Some loss factors and processing methods of dried fishes used in the Sahelian domain of Cameroon

Fish is an important source of protein, minerals and essential fatty acids for human nutrition. To ensure its permanent availability, this highly perishable commodity is processed and stored. However, during storage, dry fishes suffer significant quantitative and qualitative losses due to the lack of efficient pest control methods and preservative processes. An intensive interview survey was conducted with 175 dry fish sellers in markets and processors' families in the Logone basin from 2020 to 2021 to identify the causes of losses and the usual tools used to limit them. It was found that losses of dry fish start during handling and transport where inadequate packaging breaks the dry fish. In addition, fishes are attacked by insect pests, losses depend on fish species and climate. In the Logone basin, 14 species of fish are caught and processed, the most represented species are Cyprinus carpio Linnaeus 1758 with 24.49%, Clarias gariepinus Burchell 1822 with 21.26% and Brevimyrus niger Günther 1866 with 15.40%. Their major pests are insects (53%), rodents (28%) and fungi (9%). Moreover, during handling/transport, 10% are lost. The insects identified are from the family of Cleridae and Dermestidae. Cleridae predominate in the surveyed areas with 67.76% against 32.23% of Dermestidae. Four species are identified: Dermestes maculatus, Dermestes frischii, Dermestes ater and Necrobia rufipes. Moreover, to protect their fish against these insect pests, stakeholders use industrial chemicals, generally sold as plant protection products, and local products derived from biodiversity local. The most popular chemical pesticide applied on dried fish has dichlorvos as active ingredient (83%). The most used are DD FORCE (48.51%) and PRIME SHI (16.26%), at the level of sellers and processors. It is an extremely hazardous pesticide, damaging health of consumers and polluting the environment. It would be necessary to consider the use of bio insecticides for the protection of the dried fish against pests and for the health safety of consumers. Modular Metabolic Engineering of Mortierella alpina by the 2A Peptide Platform to Improve Arachidonic Acid Production.

Arachidonic acid (ARA) is an essential fatty acid in human nutrition. Mortierella alpina, a filamentous fungus, has been widely used for the production of ARA. Here, we report a modular engineering approach that systematically eliminates metabolic bottlenecks in the multigene elongase/desaturase pathway and has led to significant improvements of the ARA titer. The elongase/desaturase pathway in Mortierella alpina was recast into two modules, namely, push and pull modules, based on its function in the ARA synthesis. Combinatorial optimization of these two modules has balanced the production and consumption of intermediate metabolites. A 2A peptide-based facile assembly platform that can achieve multigene expression as a polycistron was first established. The platform was then applied to express the push and pull modules in Mortierella alpina. In the shake-flask fermentation, the lipid and ARA contents of the engineered strain MA5 were increased by 1.2-fold and 77.6%, respectively, resulting in about fivefold increase of the ARA yield. The final ARA titer reached 4.4 g L-1 in shake-flask fermentation. The modular engineering strategies presented in this study demonstrate a generalized approach for the engineering of cell factories in the production of valuable metabolites.

Quantitative analysis of fatty acids and vitamin E and total lipid profiling of dietary supplements from the German market

Certain polyunsaturated fatty acids with n-3 double bonds are essential nutrients for the human body and are part of the bilayer of cell membranes or precursors of tissue hormones. The most abundant dietary n-3 fatty acids in human nutrition are α-linolenic, eicosapentaenoic, and docosahexaenoic acid and can be taken up through dietary sources such as vegetable oils or fish or, alternatively, dietary supplements with high levels of n-3 fatty acids. In previous studies, considerable variation of lipid patterns and quantities of n-3 fatty acids were observed. In this study, 33 dietary supplements from the German market, based on fish-, krill-, microalgae, and plant oil, have been analyzed. Lipid profiling (LC–MS) revealed triacylglycerols as the dominant lipid species in most samples. However, krill oil was rich in phospholipids and samples containing fatty acid concentrates featured abundant fatty acid ethyl esters and diacylglycerols. Furthermore, total lipid profiles showed considerable variance depending on the lipid sources (e.g., fish or plant oil), which was also apparent in fatty acid analysis. The contents of n-3 fatty acids ranged between 150 and 570 mg/g capsule content (GC–MS) and vitamin E (α-tocopherol and tocopheryl acetate) were found in quantities ranging from 1.2 to 86.1 mg/g capsule content (HPLC–UV/Vis). While our analyses indicated a good agreement between labeled and present quantities of total n-3 fatty acids and vitamin E for the majority of samples, significant differences in agreement between individual fatty acids were observed, as well as frequent mismatches between declared and present vitamin E derivatives.

