Peroxisome proliferator-activated receptor gamma1 expression is induced during cyclic adenosine monophosphate-stimulated differentiation of alveolar type II pneumonocytes.

The primary function of lung alveolar type II cells is to synthesize pulmonary surfactant, a lipoprotein enriched in dipalmitoylphosphatidylcholine. Because type II pneumonocytes are highly lipogenic, we considered the possible role of the adipogenic nuclear hormone receptor, peroxisome proliferator-activated receptor gamma (PPARgamma), in their differentiation from epithelial cell precursors. A degenerate PCR-screening strategy revealed that multiple PPARs, including PPARgamma, are present in differentiated type II cells. A PCR-amplified PPARgamma DNA-binding domain was used to isolate a full-length PPARgamma1 complementary DNA clone from a rabbit type II cell complementary DNA library. Although another PPARgamma isoform, PPARgamma2, is known to be highly expressed in adipocytes, only PPARgamma1 was detected in rabbit type II cells by use of RT-PCR and by library screening. Rabbit PPARgamma1 has 90% nucleotide sequence identity and 95% amino acid identity to mouse PPARgamma1. PPARgamma1 messenger RNA was readily detected in total RNA isolated from rabbit type II pneumonocytes cultured in the presence of cAMP, which causes enlargement of the prealveolar ducts, accelerates the rate of type II cell differentiation, and induces transcription of the major surfactant associated protein, surfactant protein-A. PPARgamma1 messenger RNA also was detected in total RNA isolated from rabbit adipose tissue but not from whole adult or fetal lung, heart, or liver. By Western blot analysis, PPARgamma protein expression was found to occur coincidentally with surfactant protein-A expression during lung type II cell differentiation. In view of the role of PPARgamma in adipocyte differentiation and lipid homeostasis, we postulate that PPARgamma1 induction by cAMP plays a role in the differentiation and expression of lipogenic enzymes in lung type II cells.

IL-10 counteracts IL-1-induced surfactant protein A (SP-A) expression in rabbit lung explants. † 1472

The production of the proinflammatory cytokine IL-1 is increased in the fetal compartment in cases of preterm labor associated with intrauterine infection. The production of EGF, on the other hand, increases with advancing gestation. Both IL-1 and EGF upregulate the expression of SP-A. However, IL-1 may also have deleterious effects on the developing lung, since an elevated IL-1 concentration in lung effluent is associated with the development of chronic lung disease in the newborn. IL-10 is an anti-inflammatory cytokine that inhibits the production of IL-1 and of other proinflammatory cytokines.Aim. We investigated 1) whether IL-10 affects SP-A expression in lung explants and 2) whether IL-10 modulates SP-A expression in explants treated with IL-1 or EGF. Methods. Fetal rabbit lung explants(gestational age 22 days) were treated with IL-1 (570 ng/ml), EGF (50 ng/ml) or IL-10 (5 ng/ml) for 24 or 72 hours. Expression of SP-A mRNA was quantitated by Northern blotting. Results. SP-A expression at 24 and 72 h as percent of the respective controls (±SE) was:Table Thus, IL-1 and EGF enhanced SP-A expression, but did not potentiate each other's effects. IL-10 alone had no effect on SP-A expression. IL-10 suppressed the stimulatory effect of IL-1 in explants treated for 72 h, but not in those treated for 24 h. IL-10 did not suppress SP-A expression in EGF-treated explants. Conclusion. IL-10 selectively counteracts the effect of IL-1 but not that of EGF on SP-A expression. IL-10 may be of value in inhibiting lung inflammation caused by proinflammatory cytokines. The potential of IL-10 in preventing inflammation in the lungs without affecting maturational events unrelated to inflammation, should be further investigated.

Interleukin-1 alpha upregulates the expression of surfactant protein-A in rabbit lung explants.

