Alterations in tryptophan metabolism and de novo NAD+ biosynthesis within the microbiota-gut-brain axis in chronic intestinal inflammation.

Inflammatory bowel disease is an incurable and idiopathic disease characterized by recurrent gastrointestinal tract inflammation. Tryptophan metabolism in mammalian cells and some gut microbes comprise intricate chemical networks facilitated by catalytic enzymes that affect the downstream metabolic pathways of de novo nicotinamide adenine dinucleotide (NAD+) synthesis. It is hypothesized that a correlation exists between tryptophan de novo NAD+ synthesis and chronic intestinal inflammation. Transcriptome analysis was performed using high-throughput sequencing of mRNA extracted from the distal colon and brain tissue of Winnie mice with spontaneous chronic colitis and C57BL/6 littermates. Metabolites were assessed using ultra-fast liquid chromatography to determine differences in concentrations of tryptophan metabolites. To evaluate the relative abundance of gut microbial genera involved in tryptophan and nicotinamide metabolism, we performed 16S rRNA gene amplicon sequencing of fecal samples from C57BL/6 and Winnie mice. Tryptophan and nicotinamide metabolism-associated gene expression was altered in distal colons and brains of Winnie mice with chronic intestinal inflammation. Changes in these metabolic pathways were reflected by increases in colon tryptophan metabolites and decreases in brain tryptophan metabolites in Winnie mice. Furthermore, dysbiosis of gut microbiota involved in tryptophan and nicotinamide metabolism was evident in fecal samples from Winnie mice. Our findings shed light on the physiological alterations in tryptophan metabolism, specifically, its diversion from the serotonergic pathway toward the kynurenine pathway and consequential effects on de novo NAD+ synthesis in chronic intestinal inflammation. The results of this study reveal differential expression of tryptophan and nicotinamide metabolism-associated genes in the distal colon and brain in Winnie mice with chronic intestinal inflammation. These data provide evidence supporting the role of tryptophan metabolism and de novo NAD+ synthesis in IBD pathophysiology.

Antioxidant Activities and Phenolic Composition of Sarcococca saligna Leaves

Abstract: Sarcococca saligna (D. DON) Muel is a vital plant with several steroidal alkaloids and is effective against syphilis, liver diseases, gastrointestinal tract disease, infections, and inflammation. The present study aimed to investigate the in vitro antioxidant activities and phenolic profile of S. saligna. High-performance liquid chromatography (HPLC) analysis has shown the presence of twelve phenolics namely kaempferol-3-O-glucoside, proanthocyanidin B1, quercetin-3-malonylglucoside- 7-glucoside, kaempferol-3-O-sophorotioside, Isorhamnetin-3-O-glucoside-7-Orhamnoside, kaempferolhexoside, 4-O-Caffeoyl-5-O-p-coumaroylquinic acid, 3,4-Di-O-caffeoyl-5- O-feruloylquinic acid, 3,4-Di-O-feruloyl-5-O-caffeoylquinic acid, flavogalloyl-HHDP-gluconic acid (lagerstannin B), 3,4-di-O-caffeoylquinic acid and kaempferol-3-(caffeoyldiglucoside)-7-glucoside. The hot water extract showed the highest anti-lipid peroxidative activities. The IC50 value for 2,2- diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity was 71.5 ± 2.1 μg/mL. The extract could chelate the iron and possess reducing activities on phosphomolybenum assay. It is concluded that extracts of S. saligna are rich in antioxidants and contain essential phytochemicals. objective: The present study was aimed to investigate thein vitro antioxidant activities and phenolic profile of S. saligna. The HPLC system used was Agilent 1260 Infinity HPLC system which consists of quaternary pump, degasser, auto-sampler and diode array detector (DAD). conclusion: The results have shown that S. saligna antioxidant rich extract can be effectively utilized in food and pharmaceutical industries with additional income. However, more detailed understanding of utilization of extract against degenerative diseases is required. There is also need to find out the safety and bioavailability of the plant extract for its use as nutraceutical and as a plant products.

