Molecular profiling of triacylglycerols in bovine and goat milk powders via ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry

Triacylglycerols (TAG) play several nutritional, technological, and functional roles in the dairy industry, representing approximately 98 % of milk fat. In the present study, the total fatty acid composition and the sn-2 fatty acid positional distribution of bovine and goat milk powders fat were analyzed by using the gas chromatography-flame ionization detector. Furthermore, the identification of TAG molecular species from milk powders was conducted using ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). The separation of milk TAG was accomplished by using isopropanol/acetonitrile (90:10, v/v) as mobile phase A, and water/acetonitrile (60:40, v/v) as mobile phase B. Myristic, palmitic, stearic, and oleic acids were identified as the most abundant fatty acids making up 11.55 and 10.71 %, 33.23 and 30.64 %, 10.86 and 12.52 %, and 26.74 and 21.80 % in the bovine and goat milk powders, respectively. In bovine milk, 221 molecular species of TAG were identified, with the predominant molecular species (LaCyO, CaLaPa, CyPaM, OCaCa, CaGCy, GLaCo) detected at a retention time of 6.362 min, with a relative abundance of 8.28 %. In goat milk, 202 molecular species were identified, with the major molecular species (CaLaM, CaPCa, CaCyS, CaCoAra, CaBuBe, LaPCy, LaCoS, LaBuAra, MPCo, MCyM, MBuS, PBuP, CyCoBe) detected at a retention time of 6.106 min, making up 8.14 % of the total TAG detected. These results provide valuable insights into milk fat composition for the dairy sector, aiding in the development of specialized dairy products.

Improvement in Muscle Fatty Acid Bioavailability and Volatile Flavor in Tilapia by Dietary α-Linolenic Acid Nutrition Strategy.

To investigate the modification of muscle quality of farmed tilapia through dietary fatty acid strategies, two diets were formulated. Diet SO, using soybean oil as the lipid source, and diet BO, using blended soybean and linseed oils, each including 0.58% and 1.35% α-linolenic acid (ALA), respectively, were formulated to feed juvenile tilapia for 10 weeks. The muscular nutrition composition, positional distribution of fatty acid in triglycerides (TAGs) and phospholipids (PLs), volatile flavor, lipid mobilization and oxidation were then analyzed. The results showed that there was no distinct difference between the SO and BO groups in terms of the nutrition composition, including crude protein, crude lipid, TAGs, PLs, and amino acid. Although the fatty acid distribution characteristics in ATGs and PLs showed a similar trend in the two groups, a higher level of n-3 PUFA (polyunsaturated fatty acid) and n-3 LC-PUFA (long-chain polyunsaturated fatty acid) bound to the glycerol backbone of TAGs and PLs was detected in the BO group than the SO group, whereas the opposite was true for n-6 PUFA. Additionally, the muscular volatile aldehyde and alcohol levels were higher in the BO group. Moreover, the expression of enzymatic genes and protein activities related to lipid mobilization (LPL, LPCAT, DGAT) and oxidation (LOX and GPX) was higher in the BO group. The results demonstrate that high-ALA diets may improve the fatty acid bioavailability and volatile flavor of tilapia by improving the lipid mobilization and oxidation, which provides new ideas for the improvement of muscle quality in farmed fish.

Assessment of the Nutritional Potential and Resistance to Oxidation of Sea Buckthorn and Rosehip Oils

Cold-pressed oils from non-typical materials, such as wild rose and sea buckthorn, should meet certain requirements to be approved for consumption. The aim of this study was to evaluate the nutritional potential and selected quality parameters with particular emphasis on oxidative stability of two commercially available cold-pressed oils: rose hip oil and sea buckthorn oil. The fatty acid composition, including nutritional indexes (polyunsaturated fatty acid/saturated fatty acid ratio PUFA/SFA; atherogenicity index IA is as follows: hypocholesterolemic/hypercholesterolemic index HH; health-promoting index HPI), positional distribution of fatty acids, melting profile and oxidative stability were analyzed. The tested oils were characterized by a high content of unsaturated fatty acids, which were mainly represented by oleic and linoleic acids. In the case of one of the analyzed rosehip oil oils (R1), α-linolenic acid was also found at the level of 26%, which contributed to obtaining the recommended ratio of omega-6 to omega-3 fatty acids. The lowest value of IA and the highest values of HH and HPI were also recorded for R1 rosehip oil. This oil was also characterized by the highest resistance to oxidation, as indicated by induction times measured at various temperatures. The distribution of fatty acids in triacylglycerols molecules in the analyzed oils was typical for vegetable oils. In the case of tested oils, similar melting profiles with endothermic peaks recorded at negative temperatures, which resulted from the presence of triacylglycerol fractions with a high proportion of mono- and polyunsaturated fatty acids, were observed.

