Integrated resistance management for acaricide use on Varroa destructor

The global beekeeping industry faces an escalating challenge in the form of Varroa destructor. Synthetic chemicals serve as a cornerstone for varroa management, although they face a major challenge in the form of acaricide resistance. Here, I examine acaricide resistance in varroa under the framework of Insecticide Resistance Management (IRM). I assess the potential of diverse IRM strategies, such as pesticide rotation and mixtures, refuge utilization, synergists and the integration of non-persistent chemicals. The peculiar life history of varroa, characterized by its incestuous breeding system, challenges conventional IRM strategies. There is little published evidence that pesticide rotation is beneficial for resistance management in varroa, with several studies showing resistance is maintained despite rotation. Fitness costs associated with pesticide resistance are often an essential component for IRM strategies, but there are no current data from varroa demonstrating such specific fitness costs (e.g., a reduced relative oviposition rate) associated with resistance. The single published experimental study directly examining relative fitness found that here was little or no reproductive fitness cost associated with pyrethroid resistance. More work is needed on fitness effects of the key acaricides, which would better guide the use of rotation and refuge strategies. A key prospect for future work that has been identified through simulation modeling is offered by pesticide mixtures and the role of synergists to elevate acaricide efficacy. Additional tools for varroa IRM include ‘soft’ acaricides, including oxalic acid, and biopesticides such as dsRNA. In light of the widespread prevalence of acaricide resistance and an increasing varroa problem, there is an urgent need for nuanced, data-driven varroa IRM strategies.

House Sparrows Vary Seasonally in Their Ability to Transmit West Nile Virus.

AbstractSeasonality in infectious disease prevalence is predominantly attributed to changes in exogenous risk factors. For vectored pathogens, high abundance, activity, and/or diversity of vectors can exacerbate disease risk for hosts. Conversely, many host defenses, particularly immune responses, are seasonally variable. Seasonality in host defenses has been attributed, in part, to the proximate (i.e., metabolic) and ultimate (i.e., reproductive fitness) costs of defense. In this study, our goal was to discern whether any seasonality is observable in how a common avian host, the house sparrow (Passer domesticus), copes with a common zoonotic arbovirus, the West Nile virus (WNV), when hosts are studied under controlled conditions. We hypothesized that if host biorhythms play a role in vector-borne disease seasonality, birds would be most vulnerable to WNV when breeding and/or molting (i.e., when other costly physiological activities are underway) and thus most transmissive of WNV at these times of year (unless birds died from infection). Overall, the results only partly supported our hypothesis. Birds were most transmissive of WNV in fall (after their molt is complete and when WNV is most prevalent in the environment), but WNV resistance, WNV tolerance, and WNV-dependent mortality did not vary among seasons. These results collectively imply that natural arboviral cycles could be partially underpinned by endogenous physiological changes in hosts. However, other disease systems warrant study, as this result could be specific to the nonnative and highly commensal nature of the house sparrow or a consequence of the relative recency of the arrival of WNV to the United States.

Fitness Costs Associated With Insecticide Resistance in Populations of Homalodisca vitripennis Germar (Hemiptera: Cicadellidae).

The glassy-winged sharpshooter, Homalodisca vitripennis (Germar) (Hemiptera: Cicadellidae), is an important vector of bacterial Xyllela diseases throughout the southern and southwestern portions of the United States. Strong insect control measures, such that population densities of the insect vector are significantly reduced, are often necessary to limit the spread of Xylella fastidiosa. Glassy-winged sharpshooter populations within the Central Valley of California have developed a high resistance to imidacloprid (resistance ratio greater than 3,200) and tolerance to pyrethroids (ratio of less than 10) due to frequent applications of these materials. The purpose of this study was to determine the potential effects of insecticide resistance upon a variety of sharpshooter life history parameters associated with reproductive fitness. Our results indicate that individuals from susceptible populations of glassy-winged sharpshooters exhibited significantly higher fecundity and longer adult lifespans than those from the resistant populations. Additionally, resistant individuals were on average slightly larger than susceptible individuals. These results provide a strong indication that resistance to neonicotinoids imposes a reproductive fitness cost in an insecticide-free environment.

