Nanopore Blockade Sensors for Quantitative Analysis Using an Optical Nanopore Assay.

Nanopore sensing is a popular biosensing strategy that is being explored for the quantitative analysis of biomarkers. With low concentrations of analytes, nanopore sensors face challenges related to slow response times and selectivity. Here, we demonstrate an approach to rapidly detect species at ultralow concentrations using an optical nanopore blockade sensor for quantitative detection of the protein vascular endothelial growth factor (VEGF). This sensor relies on monitoring fluorescent polystyrene nanoparticles blocking nanopores in a nanopore array of 676 nanopores. The fluorescent signal is read out using a wide-field fluorescence microscope. Nonspecific blockade events are then distinguished from specific blockade events based on the ability to pull the particles out of the pore using an applied electric field. This allows the detection of VEGF at sub-picomolar concentration in less than 15 min.

Review on applications of Pullulan in bone tissue engineering: Blends and composites with natural and synthetic polymers

Pullulan (PUL) has a diverse range of applicationsdue to its many therapeutic benefits, including biodegradability, biocompatibility, nontoxicity, antimicrobial activity, and adsorption. They are combined with chitosan, polyvinyl pyrrolidone (PVP), polycaprolactone (PCL), heparin, fluorescent polystyrene nanoparticles (PS-NPs), and carboxyl Pullulan to develop properties such as thermal stability, mechanical properties, pH resistance, chemical stability, toughness. The effects of Pullulan content on the properties of the solution, as well as the morphology of the resultant nanofibers, were investigated >80%. The concept of a scaffold can be a useful notion to improve the mechanical behavior of hydrogel-based scaffolds. Compositional analysis by Differential scanning calorimetry (DSC) revealed that Pullulan might enhance the mechanical properties of the nanofibers. This review focuses on the combination and analysis of Pullulan blends and composites of natural and synthetic polymers, as well as their capability in biomedical fields and bone tissue engineering, for example in drug delivery, insulin delivery, food industry, medicinal and biomedical applications, antimicrobial wound dressings, cancer cell targeting, anticancer vaccine improvement, new biopolymer development, food product development and sensing. The electro spinning procedure and the materials employed in it will be covered in this review. The use of Pullulan electrospun nanofibers structures in tissue engineering will also be covered in this paper. The benefits, restrictions, and future opinions were studied. This is because of Pullulan-based polymers have a variety of properties.

Fluorescent molecularly imprinted polymer nanocomposite for solid-phase extraction and fluorimetric determination of hydrochlorothiazide.

We report herein a fluorescent molecularly imprinted polymer (FMIP) for the solid-phase extraction (SPE) and fluorimetric determination of hydrochlorothiazide (HCTZ) in water. The FMIP is based on fluorescent polystyrene nanoparticles embedded within a molecularly imprinted polyaniline (PANI) matrix. The operational adsorption parameters such as the initial HCTZ concentration, incubation time and the solution pH were found to influence the removal efficiency. At optimum conditions, a high adsorption capacity of the FMIP was found (2.08 mg g-1). Evidence of the adsorption process was confirmed by the change in the FMIP physicochemical properties measured by FTIR absorption spectroscopy and electron microscopy. Based on the regression R2 values and the consistently low values of the adsorption statistical error functions, equilibrium data were best fitted to both Freundlich and Temkin isotherms. Moreover, the pseudo-second-order kinetic model described the adsorption kinetics, and the mechanism of the adsorption process was explained by the intraparticle diffusion model. Upon studying adsorption thermodynamics, negative ΔG values (-26.18 kJ mol-1 at room temperature) were obtained revealing that the adsorption process is spontaneous. Interestingly, the maximum adsorption capacity was obtained at 298 K, pH 7.0, and using a high HCTZ concentration, thus revealing the suitability of the proposed FMIP for easy and fast SPE of HCTZ. The FMIP showed an imprinting factor of 1.19 implying the selectivity over the corresponding FNIP. Eventually, the proposed FMIP was successfully applied to the spectrofluorimetric determination of HCTZ in aqueous samples with %recovery values close to 100%.