Detection and analysis of triacylglycerol regioisomers via electron activated dissociation (EAD) tandem mass spectrometry

Triacylglycerols (TGs) are important components of human diet. The positional distribution of fatty acids (FAs) on the glycerol backbone affects the chemistry and physical properties of fats. Especially for infants, the structure of TGs plays an important role in the growth and development. However, limited by detecting technology, accurately identifying regioisomers of ABA/AAB and BAC/ABC/ACB type TGs is a significant challenge for human milk utilization and the development of infant formula. For this, we exploit a novel method for identifying the regioisomers of ABA/AAB and BAC/ABC/ACB type TGs within complex lipid mixtures, via used electron activated dissociation (EAD) tandem mass spectrometry. The distribution information of acyl chains at the sn-2 and sn-1/3 positions of glycerol backbone and double bonds in unsaturated FAs can be easily obtained by fragmenting TG ions with energetic electrons (15 eV). Then, the standard curve was established by correlating the peak area intensity of sn-2 characteristic product ion with the content of TG regioisomers standard. These analytical methods successfully enabled the identification and quantification of TG regioisomers in human milk, cow milk, infant formula, palm oil, and sunflower oil. Additionally, the distribution of the double-bond positions of unsaturated FAs in these samples was also identified. Compared to traditional methods, this approach eliminates the need for complex processing and analysis procedures, enabling rapid structural characterization of ABA/AAB and BAC/ABC/ACB type TGs within 17 min. Hence, we provide a rapid and convenient methodology for detecting and analyzing ABA/AAB and BAC/ABC/ACB type TG regioisomers, thereby offering valuable assistance in the development of specialized formulations and facilitating effective process control for ensuring the quality of edible oils and fats.

Effects of Chemically Interesterified Palm Olein on Lipid Profiles in Hamsters

Palm olein (POo) has been perceived as atherogenic due to its high proportion of palmitic acid (41.2%) content. It is interesting that most of the palmitic acid of POo is located at stereospecific numbering sn-1 and sn-3 positions of the triacylglycerol (TAG) backbone. The present study aims to investigate the effects of positional distribution of fatty acids on the lipid profiles of POo or chemically interesterified palm olein (CIE POo) fed hamsters in comparison to high oleic sunflower oil (HOSO) fed hamsters. Male weanling Syrian golden hamsters (n=10 for each group), were fed diets formulated with the above oils for 12 weeks. There was no significant difference between CIE POo and HOSO groups for total cholesterol (TC). CIE POo with increased amount of palmitic acid (43.2%) at sn-2 position did not cause significant increases in TC levels compared to the HOSO group. In addition, the POo group has significantly higher high-density lipoprotein cholesterol (HDL-C) than that of the HOSO group, P = 0.011 (< 0.05) while the HOSO group has significantly lower total cholesterol (TC) levels than that of the POo group, P = 0.012 (< 0.05).

The Positional Distribution of Fatty Acids in Palm Oil and Lard Influences Their Biologic Effects in Rats

Dietary fatty acids in the sn-2 position are preferentially absorbed as monoacylglycerols. To determine whether they also have more important biologic effects, rats were fed for 2 and 4 mo a purified diet containing native palm oil, interesterified palm oil, native lard or interesterified lard. Interesterification that increased or decreased the level of fatty acids in position 2 depending on the fat, resulted in significant corresponding changes in the fecal excretion of saturated fatty acids. Fecal excretion of saturated fatty acids was associated with significant changes in some plasma fatty acids. Interesterification in lard resulted in significantly lower plasma triglycerides, and in palm oil, increased platelet aggregation induced by ADP. Lipemia, platelet aggregation and associated plasma fatty acids (palmitic, heneicosanoic and docosahexaenoic acids) were significantly affected only by dietary fatty acids at the sn-2 position. Even without changes in absorption, only linoleic acid in position 2 was correlated with the plasma concentration of the corresponding longer chain arachidonic acid. These results in rats confirm that the fatty acids (saturated and unsaturated) in position 2 of dietary triglycerides play a crucial role in the metabolism and biologic effects of these fatty acids.

Ostrich oil as a fat substitute in milk-based infant formula.