Pyrokinin receptor silencing in females of the southern cattle tick Rhipicephalus (Boophilus) microplus is associated with a reproductive fitness cost

BackgroundRhipicephalusmicroplus is the vector of deadly cattle pathogens, especially Babesia spp., for which a recombinant vaccine is not available. Therefore, disease control depends on tick vector control. However, R.microplus populations worldwide have developed resistance to available acaricides, prompting the search for novel acaricide targets. G protein-coupled receptors (GPCRs) are involved in the regulation of many physiological processes and have been suggested as druggable targets for the control of arthropod vectors. Arthropod-specific signaling systems of small neuropeptides are being investigated for this purpose. The pyrokinin receptor (PKR) is a GPCR previously characterized in ticks. Myotropic activity of pyrokinins in feeding-related tissues of Rhipicephalussanguineus and Ixodesscapularis was recently reported.MethodsThe R.microplus pyrokinin receptor (Rhimi-PKR) was silenced through RNA interference (RNAi) in female ticks. To optimize RNAi, a dual-luciferase assay was applied to determine the silencing efficiency of two Rhimi-PKR double-stranded RNAs (dsRNA) prior to injecting dsRNA in ticks to be placed on cattle. Phenotypic variables of female ticks obtained at the endpoint of the RNAi experiment were compared to those of control female ticks (non-injected and beta-lactamase dsRNA-injected). Rhimi-PKR silencing was verified by quantitative reverse-transcriptase PCR in whole females and dissected tissues.ResultsThe Rhimi-PKR transcript was expressed in all developmental stages. Rhimi-PKR silencing was confirmed in whole ticks 4 days after injection, and in the tick carcass, ovary and synganglion 6 days after injection. Rhimi-PKR silencing was associated with an increased mortality and decreased weight of both surviving females and egg masses (P < 0.05). Delays in repletion, pre-oviposition and incubation periods were observed (P < 0.05).ConclusionsRhimi-PKR silencing negatively affected female reproductive fitness. The PKR appears to be directly or indirectly associated with the regulation of female feeding and/or reproductive output in R.microplus. Antagonists of the pyrokinin signaling system could be explored for tick control.Graphical abstract

Lack of Evidence for Fitness Costs in Soybean Aphid (Hemiptera: Aphididae) With Resistance to Pyrethroid Insecticides in the Upper Midwest Region of the United States.

Twenty years after the arrival of soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), in the United States, it remains the most economically important arthropod pest of soybean in the Upper Midwest Region. After years of repeated and sustained insecticidal pressures placed on the aphid, resistance to the pyrethroid class of insecticides has been documented in multiple years over a large geographic area. In this study, the fitness of aphid isolates displaying resistant and susceptible phenotypes to λ-cyhalothrin were compared within several experiments over three soybean-growing seasons. Rates of population increase were evaluated on whole plants in the greenhouse, intrinsic rates of increase were calculated from leaf discs in the laboratory, and aphid size and asymmetry were compared through tibial measurements. No evidence of a fitness cost associated with the resistant phenotype was seen in any of our experiments. In contrast, individual resistant isolates occasionally supported significantly higher fitness values than some susceptible isolates. Additionally, a pooled analysis comparing resistant and susceptible phenotypes across years and isolates revealed that, on average, the resistant phenotype had significantly higher fitness values than the susceptible phenotype in most experiments. The lack of reproductive fitness costs associated with the pyrethroid-resistant phenotype raises concerns for longevity of pyrethroid use in soybean aphid management.

A retrospective view of early research on dominance, stress and reproduction in cooperatively breeding carnivores

Social carnivores have been central in studies of cooperative breeding, and research using noninvasive methods to examine behavioral and endocrine mechanisms of reproductive suppression started in the 1980s with dwarf mongooses in Serengeti National Park. Here, I synthesize the methods, findings and limitations of a research program that examined relationships between social dominance, age, mass, aggression, mating, gonadal steroids, glucocorticoids and reproduction in female and male dwarf mongooses, African wild dogs and wolves. Infanticide is a reliable backstop for reproductive suppression in females, and reproduction is energetically costly in these species. These conditions favor hypothalamic – pituitary – gonadal (HPG) adaptations that reduce the fertility of subordinate females to avoid the cost of producing doomed offspring. Infanticide also favors close synchronization of reproduction when subordinate females do become pregnant. In males, infanticide is a less reliable backstop and reproduction is less costly, so direct effects of subordination on fertility are less pronounced. Age is a strong predictor of social dominance in these species, but the evolutionary reason for this is not clear. In dwarf mongooses and wild dogs, alpha females were never deposed by younger packmates, but alpha males were: this difference is also not understood. Patterns of reproduction supported models predicting that alphas are less likely to share reproduction when the fitness costs of reproduction are high, when the fitness expected for dispersers is low, and with young subordinates to whom they are more closely related. Correlations between dominance and adrenal glucocorticoid concentrations varied between species and sexes, but did not support the hypothesis that chronic stress causes reproductive suppression.

