Optimization of the ultrasound-assisted extraction of phenolic antioxidants from Cistus salvifoliusL. using response surface methodology.

The current work aims to optimize the ultrasound-assisted extraction (UAE) of Cistus salviifolius L. (aerial parts) antioxidative phenolic compounds using response surface methodology. A Box-Behnken design has been conducted to investigate the effect of four factors, namely: (i) percentage of ethanol (50-90%, v/v), (ii) temperature (40-80°C), (iii) solvent-solid ratio (10-50 mL g-1) and (iv) extraction time (5-25min) on four responses, namely: total phenolic content (TPC), total flavonoid content (TFC) 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical inhibition, and ferric reducing antioxidant power (FRAP). Based on the desirability index, UAE with 50% (v/v) ethanol, at 80 oC, using a solvent-solid ratio of 32.24 mL g-1, for 21 min resulted in the maximum recovery of phenolic antioxidants. Under optimum conditions, the experimental values of TPC, TFC, % DPPH scavenging activity, and FRAP were 171.67 ± 4.69 mg GAE g-1, 26.87 ± 0.78 mg CE g-1, 81.31 ± 0.16%, and 1038.22 ± 7.69 μmol TE g-1, respectively. Results highlight that the developed eco-friendly method is appropriate for the improved recovery of phenolic antioxidants from C. salviifolius L.

In vitro and computational investigation of antioxidant and anticancer properties of Streptomyces coeruleofuscus SCJ extract on MDA-MB-468 triple-negative breast cancer cells

This study aimed to explore the antioxidant potential of the ethyl acetate extract of Streptomyces coeruleofuscus SCJ strain, along with its inhibitory effects on the triple-negative human breast carcinoma cell line (MDA-MB-468). The ethyl acetate extract’s total phenolic and flavonoid contents were quantified, and its antioxidant activity was investigated using DPPH (1,1-Diphenyl-2-picrylhydrazyl), ABTS (2,2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid), and FRAP (Ferric Reducing Antioxidant Power) assays. Furthermore, the cytotoxic effect of the organic extract from Streptomyces coeruleofuscus SCJ on MDA-MB-468 cancer cells was assessed via the crystal violet assay. In tandem, a thorough computational investigation was conducted to explore the pharmacokinetic properties of the identified components of the extract, utilizing the SwissADME and pKCSM web servers. Additionally, the molecular interactions between these components and Estrogen Receptor Beta, identified as a potential target, were probed through molecular docking studies. The results revealed that ethyl acetate extract of SCJ strain exhibited remarkable antioxidant activity, with 39.899 ± 1.56% and 35.798 ± 0.082% scavenging activities against DPPH and ABTS, respectively, at 1 mg/mL. The extract also displayed significant ferric reducing power, with a concentration of 1.087 ± 0.026 mg ascorbic acid equivalents per mg of dry extract. Furthermore, a strong positive correlation (p < 0.0001) between the antioxidant activity, the polyphenol and the flavonoid contents. Regarding anticancer activity, the SCJ strain extract demonstrated significant anticancer activity against TNBC MDA-MB-468 cancer cells, with an inhibition percentage of 62.76 ± 0.62%, 62.67 ± 0.93%, and 58.07 ± 4.82% at 25, 50, and 100 µg/mL of the extract, respectively. The HPLC-UV/vis analysis revealed nine phenolic compounds: gallic acid, sinapic acid, p-coumaric acid, cinnamic acid, trans-fereulic acid, syringic acid, chloroqenic acid, ellagic acid, epicatechin. Streptomyces coeruleofuscus SCJ showed promise for drug discovery, exhibiting antioxidant and anticancer effects.

Variations of Major Flavonoids, Nutritional Components, and Antioxidant Activities in Mung Beans (Vigna radiate L.) of Different Seed Weights

