Long-term intermittent theta burst stimulation enhanced hippocampus-dependent memory by regulating hippocampal theta oscillation and neurotransmitter levels in healthy rats

Intermittent theta burst stimulation (iTBS), an updated pattern of high-frequency repetitive transcranial magnetic stimulation, is a potential candidate for improving memory. The hippocampus has been shown to be involved in the memory-enhancing effect induced by iTBS. However, it remains largely unknown whether this effect is achieved by regulating hippocampal theta oscillation and neurotransmitters gamma-aminobutyric acid (GABA) and glutamate, which are strongly related to memory. Thus, we investigated the effect of 14 days of iTBS on hippocampus-dependent memory and further explored the roles of hippocampal theta oscillation and neurotransmitters GABA and glutamate in this effect. We found that compared to sham iTBS, real iTBS enhanced hippocampus-dependent memory measured by hole-board test and object place recognition test. Further, real iTBS increased the density of c-Fos positive neurons and normalized power of theta oscillation in the dorsal hippocampus (dHip) compared to sham iTBS. Interestingly, we observed a decrease in the level of extracellular GABA and an increase in the level of extracellular glutamate in the dHip after real iTBS. Our results suggest that long-term iTBS improved hippocampus-dependent memory, which may be attributed to the enhancement of theta oscillation and altered levels of extracellular GABA and glutamate in the dHip.

Effect of cortical extracellular GABA on motor response.

To elucidate how the flattening of sensory tuning due to a deficit in tonic inhibition slows motor responses, we simulated a neural network model in which a sensory cortical network ([Formula: see text]) and a motor cortical network ([Formula: see text]) are reciprocally connected, and the [Formula: see text] projects to spinal motoneurons (Mns). The [Formula: see text] was presented with a feature stimulus and the reaction time of Mns was measured. The flattening of sensory tuning in [Formula: see text] caused by decreasing the concentration of gamma-aminobutyric acid (GABA) in extracellular space resulted in a decrease in the stimulus-sensitive [Formula: see text] pyramidal cell activity while increasing the stimulus-insensitive [Formula: see text] pyramidal cell activity, thereby prolonging the reaction time of Mns to the applied feature stimulus. We suggest that a reduction in extracellular GABA concentration in sensory cortex may interfere with selective activation in motor cortex, leading to slowing the activation of spinal motoneurons and therefore to slowing motor responses.

A stable naked-eye colorimetric sensor for monitoring release of extracellular gamma-aminobutyric acid (GABA) neurotransmitter from SH-SY5Y cells

A novel optical γ-aminobutyric acid (GABA)-based sensor was developed on interacting thiol compounds and o-phthalaldehyde (OPA) to form thiacetal compounds. Then, the thiacetal interacts with the GABA molecule to form an isoindole compound. The effects of four thiol compounds on the stability of the resulting isoindole compound were assessed. The 2-mercaptoethanol, “one of the most used derivatizing agents,” is unexpectedly the least stable; while, 16-mercaptohexadecanoic acid resulted in the most durable isoindole compound. The developed sensor showed the capability for detecting GABA within a wide concentration range spanning from 500 nmol L–1 to 100 µmol L–1. The detection limit was about 330 nmol L–1, which indicated the high sensitivity of the developed sensor compared with those previously reported. The findings illustrated the ability to detect GABA at the physiological pH (pH = 7.4) without adjusting the pH value, opening the door for real applications. Furthermore, the sensor could detect various GABA concentrations in human serum with good recovery percentages (98% to 101.4%). In addition, this assay was applied to monitor GABA release from the SH-SY5Y cell line to convert glutamate into GABA. This result indicates the capability of the proposed assay for visually monitoring the release of GABA neurotransmitters.

