Novel fast dissolving freeze dried sublingual baicalin tablets for enhanced hepatoprotective effect: in-vitro characterization, cell viability, and in-vivo evaluation

Baicalin (BG), a natural product, has been used in the prevention and treatment of drug-induced liver injury (DILI); however, its poor solubility and extensive liver metabolism limit its pharmacological use. The aim of the present study was the formulation of fast-dissolving freeze-dried sublingual tablets (FFSTs) to increase BG dissolution, avoid first-pass metabolism, and overcome swallowing difficulties. FFSTs were prepared following a 23 factorial design. The effect of three independent variables namely matrix former, Maltodextrin, concentration (4%, and 6%), binder concentration (2%, and 3%), and binder type (Methocel E5, and Methocel E15) on the FFSTs’ in-vitro disintegration time and percentage dissolution was studied along with other tablet characteristics. Differential scanning calorimetry, scanning electron microscopy, in-vitro HepG2 cell viability assay, and in-vivo characterization were also performed. F8 (6% Maltodextrin, 2% Mannitol, 2% Methocel E5), with desirability of 0.852, has been furtherly enhanced using 1%PEG (F10). F10 has achieved an in-vitro disintegration time of 41 secs, and 60.83% in-vitro dissolution after 2 min. Cell viability assay, in-vivo study in rats, and histopathological studies confirmed that pretreatment with F10 has achieved a significant hepatoprotective effect against acetaminophen-induced hepatotoxicity. The outcome of this study demonstrated that FFSTs may present a patient-friendly dosage form against DILI.

Current research aimed to prepare and estimate chronology-based mucoadhesive PDDS of antihypertensive drug losartan potassium through formulation of fast-dissolving core tablet and incorporation of core tablet to polymer coating to formulate bioadhesive PDDS through direct compression system. The coating was done by using polymers ethyl cellulose and carbopol 934. Pre-compression and post-compression parameters, drug release, lag time and mucoadhesive examination was assessed for the formulation. The expected lag time for hypertension is 8 hours; hence, this lag time was achieved by using a bioadhesive pulsatile system. The optimized formulation showed 8 hours lag time with appropriate mucoadhesion for an equivalent period. Moreover, in-vitro as well as in-vivo mucoadhesion investigations reflected the positive outcomes. Consequently, the BPDDS was the finest preventative substitute for drugs with the highest absorption in the stomach and for drugs used to treat diseases related to circadian rhythm

A Comparative Study on Cyclodextrin Derivatives in Improving Oral Bioavailability of Etoricoxib as a Model Drug: Formulation and Evaluation of Solid Dispersion-Based Fast-Dissolving Tablets.

Etoricoxib, as a model drug, has a poor solubility and dissolution rate. Cyclodextrin derivatives can be used to solve such a problem. A comparative study was run on three cyclodextrin derivatives, namely β-CD, HP β-CD, and SBE β-CD, to solve the drug problem through the formulation of solid dispersions and their preparation into fast-dissolving tablets. Preparations utilized different (1:1, 1:2, and 1:4) drug:carrier ratios. Nine fast-dissolving tablets (containing 1:4 drug: carrier) were formulated using Prosolv ODT® and/or F-melt® type C as super-disintegrants. Optimized formulation was chosen based on a 32 factorial design. The responses chosen were the outcomes of the in vitro evaluation tests. The optimized formulation that had the highest desirability (0.86) was found to be SD-HP3, which was prepared from etoricoxib: HP β-CD at a 1:4 ratio using equal amounts of Prosolv ODT® and F-melt® type C. An in vivo evaluation of SD-HP3 on a rabbit model revealed its superiority over the marketed product Arcoxia®. SD-HP3 showed a significantly lower Tmax (13.3 min) and a significantly higher Cmax (9122.156 μg/mL), as well as a significantly higher AUC, than Arcoxia®. Thus, the solubility, dissolution, and bioavailability of etoricoxib were significantly enhanced.