DNA-Immobilized Fluorescent Polystyrene Nanoparticles as Probes with Tunable Detection Limits.

DNA-immobilized nanoparticle probes show high target specificity; thus, they are employed in various bioengineering and biomedicine applications. When the nanoparticles employed are dye-loaded polymer particles, the resulting probes have the additional benefit of biocompatibility and versatile surface properties. In this study, we construct DNA-immobilized fluorescent polystyrene (PS) nanoparticles through controlled surface reactions. PS nanoparticles with surface carboxyl groups are utilized, and amine-functionalized dye molecules and capture DNAs are covalently immobilized via a one-pot reaction. We show that the surface chemistry employed allows for quantitative control over the number of fluorescent dyes and DNA strands immobilized on the PS nanoparticle surfaces. The nanoparticles thus prepared are then used for DNA detection. The off state of the nanoprobe is achieved by hybridizing quencher-functionalized DNAs (Q-DNAs) to the capture DNAs immobilized on nanoparticle surfaces. Target-DNAs (T-DNAs) are detected by the displacement of the prehybridized Q-DNAs. The nanoprobes show successful detection of T-DNAs with high sequence specificity and long-term stability. They also show excellent detection sensitivity, and the detection limit can be tuned by adjusting the capture DNA-to-dye ratio.

Near-Infrared-Triggered On-Demand Controlled Release of Adeno-Associated Virus from Alginate Hydrogel Microbeads with Heat Transducer for Gene Therapy.

Gene therapy using adeno-associated virus (AAV) has potential as a radical treatment modality for genetic diseases such as sensorineural deafness. To establish clinical applications, it is necessary to avoid immune response to AAV by controlled release system of AAV. Here, a near-infrared (NIR)-triggered on-demand AAV release system using alginate hydrogel microbeads with a heat transducer is proposed. By using a centrifuge-based microdroplet shooting device, the microbeads encapsulating AAV with Fe3 O4 microparticles (Fe3 O4 -MPs) as a heat transducer are fabricated. Fe3 O4 -MPs generated heat by NIR enhanced the diffusion speed of the AAV, resulting in the AAV being released from the microbeads. By irradiating the microbeads encapsulating fluorescent polystyrene nanoparticles (FP-NPs) (viral model) with NIR, the fluorescence intensity decreased only for FP-NPs with a diameter of 20nm and not for 100 or 200nm, confirming that this system can release virus with a diameter of several tens of nanometers. By irradiating NIR to the AAV-encapsulating microbeads with Fe3 O4 -MPs, the AAV is released on demand, and gene transfection to cells by AAV is confirmed without loss of viral activity. The NIR-triggered AAV release system proposed in this study increases the number of alternatives for the method of drug release in gene therapy.

Size-Dependent Effects of Polystyrene Nanoparticles (PS-NPs) on Behaviors and Endogenous Neurochemicals in Zebrafish Larvae.

Microplastics, small pieces of plastic derived from polystyrene, have recently become an ecological hazard due to their toxicity and widespread occurrence in aquatic ecosystems. In this study, we exposed zebrafish larvae to two types of fluorescent polystyrene nanoparticles (PS-NPs) to identify their size-dependent effects. PS-NPs of 50 nm, unlike 100 nm PS-NPs, were found to circulate in the blood vessels and accumulate in the brains of zebrafish larvae. Behavioral and electroencephalogram (EEG) analysis showed that 50 nm PS-NPs induce abnormal behavioral patterns and changes in EEG power spectral densities in zebrafish larvae. In addition, the quantification of endogenous neurochemicals in zebrafish larvae showed that 50 nm PS-NPs disturb dopaminergic metabolites, whereas 100 nm PS-NPs do not. Finally, we assessed the effect of PS-NPs on the permeability of the blood–brain barrier (BBB) using a microfluidic system. The results revealed that 50 nm PS-NPs have high BBB penetration compared with 100 nm PS-NPs. Taken together, we concluded that small nanoparticles disturb the nervous system, especially dopaminergic metabolites.