Cell surface display of Neurospora crassa glutamate decarboxylase on Escherichia coli for extracellular Gamma-aminobutyric acid production from high cell density culture

GABA is produced from the decarboxylation of glutamate by glutamate decarboxylase enzyme (GadB). Various hetrologous GadB enzymes has been indentified and introduced in Escherichia coli to improve the productivity of GABA. In this study, cell surface display strategy was employed by displaying Neurospora crassa glutamate decarboxylase (GadB) on the surface of Escherichia coli using OmpC as an anchoring motif. To construct the display system, the GadB was fused to truncated C-terminus of OmpC and expressed in E. coli. The recombinant E. coli displaying GadB has produced 3.03 g/L of GABA from 10 g/L glutamate within 12 h of culture period. When the GadB display system was cultured at high cell density, 100% GABA yeild was achieved from 50 g/L of glutamate. The results suggest that the GadB expression on the E. coli surface is stable and effective on GABA production. The surface display strategy can be applied on other potential enzyme candidates and used in Industry for bulk chemical production.

Rapid quantification of extracellular neurotransmitters in mouse brain by PESI/MS/MS and longitudinal data analysis using the R and Stan-based Bayesian state-space model

We developed a methodology for rapid quantification of extracellular neurotransmitters in mouse brain by PESI/MS/MS and longitudinal data analysis using the R and Stan-based Bayesian state-space model. We performed a rapid analysis for quantifying extracellular l-glutamic acid (L-Glu) and gamma-aminobutyric acid (GABA) in the mouse striatum by combined use of probe electrospray ionization/tandem mass spectrometry (PESI/MS/MS) and in vivo brain microdialysis. We optimized the PESI/MS/MS parameters with the authentic L-Glu, GABA, L-Glu-13C5,15N1, and GABA-D6 standards. We constructed calibration curves of L-Glu and GABA with the stable isotope internal standard correction method (L-Glu-13C5,15N1, and GABA-D6), demonstrating sufficient linearity (R > 0.999). Additionally, the quantitative method for L-Glu and GABA was validated with low-, middle-, and high-quality control samples. The intra- and inter-day accuracy and precision were 0.4%–7.5% and 1.7%–5.4% for L-Glu, respectively, and 0.1%–4.8% and 2.1%–5.7% for GABA, respectively, demonstrating high reproducibility of the method. To evaluate the feasibility of this method, microdialyses were performed on free-moving mice that were stimulated by high-K+-induced depolarization under different sampling conditions: 1) every 5 min for 150 min (n = 2) and 2) every 1 min for 30 min (n = 3). We applied the R and Stan-based Bayesian state-space model to each mouse's time-series data considering autocorrelation, and the model successfully detected abnormal changes in the L-Glu and GABA levels in each mouse. Thus, the L-Glu and GABA levels in all microdialysates approximately increased up to two- and seven-fold levels through high-K+-induced depolarization. Additionally, a 1-min temporal resolution was achieved using this method, thereby successfully monitoring microenvironmental changes in the extracellular L-Glu and GABA of the mouse striatum. In conclusion, this methodology using PESI/MS/MS and Bayesian state-space model allowed easy monitoring of neurotransmitters at high temporal resolutions and appropriate data interpretation considering autocorrelation of time-series data, which will reveal hidden pathological mechanisms of brain diseases, such as Parkinson's disease and Huntington's disease in the future.

Adolescent Exposure to WIN 55212-2 Render the Nigrostriatal Dopaminergic Pathway Activated During Adulthood.

