Asparagine sustains cellular proliferation and c‑Myc expression in glutamine‑starved cancer cells.

During tumorigenesis, oncogene activation and metabolism rewiring are interconnected. Activated c‑Myc upregulates several genes involved in glutamine metabolism, making cancer cells dependent on high levels of this amino acid to survive and proliferate. After studying the response to glutamine deprivation in cancer cells, it was found that glutamine starvation not only blocked cellular proliferation, but also altered c‑Myc protein expression, leading to a reduction in the levels of the canonical c‑Myc isoform and an increase in the expression of c‑Myc1, a c‑Myc isoform translated from an in‑frame 5' CUG codon. In an attempt to identify nutrients able to counteract glutamine deprivation effects, it was shown that, in the absence of glutamine, asparagine permitted cell survival and proliferation, and maintained c‑Myc expression as in glutamine‑fed cells, with high levels of canonical c‑Myc and c‑Myc1 almost undetectable. In asparagine‑fed cells, global protein translation was higher than in glutamine‑starved cells, and there was an increase in the levels of glutamine synthetase (GS), whose activity was essential for cellular viability and proliferation. In glutamine‑starved asparagine‑fed cells, the inhibition of c‑Myc activity led to a decrease in global protein translation and GS synthesis, suggesting an association between c‑Myc expression, GS levels and cellular proliferation, mediated by asparagine when exogenous glutamine is absent.

Metabolic transcriptional analysis on copper tolerance in moderate thermophilic bioleaching microorganism Acidithiobacillus caldus

Bioleaching, an alternative environmental smelting technology, typically uses high concentrations of heavy metal ions, especially in the subsequent phase, due to metal ion accumulation from the mineral. In this study, we analyzed the overall response of the bioleaching microorganism Acidithiobacillus caldus to copper stress through physiological and transcriptomic analyses. Scanning electron microscopy results showed higher extracellular polymeric substances secretion and cell aggregation under copper stress. Intracellular levels of glutamic acid, glycine and cysteine increased, favoring the synthesis of glutathione for maintenance of the oxidation-reduction state. GSH, during copper stress conditions, the activity of GSH-PX and CAT increased, resulting in reduced oxidative damage while maintaining stable intracellular pH. Higher unsaturated and cyclopropane fatty acid levels resulted in increased membrane fluidity and compactness and decreased ATP levels to support the energy requirements for stress resistance. Initially, H+-ATPase activity increased to provide energy for proton output and decreased later at higher copper ion stress. From transcriptome analysis, 140 genes were differentially expressed under low copper stress (1g/L), while 250 genes exhibited altered transcriptional levels at higher copper stress (3g/L). These differentially expressed genes were involved primarily in metabolic pathways such as energy metabolism, two-component systems, amino acid metabolism, and signal transduction. The Sox family cluster gene cluster involved in the conversion of thiosulfate to sulfate was upregulated in the sulfur metabolism pathway. In the oxidative phosphorylation pathway, genes participating in the synthesis of NADH oxidoreductase and cytochrome c oxidase, nuoL, cyoABD (cyoA, cyoB and cyoD) and cydAB (cydA and cydB), were downregulated. The TCS element ompR, closely associated with the osmotic pressure, exhibited active response, while Cu2+ efflux system gene cusRS was upregulated. In the amino acid metabolism, the glnA involved in nitrogen fixation was upregulated and promoted the synthesis of glutamine synthetase for reducing excessive oxidative stress. This study provides new insights into the mechanism underlying A. caldus response to heavy-metal ion stress under harsh bioleaching conditions.

The Distinctive Regulation of Cyanobacterial Glutamine Synthetase.

Glutamine synthetase (GS) features prominently in bacterial nitrogen assimilation as it catalyzes the entry of bioavailable nitrogen in form of ammonium into cellular metabolism. The classic example, the comprehensively characterized GS of enterobacteria, is subject to exquisite regulation at multiple levels, among them gene expression regulation to control GS abundance, as well as feedback inhibition and covalent modifications to control enzyme activity. Intriguingly, the GS of the ecologically important clade of cyanobacteria features fundamentally different regulatory systems to those of most prokaryotes. These include the interaction with small proteins, the so-called inactivating factors (IFs) that inhibit GS linearly with their abundance. In addition to this protein interaction-based regulation of GS activity, cyanobacteria use alternative elements to control the synthesis of GS and IFs at the transcriptional level. Moreover, cyanobacteria evolved unique RNA-based regulatory mechanisms such as glutamine riboswitches to tightly tune IF abundance. In this review, we aim to outline the current knowledge on the distinctive features of the cyanobacterial GS encompassing the overall control of its activity, sensing the nitrogen status, transcriptional and post-transcriptional regulation, as well as strain-specific differences.

