Development of an Elementary Reaction-Based Kinetic Model to Predict the Aqueous-Phase Fate of Organic Compounds Induced by Reactive Free Radicals.

ConspectusAqueous-phase free radicals such as reactive oxygen, halogen, and nitrogen species play important roles in the fate of organic compounds in the aqueous-phase advanced water treatment processes and natural aquatic environments under sunlight irradiation. Predicting the fate of organic compounds in aqueous-phase advanced water treatment processes and natural aquatic environments necessitates understanding the kinetics and reaction mechanisms of initial reactions of free radicals with structurally diverse organic compounds and other reactions. Researchers developed conventional predictive models based on experimentally measured transformation products and determined reaction rate constants by fitting with the time-dependent concentration profiles of species due to difficulties in their measurements of unstable intermediates. However, the empirical treatment of lumped reaction mechanisms had a model prediction limitation with respect to the specific parent compound's fate. We use ab initio and density functional theory quantum chemical computations, numerical solutions of ordinary differential equations, and validation of the outcomes of the model with experiments. Sensitivity analysis of reaction rate constants and concentration profiles enables us to identify an important elementary reaction in formating the transformation product. Such predictive elementary reaction-based kinetics models can be used to screen organic compounds in water and predict their potentially toxic transformation products for a specific experimental investigation.Over the past decade, we determined linear free energy relationships (LFERs) that bridge the kinetic and thermochemical properties of reactive oxygen species such as hydroxyl radicals (HO•), peroxyl radicals (ROO•), and singlet oxygen (1O2); reactive halogen species such as chlorine radicals (Cl•) and bromine radicals (Br•); reactive nitrogen species (NO2•); and carbonate radicals (CO3•-). We used literature-reported experimental rate constants as kinetic information. We considered the theoretically calculated aqueous-phase free energy of activation or reaction to be a kinetic or a thermochemical property, and obtained via validated ab initio or density functional theory-based quantum chemical computations using explicit and implicit solvation models. We determined rate-determining reaction mechanisms involved in reactions by observing robust LFERs. The general accuracy of LFERs to predict aqueous-phase rate constants was within a difference of a factor of 2-5 from experimental values.We developed elementary reaction-based kinetic models and predicted the fate of acetone induced by HO• in an advanced water treatment process and methionine by photochemically produced reactive intermediates in sunlit fresh waters. We provided mechanistic insight into peroxyl radical reaction mechanisms and critical roles in the degradation of acetone and the formation of transformation products. We highlighted different roles of triplet excited states of two surrogate CDOMs, 1O2, and HO•, in methionine degradation. Predicted transformation products were compared to those obtained via benchtop experiments to validate our elementary reaction-based kinetic models. Predicting the reactivities of reactive halogen and nitrogen species implicates our understanding of the formation of potentially toxic halogen- and nitrogen-containing transformation products during water treatment processes and in natural aquatic environments.

Transformation mechanisms of antidepressants in biological wastewater treatment: Removal kinetic, transformation products and pathways

The extensive detection of antidepressants in wastewater has raised increasing concerns for human beings. During biological wastewater treatment processes, antidepressants cannot be completely removed and mineralized, which caused the formation of transformation products (TPs). To date, knowledges on TPs that participated in the transformation processes of antidepressants were scarce. To fill this gap, molecular networking nontarget screening based on lab-scale batch assays were employed to explore the removal and transformation of five antidepressants (amitriptyline (AMI), clomipramine (CLO), duloxetine (DUL), nortriptyline (NOR) and paroxetine (PAR)). The removal rate constants per unit of biomass (kbio) of the five antidepressants were found to range from 0.29 to 0.56 L/(gMLSS*d). Comparatively, clomipramine was removed more slowly than other antidepressants, which attributed to its lower electrophilic properties. Subsequently, 22 TPs were tentatively identified, among which 19 ones were newly found in the present study. The proposed transformation pathways indicated that demethylation, N-formylation, N-acetylation, N-succinylation, N-hydroxylation, dealkylation, hydrolysis and nitrosation participated in the transformation of antidepressants in wastewater. Particularly, N-acylation and N-hydroxylation were found as characteristics reactions for antidepressants in wastewater and these reactions consistently occurred at the site of amino functional group, where high electrophilic activity was the dominant causes for the observed characteristics reactions. In silico results revealed that TPs formed during biological wastewater treatment generally had lower toxicity, lower bioaccumulation and higher bioavailability than their parent compounds. Collectively, the present study provides insights into the transformation processes of antidepressants in wastewater, indicating that characteristic transformation reactions occur across different antidepressants, which is theoretically useful to effectively remove pharmaceuticals from wastewater.

