Chlorosulfonation of Some Aldimine-Isocyanate Cycloadducts

Attempts to chlorosulfonate 1,4-diphenyl-1,3-diazetidin-2-one (1) failed, but the 3-methyl derivative (2) reacted with chlorosulfonic acid to give the bis-sulfonyl chloride (3), characterized as the sulfonamides 4 and 5. 2,3,6-Triphenyl-2,3-dihydro-1,3,5-thiadiazin-4-one (6) with chlorosulfonic acid suffered an acid-catalyzed ring-opening reaction forming the sulfonyl derivatives (8, 9) of N-phenyl-N′-thiobenzoylurea (7). Condensation of 8 and 9 with diethylamine afforded the diethyl-sulfonamide (10). Dibenzylideneethylenediamine (11) reacted with thiobenzoyl isocyanate at room temperature to yield the cycloadduct 12; however at 90°C, N,N′-di (thiobenzoylcarbamoyl)ethylenediamine (13) was obtained. The cycloadduct 12 with chlorosulfonic acid gave the ring-opened disulfonyl chloride 14 and the diethylsulfonamide 15. 1,6-Diphenylhexahydro-s-triazine-2,4-dione (17) was converted into the dimethyl derivative (18), which with chlorosulfonic acid afforded the bis-sulfonyl chloride (19), characterized as the sulfonamides 20–22. BIMU1 Increases Associative Memory in Rats by Activating 5-HT4 Receptors

Olfactory association learning was used to investigate the involvement of 5-HT4 receptors in learning and long-term memory. The behavioral role of the 5-HT4 receptors was studied by using BIMU1 (3-ethyl-2,3-dihydro-N-[endo-8-methyl-8-azabicyclo(3.2.1)oct-3-yl]-2-oxo-1H-benzimidazole-1-carboxamide, hydrochloride (Boehringer Ingelheim, Italy); a mixed 5-HT4 agonist/5-HT3 antagonist, and GR125487 (1-[2-[methyl sulphonyl)-amino]ethyl]-4-piperidinyl-methyl 5-fluoro-2-methoxy-1H-indole-3-carboxylate; Glaxo Group Research, Hertfordshire, U.K.), a specific 5-HT4 antagonist. The intraperitoneal injections of BIMU1 at 1, 5, and 10 mg/kg were followed by an substantial improvement (>15% in percentage of correct responses at the dose of 10 mg/kg) in associative memory. Difficulty rapidly reversing behavioral responses to previously learned association, 1 month later indicated that the BIMU1 effect at 10 mg/kg was not transient, but correlated to long-term memory. The effects of BIMU1 are most likely to be mediated by 5-HT4 receptors since they were blocked by GR125487 at 10 mg/kg. These data suggest that activation of 5-HT4 receptors may modulate cognitive processes like learning and memory. © 1997 Elsevier Science Ltd.

Case study Client and project

This paper describes the use of IT on a major research campus for Glaxo Group research. Ove Arup & Partners were appointed with Sheppard Robson and Davis Langdon & Everest as the Project Engineer Architect. 350 architects and engineers were assembled in a single office and 4,500 drawings produced using CAD with linked networks of Autocad and Intergraph systems. The M & E subcontractors were invited to use the Autocad system which had been bought by the client. CAD was essential to the delivery of the project on time. Outsourcing was rejected. Good management and standards were essential to overcome difficulties and deliver a very complex project.

R&D networking in a pharmaceutical company: Some implications for human resource management

AbstractA case‐study of the R&D networking activities of Glaxo Group Research was carried out between June and November 1993. Different types of networking situation were identified using a simple 2times2 matrix –‐ the ‘networking uncertainty map’. This paper describes the background to Glaxo's R&D networking activities, introduces and illustrates the uncertainty map and then suggests the implications of each type of network. There are implications in general for R&D management and more specifically for the management of human resources in R&D. Managers might easily apply the uncertainty map to their own R&D networking situations.

Experience of International Data Management for Drug Development in a Local Operating Company

Success in the pharmaceutical industry is dependent on many factors. Among them, the efficient management of clinical data is increasingly important. Glaxo Group Research (GGR), to accelerate the preparation of international registration dossiers (IRDs), has devolved the data management of international multicenter studies to the local operating companies (LOCs). The mother company has set up a standalone package that enables LOCs to process data according to central standards.The Italian subsidiary was recently involved in international development. This paper attempts to provide an overview of this experience, with special reference to data management activities in the context of international development—the issues encountered, the acquisition of knowledge (methodologies, computer tools), and the advantages of this working method.

The Glaxo International Postmarketing Surveillance System: Present and Future

The Glaxo worldwide system for monitoring the safety profile of its marketed and investigational products is described. This paper specifically focuses on the coordinated development of drug surveillance activities between Glaxo Group Research (United Kingdom) and Glaxo Inc. (United States). Central to this dedicated effort is a single worldwide computer system for the storage, monitoring and retrieval of adverse event data.

Symbiotic relationship between a research institute and a pharmaceutical company: the Baker Institute/Glaxo Australia story.

1. The Baker Medical Research Institute is a non-profit, charitable institution to advance knowledge and solve problems in the area of cardiovascular medicine. This allows scientists to collaborate within a critical mass and be flexible in their approach to research. 2. The Institute has interactions with the pharmaceutical industry at three levels: (i) contract research on a specific molecule; (ii) clinical trials; and (iii) collaborative basic research agreements. 3. The Glaxo Australia-Baker Medical Research Institute Agreement is for curiosity driven research in specified areas of vascular pharmacology of interest to Glaxo Group Research. 4. The relationship between universities, research institutes and the pharmaceutical industry is discussed in terms of the starting point for drug development. 5. The enormous cost (US$100 million), lead time (10 years) and uncertainty of a successful profitable drug are compelling reasons why an independent pharmaceutical company is unlikely to succeed in Australia today.
