Investigation of transferrin polymorphism in Garole sheep

The aim of this study was to determine the genetics of polymorph systems of Transferrin in Garole sheep breed. The present study was conducted on 95 adult Garole sheep comprising 52 ewes and 43 rams, maintained at Sheep and Goat Breeding Farm of West Bengal University of Animal and Fishery Sciences, West Bengal, during the period from April-September, 2009. The polymorphism of transferrin was determined through SDS-Polyacrylamide gel electrophoresis technique. It was found that the transferrin type was controlled by five codominant alleles (TfA, TfB, TfC TfD and TfE) in Garole sheep. These five alleles, because of co-dominant nature of inheritance, determined the occurrence of nine transferrin genotypes in the analyzed flock. Four (TfAA, TfBB, TfCC and TfDD) of these were homozygous and the remaining five (TfAD, TfBC, TfBD, TfCD and TfDE) heterozygous. It was found that the TfDD genotype (0.263) was predominant while TfDE genotype (0.042) was least common in the analyzed flock. Frequencies of other genotypes were as: TfCD(0.242), TfBD(0.126), TfCC(0.084), TfBB(0.074), TfAA(0.063), TfAD and TfBC (0.053 for each genotype ) in whole population. From the result it was found that in whole population combined, the heterozygotic genotypic frequency (0.516) was more than that of homozygotic genotypic frequency (0.484). Considerable variations were recognized in the frequencies of transferrin alleles. In the whole population frequencies of transferrin alleles were found to be TfA = 0.089, TfB = 0.163, TfC = 0.232, TfD = 0.495 and TfE = 0.021. Transferrin system has shown an absence of genetic equilibrium among the analyzed herd (chi2 value = 51.31). In conclusion, there were polymorphism in Transferrin types and the presence of differences among the frequencies of the five alleles by categories could be a source of genetic variation in Garole sheep.

Geographical variation of transferrin allele frequencies in three deer species from Scandinavia.

Blood samples from 871 moose (Alces alces L.), 91 red deer (Cervus elaphus L.), and 33 roe deer (Capreolus capreolus L.) from Scandinavia were analysed for serum transferrin variability by agarose electrophoresis. The patterns of variability suggest two alleles in the moose, three in the red deer, but only one in the roe deer. Spatial heterogeneity of allele frequencies was found in both variable species. Differences in the level of transferrin variation among closely related species of Cervidae may be related to the social structure of each species and its effects on the transmission of disease organisms.

SELECTION IN A CYCLING POPULATION: DIFFERENTIAL RESPONSE AMONG SKELETAL TRAITS

Population density cycles influence phenotypic evolution through both density-dependent selection during periods of high density and through enhanced genetic drift during periods of low density. We investigated the response of different phenotypic traits to the same density cycles in a population of the yellow-necked mouse, Apodemus flavicollis, from Białowieza National Park in Poland. We examined nonmetric skull traits, skull and mandible size, skull and mandible shape, and transferrin allele frequencies. We found that all of the traits changed significantly over the seven-year study period. The greatest changes in nonmetric traits and mandible size occurred during periods of increasing density, and the magnitude of changes in skull and mandible shape was correlated with the magnitude of density changes. Frequencies of transferrin alleles changed the most when population density was in decline. Changes among the five phenotypic traits were generally uncorrelated with one another, except for skull and mandible shape. Nonmetric traits were selectively neutral when assessed with Q(ST)/F(ST) analysis, whereas mandible size, mandible shape, and skull shape showed evidence of fairly strong selection. Selection on skull size was weak or nonexistent. We discuss how different assumptions about the genetic components of variance affect Q(ST) estimates when phenotypic variances are substituted for genetic ones. We also found that change in mandible size, mandible shape, skull size, and skull shape were greater than expected under a neutral model given reasonable assumptions about heritability and effective population size.