Poultry Meat and Eggs as an Alternative Source of n-3 Long-Chain Polyunsaturated Fatty Acids for Human Nutrition.

The beneficial effects of n-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) on human health are widely known. Humans are rather inefficient in synthesizing n-3 LC-PUFA; thus, these compounds should be supplemented in the diet. However, most Western human diets have unbalanced n-6/n-3 ratios resulting from eating habits and the fact that fish sources (rich in n-3 LC-PUFA) are not sufficient (worldwide deficit ~347,956 t/y) to meet the world requirements. In this context, it is necessary to find new and sustainable sources of n-3 LC-PUFA. Poultry products can provide humans n-3 LC-PUFA due to physiological characteristics and the wide consumption of meat and eggs. The present work aims to provide a general overview of the main strategies that should be adopted during rearing and postproduction to enrich and preserve n-3 LC-PUFA in poultry products. The strategies include dietary supplementation of α-Linolenic acid (ALA) or n-3 LC-PUFA, or enhancing n-3 LC-PUFA by improving the LA (Linoleic acid)/ALA ratio and antioxidant concentrations. Moreover, factors such as genotype, rearing system, transport, and cooking processes can impact the n-3 LC-PUFA in poultry products. The use of a multifactorial view in the entire production chain allows the relevant enrichment and preservation of n-3 LC-PUFA in poultry products.

Dietary strategies to enrich milk with healthy fatty acids – A review

Abstract Feed is the main factor impacting the composition and quality of milk of dairy animals. Therefore, the present review explores the effects of feed and nutrition on milk fat content and levels of healthy fatty acids (FA) in milk consumed by humans. Milk and dairy products are two main sources of healthy and unhealthy FA in human nutrition. The concentrations of FA in milk depend mainly on diets; therefore, milk FA concentrations and ratios can be greatly altered by some feeding strategies. Dietary supplementation of the diets of dairy livestock with vegetable seeds or oils, microalgae and phytogenic feed additives, and feeding of some grasses can enhance the contents of healthy FA, including n-3 FA, α-linolenic acid, conjugated linoleic acid (CLA) and, generally, unsaturated FA in milk and dairy products. Enrichment of milk with healthy FA may make milk a source of anticarcinogens (CLA and polyphenols) for human health. This review, therefore, focusses on the current research findings on enrichment of milk with healthy FA and summarizes some effective supplementation strategies to alter milk FA profile.

Effect of dietary black soldier fly larvae meal on fatty acid composition of lipids and sn-2 position of triglycerides of marketable size gilthead sea bream fillets

Insects are an alternative protein source recently adopted in aquacultured carnivorous species. However, the replacement of marine ingredients with insect meal might cut down the n−3 polyunsaturated fatty acid (PUFA) of fish fillet, a fraction of great interest for human nutrition. Recently, it has been shown that different animal species and the human species better absorb the dietary fatty acids esterified in the sn-2 position of the triglyceride (TG) than those contained in the sn-1 and sn-3 positions. Hence, the present paper aimed to evaluate in which extent replacing fishmeal with insect meal could alter not only the fillet fatty acid (FA) profile, but even the FA distribution inside the TGs. Specifically, three-hundred and sixty gilthead sea bream (Sparus aurata L.) were randomly divided into four groups with three replicates each. The fish were fed for 120 days four isoenergetic, isolipidic and isoproteic diets where fishmeal was substituted by Hermetia illucens (HI) larvae meal. The inclusion levels were 0% (HI0), 9.2% (HI9), 18.4% (HI18) and 27.6% (HI27), corresponding to 0, 25, 50 and 75% of fishmeal substitution, respectively. At the end of the trial, 10 fish per group were analyzed for marketable traits, fillet color values and lipid composition. Dietary intervention did not affect any of the considered marketable traits and the fillet color. Major changes were observed in the FA profile of the fillet lipids. The fillets from fish fed HI18 and HI27 contained the highest amount of saturated fatty acids while their n-3 PUFA were significantly reduced in comparison with HI0 and HI9. Analogous variations were observed in the FA composition of the TG fraction. On the contrary, dietary HI did not reduce the overall n-3 PUFA positioned in the sn-2 of TG, nor eicosapentaenoic acid (EPA) percentage. In conclusion, dietary HI inclusion did not substantially modify the presence of important fatty acids for human nutrition (i.e., EPA and DHA) in the sn-2 position of the fillet triglycerides, increasing the chances to be better assimilated and absorbed by humans.