Context-dependent fitness costs of reproduction despite stable body mass costs in an Arctic herbivore.

The cost of reproduction on demographic rates is often assumed to operate through changing body condition. Several studies have found that reproduction depresses body mass more if the current conditions are severe, such as high population densities or adverse weather, than under benign environmental conditions. However, few studies have investigated the association between the fitness components and body mass costs of reproduction. Using 25years of individual-based capture-recapture data from Svalbard reindeer Rangifer tarandus platyrhynchus, we built a novel Bayesian state-space model that jointly estimated interannual change in mass, annual reproductive success and survival, while accounting for incomplete observations. The model allowed us to partition the differential effects of intrinsic and extrinsic factors on both non-reproductive mass change and the body mass cost of reproduction, and to quantify their consequences on demographic rates. Contrary to our expectation, the body mass cost of reproduction (mean=-5.8kg) varied little between years (CV=0.08), whereas the between-year variation in body mass changes, that were independent of the previous year's reproductive state, varied substantially (CV=0.4) in relation to autumn temperature and the amount of rain-on-snow in winter. This body mass loss led to a cost of reproduction on the next reproduction, which was amplified by the same environmental covariates, from a 10% reduction in reproductive success in benign years, to a 50% reduction in harsh years. The reproductive mass loss also resulted in a small reduction in survival. Our results show how demographic costs of reproduction, driven by interannual fluctuations in individual body condition, result from the balance between body mass costs of reproduction and body mass changes that are independent of previous reproductive state. We illustrate how a strong context-dependent fitness cost of reproduction can occur, despite a relatively fixed body mass cost of reproduction. This suggests that female reindeer display a very conservative energy allocation strategy, either aborting their reproductive attempt at an early stage or weaning at a relatively constant cost. Such a strategy might be common in species living in a highly stochastic and food limited environment.

Evaluation of Developmental and Reproductive Fitness of Laboratory Selected Thiamethoxam Resistance in Black Legume Aphid Aphis craccivora Koch.

Thiamethoxam is a neonicotinoid group of insecticide which are selective agonists of the insect nicotinic acetylcholine receptor (nAChR), a pentameric cys-loop ligand-gated ion channel located in the central nervous system of insects. They provide farmers with invaluable, highly effective tools against some of the world’s most destructive crop pests. Black legume aphids, Aphis craccivora is one of the devastating polyphagous pests in agroecosystem and to manage their population, farmers are still relying on application of synthetic insecticides like Thiamethoxam. To evaluate the risk of resistance development, selection of A. craccivora was done to characterize Thiamethoxam resistance along with evaluation of developmental and reproductive fitness cost of the resistance. About 86.19 folds of resistance were developed after selecting for 24 generations. In the first twelve generations steady development of resistance was noticed followed by development become stiffer till 16th generation and more or less stable till 24th generations. Relative fitness of the selected resistant strain of aphids was decreased with longer nymphal duration and adult longevity. The mean nymphal duration was increased from 4.35±1.02 days (in F0) to 7.9±0.57 days (in F24). The fecundity rate was significantly less in resistant population (16.0±5.34) as compared to the susceptible strain (54.71±7.63) whereas, oviposition periods were significantly longer (12.49±1.44 days) in resistant population as compare to susceptible strain (9.50±1.22 days).

Male and female reproductive fitness costs of an immune response in natural populations.

Parasites can mediate host fitness both directly, via effects on survival and reproduction, or indirectly by inducing host immune defense with costly side-effects. The evolution of immune defense is determined by a complex interplay of costs and benefits of parasite infection and immune response, all of which may differ for male and female hosts in sexual lineages. Here, we examine fitness costs associated with an inducible immune defense in a fish-cestode host-parasite system. Cestode infection induces peritoneal fibrosis in threespine stickleback (Gasterosteus aculeatus), constraining cestode growth and sometimes encasing and killing the parasite. Surveying two wild populations of stickleback, we confirm that the presence of fibrosis scar tissue is associated with reduced parasite burden in both male and female fish. However, fibrotic fish had lower foraging success and reproductive fitness (reduced female egg production and male nesting success), indicating strong costs of the lingering immunopathology. Consistent with substantial sexually concordant fitness effects of immune response, we find alignment of multivariate selection across the sexes despite sexual antagonism over morphological shape. Although both sexes experienced costs of fibrosis, the net impacts are unequal because in the two study populations females had higher cestode exposure. To evaluate whether this difference in risk should drive sex-specific immune strategies, we analyze a quantitative genetic model of host immune response to a trophically transmitted parasite. The model and empirical data illustrate how shared costs and benefits of immune response lead to shared evolutionary interests of male and female hosts, despite unequal infection risks across the sexes.