제주마 Transferrin Gene Exon 13, 15 및 16의 다형현상

본 연구는 제주마집단(GroupⅠ, 제주도 축산진흥원 사육, 137두; Group II, 농가사육, 30두)과 더러브렛 품종집단(한국마사회 육성마목장, 43두)을 이용하여 SSCP를 통한 Transferrin exon 13, 15, 16의 다형현상 확인과 각 SSCP 유전자형의 염기서열을 분석하기 위하여 수행하였다. 공시재료에서 SSCP에서 관찰된 band에 의한 분석결과 대립인자는 exon 13, 15 및 16에서 각각 2개(A,B), 3개(A,B,C) 및 3개(A,B,C)가 존재하는 것으로 확인되었다. Transferrin exon 13에서 제주마와 더러브렛 집단 모두 A인자가 매우 높게 분포하고 있음이 확인되었다. exon 15에서는 그룹간의 빈도차를 확인 할 수 있었다. exon 15에서 높게 출현되고 있는 유전자형은 GroupⅠ에서 AB (0.445)형, GroupⅡ에서 AA(0.367)형, 더러브렛 품종에서는 AA(0.767) 유전자형이 가장 높은 빈도로 출현되어 제주마 집단간 또는 품종간에 빈도의 차이를 관찰할 수 있었다. exon 16에서는 GroupⅠ은 A, B, C 인자, GroupⅡ에서는 A 및 B 2종류의 인자형이 확인되었고 더러브렛 품종에서는 A인자형만 검출되었다. exon 16에서도 그룹간에 유전인자의 빈도차를 확인 할 수 있었다. 또한 exon 13, 15 및 16의 조합으로 형성된 개체의 유전자형은 전체 13종류가 출현되었고 이 조합도 그룹간 차이를 확인 할 수 있었다. SSCP 유전자형에 따른 각 인자들에 대한 염기서열을 분석한 결과 exon 13과 16에서 각 1개의 새로운 SNP가 발견되었다. 본 연구결과 제주마 transferrin exon 13, 15, 16은 더러브렛 품종에서와 같이 높은 대립인자의 다형성을 보였으며, 각 Group 간 빈도차를 확인 할 수 있었다. This study was conducted to determine the polymorphism of transferrin exons 13, 15 and 16 by Single-Strand Conformation Polymorphism(SSCP) analysis and to compare their genotypes of Cheju horse Group I (Cheju Institute), Cheju horse Group II (farms), and Thoroughbred (KRA). SSCP of transferrin exon 13, 15, and 16 showed two (A, B), three (A, B, C) and three (A, B, C) codominant alleles, respectively. The Group I and Thoroughbred showed the similar frequencies of allele A and B in transferrin exon 13, but only allele A was observed in Group Ⅱ. In transferrin exons 15 and 16, the frequencies of each allele were different in each Groups. The multiple allele frequencies in exons 15 and 16 suggested that the genotyping of this locus could be used to identify an individual and to test the parentage of offspring. The probability for parentage exclusion were 0.46 and 0.374 for exons 15 and 16 for Cheju horse Group I. Among the 13 combined genotypes of exons 13, 15 and 16, the genotype AA-AB-AB (0.372) is the most common in Cheju horse Group I, but genotype AA-AA-AA is common in the Cheju horse Group II (0.366) and Thoroughbred (0.767). The present study showed two new SNP, which was at the cDNA position 1626 (A/G) in B allele of the exon 13 and 2075 (C/T) in C allele of the exon 16 resulting in amino acid change (Threonine <TEX>$\longrightarrow$</TEX> Methionine). Result showed that polymorphism of exons 13, 15 and 16 in Cheju horses was as high as in Thoroughbred and there was a differences of transferrin allele frequencies in Cheju horses.

