The Mysterious Food-Entrainable Oscillator: Insights from Mutant and Engineered Mouse Models.

The food-entrainable oscillator (FEO) is a mysterious circadian clock because its anatomical location(s) and molecular timekeeping mechanism are unknown. Food anticipatory activity (FAA), which is defined as the output of the FEO, emerges during temporally restricted feeding. FAA disappears immediately during ad libitum feeding and reappears during subsequent fasting. A free-running FAA rhythm has been observed only in rare circumstances when food was provided with a period outside the range of entrainment. Therefore, it is difficult to study the circadian properties of the FEO. Numerous studies have attempted to identify the critical molecular components of the FEO using mutant and genetically engineered mouse models. Herein we critically review the experimental protocols and findings of these studies in mouse models. Several themes emerge from these studies. First, there is little consistency in restricted feeding protocols between studies. Moreover, the protocols were sometimes not optimal, resulting in erroneous conclusions that FAA was absent in some mouse models. Second, circadian genes are not necessary for FEO timekeeping. Thus, another noncanonical timekeeping mechanism must exist in the FEO. Third, studies of mouse models have shown that signaling pathways involved in circadian timekeeping, reward (dopaminergic), and feeding and energy homeostasis can modulate, but are not necessary for, the expression of FAA. In sum, the approaches to date have been largely unsuccessful in discovering the timekeeping mechanism of the FEO. Moving forward, we propose the use of standardized and optimized experimental protocols that focus on identifying genes that alter the period of FAA in mutant and engineered mouse models. This approach is likely to permit discovery of molecular components of the FEO timekeeping mechanism.

MPeriod2Brdm1 and other single Period mutant mice have normal food anticipatory activity

Animals anticipate the timing of food availability via the food-entrainable oscillator (FEO). The anatomical location and timekeeping mechanism of the FEO are unknown. Several studies showed the circadian gene, Period 2, is critical for FEO timekeeping. However, other studies concluded that canonical circadian genes are not essential for FEO timekeeping. In this study, we re-examined the effects of the Per2Brdm1 mutation on food entrainment using methods that have revealed robust food anticipatory activity in other mutant lines. We examined food anticipatory activity, which is the output of the FEO, in single Period mutant mice. Single Per1, Per2, and Per3 mutant mice had robust food anticipatory activity during restricted feeding. In addition, we found that two different lines of Per2 mutant mice (ldc and Brdm1) anticipated restricted food availability. To determine if FEO timekeeping persisted in the absence of the food cue, we assessed activity during fasting. Food anticipatory (wheel-running) activity in all Period mutant mice was also robust during food deprivation. Together, our studies demonstrate that the Period genes are not necessary for the expression of food anticipatory activity.

Effects of bilateral anterior agranular insula lesions on food anticipatory activity in rats.

Food anticipatory activity (FAA) refers to a daily rhythm of locomotor activity that emerges under conditions of food restriction, whereby animals develop an intense, predictable period of activity in the few hours leading up to a predictable, daily delivery of food. The neural mechanisms by which FAA is regulated are not yet fully understood. Although a number of brain regions appear to be involved in regulating the development and expression of FAA, there is little evidence to date concerning the role of the anterior agranular insular cortex (AICa). The AICa plays a critical role in integrating the perception of visceral states with motivational behaviour such as feeding. We assessed the effect of bilateral electrolytic or ibotenic acid lesions of the AICa on FAA in male Wistar rats receiving food for varying lengths of time (2 h, 3 h, or 5 h) during the middle of the light phase (starting at either ZT4 or ZT6). Contrary to our initial expectations, we found that both electrolytic and ibotenic acid lesions significantly increased, rather than decreased, the amount of FAA expressed in lesioned rats. Despite increased FAA, lesioned rats did not eat significantly more during restricted feeding (RF) periods than control rats. Similar to controls, AlCa-lesioned rats showed negligible anticipatory activity to a restricted treat suggesting that the increased anticipatory activity in lesioned rats is associated with food restriction, rather than the appetitive value of the meal. Monitoring behaviour in an open field indicated that increased FAA in AlCa-lesioned rats was not explained by a general increase in locomotor activity. Together, these findings suggest that the AICa contributes to the network of brain regions involved in FAA.