BackgroundDuring adolescence, neuronal circuits exhibit plasticity in response to physiological changes and to adapt to environmental events. Nigrostriatal dopaminergic pathways are in constant flux during development. Evidence suggests a relationship between early use of cannabinoids and psychiatric disorders characterized by altered dopaminergic systems, such as schizophrenia and addiction. However, the impact of adolescent exposure to cannabinoids on nigrostriatal dopaminergic pathways in adulthood remains unclear. The aim of this research was to determine the effects of repeated activation of cannabinoid receptors during adolescence on dopaminergic activity of nigrostriatal pathways and the mechanisms underlying this impact during adulthood.MethodsMale Sprague-Dawley rats were treated with 1.2 mg/kg WIN 55212-2 daily from postnatal day 40 to 65. Then no-net flux microdialysis of dopamine in the dorsolateral striatum, electrophysiological recording of dopaminergic neuronal activity, and microdialysis measures of gamma-aminobutyric acid (GABA) and glutamate in substantia nigra par compacta were carried out during adulthood (postnatal days 72–78).ResultsRepeated activation of cannabinoid receptors during adolescence increased the release of dopamine in dorsolateral striatum accompanied by increased population activity of dopamine neurons and decreased extracellular GABA levels in substantia nigra par compacta in adulthood. Furthermore, perfusion of bicuculline, a GABAa antagonist, into the ventral pallidum reversed the increased dopamine neuron population activity in substantia nigra par compacta induced by adolescent cannabinoid exposure.ConclusionsThese results suggest that adolescent exposure to cannabinoid agonists produces disinhibition of nigrostriatal dopamine transmission during adulthood mediated by decreased GABAergic input from the ventral pallidum.

Dynamic Cerebral Microdialysis during Pallidotomy and Thalamotomy in Parkinson’s Disease: A Preliminary Neurochemical Study

In Parkinson’s disease (PD), dopaminergic neurons reduce the regulation of glutamatergic (glutamate-Glu) input from the cortex to neostriatum (caudate and putamen nuclei) consequently leading to a hyperactivity of globus pallidus internae (GPi) neurons that release gamma-amino-butyric acid (GABA) into the thalamic ventrolateral (VL) nucleus. The objective of the present experiment was to measure changes in GABA and Glu in the caudate and the thalamus of 2 patients during the application of electrical stimuli following either a pallidotomy or a thalamotomy. Proper insertion of the electrode was tested by applying high frequency electrical pulses (HFEP). During these procedures, we obtained neurochemical information placing cerebral (CMD) microdialysis probes in caudate nucleus and VL nucleus of ipsi- and contra-lateral thalamus. In VL thalamus, extracellular GABA decreased during HFEP, tending to reach previous levels once HFEP was finalized. Following the pallido- or thalamotomy GABA decreased again. Similarly, in the contralateral VL thalamus, extracellular GABA levels showed a similar but less pronounced profile but did not show any decrement after the lesion. Caudate Glu decreases when HFEP is applied to the GPi and recovers to previous levels after HFEP, but did not decrease again after lesion (GPi-tomy), instead it continued to rise. These results suggest that HFEP exerts a similar but reversible biochemical effect as thermopallido- or thermothalamotomy on GABA extracellular concentration in the ipsilateral VL thalamus. We also observe a distant effect of HFEP, but not of thermolesion, on contralateral thalamic GABA and ipsilateral caudate Glu.

Challenges of simultaneous measurements of brain extracellular GABA and glutamate in vivo using enzyme-coated microelectrode arrays

BackgroundAlthough GABA is the major inhibitory neurotransmitter in the CNS, quantifying in vivo GABA levels has been challenging. The ability to co-monitor both GABA and the major excitatory neurotransmitter, glutamate, would be a powerful tool in both research and clinical settings. New methodCeramic-based microelectrode arrays (MEAs) were used to quantify gamma-aminobutyric acid (GABA) by employing a dual-enzyme reaction scheme including GABase and glutamate oxidase (GluOx). Glutamate was simultaneously quantified on adjacent recording sites coated with GluOx alone. Endogenous glutamate was subtracted from the combined GABA and glutamate signal to yield a pure GABA concentration. ResultsElectrode sensitivity to GABA in conventional, stirred in vitro calibrations at pH 7.4 did not match the in vivo sensitivity due to diffusional losses. Non-stirred calibrations in agarose or stirred calibrations at pH 8.6 were used to match the in vivo GABA sensitivity. In vivo data collected in the rat brain demonstrated feasibility of the GABA/glutamate MEA including uptake of locally applied GABA, KCl-evoked GABA release and modulation of endogenous GABA with vigabatrin. Comparison with existing methodsImplantable enzyme-coated microelectrode arrays have better temporal and spatial resolution than existing off-line methods. However, interpretation of results can be complicated due to the multiple recording site and dual enzyme approach. ConclusionsThe initial in vitro and in vivo studies supported that the new MEA configuration may be a viable platform for combined GABA and glutamate measures in the CNS extending the previous reports to in vivo GABA detection. The challenges of this approach are emphasized.