Bovine serum albumin-gold nanoclusters protein corona stabilized polystyrene nanoparticles as dual-color fluorescent nanoprobes for breast cancer detection

Breast cancer is the most prevalent malignancy and the first leading cause of cancer-related mortality among the female population worldwide. Approaches for precise and reliable detection of breast cancer cells, particularly in the nascent state, are desperately needed for elevating the survival rate of patients bearing the breast tumor. In this work, we successfully performed the sensitive, precise, and reliable breast cancer cell detection using facilely fabricated bovine serum albumin-gold nanocluster (BSA-AuNCs) protein corona stabilized, epithelial cell adhesion molecule (EpCAM) aptamer linked fluorescent polystyrene nanoparticle (PS NP), termed as PS-BSA-AuNCs-Apt. The rapidly adsorbed BSA-AuNCs hard protein corona without complicated covalent conjugation not only imparted excellent colloidal stability to the PS nanoparticles, but also offered numerous active anchors for the targeted EpCAM aptamers to locate. With the remarkable aid of the aptamers specifically targeting the EpCAM-positive breast cancer cells, the PS-BSA-AuNCs-Apt emitted strong and photostable dual-color fluorescent signals for precise and reliable cancer cell detection by overcoming the false signals. The specific identification potency of the PS-BSA-AuNCs-Apt system was further verified by successfully detecting the xenografted breast tumor tissue. Notably, to the best of our knowledge, the protein corona formed nanoprobes was exploited for direct tumor cell and tissue detection with high efficacy for the first time, demonstrating their promising potential in clinical tumor detection.

β-Lactamase-Responsive Hydrogel Drug Delivery Platform for Bacteria-Triggered Cargo Release

Antibiotic resistance is a growing public health threat that complicates the treatment of infections. β-Lactamase enzymes, which hydrolyze the β-lactam ring present in many common antibiotics, are a major cause of this resistance and are produced by a broad range of bacterial pathogens. Here, we developed hydrogels that degrade specifically in the presence of β-lactamases and β-lactamase-producing bacteria as a platform for bacteria-triggered drug delivery. A maleimide-functionalized β-lactamase-cleavable cephalosporin was used as a crosslinker in the fabrication of hydrogels through end-crosslinked polymerization with multiarm thiol-terminated poly(ethylene glycol) macromers via Michael-type addition. We demonstrated that only hydrogels containing the responsive crosslinker were degraded by β-lactamases and β-lactamase-producing bacteria in vitro and in an ex vivo porcine skin infection model. Fluorescent polystyrene nanoparticles, encapsulated in the hydrogels as model cargo, were released at rates that closely tracked hydrogel wet mass loss, confirming β-lactamase-triggered controlled cargo release. Nonresponsive hydrogels, lacking the β-lactam crosslinker, remained stable in the presence of β-lactamases and β-lactamase-producing bacteria and exhibited no change in mass or nanoparticle release. Furthermore, the responsive hydrogels remained stable in non-β-lactamase enzymes, including collagenases and lipases. These hydrogels have the potential to be used as a bacteria-triggered drug delivery system to control unnecessary exposure to encapsulated antimicrobials, which can provide effective infection treatment without exacerbating resistance.

Synthesis of fluorescent polystyrene nanoparticles: a reproducible and scalable method

Methods Polystyrene nanoparticles (PS NPs) were fabricated from an emulsion of PS/toluene in water using several surfactants, and purified via dialysis in a simple procedure. The synthesis process was carried out at room temperature, without hazardous chemicals, and with a workload of 5 h. All relevant parameters–surfactant type, component concentration, solvent volume, polymer chain length, sonication time–were varied and the effect on the size of the resulting PS NPs were determined. A robust PS NP synthesis procedure was developed, repeated, and tested by three independent researches. The procedure was up-scaled to prove the applicability of the method and the NPs were prepared with four different hydrophobic dyes. Results All products were found to be comparable, and we conclude that the method can provide PS NPs with or without dye dopants with an average diameter of 25 nm in a reproducible size distribution.