Ulcerative Colitis diagnosed for the First Time in a 93 year old Nigerian Woman: A Case Report

Inflammatory bowel disease (IBD) is a chronic and life-long disease characterized by gastrointestinal tract inflammation caused by the interplay of the host’s genetic predisposition, immune responses, and various environmental factors. It is characterized by chronic diarrhea, among other symptoms. Its spectrum comprises Crohn’s disease (CD), Ulcerative Colitis (UC), and indeterminate colitis (IC). Inflammatory bowel disease occurs in two peaks, the first at 15-25 years, and the second at 40-60 years. It is rarely reported in the extremes of age. Here we report the case of a 93-year-old woman with previously undiagnosed ulcerative colitis presenting as large bowel obstruction.

Bifidobacterium species viability in dairy-based probiotic foods: challenges and innovative approaches for accurate viability determination and monitoring of probiotic functionality.

Bifidobacterium species are essential members of a healthy human gut microbiota. Their presence in the gut is associated with numerous health outcomes such as protection against gastrointestinal tract infections, inflammation, and metabolic diseases. Regular intake of Bifidobacterium in foods is a sustainable way of maintaining the health benefits associated with its use as a probiotic. Owing to their global acceptance, fermented dairy products (particularly yogurt) are considered the ideal probiotic carrier foods. As envisioned in the definition of probiotics as "live organisms," the therapeutic functionalities of Bifidobacterium spp. depend on maintaining their viability in the foods up to the point of consumption. However, sustaining Bifidobacterium spp. viability during the manufacture and shelf-life of fermented dairy products remains challenging. Hence, this paper discusses the significance of viability as a prerequisite for Bifidobacterium spp. probiotic functionality. The paper focuses on the stress factors that influence Bifidobacterium spp. viability during the manufacture and shelf life of yogurt as an archetypical fermented dairy product that is widely accepted as a delivery vehicle for probiotics. It further expounds the Bifidobacterium spp. physiological and genetic stress response mechanisms as well as the methods for viability retention in yogurt, such as microencapsulation, use of oxygen scavenging lactic acid bacterial strains, and stress-protective agents. The report also explores the topic of viability determination as a critical factor in probiotic quality assurance, wherein, the limitations of culture-based enumeration methods, the challenges of species and strain resolution in the presence of lactic acid bacterial starter and probiotic species are discussed. Finally, new developments and potential applications of next-generation viability determination methods such as flow cytometry, propidium monoazide-quantitative polymerase chain reaction (PMA-qPCR), next-generation sequencing, and single-cell Raman spectroscopy (SCRS) methods are examined.

Endoscopic Management of Crohn’s Disease Strictures

Crohn’s disease is a chronic, progressive inflammatory condition characterised by gastrointestinal tract inflammation and extra-intestinal manifestations. Some patients may develop stricturing disease, which may either be of inflammatory, fibrotic or post-anastamotic aetiologies. The management of fibrostenosis is challenging, necessitating preventative strategies and a multidisciplinary approach to its diagnosis and management once established. Although several treatment options are currently used in the management of strictures, there are currently no targeted anti-fibrotic therapies. Endoscopically, there are several modalities that can be employed in the symptomatic patient including balloon dilatation and stenting. In this article we will briefly discuss the pathophysiology and investigations with focus centred around endoscopic management of Crohn’s strictures, before suggesting a strategy to employ when approaching these cases.

Identification and bioinformatic analysis of invA gene of Salmonella in free range chicken.

Salmonella is a serious cause of the health issues in human and animal worldwide. Salmonella has been isolated from different biological samples and it considers as the key role in induction of inflammation of gastrointestinal tract which in turn cause diarrhoea in different species. To further understand the involvement of Salmonella in contaminating and infecting fresh eggs and meat of free-range chicken. This study aimed to establish the microbiological and molecular detections of Salmonella in the cloaca of the free-range chicken and to identify predicted biological functions using Kyoto Encyclopedia of Gene and Genomic (KEGG) pathways and protein-protein interaction. Cloacal swabs were collected from free range chicken raised in the local farm in Duhok city. The isolates were cultured and biochemical test performed using XLD and TSI, respectively. Molecular detection and functional annotation of invA gene was carried out using Conventional PCR and bioinformatics approaches. The present study found that Salmonella was detected in 36 out of 86 samples using microbiological methods. To confirm these findings, invA gene was utilised and 9 out of 36 Salmonella isolates have shown a positive signal of invA by agarose gel. In addition, bioinformatic analysis revealed that invA gene was mainly associated with bacterial secretion processes as well as their KEGG terms and Protein-Protein Interaction were involved in bacterial invasion and secretion pathways. These findings suggested that invA gene plays important role in regulating colonization and invasion processes of Salmonella within the gut host in the free range chicken.