ANGIOTENSIN II TYPE 1 RECEPTOR MEDIATED INHIBITORY MODULATION OF THE BAROREFLEX BY ANGIOTENSIN II INVOLVES NITRIC OXIDE DEPENDENT GABA RELEASE IN THE NUCLEUS TRACTUS SOLITARII

Objective: It is well established that angiotensin II exerts a dampening effect on the baroreflex within the nucleus tractus solitarii, the principal brainstem site for termination of baroreceptor afferents and which is densely populated with gamma-aminobutyric acid-ergic neurons and nerve terminals. The present study was designed to investigate whether local release of gamma-aminobutyric acid is involved in the effects mediated by local angiotensin II within the nucleus tractus solitarii. Design and method: In vivo microdialysis was used for measurement of extracellular glutamate and gamma-aminobutyric acid levels and for infusion of angiotensin II and other compounds within the nucleus tractus solitarii of normotensive Wistar rats. The mean arterial pressure and heart rate responses were monitored with a pressure transducer. Angiotensin II type 1 receptor selectivity was assessed using the angiotensin II type 1 receptor antagonist candesartan; the nitric oxide synthase inhibitor was used to assess the involvement of nitric oxide in the evoked responses by infusion of angiotensin II. Results: Local infusion of angiotensin II into the nucleus tractus solitarii significantly increased extracellular gamma-aminobutyric acid levels as well as mean arterial pressure. These responses were both abolished by co-infusion of either the angiotensin II type 1 receptor antagonist candesartan, or the nitric oxide synthase inhibitor, N (ω)-nitro-L-arginine methyl ester. The pressor response to angiotensin II was reversed by co-infusion with the gamma-aminobutyric acid A receptor antagonist, bicuculline. Local blockade of nitric oxide synthase decreased both, gamma-aminobutyric acid and glutamate concentrations. Conclusions: Our results suggest that the inhibitory modulation of the baroreflex by angiotensin II involves angiotensin II type 1 receptor mediated nitric oxide dependent local gamma-aminobutyric acid release within the nucleus tractus solitarii in normotensive rats. Nitric oxide produced within the nucleus tractus solitarii tonically potentiates local gamma-aminobutyric acid and glutamate release.

Investigation of the Role of AT2 Receptors in the Nucleus Tractus Solitarii of Normotensive Rats in Blood Pressure Control.

AimThe nucleus tractus solitarii (NTS) densely expresses angiotensin II type 2 receptors (AT2R), which are mainly located on inhibitory gamma-aminobutyric acid (GABA) neurons. Central AT2R stimulation reduces blood pressure, and AT2R stimulation in the rostral ventrolateral medulla (RVLM), mediates a hypotensive response through a GABAergic mechanism. We aimed to test the hypothesis that an AT2R mediated inhibition of the GABA release within the NTS might be involved in this hypotensive response, by assessing possible alterations in blood pressure and heart rate, as well as in GABA levels in normotensive Wistar rats.MethodsIn vivo microdialysis was used for measurement of extracellular GABA levels and for perfusion of the selective AT2R agonist, Compound 21, within the NTS. Our set-up allowed to determine simultaneously the excitatory glutamate dialysate levels. The mean arterial pressure and heart rate responses were monitored with a pressure transducer.ResultsLocal perfusion of Compound 21 into the NTS did not modify blood pressure and heart rate, nor glutamate and GABA levels compared to baseline concentrations. A putative effect was also not unmasked by concomitant angiotensin II type 1 receptor blockade with candesartan. Positive control experiments confirmed that the experimental set up had enough sensitivity to detect a reduction in GABA dialysate levels and blood pressure.ConclusionThe results did not provide evidence for a role of the AT2R within the NTS in the control of blood pressure, nor for an interaction with local GABAergic signaling in normotensive rats.