This study examined the levels of major flavonoids, nutritional components, total secondary metabolite contents, and antioxidant activities in 136 mung bean accessions and statistically analyzed the effect of seed weight difference on each. Vitexin and isovitexin were detected in all the mung bean accessions, with isovitexin being in a higher concentration regardless of seed weight difference. The contents of total protein and total starch were in the ranges of 22.01–28.96 and 32.62–49.03 g/100 g, respectively. Five fatty acids were detected by GC–FID analysis in all mung bean accessions, with linoleic acid being the most dominant (37.96–50.71 g/100 g). Total saponin content (TSC), total phenol content (TPC), DPPH• scavenging activity, ABTS•+ scavenging activity, and ferric reducing antioxidant power (FRAP) showed more than five-fold differences. Analysis of variance supported by multivariate analysis demonstrated that seed weight difference had a significant effect on total starch, all individual fatty acids except for stearic acid and oleic acid, TSC, and all antioxidant activities except for ABTS•+ scavenging activity. On the other hand, vitexin, isovitexin, total protein, total phenol, and total fatty acid contents remained unaffected by seed weight difference. Overall, this study showed the diversity of key flavonoids, nutritional components, total secondary metabolite contents, and antioxidant activities in mung bean genetic materials. Moreover, the study unveiled how seed weight affects the analyzed parameters in mung beans for the first time. These findings could maximize the use of mung beans in food industries and breeding programs as well as lead to more studies in metabolomics and genomics.

Antioxidant and Anti-Proliferative Effects of Ornithogalum balansae Extracts Collected in Black Sea Region Against Lung Cancer Cells

Ornithogalum is a genus of wild herb species widely used as food and in traditional medicine. This study investigated some of the biologically active properties of Ornithogalum balansae grown in the eastern Black Sea region of Türkiye. The investigations were carried out using the methanolic extracts of the plant’s aerial and bulbs (B1 and B2) parts. The phenolic composition was examined as total phenolic content (TPC), total flavonoid content (TFC), and HPLC-PDA. The antioxidant capacity of the extracts was tested based on ferric reducing/antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging methods. The anti-proliferative activity was tested against metastatic cell lines H1299 and H209, non-small-cell lung A549, and fibroblast MRC-5 cell lines using MTT and trypan blue methods. Wound-healing and invasion chamber assays were used to determine the inhibitory effects of the extracts on migration and invasion, respectively. The extract of the aerial part contained a large number of phenolic substances and high antioxidant capacity. The extract exhibited a significant anti-proliferative effect on the human lung cancer cells (A549 and H209), with IC50 values of 0.97 ± 0.04 and 1.06 ± 0.07 µg/mL, respectively. Moreover, the aerial part exhibited inhibition of migratory and invasive capacities in A549 cells at a concentration of 1.50 µg/mL. The findings associated with O. balansae suggest a promising therapeutic potential against lung cancer cells.

Immunomodulatory, Anticancer, and Antioxidative Activities of Bioactive Peptide Fractions from Enzymatically Hydrolyzed White Jellyfish (Lobonema smithii)

This study aimed to develop bioactive protein hydrolysates from low-value edible jellyfish obtained from local fisheries using enzymatic hydrolysis. Fresh white jellyfish were hydrolyzed using several commercial proteases, including alcalase (WJH-Al), flavourzyme (WJH-Fl), and papain (WJH-Pa). The antioxidant, immunomodulatory, and anticancer activities of these white jellyfish hydrolysates (WJH) were investigated. The results demonstrated that the crude WJH exhibited strong antioxidant properties, including DPPH, ABTS, and hydroxyl radical scavenging activities, as well as ferric-reducing antioxidant power. Additionally, the hydrolysates showed notable immunomodulatory activity. However, all WJH samples displayed relatively low ability to inhibit HepG2 cell proliferation at the tested concentrations. Among the hydrolysates, WJH-Pa demonstrated the highest antioxidant and immunomodulatory activities and was therefore selected for further bioactive peptide isolation and characterization. Ultrafiltration membranes with three molecular weight (MW) cut-offs (1, 3, 10 kDa) were used for peptide fractionation from WJH-Pa. Six potential peptides were identified with the MW range of 1049–1292 Da, comprising 9–12 residues, which exhibited strong antioxidant and immunomodulatory activities.

Antioxidant potential of olive leaf (&lt;i&gt;Olea europaea&lt;/i&gt; L.) sustainable extracts evaluated &lt;i&gt;in vitro&lt;/i&gt; and minced beef meat