Interleukin-1 (IL-1) is an important participant in infectious and inflammatory conditions. Interleukin-1 receptor antagonist (IL-1ra) prevents the effect of IL-1. We have shown that injection of interleukin-1 alpha (IL-1 alpha) into the amniotic fluid of pregnant rabbits stimulates the expression of surfactant protein-A (SP-A) in the lungs of the fetuses. We hypothesized that IL-1 alpha similarly enhances the expression of SP-A in rabbit lung explants in vitro. Explants obtained from 22-day fetal rabbit lungs were cultured in Waymouth's medium on a rotating platform in the presence or absence of IL-1 alpha (5.7-570 ng/ml) or IL-1ra (1-10 micrograms/ml). Dibutyryl cAMP (1 mM) served as a positive control. After 3 days in culture, the explants were harvested and Northern analysis of SP-A was performed using the 1.9-kb rabbit SP-A cDNA probe. IL-1 alpha and dibutyryl cAMP increased the expression of SP-A twofold, as judged by video densitometry. IL-1ra did not change SP-A expression as compared with controls, suggesting that endogenous IL-1 activity was not responsible for the basal level of SP-A expression in the explants. Dibutyryl cAMP increased the expression of SP-B mRNA, whereas IL-1 alpha had no effect on SP-B mRNA concentration. We conclude that inflammatory mediators interact with lung cells to alter synthesis of important components of the surfactant system.

Surfactant protein gene expression in metastatic and micrometastatic pulmonary adenocarcinomas and other non-small cell lung carcinomas: Detection by reverse transcriptase-polymerase chain reaction

Reverse transcriptase (RT)-PCR with primers specific for surfactant protein A (SP-A), B (SP-B), C (SP-C), and D (SP-D) genes was applied to detect metastatic non-small cell lung carcinomas. Forty-one paratracheal and subcarinal lymph nodes obtained from 41 patients with non-small cell lung carcinomas, 11 lymph nodes from 11 patients with extrapulmonary adenocarcinomas, and eight control lymph nodes from patients without cancer were analyzed using RT-PCR. PCR products corresponding to SP-B gene products were found in all eight control lymph nodes, offering evidence of SP-B gene expression in cells of lymphatic tissue. SP-A, SP-C, and/or SP-D transcripts were detected in 11 (84.6%) of 13 lymph nodes with histologically identifiable metastases of pulmonary adenocarcinomas and in 10 (55.5%) of 18 lymph nodes that were tumor free on routine histological examination. These findings provide evidence of micrometastatic nodal involvement which remains undetectable by conventional light microscopy but can be evaluated by surfactant RT-PCR. Gene expression of SP-A and SP-C was restricted to metastatic pulmonary adenocarcinomas but SP-D gene activity has been also detected in two of four metastases of pulmonary large cell carcinomas, one adenosquamous carcinoma, and nine extrapulmonary adenocarcinomas as well.

The structure and function of surfactant protein A. Hydroxyproline- and carbohydrate-deficient mutant proteins.

Pulmonary surfactant protein A (SP-A) regulates the uptake and secretion of phospholipid by alveolar type II cells and is an important component of surfactant lipid aggregates. In an attempt to understand how specific structural domains of SP-A relate to the function of the protein, we used site-directed mutagenesis of the cDNA for SP-A and heterologous expression with baculovirus vectors. Synthesis of the wild type SP-A in insect cells resulted in a form of the protein in which proline residues were not hydroxylated and that is denoted SP-Ahyp. Three mutant SP-As with substitutions in the consensus sequences for glycosylation (SP-Ahyp,glc) to prevent N-linked oligosaccharide attachment at Asn1 and Asn187 were produced, individually and in tandem. The SP-Ahyp was glycosylated at both the Asn1 and Asn187 positions, demonstrated partial sulfhydryl-dependent oligomerization, and formed incomplete oligomers in solution. The SP-Ahyp and SP-Ahyp,glc bound to immobilized carbohydrate and to phospholipid liposomes and partially competed for occupancy of a plasma-membrane receptor for SP-A. The SP-Ahyp and the SP-Ahyp,glc were equally effective inhibitors of the secretion of surfactant lipids from isolated type II cells (IC50 = 0.5 microgram/ml) and aggregated phospholipid liposomes at 20 degrees C. All of the recombinant SP-As demonstrated markedly reduced aggregation of lipid at 37 degrees C. We conclude that the hydroxylation of proline residues is required for perfect oligomerization of SP-A and for thermal stability in the interaction with lipid. Furthermore, recombinant SP-A is able to inhibit the secretion of phospholipid from isolated type II cells and to aggregate lipid vesicles independent of the presence of N-linked carbohydrate or the site of glycosylation.

Genomic elements involved in transcriptional regulation of the rabbit surfactant protein-A gene.

