Enhancing efficiency of ultrasound-assisted biodiesel production catalyzed by Eversa® Transform 2.0 at low lipase concentration: Enzyme characterization and process optimization

This study focused on the ultrasound-assisted transesterification of simulated low-quality feedstocks using a low-cost liquid lipase Eversa® Transform 2.0 (ET2). Enzyme characterization was also performed to investigate the effect of ultrasound parameters on enzyme structure. The optimal ultrasound parameters, 40 % amplitude, and 5 % duty cycle effectively enhanced the reaction rate compared to the conventional stirring method while retaining 95 % of the enzyme activity. Analysis of circular dichroism (CD) spectra revealed the preservation of the secondary structure of ET2 under the optimal ultrasound intensities, while fluorescence spectra indicated a slight change in its tertiary structure. The implementation of a two-stage methanol dosing strategy in the ultrasound-assisted reaction effectively mitigated lipase inhibition, yielding a remarkable fatty acid methyl ester (FAME) content of 92.2 % achieved within a 12-h reaction time. Notable, this high FAME content was achieved with only a 4:1 methanol-to-oil molar ratio and a 0.5 wt% enzyme concentration. Under these optimized conditions, the ultrasound-assisted reaction also demonstrated a 15 % improvement in the final FAME content compared to the conventional stirring method. These promising results hold significant potential for advancing the field of biodiesel production via ultrasound technology, contributing substantively to sustainable energy sources.

Optimization of Enzymatic Transesterification of Acid Oil for Biodiesel Production Using a Low-Cost Lipase: The Effect of Transesterification Conditions and the Synergy of Lipases with Different Regioselectivity.

This study describes the enzymatic production of second-generation biodiesel using low-quality acid oil as a substrate. Biolipasa-R, a commercially available and low-cost lipase, was employed for enzymatic transesterification. Response surface methodology was applied to optimize the enzymatic transesterification process. The optimal conditions for biodiesel production, which comprised 42% lipase concentration (per weight of oil), 32% water content (per weight of oil), a methanol to oil molar ratio of 3:1, pH 7.0 and reaction temperature 30°C, resulted in the highest fatty acid methyl ester (FAME) content (71.3%). Subsequently, the synergistic effect of two lipases with different regioselectivities under the optimum transesterification conditions was studied, aiming at the enhancement of process efficiency. The transesterification efficiency of immobilized Biolipasa-R was determined and compared to that of Biolipasa-R in its free form. The results revealed a good performance on FAME content (66.5%), while the recycling of immobilized lipase resulted in a decrease in transesterification efficiency after three consecutive uses.

Saccharomyces cerevisiae cellular engineering for the production of FAME biodiesel

The unsustainable and widespread utilization of fossil fuels continues to drive the rapid depletion of global supplies. Biodiesel has emerged as one of the most promising alternatives to conventional diesel, leading to growing research interest in its production. Microbes can facilitate the de novo synthesis of a type of biodiesel in the form of fatty acid methyl esters (FAMEs). In this study, Saccharomyces cerevisiae metabolic activity was engineered to facilitate enhanced FAME production. Initially, free fatty acid concentrations were increased by deleting two acetyl-CoA synthetase genes (FAA1, FAA4) and an acyl-CoA oxidase gene (POX1). Intracellular S-adenosylmethionine (SAM) levels were then enhanced via the deletion of an adenosine kinase gene (ADO1) and the overexpression of a SAM synthetase gene (SAM2). Lastly, the S. cerevisiae strain overproducing free fatty acids and SAM were manipulated to express a plasmid encoding the Drosophila melanogaster Juvenile Hormone Acid O-Methyltransferase (DmJHAMT). Using this combination of engineering approaches, a FAME concentration of 5.79 ± 0.56 mg/L was achieved using these cells in the context of shaking flask fermentation. To the best of our knowledge, this is the first detailed study of FAME production in S. cerevisiae. These results will provide a valuable basis for future efforts to engineer S. cerevisiae strains for highly efficient production of biodiesel.

