Association between dietary antioxidant capacity and type 2 diabetes mellitus in Chinese adults: a population-based cross-sectional study

BackgroundHigher intakes of dietary antioxidants have been linked to a lower type 2 diabetes mellitus (T2DM) risk. However, few studies have comprehensively examined the overall dietary antioxidant capacity, assessed by dietary antioxidant quality scores (DAQS) and dietary total antioxidant capacity (DTAC), related to T2DM risk, especially in populations consuming relatively monotonous diets. This study aimed to evaluate the associations of DAQS, DTAC, and T2DM among rural Chinese adults.MethodsData from 12,467 participants from the Natural Population Cohort of Northwest China: Ningxia Project was analyzed. Dietary intake was assessed using a validated semi-quantitative food frequency questionnaire. DAQS were calculated based on vitamins A, C, and E, zinc (Zn), and selenium (Se) intake. DTAC was estimated using the ferric-reducing ability of plasma assay. Logistic regression models were used to evaluate the associations of DAQS and DTAC with T2DM risk. Restricted cubic splines were used to assess potential non-linear relationships between DTAC and T2DM.ResultsT2DM was observed in 1,238 (9.9%) participants. After adjusting for confounders, compared to the lowest tertiles (T1) of DAQS, the odds ratios (ORs) for T2DM were 1.03 (95% CI 0.82–1.30) in T2 and 0.85 (95% CI 0.68–1.06) in T3 (P = 0.010). Compared to T1, the ORs for T2DM in the highest T3 were 0.78 (95% CI 0.67–0.91, P-trend = 0.008) for vitamin A, 1.34 (95% CI 1.15–1.56, P-trend < 0.001) for vitamin E, 0.83 (95% CI 0.71–0.97, P-trend = 0.007) for Se, and 0.86 (95% CI 0.74–1.01, P-trend = 0.033) for Zn. Compared to the lowest quartile(Q1) of DTAC, the OR in the highest Q4 was 0.96 (95% CI 0.80–1.17, P-trend = 0.024) for T2DM. A non-linear relationship was observed between DATC and T2DM.ConclusionHigher DAQS and DATC were associated with a lower T2DM risk, suggesting that consuming antioxidant-rich foods may reduce the T2DM risk.

High dietary salt intake attenuates nitric oxide mediated endothelium-dependent vasodilation and increases oxidative stress in pregnancy.

This study aimed to investigate the impact of dietary salt intake during normal pregnancy on maternal microvascular and macrovascular endothelium-dependent reactivity and oxidative stress level. In this cross-sectional study, based on their 24-h urinary sodium excretion, pregnant women (37-40 weeks of gestation) were divided into three groups: normal salt (<5.75 g/day, N = 12), high salt (5.75-10.25 g/day, N = 36), and very high salt (VHS;>10.25 g/day, N = 17). Forearm skin microvascular reactivity in response to vascular occlusion, local heating (LTH) and iontophoresis of acetylcholine (AChID), as well as brachial artery flow mediated dilation (FMD) were measured. Serum nitric oxide, endocan, 8-iso-prostaglandin F2α (8-iso-PGF2α), thiobarbituric acid reactive substances (TBARS), and ferric-reducing ability of plasma assay were measured as biomarkers of endothelial function/activation and oxidative stress. Brachial artery FMD, microvascular AChID, and LTH were significantly decreased in VHS compared with NS group, while LTH was also decreased in normal salt compared with high salt group. Nitric oxide was significantly decreased in both high salt and VHS groups compared with normal salt. Endocan, 8-iso-PGF2α, and TBARS were significantly increased in VHS compared with the normal salt group. High dietary salt intake is associated with decreased nitric oxide mediated endothelium-dependent vasodilation in peripheral microcirculation and macrocirculation of healthy pregnant women due to increased oxidative stress.

The Effect of Lactiplantibacillus plantarum I-Enriched Diet on the Phenolic Content and Antioxidant Capacity of Queen Scallop (Aequipecten opercularis Linnaeus, 1758) Extracts.