In this study, the possibility of replacing vegetable fats with ostrich oil in infant formula (IF) production was investigated. The fatty acid profile, the positional distribution of fatty acids in the triacylglycerols, the cholesterol content, and the physicochemical properties of ostrich oil were determined and compared with breast milk fat and vegetable oils. In the next step, two infant formulas were produced using ostrich oil and vegetable oils and the physicochemical properties, rheological properties, color parameters, and sensory analysis of the resultant powders were compared. The results showed that the predominant fatty acids in ostrich oil are palmitic acid, oleic acid, and linoleic acid which is similar to breast milk fat and vegetable oils. The presence of appropriate cholesterol content in ostrich oil makes it more similar to breast milk fat compared to vegetable fats. Palmitic acid was located at sn-2 position in 15% triacylglycerol from ostrich fat, which was equal to the amount reported for vegetable fats. The incorporation of ostrich oil in infant formula production showed that there is no statistically significant difference between quality attributes of powder formulated with ostrich oil or vegetable oils. Therefore, ostrich oil can be introduced as a new source of edible oil, and addition of ostrich oil is an effective way to reduce the gap between the composition of breast milk and infant formula.

Positional Distribution of Fatty Acids in Processed Chinook Salmon Roe Lipids Determined by 13C Magnetic Resonance Spectroscopy (NMR)

Recently, there has been great interest in the lipidomic of marine lipids and their potential health benefits. Processing of seafood products can potentially modify the characteristics and composition of lipids. The present study investigated the effect of processing methods (salting and fermentation) on the positional distribution of fatty acids of Chinook salmon roe using 13C nuclear magnetic resonance spectroscopy (NMR). The NMR analysis provided information on the carbonyl atom, double bond/olefinic, glycerol backbone, aliphatic group, and chain ending methyl group regions. The obtained data showed that docosahexaenoic acid (DHA) is the main fatty acid esterified at the sn-2 position of the triacylglycerides (TAGs), while other fatty acids, such as eicosapentaenoic acid (EPA) and stearidonic acid (SDA), were randomly distributed or preferentially esterified at the sn-1 and sn-3 positions. Fermentation of salmon roe was found to enrich the level of DHA at the sn-2 position of the TAG. The processing of roe by both salt drying and fermentation did not appear to affect the proportion of EPA at the sn-2 position. This present study demonstrated that fish roe processing can enhance the proportion of DHA at the sn-2 position and potentially improve its bioavailability.

Isochrysis sp. cultivation in pilot-scale to concurrently produce sustainable triacylglycerols for human milk fat substitutes and fucoxanthin

Isochcrysis strains as versatile cell factories can produce varieties of high-valued bioproducts for food, nutraceuticals and pharmaceuticals. This work aimed to evaluate the ability of Isochrysis sp. to produce high-valued triacylglycerols (TAGs) for human milk fat substitutes (HMFs) and fucoxanthin by batch, fed-batch and semi-continuous cultivation modes in 50-L column photobioreactors. It was found that, Isochrysis cells by three cultivation modes had individual growth characteristics, nutrients consumption, and biochemical composition. In all cultivation modes, microalga cultivated by the semi-continuous cultivation at 10 % renewable rate achieves the highest TAGs and fucoxanthin productivities with 8.2 and 0.5 mg/(L·d), respectively. Additionally, the obtained results showed that the microalgal growth phase and the levels of nitrogen and phosphorus had distinct impacts on the deducting G value for microalgal TAGs produced by three cultivation modes. Based on the fatty acid composition and positional distribution, Isochrysis TAGs by the semi-continuous process at 10 % renewable rate had 73.0 deducting G value that were close to the ones by the batch cultivation (13th day) and fed-batch cultivation. Taken together, Isochrysis sp. was the promising marine microalga to concurrently produce sustainable high-valued TAGs for HMFs and fucoxanthin by semi-continuous cultivation in pilot-scale photobioreactors. This work provides a new marine microalgal platform to exploit Isochrysis-derived TAGs as human milk fat subsitutes for infant formulas and fucoxanthin for nutraceuticals and pharmaceuticals.

Similarity Index for the Fat Fraction between Breast Milk and Infant Formulas.