Ozone Strong Water Dosing as Optimized Ozonation Process for Micropollutants Reduction in Wastewater Treatment Plants

ABSTRACT Growing concern over potential ecotoxicological problems caused by micropollutants (MPs) in water has led to the application of adsorptive and oxidative advanced treatment technologies at European wastewater treatment plants (WWTPs). Direct dosage of gaseous ozone via diffusers or pump-injection systems is the most common large-scale process applied in water and wastewater treatment. Studies point out that MP removal may take place via the direct, very selective reaction with the ozone molecule as well as the indirect reaction with the very reactive but nonselective hydroxyl radicals. Several process parameters and set-ups may affect the MPs elimination and inhibit or even cause the formation of transformation products (TPs) such as bromate. This work compares an alternative ozone application procedure (the so-called ‘ozone strong water’ dosing – OSW) with standard systems regarding MP elimination, ozone consumption, reaction time, and required plant space. A full-scale OSW system was installed and operated at the WWTP Duisburg-Vierlinden. A significant reduction in the time required to achieve the maximum possible elimination was observed for all six MPs investigated, with a hydraulic reaction time of 8 min. According to a theoretical calculation, the reactor volume could be reduced by two-thirds of the installed volume.

Should Transformation Products Change the Way We Manage Chemicals?

When chemical pollutants enter the environment, they can undergo diverse transformation processes, forming a wide range of transformation products (TPs), some of them benign and others more harmful than their precursors. To date, the majority of TPs remain largely unrecognized and unregulated, particularly as TPs are generally not part of routine chemical risk or hazard assessment. Since many TPs formed from oxidative processes are more polar than their precursors, they may be especially relevant in the context of persistent, mobile, and toxic (PMT) and very persistent and very mobile (vPvM) substances, which are two new hazard classes that have recently been established on a European level. We highlight herein that as a result, TPs deserve more attention in research, chemicals regulation, and chemicals management. This perspective summarizes the main challenges preventing a better integration of TPs in these areas: (1) the lack of reliable high-throughput TP identification methods, (2) uncertainties in TP prediction, (3) inadequately considered TP formation during (advanced) water treatment, and (4) insufficient integration and harmonization of TPs in most regulatory frameworks. A way forward to tackle these challenges and integrate TPs into chemical management is proposed.

Identifying organic micropollutants' transformation products from the soil dissipation experiment by non-targeted high-resolution mass spectrometry approach: Can we gain more than transformation product identity?

Risk assessment of environmental hazards originating from xenobiotics extensively used worldwide (e.g., pharmaceuticals, bisphenols, or preservatives) requires a combined study of their effects, mobility, dissipation mechanisms, and subsequent transformation product identification and evaluation. We have developed an efficient accelerated solvent extraction method for a broad range of micropollutants of variable physical-chemical properties in soils to enable more accurate hazard characterisation. Micropollutant recoveries from freeze-dried soils were 60–120%, with the exception of atorvastatin, fexofenadine, and telmisartan, which had reduced recoveries (40–66%). The observed matrix effect ranged from −26% to 17% and was corrected by the matrix matching standard for quantitative analysis. The method allows sensitive and reliable determination of a wide range of analytes in soil samples and, consequently, qualitative analysis of transformation products (TP) with variable physicochemical properties. We identified TPs of five compounds (venlafaxine, telmisartan, valsartan, atorvastatin, and sertraline) by applying suspect and non-targeted data analyses. To our knowledge, the transformation product of atorvastatin was reported for the first time. All others were found in soil or other matrices. Valsartan (formed valsartan acid) and atorvastatin (transformed probably by oxidative decarboxylation of beta, delta dihydroxy heptanoic acid chain to propionic acid) were modified to a relatively large extent. All other compounds identified were only hydroxylated (sertraline and telmisartan) or demethylated (venlafaxine). We estimated the stability and presence of the identified TPs based on the constructed time trends and the ratio between TP formation and degradation rates. We demonstrated how valuable a non-targeted approach can be for complex evaluation of the fate and effect of soil pollutants.

The potential of soil microbial communities to transform deoxynivalenol in agricultural soils-a soil microcosm study.