The use of probiotics in the diet of bivalves poses a great potential in aquaculture as an alternative to antibiotics. The aim of this study was to assess the effect of Lactiplantibacillus plantarum I on the phenolic content and antioxidant capacity (AC) of queen scallop extracts after one month of feeding. Total phenols (TP) ranged from 28.17 ± 3.11 to 58.58 ± 8.57 mg GAE/100 g, total non-flavonoids (TNF) from 23.33 ± 3.66 to 36.56 ± 9.91 mg GAE/100 g, and total flavonoids (TF) from 10.56 ± 5.57 to 30.16 ± 1.69 mg CE/100 g. AC was assessed via three different methods: the ferric-reducing ability of plasma assay (FRAP), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic) acid assay (ABTS), and 2,2-diphenyl-1-picryhydrazyl assay (DPPH). FRAP values ranged from 0.13 ± 0.03 to 0.17 ± 0.02 µM AA/g, ABTS from 0.68 ± 0.11 to 2.79 ± 0.34 µM AA/g, and DPPH from 1.75 ± 0.17 to 2.98 ± 0.53 µM AA/g. Among all extracts, the best phenolic content and AC were observed in water extracts from queen scallops. The bivalves treated with the Lactiplantibacillus plantarum I-enriched diet showed higher AC according to the FRAP assay in all extracts. A significant correlation was observed between AC and TP and TNF in control and Lactiplantibacillus plantarum I-treated scallops.

Seasonal Variability of Juniperus communis L. Berry Ethanol Extracts: 2. In Vitro Ferric Reducing Ability of Plasma (FRAP) Assay.

In the present study, the seasonal variability of the in vitro ferric reducing ability of plasma (FRAP), total phenols, and terpene hydrocarbon content in 70% ethanol extracts were evaluated. The samples of crushed (CBs) and non-crushed ripe juniper berries (NCBs) collected at five localities in North-East Slovakia during the years 2012-2014 were compared. The method of preparation of the extract influenced the amount of dry matter (DM) in the extracts. In the CB extracts were statistically higher contents of DM (from 13.91 ± 0.11 g·L-1 to 23.84 ± 0.14 g·L-1) compared to NCB extracts (from 1.39 ± 0.01 g·L-1 to 16.55 ± 0.09 g·L-1). The differences in antioxidant activity between the investigated localities were statistically significant for both types of extract. For example, in 2013 in the locality of Zbojné, the FRAP in NCBs was 76.62 µmol·L-1·g-1 DM and in CBs was 138.27 µmol·L-1·g-1 DM, while in the Miľpoš locality, in NCBs there was 232.66 µmol·L-1·g-1 DM and in CBs there was 1178.98 µmol·L-1·g-1 DM. The differences in the antioxidant activity between the studied years in the case of NCB extracts were not statistically significant. In the case of CB extracts, significant differences between the evaluated years were found. Statistics by ANOVA confirmed that CB extracts prepared from berries in the year 2013 showed significantly higher activity compared to CB extracts from berries from the years 2012 and 2014. Based on the Pearson we found a negative correlation coefficient between the FRAP assay and the content of total polyphenols in NCB extracts (-0.531 in 2012; -0.349 in 2013; and -0.224 in 2014). In contrast, CB extracts showed a positive correlation coefficient (0.843 in 2012; 0.742 in 2013; 0.617 in 2014).

Possibility to use the different sizes and colors of carrots for the production of juices - comparison of bioactive compounds, nutritional quality, pro-health properties, and sensory evaluation.

Carrot is a popular vegetable consumed by people of all age groups and is used in various food products because of its high nutritional content, especially vitamin A. In the present study, colorful fresh carrot juices of 12 carrot varieties were investigated for in vitro antidiabetic, anti-aging, and anti-obesity activities with antioxidant potential by ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and FRAP (ferric reducing ability of plasma) assays. The studied juices were also compared for physicochemical characteristics: titratable acidity, pH, pectin content, total soluble solids, dry mass, ash, viscosity, turbidity, osmolality, and color. The results of the study showed that normal purple carrot juice exhibited the best activities in all biological and antioxidant tests, except for anti-α-glucosidase activity. Normal purple carrot juice also had the highest total mineral content with elevated results for titratable acidity, pH, total soluble solids, dry mass, ash, viscosity, and osmolality. Purple carrot juices demonstrated elevated health-promoting activities and could be used in blended beverage recipes to attract children's attention. The results of sensorial characteristics (appearance, color, and taste) of juices, however, showed that people are more familiar with orange carrot products. © 2022 Society of Chemical Industry.