In this work, by using water steam only, two antioxidant extracts were obtained from olive leaves (Olea europaea L.), a by-product of olive oil chain. Olive leaf extracts (OLEs) were tested as such (water extract: WE) and partially purified with ethyl acetate (ethyl acetate [EA] extract). Total phenols were 7.4 mg/mL and 3.8 mg/mL in WE and EA final solutions, respectively, evidencing a different composition by high-performance liquid chromatography analysis. Both extracts were evaluated in vitro in comparison to pure hydroxytyrosol (Hy). A 2,2-diphenyl-1-picrylhydrazyl (DPPH) EC50 of 57.6, 76.5, and 39.7 µg/mL and a ferric reducing antioxidant power EC50 of 84.8, 69.9, 41.2 µg/mL were determined for WE, EA, and Hy solution, respectively. The Rancimat induction time determined at 120°C in a lard sample with 200-ppm total phenol equivalent addition of WE, EA, and Hy was 8.92 h, 12.74 h, and 7.27 h, respectively (vs. 2.24 h for lard only). Extracts were added at the same dose (200 ppm) to minced beef patties that were put in closed containers under controlled air headspace composition (NA, natural air; HOA, modified air with 80% O2; and 20% N2) and stored at 4°C up to 10 days. Extracts showed significant effectiveness in contrasting decrease of O2 level in containers as well as pH and color changes of patties. A significant increase (expressed as mg of malondialdehyde MDA/kg; thiobarbituric acid-reactive substance (TBARS) assay was conducted in control samples at an interval of 0–10 days (from 0.52 to 0.78 mg of MDA/kg in NA samples; and from 0.55 to 1.31 mg of MDA/kg in HOA samples), while minimal changes were observed in treated samples. These findings suggest a potential use of OLEs for maintaining quality and oxidative stability of beef patties during storage.

Improvement of Hygienic and Phytochemical Qualities of Mangosteen (Garcinia Mangostana) Peel by Irradiation

The aim of this study was to compare the effects of gamma ray and x-ray irradiation at different dose levels 95, 10, 15 and 20 kGy) on in mangosteen peel. The antioxidant activities were assessed using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP), while total phenolic contents and antibacterial activities against Staphylococcus aureus and Escherichia coli of 60 % ethanolic mangosteen powder extracts were analyzed. The results indicated that both types of ionizing radiations showed a non-significant decrease in measured parameters with increasing doses. The minimum inhibitory concentration (MIC) values of irradiated mangosteen peel extracted against S. aureus and E. coli ranged from 0.125 to 2.0 mg/mL. The quantity of bioactive compound mangostin determined via high-performance liquid chromatography (HPLC), did not show significant decrease with different irradiation sources and doses. In microbiological aspect, total yeast and mold from both irradiation sources were reduced by 1 log cycle after irradiation at 5 kGy. The irradiation induced color changes corresponding to Hunter color L a b value. X-ray irradiation caused slight color alteration compared to gamma rays. The advantage of x-ray irradiation was that it revealed non-significant changes in b values, whereas gamma irradiation showed the significant change in L, and b values with increasing dosage. The x-ray irradiation at the dose 5 kGy effectively decreased microbial contamination while causing minimal changes to phytochemical qualities and color. HIGHLIGHTS Irradiation at 5 kGy was sufficient to control microbial contamination There were no significant changes in antioxidant activities (DPPH, FRAP), a-mangostin contents, total phenolic contents (TPC) and antibacterial activities: after irradiation up to 20 kGy X-ray irradiation was an excellent alternative to gamma irradiation for decontamination with a non-significant alteration in b values. Evaluating the nutrient composition and antioxidant properties of orange (Citrus sinensis) peels through Penicillium camemberti-based solid-substrate fermentation

Citrus by-products, especially orange peels, are a significant global waste issue, driving research into management solutions. Solid-substrate fermentation is a promising method to convert this waste into valuable resources, enhancing its medicinal and nutritional potential while addressing environmental and economic concerns in food waste management. Therefore, this study was carried out to evaluate the impact of solid-substrate fermentation (SSF) with Penicillium camemberti on the nutrient profile, antioxidative properties, and potential neuroprotective effects of orange peels in AlCl3 (Al)-induced neurotoxicity in fruit fly (Drosophila melanogaster) model. Orange peels were collected, air-dried to a constant weight, and subjected to a 10-day SSF process with Penicillium camemberti. Post-fermentation, in vitro nutritional profiling, and antioxidant assays were conducted, alongside in vivo assessments of the neuroprotective effects on survival rate, memory index, monoamine oxidase (MAO) activity, and endogenous antioxidant markers on Al-induced fruit flies were determined. The results showed that fermentation with Penicillium camemberti significantly enhanced the nutritional composition of the orange peels compared to their unfermented counterparts. The antioxidant properties of fermented orange peels were significantly (p < 0.05) increased, as evidenced by increased DPPH scavenging activity (81.68 ± 0.18) and ferric reducing antioxidant power (10.77 ± 0.27). Additionally, biochemical assays in vivo revealed a significant improvement in survival rates and memory indices in Al-induced flies. Also, monoamine oxidase (MAO) levels were significantly reduced, while glutathione S-transferase (GST) levels were significantly (p < 0.05) elevated in the induced flies fed with the fermented samples. Conclusively, this study highlights the capability of Penicillium camemberti fermentation in converting waste into valuable resources, enhancing the nutrient profile of citrus peels for applications in livestock feed and potential therapeutic interventions for degenerative disorders.