Postglacial dispersal of longnose suckers, Catostomus catostomus, in the Mackenzie and Yukon Drainages

The role played by stream transfers and reversals in the postglacial dispersal of fish populations in the Yukon and Mackenzie River drainages of Canada was analysed using genetic and meristic data from longnose sucker ( Catostomus catostomus Forrester) populations. The hypothesis the suckers dispersed from the Yukon River into the Mackenzie River via the Peel River was evaluated. Meristic data (gill rakers and lateral line scales) were uninformative for this analysis. Genetic affinity between the Yukon River and upper Peel River populations, and between Mackenzie and lower Peel River populations is suggested by transferrin allele frequencies. These affinities support the view that longnose suckers inhabiting the middle and upper regions of the Peel River are derived from Beringian and Mississippian ancestors, while suckers in the lower Peel are descended from Mississippian stock. Evidence for dispersal by way of the Eagle River was not found. Affinities of sucker populations in other river systems in the region are also described.

Genetic variation in transferrin as a predictor for differentiation and evolution of caribou from eastern Canada

Polycrylamide gel electrophoresis was used to analyse tranferrrin variation in caribou populations from Manitoba, Ontario, Qu&amp;eacute;bec/Labrador, and from Baffin Island, Northwest Territories in eastern Canada. The transferrin allele frequencies in these populations were compared with those previously reported for Canadian barren-ground caribou, Rangifer tarandus groenlandicus, Alaska caribou, R.t. grand, Peary caribou, R.t. pearyi, Svalbard reindeer, R.t. pla-tyrhynchus, and Eurasian tundra reindeer, R.t. tarandus. A total of twenty different alleles was detected in the analysed material, considerable genetic heterogeneity being detected among regions. Three alleles that were relatively common in caribou from Ontario, Manitoba and Qu&amp;eacute;bec/Labrador, were not present in R.t. grand, R.t. pearyi, R.t. tarandus or R.t. platyrhynchus, and present only at very low frequencies 'm R.t. groenlandicus. These findings, together with genetic identity analyses, suggest that the caribou in Manitoba, Ontario, and Qu&amp;eacute;bec/Labrador are mainly of the R.t. caribou type, and that little interbreeding has occurred with other subspecies. The large genetic distance in the transferrin locus between R.t. caribou and other subspecies of reindeer/caribou suggests that, during the Wisconsin glaciation the ancestral populations of R.t. caribou survived in a refugium different from that of the ancestral populations of the other subspecies. Significant genetic differences between Baffin Island caribou and all other populations were mainly due to the presence of one allele that was in high frequency in Baffin Island caribou, but that was absent, or present in very low frequencies, in all other reindeer/caribou populations. The genetic differences between Baffin Island caribou and the other subspecies were greater than the differences between several of the currently recognized subspecies.

Spatial Correlations of Transferrin Allele Frequencies in Atlantic Salmon (Salmo salar) Populations from North America

Existing analyses of spatial patterns of transferrin (Tf) allele frequency variation in anadromous Atlantic salmon (Salmo salar) from rivers in North America have not utilized all available data. Further analysis shows that a latitudinal dine in Tf-4 allele frequency previously reported is an artefact of a major regional difference between populations from Newfoundland/Labrador and those sampled elsewhere. Furthermore, the allele frequencies of populations outside Newfoundland/Labrador show a significant association with the proportion of spawning fish in the populations having spent only one winter at sea. Fequencies in Newfoundland/Labrador are collectively consistent with this relationship but by themselves show no similar correlation. These results, combined with previously reported differences between anadromous and nonanadromous fish, suggest that selective forces, which vary with life history pattern, may operate on the polymorphism.

Studies were made on Apodemus agrarius inhabiting three different urban and suburban environments. Statistically significant seasonal differences in transferrin allele frequencies were observed only in the field population. The greater number of heterozygotes in the island population than in the others in spring was followed by large numbers of mice there in autumn.

On the basis of 3-year studies transferrin polymorphism in Apodemus agrarius in chosen differently isolated urban (Warsaw), suburban and field (Turew) populations it was stated as follows: (a) a lack of differences in variability of transferrin related to sex, sexual activity and season (summer-early autumn), (b) differences in frequencies of transferrin alleles between populations during two years of studies when the gradient of genetic variability in populations was opposite to gradient of isolation and urbanization of habitat studied, (c) significantly different frequencies of transferrin alleles from year to year in isolated population from the city centre.