Seaweeds as Valuable Sources of Essential Fatty Acids for Human Nutrition.

The overexploitation of terrestrial habitats, combined with the ever-growing demand for food, has led to the search for alternative food sources. The importance of seaweeds as food sources has been growing, and their potential as sources of fatty acids (FA) make seaweeds an interesting feedstock for the food and nutraceutical industries. The aim of this study is to assess the potential of five red seaweeds (Asparagospis armata, Calliblepharis jubata, Chondracanthus teedei var. lusitanicus, Gracilaria gracilis, and Grateloupia turuturu) and three brown seaweeds (Colpomenia peregrina, Sargassum muticum and Undaria pinnatifida), harvested in central Portugal, as effective sources of essential FA for food or as dietary supplements. FA were extracted from the biomass, transmethylated to methyl esters, and analyzed through gas chromatography-mass spectrometry. G. gracilis presented the highest content of saturated fatty acids (SFA) (41.49 mg·g−1), whereas C. jubata exhibited the highest content of highly unsaturated fatty acids (HUFA) (28.56 mg·g−1); the three G. turuturu life cycle stages presented prominent SFA and HUFA contents. Omega-6/omega-3 ratios were assessed and, in combination with PUFA+HUFA/SFA ratios, it is suggested that C. jubata and U. pinnatifida may be the algae with highest nutraceutical potential, promoting health benefits and contributing to a balanced dietary intake of fatty acids.

Role of n-3 long-chain polyunsaturated fatty acids in human nutrition and health: review of recent studies and recommendations.

Long-chain (LC) n-3 polyunsaturated fatty acids (n-3 PUFAs), in particular docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), are nutrients involved in many metabolic and physiological processes, and are referred to as n-3 LCPUFA. They have been extensively studied for their effects in human nutrition and health. This paper provides an overview on metabolism, sources, dietary intake, and status of n-3 LCPUFA. A summary of the dietary recommendations for n-3 LCPUFAs for different age groups as well as specific physiological conditions is provided. Evidence for n-3 LCPUFA in cardiovascular diseases, including new studies, is reviewed. Expert recommendations generally support a beneficial effect of n-3 LCPUFA on cardiovascular health and recommend a daily intake of 500 mg as DHA and EPA, or 1–2 servings of fish per week. The role of n-3 LCPUFA on brain health, in particular neurodegenerative disorders and depression, is reviewed. The evidence for beneficial effects of n-3 LCPUFA on neurodegenerative disorders is non-conclusive despite mechanistic support and observational data. Hence, no definite n-3 LCPUFA expert recommendations are made. Data for the beneficial effect of n-3 LCPUFA on depression are generally compelling. Expert recommendations have been established: 200–300 mg/day for depression; up to 1–2 g/day for major depressive disorder. Recent studies support a beneficial role of n-3 LCPUFAs in reducing the risk for premature birth, with a daily intake of 600–800 mg of DHA during pregnancy. Finally, international experts recently reviewed the scientific evidence on DHA and arachidonic acid (ARA) in infant nutrition and concluded that the totality of data support that infant and follow-on formulas should provide both DHA and ARA at levels similar to those in breast milk. In conclusion, the available scientific data support that dietary recommendations for n-3 LCPUFA should be established for the general population and for subjects with specific physiological conditions.