An injectable subcutaneous colon-specific immune niche for the treatment of ulcerative colitis.

As a chronic autoinflammatory condition, ulcerative colitis is often managed via systemic immunosuppressants. Here we show, in three mouse models of established ulcerative colitis, that a subcutaneously injected colon-specific immunosuppressive niche consisting of colon epithelial cells, decellularized colon extracellular matrix and nanofibres functionalized with programmed death-ligand 1, CD86, a peptide mimic of transforming growth factor-beta 1, and the immunosuppressive small-molecule leflunomide, induced intestinal immunotolerance and reduced inflammation in the animals' lower gastrointestinal tract. The bioengineered colon-specific niche triggered autoreactive T cell anergy and polarized pro-inflammatory macrophages via multiple immunosuppressive pathways, and prevented the infiltration of immune cells into the colon's lamina propria, promoting the recovery of epithelial damage. The bioengineered niche also prevented colitis-associated colorectal cancer and eliminated immune-related colitis triggered by kinase inhibitors and immune checkpoint blockade.

Toxic metabolites and metabolic soft spots of celastrol based on glutathione metabolic capture and high-resolution mass spectrometry

ABSTRACT Background Celastrol is known as one of the most medicinally valuable compounds. However, the pharmaceutical application of celastrol is significantly limited due to high toxicity, while there are few reports on the mechanism of toxicity. Methods This study searched for possible toxic metabolites through phase I in vitro metabolism and glutathione capture experiments. Then in vivo metabolism experiments in mice and rats were conducted to look for metabolites in vivo. Finally, mice in vivo toxicity experiment was conducted to verify the toxicity of different doses of celastrol to mice. Results In the in vivo and in vitro metabolism experiments, we found 7 phase I metabolites in vitro, 9 glutathione conjugation metabolites in vitro, and 20 metabolites in vivo. The metabolic soft points of celastrol could be the quinone methyl structure at C3-OH and C6. In vivo toxicity experiments show that celastrol causes weight loss, diarrhea, gastrointestinal tract and liver inflammation in mice. Conclusions This study analyzed the metabolites and possible metabolic soft spots of celastrol, and its hepatotoxicity and gastrointestinal toxicity were demonstrated through in vivo studies for the first time. The results might provide an important basis for potential structural modification to increase the druggability of celastrol.

Ulcerative Colitis with Henoch Schonlein Purpura in Pediatric Patient: A Case-Report

Background: Inflammatory bowel disease (IBD) is defined as idiopathic disorder which associated with inflammation of gastrointestinal tract. Ulcerative colitis (UC) and Chron's disease are the predominant forms of IBD. Henoch Schonlein Purpura and UC share some similar symptoms and can mimic each other especially in children. In this case report, we present an eight-year-old boy who had main complaint of recurrent bloody stools and abdominal pain. Diagnosis was made through history taking, physical examination, laboratory examination and colonoscopy in this patient. Case: A boy aged 8 years and 1 month had main complaint of bloody stools with fluid consistency. This complaint was accompanied by abdominal pain all over the abdominal region and vomiting. Upon physical examination, the child was in pain. The vital signs were within normal limits. Laboratory examination showed leukocytosis. The routine stool examination showed malabsorption of fat, protein, and carbohydrates, followed with positive erythrocytes, leukocytes, epithelium, bacteria, and yeast cells; but the stool culture was sterile. Gastroscopy and colonoscopy were performed which showed erosive gastritis, duodenitis, and proctitis. Discussion: The relationship between ulcerative colitis and HSP remains unknown. Recent research showed overproduction of IgA may induce chronic inflammation in the intestinal. While HSP is IgA/immune complex mediated, IBD is thought to be predominantly T-cell driven. Recently, in IBD patients especially those with ulcerative colitis, IgA were found in their histopathologic biopsy result. Conclusion: The relationship between ulcerative colitis and HSP remains unknown. Recent research has shown that the overproduction of IgA may induce chronic inflammation in the intestinal. As such, symptoms of both diseases may mimic each other as seen in our patients.