A Study of Karabaghian Skullcap (Scutellaria platystegia Juz.): Antioxidant and Antibacterial Activity Assays, Essential Oil Analysis, and Isolation of Its Phenolic Compounds

Background: Scutellaria platystegia Juz. is an herbaceous perennial, distributed in the northwest of Iran and southern Caucasian countries? Objectives: This study aimed to examine the antioxidant and antibacterial activities of the aerial part of S. platystegia and to determine the phytochemical constituents of its extract and essential oil. Methods: The ferric reducing ability of plasma (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods were used to examine the antioxidant effects of fractions obtained from the hydroalcoholic extract of the aerial part of S. platystegia. The antibacterial activity screening was also performed using the disk diffusion and microdilution methods. The phytochemical constituents of the most active fractions were investigated using column chromatography. Nuclear magnetic resonance (NMR) imaging and UV-Vis spectral analysis were used to determine the chemical structure of the isolated compounds. The GC-MS technique was also employed to identify the essential oil composition of the plant. Results: In the antioxidant activity assay, the n-butanol fraction was the most potent fraction, with a half maximal inhibitory concentration (IC50) of 16.14 ± 0.8 µg/mL on the DPPH assay and 736.4 ± 4.6 mmol FeSO4.7H2O equivalent per gram of sample on the FRAP assay. The n-butanol fraction exhibited very strong antibacterial activities against Staphylococcus aureus, Staphylococcus epidermidis, and Shigella dysenteriae (inhibition zone diameter: 20 - 25 mm, MIC: 125 - 250 µg/mL). The phytochemical analysis of the most active fractions resulted in the isolation of the following components from the chloroform fraction: 5,7,2’-trihydroxy-8,6’-dimethoxyflavone; 5-hydroxy-6,7,8,2’,6’-pentamethoxyflavone; 5,2’,6’-trihydroxy-7,8-dimethoxyflavone; 5,2’,6’-trihydroxy-6,7,8-trimethoxyflavone; and 5,4’-dihydroxy-7-methoxyflavone. Besides, the following components were extracted from the n-butanol fraction: luteolin-7-O-β-D-glucopyranoside; verbascoside; apigenin; kaempferol; caffeic acid; rosmarinic acid; apigenin-7-O-β-D-glucopyranoside; apigenin-7-O-β-D-(-6’’-(E)-caffeoyl)-glucopyranoside; and luteolin. Fourteen compounds were also identified in the plant essential oil; terpinen-4-ol (44.41%), α-terpineol (10.75%), caryophyllene oxide (9.61%), and thymol (8.73%) were the main compounds. Conclusions: This study introduced S. platystegia as a plant rich in bioactive phenolic compounds, with considerable antioxidant and antimicrobial activities. Therefore, it can be suggested as a potential candidate to be evaluated in future biological studies.

A comparative study on the effect of blood collection tubes on stress oxidative markers.

Oxidative stress has a major role in disease pathogenesis. However, limited studies have investigated the effect of various sample collection tubes on oxidative biomarkers. The present study aimed to evaluate the effect of different collection tubes on the variation of malondialdehyde (MDA), nitric oxide (NO), total thiol (t-SH), and ferric reducing ability of plasma (FRAP) levels. A total of 35 individuals participated in this study and each collected sample was separated into three different tubes: glass tubes (GTs), plain plastic tubes (PTs), and gel separator tubes (GSTs). The results of PTs and GSTs were compared to those of GTs as the reference tube. The comparison between the means of biomarkers in various tubes indicated that there was no significant difference in MDA results between tubes. In contrast, t-SH and NO content were significantly decreased in GSTs and PTs compared to GTs. However, the Bland-Altman analysis showed an acceptable concordance for the mentioned analytes and the statistically significant differences were not clinically significant for NO, MDA, and t-SH antioxidant parameters. Moreover, the FRAP level was considerably lower in GSTs compared to GTs. Nevertheless, the Bland-Altman analysis showed a high bias percentage for the FRAP assay when using PTs and GSTs. According to the present results, it can be concluded that switching to plastic blood collection tubes or serum separation tubes could influence the FRAP results. However, there was no interference for the interpretation of other antioxidant assays in different types of collection tubes. Hence, it is suggested to use GTs for total antioxidant capacity evaluations, especially the FRAP assay.