Infestation of cereal fields with toxigenic Fusarium species is identified as an environmental source for the mycotoxin deoxynivalenol (DON). During rain events, DON may be washed off from infested plants and enter the soil, where microbial transformation may occur. Although some studies showed DON transformation potential of soil microbial communities in liquid soil extracts, these findings can not be transferred to environmental conditions. Accordingly, microbial transformation of DON in soil has to be investigated under realistic conditions, e.g., microcosms mimicking field situations. In this study, we investigated the potential of soil microbial communities to transform DON in six different agricultural soils at two levels (0.5 and 5 µg g-1). The dissipation and the formation of transformation products were investigated in a period of 35 days and compared to a sterilized control. In addition, we measured soil respiration and applied the phospholipid-derived fatty acid (PLFA) analysis to assess whether soil microbial community characteristics are related to the microbial transformation potential. Dissipation of DON in non-sterilized soils was fast (50% dissipation within 0.6-3.7 days) compared to the sterile control where almost no dissipation was observed. Thus, dissipation was mainly attributed to microbial transformation. We verified that small amounts of DON are transformed to 3-keto-deoxynivalenol (3-keto-DON) and 3-epi-deoxynivalenol (3-epi-DON), which were not detectable after 16-day incubation, indicating further transformation processes. There was a trend towards faster transformation in soils with active and large microbial communities and low fungi-to-bacteria ratio.

Environmental consequences of transformation products from an antibiotic mixture and their mitigation in a wastewater microbiome using an HCl-modified adsorbent

This study enhanced our understanding of antibiotic mixtures' occurrence, transformation, toxicity, and ecological risks. The role of acid-modified biochar (BC) in treating antibiotic residues was explored, shedding light on how BC influences the fate, mobility, and environmental impact of antibiotics and transformation products (TPs) in an activated sludge (AS) microbiome. A mixture of oxytetracycline and sulfamethoxazole was found to synergistically (or additively) inhibit cell growth of AS and disrupt the microbiome structure, species richness/diversity, and function. The formation of TPs with potentially higher toxicity and persistence than the original compounds was identified, explaining the microbiome disruption. Agricultural waste-derived BC was optimized for contaminant adsorption, leading to a reduction in toxicity when added to AS by sequestering TPs on its surface. This work highlighted adsorbents as a practical engineering strategy for mitigating liquid-phase contaminants' toxicological consequences, proactively controlling the fate and effects of antibiotics and TPs.

Degradation of micropollutants by gamma irradiation: Insight of transformation product formation explored by suspect and non-target screening using LC-HRMS

Conventional wastewater treatment systems have limitations in removing diverse micropollutants present with the complex matrix. As an alternative, advanced oxidation processes such as gamma-irradiation have been proposed. The aim of the study was to get insight into the decomposition of persistent pharmaceuticals (carbamazepine, cetirizine, tramadol) by gamma irradiation in pure water and effluent, and the formation and pathway of transformation products (TPs) identified via suspect/non-target screening using LC-HRMS. For TPs, the environmental risk assessment was also conducted with Quantitative Structure Activity Relationship (QSAR) based estimations. The decomposition effect was estimated with the removal rate and radiation chemical yield (G-value). The results indicated that the removal rate increased as the absorbed dose increased, up to 5 kGy, completely eliminating the target pharmaceuticals (initial nominal concentration: 10 mg/L) in pure water. However, the degradation in effluent was negatively affected by the organic matrix. By employing suspect/non-target screening techniques, a tentative identification of 30 transformation products (TPs) was achieved, providing insights into the degradation pathway. They were mainly formed through hydroxylation reaction by OH⸱. As a result of the QSAR estimation for the hazard of TPs, most of the TPs showed a lower risk than the parent compound. It indicates that the gamma-irradiation can be used as an alternative process to eliminate hardly degradable micropollutants in conventional wastewater treatment plant in a way to mitigate the environmental risk.