Effects of a multispecies direct-fed microbial on gastrointestinal permeability during feed restriction in a growing heifer model

The objectives were to evaluate the effects of a 4-strain direct-fed microbial (DFM) on gastrointestinal tract (GIT) permeability and inflammation during feed restriction (FR) in heifers. Holstein heifers (n = 32; mean ± standard deviation; 295 ± 25 kg body weight; 287 ± 17 d of age) were used in an experiment conducted in 2 replicates (16/replicate). Heifers were randomly assigned to 1 of 2 top-dressed dietary treatments: (1) control (CON; 10 g/d dried lactose; n = 16) or (2) DFM containing a commercial blend of Lactobacillus animalis, Propionibacterium freudenreichii, Bacillus licheniformis, and Bacillus subtilis at 11.8 × 109 cfu/d (PRO; 10 g/d 4-strain DFM; n = 16). The trial consisted of 2 experimental periods (P): P1 (14 d) served as baseline for P2 (5 d), when all heifers were restricted to 40% of their P1 dry matter intake (DMI). On P1 d 12 and P2 d 2 and 5, GIT permeability was evaluated using oral chromium (Cr)-EDTA. By design, FR decreased DMI (60%) and body weight (∼18 kg) in all heifers. Regardless of treatment, during FR, all heifers had decreased circulating glucose, β-hydroxybutyrate, insulin, and l-lactate (4, 14, 45, and 19%, respectively), but increased nonesterified fatty acids, serum amyloid A, and haptoglobin (3.0-, 1.7-, and 5.0-fold, respectively). Circulating white blood cells, neutrophils, lymphocytes, and basophils decreased (4, 7, 5, and 6%, respectively), whereas eosinophils increased (41%) during P2 irrespective of dietary treatment. Circulating IFN-γ inducible protein-10 increased (23%) during FR compared with P1 regardless of treatment. Plasma Cr area under the curve increased in all heifers on d 2 and 5 (10 and 14%, respectively) of P2 relative to P1, but this was unaltered by dietary treatment. In summary, FR compromised GIT barrier function and stimulated an inflammatory response, but this did not appear to be ameliorated by PRO.

21 Intentionally Induced Barrier Dysfunction Causes Inflammation, Decreases Growth, and Alters Carcass Characteristics of Feedlot Cattle

Abstract There are a variety of acute and chronic psychological and physical stressors associated with management practices used in beef production systems. Primary challenges in feedlot cattle include heat stress, acute and subacute acidosis, weaning, transportation, feed deprivation/restriction, and social mixing/crowding. These stressors disturb the homeostatic state of the host and microbiome and cause acute or chronic inflammation resulting in damage to the gastrointestinal tract (GIT) epithelial barrier that leaves the animal vulnerable to enteric pathogens and toxins. A breach of the small intestinal barrier (leaky gut) allows unwanted pathogens and toxins to infiltrate into blood, causing infection, depressed feed intake, and diverts nutritional resources away from anabolic processes. We have demonstrated that intentionally inducing GIT inflammation in feedlot cattle with aspirin, which is known to cause mucosal injury and increase GIT barrier dysfunction, provides a model system to better understand the metabolic cost of leaky gut in grain-fed feedlot cattle. Indeed, aspirin increased leakage of chromium-EDTA into urine, raised serum concentrations of lipopolysaccharide binding protein (LBP), decreased growth by 4.5 to 8.5%, and re-partitioned carcass components from lean tissue (rib-eye area) into fat. Thus, the chronic inflammatory effects of leaky gut have the potential to create a significant economic loss. Our work also shows that in-feed administration of monensin and tylosin have offered a solution to the non-specific immune response observed in grain-fed cattle by increasing jejunal villus height to crypt depth ratio, diminishing leaky gut, and decreasing serum LBP. Finally, we have observed that molasses based liquid supplements increase ruminal butyrate concentration, resulting in improved gut barrier function, decreased serum inflammatory markers, and greater DMI and ADG in newly received feedlot cattle fed a 30% roughage diet.