Sex-related difference in food-anticipatory activity of mice

The expression of food-anticipatory activity (FAA) is induced by restricted feeding (RF), and its entrainment requires food-entrainable oscillators, the neuroanatomical basis of which is currently unclear. Although RF impacts various hormones, sex-related differences in FAA are unclear. 'Here, we report significantly more food-anticipatory wheel-running activity in male than in female mice during RF. In parallel with the sex-related difference in FAA, male and female mice display different food intake and body weight in response to RF. Since gonadal hormones could be involved in the sex-specific difference in FAA, we compared sham and gonadectomized male and female wild-type mice. In gonadectomized mice, the sex difference in FAA was abolished, indicating a role for gonadal hormones in FAA. Further, plasma concentrations of the hormone ghrelin were higher in female than in male mice during ad libitum (AL) feeding, and RF induced a temporal advance in its peak in both sexes. RF also shifted the expression peak of the circadian gene mPer1 in the hippocampus and liver, although no sex difference was found in either the level or the cyclic phase of its expression. Per1Brdm1 mutant mice were still sexually dimorphic for FAA, but diminished FAA was noted in both male and female Per2Brdm1 mutant mice. In summary, our results imply that gonadal hormones contribute to the sex difference in FAA, possibly through modulating ghrelin activity.

Assessing interactions between Ghsr and Mc3r reveals a role for AgRP in the expression of food anticipatory activity in male mice.

The stomach hormone ghrelin and hypothalamic melanocortin neurons belong to a gut-brain circuit controlling appetite and metabolic homeostasis. Mice lacking melanocortin-3 receptor (Mc3rKO) or growth hormone secretagogue receptor (GhsrKO) genes exhibit attenuated food anticipatory activity (FAA), a rise in locomotor activity anticipating mealtime, suggesting common circuitry regulating anticipatory responses to nutrient loading. To investigate the interaction between Ghsrs and Mc3rs, we compared food anticipatory responses in GhsrKO, Mc3rKO, and double Ghsr;Mc3r knockout (DKO) mice subjected to a hypocaloric restricted feeding (RF) protocol in constant dark or 12-hour light, 12-hour dark settings. DKO are viable, exhibiting no overt behavioral or metabolic phenotypes in ad libitum or fasting conditions. FAA was initially attenuated in all mutant strains in constant darkness. However, GhsrKO eventually exhibited a robust food anticipatory response, suggesting compensation. Mc3rKO and DKO did not compensate, indicating a continued requirement for Mc3rs in maintaining the expression of FAA in situations of RF. Abnormal regulation of hypothalamic agouti-related peptide/neuropeptide Y (AgRP/Npy) neurons previously observed during fasting may contribute to attenuated FAA in Mc3rKO. AgRP and Npy expression measured 1 hour before food presentation correlated positively with FAA. Absence of Mc3rs (but not Ghsrs) was associated with lower AgRP/Npy expression, suggesting attenuated responses to signals of negative energy balance. These observations support the importance of Mc3rs as modulators of anticipatory responses to feeding, with mice able to compensate for loss of Ghsrs. The behavioral deficits of Mc3rKO displayed during RF may be partially explained by reduced hunger sensations owing to abnormal regulation of orexigenic AgRP/Npy neurons.

Behavioral and neural correlates of acute and scheduled hunger in C57BL/6 mice.

In rodents, daily feeding schedules induce food anticipatory activity (FAA) rhythms with formal properties suggesting mediation by food-entrained circadian oscillators (FEOs). The search for the neuronal substrate of FEOs responsible for FAA is an active area of research, but studies spanning several decades have yet to identify unequivocally a brain region required for FAA. Variability of results across studies leads to questions about underlying biology versus methodology. Here we describe in C57BL/6 male mice the effects of varying the ‘dose’ of caloric restriction (0%, 60%, 80%, 110%) on the expression of FAA as measured by a video-based analysis system, and on the induction of c-Fos in brain regions that have been implicated in FAA. We determined that more severe caloric restriction (60%) leads to a faster onset of FAA with increased magnitude. Using the 60% caloric restriction, we found little evidence for unique signatures of neuronal activation in the brains of mice anticipating a daily mealtime compared to mice that were fasted acutely or fed ad-libitum–even in regions such as the dorsomedial and ventrolateral hypothalamus, nucleus accumbens, and cerebellum that have previously been implicated in FAA. These results underscore the importance of feeding schedule parameters in determining quantitative features of FAA in mice, and demonstrate dissociations between behavioral FAA and neural activity in brain areas thought to harbor FEOs or participate in their entrainment or output.