Orally administered fluorescent nanosized polystyrene particles affect cell viability, hormonal and inflammatory profile, and behavior in treated mice

Commercially manufactured or generated through environmental degradation, microplastics (MPs) and nanoplastics (NPs) considerably contribute to environmental pollution. There is a knowledge gap in how exposure to MPs/NPs changes cellular function and affects animal and human health. Here, we demonstrate that after oral uptake, fluorescent polystyrene (PS) nanoparticles pass through the mouse digestive system, accumulate and aggregate in different organs, and induce functional changes in cells and organs. Using cochlear explant as a novel in vitro system, we confirmed the consequences of PS-MP/NP interaction with inner ear cells by detecting aggregates and hetero-aggregates of PS particles in hair cells. The testes of treated males accumulated MPs/NPs in the interstitial compartment surrounding the seminiferous tubules, which was associated with a statistically significant decrease in testosterone levels. Male mice showed increased secretion of interleukins (IL-12p35 and IL-23) by splenocytes while cyto- and genotoxicity tests indicated impaired cell viability and increased DNA damage in spleen tissue. Males also showed a broad range of anxiogenic responses to PS nanoparticles while hippocampal samples from treated females showed an increased expression of Bax and Nlrp3 genes, indicating a pro-apoptotic/proinflammatory effect of PS treatment. Taken together, induced PS effects are also gender-dependent, and therefore, strongly motivate future research to mitigate the deleterious effects of nanosized plastic particles.

Simultaneous, Single-Particle Measurements of Size and Loading Give Insights into the Structure of Drug-Delivery Nanoparticles.

Nanoparticles are a promising solution for delivery of a wide range of medicines and vaccines. Optimizing their design depends on being able to resolve, understand, and predict biophysical and therapeutic properties, as a function of design parameters. While existing tools have made great progress, gaps in understanding remain because of the inability to make detailed measurements of multiple correlated properties. Typically, an average measurement is made across a heterogeneous population, obscuring potentially important information. In this work, we develop and apply a method for characterizing nanoparticles with single-particle resolution. We use convex lens-induced confinement (CLiC) microscopy to isolate and quantify the diffusive trajectories and fluorescent intensities of individual nanoparticles trapped in microwells for long times. First, we benchmark detailed measurements of fluorescent polystyrene nanoparticles against prior data to validate our approach. Second, we apply our method to investigate the size and loading properties of lipid nanoparticle (LNP) vehicles containing silencing RNA (siRNA), as a function of lipid formulation, solution pH, and drug-loading. By taking a comprehensive look at the correlation between the intensity and size measurements, we gain insights into LNP structure and how the siRNA is distributed in the LNP. Beyond introducing an analytic for size and loading, this work allows for future studies of dynamics with single-particle resolution, such as LNP fusion and drug-release kinetics. The prime contribution of this work is to better understand the connections between microscopic and macroscopic properties of drug-delivery vehicles, enabling and accelerating their discovery and development.

Modular fluorescent nanoparticle DNA probes for detection of peptides and proteins

Fluorescently labeled antibody and aptamer probes are used in biological studies to characterize binding interactions, measure concentrations of analytes, and sort cells. Fluorescent nanoparticle labels offer an excellent alternative to standard fluorescent labeling strategies due to their enhanced brightness, stability and multivalency; however, challenges in functionalization and characterization have impeded their use. This work introduces a straightforward approach for preparation of fluorescent nanoparticle probes using commercially available reagents and common laboratory equipment. Fluorescent polystyrene nanoparticles, Thermo Fisher Scientific FluoSpheres, were used in these proof-of-principle studies. Particle passivation was achieved by covalent attachment of amine-PEG-azide to carboxylated particles, neutralizing the surface charge from − 43 to − 15 mV. A conjugation-annealing handle and DNA aptamer probe were attached to the azide-PEG nanoparticle surface either through reaction of pre-annealed handle and probe or through a stepwise reaction of the nanoparticles with the handle followed by aptamer annealing. Nanoparticles functionalized with DNA aptamers targeting histidine tags and VEGF protein had high affinity (EC50s ranging from 3 to 12 nM) and specificity, and were more stable than conventional labels. This protocol for preparation of nanoparticle probes relies solely on commercially available reagents and common equipment, breaking down the barriers to use nanoparticles in biological experiments.