Facing the challenge of discarded fish: improving nutritional quality of two insect species larvae for use as feed and food

Fishery discards represent 10% of total fishery catches, and insect rearing can be accomplished using this wasted resource. Considering that fish are the main source of n-3 very long-chain polyunsaturated fatty acids for human nutrition, and that fish contain both eicosapentaenoic acid and docosahexaenoic acid, this study focused on monitoring the accumulation of such n-3 very long-chain polyunsaturated fatty acids in insect larvae. To determine the feasibility of this process, we monitored nutritional changes achieved in two insect larvae – black soldier fly (Hermetia illucens Linnaeus, 1758) and mealworm (Tenebrio molitor Linnaeus, 1758) – fed using two different fish species from discards, i.e. round sardinella (Sardinella aurita Valenciennes, 1847) and blackspot seabream (Pagellus bogaraveo, Brünnich, 1768). Five different diets were prepared: control (broiler feed), 50% discarded fish (round sardinella and blackspot seabream) +50% broiler feed, and 100% discarded fish. The 100% blackspot seabream fedH. illucens accumulated eicosapentaenoic acid and docosahexaenoic acid up to 2.4 g /100 g and 0.8 g /100 g, respectively.T. molitor accumulated lower amounts of both n-3 very long-chain polyunsaturated fatty acids due to the low intake of fish-containing feed by the larvae.

Evaluation of growth and fatty acid profile of Nile tilapia (Oreochromis niloticus) fed with Schizochytrium sp.

The objective of this study was to evaluate growth, body chemical composition and lipid profile of Nile tilapia juvenile fed with Schizochytrium sp. Two hundred and forty Nile tilapia (Oreochromis niloticus) juvenile (1.33 ± 0.11 g) were distributed in 20 aquariums, at the density of 12 fish per aquarium. The juvenile were fed with five levels of Schizochytrium sp.: Control (0 g/kg of Schizochytrium sp. in feed); AS10 (10 g/kg of Schizochytrium sp. in feed); AS20 (20 g/kg of Schizochytrium sp. in feed); AS30 (30 g/kg of Schizochytrium sp. in feed) and AS40 (40 g/kg of Schizochytrium sp. in feed). The inclusion of Schizochytrium sp. increase the body weight, weight gain, final biomass and biomass gain of tilapia juvenile. The body crude protein of tilapia was increased after addition of microalgae. Juvenile lipid profile also was influenced when fed with 40 g of Schizochytrium sp. per kg and have it levels of omega-3 and docosahexaenoic acid (DHA) increased, and omega 6:3 ratio decreased. Tilapia juvenile fed with Schizochytrium sp. per kg have better growth, omega-3 and DHA levels rising in the body, being a considerable source of fatty acids for human nutrition. Levels above 20 g of Schizochytrium sp. per kg in the feed favour the increase of protein in Nile tilapia body.

Seasonal variations in the fatty acid composition of Greek wild rabbit meat

The fatty acid (FA) profile of the Longissimus thoracis et lumborum muscle (LL) was used to investigate seasonal variation (September, November and March) in wild rabbits from Lemnos Island (Greece). The n-3 FA were particularly high during early March in comparison (P<0.05) with late September and late November. Thrombogenicity index (TI) values were lower in March (P<0.05) compared to the other periods. High concentrations of odd- and branched-chain FA were found in the meat of wild rabbits; however, they were not different among the considered periods of the year. The present results showed that wild rabbit meat has a desirable FA profile, particularly during early spring, and it could be a good source of bioactive FA in human nutrition.

Fatty Acid Profile of the Amazon Caiman Protein Concentrate

Some species of fish and other aquatic organism are important sources of protein and fatty acids that are beneficial to human health and can be industrially processed. The fatty acid profile of Melanosuchus niger (native to the Brazilian Amazon flooded forest) was determined in samples of a protein concentrate (PC) there was obtained from processing residues. The PC was prepared from cooked muscle portions and NaCl (1.5%) using an adiabatic process. Saturated fatty acids such as stearic acid (0.59%) and palmitic acid (1.43%) were present. The levels of the unsaturated fatty acids omega-6 (ω-6) and omega-3 (ω-3) linolenic acids were 0.32% and 0.15%, respectively. In conclusion, Black caiman PC seems to provide essential fatty acids for human nutrition. Clinical studies are necessary to assess the influence of fatty acids from Amazon Caimans on human diet and the feasibility of obtaining new products such as nutraceuticals.

Aflatoxin Contamination Detected in Nutrient and Anti-Oxidant Rich Edible Stink Bug Stored in Recycled Grain Containers.

