Evaluation of the creditability of forensic tests through competency tests

The paper aims to evaluate a round of Competence Tests with qualitative results (non-destructive tests) in the field of classical forensics. The purpose of organizing the competency test rounds is to evaluate the performance of the forensic laboratories. The performance of the laboratories is estimated by the statistical evaluation of the results obtained by each participant. In this particular case, the assignment of the values of the competence test objects (CTO) was carried out by the group of experts, and the Competence Test Scheme (CTS) was of the simultaneous participation type; also, the group of experts established the performance evaluation criteria. The results obtained by the participants were satisfactory.

The need for a marine forensic laboratory for India's security and Indian maritime archaeology

An interdisciplinary discipline called maritime archaeology studies human cultures' submerged cultural legacy. India offers tremendous opportunities for marine archaeology study due to its extensive coastline and maritime heritage. To strengthen India's security measures, this article discusses the relevance of maritime archaeology in India and emphasizes the urgent necessity for a specialized marine forensic laboratory. The numerous facets of marine archaeology are covered in the research, along with their historical significance, archaeological methods, and national security concerns. The need to create a marine forensic lab to investigate crimes involving India's undersea cultural treasures and improve maritime security is further emphasized.

A beetle for prosecution: exogenous DNA detection from larval and adult gut of a Neotropical carrion beetle.

The use of recovered DNA ingested by necrophagous or hematophagous insects has increased in forensic sciences, mainly with representatives of flies. However, some beetles are also important for medico-legal forensic entomology because they feed on carcasses until advanced decomposition. This study evaluated whether the Neotropical carrion beetle Oxelytrum discicolle (Silphidae) has the potential for the detection of exogenous DNA into the gut. The whole gut or the gut contents were extracted from O. discicolle larvae and adult previously fed on pig carcass. The pig DNA recovery rate was 33.3% in larvae and 25% in adults, indicating that the carrion beetle's gut may be useful for DNA identification of ingested food. Samples with the whole gut or only gut contents showed the same DNA recovery rate. Exogenous DNA from the whole gut was recovered from samples stored in ethanol at -20 ºC for 11 days, showing that samples of O. discicolle can be stored in the forensic laboratory without loss in DNA recovery rate.

Variation in assessments of suitability and number of contributors for DNA mixtures

The interpretation of a DNA mixture (a sample that contains DNA from two or more people) depends on a laboratory/analyst’s assessment of the suitability of the sample for comparison/analysis, and an assessment of the number of contributors (NoC) present in the sample. In this study, 134 participants from 67 forensic laboratories provided a total of 2272 assessments of 29 DNA mixtures (provided as electropherograms). The laboratories’ responses were evaluated in terms of the variability of suitability assessments, and the accuracy and variability of NoC assessments. Policies and procedures related to suitability and NoC varied notably among labs. We observed notable variation in whether labs would assess a given mixture as suitable or not, predominantly due to differences in lab policies: if two labs following their standard operating procedures (SOPs) were given the same mixture, they agreed on whether the mixture was suitable for comparison 66% of the time. Differences in suitability assessments have a direct effect on variability in interpretations among labs, since mixtures assessed as not suitable would not result in reported interpretations. For labs following their SOPs, 79% of assessments of NoC were correct. When two different labs provided NoC responses, 63% of the time both labs were correct, and 7% of the time both labs were incorrect. Incorrect NoC assessments have been shown to affect statistical analyses in some cases, but do not necessarily imply inaccurate interpretations or conclusions. Most incorrect NoC estimates were overestimates, which previous research has shown have less of an effect on likelihood ratios than underestimates.

Morphometrics as a Tool for the Species Identification of North American Bald and Golden Eagle Primary Feathers

ABSTRACT Eagle feathers are among the most commonly trafficked items in the illegal wildlife trade in the United States. Although the tail and secondary wing feathers of Bald Eagles (Haliaeetus leucocephalus) and Golden Eagles (Aquila chrysaetos) generally exhibit identifiable patterns by which the species of origin can be determined, there is a lack of data allowing for the morphological identification of the commonly encountered, unpatterned outer primary wing feathers. To test if the shape of the primary feathers differed between the two species, I measured the notch and emargination heights of the outer five primaries (P10–P6) from specimens at the National Fish and Wildlife Forensic Laboratory collection and the National Eagle Repository. Although there was some overlap in lengths of the feather emargination and notch heights of both species, the results demonstrate that measurements of the outermost primaries do allow for the identification of many loose feathers. Overall, Golden Eagle feathers exhibit longer emarginations and notch heights than those of the Bald Eagle. These results will be valuable for law enforcement officials, ornithologists, and anthropologists studying the remains of North American eagles.