Possibilities of Utilising Biomass Collected from Road Verges to Produce Biogas and Biodiesel

Grass collected as part of roadside maintenance is conventionally subjected to composting, which has the disadvantage of generating significant CO2 emissions. Thus, it is crucial to find an alternative method for the utilisation of grass waste. The aim of this study was to determine the specific biogas yield (SBY) from the anaerobic mono-digestion of grass from road verges and to assess the content of Fatty Acid Methyl Esters (FAMEs) in grass in relation to the time of cutting and the preservation method of the studied material. The biochemical biogas potential (BBP) test and the FAMEs content were performed on fresh and ensiled grass collected in spring, summer, and autumn. The highest biogas production was obtained from fresh grass cut in spring (715.05 ± 26.43 NL kgVS−1), while the minimum SBY was observed for fresh grass cut in summer (540.19 ± 24.32 NL kgVS−1). The methane (CH4) content in the biogas ranged between 55.0 ± 2.0% and 60.0 ± 1.0%. The contents of ammonia (NH3) and hydrogen sulphide (H2S) in biogas remained below the threshold values for these inhibitors. The highest level of total FAMEs was determined in fresh grass cut in autumn (98.08 ± 19.25 mg gDM−1), while the lowest level was detected in fresh grass cut in spring (56.37 ± 7.03 mg gDM−1). C16:0 and C18:0, which are ideal for biofuel production, were present in the largest amount (66.87 ± 15.56 mg gDM−1) in fresh grass cut in autumn. The ensiling process significantly impacted the content of total FAMEs in spring grass, leading to a reduction in total saturated fatty acids (SFAs) and an increase in total unsaturated fatty acids (USFAs). We conclude that grass biomass collected during the maintenance of road verges is a valuable feedstock for the production of both liquid and gaseous biofuels; however, generating energy from biogas appears to be more efficient than producing biodiesel.

Utilization of Dairy Waste Scum Oil for Microwave-Assisted Biodiesel Production over KOH-Waste Eggshell based Calcium Oxide Catalyst

The sustainability can be maintained by utilizing the available waste as feedstock and catalysts such as dairy and eggshell waste respectively for biodiesel production. In this study, the calcium oxide (CaO) synthesized from calcined eggshell was doped with potassium hydroxide (KOH-ECaO) via wet impregnation method and analyzed the catalyst performance on biodiesel production from dairy waste scum oil (DWSO) via microwave-assisted transesterification. The catalyst was characterized by X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy equipped with Energy Dispersive X-ray (SEM-EDX), Brunauer-Emmett-Teller (BET) and Thermogravimetric analysis (TGA). The fatty acid methyl ester (FAME) contents were deduced by Gas Chromatography-Mass Spectrometry (GC-MS). The KOH-ECaO catalyst shows good potential based on the characterization analysis such as high pore size (25.5 nm) which is supported by SEM pattern analysis. The highest biodiesel production (75%) was obtained at optimum reaction parameters conditions. The optimized conditions were discovered to be 3 wt.% of catalyst, 16:1 of methanol to oil molar ratio, reaction temperature of 65°C and 15 minutes of reaction time as microwave provided faster reaction for the transesterification. These innovative results showed that KOH-ECaO could enhance the biodiesel production from DWSO which encourage the usage of waste for wealth product.

The continuous and increasing use of fuel, causing depleting and non-renewable petroleum reserves, has the potential to cause an economic crisis in the future. With the current depletion of petroleum reserves, it can encourage efforts to find alternative fuels. Biodiesel is a type of alternative fuel that is environmentally friendly and can be renewed because it is made from vegetable oil (biodiesel) mixed with diesel oil. This research was conducted to determine the specifications for the content of Fatty Acid Methyl Ester (FAME) in biodiesel B30 using the Fourier Transform Infrared (FTIR) method produced at the Laboratory of PT. X. The results of the analysis of the Fatty Acid Methyl Ester (FAME) content of BioSolar B30 products at PT. X on March 25, 2021, the fame earned 30%, on April 7, 2021 the FAME content obtained 29% met the specifications (On Specifications) stipulated by the Decree of the Director General of Oil and Gas SK No. 146.K/10/DJM/2020.