The similarity of the fat fraction in infant formulas rich in either bovine milk fat (MF) or vegetable oil (VO) to breast milk was evaluated by analyzing their lipid composition. Milk fat-rich formulas were highly similar (average similarity index 0.68) to breast milk compared to the VO-rich formulas (average similarity index 0.56). The highest difference in the indices was found in the contents of cholesterol (0.66 vs 0.28 in MF- and VO-rich formulas, respectively, on average) and polar lipids (0.84 vs 0.53), the positional distribution of fatty acids in the sn-2 position of triacylglycerols (0.53 vs 0.28), and fatty acid composition (0.72 vs 0.54). The VO-based formulas were superior in similarity in n – 6 PUFA. Thus, the addition of bovine MF fractions is an effective way to increase the similarity between the lipid composition of infant formulas and human milk.

Bioaccessibility of phospholipids in homogenized goat milk: Lipid digestion ecology through INFOGEST model

Phospholipids-rich goat milk provides health benefits to consumers. The effects of homogenization on the disruption and recombination of milk fat globule membrane and change the fatty acid positional distribution in glycerophospholipids profile by phosphatidylcholine metabolism pathways were investigated. Goat milk was homogenized at different intensity pressure. Homogenized samples were introduced into harmonized INFOGEST digestion model. Results showed that phosphatidylcholine increased significantly during storage in 30 MPa and were approximately twice that in raw milk (LOD 0.27–1.49 μg/L and LOQ 0.89–4.92 μg/L, respectively). Meanwhile, both linoleic acid (C18:2) and α-linolenic acid (C18:3ω-3), the foremost polyunsaturated acyl chains in homogenized milk extracts, showed upward trends. Notably, homogenization increased the number and altered the composition of Sn-1, 2 diacylglycerols via increasing trypsin and pancreatic lipase (PLRP2, MAUC15, CD36 and BSSL) expression and accelerated the phosphatidylcholine conversion. Ultimately, the relationship between homogenization and milk fat globule recombination and phospholipids bioaccessibility was preliminary established.

Bovine milk fatty acid and triacylglycerol composition and structure differ between early and late lactation influencing milk fat solid fat content

Milk samples were collected in early lactation (8–14 days in milk; DIM) and late lactation (199–326 DIM) from 11 cows to assess the differences in bovine milk fatty acid (FA) and triacylglycerol (TAG) composition, FA positional distribution in the TAG structure, and milk solid fat content (SFC). Cows in early lactation that were in negative energy balance, mobilised C16:0, C18:0, and C18:1cis9 from their body fat stores, increasing the concentrations of these FA in milk fat. These high concentrations of C18:0 and C18:1cis9 enhanced the synthesis of high molecular weight TAG and decreased milk fat SFC in early lactation. For both lactation stages, alterations in the total FA concentrations in the TAG structures resulted in changes in the abundances and the proportions of the FA at the sn-2 and sn-1 (3) positions in the TAG structure, yet without affecting the main esterification preferences of the FA.

Feeding hydrogenated palm fatty acids and rumen-protected protein to lactating Holstein-Friesian dairy cows modifies milk fat triacylglycerol composition and structure, and solid fat content

The aim of this study was to analyze the effect of fat and protein supplementation to dairy cattle rations on milk fat triacylglycerol (TAG) composition, fatty acid (FA) positional distribution in the TAG structure, and milk solid fat content (SFC). Fifty-six lactating Holstein-Friesian cows were blocked into 14 groups of 4 cows and randomly assigned 1 of 4 dietary treatments fed for 28 d: (1) low protein, low fat, (2) high protein, low fat, (3) low protein, high fat, and (4) high protein, high fat. The high protein and high fat diets were obtained by isoenergetically supplementing the basal ration (low protein, low fat) with rumen-protected soybean meal and rumen-protected rapeseed meal, and hydrogenated palm FA (mainly C16:0 and C18:0), respectively. Fat supplementation modified milk TAG composition more extensively compared with protein supplementation. Fat supplementation resulted in decreased concentrations of the low molecular weight TAG carbon number (CN) 26 to CN34 and medium molecular weight TAG CN40, CN44, and CN46, and increased concentrations of CN38 and the high molecular weight TAG CN50 and CN52. Increased contents of C16:0, C18:0, and C18:1cis-9 in TAG in response to fat supplementation were related to increases in the relative concentrations of C16:0 and C18:0 at the sn-2 position and C18:0 and C18:1cis-9 at the sn-1(3) positions of the TAG structure. Increased concentrations of high molecular weight TAG species CN50 and CN52 in response to fat supplementation was associated with increased milk SFC at 20, 25, and 30°C. Our study shows that important alterations in milk TAG composition and structure occur when feeding hydrogenated palm FA to lactating dairy cattle, and that these alterations result in an increased SFC of milk fat. These changes in milk SFC and TAG composition and structure may improve absorption of both fat and minerals in milk-based products for infants and may affect processing of milk fat.