Simplified COI barcoding of blow, flesh, and scuttle flies encountered in medicolegal investigations.

Accurate insect identification is critical to the estimation of time of colonization (TOC) and post-mortem interval (PMI) in medicolegal death investigations. DNA testing is advantageous because it enables the identification of immature specimens that may not be identified based on morphology alone. We describe here a simplified DNA barcoding method for identifying relevant species that may be implemented by forensic genetics laboratories. A cytochrome oxidase (COI) fragment is analyzed after PCR amplification with a single primer set. The method is effective for many species commonly encountered in death investigations in the USA: members of blowfly genera Calliphora, Chrysomya, Cochliomyia, Lucilia, and Phormia; members of the flesh fly genera Blaesoxipha, Oxysarcodexia, Ravinia, and Sarcophaga; and the scuttle fly Megaselia scalaris. We tested the method on specimens with verified identifications and used it to build a collection of reference sequences from specimens collected in Harris County, Texas. We show here the correct identification of larvae, pupae, and pupal exuviae from the medicolegal casework.

DNA profiling in forensic investigation – A review

DNA Profiling is a revolutionary method for individual and relationship analysis, crime investigation, hereditary disorders, etc. It is a universal method used to establish accurate results during the process of forensic investigation. DNA profiling techniques, which are based on repetitive sequences within DNA, have proven to be of paramount importance, albeit the complete utilization of knowledge still remaining unexplored. Even a hair strand, blood drop or even skin flakes can be used to identify DNA sequences. It has a wide range of applications both in forensics and law. Because of the advancement in the field of forensics in the past four decades, DNA evidence now stands as one of the most reliable forms of proof in a court of law. In the following article, the authors explore the main concepts of DNA Profiling, and the techniques which are widely used in forensic laboratories such as RFLP, VNTR, STR, AFLP, mtDNA analysis, Y-chromosome analysis and gender typing.

A comparison of five DNA extraction methods from degraded human skeletal remains

Extracting DNA from degraded human remains poses a challenge for any forensic genetics laboratory, as it requires efficient high-throughput methods. While little research has compared different techniques, silica in suspension has been identified in the literature as the best method for recovering small fragments, which are often present in these types of samples. In this study, we tested five DNA extraction protocols on 25 different degraded skeletal remains. Including the humerus, ulna, tibia, femur, and petrous bone. The five protocols were organic extraction by phenol/chloroform/isoamyl alcohol, silica in suspension, High Pure Nucleic Acid Large Volume silica columns (Roche), InnoXtract™ Bone (InnoGenomics), and PrepFiler™ BTA with AutoMate™ Express robot (ThermoFisher). We analysed five DNA quantification parameters (small human target quantity, large human target quantity, human male target quantity, degradation index, and internal PCR control threshold), and five DNA profile parameters (number of alleles with peak height higher than analytic and stochastic threshold, average relative fluorescence units (RFU), heterozygous balance, and number of reportable loci) were analysed. Our results suggest that organic extraction by phenol/chloroform/isoamyl alcohol was the best performing method in terms of both quantification and DNA profile results. However, Roche silica columns were found to be the most efficient method.

The influence of storage conditions on fingermarks developed with 1,2-indanedione-ZnCl