Antioxidant activities of Saudi honey samples related to their content of short peptides

This study explored the effect of geographical and floral origins on the antioxidant activities of Saudi honey samples related to their content of short peptides originated from honeybee proteins. The studied antioxidants were the total protein concentration, catalase activity, phenolic acids and flavonoids. The antioxidant activity assays included were the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, the ferric reducing antioxidant power (FRAP) assay and Ascorbic acid Equivalent Antioxidant Capacity (AEAC). The studied honey samples were obtained from the southwestern region of Saudi Arabia, namely Asir (65) and Jazan (25). The floral origins of the honey samples were Acacia (51), Ziziphus (4) and polyfloral (35). The LC/MS technique was used to detect the short peptides and the mascot database was used to identify the short peptides, their precursor proteins and the protease enzymes that produce them. Jazan honey was characterized by high number of short peptides. The short peptides were originated from honeybee proteins by the action of proteases from the honeybees and bacteria. The antioxidant activity of the honey samples increase with the increase of their content of short peptides and proteins. The amino acids type and sequence of the short peptides qualify them to act as antioxidant, antimicrobial, anti-diabetic, anti-hypertension, immunomodulatory and cholesterol lowering peptides.

Association of healthy eating index (HEI), alternative healthy eating index (AHEI) with antioxidant capacity of maternal breast milk and infant’s urine: a cross-sectional study

Maternal dietary quality may alter the nutrient content of breast milk. In this study, we aimed to investigate the relationship between the healthy eating index (HEI) and alternative healthy eating index (AHEI) of a breastfeeding mother’s diet with the antioxidant profile of her breast milk and her infant’s urine. This study included 300 healthy mother-infant pairs. The participants’ dietary intake was estimated using a validated semi-quantitative food frequency questionnaire. The diet quality of participants was assessed using the HEI and AHEI. The total antioxidant content of the breast milk and infant’s urine was evaluated using ferric reducing antioxidant power (FRAP), 2, 2′-diphenyl-1-picrylhydrazyl (DPPH), thiobarbituric acid reactive substances (TBARs), and Thiol quantification assays. After adjusting for confounding factors, the odds of a low malondialdehyde (MDA) content of breast milk were significantly higher in the highest quartile of HEI than in the lowest quartile. The odds of low DPPH and FRAP in infant urine decreased in the highest quartile of HEI compared to the lowest quartile. No significant relationship was found between AHEI and antioxidant levels of breast milk and the infant’s urine. Our findings demonstrate that a high quality diet of breastfeeding mothers, identified by a higher HEI, can affect the oxidant-antioxidant balance of a mother’s breast milk and her infant’s urine.

Exploring the Antimalarial Potential ofEntandrophragma utileandMelochia umbellataExtracts.

Resistance remains the fundamental problem with antiplasmodial treatments. The study investigated the antiplasmodial, cytotoxic, and antioxidant properties of two Cameroonian medicinal plants, Entandrophragma utile and Melochia umbellata. Antimalarial activity against Plasmodium falciparum strains 3D7 and Dd2 was assessed using the SYBR Green I assay. Cytotoxicity was assessed against Raw and Vero cells using the resazurin assay, and against red blood cells using a hemoglobin release quantification assay. Antioxidant potential was determined using DPPH (2, 2-diphenyl-1-picrylhydrazine), ABTS (2, 2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), and FRAP (ferric-reducing antioxidant power) assays. The most bioactive extract was further analyzed using ultra-high-performance liquid chromatography-mass spectrometry (UHPLC-MS) to determine its phytochemical composition. Extracts from E. utile exhibited moderate anti-malarial activity against Plasmodium falciparum (IC50 = 32.81-100 µg/mL), while M. umbellata leaf extract (MULAE) showed strong activity (IC50 = 11.62 and 11.91 µg/mL). All extracts demonstrated antioxidant activity (9.22 to 135.8 µg/mL), were selective for Raw and Vero cells, and were not toxic to red blood cells. UHPLC-MS analysis annotated potential pyrimidones, phenolic compounds, and terpenoids in MULAE. MULAE exhibited better antiplasmodial activity, suggesting the presence of unique bioactive compounds. Further research, including in vivo investigations, is needed to develop safe and effective antiplasmodial therapies.