Utilization of microalgae for agricultural runoff remediation and sustainable biofuel production through an integrated biorefinery approach

Generally wastewater such agricultural runoff is considered a nuisance; however, it could be harnessed as a potential source of nutrients like nitrates and phosphates in integrated biorefinery context. In the current study, microalgae Chlorella sp. S5 was used for bioremediation of agricultural runoff and the leftover algal biomass was used as a potential source for production of biofuels in an integrated biorefinery context. The microalgae Chlorella sp. S5 was cultivated on Blue Green (BG 11) medium and a comprehensive optimization of different parameters including phosphates, nitrates, and pH was carried out to acquire maximum algal biomass enriched with high lipids content. Dry biomass was quantified using the solvent extraction technique, while the identification of nitrates and phosphates in agricultural runoff was carried out using commercial kits. The algal extracted lipids (oils) were employed in enzymatic trans-esterification for biodiesel production using whole-cell biomass of Bacillus subtilis Q4 MZ841642. The resultant fatty acid methyl esters (FAMEs) were analyzed using Fourier transform infrared (FTIR) spectroscopy and gas chromatography coupled with mass spectrometry (GC–MS). Subsequently, both the intact algal biomass and its lipid-depleted algal biomass were used for biogas production within a batch anaerobic digestion setup. Interestingly, Chlorella sp. S5 demonstrated a substantial reduction of 95% in nitrate and 91% in phosphate from agricultural runoff. The biodiesel derived from algal biomass exhibited a noteworthy total FAME content of 98.2%, meeting the quality standards set by American Society for Testing and Materials (ASTM) and European union (EU) standards. Furthermore, the biomethane yields obtained from whole biomass and lipid-depleted biomass were 330.34 NmL/g VSadded and 364.34 NmL/g VSadded, respectively. In conclusion, the findings underscore the potent utility of Chlorella sp. S5 as a multi-faceted resource, proficiently employed in a sequential cascade for treating agricultural runoff, producing biodiesel, and generating biogas within the integrated biorefinery concept.Graphical

Increasing lipid production in Chlamydomonas reinhardtii through genetic introduction for the overexpression of glyceraldehyde-3-phosphate dehydrogenase.

Microalgae, valued for their sustainability and CO2 fixation capabilities, are emerging as promising sources of biofuels and high-value compounds. This study aimed to boost lipid production in C. reinhardtii by overexpressing chloroplast glyceraldehyde-3-phosphate dehydrogenase (GAPDH), a key enzyme in the Calvin cycle and glycolysis, under the control of a nitrogen-inducible NIT1 promoter, to positively impact overall carbon metabolism. The standout transformant, PNG#7, exhibited significantly increased lipid production under nitrogen starvation, with biomass rising by 44% and 76% on days 4 and 16, respectively. Fatty acid methyl ester (FAME) content in PNG#7 surged by 2.4-fold and 2.1-fold, notably surpassing the wild type (WT) in lipid productivity by 3.4 and 3.7 times on days 4 and 16, respectively. Transcriptome analysis revealed a tenfold increase in transgenic GAPDH expression and significant upregulation of genes involved in fatty acid and triacylglycerol synthesis, especially the gene encoding acyl-carrier protein gene (ACP, Cre13. g577100. t1.2). In contrast, genes related to cellulose synthesis were downregulated. Single Nucleotide Polymorphism (SNP)/Indel analysis indicated substantial DNA modifications, which likely contributed to the observed extensive transcriptomic and phenotypic changes. These findings suggest that overexpressing chloroplast GAPDH, coupled with genetic modifications, effectively enhances lipid synthesis in C. reinhardtii. This study not only underscores the potential of chloroplast GAPDH overexpression in microalgal lipid synthesis but also highlights the expansive potential of metabolic engineering in microalgae for biofuel production.

Development of a fast GC-response factor method to quantify total FAMEs in biodiesel from various fatty acid sources

Biodiesel is a renewable energy source that can be employed as an alternative fuel to fossil diesel. Monitoring samples in terms of fatty acid methyl esters (FAMEs) content using gas chromatography (GC) is a useful way to evaluate the quality of biodiesel produced. Fast GC can be used to reduce the time of analysis and consequently the costs, even though the equipment required for this may be expensive. Therefore, we developed and validated a Fast GC-response factor (Fast-GCRF) methodology using a low-priced nonpolar column, flame ionization detection (FID) and a standard chromatograph usually destined for conventional GC separations. The method was validated and applied to the quantification of FAMEs produced from various raw materials (soybean, cottonseed, canola, coconut, palm, sesame, sunflower, castor, corn, macauba, almond, peanut, chia seeds, residual frying oils, and chicken fat), being both selective and robust even when subjected to changes in sample preparation. The Fast GCRF method presented respective maximum relative standard deviation (RSD) values for instrumental precision, repeatability, and intermediate precision of 2.3 %, 3.6 %, and 2.8 %. When compared to conventional GC methodologies, accurate results with high degrees of agreement were obtained using the Fast-GCRF method, within an at least four times shorter analysis time.