The most important class of reagents for the enhancement of fingermarks on porous surfaces are those that react with the amino acid constituents of fingermarks. Ninhydrin, DFO (1,8-diazafluoren-9-one), and 1,2-indanedione are the three most common techniques widely known in forensic laboratories for the visualization of latent fingermarks on porous surfaces. In 2012 the Netherlands Forensic Institute replaced DFO by 1,2-indanedione-ZnCl after an internal validation, just like an increasing numbers of laboratories. In 2003, Gardner et al. published an article showing that fingermarks treated with 1,2-indanedione (without ZnCl), that were stored in daylight only decreased by 20% in fluorescence in 28 days. However, during casework we observed that the fluorescence of fingermarks treated with 1,2-indanedione with ZnCl decreased more rapidly. In this study, the effect of various storage conditions and aging times on the fluorescence of marks after treatment with 1,2-indanedione-ZnCl were assessed. Latent printed fingermarks from the digital matrix printer (DMP) and natural fingermarks from a known donor were used. The results showed that storing fingermarks in daylight (wrapped and un-wrapped) drastically decrease (over 60% loss) in fluorescence in approximately three weeks. Storage of the marks in a dark environment (at room temperature, in the refrigerator or even in the freezer) resulted in a decrease in fluorescence of less than 40%. Our recommendation is to always store treated fingermarks with 1,2-indanedione-ZnCl in a dark environment and, if possible, photographing them directly (within 1–2 days after treatment) to minimize the decrease of fluorescence.

DNA transfer to placed, stored, and handled drug packaging and knives in houses

Forensic laboratories often sample weapons and clip-seal plastic bags (CSPB) used to package illicit material for the purpose of identifying the handler(s). However, there may be other explanations as to how a person’s DNA was transferred to such items. This may include an individual storing the item among their personal belongings for somebody else or the item being stored among their belongings without their knowledge. Here we investigate the direct transfer of DNA to knives and CSPB during handling and explore two feasible alternative explanations related to the indirect transfer of DNA to these items in residential environments. The handling of DNA-free items was performed by 10 individuals who were instructed, on separate occasions, to cut a foam board in half and fill a CSPB with a drug substitute. To explore indirect transfer, sets of these items were (a) placed on kitchen benches and coffee/dining tables for ∼1 min, or (b) stored for two days in kitchen and bedroom drawers within the homes of 10 individuals. After each of the three scenarios, samples were collected from the knife handle and blade, the body and seal of the CSPB, and the surface the items were placed on, the latter as a measure to gain insight into the presence of prevalent and/or background DNA. DNA transfer was observed under all three scenarios, though more frequently when items were handled or stored for 2 days, compared to when placed on a surface for ∼1 min. Under the latter scenario, DNA, if present, was below the level of detection in many samples and produced no profile, suggesting that detectable DNA transfer occurs to a lesser degree from static brief contacts. The study results and associated probabilities will assist forensic examiners with their interpretation of case circumstances regarding the transfer and recovery of DNA from these items.

QR code technology in individual identification training provides an introduction in hands-on forensic DNA genotyping to medical students

BackgroundForensic biology is a subject in the field of forensic science that stresses practical teaching and training in laboratory skills. Visualization of deoxyribonucleic acid (DNA) profiles is important in individual identification and is easily performed by well-trained examiners. Therefore, developing a novel training project for obtaining individual DNA profiles can improve the quality of teaching for medical students or trainees. DNA profiles based on quick response (QR) codes can also be applied to practical teaching and operation training for individual identification.MethodsA novel training project was developed through an experimental course in forensic biology. Blood samples and buccal swabs with oral epithelial cells, as used in the forensic DNA laboratory, were obtained from medical students at Fujian Medical University. DNA was isolated, and a number of short tandem repeat (STR) loci were used as genetic markers to generate DNA profiles. The students converted DNA profiles and individual information into a QR code. The QR code could then be scanned by a mobile phone for consulting and retrieval. Gene identity cards with QR codes were produced and provided to every student. The participation rate and passing rate of students who participated in the novel training project were calculated and compared with those of students in the traditional experimental course, and a chi-square test was carried out by SPSS 23.0 software to evaluate the teaching effectiveness. p < 0.05 indicated significant differences. In addition, a survey was conducted to investigate the likelihood of using of gene identity cards with QR codes in the future.ResultsA total of 54 of 91 medical students who studied forensic biology participated in the novel training project in 2021. Only 31 of 78 students who studied forensic biology participated in the traditional experimental course in 2020. The participation rate in the novel training project was 24% higher than that of the traditional experimental course. The participants in the novel training project showed better performance in forensic biological handling techniques. The passing rate of the students in the forensic biology course with the novel training project was approximately 17% higher than that of the students in the former course. The participation rates and passing rates of the two groups were significantly different (χ = 6.452, p = 0.008 and χ = 11.043, p = 0.001). In the novel training project, all participants made 54 gene identity cards with QR codes. Furthermore, in the DNA profiles of four African students who participated, we found two rare alleles that were not discovered in Asians. The survey showed that the use of gene identity cards with QR codes was accepted by most participants, and the likelihood of future utilization was 78%.ConclusionWe established a novel training project to promote the learning activities of medical students in experimental forensic biology courses. The participants showed great interest in using gene identity cards with QR codes to store general individual identity information and DNA profiles. They also examined the genetic population differences between different races based on DNA profiles. Hence, the novel training project could be useful for training workshops, forensic experimental courses, and medical big data research.