Physiological and reproductive fitness cost in Aedes aegypti on exposure to toxic xenobiotics in New Delhi, India

Aedes aegypti, is a well-known vector of dengue, Chikungunya and Zika at the global level. Primary use of pyrethroids as control interventions has caused the development of a considerable level of immunity in Ae. aegypti. The current study assessed the efficacy of a pyrethroid, ?-cypermethrin on the survival and various life parameters of Ae. aegypti. The larvicidal studies with ?-cypermethrin revealed the respective LC50 and LC90 values as 0.26526 mg/L and 0.60211 mg/L. The impact of LC50 level was assessed on the growth and life attributes; such as gonotrophic cycle, egg development, hatchability, development and survival of immature stages, adult longevity, reproduction rate and generation time; of fourth instar of susceptible (S) and ?-cypermethrin-exposed population (E). The exposed population showed diminished fitness as compared to the susceptible population. The individual female fecundity in susceptible population was recorded as 79.6 with 61.6% hatchability rate as compared to the 28 eggs/female and 25% hatchability in the exposed population. The mean egg hatch time in S strain increased by 2-fold in E strain. The proportion of immature survival observed in S strain was 0.88 for fourth instar to pupa (P/I), 0.94 for pupa to adult (A/P) and an overall 0.83 for fourth larva to adult (A/I), which respectively reduced to 0.32, 0.86 and 0.27 in E strain of Ae. aegypti. Likewise, the net reproductive rate, birth rate and death rate were significantly (p &lt; 0.05) higher in S than in E strain. This study demonstrates the negative impact of ?-cypermethrin on the physiological and reproductive fitness of Ae. aegypti.

A random small molecule library screen identifies novel antagonists of the kinin receptor from the cattle fever tick, Rhipicephalus microplus (Acari: Ixodidae).

The southern cattle tick, Rhipicephalus microplus, is a primary vector of the deadly bovine disease babesiosis. Worldwide populations of ticks have developed resistance to acaricides, underscoring the need for novel target discovery for tick control. The arthropod-specific R. microplus kinin receptor is such a target, previously validated by silencing, which resulted in female reproductive fitness costs, including a reduced percentage of eggs hatching. In order to identify potent small molecules that bind and activate or inhibit the kinin receptor, a high-throughput screening (HTS) assay was developed using a CHO-K1 cell line expressing the recombinant tick kinin receptor (BMLK3 ). A total of ~20 000 molecules from a random in-house small molecule library were screened in a 'dual-addition' calcium fluorescence assay. This was followed by dose-response validation of the hit molecules identified both from HTS and an in silico screen of ~390 000 molecules. We validated 29 antagonists, 11 of them were full antagonists with IC50 values between 0.67 and 8 μmol L-1 . To explore the structure-activity relationships (SAR) of the small molecules, we tested the activities of seven analogs of the most potent identified antagonist, additionally discovering three full antagonists and four partial antagonists. These three potent antagonists (IC50 < 3.2 μmol L-1 ) were validated in vitro using the recombinant mosquito kinin receptor and showed similar antagonistic activities. In vivo, these three compounds also inhibited the mosquito hindgut contraction rate induced by a myotropic kinin agonist analog 1728. Antagonists identified in this study could become pesticide leads and are reagents for probing the kinin signaling system. © 2021 Society of Chemical Industry.

Forage availability and maternal characteristics affect costs of reproduction in a large marsupial.