Evaluating nutrient limitation in co-culture of Chlorella pyrenoidosa and Rhodobacter sphaeroides

The influence of nitrogen deficiency on microalgae-bacteria co-culture has been studied mostly with nitrogen-fixing bacteria. Photosynthetic bacteria (PSB), which are non-nitrogen-fixing bacteria, the impact of N deficiency on its co-culture with microalgae is unknown. In this study, Chlorella pyrenoidosa and Rhodobacter sphaeroides co-culture was cultivated photoheterotrophically with acetate. The impact of N starvation and different P supply levels on oil production were examined. When phosphorus was sufficient, N starvation increased the fatty acid methyl ester (FAME) content from 21.7 % to 28.2 %, and also increased the FAME yield (g CODFAME/g CODAcetate) from 0.17 to 0.22. However, the biomass and FAME productivities decreased. Sufficient phosphorus was also essential for a high growth rate and FAME productivity. Deficiencies in either N or P led to a decrease in the proportion of unsaturated FAMEs. iTRAQ analysis indicated N starvation promoted oil accumulation by driving the carbon flow to fatty acid synthesis in microalgae from co-culture. This study improves the understanding of biomass and lipid production via microalgae-PSB co-culture in photoheterotrophic cultivation. The mechanism of interaction between microalgae and bacteria needs further study.

A bio-augmented system with Methylosarcina sp. LC-4 immobilized on bio-carriers: Towards an integrated approach to mitigate and valorize methane emissions from landfills to biodiesel

Bio-augmented systems based on methanotrophs are indispensable in curbing anthropogenic methane emissions from engineered landfills or dumpsites to curtail rising levels of greenhouse gases. Using a defined methanotroph culture immobilized on an inert material-based bio-carrier makes it possible to harness these methane emissions for creating value-added products, thus contributing to the circular bio-economy. The methane oxidation capacity of the model methanotroph Methylosarcina sp. LC-4, a prospective organism for biodiesel production using methane present in landfill gas, immobilized on several inert bio-carriers, was evaluated to identify a bio-carrier that provided optimum conditions for the process.Among the several bio-carriers evaluated, perlite and vermiculite were selected due to their high specific surface area and superior water-holding capacity, which result in the retention of nutrients and biomass and higher methane elimination capacity. While perlite showed high biomass holding capacity and methane transport, vermiculite supported a high growth of methanotrophs. LC-4 immobilized on perlite and vermiculite as the bio-carrier showed maximum methane elimination capacity (MEC) of 291.3 g m−2 day−1 and 155.5 g m−2 day−1, respectively. The low bed height of only 0.13 m and a short start-up period of 2–4 days are promising for use as alternate daily cover in a landfill. The recovered biomass had 12% (w/w) fatty acid methyl ester (FAME), with a high fraction of (∼85%) of C14–C18 saturated and monounsaturated fatty acids, suitable for biodiesel production. The combination of perlite and vermiculite increased MEC and FAME content levels. The current study demonstrated a new bio-augmented system designed with a pure methanotroph for methane elimination with a short start-up time and the valorization of the assimilated methane.