Evaluation of Co-Processed Excipients for Fast Disintegration of Aspirin Tablets Prepared by Direct Compression

Background: Co-processed excipients are popular for direct tablet compression and give added economic value to product formulation. Specific benefits of co-processed excipients include improved flow, compressibility, disintegrating effect, and masking undesirable properties of individual excipients.
 Objective: The aim of this study is to co-process physically modified (pregelatinized) Musa sapientum starch with lactose by fusion and evaluate the effect of the co-processed excipients on the disintegration properties of Aspirin tablet.
 Methods: Co-processed excipients were prepared from pregelatinized Musa sapientium starch and lactose by fusion in ratios of 1:1, 1:2 and 2:1 (BL1, BL2, and BL3 respectively) and evaluated for particle size, pH, moisture content, flow and swelling properties. Aspirin tablets were formulated using the excipients at ratio 5% W/W by direct compression. (FM1, FM2, FM3 respectively). Sodium starch glycolate (FSG) tablet was also prepared as the standard and the formulations were assessed for hardness, friability and disintegration time. Data were analyzed using ANOVA.
 Results: The co-processed excipients possessed excellent flow having a Carr’s index between 18.39-20.78 and Hausner’s ratio ≤ 1.25.BL1 had the highest swelling profile while BL3 had the lowest. Formulation FM1 and FM2 had the highest tensile strength of (0.16 N/cm2). Disintegration time of FM3 (4.28 min) was comparable to that of FSG (4.15 min).
 Conclusion: Co-processing pregelatinized Musa sapientium starch and lactose by fusion influenced tablet disintegration and has potential to be used in manufacture of fast dissolving tablet formulations.

Quality by design (QbD) approach to develop fast-dissolving tablets using melt-dispersion paired with surface-adsorption method: formulation and pharmacokinetics of flurbiprofen melt-dispersion granules.

Developing amorphous solid dispersions with good flow properties is always challenging for formulation scientists to convert into tablets. Hence, the present study investigates the impact of the combination of melt-dispersion and surface-adsorption methods to prepare melt-dispersion granules with enhanced dissolution rate and flow properties. This study covers the formulation and pharmacokinetic study of fast-dissolving flurbiprofen tablets using PEG 6000 (hydrophilic carrier) and lactose (adsorbent). Response surface methodology (RSM) using the central composite design (CCD) was used to optimize independent variables like carrier concentrations and adsorbent concentrations, and their interactions with the dependent variables (responses), including solubility, angle of repose, Carr's index, and cumulative % drug release, were investigated. The optimized formulation was selected based on the numerical optimization method and further investigated for FTIR spectroscopy, differential scanning calorimetry, and X-ray diffractometry. Then, the optimized formulation was compressed into tablets and evaluated for both in vitro dissolution and in vivo pharmacokinetics parameters. In vitro dissolution studies revealed that the prepared fast-dissolving tablets released the drug entirely within 15min (Q15 of F4 tablets: 99.34 ± 1.24%), whereas conventional tablets took around 60min for complete dissolution. Pharmacokinetic studies in rats revealed that fast-dissolving tablets showed 1.38-fold higher peak-plasma concentration (Cmax) and 1.39-fold higher bioavailability than conventional tablets. Overall, this study revealed the successful fabrication of fast-dissolving tablets via melt-dispersion paired with the surface-adsorption method to enhance the flow properties and the dissolution rate.

Efficacy of jackfruit components in prevention and control of human disease: A scoping review.

The jackfruit (Artocarpus heterophyllus) is one of the natural remedies significantly used in folk medicine. The ethnopharmacological applications of jackfruit are mainly concerned with the management of inflammation, diarrhea, and diabetes mellitus. Flavonoids, stilbenoids, aryl benzofurans, and lectin jacalin are abundant in jackfruit species. Jacalin is a good indicator for evaluating the immunological state of HIV-1 patients. The extracts and metabolites of jackfruit, particularly those from the leaves, bark, stem, and fruit, contain several beneficial bioactive mixtures. New studies are focused on exploring these bioactive compounds used in various biological activities such as antiviral, antiplatelet, anticancer, antiatherosclerotic, immunomodulatory effects, inhibitors of 5-alpha reductase activity, and the formulation of fast-dissolving tablets (orodispersible, rapid melts porous). Multidisciplinary programs that integrate traditional and modern technology play a crucial role in the lies ahead expansion of jackfruit as the prospective inception of therapeutic compounds. This review aims to highlight significant results on the identification, production, and bioactivity of metabolites found in jackfruit, with current developments in jackfruit research in the control and prevention of human diseases.