Dielectrophoretic Trapping for Nanoparticles, High-Molecule-Weight DNA, and SYBR Gold Using Polyimide-Based Printed Circuit Board

Microelectrodes for electrokinetic detection have been employed to manipulate and collect nanomaterials and biomaterials. However, the process of microelectrode patterning has been researched in the semiconductor fabrication process, which is expensive and time-consuming. Here, we demonstrate dielectrophoretic trapping of 200 nm fluorescent polystyrene nanoparticles and λ-deoxyribonucleic acid with SYBR gold using dielectrophoretic trapping onto an interdigitated circular electrode (ICE). For dielectrophoretic trapping, an ICE is designed and an ordered flexible printed circuit board (FPCB), possessing a non-porous polyimide substrate, is constructed. The ICE FPCB could identify the dielectrophoretic trapping conditions and simulate them by the finite element method. The fluorescent molecules in the ICE FPCB are dielectrophoretically trapped at the finger edges of the ICE when a peak-to-peak voltage of 12 V is applied at 6 kHz for 15-20 min. Moreover, monitoring of the dielectrophoretic trapping effect of SYBR gold eliminated the false negative detection error.

The Effects of Combined Ocean Acidification and Nanoplastic Exposures on the Embryonic Development of Antarctic Krill

In aquatic environments, plastic pollution occurs concomitantly with anthropogenic climate stressors such as ocean acidification. Within the Southern Ocean, Antarctic krill (Euphausia Superba) support many marine predators and play a key role in the biogeochemical cycle. Ocean acidification and plastic pollution have been acknowledged to hinder Antarctic krill development and physiology in singularity, however potential multi-stressor effects of plastic particulates coupled with ocean acidification are unexplored. Furthermore, Antarctic krill may be especially vulnerable to plastic pollution due to their close association with sea-ice, a known plastic sink. Here, we investigate the behaviour of nanoplastic [spherical, aminated (NH2), and yellow-green fluorescent polystyrene nanoparticles] in Antarctic seawater and explore the single and combined effects of nanoplastic (160 nm radius, at a concentration of 2.5 μg ml–1) and ocean acidification (pCO2 ∼900, pHT 7.7) on the embryonic development of Antarctic krill. Gravid female krill were collected in the Atlantic sector of the Southern Ocean (North Scotia Sea). Produced eggs were incubated at 0.5 °C in four treatments (control, nanoplastic, ocean acidification and the multi-stressor scenario of nanoplastic presence, and ocean acidification) and their embryonic development after 6 days, at the incubation endpoint, was determined. We observed that negatively charged nanoplastic particles suspended in seawater from the Scotia Sea aggregated to sizes exceeding the nanoscale after 24 h (1054.13 ± 53.49 nm). Further, we found that the proportion of embryos developing through the early stages to reach at least the limb bud stage was highest in the control treatment (21.84%) and lowest in the multi-stressor treatment (13.17%). Since the biological thresholds to any stressors can be altered by the presence of additional stressors, we propose that future nanoplastic ecotoxicology studies should consider the changing global ocean under future climate scenarios for assessments of their impact and highlight that determining the behaviour of nanoplastic particles used in incubation studies is critical to determining their toxicity.

An ex Vivo Model Enables Systematic Investigation of the Intestinal Absorption and Transcytosis of Oral Particulate Nanocarriers.

Nanoparticulate formulations are being developed toward enhancing the bioavailability of orally administrated biologics. However, the processes mediating particulate carriers' intestinal uptake and transport remains to be fully elucidated. Herein, an optical clearing-based whole tissue mount/imaging strategy was developed to enable high quality microscopic imaging of intestinal specimens. It enabled the distribution of nanoparticles within intestinal villi to be quantitatively analyzed at a cellular level. Two-hundred and fifty nm fluorescent polystyrene nanoparticles were modified with polyethylene glycol (PEG), Concanavalin A (ConA), and pectin to yield mucopenetrating, enterocyte targeting, and mucoadhesive model nanocarriers, respectively. Introducing ConA on the PEGylated nanoparticles significantly increased their uptake in the intestinal epithelium (∼4.16 fold for 200 nm nanoparticle and ∼2.88 fold for 50 nm nanoparticles at 2 h). Moreover, enterocyte targeting mediated the trans-epithelial translocation of 50 nm nanoparticles more efficiently than that of the 200 nm nanoparticles. This new approach provides an efficient methodology to obtain detailed insight into the transcytotic activity of enterocytes as well as the barrier function of the constitutive intestinal mucus. It can be applied to guide the rational design of particulate formulations for more efficient oral biologics delivery.