Agonistic acts as possible indicator of food anticipatory activity (FAA) in rainbow trout (Oncorhynchus mykiss)

Summary Food anticipatory activity (FAA) was assessed in rainbow trout (Oncorhynchus mykiss) in two small raceways using demand-feeding (T2) and hand-feeding (t2). The fish of both raceways were subjected to restricted feeding (RF) at two times in two places and the fish distribution and/or trigger actuation, total agonistic behaviour between fish, and swimming speed were measured. Food anticipatory activity did not appear clearly when using the usual measures. The present study suggests that the expression of FAA is not limited to trigger actuations, fish positioning or swimming activity but may also be described by swimming speed and agonistic behaviour.

Robust Food Anticipatory Activity in BMAL1-Deficient Mice

Food availability is a potent environmental cue that directs circadian locomotor activity in rodents. Even though nocturnal rodents prefer to forage at night, daytime food anticipatory activity (FAA) is observed prior to short meals presented at a scheduled time of day. Under this restricted feeding regimen, rodents exhibit two distinct bouts of activity, a nocturnal activity rhythm that is entrained to the light-dark cycle and controlled by the master clock in the suprachiasmatic nuclei (SCN) and a daytime bout of activity that is phase-locked to mealtime. FAA also occurs during food deprivation, suggesting that a food-entrainable oscillator (FEO) keeps time in the absence of scheduled feeding. Previous studies have demonstrated that the FEO is anatomically distinct from the SCN and that FAA is observed in mice lacking some circadian genes essential for timekeeping in the SCN. In the current study, we optimized the conditions for examining FAA during restricted feeding and food deprivation in mice lacking functional BMAL1, which is critical for circadian rhythm generation in the SCN. We found that BMAL1-deficient mice displayed FAA during restricted feeding in 12hr light:12hr dark (12L:12D) and 18L:6D lighting cycles, but distinct activity during food deprivation was observed only in 18L:6D. While BMAL1-deficient mice also exhibited robust FAA during restricted feeding in constant darkness, mice were hyperactive during food deprivation so it was not clear that FAA consistently occurred at the time of previously scheduled food availability. Taken together, our findings suggest that optimization of experimental conditions such as photoperiod may be necessary to visualize FAA in genetically modified mice. Furthermore, the expression of FAA may be possible without a circadian oscillator that depends on BMAL1.

Restricted feeding schedules phase shift daily rhythms of c-Fos and protein Per1 immunoreactivity in corticolimbic regions in rats

Entrainment by daily restricted feeding schedules (RFS) produces food anticipatory activity (FAA) which involves motivational processes which may be regulated by corticolimbic structures and the nucleus accumbens. The present study aimed first to determine whether corticolimbic structures participate in the expression of FAA, therefore c-Fos immunoreactivity (Fos-IR) was employed as marker of neuronal activity. The second goal was to characterize diurnal rhythms of the clock protein protein Per1 (PER1) in corticolimbic structures and to determine the influence of RFS on the diurnal temporal pattern. Rats were maintained under RFS with food access for 2 h daily, a control group was fed ad libitum. Food entrainment produced a pattern of increased Fos-IR during FAA and after mealtime in the two sub-regions of the nucleus accumbens (ACC), in the basolateral and central amygdala, in the bed nucleus of the stria terminalis (BNST), in the lateral septum (LS), in the prefrontal cortex (PFC), and in the paraventricular thalamic nucleus (PVT). No increased Fos-IR was observed in the hippocampus. Under ad libitum conditions all structures studied showed daily oscillations of PER1, excluding both amygdalar nuclei and the PFC. RFS shifted and set the daily peaks at zeitgeber time (ZT) 12 for both sub-regions in the accumbens, the hippocampus, lateral septum and PFC. RFS enhanced the amplitude at ZT12 of the BNST and shifted the peak of the PVT to ZT6. No changes were observed in the amygdalar nuclei. Present data indicate that cellular activation of corticolimbic structures is associated with behavioral events related to food anticipatory activity and that mealtime is a relevant signal that shifts daily oscillations of PER1 in corticolimbic structures. Data suggest a relevant role of corticolimbic structures as oscillators for FAA.