Multifunctional effects of Lactobacillus sakei HEM 224 on the gastrointestinal tract and airway inflammation

Mucosal tissues serve as the first defense line and their commensal microbiota play a role in sustaining of host health. This study aimed to isolate and evaluate a putative probiotic strain on various mucosal regions. Lactobacillus sakei HEM 224 was isolated from traditional Korean kimchi and identified. In the safety assessment L. sakei HEM 224 showed negative results for hemolysis, biogenic amine production and transferable antibiotic resistance. The probiotic potential of strain HEM 224 in diverse mucosal areas was shown in two different models, viz. a murine model with colitis induced by dextran sulfate sodium (DSS) and an allergic airway inflammation model induced by ovalbumin (OVA). In the colitis model, oral administration of L. sakei HEM 224 improved colitis physiology with immunomodulation, enhancing barrier components and gut microbiota alteration. In the allergic airway inflammation model, the intranasal administration of the strain decreased type 2 inflammation and enhanced epithelial barrier integrity from the airways. These results demonstrate that L. sakei HEM 224 can ameliorate inflammatory conditions in both the gastrointestinal and respiratory tracts through the reinforcement of the epithelial barrier and immunomodulation.

Giardia duodenalis (Styles, 1902) in Cattle: Isolation of Calves with Diarrhoea and Manure Treatment in the Lagoon Presented as Risk Factors in Latvian Herds.

Giardia duodenalis is a waterborne zoonotic protozoan that causes gastrointestinal tract inflammation in humans, cattle, and other animals. The aim of the present study was to estimate the prevalence and potential risk factors for Giardia infection in cattle in Latvia. During 2020-2021, a total of 973 individual faecal samples from cattle aged from 1 day to 12 years old, from 32 cattle herds, were tested for Giardia cyst presence with immunofluorescence staining followed by Giardia assemblage differentiation targeting beta-giardin gene. Using a questionnaire, information was collected to estimate the potential risk factors for G. duodenalis infection in cattle herds. Giardia was found in 8.4% of the examined cattle with a mean intensity of 5756 cysts per gram of faeces. The highest prevalence was observed in the 0 to 3-month-old calves (16.4%). At least one Giardia shedding animal was found in 27 herds with an overall prevalence of 84.4%. Significantly higher prevalence was found for cattle infected with G. duodenalis assemblage E compared to that infected with assemblage A: 88.7% and 11.3%, respectively. Protective factors such as age and rodent control and change of shoes were found to be significant for Giardia infection, while isolating calves for diarrhoea and water bodies (ponds/lakes) in pasture were potential risk factors in Latvian cattle.

Effects of Bacillus subtilis PB6 supplementation on production, metabolism, inflammatory biomarkers, and gastrointestinal tract permeability in transition dairy cows

