast imaging (DPCi). Our model includes the noise transfer characteristics of the x-ray detector and the jitter of the phase steps. We find that the noise in DPCi depends strongly on the phase stepping visibility and the sample properties. The results are supported by experimental evidence acquired with our new instrument with a field of view of 50x70 mm(2). Our conclusions provide general guidelines to optimize grating interferometers for specific applications and problems.

Advanced phase-contrast imaging using a grating interferometer

Phase-sensitive X-ray imaging methods can provide substantially increased contrast over conventional absorption-based imaging, and therefore new and otherwise inaccessible information. Differential phase-contrast (DPC) imaging, which uses a grating interferometer and a phase-stepping technique, has been integrated into TOMCAT, a beamline dedicated to tomographic microscopy and coherent radiology experiments at the Swiss Light Source. Developments have been made focusing on the fast acquisition and post-processing of data to enable a high-throughput of samples, with obvious advantages, also through increasing the efficiency of the detecting system, of helping to reduce radiation dose imparted to the sample. A novel aquarium design allows a vertical rotation axis below the sample with measurements performed in aqueous environment. Optimization of the data acquisition procedure enables a full phase volume (1024 x 1024 pixels x 1000 projections x 9 phase steps, i.e. 9000 projections in total) to be acquired in 20 min (with a pixel size of 7.4 microm), and the subsequent post-processing has been integrated into the beamline pipeline for sinogram generation. Local DPC tomography allows one to focus with higher magnification on a particular region of interest of a sample without the presence of local tomography reconstruction artifacts. Furthermore, 'widefield' imaging is shown for DPC scans for the first time, enabling the field of view of the imaging system to be doubled for samples that are larger than the magnification allows. A case study is illustrated focusing on the visualization of soft tissue features, and particularly the substantia nigra of a rat brain. Darkfield images, based on local X-ray scattering, can also be extracted from a grating-based DPC scan: an example of the advantages of darkfield contrast is shown and the potential of darkfield X-ray tomography is discussed.