AT1 Receptor Mediated Hypertensive Response to Ang II in the Nucleus Tractus Solitarii of Normotensive Rats Involves NO Dependent Local GABA Release.

The angiotensin II type 1 receptor (AT1R) antagonist, candesartan, was used to assess whether responses were AT1R dependent and the nitric oxide (NO) synthase inhibitor, N(ω)-nitro-L-arginine methyl ester (L-NAME), was used to assess the involvement of NO in the evoked responses by infusion of angiotensin II. The MAP and heart rate responses were monitored with a pressure transducer.ResultsLocal infusion into the NTS of angiotensin II induced a significant to ninefold significantly increase in extracellular GABA levels; as well as MAP was increased by 15 mmHg. These responses were both abolished by co-infusion of either, the angiotensin II type 1 receptor antagonist, candesartan, or the NO synthase inhibitor, L-NAME, demonstrating that the effect is not only AT1R dependent but also NO dependent. The pressor response to angiotensin II was reversed by co-infusion with the GABAA receptor antagonist, bicuculline. Local blockade of NO synthase decreased both, GABA and glutamate concentrations.ConclusionOur results suggest that the AT1R mediated hypertensive response to angiotensin II within the NTS in normotensive rats is GABA and NO dependent. Nitric oxide produced within the NTS tonically potentiates local GABA and glutamate release.

Gene mutations in paediatric epilepsies cause NMDA-pathy, and phasic and tonic GABA-pathy.

The aim of this study was to disentangle mechanisms of epileptogenesis in monogenic epilepsies in children. We reviewed paediatric monogenic epilepsies excluding brain malformation or an inborn error of metabolism, but including the gene function whether there is loss-of-function or gain-of-function, age at gene expression when available, and associated epilepsy syndrome. Genes for which at least five patients with similar epilepsy phenotype had been reported were selected. Three mechanisms are shared by most monogenic epilepsies: (1) excess of N-methyl-d-aspartate (NMDA) transmission activation (NMDA-pathies); (2)abnormal gamma-aminobutyric acid (GABA) transmission with reduced inhibition (phasic GABA-pathies); and (3) tonic activation of extrasynaptic GABAA receptors by extracellular GABA (tonic GABA-pathies). NMDA-pathies comprise early epileptic encephalopathy with suppression-burst, neonatal/infantile benign seizures, West and Lennox-Gastaut syndromes, and encephalopathy with continuous spike waves in slow sleep, thus brief seizures with major interictal spiking. Phasic GABA-pathies comprise mostly generalized epilepsy with febrile seizures plus and Dravet syndrome, thus long-lasting seizures with mild interictal spiking. Tonic GABA-pathies cause epilepsy with myoclonic-atonic seizures and Angelman syndrome, thus major high-amplitude slow-wave activity. This pathophysiological approach to monogenic epilepsies provides diagnostic clues and helps to guide treatment strategy. WHAT THIS PAPER ADDS: In paediatric monogenic epilepsies, electroclinical patterns point to three main mechanisms: NMDA-pathies, and phasic and tonic GABA-pathies. Antiepileptic treatment choice could be guided by each of these mechanisms.

Hyperbaric oxygen (HBO2) causes inhibition of gamma-aminobutyric acid (GABA) synthesis in the brain, which leads to a decrease in GABAergic neurotransmission and the development of convulsive syndrome like epileptic seizures. We tested the hypothesis that inhibition of GABA transporters can compensate a decrease in GABA synthesis, enhance GABAergic transmission, and prevent against oxygen seizures. HBO2-induced seizures were analyzed in awake rats pretreated with GABA transporter inhibitors (NO-711, nipecotic acid) by intracerebroventricular injection and exposed to oxygen at 5 ATA (absolute atmospheres). In a separate group of rats, GABA was measured in the striatum using intracerebral microdialysis coupled with liquid chromatography. Inhibition of neuronal and glial GABA transporters increased extracerebral GABA content in the brain and protected against HBO2-induced seizures. A more effective anti-convulsive effect was observed after the treatment with a non-selective inhibitor of GABA transporters. The data obtained indicate that blocking the functions of neuronal and glial GABA transporters increases the level of extracellular GABA in the brain and prevent the development of convulsive syndrome in HBO2. The anti-convulsive effects of the GABA transporter inhibition in HBO2 are apparently due to the enhancement of GABA-mediated synaptic and extrasynaptic neurotransmissions. Inhibition of GABA transporters may be a promising approach for the development of effective methods to prevent oxygen seizures.