Comparison of monochloramination and chlorination of 1,3-diphenylguanidine (DPG): Kinetics, transformation products, and cell-based in-vitro testing

As a widely used secondary vulcanization accelerator in the rubber industry, 1,3-diphenylguanidine (DPG) poses risks to human health and the environment. To compare and comprehend the disinfection process of DPG, this work investigates the reaction kinetics, toxicity, and transformation products (TPs) of DPG during chlorination and monochloramination. It has been revealed that the reactivity of monochloramine is significantly slower compared to chlorination of DPG, with the maximum efficiency observed at pH 7 to pH 8. Cytotoxicity assessment using HepG2 and THP-1 cells reveals that cytotoxicity hierarchy is as follows: chlorine TPs > monochloramine TPs > DPG. Moreover, oxidant-to-DPG molar ratios 10 and 20 lead to higher cytotoxicity in both chlorination and monochloramination compared to ratio 5 and 100. Additionally, cell bioenergetics experiments demonstrate that chlorine and monochloramine TPs induce mitochondrial dysfunction and enhance glycolytic function in HepG2 cells. The genotoxic response from p53 signaling further suggested genotoxic effects of certain TPs. Furthermore, analysis of TPs using high-resolution mass spectrometry (HRMS) identifies ten TPs, with chlorination yielding more TPs than monochloramination. Generally, a chlorine or monochloramine molar ratio to DPG of 10–20 results in an increased formation of TPs and heightened cytotoxicity. Notably, higher oxidant molar ratios increased the formation of monoguanidine TPs and DPG hydroxylation during chlorination, whereas monochloramination lead to DPG substitution predominantly generating chlorinated DPG due to weaker oxidation effects. These findings provide valuable information for the appropriate treatment of DPG and disinfection processes in water facilities to mitigate potential risks to human health and the ecosystem.

Methylation and Demethylation of Emerging Contaminants Changed Bioaccumulation and Acute Toxicity in Daphnia magna.

Contaminants of emerging concern (CECs) in the environment undergo various transformations, leading to the formation of transformation products (TPs) with a modified ecological risk potential. Although the environmental significance of TPs is increasingly recognized, there has been relatively little research to understand the influences of such transformations on subsequent ecotoxicological safety. In this study, we used four pairs of CECs and their methylated or demethylated derivatives as examples to characterize changes in bioaccumulation and acute toxicity in Daphnia magna, as a result of methylation or demethylation. The experimental results were further compared to quantitative structure-activity relationship (QSAR) predictions. The methylated counterpart in each pair generally showed greater acute toxicity in D. magna, which was attributed to their increased hydrophobicity. For example, the LC50 values of methylparaben (34.4 ± 4.3 mg L-1) and its demethylated product (225.6 ± 17.3 mg L-1) differed about eightfold in D. magna. The methylated derivative generally exhibited greater bioaccumulation than the demethylated counterpart. For instance, the bioaccumulation of methylated acetaminophen was about 33-fold greater than that of acetaminophen. In silico predictions via QSARs aligned well with the experimental results and suggested an increased persistence of the methylated forms. The study findings underline the consequences of simple changes in chemical structures induced by transformations such as methylation and demethylation and highlight the need to consider TPs to achieve a more holistic understanding of the environmental fate and risks of CECs.

Emissions of the Urban Biocide Terbutryn from Facades: The Contribution of Transformation Products.

Biocides are added to facade paints and renders to prevent algal and fungal growth. The emissions of biocides and their transformation products from building facades during wind-driven rain can contaminate surface waters, soil, and groundwater. Although the emissions of biocide transformation products may be higher than those of the parent biocide, knowledge of the emissions of transformation products over time is scarce. Combining field- and lab-scale experiments, we showed that solar irradiation on facades controls the formation of transformation products and can be used with runoff volume to estimate the long-term emissions of terbutryn transformation products from facades. The slow (t1/2 > 90 d) photodegradation of terbutryn in paint under environmental conditions was associated with insignificant carbon isotope fractionation (Δδ13C < 2 ‰) and caused 20% higher emission of terbutryn-sulfoxide than terbutryn in leachates from facades. This indicated continuous terbutryn diffusion toward the paint surface, which favored terbutryn photodegradation and the concomitant formation of transformation products over time. The emissions of terbutryn transformation products (77 mg m-2) in facade leachates, modeled based on irradiation and facade runoff, were predicted to exceed those of terbutryn (42 mg m-2) by nearly 2-fold after eight years. Overall, this study provides a framework to estimate and account for the long-term emissions of biocide transformation products from building facades to improve the assessment of environmental risks.