New Mutant Amaranth Varieties as a Potential Source of Biologically Active Substances

Amaranth species represent a diverse group of plants. Many of them are a rich source of secondary metabolites with many positive biological effects. Total phenolic, total flavonoid and rutin content, antioxidant activity against superoxide and hydroxyl radicals, FRAP (Ferric-reducing ability of plasma) assay and DPPH (2,2-Diphenyl-1-picrylhydrazyl) radical scavenging assay were determined in ethanol extracts of dried leaves of the new Slovak amaranth varieties ‘Pribina’ and ‘Zobor’. The amount of total phenolic substances (‘Pribina’ GAE 38.3 mg.g−1 DM and ‘Zobor’ GAE 26.1 mg.g−1 DM), content of total flavonoids (‘Pribina’ QE 26.5 mg.g−1 DM and ‘Zobor’ QE 20.3 mg.g−1 DM) and rutin (‘Pribina’ 50.8 mg.g−1 DM and ‘Zobor’ 15.2 mg.g−1 DM) were higher in the variety ‘Pribina’, compared to the variety ‘Zobor’. A statistically higher antioxidant activity against superoxide radical (1.63%·mg−1g−1 DM), hydroxyl radical (3.20%.mg−1g−1 DM), FRAP assay (292.80 µmol.L−1·mg−1.g−1 DM) and DPPH (54.2 ± 1.78 µg.mL−1 DM) were detected in the ‘Pribina’ variety. Antiradical and antioxidant activities of both extracts showed high positive correlations in relation to the content of total phenolic substances, total flavonoids and rutin. Amaranth is an undemanding crop on specific environmental conditions and is resistant to abiotic and biotic stress.

Highland Barley and Its By-Products Enriched with Phenolic Compounds for Inhibition of Pyrraline Formation by Scavenging α-Dicarbonyl Compounds.

Pyrraline, a typical kind of advanced glycation end product, has been found to contribute to the development of pathologies associated with ageing and diabetes mellitus. In the study, phenolic compounds extracted from highland barley whole grain (HBWG) and vinasse (HBVN) were used to inhibit pyrraline formation in a simulated food. The optimal extraction condition for HBWG and HBVN was using 8 mL of 50% acetone solution at 50 °C for 60 min. The extraction and identification of phenolic compounds from HBWG and HBVN were performed by UPLC–PAD–MS/MS. The inhibitory effects of pyrraline in the simulated food were 52.03% and 49.22% by HBVN and HBWG, respectively. The diphenyl picrylhydrazyl radical- and ferric-reducing ability of plasma assays was used to evaluate the antioxidant activity of the extracts. The main inhibition pathways and molecular mechanism of phenolic compounds on pyrraline regulation were explored by scavenging α-dicarbonyl compounds. The study demonstrated that highland barley and its by-products can potentially be used as a functional food to regulate pyrraline formation during food processing.

High taurine concentrations negatively effect stallion spermatozoa parameters in vitro

The target of present study was to evaluate the effect of various taurine concentrations on stallion spermatozoa during 37°C cultivation. Fresh semen was collected from 10 breeding stallions. The experimental groups were supplemented with six different concentration of taurine (in mg/ml): A – 2.5, B – 5, C – 7.5, D – 10, E – 15, F – 20 and compared to control (CON – 0). Spermatozoa motility was assessed using the Computer Assisted Semen Analyzer (CASA) system in 6 time periods (0, 1, 2, 3, 4 and 5 hours). The MTT test was used for detection of viability. For measuring antioxidant activity FRAP and TOS methods were used. Significantly negative effect was observed in the samples with the highest concentration of taurine (20 mg/ml). Spermatozoa viability was not significantly affected in analysed concentrations of taurine. Significant higher antioxidant activity was detected in the sample with the highest taurine concentration. Phytochemical and Biological Activity Studies on Nasturtium officinale (Watercress) Microshoot Cultures Grown in RITA® Temporary Immersion Systems.

The main compounds in both extracts were gluconasturtiin, 4-methoxyglucobrassicin and rutoside, the amounts of which were, respectively, determined as 182.93, 58.86 and 23.24 mg/100 g dry weight (DW) in biomass extracts and 640.94, 23.47 and 7.20 mg/100 g DW in plant herb extracts. The antioxidant potential of all the studied extracts evaluated using CUPRAC (CUPric Reducing Antioxidant Activity), FRAP (Ferric Reducing Ability of Plasma), and DPPH (1,1-diphenyl-2-picrylhydrazyl) assays was comparable. The anti-inflammatory activity of the extracts was tested based on the inhibition of 15-lipoxygenase, cyclooxygenase-1, cyclooxygenase-2 (COX-2), and phospholipase A2. The results demonstrate significantly higher inhibition of COX-2 for in vitro cultured biomass compared with the herb extracts (75.4 and 41.1%, respectively). Moreover, all the studied extracts showed almost similar antibacterial and antifungal potential. Based on these findings, and due to the fact that the growth of in vitro microshoots is independent of environmental conditions and unaffected by environmental pollution, we propose that biomass that can be rapidly grown in RITA® bioreactors can serve as an alternative source of bioactive compounds with valuable biological properties.