Pharmacological Potential and Electrochemical Characteristics of Typha angustifolia Pollen

Typha angustifolia L. (TA) pollen has been utilized as a traditional Chinese medicine for treating various internal and external traumas. Moreover, bioactive compounds possess diverse pharmacological activities. This study aims to evaluate the antiviral properties of TA based on its ability to generate bioenergy, capable of inhibiting viruses. TA pollens were extracted using water and ethanol solvents. These extracts were utilized to identify the phytochemical contents and correlate with the antioxidant activity via 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. HPLC analysis was conducted to identify its electron-shuttling compositions. The bioenergy-generating characteristics were determined via microbial fuel cells. The water extract (TA-W) showed higher antioxidant activity due to a higher phenolic and flavonoid content compared to the ethanol extract (TA-E). Quercetin-3-O-(2G-α-L-rhamnosyl)-rutinoside, quercetin-3-O-neohesperidoside, and quercetin are the electron shuttles (ES) identified out of the 11 compounds. TA obtained a 1.39 ± 0.10 amplification factor of power generation that indicates potential bioenergy-generating and associated antiviral characteristic properties. The findings may provide a foundation for developing antiviral medications specifically designed to target virus-related diseases, while minimizing the risk of drug toxicity and reducing the costs of drug development.

Assessment of the Antioxidative Properties of Extracts from the Fruits of Pyrus pyraster (L.) Burgsd and Pyrus ×myloslavensis Czarna &amp; Antkowiak Grown under Natural Environmental Conditions

Analyses were conducted on extracts from the fruits of P. pyraster and P. ×myloslavensis. Extraction with 80% methanol was performed at room temperature. The total phenolic content was determined by spectrophotometry using the Folin–Ciocalteu reagent, with gallic acid as the reference standard. Phenolic compounds and organic acids were identified on a liquid chromatograph. The antioxidative activity of the extracts was tested in relation to linoleic acid incubation of the emulsions for 19 h based on the neutralization of the DPPH radical (2,2-diphenyl-1-picrylhydrazyl) and the ABTS cation radical (2,2′-azino-bis[3-ethylbenzothiazoline-6-sulfonic acid]) as well as by the ferric reducing antioxidant power (FRAP) assay. The analyses showed that the extract from P. pyraster fruits is characterized by a higher content of phenolic compounds and a higher antioxidative potential compared with that from P. ×myloslavensis. In extracts of both pear species, seven phenolic compounds and four organic acids were identified. The total fiber content in pears of P. pyraster and P. ×myloslavensis was determined at 36.45 g and 24.74 g/100 g d.m. of the pear fruits, of which most comprised the insoluble fraction (32.49 g and 20.86/100 g, respectively). The results of the conducted research are highly significant, as they confirm that pears contain many valuable nutrients and biologically active compounds, including antioxidants and dietary fiber. Adding pear extracts to food products may offer a way to boost their health benefits while also broadening the variety of items that have appealing sensory characteristics. Moreover, research has shown that fruit extracts can help to prolong the shelf life of food products by safeguarding them against lipid oxidation and the decline in their nutritional value.

Valorization of Pineapple Core Waste for Sequential Extraction of Phenolic Compounds and Carotenoids: Optimization Through Ultrasound-Assisted Method and Box–Behnken Design

AbstractIn this study, a pioneering cascade method involving ultrasound-assisted extraction (UAE) and Box–Behnken Design (BBD) was optimized to valorize pineapple core waste by the sequential extraction of, firstly phenolic compounds and, secondly, carotenoids. The effectiveness of the extraction was evaluated based on total polyphenol content (TPC) and antioxidant activity using 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferric reducing antioxidant power (FRAP) assays. Characterization of the carotenoids was performed using high-performance liquid chromatography with a diode array detector (HPLC–DAD). The initial characterization of dry pineapple core (DPC) samples revealed their nutritional composition, including protein, lipid, and carbohydrate weight percentages of 1.20 ± 0.05%, 5.3 ± 0.4%, and 88.6 ± 0.5%, respectively. The high extractives content (40.0 ± 4.5%) suggests a substantial presence of phenolic compounds, making the pineapple core a valuable source of natural antioxidants. The optimal UAE conditions for phenolic compound extraction were 70% amplitude, 5 min extraction time, and 2 cycles, yielding an antioxidant extract rich in phenolic compounds with a desirability value of 81.2%. Therefore, DPC was considered a valuable source of natural antioxidants. The extraction of β-carotene also showed promising results with optimal UAE conditions of 20% amplitude, 3 min extraction time, and 2 cycles. This research promotes the sustainable use of pineapple waste and demonstrates the potential to obtain valuable additives for the food, pharmaceutical, and cosmetic industries, encouraging a more circular and efficient use of resources in the pineapple processing industry.