Hepatic glutamate transport and glutamine synthesis capacities are decreased in finished vs. growing beef steers, concomitant with increased GTRAP3-18 content

Hepatic glutamate uptake and conversion to glutamine is critical for whole-body N metabolism, but how this process is regulated during growth is poorly described. The hepatic glutamate uptake activities, protein content of system {text{X}}^{ - }_{text{AG}} transporters (EAAC1, GLT-1) and regulatory proteins (GTRAP3-18, ARL6IP1), glutamine synthetase (GS) activity and content, and glutathione (GSH) content, were compared in liver tissue of weaned Angus steers randomly assigned (n = 8) to predominantly lean (growing) or predominantly lipid (finished) growth regimens. Steers were fed a cotton seed hull-based diet to achieve final body weights of 301 or 576 kg, respectively, at a constant rate of growth. Liver tissue was collected at slaughter and hepatic membranes fractionated. Total (75%), Na+-dependent (90%), system {text{X}}^{ - }_{text{AG}}-dependent (abolished) glutamate uptake activity, and EAAC1 content (36%) in canalicular membrane-enriched vesicles decreased as steers developed from growing (n = 6) to finished (n = 4) stages, whereas Na+-independent uptake did not change. In basolateral membrane-enriched vesicles, total (60%), Na+-dependent (60%), and Na+-independent (56%) activities decreased, whereas neither system {text{X}}^{ - }_{text{AG}}-dependent uptake nor protein content changed. EAAC1 protein content in liver homogenates (n = 8) decreased in finished vs. growing steers, whereas GTRAP3-18 and ARL6IP1 content increased and GLT-1 content did not change. Concomitantly, hepatic GS activity decreased (32%) as steers fattened, whereas GS and GSH contents did not differ. We conclude that hepatic glutamate uptake and GS synthesis capacities are reduced in livers of finished versus growing beef steers, and that hepatic system {text{X}}^{ - }_{text{AG}} transporter activity/EAAC1 content is inversely proportional to GTRAP3-18 content.

Biological Design Principles of Complex Feedback Modules in theE. coliAmmonia Assimilation System

To synthesize natural or artificial life, it is critically important to understand the design principles of how biochemical networks generate particular cellular functions and evolve complex systems in comparison with engineering systems. Cellular systems maintain their robustness in the face of perturbations arising from environmental and genetic variations. In analogy to control engineering architectures, the complexity of modular structures within a cell can be attributed to the necessity of achieving robustness. To reveal such biological design, the E. coli ammonia assimilation system is analyzed, which consists of complex but highly structured modules: the glutamine synthetase (GS) activity feedback control module with bifunctional enzyme cascades for catalyzing reversible reactions, and the GS synthesis feedback control module with positive and negative feedback loops. We develop a full-scale dynamic model that unifies the two modules, and we analyze its robustness and fine tuning with respect to internal and external perturbations. The GS activity control is added to the GS synthesis module to improve its transient response to ammonia depletion, compensating the tradeoffs of each module, but its robustness to internal perturbations is lost. These findings suggest some design principles necessary for the synthesis of life.

Influence of insulin on glutamine synthetase in the Müller glial cells of retina

Glutamine synthetase (GS), a Müller cell specific enzyme in the retina, is the key enzyme involve in glutamate metabolism. The goal of this study was to investigate the expression and regulation of GS by insulin in the cultured rat retinal Müller cells. Immunocytochemical and immunoblotting experiments showed that the cultured Müller cells express GS protein under normal cell culture conditions. Insulin treatments decreased the GS expression both in a time and dose dependent manner. Insulin also decreased the hydrocortisone induced GS expression. Furthermore, we investigated the expression and regulation of two other Müller cell specific enzymes known to be involved in glutamate metabolism, the mitochondrial branched chain aminotransferase (BCATm) and pyruvate carboxylase (PC). Immunoblotting experiments showed that Müller cells expressed both BCATm and PC. Treatments of cells with hydrocortisone or insulin did not influence the BCATm expression level. Hydrocortisone treatment of cells increased the PC expression but this induced expression was suppressed by insulin treatment. Müller cells expressed insulin receptor proteins (IRβ and IRS-1) and insulin activation induced the phosphotyrosine level of insulin receptor proteins. Moreover, hydrocortisone did not influence the expression or activation of these receptor proteins. The data suggests that insulin modulates the GS synthesis and may influence glutamate metabolism in the cultured retinal Müller cells but not by influencing the insulin signaling pathway.