Alteration of Apoptotic Gene Expression and Reproductive Parameters through Thymus vulgaris Administration Following Paclitaxel-Induced Testopathy in Mice

Objective: Paclitaxel (PAC) is a chemotherapy drug and has an active role in the treatment of lung, breast, and ovarian cancers; however, its use causes increased levels of free radicals, leading to severe and irreversible organ damage. Thymus vulgaris is a species of flowering plant that contains various antioxidant chemical compounds. This study aimed to investigate the effect of T. vulgaris on PAC-induced oxidative damage in mice testis and sperm parameters. Methods: In this study, 40 BALB/c mice were divided into five groups: normal control (NC); positive control (20 mg/kg of PAC); and three treatment groups (4.5, 9, and 18 mg/kg doses of T. vulgaris extract + 20 mg/kg PAC). Treatments were administered intraperitoneally daily for 50 days. Finally, the levels of serum luteinizing hormone (LH), follicle-stimulating hormone (FSH), and testosterone were measured using immunoassay method. Testicular stereological features and sperm parameters were also calculated. Antioxidant parameters were measured using nitrite oxide (NO) and perioxidation levels and ferric-reducing ability of plasma assay. The expressions of p53, caspase-3, Bax, and Bcl-2 were measured through real-time quantitative polymerase chain reaction. Results: T. vulgaris + PAC treatments at all doses significantly increased all parameters in NC and three treatment groups compared with the positive control group (20 mg/kg of PAC) (P Conclusions: T. vulgaris administration attenuates the toxic effects of PAC on male reproductive parameters.

Characterization of European cranberrybush (Viburnum opulus L.) genetic resources in Turkey

In this study, a comprehensive morphological and biochemical characterization of 14 European Cranberrybush (Viburnum opulus L.) genotypes that were collected from Gumushane province of Turkey is provided. Additionally, their potentials as cultivar candidate, natural antioxidants, colorants and ingredients were exploited. Yield per bush, fruit weight, number of fruits per raceme and number of racemes per bush were ranged from 4.7 kg to 6.2 kg; 0.53 to 0.86 g; 34.4–53.1 and 119–307 among genotypes indicating high diversity. The majority of genotypes (8) had red fruit skin color and rest of the genotypes had equally light red and dark red fruit skin color. European Cranberrybush fruits had medium level vitamin C content (28−43 mg per 100 g fresh fruit) and the genotypes had SSC (Soluble Solid Content) between 10.40 and 12.20 %. Crude protein and crude cellulose of the genotypes varied from 5.67 to 6.71 (%) and 16.6–18.7 (%), respectively. Total phenol and total anthocyanin content ranged from 696 to 830 mg gallic acid equivalent per 100 g fresh weight and 27.6–54.3 mg per 100 g fresh fruit expressed cyanidin-3-glucoside. Antioxidant activity were found between 27.67 and 35.65 μmol trolox equivalents per g fresh fruit based on FRAP (Ferric Reducing Ability of Plasma) assay. Total lipid content ranged from 10.3–13.3 (%) and oleic acid was dominant fatty acids for all genotypes (57.3–70.4). Local genotypes are found to be promising for further valorization through horticultural characteristics and novel antioxidants, colorants and ingredients.