Quality by Design approach for Optimization and Development of Cyclodextrin-Surfactant Complex Based Formulations for Bioavailability Enhancement of Valsartan

The main objective of the present research is to increase the oral bioavailability of Valsartan by inclusion complexes (ICVs) with a cyclodextrin-surfactant combination followed by the formulation of fast-dissolving tablets (FDTs). The solvent evaporation method was used for the preparation of ICVs. Methyl-ß-cyclodextrin and Hydroxypropyl-ß- cyclodextrin were evaluated with the combination of poloxamer 188 to get the formulations with the desired solubility. Central composite design (CCD) was used as the experimental design as a part of the quality by design (QbD) approach. The optimized ICVs were further developed into FDTs by direct compression technique. Taking concentration of povidone, type and concentration of disintegrant as the formulation factors, the FDTs were optimized using CCD. In-vivo bioavailability study in rats was performed for the optimized FDTs against the marketed tablets. The optimized ICVs were found to have a 3.12 mg/mL solubility. The optimized FDTs were found to be disintegrated in 18.7 sec and dissolved 90% of the dose in 6.3 min. The In-vivo results indicated that the FDTs exhibited rapid absorption and an increase in bioavailability by 24.1% against the marketed tablets. The results indicated that the QbD approach successfully improved Valsartan's oral bioavailability through cyclodextrin-surfactant complexation.

FORMULATION AND EVALUATION OF FAST DISSOLVING TABLETS OF ACECLOFENAC USING NATURAL SUPERDISINTEGRANT

On contact with saliva, FDTs (Fast dissolving tablets) which are designed to disintegrate rapidly, enables tremendous oral administration without contact with water. Also chewing these formulations produces an increased convenience and ease of administration with a significant potential efficacy to improve the patient compliance, predominantly in certain populations who were facing difficulties for swallowing the conventional solid oral dosage forms. In our present study, the effect of a natural superdisintegrant namely, Fenugreek and some synthetic superdisintegrants such as SSG (Sodium Starch Glycolate), MCC (Micro Crystalline Cellulose), CP (Cross Povidone) and CCS (Cross Carmaellose sodium) were employed for this study to produce the formulations of FDTs. By direct compression method, FDTs of Aceclofenac were prepared and evaluated as per IP standards. From our study it was confirmed that Fenugreek showed excellent swelling index than the synthetic superdisintegrants. Hence the present study reveals that this natural superdisintegrants showed significant disintegration.

Preclinical Safety and Pharmacokinetics of Heat Stable Oxytocin in Sublingual Fast-Dissolving Tablet Formulation.

The work reported here focuses on an evaluation of a novel heat stable formulation of a uterotonic peptide drug oxytocin involving stability testing under elevated temperatures and toxicokinetic response generated by sublingual (SL) administration in rabbits. The formulation was thermotolerant, maintaining the potency of oxytocin in the form of a fast-dissolving tablet at the end of 2-year storage at 30 °C/65% relative humidity with less than 5% loss in oxytocin content based on analytical high performance liquid chromatography (HPLC). The toxicokinetic results in rabbits showed that the fast-dissolving tablet was safe without any reactogenicity or toxicity associated with SL administration or the excipients present in the formulation. The SL route elicited rapid absorption of oxytocin in plasma within 5 min of administration although lower than intramuscular (IM) administration. IM resulted in area under the curve (AUC) values approximately 5 times higher than SL oxytocin. However, due to the limitations encountered during SL administration in an anesthetized rabbit model, the relevance of heat stable oxytocin formulation that has the flexibility to be adapted in different formats may warrant a human clinical study to determine whether therapeutically relevant plasma levels for treating postpartum hemorrhage can be generated via alternate non-injectable routes of administration.