Total dietary antioxidant capacity and food groups and their relationship with the sleep time of Brazilian graduates (CUME Study)

ABSTRACT To investigate the association between the Dietary Total Antioxidant Capacity (dTAC) and the Total Antioxidant Capacity of food groups (fgTAC) with the sleep time of Brazilian graduates participating in the Cohort of Universities of Minas Gerais (CUME Study). This cross-sectional study analyzed 6,387 graduates (2,052 men, 4,335 women, 35.3 ± 9.3 years old) from the CUME Study. Data was collected online, and dTAC was obtained by the Ferric Reduction Antioxidant Power (FRAP) method. Daily sleep time was classified as short sleep, normal sleep, and long sleep (≤6, 7–8, and ≥9 h, respectively). Multinomial logistic regression models were used to estimate the Odds Ratio (OR) and its 95% Confidence Interval (95% CI) between short sleep and long sleep with quartiles of dTAC and the fgTAC. Lower odds of short sleep was observed for the third quartile of dTAC and for fourth quartile of fgTAC of fruits, beans, and lentils, and for the third quartile of fgTAC of vegetables and oils and fats. Higher odds of short sleep for the fourth quartile of fgTAC of teas and coffees. For long sleep, inverse associations were observed for the fourth quartile of fgTAC of oilseeds and the third quartile of fgTAC of teas and coffees. Higher odds of long sleep were observed for the third quartile of artificial juices and sodas. We cannot independently assert an association between higher dTAC and sleep time. In turn, the associations between sleep time and fgTAC show the importance of the food matrix that antioxidants are inserted, requiring longitudinal studies to observe the direction of associations.

Clean label extraction of bioactive compounds from Chenopodium album and their role in the characterization and stability of ostrich meat

The demand for clean-label products continues to rise, as consumers increasingly prioritize natural and transparent ingredient lists. Natural substances are generally deemed safe for consumption by consumers. This study was focused on the development of clean-label ostrich meat patties with Chenopodium album extract and their storage stability. To prepare C. album extract, microwave-assisted extraction (MAE) and ultrasound-assisted extraction (UAE) were employed. Ostrich meat patties were prepared using diverse combinations of extract, including 1% UAE, 2% UAE, 1% MAE, 2% MAE, 0.5% UAE+0.5% MAE, and 1% UAE + 1% MAE. The highest pH was observed for MAE in T3 on the 14th day (6.19 ± 0.03). The L* value was observed between 39.12 ± 1.09 and 44.00 ± 1.1. As storage intervals passed, the a* and b* values of ostrich meat patties decreased. After the 14th day of storage, the best results were obtained from 2% UAE (T2), with the lowest TBRAS, Peroxide value (POV), and Total of volatile basic nitrogen (TVBN) readings recorded for T2 (0.74 ± 0.02 MDA/kg, 0.56 ± 0.01 meq peroxide/kg, and 6.28 ± 0.40 mg/100 mL, respectively). At the end of the storage study, the lowest Total Microbial Count (TMC) and coliform count were recorded for T2 (8.08 ± 0.03 and 4.97 ± 0.05 cfu/mL, respectively). At the end of the storage study, T2 exhibited the highest values for total phenolic content (TPC), diphenyl-1-picrylhydrazyl (DPPH), Ferric reducing antioxidant power (FRAP), and azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay, namely of T2 122 ± 0.28 mg GAE/100 g, 73 ± 0.27%, 5.9 ± 0.01 mmol FSE/100 g, and 83 ± 0.08 mmol/L, respectively. The current study concludes that the UAE extract of C. album incorporated into ostrich meat patties manifests improved safety, quality, and storage stability. The implementation of clean label strategies can facilitate food manufacturers to align with consumer preferences for product transparency and sustainability while ensuring product safety and quality.