Effects of aerobic versus anoxic conditions on glutamine synthetase activity in eelgrass ( Zostera marina L.) roots: regulation of ammonium assimilation potential

Root glutamine synthetase (GS; EC 6.3.1.2) activity was measured daily (0 to 4 days) for eelgrass ( Zostera marina L.) plants held under continuous darkness rooted in sediments, continuous darkness without sediments, continuous light without sediments, and control light/dark cycle (Control L/D). Roots experiencing prolonged aerobiosis exhibited lower activity in vitro than controls, whereas roots experiencing prolonged anoxia exhibited increased activity. Plants held in darkness without sediments had activity intermediate between controls and anoxic roots. One-hour pretreatment of root extracts with ATP slightly reduced in vitro glutamine synthetase activity, whereas pretreatment with ADP and AMP increased activity ≈50%. While glutamine synthetase activity increased with higher adenylate energy charge (AEC) in the reaction mixture, pretreatment of enzyme extracts at high adenylate energy charges decreased subsequent activity relative to pretreatment at lower energy charges. One-hour pretreatment with l-alanine (Ala) had little effect on enzyme activity. Pretreatment with l-glutamine (Gln), l-glutamate (Glu), and γ-amino butyric acid (GABA) increased activity ≈75%. Incubation of excised roots under anoxic conditions for 24 h nearly doubled enzyme activity. However, addition of cycloheximide to anoxic root incubations lessened or prevented the increase in activity. It appears that enhanced glutamine synthetase activity following periods of root anoxia results from interactions with metabolites that fluctuate between aerobic and anoxic conditions, particularly adenylates, and from de novo synthesis of glutamine synthetase or some other protein synthesis-dependent process.

Regulation, purification and partial characterization of glutamine synthetase fromStreptomyces aureofaciens

The activity of glutamine synthetase (GS) fromStreptomyces aureofaciens was regulated by the availability of the nitrogen source. Rich nitrogen sources repressed GS synthesis and increased GS adenylylation. The enzyme was purified 270-fold to virtual homogeneity with 37% recovery. The molar mass of the native enzyme and its subunits was determined to be 620 and 55 kDa, respectively, indicating that GS is composed of 12 identical subunits. The enzyme has a hexagonal-bilayered structure as observed by electron microscopy. The isoelectric point of the purified GS was at pH 4.2. The enzyme was stable for 1 h at 50°C but lost activity rapidly when incubated at 65 and 70°C. Mg2+ supported relative synthetic activity of 100 and 72%, respectively, with the corresponding pH optima of 7.3 and 7.0. Mn2+ ions activated transferase activity at a pH optimum of 7.0. The temperature optimum for all GS activities was 50°C. Intermediates of the citric acid cycle exerted insignificant effects on the synthetic activities. There was no SH-group essential for the GS activity.

Ammonia-assimilating enzymes in the basidiomycete fungus Pleurotus ostreatus

l-Glutamate and l-aspartate are the natural nitrogen sources preferred by the fungus Pleurotus ostreatus. In the vegetative mycelium NAD-dependent glutamate dehydrogenase (NAD-GDH) and glutamine synthetase (GS) were both present, but the highest level of these enzymes was found in the fruiting body. NAD-GDH was derepressed by ammonia and repressed by higher concentrations of l-glutamate. Otherwise, the level of the enzyme in mycelium was independent of the nitrogen source. GS was derepressed by ammonia, l-glutamate, l-aspartate and l-alanine. Repression of GS was observed in the presence of l-glutamine, l-arginine, nitrate, urea and low concentrations of ammonia. An increase in the ratio of glucose to nitrogen resulted in a decrease in the level of GS in the mycelium. The K m values of NAD-GDH for 2-oxoglutarate, ammonia, NADH, l-glutamate and NAD+ were 2·9 mM, 3·3 mM, 11 μM, 0·18 mM and 62 μM, respectively. The K m values of GS for l-glutamate and ammonia were 2·5 and 0·27 mM, respectively. The repression of GS and derepression of NAD-GDH in the presence of low concentrations of ammonia together with glutamate or aspartate could be explained by an inhibition or repression of some amino acid permeases by ammonia. The absence of NADP-dependent glutamate dehydrogenase and glutamate synthase together with the apparent mode of regulation of NAD-GDH and GS synthesis suggest that P. ostreatus assimilates ammonia via GS and NAD-GDH. However, a dissimilatory role of NAD-GDH in the deamination of l-glutamate, due to its very low K m for l-glutamate, is not excluded.