Electrochemistry-mass spectrometry bridging the gap between suspect and target screening of valsartan transformation products in wastewater treatment plant effluent

Degradation of xenobiotics in wastewater treatment plants may lead to the formation of transformation products with higher persistence or increased (eco-)toxic potential compared to the parent compounds. Accordingly, the identification of transformation products from wastewater treatment plant effluents has gained increasing attention. Here, we show the potential of electrochemistry hyphenated to liquid chromatography and mass spectrometry for the prediction of oxidative degradation in wastewater treatment plants using the antihypertensive drug valsartan as a model compound. This approach identifies seven electrochemical transformation products of valsartan, which are used to conduct a suspect screening in effluent of the main wastewater treatment plant in the city of Münster in Germany. Apart from the parent compound valsartan, an electrochemically predicted transformation product, the N-dealkylated ETP336, is detected in wastewater treatment plant effluent. Subsequently, a targeted liquid chromatographytandem mass spectrometry method for the detection of valsartan and its electrochemical transformation products is set up. Here, electrochemical oxidation is used to generate reference materials of the transformation products in situ by hyphenating electrochemistry online to a triple quadrupole mass spectrometer. Using this setup, multiple reaction monitoring transitions are set up without the need for laborious and costly synthesis and isolation of reference materials for the transformation products. The targeted method is then applied to extracts from wastewater treatment plant effluent and the presence of ETP336 and valsartan in the samples is verified. The presented workflow can be used to set up targeted analysis methods for previously unknown transformation products even without the need for expensive high-resolution mass spectrometers.

Transformation of Algal Toxins during the Oxidation/Disinfection Processes of Drinking Water: From Structure to Toxicity.

With the increase of algal blooms worldwide, drinking water resources are threatened by the release of various algal toxins, which can be hepatotoxic, cytotoxic, or neurotoxic. Because of their ubiquitous occurrence in global waters and incomplete removal in conventional drinking water treatment, oxidation/disinfection processes have become promising alternative treatment options to destroy both the structures and toxicity of algal toxins. This Review first summarizes the occurrence and regulation of algal toxins in source water and drinking water. Then, the transformation kinetics, disinfection byproducts (DBPs)/transformation products (TPs), pathways, and toxicity of algal toxins in water oxidation/disinfection processes, including treatment by ozonation, chlorination, chloramination, ultraviolet-based advanced oxidation process, and permanganate, are reviewed. For most algal toxins, hydroxyl radicals (HO•) exhibit the highest oxidation rate, followed by ozone and free chlorine. Under practical applications, ozone and chlorine can degrade most algal toxins to meet water quality standards. However, the transformation of the parent structures of algal toxins by oxidation/disinfection processes does not guarantee a reduction in toxicity, and the formation of toxic TPs should also be considered, especially during chlorination. Notably, the toxicity variation of algal toxins is associated with the chemical moiety responsible for toxicity (e.g., Adda moiety in microcystin-LR and uracil moiety in cylindrospermopsin). Moreover, the formation of known halogenated DBPs after chlorination indicates that toxicity in drinking water may shift from toxicity contributed by algal toxins to toxicity contributed by DBPs. To achieve the simultaneous toxicity reduction of algal toxins and their TPs, optimized oxidation/disinfection processes are warranted in future research, not only for meeting water quality standards but also for effective reduction of toxicity of algal toxins.

Transformation products of contaminants of emerging concern in water by UV-based processes

Abstract Contaminants of emerging concern (CECs) and their respective transformation products (TPs) formed following photodegradation pose considerable threats to the environment and our health. The formation of TPs during UV-LED-based degradation of three target pollutants in the EU Watch List of CECs was accessed by LC-MS-Orbitrap, and their reaction pathways were elucidated. The influence of different matrices and treatments of choice on TP formation was investigated. Results showed that matrix changes did not produce different reaction pathways during UV-A photocatalysis, although plots of TP peak areas vs. time were different for each case. A new TP was found for the antidepressant venlafaxine, (1-[2-(dimethylnitroryl)-1-(4-methoxyphenyl)ethyl]cyclohexanol. When comparing UV-A photocatalysis with UV-C photolysis, dissimilar pathways were observed due to the distinct reaction mechanisms of each process, since photocatalysis, unlike photolysis, relies on radical-based reaction routes. Different levels of confidence were obtained for each TP depending on the availability of MS2 data in the literature and of standards for comparison. All the found TPs had similar molecular masses in comparison to their respective parent compounds. Most of the TPs remained in the effluent after 6 hours of photodegradation, which highlights the importance of their control, close-monitoring, and further toxicity assessments.

Guidance document on the impact of water treatment processes on residues of active substances or their metabolites in water abstracted for the production of drinking water.