Recently, there has been multi-agency promotion of entomophagy as an environmentally-friendly source of food for the ever increasing human population especially in the developing countries. However, food quality and safety concerns must first be addressed in this context. We addressed these concerns in the present study using the edible stink bug Encosternum delegorguei, which is widely consumed in southern Africa. We analysed for mycotoxins, and health beneficials including antioxidants, amino acids and essential fatty acids using liquid chromatography coupled to quadrupole time of flight mass spectrometry (LC-Qtof-MS) and coupled gas chromatography (GC)-MS. We also performed proximate analysis to determine nutritional components. We identified the human carcinogen mycotoxin (aflatoxin B1) at low levels in edible stink bugs that were stored in traditonally woven wooden dung smeared baskets and gunny bags previously used to store cereals. However, it was absent in insects stored in clean zip lock bags. On the other hand, we identified 10 fatty acids, of which 7 are considered essential fatty acids for human nutrition and health; 4 flavonoids and 12 amino acids of which two are considered the most limiting amino acids in cereal based diets. The edible stink bug also contained high crude protein and fats but was a poor source of minerals, except for phosphorus which was found in relatively high levels. Our results show that the edible stink bug is a nutrient- and antioxidant-rich source of food and health benefits for human consumption. As such, use of better handling and storage methods can help eliminate contamination of the edible stink bug with the carcinogen aflatoxin and ensure its safety as human food.

Linoleic Acid-Induced Growth Inhibition of Human Gastric Epithelial Adenocarcinoma AGS Cells is Associated with Down-Regulation of Prostaglandin E2 Synthesis and Telomerase Activity.

Linoleic acid is the most abundant polyunsaturated fatty acid in human nutrition and found in most vegetable oils and certain food products. In the present study, we investigated the effects of linoleic acid on the growth of human epithelial adenocarcinoma AGS cells. MTT assay, flow cytometry, RT-PCR and Western-blot analyses were used to investigate the effects and underlying mechanisms of linoleic acid on AGS cells. The effects of this compound were also tested on prostaglandin E2 (PGE2) production and telomerase activity. Our data indicated that growth inhibition of AGS cells by linoleic acid treatment was associated with induction of apoptosis. Linoleic acid treatment decreased the expression levels of the cyclooxygenase (COX)-2 mRNA and protein without causing significant changes in the COX-1 levels, which was correlated with the inhibition of PGE2 synthesis. Linoleic acid treatment also decreased the expression of human telomerase reverse transcriptase (hTERT), a main determinant of the telomerase enzymatic activity, and activity of telomerase, with inhibiting the expression of c-myc in a concentration-dependent manner. Taken together, our results indicate that linoleic acid inhibits the production of PGE2 and activity of telomerase by suppressing COX-2 and hTERT expression.

Can a marine pest reduce the nutritional value of Mediterranean fish flesh?

Biological invasions are deemed as one of the most severe threats to biodiversity and can result in huge economic impacts on natural resources. Many studies have assessed structural changes caused by the introduction of nonindigenous species, but urgent questions remain to be answered about the complex indirect effects of invasive pests on marine biodiversity at the species and ecosystem level. Previous investigations focusing on the biological invasion by Caulerpa racemosa in the Mediterranean indicated that the lipophilic algal metabolite caulerpin accumulates in the tissues of the native edible white sea bream Diplodus sargus feeding on the exotic alga. The level of caulerpin in D. sargus has been thus used as an indicator of the trophic exposure of the fish to the seaweed and related with adverse effects on the fish health. In the present study, we show that the C. racemosa-based diet can also alter muscle fatty acids composition in the white sea bream by reducing the percentage of polyunsaturated fatty acids of the n-3 and n-6 series, such as eicosapentaenoic (C20:5), docosahexaenoic (C22:6) and arachidonic acids (C20:4). This represents both a real threat to the health of fish unable to biosynthesize these essential fatty acids and an impoverishment of an important source of essential fatty acids for human nutrition, which helps to prevent cardiovascular diseases, diabetes, hepatic steatosis, cancer and neurological disorders. On the whole, this work sheds light on an unexplored and critical aspect of biological invasions with implication on the health of both humans and the environment.

