Design and synthesis of glycolic and mandelic acid derivatives as factor Xa inhibitors

A series of glycolic and mandelic acid derivatives was synthesized and investigated for their factor Xa inhibitory activity. These analogues are highly potent and selective inhibitors against fXa. In a rabbit deep vein thrombosis model, compound 26 showed significant antithrombotic effects (81% inhibition of thrombus formation) at 1.1 μM plasma concentration following intravenous administration.

New access to fluorinated glycolic acid derivatives from trifluoropyruvamides

Trifluoropyruvamide in their hydrates forms participate in various Friedel-Crafts type reactions with electron rich aromatic compounds. Good diastereoisomeric excess have been obtained starting from the chiral isobornylamine derivative.

Copper catalyzed chlorine transfer cyclizations of glycine and glycolic acid derivatives

Chloroglycine and chloroglycolic acid derivatives smoothly cyclize in the presence of a catalytic amount of cuprous chloride and 2,2′-bipyridine to produce functionalized proline and 2-tetrahydrofurancarboxylic esters, respectively, in an atom transfer process.

Preformulation Prodrug Research - Chemical and Enzymatic Hydrolysis Kinetics of the Glycerol, Glycolic Acid and Morpholino Ethyl Ester Derivatives of a Developmental Analgesic Agent (RS-82917)

The chemical and enzymatic hydrolysis of the glycerol-(2), glycolic acid-(3), and morpholino ethyl-(4) ester derivatives of 7-(4-methylthiobenzoyl) benzofuran-5-yl-acedtic acid (RS-82917,1) were evaluated for potential application as prodrugs. In aqueous solution with pH from 1-10, all three ester derivatives hydrolyzed quantitatively to 1. Rate expressions were derived from the log(rate) pH profiles for each ester, and pH-dependent intramolecular catalysis was proposed to account for the hydrolysis kinetics of all three esters. None of the ester aqueous solutions can provide two-year shelf-life at 25°C. The rate of hydrolysis was accelerated significantly by human and mouse plasma enzymes for 2 and 4 and by mouse liver homogenate for 2, 3 and 4.

Erucoyl Glycolates as Lubricants for Cotton Yarns

Long-chain fatty acid derivatives of glycolic acid were evaluated as yarn lubricants for the preparation of cotton knitted fabrics. All experimental compounds imparted excellent lubricity, as measured by coefficient of friction, to both bleached and dyed cotton yarns. Yarns treated with polymeric sizing agents then topped with these compounds also exhibited improved lubricity. These treatments, therefore, have potential for greater efficiency in weaving as well as knitting fabrics. Frictional prop erties of these glycolic acid derivatives were among the best compared to commer cially available yarn lubricants.

Prodrugs as drug delivery systems. 68. Chemical and plasma-catalyzed hydrolysis of various esters of benzoic acid: a reference system for designing prodrug esters of carboxylic acid agents

The hydrolysis of a series of esters of benzoic acid including various glycolic acid derivatives was studied in alkaline solution and in human plasma at 37 ° C. For the hydroxide ion-catalyzed hydrolysis a linear free-energy relationship between log kOH and the Taft polar parameter σ ∗ for the substituents in the alcohol portion was derived. The linear correlation equation obtained covered esters with a 100-fold variation in reactivity. All esters hydrolyzed to give benzoic acid except for various benzoylglycolic acid esters which predominantly hydrolyzed to give benzoylglycolic acid. The susceptibility of the ester derivatives to undergo enzyme-catalyzed hydrolysis by human plasma was strongly influenced by the structure of the alcohol moiety and was unrelated to the chemical reactivity of the compounds. Among the alkyl esters the ethyl ester showed the least enzymatic lability whereas the choline, N,N-dimethylaminoethanol and N,N-dimethylglycolamide esters were hydrolyzed extremely rapidly. The benzoylglycolic acid esters were predominantly, although incompletely, cleaved to benzoic acid by plasma enzymes. The results obtained were discussed in relation to design of ester prodrugs of carboxylic acid agents.

Preparation of some fatty glycolic acid derivatives and screening for antimicrobial activity

AbstractNine glycolic acid derivatives were prepared and screened as primary plasticizers for polyvinyl chloride (PVC) and for antimicrobial activity. Two types of compounds were represented: (a) those with a methyl or ethyl ester grouping at the carboxyl function of glycolic acid and a fatty acyl group at the hydroxyl function; and (b) those with three acyl groups attached at the hydroxyl functions of the amide, resulting from the reaction of glycolic acid and diethanolamine. None of the compounds was compatible with PVC at the 35% level of incorporation, and the compounds were not further evaluated as plasticizers. They were screened for antimicrobial activity against a gram‐positive bacterium,Staphylococcus aureus; a gram‐negative bacterium,Escherichia coli; a yeast,Candida utilis; and a mold,Penicillium sp. Six of the compounds inhibited two or more of the test organisms suggesting some of these materials merit further testing as biostatic agents.

Halovinylene Carbonates in Organic Synthesis. New synthetic methods (2)

AbstractMonohalo‐ and dihalovinylene carbonates constitute a new class of cyclophiles which permit simultaneous introduction of masked α‐hydroxyketo and α‐diketo functions, respectively, into the cycloadducts. Demasking can be performed by simple hydrolysis. Solvolytic opening of the carbonate ring leads to glycolic acid derivatives in the case of the monohalo compounds and to glyoxylic acid derivatives with the dihalo compounds. Preparation of the title compounds, their potential as synthetic reagents, and the chemistry of their simple reaction products are surveyed from a preparative viewpoint.

A SPECIFIC AND SENSITIVE METHOD FOR THE DETERMINATION OF BENZILIC ACID

Abstract A specific, sensitive and relatively simple procedure is described for the quantitative analysis of benzilic acid. The method is based on α-naphthofuchsone formation obtained by condensation of benzilic acid with α-naphthol in the presence of concentrated sulfuric acid and measuring the resultant hydroxytriaryl methyl cation at 558 mμ. No interference was observed with other glycolic acid derivatives or oxygen-bearing compounds. As little as 5 × 10−6 M benzilic acid is determinable.

Role of adrenergic mechanisms in the development of motor excitation in rats caused by benactyzine

An investigation was made of behavior reactions of rats under the effects of benactyzine and phenamine, along with the influence exerted by adrenergic blocking agents upon the motor excitation, produced by these preparations. Chlorpromazine was effective in fully eliminating disturbed behavior reactions produced by benactyzine, dihydroergotamine, benzolin (prescolin) while sympatholytin—did this only partially. Under the effect of amphetamine all of the adrenergic blocking agents were capable of restoring the latent reflex period; but stereotype reactions, produced by the action of amphetamine, could be abolished only by chlorpromazine. The above data prove that these disturbances appearing under the effect of the glycolic acid derivatives are caused by the activation of adrenergic mechanisms of the brain.

SOME ANTAGONISTS OF ATROPINE-LIKE PSYCHOTOMIMETICS.

Abstract The peripheral pharmacological effects of an anti-acetylcholine psychotomimetic drug, ethylpiperidylcyclopentylphenyl glycolate hydrochloride (JB-329) were found to be essentially similar to those of atropine. Both drugs inhibited parasympathetic effects and acetylcholine responses and produced potentiation of the pressor responses to adrenaline and noradrenaline. The compound tetrahydroaminoacridine, shown to be an antagonist of JB-329, was studied and its actions were compared with those of other cholinesterase inhibitors. Certain glycolic acid derivatives, some of which have been shown to antagonise the effects of JB-329, were also studied and their pharmacology and interactions are presented. The mode of action of the antagonists are discussed.