Development and validation of a rapid GC–MS method for seized drug screening

Controlled substances are one of the most commonly analyzed types of evidence in forensic laboratories. Persistent, high levels of case submissions in recent years have created a need for rapid screening techniques to decrease case backlogs and expedite confirmatory analyses. The most utilized technique for the analysis of seized drugs is gas chromatography mass spectrometry (GC-MS). While this approach typically takes tens of minutes, advances in the technique have enabled approximately 1 min run times with a short column and a rapid (10’s °C/s) temperature program. These advances were used in this work to develop and validate a rapid GC-MS method for screening seized drug samples. Method validation was completed to determine retention time repeatability and reproducibility, limits of detection, identification accuracy, selectivity, and carryover. Validation results showed the utility of this method for accurate and precise qualitative analyses for the screening of seized drugs across seven different drug classes. The optimized method was then applied to 15 real and mock case samples from the Maryland State Police Forensic Sciences Division. Controlled substances, cutting agents, and other diluents were identified from these samples using acceptance criteria of retention time and mass spectral comparisons currently used in casework. Using rapid GC-MS methods in casework could enable faster and more reliable presumptive testing results for the analysis of seized drug exhibits.

Comparative evaluation of the MAPlex, Precision ID Ancestry Panel, and VISAGE Basic Tool for biogeographical ancestry inference

Biogeographical ancestry (BGA) inference from ancestry-informative markers (AIMs) has strong potential to support forensic investigations. Over the past two decades, several forensic panels composed of AIMs have been developed to predict ancestry at a continental scale. These panels typically comprise fewer than 200 AIMs and have been designed and tested with a limited set of populations. How well these panels recover patterns of genetic diversity relative to larger sets of markers, and how accurately they infer ancestry of individuals and populations not included in their design remains poorly understood. The lack of comparative studies addressing these aspects makes the selection of appropriate panels for forensic laboratories difficult. In this study, the model-based genetic clustering tool STRUCTURE was used to compare three popular forensic BGA panels: MAPlex, Precision ID Ancestry Panel (PIDAP), and VISAGE Basic Tool (VISAGE BT) relative to a genome-wide reference set of 10k SNPs. The genotypes for all these markers were obtained for a comprehensive set of 3957 individuals from 228 worldwide human populations. Our results indicate that at the broad continental scale (K=6) typically examined in forensic studies, all forensic panels produced similar genetic structure patterns compared to the reference set (G'≈90%) and had high classification performance across all regions (average AUC-PR > 97%). However, at K=7 and K=8, the forensic panels displayed some region-specific clustering deviations from the reference set, particularly in Europe and the region of East and South-East Asia, which may be attributed to differences in the design of the respective panels. Overall, the panel with the most consistent performance in all regions was VISAGE BT with an average weighted AUC̅W score of 96.26% across the three scales of geographical resolution investigated.

Maintaining the chain of custody: Anti-contamination measures for trace DNA evidence

Trace DNA evidence has become an essential tool in forensic investigations, but its reliability is dependent on strict adherence to protocols and procedures that ensure the integrity of the evidence is preserved throughout the investigation. Maintaining a clear chain of custody is critical to the admissibility of evidence in court, and its absence can result in the exclusion of evidence or even a mistrial being declared. Contamination is a significant issue when collecting DNA evidence at a crime scene, and anti-contamination measures must be taken to ensure the reliability and accuracy of the evidence. The use of technology, such as electronic chain of custody (eCOC), automated DNA extraction systems, real-time PCR analysis, DNA profiling software, and environmental monitoring systems, can enhance the accuracy and reliability of evidence. Law enforcement agencies and forensic labs must establish strict guidelines for the collection, preservation, and analysis of trace DNA evidence to maintain its integrity and admissibility in court.