Rhizopus stolonifer biomass catalytic transesterification capability: optimization of cultivation conditions

BackgroundUsing fungal biomass for biocatalysis is a potential solution for the expensive cost of the use o enzymes. Production of fungal biomass with effective activity requires optimizing the cultivation conditions.ResultsRhizopus stolonifer biomass was optimized for transesterification and hydrolysis of waste frying oil (WFO). Growth and biomass lipolytic activities of R. stolonifer improved under shaking conditions compared to static conditions, and 200 rpm was optimum. As biomass lipase and transesterification activities inducer, olive oil was superior to soybean, rapeseed, and waste frying oils. Biomass produced in culture media containing fishmeal as an N-source feedstock had higher lipolytic capabilities than corn-steep liquor and urea. Plackett Burman screening of 9 factors showed that pH (5–9), fishmeal (0.25–1.7%, w/v), and KH2PO4 (0.1–0.9%, w/v) were significant factors with the highest main effect estimates 11.46, 10.42, 14.90, respectively. These factors were selected for response surface methodology (RSM) optimization using central composite design (CCD). CCD models for growth, biomass lipase activity, and transesterification capability were significant. The optimum conditions for growth and lipid modification catalytic activities were pH 7.4, fishmeal (2.62%, w/v), and KH2PO4 (2.99%, w/v).ConclusionOptimized culture conditions improved the whole cell transesterification capability of Rhizopus stolonifer biomass in terms of fatty acid methyl ester (FAME) concentration by 67.65% to a final FAME concentration of 85.5%, w/w.

An experimental evaluation of cetane improving techniques for enhancing the performance and emission trade-off in diesel engine: A comparative study

The polyunsaturated fatty acid methyl ester concentration in Prosopis juliflora biodiesel drops the cetane number and affects ignition quality. In this study, two fuel reformulation strategies, viz. partial hydrogenation and the inclusion of Di-tert-butyl peroxide (DTBP), are investigated for their ability to improve the combustion and performance of a diesel–biodiesel (B20) blend without compromising the emissions trade-off. An autoclave reactor utilizes partially hydrogenated juliflora biodiesel and gas chromatography to determine the altered fuel composition. The cetane improver is DTBP, which is mixed with B20 at a concentration of 2000 ppm. The filterability of the test fuel was evaluated using the Tamson filter blocking tendency apparatus. Engine parameters and emissions of the base diesel (D0), JB20, HJB20, and JB20 + DTBP test fuels were examined in a diesel engine. According to the FBT results, JB20 (1.36) and HJB20 (1.28) have good filterability, which is consistent with the ASTM standard (D2068-14). While comparing JB20 blend with modified JB20 fuels decreases NOx (up to 5.24%) and increases brake thermal efficiency (7.5%) at full load. Compared to diesel fuel, partially hydrogenated blend fuel emissions viz. HC, CO, and smoke were reduced by 15.24%, 7.76%, and 10.14%, respectively. Based on the above results, partial hydrogenation is more beneficial than the inclusion of DTBP in enhancing biodiesel's trade-off qualities.

Biodiesel Production from the Marine Alga Nannochloropsis oceanica Grown on Yeast Wastewater and the Effect on Its Biochemical Composition and Gene Expression.

Microalgae-based biodiesel synthesis is currently not commercially viable due to the high costs of culture realizations and low lipid yields. The main objective of the current study was to determine the possibility of growing Nannochloropsis oceanica on Saccharomyces cerevisiae yeast wastewater for biodiesel generation at an economical rate. N. oceanica was grown in Guillard F/2 synthetic medium and three dilutions of yeast wastewater (1, 1.25, and 1.5%). Biodiesel properties, in addition to carbohydrate, protein, lipid, dry weight, biomass, lipid productivity, amino acids, and fatty acid methyl ester (FAMEs) content, were analyzed and the quality of the produced biodiesel is assessed. The data revealed the response of N. oceanica to nitrogen-deficiency in the three dilutions of yeast wastewater. N. oceanica in Y2 (1.25%) yeast wastewater dilution exhibited the highest total carbohydrate and lipid percentages (21.19% and 41.97%, respectively), and the highest lipid productivity (52.46 mg L-1 day -1) under nitrogen deficiency in yeast wastewater. The fatty acids profile shows that N. oceanica cultivated in Y2 (1.25%) wastewater dilution provides a significant level of TSFA (47.42%) and can be used as a feedstock for biodiesel synthesis. In addition, N. oceanica responded to nitrogen shortage in wastewater dilutions by upregulating the gene encoding delta-9 fatty acid desaturase (Δ9FAD). As a result, the oleic and palmitoleic acid levels increased in the fatty acid profile of Y2 yeast wastewater dilution, highlighting the increased activity of Δ9FAD enzyme in transforming stearic acid and palmitic acid into oleic acid and palmitoleic acid. This study proved that the Y2 (1.25%) yeast wastewater dilution can be utilized as a growth medium for improving the quantity of specific fatty acids and lipid productivity in N. oceanica that affect biodiesel quality to satisfy global biodiesel requirements.