Dynamic changes of 'substantianigra-ventralislateralis-cortex' pathway neural activity coherence and neurotransmitters in rat during exhausting exercise

To reveal the possible mechanism of changes of 'substantianigra-ventralislateralis-cortex' pathway neural activity during one bout of exhausting exercise through observing the neural activity coherence between different nucleus and the concentration of extra-cellular glutamate (Glu) and gamma-aminobutyric acid (GABA). Male Wistar rats were randomly divided into neural activity real-time observation group, substantianigra (SNr) extracellular neurotransmitters observation group, ventralislateralis (VL) extracellular neuro-transmitters observation group and supplementary motor area (SMA) extracellular neurotransmitters observation group, 10 rats in each group. For rats of neural activity real-time observation group, by using LFPs and ECoG recording technique, and self-comparison, we simultaneously recorded the dynamic changes of neural activity of rat SNr, VL and SMA during one bout of exhausting exercise. The dynamic changes of ex-tracellular Glu and GABA in rat SNr, VL and SMA were also observed through microdialysis combined high performance liquid chromatography (HPLC) technique and self-comparison method. Based on the behavioral performance, the exhausting exercise process could be di-vided into 5 different stages, the rest condition, auto exercise period, early fatigue period, exhaustion condition and recovery period. The elec-trophysiological study results showed that, the coherence between neural activity in rat SNr, VL and SMA was significant between 0~30 Hz during all the procedure of exhausting exercise. Compared with the rest condition, the microdialysis study showed that the Glu concentrations and Glu/GABA ratio in SNr were decreased significantly during automatic exercise period (P < 0.05, P < 0.01), the GABA concentrations were increased significantly (P < 0.05, P < 0.01), while, in VL and cortex, the Glu concentrations and Glu/GABA ratio were increased significantly (P < 0.05, P < 0.01), the GABA concentrations were decreased significantly (P < 0.05, P < 0.01). Under early fatigue and ex-haustion conditions, compared with the rest condition,the Glu concentrations and Glu/GABA ratio in SNr were increased significantly (P < 0.05, P < 0.01), the GABA concentrations were decreased significantly (P < 0.05, P < 0.01), while the Glu concentrations and Glu/GABA ratio in VL and cortex were decreased significantly (P < 0.05, P < 0.01), the GABA concentrations were increased significantly (P < 0.05, P < 0.01). The neural net work communication between 'substantianigra-ventralislateralis-cortex' pathway exists, changes of Glu and GABA in the nucelus of the pathway are one of the factors resulting in the changes of neural activity.

The Role of Ventral Tegmental Area Gamma-Aminobutyric Acid in Chronic Neuropathic Pain after Spinal Cord Injury in Rats