This guidance document provides a tiered framework for risk assessors and facilitates risk managers in making decisions concerning the approval of active substances (AS) that are chemicals in plant protection products (PPPs) and biocidal products, and authorisation of the products. Based on the approaches presented in this document, a conclusion can be drawn on the impact of water treatment processes on residues of the AS or its metabolites in surface water and/or groundwater abstracted for the production of drinking water, i.e. the formation of transformation products (TPs). This guidance enables the identification of actual public health concerns from exposure to harmful compounds generated during the processing of water for the production of drinking water, and it focuses on water treatment methods commonly used in the European Union (EU). The tiered framework determines whether residues from PPP use or residues from biocidal product use can be present in water at water abstraction locations. Approaches, including experimental methods, are described that can be used to assess whether harmful TPs may form during water treatment and, if so, how to assess the impact of exposure to these water treatment TPs (tTPs) and other residues including environmental TPs (eTPs) on human and domesticated animal health through the consumption of TPs via drinking water. The types of studies or information that would be required are described while avoiding vertebrate testing as much as possible. The framework integrates the use of weight-of-evidence and, when possible alternative (new approach) methods to avoid as far as possible the need for additional testing.

Investigation of degradation and transformation product formation during the UV/H2O2-based oxidation process for chemicals of emerging concern

This study aimed to optimize the ultraviolet (UV)/hydrogen peroxide (H2O2) process for degrading chemicals of emerging concern (CECs), such as caffeine (CAF), atenolol (ATL), acetaminophen (ACT), and chlortetracycline (CTC). The CECs used in this study were chosen because they are used in various industries, affect water bodies, and are difficult to degrade using conventional oxidation and biological processes. The individual degradation results revealed that the CAF degradation rate increased from 23 ± 2%–80 ± 4% as the UV/H2O2 dose increased from 1 to 10 mg·L−1. Similarly, the ATL, ACT, and CTC degradation rates increased from 59.23 ± 2%, 34.21 ± 1.9%, and, 41.36 ± 2.3%–91.24 ± 3%, 68.12 ± 3%, and 91.93 ± 3.1%, respectively. The complex feed water analysis results revealed that CTC had a higher degradation rate than the other targeted contaminants owing to its higher degradation potential. The rate of reaction results revealed that the degradation of these CECs followed the pseudo-first-order reaction, with R2 > 0.95 in all cases. liquid chromatography with tandem mass spectrometry (LC–MS/MS) was used to identify the degradation products, and the results revealed the presence of low-molecular-weight organic products due to oxidation. The organic products were formed because of the hydroxylation and demethylation of intermediates and photoproducts, which caused a ring opening and generated several low-molecular-weight carboxylic acids before complete mineralization. These results illustrate that the UV/H2O2 process had some significant effects on the complex mixture. However, CTC exhibited a relatively high degradation rate, which is believed to have inhibited the degradation of other chemicals.

Investigating the Transformation Products of Selected Antibiotics and 17 α-Ethinylestradiol under Three In Vitro Biotransformation Models for Anticipating Their Relevance in Bioaugmented Constructed Wetlands.

The degradation of three antibiotics (sulfamethoxazole, trimethoprim, and ofloxacin) and one synthetic hormone (17 α-ethinylestradiol) was investigated in three in-vitro biotransformation models (i.e., pure enzymes, hairy root, and Trichoderma asperellum cultures) for anticipating the relevance of the formation of transformation products (TPs) in constructed wetlands (CWs) bioaugmented with T. asperellum fungus. The identification of TPs was carried out employing high-resolution mass spectrometry, using databases, or by interpreting MS/MS spectra. An enzymatic reaction with β-glucosidase was also used to confirm the presence of glycosyl-conjugates. The results showed synergies in the transformation mechanisms between these three models. Phase II conjugation reactions and overall glycosylation reactions predominated in hairy root cultures, while phase I metabolization reactions (e.g., hydroxylation and N-dealkylation) predominated in T. asperellum cultures. Following their accumulation/degradation kinetic profiles helped in determining the most relevant TPs. Identified TPs contributed to the overall residual antimicrobial activity because phase I metabolites can be more reactive and glucose-conjugated TPs can be transformed back into parent compounds. Similar to other biological treatments, the formation of TPs in CWs is of concern and deserves to be investigated with simple in vitro models to avoid the complexity of field-scale studies. This paper brings new findings on the emerging pollutants metabolic pathways established between T. asperellum and model plants, including extracellular enzymes.