Box–Behnken optimization of biodiesel production using trisodium phosphate catalyst from monazite ore processes

AbstractTrisodium phosphate (TSP) is a non‐toxic waste obtained from alkaline cracking as part of the processing of monazite ore. After simple purification, TSP can be utilized in many applications; however, it is used as a catalyst in this study. It is characterized using Thermogravimetric analysis , X‐ray diffraction analysis, Brunauer‐Emmett‐Teller, and Temperature‐programmed desorption of carbon dioxide and optimized in the transesterification reaction. After recrystallization and calcination at 350 °C for 0.5 h, the form of the TSP was tetragonal, with a 2.61 m2/g surface area and 537.4 x 10‐6 mol/g basic sites. Trisodium phosphate can therefore be utilized effectively as a solid base catalyst for the transesterification of palm oil. Three biodiesel production parameters were investigated: the methanol to oil molar ratio (6:1–24:1), catalyst concentration (1–10%), and reaction time (30 to 240 min). The experiments were designed using the Box–Behnken response surface method. As a result, the optimum conditions for biodiesel production are a 22:1 methanol to oil molar ratio, 4% catalyst concentration, and 200 min reaction time. The optimal fatty acid methyl ester (FAME) content is 99.79%, with an error between actual and predicted FAME of 0.21%. © 2023 Society of Industrial Chemistry and John Wiley & Sons Ltd.

Enhancing Lipid Production of Chlorella sp. by Mixotrophic Cultivation Optimization

Mixotrophic microalgal cultivation can utilize CO2 and organic carbon sources. This study optimized the cultivation nitrogen source (peptone, urea, yeast extract, NH4Cl, (NH4)2SO4, NH4NO3, NaNO3 and KNO3), carbon combination (glucose, glycerol, sucrose), (NH4)2SO4 nitrogen source and pH (6–11) in a local microalgae species with three characteristics (high pH-resistant, high growth rate and high lipid content). Chlorella sp. G3H3-1-2 biomass production and lipid accumulation were estimated using the fatty acid methyl esters (FAMEs) concentration. The Chlorella sp. G3H3-1-2 FAME content was strongly influenced by the carbon, nitrogen sources and pH variations. The pH ranged from 6.0 to 8.0, which produced the highest specific growth rate of 1.22 day−1 for Chlorella sp. G3H3-1-2 while using glucose as the single carbon source. However, the highest total FAMEs content of 59% in the Chlorella sp. G3H3-1-2 biomass of 1.75 g/L was obtained while using the combination of 1 g-glucose/L as the carbon source and 0.2 g-(NH4)2SO4/L as the nitrogen source at the high pH value of 10.

Urban sewage sludge valorization to biodiesel production: Solvent-free lipid recovery through adsorption on used 3 Ply Safety Face Masks

The use of urban sewage sludge as the feedstock of lipids to produce biodiesel is a topic of great interest and potential. However, the extractive step, conventionally operated with organic solvents, makes this use unfeasible and with a high environmental impact. In this work, for the first time, the solvent-free recovery of lipids from sewage sludge was studied through adsorption onto non-polar polymeric materials (Spunbonded nonwoven Polypropylene (s-nwPP)). After being immersed in sedimented sewage sludge (TS: 6.9 wt%), s-nwPP sheets were easily separated, dried and analyzed: sewage sludge’s lipids were preferentially adsorbed onto the polymeric surface. Free fatty acids (FFAs) were the dominant species and were recovered from s-nwPP sheets through acidic methanol in a separated reactive washing step as fatty acid methyl esters (FAMEs) A crude product was obtained through this sequence of adsorption and reactive desorption operations, with a FAMEs content of over 55 wt%. After preliminary investigations on commercial s-nwPP sheets, 3 Ply Safety Face Masks were also positively tested. pH, weight ratio polymer/wet sludge (R) and temperature were the parameters optimized: operating at 343 K under acidic conditions (pH of sludge<2), with an R equal to 6%, a total FFAs recovery yield of 70% as FAMEs was already achieved after two adsorption cycles. A preliminary evaluation of the economic feasibility demonstrated that used PP surgical facial masks allow the recovery and generation of crude biodiesel from sewage sludge at a very reasonable price with minimal environmental impacts.