Life history theory predicts trade-offs in allocation between survival, maintenance, growth, and reproduction, especially when resources are scarce. Individual variation in resource acquisition can affect trade-offs, but is often unaccounted for. We quantified the fitness costs of reproduction, accounting for environmental conditions, maternal characteristics and individual variation. We analyzed 10years of data from marked kangaroos to evaluate how reproductive allocation affected annual mass change and skeletal growth, subsequent fecundity and weaning success, and survival, accounting for maternal mass or size and forage availability. Through repeated measurements of 76-91 females, we investigated how trade-offs varied within and between individuals, assessing whether individual variation could mask population-level trade-offs. In poor environments, females that weaned an offspring lost mass. Females that nursed an offspring for > 7months had reduced skeletal growth. Females that did not gain mass over the previous 12months rarely reproduced, especially if they had nursed an offspring for > 7months the previous year. Reproductive allocation had no effect on weaning success, which was very low, and did not affect maternal survival, suggesting a conservative strategy. Disentangling within- and between-individual responses revealed trade-offs within individuals, but because individuals did not vary in their responses to earlier effort, these trade-offs did not drive population trends. The interacting effects of environmental conditions, maternal characteristics and individual variation on allocation trade-offs demonstrate the importance of long-term monitoring for understanding life history variations in changing environments.

Exposure of Anophelesmosquitoes to trypanosomes reduces reproductive fitness and enhances susceptibility to Plasmodium.

During a blood meal, female Anopheles mosquitoes are potentially exposed to diverse microbes in addition to the malaria parasite, Plasmodium. Human and animal African trypanosomiases are frequently co-endemic with malaria in Africa. It is not known whether exposure of Anopheles to trypanosomes influences their fitness or ability to transmit Plasmodium. Using cell and molecular biology approaches, we found that Trypanosoma brucei brucei parasites survive for at least 48h after infectious blood meal in the midgut of the major malaria vector, Anopheles coluzzii before being cleared. This transient survival of trypanosomes in the midgut is correlated with a dysbiosis, an alteration in the abundance of the enteric bacterial flora in Anopheles coluzzii. Using a developmental biology approach, we found that the presence of live trypanosomes in mosquito midguts also reduces their reproductive fitness, as it impairs the viability of laid eggs by affecting their hatching. Furthermore, we found that Anopheles exposure to trypanosomes enhances their vector competence for Plasmodium, as it increases their infection prevalence. A transcriptomic analysis revealed that expression of only two Anopheles immune genes are modulated during trypanosome exposure and that the increased susceptibility to Plasmodium was microbiome-dependent, while the reproductive fitness cost was dependent only on the presence of live trypanosomes but was microbiome independent. Taken together, these results demonstrate multiple effects upon Anopheles vector competence for Plasmodium caused by eukaryotic microbes interacting with the host and its microbiome, which may in turn have implications for malaria control strategies in co-endemic areas.

Climate change, sex reversal and lability of sex-determining systems.

Sex reversal at high temperatures during embryonic development (e.g., ZZ females) provides the opportunity for new genotypic crosses (e.g., ZZ male×ZZ female). This raises the alarming possibility that climatic warming could lead to the loss of an entire chromosome-one member of the sex chromosome pair (the Y or W)-and the transition of populations to environmental sex determination (ESD). Here we examine the evolutionary dynamics of sex-determining systems exposed to climatic warming using theoretical models. We found that the loss of sex chromosomes is not an inevitable consequence of sex reversal. A large frequency of ZZ sex reversal (50% reversal from male to female) typically divides the outcome between loss of the ZW genotype and the stable persistence of ZZ males, ZW females and ZZ females. The amount of warming associated with sex chromosome loss depended on several features of wild populations-environmental fluctuation, immigration, heritable variation in temperature sensitivity and differential fecundity of sex-reversed individuals. Chromosome loss was partially or completely buffered when sex-reversed individuals suffered a reproductive fitness cost, when immigration occurred or when heritable variation for temperature sensitivity existed. Thus, under certain circumstances, sex chromosomes may persist cryptically in systems where the environment is the predominant influence on sex.