Modifications of local cerebral glucose utilization during circadian food-anticipatory activity

Food-anticipatory activity that animals express before a daily timed meal is considered as the behavioral output of a feeding-entrainable oscillator whose functional neuroanatomy is still unknown. In order to identify the possible brain areas involved in that timing mechanism, we investigated local cerebral metabolic rate for glucose during food-anticipatory activity produced either by a 4-h daily access to food starting 4 h after light onset or by a hypocaloric feeding provided at the same time. Local cerebral metabolic rate for glucose measured by the labeled 2-[ 14C]-deoxyglucose technique was quantified in 40 structures. In both groups of food-restricted rats, three brain regions (the nucleus of the solitary tract, the cerebellar cortex and the medial preoptic area) showed a decrease in local cerebral metabolic rate for glucose, compared with control ad libitum animals. In addition, only one structure, the paraventricular thalamic nucleus, was affected by temporal restricted feeding, and not by hypocaloric feeding, compared with ad libitum rats. By contrast, three brain regions, i.e. the intergeniculate leaflets, the paraventricular hypothalamic and the arcuate nuclei, showed specifically metabolic decreases during anticipation of hypocaloric feeding, and not during anticipation of temporal restricted feeding, compared with the ad libitum group. Expression of food-anticipatory activity appears to be regulated by an integrated neural circuit of brainstem and hypothalamic pathways, with hypocaloric feeding involving more extensive forebrain areas than temporal restricted feeding.

Housing conditions influence the expression of food-anticipatory activity in mice

If food is presented for a limited duration once every 24 h, rats will gradually develop increased activity in the hours preceding mealtime, even if this is during a time of day when they are normally inactive. This food-anticipatory activity (FAA) is mediated by the entrainment of an endogenous self-sustaining circadian pacemaker. Mice are increasingly being used for the molecular analysis of behavior because of the large number of genetically manipulated mouse models available, but little is known about food entrainment in this species. This study assessed the impact of different housing conditions on the expression of FAA in a temporally restricted feeding paradigm. Wheel-running activity rhythms were recorded from mice ( Mus musculus) undergoing food restriction while housed under one of two conditions. The results demonstrated that mice housed on open shelves showed robust FAA while those housed in isolation boxes did not. These results indicate that differences in susceptibility to food entrainment among mice (e.g., those with different genotypes) should be interpreted cautiously, since at least one strain is strongly affected by a relatively minor procedural difference that was not anticipated to have a significant impact.

Food anticipatory activity and photic entrainment in food-restricted BALB/c mice

The BALB/c mouse was evaluated as a model for the study of entrainment of circadian rhythms by feeding schedules. Mice were housed in a 12:12-h light–dark (LD) environment with food available for 3–5 h/day (5 h before dark onset). Food anticipatory activity (FAA) rhythms were evident in all mice, ranging from robust in some to weak and variable in others. Advancing transients of the end of nocturnal activity were evident in many cases, culminating in a significant shortening of the main bout of nocturnal activity. Transients and contraction of nocturnal activity were not dependent on the expression of FAA. Following restricted feeding, nocturnal activity expanded by a series of delaying transients. On the first day of constant dark (DD) with ad libitum food access following restricted feeding in LD, the phase from which activity free-ran was advanced by comparison with control tests. Transients, compressed nocturnal activity, and advanced phase of free-run suggest that feeding schedules cause phase advancement of light-entrained rhythms in BALB/c mice. When restricted feeding was imposed in DD, several mice expressed robust FAA concurrent with a free-running activity component. In some cases, free-running rhythms entrained to feeding time, and in other cases, the period of the free run lengthened toward 24 h. These data show that restricted feeding in BALB/c mice can engage a circadian mechanism driving FAA rhythms and can also modulate the phase of photic entrainment, possibly by a direct entraining effect on the light-entrained rhythm. The BALB/c mouse strain, in several respects, appears to be a useful model for the study of scheduled feeding and circadian rhythms.