Spinal cord injury (SCI) frequently results in chronic neuropathic pain (CNP). However, the understanding of brain neural circuits in CNP modulation is unclear. The present study examined the changes of ventral tegmental area (VTA) putative GABAergic and dopaminergic neuronal activity with CNP attenuation in rats. SCI was established by T10 clip compression injury (35 g, 1 min) in rats, and neuropathic pain behaviors, in vivo extracellular single-cell recording of putative VTA gamma-aminobutyric acid (GABA)/dopamine neurons, extracellular GABA level, glutamic acid decarboxylase (GAD), and vesicular GABA transporters (VGATs) were measured in the VTA, respectively. The results revealed that extracellular GABA level was significantly increased in the CNP group (50.5 ± 18.9 nM) compared to the sham control group (10.2 ± 1.7 nM). In addition, expression of GAD65/67, c-Fos, and VGAT exhibited significant increases in the SCI groups compared to the sham control group. With regard to neuropathic pain behaviors, spontaneous pain measured by ultrasound vocalizations (USVs) and evoked pain measured by paw withdrawal thresholds showed significant alteration, which was reversed by intravenous (i.v.) administration of morphine (0.5-5.0 mg/kg). With regard to in vivo electrophysiology, VTA putative GABAergic neuronal activity (13.6 ± 1.7 spikes/sec) and putative dopaminergic neuronal activity (2.4 ± 0.8 spikes/sec) were increased and decreased, respectively, in the SCI group compared to the sham control group. These neuronal activities were reversed by i.v. administration of morphine. The present study suggests that chronic increase of GABAergic neuronal activity suppresses dopaminergic neuronal activity in the VTA and is responsible for negative emotion and motivation for attenuation of SCI-induced CNP.

Effect of levetiracetam on extracellular amino acid levels in the dorsal hippocampus of rats with temporal lobe epilepsy

Levetiracetam (LEV) is an anticonvulsant drug with a unique mechanism of action that is not completely understood. However, its activity profile may involve effects on excitatory and/or inhibitory neurotransmission since the primary target of LEV, synaptic vesicle protein 2A, is ubiquitously expressed in all types of synaptic vesicles. Therefore, the objective of the present study was to explore the effect of LEV (300 mg/kg/day for one week, administered via osmotic mini-pumps) on neurotransmitter release and its probable selective effect on extracellular gamma-amino butyric acid (GABA), glutamate (Glu), aspartate (Asp), glutamine (Gln), taurine (Tau) and glycine (Gly) concentrations (using in vivo microdialysis under basal and high-K+ conditions) in the dorsal hippocampus (DH), a region that undergoes major synaptic changes during epilepsy. Epileptic rats developed clear signs of hyperexcitability, i.e., an elevated Glu/GABA ratio in the DH. The LEV concentration in blood after 7 days of treatment was within the therapeutic range. In contrast, LEV was not detected four days after mini-pump removal (washout period). Furthermore, LEV restored the Glu/GABA ratio to approximately the control level and significantly increased the GABA concentration after the initiation of high-K+ conditions. Based on these data, LEV treatment restored the lost balance between the excitatory and inhibitory systems under basal conditions. Moreover, LEV showed a selective effect by preferentially increasing vesicular release of GABA, a mechanism by which LEV could reduce epileptic seizures.

Hypotensive Response to Angiotensin II Type 2 Receptor Stimulation in the Rostral Ventrolateral Medulla Requires Functional GABA-A Receptors.

Objectives: Angiotensin II, glutamate and gamma-aminobutyric acid (GABA) interact within the rostral ventrolateral medulla (RVLM) and the paraventricular nucleus (PVN) modulating the central regulation of blood pressure and sympathetic tone. Our aim was to assess the effects of local angiotensin II type 2 receptor stimulation within the RVLM and the PVN on neurotransmitter concentrations and mean arterial pressure (MAP).Methods: In vivo microdialysis was used for measurement of extracellular glutamate and GABA levels and for local infusion of the angiotensin II type 2 receptor agonist Compound 21 in the RVLM and the PVN of conscious normotensive Wistar rats. The MAP response to local Compound 21 was monitored with a pressure transducer under anaesthesia. Angiotensin II type 2 receptor selectivity was assessed using the angiotensin II type 2 receptor antagonist PD123319; the GABA-A receptor antagonist bicuculline was used to assess the involvement of GABA-A receptors.Results: Infusion of Compound 21 (0.05 μg/μl/h) in the RVLM significantly increased GABA levels and lowered blood pressure. These effects were abolished by co-infusion with PD123319. No changes in neurotransmitter levels or effects on blood pressure were seen with PD123319 infusion alone. Co-infusion of bicuculline abolished the Compound 21 evoked decrease in MAP. Infusion of Compound 21 within the PVN did not change extracellular neurotransmitter levels nor MAP.Conclusion: Selective stimulation of angiotensin II type 2 receptor within the RVLM by local Compound 21 infusion reduces blood pressure and increases local GABA levels in normotensive rats. This hypotensive response requires functional GABA-A receptors, suggesting that GABAergic neurons are involved in the sympatho-inhibitory action underlying this hypotensive response.