Differences in growth, development and innate seed dormancy of susceptible and fenoxaprop-P non-target site resistant black-grass sub-populations

Alopecurus myosuroides Huds. is the most abundant herbicide-resistant weed species in Western Europe. Herbicide resistance management programs may rely, amongst other things, on information about fitness of resistant sub-population i.e. the number of offspring a genotype produces and contributes to next generation. Growth and phenological development of susceptible (S) and fenoxaprop-P resistant (R) A. myosuroides sub-populations from a non-target site resistant (NTSR) Danish population was studied for two years, while the primary seed dormancy of the populations was evaluated for three years. The genetic background of the S and R sub-populations was controlled using a plant cloning technique. No statistically significant differences were observed in the development of vegetative growth stages between the R and S sub-populations. In contrast, the R sub-population required more thermal time (at least c. 40 °C d) compared with the S sub-population to reach the reproductive phenological stages including booting and flowering. There were no vegetative (biomass) and reproductive (seed production) fitness costs in the R sub-population, however, the harvest index of the R sub-population was significantly lower than of the S sub-population. Moreover, the R sub-population had a lower primary dormancy than the S sub-population. The lower primary dormancy of the R sub-population can be considered as agronomical fitness costs of the NTSR to fenoxaprop-P, if pre-sowing seedbed preparation are implemented. The results can be implemented in models simulating phenological development and in models predicting the proportion of resistant individuals in a population.

Alterations in Life-History Associated With Non-target-site Herbicide Resistance in Alopecurus myosuroides.

The evolution of resistance to herbicides is a classic example of rapid contemporary adaptation in the face of a novel environmental stress. Evolutionary theory predicts that selection for resistance will be accompanied by fitness trade-offs in environments where the stress is absent. Alopecurus myosuroides, an autumn-germinating grass weed of cereal crops in North-West Europe, has evolved resistance to seven herbicide modes-of-action, making this an ideal species to examine the presence and magnitudes of such fitness costs. Here, we use two contrasting A. myosuroides phenotypes derived from a common genetic background, one with enhanced metabolism resistance to a commercial formulation of the sulfonylurea (ALS) actives mesosulfuron and iodosulfuron, and the other with susceptibility to these actives (S). Comparisons of plant establishment, growth, and reproductive potential were made under conditions of intraspecific competition, interspecific competition with wheat, and over a gradient of nitrogen deprivation. Herbicide dose response assays confirmed that the two lines had contrasting resistance phenotypes, with a 20-fold difference in resistance between them. Pleiotropic effects of resistance were observed during plant development, with R plants having a greater intraspecific competitive effect and longer tiller lengths than S plants during vegetative growth, but with S plants allocating proportionally more biomass to reproductive tissues during flowering. Direct evidence of a reproductive cost of resistance was evident in the nitrogen deprivation experiment with R plants producing 27% fewer seed heads per plant, and a corresponding 23% reduction in total seed head length. However, these direct effects of resistance on fecundity were not consistent across experiments. Our results demonstrate that a resistance phenotype based on enhanced herbicide metabolism has pleiotropic impacts on plant growth, development and resource partitioning but does not support the hypothesis that resistance is associated with a consistent reproductive fitness cost in this species. Given the continued difficulties associated with unequivocally detecting costs of herbicide resistance, we advocate future studies that adopt classical evolutionary quantitative genetics approaches to determine genetic correlations between resistance and fitness-related plant life history traits.

Long-term studies of bighorn sheep and mountain goats reveal fitness costs of reproduction.

Fitness costs of reproduction are expected when resources are limited. Costs drive the evolution of life-history strategies and can affect population dynamics if females change their allocation of resources to reproduction. We studied fitness costs of reproduction in mountain ungulates in Alberta, Canada. We monitored two populations of bighorn sheep (Ovis canadensis) for 44 and 30years, and one of mountain goats (Oreamnos americanus) for 30years. Both species are highly iteroparous. Heterogeneity in individual reproductive potential makes fitness costs of reproduction difficult to detect and quantify without manipulations. In capital breeders, individual differences can be partly accounted for by considering body mass and other correlates of reproductive potential. Long-term monitoring can reveal costs that only manifest under stressful conditions such as disease or resource scarcity. Despite individual differences in reproductive potential, we detected fitness costs of reproduction in females. Costs, in terms of mass gain and survival, are almost entirely born by subsequent offspring, as mothers prioritize their own maintenance and survival. Costs are greater for primiparous females, decrease with increasing body mass and increase as resource availability declines, and sons are costlier than daughters. Costs may increase for senescent females that appear to reduce allocation to reproduction. In bighorn sheep, costs mostly involve reduced mass gain and lower survival of subsequent offspring. In mountain goats, costs include reductions in mass gain, subsequent fecundity and juvenile survival. In males, fitness costs derive mostly from attempts to reproduce rather than from siring success and likely depend upon individual competitiveness. In the absence of selective harvests, dominant males may enjoy high fitness and possibly lower costs compared to subordinates. The conservative reproductive tactic of mountain ungulate females likely explains why density dependence mostly involves later primiparity and lower recruitment, but rarely affects adult survival. Future research will seek to better account for heterogeneity in reproductive potential, assess cumulative reproductive costs and investigate the potential effects of fathers on maternal allocation tactics.