Molecular characterization of the gene encoding glutamine synthetase in the cyanobacterium Calothrix sp. PCC 7601

In order to study the regulation of the synthesis of glutamine synthetase in response to changes in environment parameters (light and nitrogen sources), we have cloned and sequenced the glnA gene from the filamentous cyanobacterium Calothrix PCC 7601. This gene consists of 472 codons and encodes a polypeptide of M r 52,290 highly homologous to that from Anabaena PCC 7120, but more distant from those identified from other procaryotes. The relative abundance of the two glnA transcripts (1.6 and 1.8 kb) is equivalent in cells grown under either red or green light, but the 1.6-kb species predominates in nitrate-grown cells and the 1.8-kb species in ammonia-grown cells. The very high identity (74%) observed between the 374-bp long nucleotide sequence upstream from the Calothrix and Anabaena glnA genes suggests the existence of similar regulatory signals for the control of glnA expression in both cyanobacteria.

Role of phosphorylated metabolic intermediates in the regulation of glutamine synthetase synthesis in Escherichia coli

Transcription of the Ntr regulon is controlled by the two-component system consisting of the response regulator NRI (NtrC) and the kinase/phosphatase NRII (NtrB), which both phosphorylates and dephosphorylates NRI. Even though in vitro transcription from nitrogen-regulated promoters requires phosphorylated NRI, NRII-independent activation of NRI also occurs in vivo. We show here that this activation likely involves acetyl phosphate; it is eliminated by mutations that reduce synthesis of acetyl phosphate and is elevated by a mutation expected to cause accumulation of acetyl phosphate. With purified components, we investigated the mechanism by which acetyl phosphate stimulates glutamine synthetase synthesis. Acetyl phosphate, carbamyl phosphate, and phosphoramidate but not ATP or phosphoenolpyruvate acted as substrates for the autophosphorylation of NRI in vitro. Phosphorylated NRI produced by this mechanism exhibited the properties associated with NRI phosphorylated by NRII, including the activated ATPase activity of the central domain of NRI and the ability to activate transcription from the nitrogen-regulated glutamine synthetase glnAp2 promoter.

Glutamine synthesis in Streptomyces — a review

The synthesis of glutamine synthetase (GS), a key enzyme in ammonium (NH 4 + assimilation, is regulated by nitrogen availability in several Streptomyces strains. In addition, the enzymatic activity of the GS enzyme is post-translationally regulated by adenylylation. Nitrogen regulation of GS synthesis is mediated at the transcriptional level in S. coelicolor, and transcription of the GS structural gene ( glnA) requires a positive regulatory protein, GlnR. The amino acid sequence of the GlnR protein is similar to that of the Escherichia coli positive regulatory proteins, OmpR and PhoB, which belong to the family of bacterial two-component regulatory systems. DNA encoding a GSII-like enzyme has been cloned from S. viridochromogenes and S. hygroscopicus, but the role of this GS isoenzyme in NH 4 + assimilation in Streptomyces is unclear.

Immunological studies of glutamine synthetase inFrankia-Alnus incanasymbioses

We have investigated the presence and form of glutamine synthetase (GS) in Frankia vesicle cluster preparations of two actively nitrogen-fixing Frankia-Alnus incana root-nodule symbioses and in cultured Frankia sp. strain CpI1 (HFP070101). The symbioses contained Frankia CpI1 or the local source of Frankia. We used Western-blot analysis with antisera raised against three types of GS. In symbiotic Frankia GS protein was not detected at a significant level when either antisera against Rhodospirillum rubrum GS or antisera against Rhizobium meliloti GSII were used. In cultured Frankia CpI1 GSI was detected both when grown with NH4+ or N2 as nitrogen source, and GSII was detected when grown on N2. Antiserum raised against the nodule-specific GSn1 of Phaseolus vulgaris crossreacted with a 43-kDa polypeptide corresponding to plant GS in root-nodule extracts from Alnus, and with a 41-kDa polypeptide corresponding to GSII in cultured Frankia CpI1 grown on N2. We conclude that both GSI and GSII are repressed in symbiotic Frankia and that NH4+ produced through nitrogen fixation is assimilated by the plant in Frankia-Alnus incana symbioses. It thus appears that vesicle formation, synthesis of nitrogenase and synthesis of GS are separately regulated in symbiotic Frankia and that the plant has to supply symbiotic Frankia with organic nitrogen in some form in addition to the carbon.