Density Measurements and PC-SAFT Modeling for Beef Tallow Fatty Acid Methyl Esters (FAME Mixture) + Alkane up to 70 MPa

Biodiesel is mainly constituted by fatty acid alkyl esters, and the effect of adding alkane compounds is of great importance in order to analyze the changes in the thermophysical properties under high pressure that is of relevance in diesel engines. This work is aimed at the study of density behavior of two systems formed by beef tallow fatty acid methyl esters (FAME mixture) + alkane (decane or dodecane) in the temperature range of 303–393 K and in the pressure range of 0.2–68.29 MPa. Experiments were carried out by means of two vibrating tube densimeters with an expanded uncertainty in density of 0.7 kg·m–3. Transesterification of waste beef tallow by using methanol at supercritical conditions was used to obtain biodiesel. The FAME mixture was constituted by methyl tetradecanoate, methyl hexadecanoate, methyl octadecenoate, and methyl (Z)-octadec-9-enoate. Density behavior was correlated using the Tammann–Tait equation and modeled with the perturbed-chain statistical associating fluid theory (PC-SAFT) equation of state. Density correlation deviations using the Tammann–Tait equation were found to be less than 0.05% and 4.15 kg·m–3 for the average absolute deviation (AAD) and standard deviation (STD), respectively. The PC-SAFT modeling yielded an AAD = 0.11% and an STD = 1.15 kg·m–3. The behavior for density and the derived thermodynamic properties were explained in terms of the fatty acid methyl ester content.

Biodiesel Production Catalyzed by Lipase Extract Powder of Leonotis nepetifolia (Christmas Candlestick) Seed

This work aimed to evaluate the ability of lipase extract powder obtained from Leonotis nepetifolia seed for enzyme-catalyzed biodiesel production using Leonotis nepetifolia oil, commercial olive oil, and waste cooking oil as substrates. The lipase extract powder showed an enzymatic activity and hydrolysis percentage of 24.7 U/g and 21.31%, respectively, using commercial olive oil as a reference. Transesterification reaction conditions were 40 g of substrate, 34 °C, molar ratio oil: methanol of 1:3, lipase extract powder 20 wt%, phosphates buffer (pH 4.8) 20 wt%, and a reaction time of 8 h. Transesterification yields of 74.5%, 71.5%, and 69.3% for commercial olive oil, waste cooking oil, and Leonotis nepetifolia oil were obtained, respectively. Biodiesel physicochemical parameters were analyzed and compared with the international standards: EN 14214 (European Union) and ASTM D6751 (American Society for Testing and Materials). The biodiesel’s moisture and volatile matter percentages, iodine index, cooper strip corrosion, and methyl esters content conformed to the standards’ specifications. The fatty acid methyl ester content of the vegetable oils showed the presence of methyl oleate after enzyme-catalyzed transesterification. This study reveals that biodiesel production catalyzed by lipase extract powder from Leonotis nepetifolia could be a viable alternative, showing that transesterification yields competitive results.

Spent coffee grounds and orange peel residues based biorefinery for microbial oil and biodiesel conversion estimation

A consolidated biorefinery was developed using spent coffee grounds (SCGs) and orange peel residues (OPR) to produce microbial oil and value-added co-products. Coffee oil and antioxidant-rich extracts were initially extracted from SCGs. Free sugars and pectin-rich extracts were obtained from OPR. Microbial oil production was evaluated using various oleaginous yeast strains cultivated in commercial carbon sources. Microbial oil production was demonstrated in 30 L bioreactor scale using combined OPR-SCGs hydrolysate resulting in 56.7% (w/w) intracellular lipid accumulation and 0.32 g/(L∙h) productivity, corresponding to 68.1 g microbial oil production per 1 kg SCGs and 360 g OPR. Principal Component Analysis has been carried out to assess the similarity of microbial oil produced in this study and literature-cited oils considering the fatty acid methyl ester content. The biodiesel properties estimated based on predictive equations showed that they generally conformed with the limits set by European standards.