Lipidomic Profiling Reveals the Reducing Lipid Accumulation Effect of Dietary Taurine in Groupers (Epinephelus coioides).

A lipidomic analysis was conducted to provide the first detailed overview of lipid molecule profiles in response to dietary lipid and taurine and associations of liver lipid-lowering effects of dietary taurine with lipid molecular species and the positional distributions of fatty acids in the liver of juvenile orange-spotted groupers (Epinephelus coioides). The results indicated that the liver was more sensitive to varied dietary lipid and taurine contents than the muscle with regard to lipid molecules. A total of 131 differential lipid molecules (DLMs) were observed in the liver of groupers when dietary taurine was increased from 0 to 1% at 15% lipid, among which all the up and down-regulated DLMs are phospholipids (PLs) and triglycerides (TGs), respectively. The liver content of TGs containing 18:2n-6 attached at the sn-2 and sn-3 positions on the glycerol backbone increased with increasing dietary lipid from 10 to 15% but decreased with increasing dietary taurine from 0 to 1%. Therefore, dietary taurine can not only reduce lipid accumulation through decreasing the contents of TGs containing 18:2n-6 at the sn-2 and sn-3 positions but also enhance the anti-inflammatory capacity and health status of groupers. This study will also provide a new insight into the function of taurine in farmed fish.

The positional distribution of fatty acids in the triacylglycerol backbones of yak milk from different pastoral areas

The regiodistribution of fatty acids (FAs) in triacylglycerols plays a critical role in the digestion and adsorption of milk lipids. However, the stereospecific location of FAs in yak milk triacylglycerols remains unknown. In this study, the intrapositional and interpositional distribution of FAs in the yak milk from different pastoral areas were compared with those of human milk and cow milk. When compared with human milk, the stereospecific locations of FAs were relatively similar between yak milk and cow milk. However, White yak milk and human milk contained similar interpositional proportions of linoleic, α-linolenic acid and polyunsaturated FAs at the sn-3 position, significantly higher than those of cow milk. Moreover, the sn-3 interpositional proportion of α-linolenic acid in Maiwa yak milk was even higher than that in human milk. The study suggests that yak milk might provide better nutritional benefits as a source of essential FAs than cow milk.

Lipidomic profiling reveals molecular modification of lipids in hepatopancreas of juvenile mud crab (Scylla paramamosain) fed with different dietary DHA/EPA ratios

Untargeted lipidomic analysis was conducted to explore how different dietary docosahexaenoic acid (DHA)/eicosapentaenoic acid (EPA) ratio and, specifically, how an optimal ratio (2.3) compared to a suboptimum ratio (0.6) impacted lipid molecular species and the positional distribution of fatty acids in hepatopancreas of mud crab. The results indicated that major category of lipid affected by dietary DHA/EPA ratio was glycerophospholipids (GPs). The optimum dietary DHA/EPA ratio increased the contents of DHA bound to the sn-2 and sn-3 positions of phosphatidylcholine (PC) and triacylglycerol, EPA bound to the sn-2 position of phosphatidylcholine and 18:2n-6 bound to the sn-2 position of phosphatidylethanolamine (PE). Increased dietary DHA/EPA ratio also led to competition between arachidonic acid (ARA) and 18:2n-6 bound to esterified sites. Appropriate dietary DHA/EPA ratio can not only improve the growth performance and nutritional quality of mud crab, but also provide higher quality products for human consumers.

Seasonal variation in the positional distribution of fatty acids in bovine milk fat

The aim of this study was to determine the seasonal variation in the positional distribution of fatty acids (FA) in bovine milk fat. Bovine milk samples were collected from May 2017 to April 2018 in the Netherlands, and the FA composition in the sn-2 position was determined by using sn-1(3)-selective transesterification of Candida antarctica lipase B. The majority of the FA showed significant variation at sn-2 and sn-1(3) positions between different seasons. The seasonal variation in sn-2 position was higher than the sn-1(3) positions. Parallel to the changes in the diet of the cows throughout a year, we observed an increase in blood-derived FA (i.e. C18:0, C18:1 cis-9) concentrations and a decrease in de novo-synthesized FA during summer. In winter, more saturated FA were esterified in sn-2 position of milk fat. Highest concentrations of palmitic acid, C16:0, was observed in sn-2 position in winter, whereas the amount of unsaturated FA at this position was highest in summer. These results showed that the FA compositions in different regiospecific positions changed due to season; however, the proportions of a specific FA within the 3 positions of the triacylglycerols in milk fat did not change upon seasonal variation.