Objectives were to evaluate the effects of Bacillus subtilis PB6 (BSP) on gastrointestinal tract permeability, metabolism, inflammation, and production parameters in periparturient Holstein cows. Multiparous cows (n = 48) were stratified by previous 305-d mature equivalent milk yield and parity and assigned to 1 of 2 top-dressed dietary treatments 21 d before expected calving through 63 DIM: (1) control (CON; 13 g/d calcium carbonate; n = 24) or (2) BSP (13 g/d BSP; CLOSTAT, Kemin Industries, Des Moines, IA; n = 24). Gastrointestinal tract permeability was evaluated in vivo using the oral paracellular marker chromium (Cr)-EDTA. Effects of treatment, time, and treatment × time were assessed using PROC MIXED of SAS version 9.4 (SAS Institute Inc.). Prepartum dry matter intake (DMI) was unaffected by treatment; however, BSP supplementation decreased postpartum DMI relative to CON (0.7 kg). Milk yield, energy-corrected milk (ECM), fat-corrected milk (FCM), and solids-corrected milk (SCM) increased in BSP cows compared with CON (1.6, 1.8, 1.6, and 1.5 kg, respectively). Decreased DMI and increased production collectively improved feed efficiency of milk yield, ECM, FCM, and SCM for BSP cows (6, 5, 5, and 5%, respectively). No treatment differences were observed for concentrations of milk fat, protein, total solids, somatic cell count, somatic cell score, body weight, or body condition score. Milk urea nitrogen concentrations decreased (5%), whereas milk protein and lactose yield increased (5 and 2%, respectively) with BSP supplementation. Prepartum fecal pH did not differ among treatments; conversely, postpartum fecal pH was increased with BSP supplementation (0.09 pH units). Prepartum fecal dry matter percentage, starch, acetic acid, propionic acid, butyric acid, and ethanol did not differ among treatments. Postpartum concentrations of the aforementioned fecal parameters were also unaffected by treatment, but fecal propionic acid concentration was decreased (24%) in BSP cows relative to CON. Circulating glucose, nonesterified fatty acids, l-lactate, and insulin were similar between treatments both pre- and postpartum. Prepartum β-hydroxybutyrate (BHB) did not differ between treatments, but postpartum BSP supplementation decreased (21%) circulating BHB relative to CON. Regardless of treatment, inflammatory markers (serum amyloid A and haptoglobin) peaked immediately following parturition and progressively decreased with time, but this pattern was not influenced by treatment. Postpartum lipopolysaccharide binding protein tended to be decreased on d 3 in BSP relative to CON cows (19%). Neither treatment nor time affected Cr-EDTA area under the curve. In summary, supplementing BSP had no detectable effects prepartum, but increased key postpartum production parameters. Bacillus subtilis PB6 consistently increased postpartum fecal pH and decreased fecal propionate concentrations but did not appear to have an effect on gastrointestinal tract permeability.

Nocturnal Pain Is Not an Alarm Symptom for Upper Gastrointestinal Inflammation but May Be an Indicator of Sleep Disturbance or Psychological Dysfunction

Alarm symptoms are widely used in pediatric gastroenterology to discern when abdominal pain needs further workup. Despite wide use, the data supporting the validity of these symptoms are not well established. This study explored one alarm symptom—nighttime waking with pain—and its associations with histologic inflammation of the upper gastrointestinal tract, psychological dysfunction, and disordered sleep. This retrospective study evaluated 240 patients with abdominal pain-related disorders of the gut–brain axis (AP-DGBI). Patients underwent questionnaires related to sleep disturbance, behavioral assessment, and gastrointestinal symptoms, including Rome IV criteria for AP-DGBI. Routine upper endoscopy with standardized biopsies was performed in 205 patients. Endoscopy results showed no association between esophageal, gastric, or duodenal histologic inflammation and nighttime waking with pain. Nocturnal pain was associated with increased scores for both psychological and sleep disorders, including social stress, depression, disorders of initiation and maintenance of sleep (DIMS), disorders of daytime somnolence (DOES), and sleep hyperhidrosis (SHY). This study concluded that nocturnal pain is not a reliable predictor of upper gastrointestinal inflammation but may be a prognosticator for psychological distress and sleep disturbances.

IL-1β/IL-1R1 signaling is involved in the propagation of α-synuclein pathology of the gastrointestinal tract to the brain.

The pathological hallmark of Parkinson's disease (PD) is the intraneuronal accumulation of misfolded alpha-synuclein (termed Lewy bodies) in dopaminergic neurons of substantia nigra par compacta (SNc). It is assumed that the α-syn pathology is induced by gastrointestinal inflammation and then transfers to the brain by the gut-brain axis. Therefore, the relationship between gastrointestinal inflammation and α-syn pathology leading to PD remains to be investigated. In our study, rotenone (ROT) oral administration induces gastrointestinal tract (GIT) inflammation in mice. In addition, we used pseudorabies virus (PRV) for tracing studies and performed behavioral testing. We observed that ROT treatments enhance macrophage activation, inflammatory mediator expression, and α-syn pathology in the GIT 6-week post-treatment (P6). Moreover, pathological α-syn was localized with IL-1R1 positive neural cells in GIT. In line with these findings, we also find pS129-α-syn signals in the dorsal motor nucleus of the vagus (DMV) and tyrosine hydroxylase in the nigral-striatum dynamically change from 3-week post-treatment (P3) to P6. Following that, pS129-α-syn was dominant in the enteric neural cell, DMV, and SNc, accompanied by microglial activation, and these phenotypes were absent in IL-1R1r/r mice. These data suggest that IL-1β/IL-1R1-dependent inflammation of GIT can induce α-syn pathology, which then propagates to the DMV and SNc, resulting in PD.