12349 Increased Total Oxidative Status/Total AntioxidantCapacityandReduced Microparticle Size In Diabetes Kidney Disease

Diabetic kidney disease (DKD) is an important cause of morbimortality in the adult population with Type 1 Diabetes Mellitus (T1DM). Our cross-sectional study explored the association of abnormalities in extracellular vehicles (EVs) and oxidative stress markers in three distinct groups of clinical outpatients: normal controls, T1DM (without DKD), and DKD. Clinical and biochemical data were obtained from these groups. The isolation and characterization of EVs from serum were obtained through a series of processes, including the ultracentrifugation and characterization of EVs was identified by specific protein markers by western blot morphology checked by Scanning Electron Microscope (FEI Tecnai 20; LAB6 emission; 200 kV). EVs particle size and concentration were determined by NanoSight NS300 instrument (Malvern Panalytical, UK). Total Oxidant Status (TOS), Total Antioxidant Capacity (TAC), Advanced Oxidation of Protein Products (AOPP), (Ferric Reducing Antioxidant Power), C reactive Protein, Uric Acid, Aloxan levels were determined in the serum or the plasma.Overall, 124 patients were evaluated: 61 normal controls, 42 T1DM (without DKD), and 21 DKD defined according the KDIGO guidelines. The three groups were similar regarding age (mean± SD) of 40.3 ± 13.7 years, BMI 25.8 ± 5.1 Kg/m2, and Abdominal Circumference 89.6 ± 12.3 cm. The albumin/creatinine ratio was statistically different among the groups, being its (median [95%CI]) of 4.0 [3.9-5.14] mg/g in Controls, 6.4[5.5-10.0]mg/g in T1DM, and 81.4 [48.5 -1415] mg/g in DKD (p&amp;lt;0.001). The DKD patients exhibited higher SBP and an increased proportion of retinopathy and neuropathy than T1DM, but HBa1c levels were similar between the two diabetes groups. Patients with DKD show an increased TOS/TAC Ratio ( μmol H2O2 Equiv/L /mmol Trolox Equiv/L) (ANOVA p&amp;lt;0.001) superior to T1DM and controls. The mean ± SD was 61.1 ± 31.0 in Controls, 103.0± 68 in T1DM, and 145.0± 62.8 in DKD. Interestingly, T1DM exhibited an increased TOS/TAC Ratio than Controls. No modifications in AOPP or FRAP could be observed among the three groups.There is a gradual decrease in EVs size (but not concentration) in patients with DKD in comparison to T1DM and controls, and T1DM with controls. The mean of the moda size of EVs was (mean + SE) 144.5 ± 3.04 nm in Controls versus 136.14 ± 3.5 nm in T1DM, and 135.4 ± 3.6nm in DKD (ANOVA p=0.04). The concentration was (mean ± SE) 4.57 x109 ± 5.57 x108 particles/mL in Controls, 3.58 x109 ± 2.9 x108 particles/mL in T1DM and 3.94 x109 ± 3.7 x108 particles/mL in DKD (NS).Our results suggest an association of increased oxidative stress status (TOS/TAC ratio) and reduced microvesicles size in the presence of T1DM; both abnormalities progress towards the development of DKD. Biogenic Silver Nanoparticles (AgNPs) Ameliorates Oxidative Biomarkers in Type-2 Diabetic Rats: In vitro and In vivo Report

Objective: Nanoparticles (NPs) are reliable biological tools for curative purposes through their application in nanomedicine. The present study synthesized and characterized silver nanoparticles (AgNPs) from Tetrapetra tetrapleura fruit. The investigation aims to examine the antidiabetic effect of the AgNPs using in vitro and in vivo models. Methods: Briefly, the synthesized AgNPs were confirmed by the application of ultraviolet-visible (UV-Vis) spectroscopy, and five other techniques, viz; transmission electron microscopy (TEM) techniques, Fourier transform infrared (FTIR) spectroscopy, energy dispersive X-ray spectroscopy (EDX), X-ray diffraction analysis (XRD) and scanning electron microscope (SEM). The in vitro model assay investigated the scavenging effect of AgNPS on 2,2-diphenyl-1-picrylhydrazyl (DPPH), superoxide anion (O2ˉ), hydroxyl anion (-OH), ferric reducing antioxidant power (FRAP), and α-amylase/α-glucosidase inhibitory activity. The in vivo model involving rats-induced type-2 diabetes with streptozotocin (STZ) was divided into six (6) groups of seven (7) rats each to assess antioxidative parameters. Results: The AgNPs scavenged free radicals (DPPH) and moderately inhibited (O2ˉ), hydroxyl anion (-OH), reduced ferric to ferrous ions, and inhibited both α-amylase and α-glucosidase activity with increasing concentrations. Similarly, AgNPs ameliorated oxidative stress imposed by type 2 diabetes on the rats’ tissues significantly (p &lt; 0.05), depleting total cholesterol, low-density lipoprotein (LDL), and increased total protein composite and high-density lipoprotein (HDL) contents. The AgNPs enhanced catalase and superoxide dismutase, reduced glutathione (GSH), and, concomitantly, decreased malondialdehyde (MDA) levels in the tissue homogenate. Conclusion: These findings provide scientific evidence for the first time, finding the application of a biogenic compound synthesized from T. tetrapleura fruit in the treatment of type 2 diabetes.

