Effects of Funneliformis mosseae on the utilization of organic phosphorus in Camellia oleifera Abel.

Arbuscular mycorrhizal (AM) fungi play an important role in the acquisition of phosphorus(P) by plants. The external hyphae of AM fungi function as an extension of plant roots and may downregulate related functions in the roots. It is not clear whether the ability of AM fungi to mineralize organicP affects root phosphatase activities. A pot experiment was conducted to investigate the effect of Funneliformis mosseae on soil organicP mineralization under phytate application and to explore root phosphatase activities, Puptake, and growth in Camellia oleifera Abel. The plants and their growth substrates were harvested 4 and 8months after planting. The results showed that organicP application had no effect on the total dry mass of nonmycorrhizal plants, but differences in dry mass under P application were observed in mycorrhizal plants in both harvests. Inoculation with F.mosseae increased soil acid phosphatase, phytase, and alkaline phosphatase activities and reduced the soil organicP content. Mycorrhizal plants had higher root activity, shoot and root P contents and root acid phosphatase and phytase activities than nonmycorrhizal plants irrespective of organicP application. In conclusion, AM fungi enhanced the mineralization of soil organicP and positively affect root phosphatase activities.

How important is the mycorrhizal pathway for plant Zn uptake?

Formation of arbuscular mycorrhizas can enhance plant uptake of immobile nutrients such as zinc (Zn) and phosphorus (P). Enhancement of Zn uptake by arbuscular mycorrhizal (AM) fungi on Zn-deficient soils has been studied previously, however, the quantity of Zn that is contributed by the AM pathway of uptake to the plant has not previously been reported for soil of any Zn status. We grew a mycorrhiza-defective mutant tomato (Solanum lycopersicum L.) genotype (rmc) and its mycorrhizal wild-type progenitor (76R) in pots containing a hyphal compartment (HC) accessible only by the external hyphae of AM fungi, and containing the radioisotope 65Zn. This was repeated at three soil Zn concentrations, ranging from low to high. We estimated the amount of Zn delivered via both the AM and direct (root) pathways. Up to 24 % of Zn in the shoots of the AM plants was delivered via the AM pathway at the lowest soil Zn treatment. This decreased significantly, to 8 %, as soil Zn concentration increased. No 65Zn was detected in the tissues of the non-mycorrhizal genotype. The relative contribution to shoot Zn by the AM pathway of uptake was highest when soil Zn was low, and decreased with increasing soil Zn concentration.

The direct response of the external mycelium of arbuscular mycorrhizal fungi to temperature and the implications for nutrient transfer

In this study we investigated the direct effects of temperature on the extra-radical mycelium (ERM) of arbuscular mycorrhizal fungi (AMF) and the resulting impact on the host plant nutrition and biomass production. Plantago lanceolata L. plants colonized by Glomus hoi (experiment 1) and either G. hoi or Glomus intraradices (experiment 2) were grown in compartmented microcosm units. AMF hyphae, but not roots, were permitted access to a second compartment containing a 15N:13C dual-labelled organic patch maintained at different temperature treatments. All plants were maintained at ambient temperature. AMF hyphal growth in the patch compartments was relatively insensitive to temperate but results were variable. G. hoi hyphal length density was 5 times higher at ambient (c. 24 °C) than cooled (c. 11 °C) temperatures but only at the end of the first experiment (105 d after patch addition). In contrast, in the second experiment (86 d after patch addition) AMF hyphal growth was unaffected by temperature in the patch compartment. These differences between experiments are likely due to large variation among replicates in the ERM produced and differences in how the organic patch was applied. In experiment 2, plant biomass and phosphate content differed according to the temperature at which the hyphae of both AMF species grew. Plant biomass was greater when the AMF were grown at c. 18 °C than c. 11 °C but was no different at c. 21 °C. These data show that direct temperature responses by the external hyphae of AMF can independently influence associated host plant growth. However, there were also important differences between the two AMF studied both in the amount of nutrients transferred and the distribution of the nutrients.

Functional diversity of external hyphae of AM fungi: Ability to colonise new hosts is influenced by fungal species, distance and soil conditions

The aim of this paper was to investigate the effects of soil conditions and distance from a host plant on the ability of hyphae of arbuscular mycorrhizal (AM) fungi to grow and colonise a new host. Two glasshouse experiments were conducted using compartmented pots. The first investigated the effects of distance between a colonised and uncolonised host plant ( Trifolium subterraneum L.) and average pore size of the growth substrate (100 μm, 38 μm) on the ability of two AM fungi, G. intraradices and G. mosseae, to colonise a new host plant. The second experiment determined if the pore size of the substrate (100 μm, 38 μm) affected the growth of AM fungi in the absence of a new host. In Experiment 1, both G. mosseae and G. intraradices grew successfully through the two sand substrates and colonised new host plants. Both fungi reached and colonised new hosts fastest when hosts were separated by the shortest distance (2.5 cm), with largest pore size substrate (100 μm). G. mosseae produced more external hyphae per unit of colonised root and colonised new host plants more rapidly than G. intraradices. However, receiver plants colonised by G. mosseae exhibited a negative mycorrhizal growth response following colonisation. Experiment 2 showed that G. mosseae grew further from its host than G. intraradices. The results support the theory that some AM fungal species may produce large amounts of external hyphae primarily to increase the probability of locating and colonising a new host plant.

Hydrolytic enzymes and ability of arbuscular mycorrhizal fungi to colonize roots

The production of hydrolytic enzymes from external mycelia associated with roots and colonized soybean roots (Glycine max L.) inoculated with different arbuscular-mycorrhizal (AM) fungi of the genus GLOMUS:, and the possible relationship between these activities and the capacity of the AM fungi to colonize plant roots was studied. There were differences in root colonization and plant growth between the GLOMUS: strains, and also between two isolates of G. mosseae. Hydrolytic activities in the root and external mycelia associated with roots differed in the AM fungi tested. Correlations were only found between the endoxyloglucanase activity of the external mycelia associated with roots of the AM fungi tested and the percentage root colonization or plant growth. However, hydrolytic activities of roots colonized by the different endophytes correlated with those of external mycelia. The hydrolytic activities were not qualitatively different because the endoxyloglucanase from AM colonized roots and the external mycelia did not show a high degree of polymorphism in the different species of fungus tested. The possible role of the hydrolytic activity of external hyphae of AM fungi was discussed as a factor affecting fungal ability to colonize the root and influence plant growth.