The ethanol extract of Scutellaria baicalensis Georgi attenuates complete Freund's adjuvant (CFA)-induced inflammatory pain by suppression of P2X3 receptor

Ethnopharmacological relevanceScutellaria baicalensis Georgi (SBG) is a perennial herb with anti-inflammatory, antibacterial, and antioxidant activities, which is traditionally used to treat inflammation of respiratory tract and gastrointestinal tract, abdominal cramps, bacterial and viral infections. Clinically, it is often used to treat inflammatory-related diseases. Research has shown that the ethanol extract of Scutellaria baicalensis Georgi (SGE) has anti-inflammatory effect, and its main components baicalin and baicalein have analgesic effects. However, the mechanism of SGE in relieving inflammatory pain has not been deeply studied. Aim of the studyThis study aimed to evaluate the analgesic effect of SGE on complete Freund's adjuvant (CFA)-induced inflammatory pain rats, and to investigate whether its effect on relieving inflammatory pain is associated with regulation of P2X3 receptor. Materials and methodsThe analgesic effects of SGE on CFA-induced inflammatory pain rats were evaluated by measuring mechanical pain threshold, thermal pain threshold, and motor coordination ability. The mechanisms of SGE in relieving inflammatory pain were explored by detecting inflammatory factors levels, NF-κB, COX-2 and P2X3 expression, and were further verified by addition of P2X3 receptor agonist (α, β me-ATP). ResultsOur results revealed that SGE can notably increase the mechanical pain threshold and thermal pain threshold of CFA-induced inflammatory pain rats, and markedly alleviate the pathological damage in DRG. SGE could suppress the release of inflammatory factors including IL-1β, IL-6, TNF-α and restrain the expression of NF-κB, COX-2 and P2X3. Moreover, α, β me-ATP further exacerbated the inflammatory pain of CFA-induced rats, while SGE could markedly raise the pain thresholds and relieve inflammatory pain. SGE could attenuate the pathological damage, inhibit P2X3 expression, inhibit the elevation of inflammatory factors caused by α, β me-ATP. SGE can also inhibit NF-κB and ERK1/2 activation caused by α, β me-ATP, and inhibit the mRNA expression of P2X3, COX-2, NF-κB, IL-1β, IL-6 and TNF-α in DRG of rats induced by CFA coupled with α, β me-ATP. ConclusionsIn summary, our research indicated that SGE could alleviate CFA-induced inflammatory pain by suppression of P2X3 receptor.

Inflammatory bowel disease (IBD) is the persistent inflammation of GIT (Gastrointestinal tract). IBD mainly divided into ulcerative colitis and Crohn’s disease while the Crohn’s disease involves the whole gastrointestinal tract while ulcerative colitis is limited to the colon. The current experimental study elucidates the antioxidative and microbial homeostatic efficacy of monolaurin, monolaurin with butyric acid and propionic acid in DSS (Dextran sulfate sodium) induced acute colitis in rat model. The results revealed that total antioxidant capacity was raised significantly (P<0.05) in all the treatment groups comparative with positive control group. While the results of total oxidative stress and relevant gene expression analysis revealed that mRNA expression of cell stress pathways such as, Traf-4, Traf-6 and MAPK-8 genes (Mitogen activated Protein Kinase) pathways and Calm-2, Gerk-2 and Pias-2 genes (Calcium signaling pathways) were significantly (P<0.05) higher expressed in the positive control group as compared to negative and all the treatment groups. While as, the total antioxidant capacity and NFR-1, Keap-1 and Nef-212 were significantly (P<0.05) down expressed in positive control group as compared to all the treatment groups. Histopathological study revealed that disruption of intestinal mucosa and immune infiltration in positive control group while, restoration and regeneration of intestinal parenchyma was observed in all the treatment groups. Overall study results concluded that monolaurin, monolaurin with butyric acid, and propionic acid can develop innovative customized remedies against DSS-induced acute colitis.