Ingestion of bivalve droppings by benthic invertebrates may lead to the transfer of nanomaterials in the aquatic food chain

BackgroundManufactured nanomaterials (MNMs) are released into the environment in increasing quantities. Consequently, MNMs also reach the aquatic environment, where they can interact with different organisms. Previous studies have already shown that filter-feeding bivalves can ingest nanomaterials from the surrounding water leading to higher concentration of the material. Furthermore, they have been shown to be vectors for environmental chemicals and pathogens to other organisms, as their feces/pseudofeces (F/pF) play a crucial role as a food source for other species. We exposed bivalves (Corbicula sp.) to MNMs and performed experiments to investigate the possible transport of MNMs by their feces to the benthic amphipod Hyalella azteca. Silver (Ag) and gold (Au) nanoparticles (NPs) as well as fluorescent polystyrene nanoparticles were used in this study. They allowed the investigation of the metal content of the bivalves’ feces and the amphipods feeding on it, as well as the localization of the fluorescent particles in the body of the animals.ResultsExamination of the feces by fluorescence microscope and determination of the total metal content by inductively coupled plasma mass spectrometry (ICP-MS) showed a high accumulation of the exposed MNMs in the F/pF. The examination of fecal matter, using transmission electron microscopy confirmed the nanoparticulate character of the metals in the examined fecal matter. After exposure of amphipods to the MNMs containing fecal matter, the fluorescent MNMs were localized in the animals gut. The chronic exposure of juvenile amphipods over 21 days to feces enriched with Au MNMs caused significant effects on the growth of the amphipods. The transfer of both metals (Ag and Au) from the fecal matter to the amphipods was confirmed after total metal measurements.ConclusionProbably, for the first time, it has been shown that when exposed to MNMs bivalves can transfer these particles to other benthic species. Transfer is via released F/pF upon which the benthic species feed and thus could ingest the particles. The high concentrations of MNMs in the fecal matter raises concerns about the potential accumulation and transfer of the materials and associated ecotoxicological effects in invertebrates such as benthic amphipods.

Single-particle fibrinogen detection using platelet membrane-coated fluorescent polystyrene nanoparticles.

Fibrinogen participates in many physiological processes and is a biomarker for a variety of diseases. On this account, the development of a sensitive method for fibrinogen assay is particularly important. Herein, we demonstrate a new color-coded single-particle detection (SPD) method for fibrinogen detection by using platelet membrane-coated fluorescent polystyrene nanoparticles (PNPs) as the probes. Due to the specific interactions between fibrinogen and integrin receptors on platelet membranes, PNPs can form aggregated structures in the presence of fibrinogen. Therefore, colocalization events between green and red PNPs and the corresponding Pearson's correlation coefficient vary with the concentrations of fibrinogen. The sensing ability shows a linear range of 30-300 μg mL-1 and a limit of detection (LOD) of 3.9 μg mL-1 (11.3 nM) for fibrinogen detection. Moreover, it has been validated that the proposed biosensor can selectively detect fibrinogen and shows a good performance in real sample applications.

Nanoparticles exhibit greater accumulation in kidney glomeruli during experimental glomerular kidney disease.