Design and development of fast dissolving liquisolid formulation

Various techniques have been employed for the solubility modification of BCS class II drugs. Out of the recent solubility enhancement techniques, Liquisolid technique based on the work of Spireas et al has explored as method that can supplement drug release. Formulations containing Etodolac were developed by dissolving the drug in a non-volatile solvent and then employing the adsorption absorption principle with a carrier and coated material admixture. The post formulation saturation solubility study has highlighted significant modification in solubility of the drug in comparison to pre solubility modification. The Liquisolid formulation then subjected to direct compression for the development of Fast dissolving tablet formulation using two varying concentrations of Sodium Starch Glycolate. The in vitro - in vivo evaluation of research study concludes adaptability and applicability of fast dissolving liquisolid tablet formulation allowing it to overcome barriers associated with the solubility. The optimized formulation found to unveil dissolution along with diffusion from the dosage form by Fickian mechanism. FORMULATION AND IN VITRO–IN VIVO PHARMACOKINETIC EVALUATION OF CARDIOVASCULAR DRUG-LOADED PULSATILE DRUG DELIVERY SYSTEMS

Objective: This study is to formulate Nebivolol into a Pulsatile liquid, solid composite compression coated tablet, which will delay the release of the drug in early morning hypertension conditions. Methods: The liquid, solid composite tablet was formulated and compressed with the ethylcellulose coating polymer. The percent in vitro drug release of the liquid solid composite compressed tablet was tested. Based on disintegration time and wetting time, the LCS2, LCS3, LSC6, LCS7 and LCS12 formulations were found to be the optimized solid-liquid compacts fast-dissolving core tablet formulations, which may be excellent candidates for further coating with polymer to transfer into press coated pulsatile tablet formulations. Coating the core tablet with varying ethyl cellulose concentrations resulted in five different formulations of the pulsatile press-coated tablet (CT1, CT2, CT3, CT4, CT5). In vitro drug release, in vitro release, kinetic studies, in vivo pharmacokinetic and stability tests were all performed for the prepared pulsatile press coated tablet. Results: CT3 tablets are coated with ethyl cellulose polymer, which shows maximum controlled drug release from the core tablet i.e. 96.34±1.2% at 8th h. It shows there was an efficient delay in drug release form core tablet i.e. up to 3 h, followed by the maximum amount of drug release of 96.34±2.4 at 8h. Which shows the core drug will be more efficiently protected from the gastric acid environment 1.2 pH, duodenal environment 4.0 pH and release drug only in the small intestine. Conclusion: According to the findings, CT3 Pulsatile press-coated tablet increased the bioavailability of Nebivolol by 3.11 percent.

In the present projected study, the effect of natural Super disintegrants was compared with synthetic Super disintegrants and conventional Super disintegrants in the of fast dissolving tablet formulation of Meclofenamate Sodium. Meclofenamate sodium NSAID is used for the treatment of mild to moderate pain in various conditions like (e.g., dental pain, osteoarthritis) and to decrease pain and blood loss during menstrual periods. It is also used for other treatments like reducing pain, swelling, and joint stiffness caused with rheumatoid arthritis. In the present work 9 formulations of FDT (Fast dissolving tablet) of Meclofenamate Sodium were prepared by using Super disintegrants was evaluated and compiles with the official parameters and specifications. Various formulations were prepared using four different super disintegrants namely natural super disintegrant Banana Powder, sodium starch glycolate, crosscarmelose sodium with three concentrations (2%, 4%, 6%) by direct compression method. The blend was evaluated for pre-compression parameters like Angle of repose, bulk density, tapped density, and then tablet evaluated with various post-compression parameters like thickness, drug content, hardness, weight variation, wetting time, friability, disintegration time, dissolution time, drug release study. Formulation F2 showed the lowest disintegration time and in-vitro dissolution studies recorded that formulation F2 showed 98.55% drug release at the end of 3 minutes. The best formulations among these were also found to be stable and optimized formulations were subjected to the stability studies as per ICH guideline.