Striatal Glutamate and GABA after High Frequency Subthalamic Stimulation in Parkinsonian Rat

ObjectiveHigh frequency stimulation (HFS) of the subthalamic nucleus (STN) is recognized as an effective treatment of advanced Parkinson’s disease. However, the neurochemical basis of its effects remains unknown. The aim of this study is to investigate the effects of STN HFS in intact and 6-hydroxydopamine (6-OHDA)-lesioned hemiparkinsonian rat model on changes of principal neurotransmitters, glutamate, and gamma-aminobutyric acid (GABA) in the striatum.MethodsThe authors examined extracellular glutamate and GABA change in the striatum on sham group, 6-OHDA group, and 6-OHDA plus deep brain stimulation (DBS) group using microdialysis methods.ResultsHigh-pressure liquid chromatography was used to quantify glutamate and GABA. The results show that HFS-STN induces a significant increase of extracellular glutamate and GABA in the striatum of 6-OHDA plus DBS group compared with sham and 6-OHDA group.ConclusionTherefore, the clinical results of STN-HFS are not restricted to the direct STN targets but involve widespread adaptive changes within the basal ganglia.

PP.35.02] EFFECT OF ANGIOTENSIN RECEPTOR STIMULATION IN THE ROSTRAL VENTROLATERAL MEDULLA ON BLOOD PRESSURE IN NORMOTENSIVE RATS

Objective: There is increasing evidence that glutamate and gamma-aminobutyric acid (GABA) interact within the rostral ventrolateral medulla (RVLM) modulating the central regulation of blood pressure and sympathetic tone. Angiotensin II (Ang II) is the most important effector in the renin-angiotensin-aldosterone system (RAAS) mediating its actions through the angiotensin II type 1 receptor (AT1R) and angiotensin II type 2 receptor (AT2R). The AT2R is part of the protective arm of the RAAS and mediates opposing effects counterbalancing the AT1R mediated actions. The selective and specific AT2R agonist Compound 21 (C21) is a useful research tool to investigate the AT2R mediated effects. Design and method: In the present study we assess through local stimulation and blockade of AT1R and AT2R the possible role of the brain RAAS at the level of the RVLM on the regulation of blood pressure. In vivo microdialysis, for measurement of extracellular glutamate and GABA levels, was carried out in the RVLM of conscious normotensive Wistar rats while locally infusing C21, Ang II, and PD123319. Glutamate and GABA levels were quantified by HPLC. Subsequently, the effect of central administration of C21, Ang II, and PD123319 on mean arterial pressure was assessed with a pressure transducer for continuous monitoring. Results: Infusion of Ang II into the RVLM significantly increased glutamate levels (P < 0.05) and significantly decreased GABA levels (P < 0.01). Infusion of the AT2R agonist C21 (0.05 μg/μl/h) tended to decrease glutamate and increase GABA levels, while absolutely no change was seen when co-infusing C21 together with the AT2 receptor antagonist PD123319. C21 infusion (0.05 μg/μl/h) significantly lowered blood pressure (P < 0.01). These responses were abolished by co-administration of C21 and PD123319. No changes in neurotransmitter levels or effects on blood pressure were seen with PD123319 infusion alone. Conclusions: These data demonstrate that acute specific stimulation of AT2R in the RVLM by local C21 infusion reduces blood pressure in normotensive rats, and suggests that decreases in local glutamate and increases in GABA levels may be involved in the sympatho-inhibitory action underlying this hypotensive response.