Purification and Characterization of a Repressor for the <italic>Bacillus cereus glnRA</italic> Operon

We report the overexpression, purification, and properties of the regulatory protein, GlnR, for glutamine synthetase synthesis of Bacillus cereus. The protein was found to be a dimer with a molecular weight of approximately 30,000, and its subunit molecular weight was 15,000 in agreement with that (15,025) of deduced amino acid sequence of GlnR. The purified GlnR protein bound specifically to the promoter region of the glnRA operon of B. cereus and Bacillus subtilis. The binding of the GlnR protein to the DNA fragment was enhanced by the presence of glutamine synthetase, the product of glnA, of B. cereus or B. subtilis, although the affinity of the GlnR protein for DNA was not affected in the presence of glutamate, glutamine, Mg2+, Mn2+, or ammonia. These results indicate the existence of an interaction between GlnR and glutamine synthetase, and support the hypothesis that the regulation of glnA expression requires both GlnR protein and glutamine synthetase in Bacillus.

Hippocampal glutamine synthetase: insensitivity to glucocorticoids and stress

Glucocorticoids enhance the neurotoxic potential of several insults to the rat hippocampus that involve overactivation of glutamatergic synapses. These hormones also stimulate the synthesis of glutamine synthetase (GS) in peripheral tissue. Because this enzyme helps regulate glutamate metabolism in the central nervous system, glucocorticoid induction of GS in the brain may underlie the observed synergy. We have measured GS activity in the hippocampus and skeletal muscle (plantaris) of adult rats after bilateral adrenalectomy (ADX), corticosterone (Cort) replacement, or stress. No significant changes in GS were observed in hippocampal tissue, whereas muscle GS was significantly elevated after Cort treatment or stress and was reduced after ADX. These results suggest that Cort-induced shifts in GS activity probably do not explain Cort neurotoxicity, although the stress-induced rise in muscle GS may be relevant to certain types of myopathy.

Regulation of glutamine synthetase synthesis and activity by glucocorticoids and adrenoceptor activation in astroglial cells

Glucocorticoids are known to induce the synthesis and activity of glutamine synthetase (GS; EC 6.3.1.2.) in astroglial cells. In the present paper, noradrenaline (NA), in itself ineffective upon GS regulation, potentiated GS activity in astroglial primary cultures in the presence of the glucocorticoid dexamethasone, the GS activity being further stimulated in the presence of glutamate (glu). Thus, adrenoceptor activation might interact with the glucocorticoid induced GS activity in astroglial primary cultures.

Nucleotide sequence and expression of the glutamine synthetase structural gene, glnA, of the archaebacterium Methanococcus voltae

The sequence of a 2,746-bp DNA fragment of Methanococcus voltae carrying the glnA gene for glutamine synthetase (GS), was established. A 1,338-bp open reading frame (ORF), encoding a 446-amino-acid polypeptide of 50,142 Da, was defined as glnA on the basis of its similarity to other glnA genes and on the ability of a DNA fragment carrying this ORF to complement an Escherichia coli Gln − mutant. No sequence homology was found between sequences flanking the M. volae glnA gene and other eubacterial glnA genes. In M. voltae, the gene was transcribed as a monocistronic unit and GS synthesis was partially repressed at high ammonia concentrations. At the amino acid sequence level, the highest similarity was found with GS of Bacillus subtilis and Clostridium acetobutylicum.

Glutamine synthetase specific activity and protein concentration in symbiotic Anabaena associated with Azolla caroliniana.

Glutamine synthetase (GS) is the primary NH4+ assimilating enzyme of cyanobacteria. The specific activities and cellular protein concentration of GS in symbiotic cyanobacteria associated with the water fern Azolla caroliniana were determined and compared to free-living cultures of Nostoc sp. strain 7801, a strain originally isolated from symbiotic association with the bryophyte Anthoceros punctatus. Both the in vitro specific activity and concentration of GS in symbiotic cyanobacteria separated from A. caroliniana were approximately 3-fold lower than the free-living Nostoc sp. strain 7801 culture. These results imply depressed synthesis of GS by the symbiont associated with A. caroliniana.