Trapped ion mobility mass spectrometry of new psychoactive substances: Isomer-specific identification of ring-substituted cathinones

New psychoactive substances (NPS) are synthetic derivatives of illicit drugs designed to mimic their psychoactive effects. NPS are typically not controlled under drug acts or their legal status depends on their molecular structure. Discriminating isomeric forms of NPS is therefore crucial for forensic laboratories. In this study, a trapped ion mobility spectrometry time-of-flight mass spectrometry (TIMS-TOFMS) approach was developed for the identification of ring-positional isomers of synthetic cathinones, a class of compounds representing two-third of all NPS seized in Europe in 2020. The optimized workflow features narrow ion-trapping regions, mobility calibration by internal reference, and a dedicated data-analysis tool, allowing for accurate relative ion-mobility assessment and high-confidence isomer identification. Ortho-, meta- and para-isomers of methylmethcathinone (MMC) and bicyclic ring isomers of methylone were assigned based on their specific ion mobilities within 5 min, including sample preparation and data analysis. The resolution of two distinct protomers per cathinone isomer added to the confidence in identification. The developed approach was successfully applied to the unambiguous assignment of MMC isomers in confiscated street samples. These findings demonstrate the potential of TIMS-TOFMS for forensic case work requiring fast and highly-confident assignment cathinone-drug isomers in confiscated samples.

Survey on the Construction Status of Forensic Virtual Autopsy Laboratory and the Applicability of Laboratory Accreditation.

To survey the development status and actual needs of virtual autopsy technology in China and to clarify the applicability of forensic virtual autopsy laboratory accreditation. The questionnaire was set up included three aspects：(1) the current status of virtual autopsy technology development; (2) the accreditation elements such as personnel, equipment, entrustment and acceptance, methods, environmental facilities; (3) the needs and suggestions of practicing institutions. A total of 130 forensic pathology institutions were surveyed by online participation through the Questionnaire Star platform. Among the 130 institutions, 43.08% were familiar with the characteristics of virtual autopsy technology, 35.38% conducted or received training in virtual autopsy, and 70.77% have establishment needs (including maintenance). Relevant elements were suitable for laboratory accreditation. Virtual autopsy identification has gained social recognition. There is a demand for accreditation of forensic virtual autopsy laboratory. After the preliminary assessment, considering the characteristics and current situation of this technology, China National Accreditation Service for Conformity Assessment (CNAS) can first carry out the accreditation pilot of virtual autopsy project at large comprehensive forensic institutions with higher identification capability, and then CNAS can popularize the accreditation in a wide range when the conditions are suitable.

Evaluation and Verification of a microRNA Panel Using Quadratic Discriminant Analysis for the Classification of Human Body Fluids in DNA Extracts.

There is significant interest in the use of miRNA analysis for forensic body fluid identification. Demonstrated co-extraction and detection in DNA extracts could make the use of miRNAs a more streamlined molecular body fluid identification method than other RNA-based methods. We previously reported a reverse transcription-quantitative PCR (RT-qPCR) panel of eight miRNAs that classified venous and menstrual blood, feces, urine, saliva, semen, and vaginal secretions using a quadratic discriminant analysis (QDA) model with 93% accuracy in RNA extracts. Herein, miRNA expression in DNA extracts from 50 donors of each body fluid were tested using the model. Initially, a classification rate of 87% was obtained, which increased to 92% when three additional miRNAs were added. Body fluid identification was found to be reliable across population samples of mixed ages, ethnicities, and sex, with 72-98% of the unknown samples classifying correctly. The model was then tested against compromised samples and over biological cycles, where classification accuracy varied, depending on the body fluid. In conclusion, we demonstrated the ability to classify body fluids using miRNA expression from DNA extracts, eliminating the need for RNA extraction, greatly reducing evidentiary sample consumption and processing time in forensic laboratories, but acknowledge that compromised semen and saliva samples can fail to classify properly, and mixed sample classification remains untested and may have limitations.

Structural Analysis and Characterization of 4-F-α-PVP Analog 4-F-3-Methyl-α- PVP Hydrochloride.