Advanced oxidation processes for removal of monocyclic aromatic hydrocarbon from water: Effects of O3/H2O2 and UV/H2O2 treatment on product formation and biological post-treatment

Several oxidative treatment technologies, such as ozonation or Fenton reaction, have been studied and applied to remove monocyclic hydroaromatic carbon from water. Despite decades of application, little seems to be known about formation of transformation products while employing different ozone- or ∙OH-based treatment methods and their fate in biodegradation. In this study, we demonstrate that O3/H2O2 treatment of benzene, toluene, ethylbenzene (BTE), and benzoic acid (BA) leads to less hydroxylated aromatic transformation products compared to UV/H2O2 as reference system – this at a similar ∙OH exposure and parent compound removal efficiency. Aerobic biodegradation tests after oxidation of 0.15 mM BA (12.6 mg C L−1 theoretical DOC) revealed that a less biodegradable DOC fraction > 4 mg C L−1 was formed in both oxidative treatments compared to the BA control. No advantage of ozonation over UV/H2O2 treatment was observed in terms of mineralization capabilities, however, we detected less transformation products after oxidation and biodegradation using high-resolution mass spectrometry. Biodegradation of BA that was not oxidized was more complete with minimal organic residual. Overall, the study provides new insights into the oxidation of monocyclic aromatics and raises questions regarding the biodegradability of oxidation products, which is relevant for several treatment applications.

Estrogenic Effect Mechanism and Influencing Factors for Transformation Product Dimer Formed in Preservative Parabens Photolysis

The environmental transformation and health effects of endocrine disruptors (EDCs) need urgent attention, particularly the formation of transformation products with higher toxicity than parent EDCs. In this paper, an important transformation product dimer (short for ethyl 4-hydroxy-3-(2-((4-hydroxybenzoyl) oxy) ethyl) benzoate) with estrogenic activity was investigated and detected in the photolysis of preservative ethyl-paraben (EPB) dissolved in actual water. The environmental factors, such as the higher initial concentration of EPB, the stronger optical power and the lower pH could stimulate the formation of the dimer. Simultaneously, the interaction of multiple environmental factors was significant, especially the initial concentration and pH using the response surface methodology. Furthermore, the relationship between the environmental factors and the formation of the product dimer was further explained and the empirical model equation was built for predicting the amount of dimer in actual water. Quantum chemical and toxicological calculations showed the estrogenic effect mechanism of the product dimer and it was revealed further that the hydrogen bonds of the dimer and ERα proteins (ARG-394, Glu-353, His-524, GYY-521) were formed, with a lowest binding energy of -8.38 Kcal/mol during molecular docking. In addition, the health effect risk of the product dimer was higher than the parent compound in the blood, cardiovascular system, gastrointestinal system, kidney and liver. In short, the present study was of great significance for the transformation product in pollution control and health effects in the photolysis of EDCs.

Photocatalytic Degradation of Pharmaceutical Amisulpride Using g-C3N4 Catalyst and UV-A Irradiation

In the present study, the photocatalytic degradation of amisulpride using g-C3N4 catalyst under UV-A irradiation was investigated. The photocatalytic process was evaluated in terms of its effectiveness to remove amisulpride from ultrapure and real municipal wastewater. High removal percentages were achieved in both aqueous matrices. However, a slower degradation rate was observed using wastewater as matrix that could be attributed to its complex chemical composition. The transformation products (TPs) were identified with liquid chromatography–mass spectrometry (LC–MS) in both ultrapure and real municipal wastewater. Based on the identified TPs, the photocatalytic degradation pathways of amisulpride are proposed which include mainly oxidation, dealkylation, and cleavage of the methoxy group. Moreover, the contribution of reactive species to the degradation mechanism was studied using well-documented scavengers, and the significant role of h+ and O2•− in the reaction mechanism was proved. The evolution of ecotoxicity was also estimated using microalgae Chlorococcum sp. and Dunaliella tertiolecta. Low toxicity was observed during the overall process without the formation of toxic TPs when ultrapure water was used as matrix. In the case of real municipal wastewater, an increased toxicity was observed at the beginning of the process which is attributed to the composition of the matrix. The application of heterogeneous photocatalysis reduced the toxicity, and almost complete detoxification was achieved at the end of the process. Our results are in accordance with literature data that reported that heterogeneous photocatalysis is effective for the removal of amisulpride from aqueous matrices.