The leucokinin-like peptide receptor from the cattle fever tick, Rhipicephalus microplus, is localized in the midgut periphery and receptor silencing with validated double-stranded RNAs causes a reproductive fitness cost

The cattle fever tick, Rhipicephalus microplus (Canestrini) (Acari: Ixodidae), is a one-host tick that infests primarily cattle in tropical and sub-tropical regions of the world. This species transmits deadly cattle pathogens, especially Babesia spp., for which a recombinant vaccine is not available. Therefore, disease control depends on tick vector control. Although R. microplus was eradicated in the USA, tick populations in Mexico and South America have acquired resistance to many of the applied acaricides. Recent acaricide-resistant tick reintroductions detected in the U.S. underscore the need for novel tick control methods. The octopamine and tyramine/octopamine receptors, both G protein-coupled receptors (GPCR), are believed to be the main molecular targets of the acaricide amitraz. This provides the proof of principle that investigating tick GPCRs, especially those that are invertebrate-specific, may be a feasible strategy for discovering novel targets and subsequently new anti-tick compounds. The R. microplus leucokinin-like peptide receptor (LKR), also known as the myokinin- or kinin receptor, is such a GPCR. While the receptor was previously characterized in vitro, the function of the leucokinin signaling system in ticks remains unknown. In this work, the LKR was immunolocalized to the periphery of the female midgut and silenced through RNA interference (RNAi) in females. To optimize RNAi experiments, a dual-luciferase system was developed to determine the silencing efficiency of LKR-double stranded RNA (dsRNA) constructs prior to testing those in ticks placed on cattle. This assay identified two effective dsRNAs. Silencing of the LKR with these two validated dsRNA constructs was verified by quantitative real time PCR (qRT-PCR) of female tick dissected tissues. Silencing was significant in midguts and carcasses. Silencing caused decreases in weights of egg masses and in the percentages of eggs hatched per egg mass, as well as delays in time to oviposition and egg hatching. A role of the kinin receptor in tick reproduction is apparent.

Impact of male alternative reproductive tactics on female costs of sexual conflict under variation in operational sex ratio and population density.

Sexual conflict over mating rate is both pervasive and evolutionarily costly. For females, the lifetime reproductive fitness costs that arise through interactions with potential mates will be influenced by the frequency of such interactions, and the fitness cost of each interaction. Both of these factors are likely to be influenced by variation in operational sex ratio (OSR) and population density. Variation in OSR‐ and density‐dependent male alternative reproductive tactics (ARTs) may be particularly important if the fitness costs that females experience vary with the reproductive tactics that males express. Using a simple model, we consider several examples of OSR‐ and/or density‐dependent variation in male ARTs and the frequency of male–female interactions, and find that variation in the expression of male ARTs has the potential to augment or diminish the costs of frequent male interactions for females. Accurately documenting variation in the expression of male ARTs and associated female fitness costs will benefit future work in this area.

Great tits trade off future competitive advantage for current reproduction

Quantifying the fitness costs of reproduction is essential to understand the evolution of reproductive behavior. Recent work shows that increased reproductive investment reduced parental survival in more competitive environments. Here we experimentally test the hypothesis that reproductive investment has a negative effect on the ability of parents to compete for resources in later life. In a nest-box population of Great tits (Parus major), we manipulated family size by reducing or enlarging broods with 2 or 3 nestlings, relative to a control group, in 2 years (N = 237 broods). Parental feeding effort was positively related to experimental family size but leveled off for the enlarged broods. In the next breeding season we manipulated the nest box quality in early spring by reducing the depth of 80% of the nest boxes (deeper boxes are safer and preferred). We analyzed parents' probability of obtaining a deep breeding box in relation to their previous year's family size manipulation. We found for both years that increased reproductive investment negatively affected the probability of parents to claim a high-quality nest box in the subsequent breeding season. We thus confirm that family size has a negative effect on the future competitive ability of parents. Such carry-over effects are important because they show that selection on individual optimal clutch size will depend on 1) resource abundance and the level of competition in the next breeding season and 2) the reproductive investment of the competitors in the current breeding season because it affects their future competitive ability as well.