L. leaf extract has been studied validating its potential application.MethodsThe plant phenolics in the leaf extracts obtained via cold-maceration with solvents viz.: n-hexane, chloroform, ethyl acetate, methanol, and water were quantitatively analyzed. The antioxidant potency was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Ferric Reducing Ability of Plasma (FRAP) assays at five concentrations (100–500 μg). The enzyme inhibition potential was performed with α-amylase, α-glucosidase, and acetylcholinesterase at seven concentrations (25–500 μg). The experiments were done in triplicates and statistically validated using Minitab-17 and SPSS 22.ResultsWater extract contain 45.08 ± 0.02 mg GAE/g, 49.30 ± 0.07 mg GAE/g, 211.21 ± 0.02 mg QE/g tannins, phenolics, flavonoids respectively. Its antioxidant activity was supported by IC50 52.43 ± 0.2 μg/mL (DPPH assay) and 41.58 ± 0.03 (FRAP assay). Methanolic extract inhibits α-amylase with IC50 1.33 ± 0.05 μg/mL. Water extract inhibits α-glucosidase and acetylcholinesterase with IC50 3.58 ± 0.02 μg/mL and 4.46 ± 0.03 μg/mL. Cytotoxicity studies with SH-SY5Y cell-line substantiate the inhibition potential of water extract with IC50 103.0035 μg/mL.Discussion and conclusionsThe extracts with potent antioxidant and enzyme-inhibiting activity were determined. The findings of the research are the first report about the inhibition effects of Evolvulus alsinoides (Linn.) Linn extracts against α-amylase, α-glucosidase and acetylcholinesterase. The extracts shall be examined in future studies to evaluate its pharmaceutical potential.

Diversity on Fruits of Wild Grown European Cranberrybush from Coruh Valley in Turkey

Wild berries-blackberry, raspberry, blueberry, strawberry, rose hip, elderberry, cranberries etc. are common across most of region of Turkey and there is lack of information about horticultural characteristics of wild grown those berries in the country. Present study aimed to determine some morphological (fruit weight, number of fruits per raceme, number of racemes per bush), and biochemical (vitamin C, soluble solid content, total phenol, total anthocyanin, and antioxidant capacity) in fruits of ten European cranberrybush (Viburnum opulus L.) genotypes naturally grown in Coruh valley in Turkey. The results showed statistically significant differences among genotypes for most of the searched parameters. Fruit weight, number of fruits per raceme and number of racemes per bush were ranged from 0.64 to 0.85 g; 35.2 to 52.6 and 326 to 483, respectively. The genotypes had SSC (Soluble solid content) and vitamin C between 10.96 and 12.65% and 29 and 41 mg per 100 g fresh fruit basis. Total phenol and total anthocyanin content ranged from 668 to 856 mg gallic acid equivalent per 100 g fresh weight, 27–53 mg per 100 g fresh fruit expressed cyanidin-3-rutinoside. Antioxidant activity were found between 28.76 and 36.41 µmol trolox equivalents per g fresh fruit based on FRAP (Ferric Reducing Ability of Plasma) assay.

Ellagic Acid Containing Nanostructured Lipid Carriers for Topical Application: A Preliminary Study.

Ellagic acid (EA) is a potent antioxidant substance of natural origin characterized by poor biopharmaceutical properties and low solubility in water that limit its use. The aim of the present study was to develop lipid-based nanoparticle formulations able to encapsulate EA for dermal delivery. The EA-loaded nanoparticles were prepared using two different lipid compositions, namely tristearin/tricaprylin (NLC-EA1) and tristearin/labrasol (NLC-EA2). The influence of formulations on size, entrapment efficiency, and stability of EA-loaded nanoparticles was investigated. Cryo-TEM and small-angle X-ray scattering (SAXS) analyses showed that no morphological differences are evident among all the types of loaded and unloaded nanostructured lipid carriers (NLCs). The macroscopic aspect of both NLC-EA1 and NLC-EA2 did not change with time. No difference in size was appreciable between empty and drug-containing NLC, thus the nanoparticle diameter was not affected by the presence of EA and in general no variations of the diameters occurred during this time. The entrapment efficiency of both EA-loaded nanoparticles was almost quantitative. In addition, NLC-EA1 maintained EA stability for almost two months, while NLC-EA2 up to 40 days. FRAP (Ferric reducing ability of plasma) assay showed an antioxidant activity around 60% for both the loaded NLC, as compared to the solution. Although both types of NLC are characterized by some toxicity on HaCaT cells, NLC-EA1 are less cytotoxic than NLC-EA2. Taken together these results demonstrated that the inclusion of EA within NLC could improve the water solubility, allowing for a reduction of the dosage. Moreover, both types of NLC-EA maintained a high antioxidant effect and low toxicity.