Expression of the surfactant protein-A (SP-A) gene is lung specific and is developmentally and hormonally regulated in fetal lung tissue. Cyclic AMP analogs and glucocorticoids stimulate transcriptional activity of the SP-A gene in fetal rabbit lung tissue in culture; an additive effect is observed when the agents are added in combination. To analyze the genomic regions that regulate SP-A promoter activity, fusion genes comprised of -1766, -991, -378, and -47 basepairs (bp) of DNA flanking the 5'-end of the SP-A gene, the transcription initiation site, and 20 bp of exon I linked to the human GH (hGH) structural gene were subcloned into a replication-defective human adenovirus vector and transfected into differentiated rat type II cells in primary culture. SP-A promoter activity was analyzed by RIA of hGH protein in the culture medium. In type II cells transfected with SP-A-1766:hGH and SP-A-991:hGH fusion genes, hGH production was induced 30- to 40-fold by (Bu)2AMP (Bt2cAMP; 1 mM). When type II cells were transfected with the SP-A-378:hGH fusion gene, basal levels of expression were reduced by more than 50%; however, Bt2cAMP caused an 11-fold increase in hGH production. In type II cells transfected with the SP-A-47:hGH fusion gene, basal levels of hGH production were essentially undetectable, and no stimulatory effect of Bt2cAMP was apparent. Cyclic AMP stimulation of expression of the SP-A-1766:hGH, SP-A-991:hGH, and SP-A-378:hGH fusion genes was limited to type II pneumonocytes in primary culture and was absent in two lung adenocarcinoma cell lines (NCl-H358 and A549), which do not express SP-A, and in cAMP-responsive adrenal Y1 cells. Mutations of a putative cAMP-responsive element (TGACCTCA) at -261 bp revealed its functional importance in mediating cAMP regulation of SP-A gene expression. Unexpectedly, dexamethasone (Dex; 10(-7) M) antagonized the stimulatory effect of Bt2cAMP on expression of SP-A:hGH fusion genes containing from -378 to -1766 bp of 5'-flanking DNA as well as that of a fusion gene construct containing -991 bp of 5'-flanking DNA, the first exon, the first intron, and 20 bp of the second exon (SP-A-991+670:hGH). The inhibitory effect of Dex was dose dependent, with half-maximal inhibition occurring at a Dex concentration of 8 x 10(-10) M. The inhibitory effect of Dex was prevented by the glucocorticoid receptor antagonist RU486.(ABSTRACT TRUNCATED AT 400 WORDS)

Pulmonary surfactant protein a in pleural effusions

Pulmonary surfactant protein A (SP-A) is known to be a major phospholipid-associated glycoprotein in pulmonary surfactant, which is specific to the lung. Immunohistochemically, expression of SP-A in tumor tissues is found in approximately 50% of patients with lung adenocarcinoma but not in the other histologic types of lung cancer of metastatic lung tumors. In this study, the SP-A content of pleural effusions was determined using an enzyme-linked immunosorbent assay. These results showed that approximately 40% of patients with lung adenocarcinomas (27 of 67) had high levels of SP-A (greater than 500 ng/ml) in their pleural effusions. By contrast, patients with other histologic types of lung cancers, adenocarcinomas of different primary sites, and tuberculosis had low levels of SP-A in their pleural effusions. The determination of SP-A in malignant effusions will contribute to distinguishing primary lung adenocarcinoma from adenocarcinomas of miscellaneous origin.

Characterization of the rat pulmonary surfactant protein A promoter.

The expression of the pulmonary surfactant protein A (SP-A) is developmentally regulated and controlled by several hormones. In an attempt to characterize cis-acting elements involved in the regulation of SP-A expression, we have cloned the 5' flanking sequence of the rat SP-A gene. The promoter region contains a TATA box but no CAAT box. The transcription start site has been identified by anchored polymerase chain reaction and S1 nuclease mapping of the mature and precursor transcripts. S1 mapping of precursor transcripts has confirmed the stimulating effect of glucocorticoids on SP-A rat gene transcription in vivo. This hormonal effect may be mediated by a putative glucocorticoid responsive element located 140 bp upstream from the initiation site and protected against DNase 1 digestion in footprinting experiments. In vitro transcription of a G-free reporter cassette linked to the 212-bp 5' flanking DNA fragment has established that this putative promoter region is functional. Efficient transcription of the G-free reporter cassette was obtained with cell-free fetal lung extracts, whereas no transcript was detectable with cell-free liver extracts. Comparative analysis of the human and rat 5' flanking sequences shows the presence of strongly conserved motifs, unrelated to previously known consensus sequences. Some of these motifs, specifically protected in DNase 1 footprinting studies, could therefore be involved in the regulation of SP-A gene expression.