Loss and dysfunction of glomerular podocytes result in increased macromolecule permeability through the glomerular filtration barrier and nephrotic syndrome. Current therapies can induce and maintain disease remission, but cause serious and chronic complications. Nanoparticle drug carriers could mitigate these side effects by delivering drugs to the kidneys more efficiently than free drug through tailoring of carrier properties. An important extrinsic factor of nanoparticle biodistribution is local pathophysiology, which may drive greater nanoparticle deposition in certain tissues. Here, we hypothesized that a “leakier” filtration barrier during glomerular kidney disease would increase nanoparticle distribution into the kidneys. We examined the effect of nanoparticle size and disease state on kidney accumulation in male BALB/c mice. The effect of size was tested using a panel of fluorescent polystyrene nanoparticles of size 20–200 nm, due to the relevance of this size range for drug delivery applications.Experimental focal segmental glomerulosclerosis was induced using an anti‐podocyte antibody that causes abrupt podocyte depletion. Nanoparticles were modified with carboxymethyl‐terminated poly(ethylene glycol) for stability and biocompatibility. After intravenous injection, fluorescence from nanoparticles of size 20 and 100 nm, but not 200 nm, was observed in kidney glomeruli and peritubular capillaries. During conditions of experimental focal segmental glomerulosclerosis, the number of fluorescent nanoparticle punctae in kidney glomeruli increased by 1.9‐fold for 20 and 100 nm nanoparticles compared to normal conditions. These findings underscore the importance of understanding and leveraging kidney pathophysiology in engineering new, targeted drug carriers that accumulate more in diseased glomeruli to treat glomerular kidney disease.

Dual function of Eosin Y in miniemulsion polymerization: Stabilizer and FRET acceptor

Recent studies have shown that water-soluble fluorescent dyes can be used as miniemulsion stabilizers to avoid a series of deficiencies in traditional stabilizers. However, visual means to demonstrate the stabilizing performance of water-soluble dyes are still scarce. In this study, the principle of fluorescence resonance energy transfer (FRET) is combined with dual emission of fluorescent polystyrene nanoparticles prepared by miniemulsion polymerization. For this purpose, 4-(1,2,2-triphenylethyl) phenylundecene-10-enoate (TPE-Aly) was employed as aggregation induced emission (AIE) dye and donor and Eosin Y as acceptor and stabilizer. The polymer molecular weight, particle size and size distribution of the nanoparticles were optimized by regulating the amount of Eosin Y and ultrasonication time, respectively. The average number of Eosin Y molecules per nanoparticle was firstly estimated by combining dialysis and Lambert-Beer law. FRET between TPE-Aly and Eosin Y indicates the presence of Eosin Y on the surface of the polymer nanoparticles. With increasing concentration of Eosin Y, the as-prepared dual-emission nanoparticles display a decreasing particle size along with an enhancing FRET efficiency. Moreover, various stability tests show that Eosin Y can be used as an excellent stabilizer for miniemulsion polymerization and has broad application prospects in the field of functional materials research.

Development of a Pemetrexed/Folic Acid Nanoformulation: Synthesis, Characterization, and Efficacy in a Murine Colorectal Cancer Model.

The folate analogue pemetrexed (PEM) is an approved therapeutic for non-small cell lung cancer and malignant pleural mesothelioma with the potential for broader application in combination therapies. Here, we report the development of a nanoformulation of PEM and its efficacy against the CT26 murine colorectal cancer cell line in vitro and in vivo. Utilizing layer-by-layer deposition, we integrate PEM, along with folic acid (FA), onto a fluorescent polystyrene nanoparticle (NP) substrate. The final nanoformulation (PEM/FA-NP) has a size of ∼40 nm and a zeta potential of approximately −20 mV. Cell uptake studies indicated increased uptake in vitro for the PEM/FA-NP compared to the uncoated NP, likely due to the presence of PEM and FA. Viability studies were performed to determine the potency of the PEM/FA-NP formulation against CT26 cells. Syngeneic CT26 tumors in BALB/c mice showed reduced growth when treated once daily (2.1 mg/kg PEM) for 3 days with PEM/FA-NP versus the vehicle (uncoated) control, with no observable signs of systemic toxicity associated with the nanoformulation. Although the current study size is limited (n = 4 animals for each group), the overall performance and biocompatibility of the PEM/FA-NP observed suggest that further optimization and larger-scale studies may be warranted for this novel formulation.