ANTIOXIDANT ACTIVITY TEST OF FRACTIONS FROM STAR FRUIT LEAVES (AVERRHOA CARAMBOLA L.) FROM THREE REGIONS IN WEST JAVA

Objectives: Previous in vitro research showed that ethyl acetate fractions of star fruit leaves exhibit strong antioxidant activities toward 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical (IC50 = 90 μg/ml). This study’s aim was to evaluate antioxidant activity of fractions from leaf extracts from star fruitsourced from three different regions in Indonesia (Depok, Sukabumi, and Subang) and, for the most active fraction (that with the highest antioxidantactivity), determine whether there was any correlation with phenolic and flavonoid content.Methods: Liquid–liquid partitioning, fractions were evaluated for in vitro antioxidant activity using DPPH radical scavenging and ferric reducingability of plasma (FRAP) assay.Results: The ethyl acetate fraction from the Subang region exhibited the strongest radical scavenging activity with both the DPPH assay (IC50 = 96 μg/ml)and the FRAP assay (FeEAC value = 1405 μmol/g).Conclusion: Ethyl acetate fractions from star fruit leaves could be used as natural antioxidants. No correlation was found between antioxidant activityand phenolic or flavonoid content.

SEASONAL VARIATIONS IN ANTIOXIDANT CAPACITIES AND PHENOLIC CONTENTS OF TEA LEAF EXTRACTS

Objectives: The objective of the study was to estimate the seasonal variations in the antioxidant capacities, total polyphenol content (TPC), total flavonoid content (TFC), and tannin content (TC) of tea leaf extracts from two different plantation sites.&#x0D; Methods: Samples were collected from two tea gardens in Tuli and Ungma situated at N 26°39’19.3 E 094°39’22.7 and N 26°17’30.6 E 094°28’29.2, respectively, under the Mokokchung district of Nagaland, India. TPC, TFC, and TC from sample extracts were determined using Folin–Ciocalteu reagent, aluminum chloride colorimetric, and Folin–Ciocalteu assay. Apart from these, antioxidant capacities were analyzed using ferric reducing ability of plasma (FRAP) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay.&#x0D; Results: The concentrations of total polyphenol, flavonoid, and tannin varied from 552.029±8.079 to 305.647±1.744 mg gallic acid equivalent/g, 238.770±0.508–148.457±1.653 mg catechin equivalent/g, and 26.453±0.485–20.173±0.173 mg tannic acid equivalent/g, respectively. FRAP and DPPH assay displayed value ranging from 2.564±0.023 to 1.074±0.023 mmol Fe(II) equivalent/g and 3.612±0.053–2.076±0.028 mmol Trolox equivalent/g. Significant seasonal variations in concentrations of these compounds were observed and a positive correlation between antioxidant capacities and phenolics of tea leaf extracts was established.&#x0D; Conclusion: Tea (Camellia sinensis (L.) O. Kuntze) has been regarded as a plant of immense medicinal and therapeutic value since time immemorial. The tea leaf extracts analyzed in this study gave high TPC, TFC, and TC, as well as high antioxidant activity in terms of DPPH and FRAP value. Studying such properties in tea leaves contributes more to our understandings of health benefit potentials in tea leaves and the quality of tea leaves on the basis of seasons and sites where they are planted.

Comparison of antioxidant levels and anti-inflammatory activities of kelulut honey harvested at different month of intervals and its chemical compositions

Background: Harvesting time might influence the biological activities of honey, which is crucial for honey quality. Objectives: This study aims to evaluate the optimum time for harvesting kelulut honey (KH) by determining its antioxidant and anti-inflammatory activities and chemical composition at different harvesting times. Materials and Methods: KH harvested at three different intervals (1, 2, and 3 months) was supplied by Rimbunan Hijau Bee Farm, located in Sarawak, Malaysia. The total phenolic content (TPC) and antioxidant level of KH were determined using Folin–Ciocalteu reagent and 2,2-diphenyl-1-picrylhydrazyl, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid), and ferric-reducing ability of plasma assays, respectively. The cytotoxicity and anti-inflammatory effects of KH were evaluated on murine macrophage cells (RAW 264.7) by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and lipopolysaccharide (LPS)-induced nitric oxide (NO) production. The chemical composition of KH was analyzed by gas chromatography-mass spectrometry (GC-MS). Results: TPC and antioxidant levels of KH collected at different intervals showed no statistically significant difference (P > 0.05). KH at 1% concentration showed no cytotoxic activity toward RAW 264.7 cells after 24 h incubation. However, 1% KH showed no inhibition on NO production in LPS-induced RAW cells compared to the control. GC-MS revealed that hydroxymethylfurfural was present with the highest concentration in all samples. Conclusion: This study suggests that harvesting time does not influence the biological activities of KH.