Evaluation of Novel co-processed excipient for fast disintegration of aspirin tablet formulations

Co-processing is a technique that ensures sub-particulate interaction of individual excipients leading to overall functionality of the resulting excipient. The aim of this study is to co-process Cyperus esculentus starch with mannitol by fusion and evaluate its effect on tablet disintegration and in vitro dissolution. Co-processed excipients were prepared from Cyperus esculentus starch and mannitol by fusion in ratios of 1:1, 1:2 and 2:1 (CM1, CM2, and CM3 respectively) and evaluated for flow and swelling properties. The excipients were incorporated into Aspirin tablet formulations at 5 %w/w by direct compression (FM1, FM2, FM3 respectively). Similar tablets were prepared using sodium starch glycolate (FSG) and the formulations were assessed for hardness, friability, wetting time, disintegrationtime and in vitro dissolution profile. All the prepared excipients possessed excellent flow with Carr’s index between 17.31 and 20.78 and Hausner ratio between 1.21 and 1.26. CM3 had the highest swelling profile (1.491) while CM2 had the lowest (1.321). Formulation FM1 had the highest tensile strength (14.12 N/cm2 ) but slower wetting time (34.33 sec) compared to FM3 with tensile strength of 11.32 N/cm2 and wetting time of 9.00 sec. Disintegration time of CM3 (4.26 min) was comparable to that of FSG (4.01 min); their dissolution profile was also found to be similar. Coprocessing Cyperus esculentus starch and mannitol by fusion (2:1) influenced tablet disintegration and in vitro dissolution and has potential to be used in manufacture of fast dissolving tablet formulations.

In the present study, the effect of superdisintegrant was compared with synthetic super disintegrants and other conventional super disintegrants in the of fast dissolving tablet formulation of Meclofenamate.
 Meclofenamate is an antihypertensive drug and in case of hypertension immediate treatment is required so the proposed investigation is totally based to provide the suitable treatment for hypertension.
 In the present work 9 formulations of Fast dissolving tablets of Cilnidipine were prepared by using Synthesized Co-proceed was evaluated and compiles with the official standards, parameters and specifications.
 Various formulations were prepared using four different superdisintegrant namely- kyron T-304, sodium starch glycolate, cross carmelose sodium with three concentrations (2%, 4%, 6%) by direct compression method.
 The blend was evaluated for pre-compression parameters like Angle of repose , bulk density , tapped density , and then tablet evaluated post-compression parameters like thickness , drug content , hardness , weight variation , wetting time , friability , disintegration time , dissolution time, drug release study.
 Formulation A8 showed the lowest disintegration time and in-vitro dissolution studies recorded that formulation A8 showed 98.64% drug release at the end of 3 minutes.
 The best formulations were also found to be stable and optimized formulations were subjected to the stability studies as per ICH guideline and standards.

Optimization and Quality by Design Approach for Piroxicam Fast Dissolving Tablet Formulations Using Box-Behnken Design

Background: The present study deals with the formulation and optimization of piroxicam fast dissolving tablets and analyzes the impact of an independent variable while selecting the optimized formulation utilizing Quality by Design (QbD) and Box-Behnken Design (BBD). Methods: Seventeen formulations were prepared by direct compression technique by altering the proportion of cross carmellose sodium, spray dried lactose and hydro propyl methyl cellulose (HPMC K4M). The BBD statistical technique was used to optimize formulations and correlate the relationship among all the variables. Also, the powder mixture characteristics and tablet physiochemical properties such as hardness, friability, drug content, Disintegration Time (DT) and dissolution test were determined using 900 ml of 0.1N HCl (pH-1.2) at 37 ± 0.5°C. Results: Significant quadratic model and second order polynomial equations were established using BBD. To find out the relationship between variables and responses, 3D response surface and 2D contour plot was plotted. A perturbation graph was also plotted to identify the deviation of the variables from the mean point. An optimized formula was prepared based on the predicted response and the resulting responses were observed to be close with the predicted value. Conclusion: The optimized formulation with the desired parameter and formulation with variables and responses can be obtained by BBD and could be used in the large experiment with the involvement of a large number of variables and responses.