Free-Range Feeding Alters Fatty Acid Composition at the sn-2 Position of Triglycerides and Subcutaneous Fat Physicochemical Properties in Heavy Pigs.

Simple SummaryFat content and the degree of fatty acid unsaturation in meat are two major concerns for consumers. Fat concentration and its molecular structure (fatty acid positional distribution) are related to the nutritional fat value and tissue rheological properties. Changes in fat concentration and/or fatty acid profile related to modifications of dietary treatments are well described in the literature. Nevertheless, studies aimed to control fatty acid positional distribution by dietary intervention in pigs are scarce, and studies have shown that the internal sn-2 position is highly regulated and resistant to dietary manipulation. However, this study demonstrated that heavy pigs fed on free-range with high levels of oleic acid can alter the fatty acid composition of the internal position of the triglyceride, thus affecting the nutritional value of their fat as well as their physicochemical properties.The nutritional value of fat consumption depends on both the fatty acid composition and the positional distribution of fatty acids within the triglyceride molecule. This research studies the effect of feeding with three different diets (4% lard-enriched; 11.5% high-oleic sunflower-enriched; and extensive feeding mainly with acorns) on the composition of fatty acids in the sn-2 position (and sn-1,3) of triglycerides and the textural properties of subcutaneous fat in heavy Iberian pigs (n = 210 castrated males). A moderate dietary enrichment with oleic acid in mixed diets did not alter the regulation of the sn-2 position of triglyceride (69.9% and 13.9% of palmitic and oleic acids, respectively), but the extremely high intake of oleic acid in pigs fed mainly on acorns changed the proportions of palmitic and oleic acids at the sn-2 position in the subcutaneous fat of pigs (55.0% and 27.2%, respectively). Hardness, adhesiveness, cohesiveness, gumminess, and chewiness showed the least values in EXT pigs, and the greatest values in LARD-fed barrows. SUN cohesiveness and gumminess did not differ from those fed LARD. In addition, Iberian pigs raised in free-range conditions had a more favorable nutritional lipid profile for human health compared to pigs fed conventional diets.

Positional distribution of fatty acids and phospholipid composition in King salmon (Oncorhynchus tshawytscha) head, roe and skin using nuclear magnetic resonance spectroscopy

This study used a novel extraction method (ETHEX) to extract the lipid content of King salmon head, skin and roe, and determined the lipid profiles using GC-FID, 13C NMR and 31P NMR spectroscopy. On a wet tissue basis, King salmon roe was found to contain the highest amount of phospholipid (26.53 µmol/g) and n-3 fatty acids (43.32%), followed by head (PL = 10.76 µmol/g; n-3 = 7.21%) and skin (PL = 4.98 µmol/g; n-3 = 8.23%). Total EPA (6.62%) and DHA (28.83%) content, along with the sn-2 positioned EPA (3.25%), DPA (1.36%) and DHA (16.35%) were also higher in roe compared with head and skin. The highest amount of EPA (7.99%) and DHA (34.47%) contents were found in the polar lipid fractions of roe, followed by skin (EPA = 4.19%; DHA = 25.95%) and head (EPA = 2.61%; DHA = 17.85%). This result suggests that salmon roe could be used for developing n-3 phospholipid enriched products.

Lipidomics study on the molecular changes of eicosapentaenoic and docosahexaenoic acyl structured glycerides during enzyme-catalysis and chemocatalysis

Enzyme-catalysis and chemocatalysis of fish oil are popularly used in industrial transesterification, but their effect on the intact molecular species is still unclear. This work aimed to trace the variation of lipid molecules and compare the performance of these two methods. During the transesterification reaction, lipidomics study was fully exploited using ultra performance liquid chromatography triple time-of-flight mass spectrometry to characterize the glycerides. The positional distribution of fatty acids in the structured triglycerides was interpreted. A total of 543 glycerides including 15 monoglycerides, 129 diglycerides and 399 triglycerides have been identified and quantified. The results of principal component analysis and orthogonal partial least squares-discriminant analysis displayed a clear molecular phenotype of different catalyzed oils, which also indicated the main difference of the molecules of TG (60:15), TG (60:14), TG (62:16), etc. In conclusion, the enrichment of EPA and DHA by enzyme-catalysis was superior to that by chemical catalysis.