Spent coffee grounds as an alternative fertilizer: impact on bioaccessibility of antioxidants and commercial quality of lettuce.

During the processing of spent coffee grounds (SCGs) several residues are obtained, which are mostly disposed of in landfills. There is an urgent need for a comprehensive waste management strategy for these residues. This study evaluates the potential of SCGs as a biofertilizer by assessing their effects on lettuce leaves and the release of antioxidants following in vitro digestion and fermentation. Lettuce plants were grown with different amounts of SCGs (0-150 g kg-1) in the substrate. High SCG concentrations in the soil generated lighter colored tissues, a decrease in the green color, less root development, and lower dry weight of leaves (P < 0.05). The SCG levels also affected the release of antioxidants by the final product. This effect was more pronounced in the digested fraction: applying the Ferric Reducing Antioxidant Power (FRAP) method, the addition of SCGs from 10 g kg-1 to 125 g kg-1 increased the amount of antioxidant from 43.88 ± 4.81 to 105.96 ± 29.09 μmol Trolox g-1 of dry weight (P < 0.05). The Indigo Carmine Reducing Capacity (ICRED) method also showed a similar trend, but in this case the highest value was obtained with 150 g kg-1 of SCGs (16.41 ± 3.93 mmol catechin g-1 of dry weight) (P < 0.05). Moreover, in the fermented fraction a significant increase in the antioxidant released was found with low levels of SCG(P<0.05), while lettuces fertilized with intermediate amounts of SCGs (25 and 50 g kg-1) presented the highest amount of insoluble antioxidant (P < 0.05). A compromise should be found in order to achieve a product with a high antioxidant capacity and an acceptable visual quality. © 2024 Society of Chemical Industry.

Effect of Different Drying Methods on Colour, Total Phenolic Content, Flavonoid Content, and Antioxidant Activity Retention of Strobilanthes crispus Leaves

Strobilanthes crispus, a medicinal herb, is recognised for its abundant phytochemicals, notably in its leaves, contributing to its high antioxidant activity. However, the crucial step of drying, aimed at extending shelf life, can impact the stability of these bioactive compounds. This study evaluates the impact of different drying methods, which include oven, microwave, freeze drying, and air drying, on the colour, phenolic and flavonoid content, and antioxidant activities of S. crispus leaves. The colour analysis of the fresh and dried leaves was assessed using the chromameter. Total phenolic content (TPC) and total flavonoid content (TFC) were determined using Folin-Ciocalteu’s and aluminium chloride colourimetric assays, respectively. Antioxidant capacities were analysed via ferric-reducing antioxidant power (FRAP) and a 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. The results showed that microwave-dried S. crispus leaves exhibited minimal alterations in colour attributes L*, a*, and b*, closely resembling the fresh leaves (p &gt; 0.05). Microwave drying significantly preserved TPC (145.42 ± 1.61 mg GAE/g), TFC (117.27 ± 5.10 mg QE/g), FRAP activity (258.92 ± 0.15 µg TE/g extract), and displayed the most potent DPPH scavenging half-maximal inhibitory concentration (7.58 ± 0.48 µg/ml) compared to other methods (p &lt; 0.05). Notably, the DPPH scavenging potency surpassed that of the synthetic antioxidant butylated hydroxytoluene. In conclusion, microwave drying appeared to be an efficient method for preserving the colour and antioxidant properties of S. crispus leaves. It highlights its potential as a favourable drying technique for conserving bioactive compounds in medicinal plant materials, offering promising applications in the nutraceutical and pharmaceutical fields.