To identify 1-(4-fluorophenyl)-2-(1-pyrrolidinyl) pentan-1-one (4-F-α-PVP) analog 1-(4-fluoro-3-methyl phenyl)-2-(1-pyrrolidinyl) pentan-1-one (4-F-3-Methyl-α-PVP) hydrochloride without reference substance. The direct-injection electron ionization-mass spectrometry (EI-MS), GC-MS, electrospray ionization-high resolution mass spectrometry (ESI-HRMS), ultra-high performance liquid chromatography-high resolution tandem mass spectrometry (UPLC-HRMS/MS), nuclear magnetic resonance (NMR), ion chromatography and Fourier transform infrared spectroscopy (FTIR) were integrated utilized to achieve the structural analysis and characterization of the unknown compound in the sample, and the cleavage mechanism of the fragment ions was deduced by EI-MS and UPLC-HRMS/MS. By analyzing the direct-injection EI-MS, GC-MS, ESI-HRMS and UPLC-HRMS/MS of the compound in the samples, it was concluded that the unknown compound was a structural analog of 4-F-α-PVP, possibly with one more methyl group in the benzene ring. According to the analysis results of 1H-NMR and 13C-NMR, it was further proved that the methyl group is located at the 3-position of the benzene ring. Since the actual number of hydrogen in 1H-NMR analysis was one more than 4-F-3-Methyl-α-PVP neutral molecule, it was inferred that the compound existed in the form of salt. Ion chromatography analysis results showed that the compound contained chlorine anion (content 11.14%-11.16%), with the structural analysis of main functional group information by FTIR, the unknown compound was finally determined to be 4-F-3-Methyl-α-PVP hydrochloride. A comprehensive method using EI-MS, GC-MS, ESI-HRMS, UPLC-HRMS/MS, NMR, ion chromatography and FTIR to identify 4-F-3-Methyl-α-PVP hydrochloride in samples is established, which will be helpful for the forensic science laboratory to identify this compound or other analog compounds.

Introducing a Rapid DNA Analysis Procedure for Crime Scene Samples Outside of the Laboratory-A Field Experiment.

Technological innovations enable rapid DNA analysis implementation possibilities. Concordantly, rapid DNA devices are being used in practice. However, the effects of implementing rapid DNA technologies in the crime scene investigation procedure have only been evaluated to a limited extent. In this study a field experiment was set up comparing 47 real crime scene cases following a rapid DNA analysis procedure outside of the laboratory (decentral), with 50 cases following the regular DNA analysis procedure at the forensic laboratory. The impact on duration of the investigative process, and on the quality of the analyzed trace results (97 blood and 38 saliva traces) was measured. The results of the study show that the duration of the investigation process has been significantly reduced in cases where the decentral rapid DNA procedure was deployed, compared to cases where the regular procedure was used. Most of the delay in the regular process lies in the procedural steps during the police investigation, not in the DNA analysis, which highlights the importance of an effective work process and having sufficient capacity available. This study also shows that rapid DNA techniques are less sensitive than regular DNA analysis equipment. The device used in this study was only to a limited extent suitable for the analysis of saliva traces secured at the crime scene and can mainly be used for the analysis of visible blood traces with an expected high DNA quantity of a single donor.

In recent years there was great growth in forensic investigations in Brazil. Forensic biology has become an area of great impact in civil and criminal investigation which encompasses several fields, among them, laboratory analysis of livestock. This article aims to present concepts, demands and ways to develop activities of a quality management system in forensic veterinary work. Forensics can be used to help in the civil and/or criminal spheres, and in studies and research in the areas of animal, public, environmental and individual health (one health). In investigations, evidence is generally present and may be physical, chemical or microbiological in nature. The collection of evidence must be based on biosecurity standards and respect the chain of custody, which refers to the capacity of guaranteeing the identity and integrity of residues of any nature. It comprises tracing and allows for its chronological order to be documented and maintained. The demand for forensic veterinary work can be implemented in public research and teaching institutions or in private services, all of which have trained professionals and specialized laboratories that can perform tests and counter tests for technical-scientific expertise. The quality of a product or service is a fundamental factor for its insertion in the market. Forensic veterinary work can develop technical-scientific activities to meet the demands in facilitating justice through research of infectious agents of diseases in animals, toxic substances and contaminants, or the occurrence of crimes involving animals, contamination of environmental fauna, and fraud in foods of animal origin for human or animal consumption.