In nutritional science, the same issues often are revisited again and again, but from deepened foundations of knowledge and the new perspectives they bring. Examples discussed include the ongoing debates surrounding the roles of dietary fiber, phytic and oxalic acids, and fish oils and omega 3 fatty acids in human nutrition. Despite the accomplishments made possible by science, consumers often view science and scientists with skepticism. Overwhelmed by contradictory and inconclusive information, consumers are increasingly demanding certainty from the nutrition, health, and physiology sciences. In the modern information age, professional and industry associations need to invest far more in establishing a credible presence in electronic and social media platforms.

Genome-wide Identification and Characterization of the Gene Families Controlling Fatty Acid Biosynthesis in Flax (Linum usitatissimum L)

Flax (Linum usitatissimum L.) is an important crop with many characteristic features such as its abundant essential ω-3 fatty acids for human nutrition. Fatty acid (FA) biosynthesis in plants, including flax, involves several consecutive steps governed by different gene families. Using in silico gene mining and comparative analysis, genome-wide gene identification and characterization were performed for six gene families related to FA biosynthesis, including KAS, SAD, FAD, KCS and FAT. We identified 91 FA-related genes from flax cv. CDC Bethune genome, from which seven previously cloned genes were validated. The newly identified 84 FA-related genes include 14 novel genes from the KAS family, two from the SAD family, 13 from the FAD2 family, three from the FAD3 family, 38 from the KCS family and 14 from the FAT family. Out of the 91 genes identified, 88 were duplicated as a consequence of recent whole genome duplication events, in which 13 FAD2 genes were hypothesized to have evolved from tandem gene duplication events followed by a whole genome duplication event and, more recently, by a single gene deletion. The six gene families described here are highly conserved in plants and have diverged anciently. These newly identified flax genes will be a useful resource for further research on FA gene cloning and expression, QTL identification, marker development and marker-assisted selection.

Carcass traits and fatty acid profile of meat from lambs fed different cottonseed by-products

The aim of this study was to evaluate the changes in carcass traits and fatty acid profile of meat by feeding lambs with cottonseed by-products. Twenty-four 5-month old ram lambs received one of four diets: 19.5% of dry matter intake (DMI) of whole cottonseed (WCS), 19.5% DMI cottonseed meal (CSM), 19.5% DMI high oil cottonseed meal (CSC) and a control group (CTL) without cottonseed by-products. After 95 experimental days, the lambs were slaughtered. Carcass weight and 12th rib composition (chemical and centesimal) were measured. Samples of Longissumus dorsi muscle were taken for fatty acid profile analysis. The animals that received CSM showed higher hot carcass weight, carcass yield and rib eye area than animals from the WCS group. Meat from CSM and CSC groups had higher levels of conjugated linolenic acid (CLA) than others and yet CSC group showed higher vaccenic acid than others. Meat from animals that received whole cottonseed had less unsaturated fatty acids, CLA and vaccenic acid. Therefore, processed cottonseed by-products (CSM and CSC) should be preferred for use in ruminant feed over whole cottonseed. The meat from animals that did not receive cotton by-products had higher n-3 fatty acids, and also better n-6 to n-3 ratio compared to others, which may indicate a problem in using these products in ruminant nutrition due to current importance given to these fatty acids in human nutrition.

Linoleic acid: Between doubts and certainties

Linoleic acid is the most abundant polyunsaturated fatty acid in human nutrition and represents about 14 g per day in the US diet. Following the discovery of its essential functions in animals and humans in the early 1920's, studies are currently questioning the real requirement of linoleic acid. It seems now overestimated and creates controversy: how much linoleic acid should be consumed in a healthy diet? Beyond the necessity to redefine the dietary requirement of linoleic acid, many questions concerning the consequences of its excessive consumption on human health arise. Linoleic acid is a direct precursor of the bioactive oxidized linoleic acid metabolites. It is also a precursor of arachidonic acid, which produces pro-inflammatory eicosanoids and endocannabinoids. A majority of the studies on linoleic acid and its derivatives show a direct/indirect link with inflammation and metabolic diseases. Many authors claim that a high linoleic acid intake may promote inflammation in humans. This review tries to (i) highlight the importance of reconsidering the actual requirement of linoleic acid (ii) point out the lack of knowledge between dietary levels of linoleic acid and the molecular mechanisms explaining its physiological roles (iii) demonstrate the relevance of carrying out further human studies on the single variable linoleic acid.