Transforming growth factor-β inhibits surfactant protein A expression in vitro

Effects of members of the transforming growth factor-β (TGF-β) family on expression of surfactant protein A (SP-A) were determined in human pulmonary adenocarcinoma cells. TGF-β decreased SP-A content in two distinct pulmonary adenocarcinoma cell lines with bronchiolar (NCI-H441-4) and alveolar (NCI-H820) cell characteristics. TGF- β 1, β 2 and β 3 were equally effective in decreasing SP-A. Effects of the TGF-β's on SP-A content were dose dependent, EC 50 approximately 20–30 pg/ml for each form of TGF-β. TGF-β decreased cellular SP-A content in association with decreased levels of SP-A mRNA. Inhibitory effects of TGF- β 1, on SP-A mRNA was time dependent, reaching maximal effects within 12–24 h, after which SP-A mRNA was approximately 10% of that present in untreated cells. Maximal inhibition of SP-A mRNA was observed at 250 pg/ml TGF- β 1. TGF-β-dependent inhibition of SP-A expression was not associated with altered cell morphology, growth, or viability. TGF-β family members act directly on pulmonary adenocarcinoma cells to inhibit SP-A expression by mechanisms which are mediated, at least in part, at a pretranslational level.

Appearance of surfactant proteins, SP-A and SP-B, in developing rat lung and the effects of in vivo dexamethasone treatment

Hydrophobic pulmonary surfactant proteins (SP-B and SP-C) promote the adsorption of phospholipids at the air/liquid interface and the addition of surfactant protein A (SP-A) enhances this function. The developmental profiles of phospholipids and SP-A in the lung have been reported, but that of SP-B and SP-C remain unknown. We recently developed an enzyme-linked immunosorbent assay (ELISA) that measures SP-B in the rat. Using ELISA for SP-A and SP-B, we measured the contents of SP-A and SP-B in lung homogenates. The developmental profiles of SP-A and SP-B during the late gestational and postnatal periods were found to be distinctly different from each other. SP-A increased during late gestation and reached its maximum on day 1 after birth. This developmental profile of SP-A in the lungs was very similar to that of disaturated phosphatidylcholine (DSPC). In contrast, the SP-B contents in fetal lungs were low and increased after birth, reaching its maximum on day 4 after birth. In vivo dexamethasone treatment resulted in significant increases of SP-A content in rat lung homogenate on day 19 and day 21 of gestation, and day 5 after birth, whereas SP-B content increased significantly only on day 19 of gestation by dexamethasone administration. SP-A synthesis may be enhanced both pre- and postnatally, but SP-B synthesis may be stimulated only during the late gestational period by in vivo dexamethasone treatment. The difference in developmental profiles and the different responses to dexamethasone treatment between SP-A and SP-B indicate that the expression of SP-A and SP-B may be regulated independently at least in developing rat lungs.

Increased synthesis and mRNA of surfactant protein A in oxygen-exposed rats.

Surfactant protein A (SP-A) is an abundant glycoprotein in surfactant that is synthesized and secreted by alveolar type II cells and likely has important roles in mediating surfactant function and metabolism. In the present study, we demonstrate that exposure to 85% oxygen increased alveolar lavage and lung SP-A, and that these increases were related to increased SP-A synthesis and mRNA. Adult rats were exposed to room air or to 85% oxygen for 3, 5, or 7 days. Continuous exposure to hyperoxia progressively increased SP-A content, with a 20-fold increase in alveolar lavage and a 10-fold increase in lung SP-A content observed after 7 days. SP-A-specific mRNA increased in the lungs of rats exposed to oxygen, occurring with a time course similar to the increase in tissue SP-A. SP-A mRNA was increased 7-fold after 7 days of oxygen exposure. Synthesis of SP-A was increased 2- to 3-fold and secretion was increased 6- to 7-fold by type II epithelial cells isolated from oxygen-exposed rats. We conclude that exposure to hyperoxia increased lung and alveolar SP-A pool sizes. Increased expression of SP-A was related, at least in part, to increased SP-A mRNA and increased SP-A synthesis and secretion by type II epithelial cells.