DESIGN AND FORMULATION DEVELOPMENT OF FAST-DISSOLVING TABLETS OF IBUPROFEN USING NOVEL NATURAL SUPERDISINTEGRANT

Objective: The objective of the study was to evaluate Ocimum gratissimum mucilage as a novel superdisintegrant in the formulation of fast-dissolving tablets (FDT) of Biopharmaceutical Classification System-II drug (Ibuprofen) employing a 23 factorial design.
 Methods: O. gratissimum mucilage was extracted by seeds and it was subjected to physical, chemical, and micrometric studies were evaluated. To establish FDT of ibuprofen with O. gratissimum mucilage as a superdisintegrants in different ratios using direct compression method employing 23 factorial design. All the formulation tablets were evaluated pre-compression and post-compression parameters like dissolution efficiency (DE%) percent of drug dissolved at 5 min.
 Results: The mucilage was to be found fine, free-flowing crystalline powder, and excellent swelling nature in all suitable solvents and buffers. The Fourier transform infrared and differential scanning calorimetry studies were indicated to no interactions between ibuprofen and O. gratissimum mucilage. All the FDT formulated employing novel mucilage shows good quality with regard drug content (98.05±0.31–99.39±0.54), hardness (3.6– 4 kg/sq. cm), and friability (0.12–0.15%). The optimized formulation batch shows less disintegrant time (30±0.06). In vitro wetting time was less (i.e., 90 s) in optimized formulation F2. The water absorption ratio of the formulated tablets was found to be in the range of 99±0.56. The cumulative drug dissolved in the optimized formulation F2 was found to be 99% in 10 min.
 Conclusion: O. gratissimum mucilage was found to be a novel superdisintegrant which enhanced the DE when combined with crospovidone and croscarmellose sodium; hence, it could be used in the formulation of FDT to provide immediate release of the contained drug within 5 min.

FORMULATION AND EVALUATION OF STATISTICALLY DESIGNED IBUPROFEN FASTDISSOLVING TABLETS EMPLOYING STARCH GLUTAMATE AS A NOVEL SUPERDISINTEGRANT

Objective: The main aim of the present work is to enhance the solubility and bioavailability of the ibuprofen by formulating it into fast-dissolving tablets employing starch glutamate as a novel superdisintegrant.
 Materials and Methods: Starch glutamate was prepared from native potato starch and glutamic acid by the esterification process. Drug-excipient compatibility studies were performed between the starch glutamate and ibuprofen with the help of Fourier transform infrared spectroscopy, and differential scanning calorimetry techniques. Ibuprofen fast dissolving tablets were formulated employing different superdisintegrants along with the starch glutamate (a novel superdisintegrant) by the direct compression method. The prepared ibuprofen fast-dissolving tablets were evaluated for various pre- and post-compression parameters along with the in vitro and in vivo release characteristics. Optimized formulation stability studies were performed at accelerated conditions for 6 months as per the International Conference on Harmonization (ICH) and WHO guidelines.
 Results: Drug-excipient compatibility studies indicated that prepared starch glutamate was compatible with ibuprofen drug, and it can be used as a superdisintegrant in the formulation of fast-dissolving tablets. Fast-dissolving tablets of ibuprofen were formulated by employing starch glutamate as a superdisintegrant showed good tablet properties and showed an increased dissolution efficiency of the drug. Among all the formulations (F1–F8), the formulation F4 which contains 5% starch glutamate and 5% croscarmellose sodium as superdisintegrants showed 99.7±0.34% drug dissolution within 5 min. Peak plasma concentration of optimized formulation F2 was achieved in a short period of time and increased relative bioavailability and F2 was found to be stable during accelerated stability testing as per the ICH stability guidelines.
 Conclusion: From drug-excipient compatibility studies, disintegration time, in vitro dissolution studies, and pharmacokinetic studies, it was concluded that starch glutamate can be used as a superdisintegrant in the formulation of fast-dissolving tablets to increase the solubility as well as bioavailability of the poorly soluble drugs.