It was suggested that the higher frequencies of common transferrin allele in female voles and male mice might indicate different selection in each sex of the two genera (as a result of different reproduction strategies). The increase in heterozygote number causing the decrease in common transferrin allele frequencies were observed during late summer in all three species.

Partial characterization of horse transferrin heterogeneity with respect to the atypical type, Tf C.

In starch gel electrophoresis of horse sera each transferrin variant is formed by a strong anodal band and a weaker cathodal band. An 'atypical' Tf C, has two zones of about equal intensity. Family data show that Tf C is genetically controlled by an allele Tf C at the Tf locus. Frequencies of transferrin alleles in various horse breeds are also presented. After isolation and fractionation of individual transferrin variants (Tf O, Tf D, Tf C) on DEAE-Sephadex, additional weak bands were detected. The two main zones of each variant were isolated in a pure state and treated with neuraminidase. In all three variants studied the electrophoretic mobility of the slower band (2a) was decreased in two steps, and the faster band (4b) in four steps. The mobilities of hands derived from the fast zone (4b) were slower than mobilities of corresponding bands derived from the slow zone (2a). These results suggest the presence of two sialic acid residues in the slow zone, and of four residues in the fast zone. Residual heterogeneity was independent of sialic acid.

Spatial Variation in Transferrin Allele Frequencies among Herds of Feral Donkeys in Death Valley National Monument, California

Serum proteins were studied to clarify interrelationships of seemingly disjunct herds of feral donkeys ( Equus asinus ) in the mountains of Death Valley National Monument, California. Sera of 162 donkeys from five localities in the Panamint Mountains were examined by starch-gel electrophoresis to assess polymorphic variation in transferrins. Four homozygous and six heterozygous transferrin phenotypes controlled by four codominant, autosomal alleles were identified with Fe59 autoradiography. Significant interlocality heterogeneity in genotype distribution and allele frequency distribution among herds was demonstrated. The maximum distance between adjacent herds was 52 km and the minimum distance was 14 km. There are no major physical barriers to movements between herds; furthermore, donkeys are highly mobile animals capable of extensive travel. Behavioral isolation of adjacent herds occurs from May through July, during the peak of reproduction. This isolation is reinforced by fidelity of individuals to their respective home ranges, and to physiological dependence on permanent sources of water. Adjacent herds may overlap during winter when breeding and dependence on water sources are reduced.

Allele Frequency Analysis of Five Soluble Protein Loci in Brook Trout, Salvelinus fontinalis (Mitchill)

Abstract Phenotypes for five soluble protein loci (serum transferrin, lactate dehydrogenase, and three eye lens proteins) in native brook trout, Salvelinus jontinalis (Mitchill), were determined by either acrylamide or starch gel electrophoresis. An allele frequency analysis involving these five loci was performed and the resultant allele frequencies were compared (chi-square of contingency) with results of a similar study conducted on samples from the same streams in 1966. All intra-stream, inter-year heterogeneities of the serum transferrin allele frequencies were found to be statistically insignificant. Intra-stream, inter-year heterogeneities of the three lens protein loci were statistically insignificant except for one population. In this one population the dominant allele (LP5 had become fixed, which seems to be the most frequent condition in natural populations. The transferrins and hemoglobins of wild Iranian sheep Ovis linnaeus

1.1. Serum transferrins and hemoglobins were analyzed by starch-gel electrophoresis from ninety-seven wild sheep collected from eight localities across northern Iran and from two Iranian domestic sheep.2.2. Comparison of transferrins from the wild sheep with domestic sheep reference sera demonstrated Tf alleles A, B, C, D and E in the former; new transferrin alleles designated Tf's A+,B+ and E− were also observed.3.3. Transferrin allele frequencies provided a means for differentiating wild sheep populations of northwestern Iran from those of northeastern Iran; natural hybrids displayed transferrin frequencies of an intermediate nature. Applications of the transferrin data to some taxonomic problems and to the origin of domestic sheep are discussed.4.4. Iranian wild sheep were found to possess Hbg B only.
