Effects of Ultraviolet B Radiation on the Function of the Testicles, Expression of Caspase-3 and NOS-2, and the Protective Role of Naringin in Mice.

In today's evolving global environment, reproductive dysfunctions brought on by various environmental toxins are of greatest concern. Radiation is a constant threat to living things, causing both genetic and cellular changes that result in mutations and cell death. It is thought that ultraviolet B (UVB) radiation we are exposed to daily has biological effects on rats and humans that are both short and long term. Due to the damaging effects of UVB radiation on the living system, this study explores the automatic mechanism by which a certain level of radiation induces oxidative stress, which is further controlled by the antioxidant activity of naringin (NG). In our study, male Swiss albino mice were exposed to UVB irradiation, which altered mice's body and testes weight, hormonal imbalance, biochemical parameters, and histo-morphometric parameter. In addition, we chose naringin's UVB irradiation deterrent effect. Twenty-four healthy adult male Swiss albino mice weighing 25-35 g were chosen at random. For 15 days of exposure, they were divided into four groups at random: group I-control, group II-UVB exposure (2 h per day), group III-UVB exposure with naringin (NG) (80 mg/kg, bw), and group IV-naringin (NG) (80 mg/kg, bw). Compared to the control group, UVB irradiation causes alterations in the animal body weight, testes weight, hormones, enzymatic and non-enzymatic assays, and histological parameters. It was seen that NG retrieved the alterations in parameters caused by UVB irradiation. The UVB radiation exposure on mice caused the testicular dysfunction drastically, while the naringin recapitulates testis functioning.

Effects of UV-B radiation on pollen germination and tube growth: A global meta-analysis

Despite widespread recognition of pollen's potential sensitivity to ultraviolet-B (UV-B) radiation (280–315 nm), there remains ongoing debate surrounding the extent and mechanisms of this effect. In this study, using published data on pollen germination and tube growth including 377 pair-wise comparisons from 77 species in 30 families, we present the first global quantification of the effects of UV-B radiation on pollen germination and tube growth, along with its underlying mechanisms. Our results showed a substantial reduction in both pollen germination and tube growth in response to UV-B radiation, affecting 90.9 % and 84.2 % of species, respectively. Notably, these reductions exhibited phylogenetic constraints, highlighting the role of evolutionary history in shaping the sensitivity of pollen germination and tube growth to UV-B radiation. A negative correlation between elevation and the sensitivity of pollen tube growth was detected, suggesting that pollens from plants at higher elevations exhibit greater resistance to UV-B radiation. Our investigation also revealed that the effects of UV-B radiation on pollen germination and tube growth were influenced by a range of abiotic and biotic factors. Nevertheless, the intensity and duration of UV-B radiation exposure exhibited the highest explanatory power for the effects on both pollen germination and tube growth. This suggests that the responses of pollens to UV-B radiation are profoundly influenced by its dose, a critical consideration within the context of global change. In conclusion, our study provides valuable insights into the diverse responses of pollen germination and tube growth to UV-B radiation, highlighting the environment and species-dependent nature of pollen's susceptibility to UV-B radiation, with substantial implications for our understanding of the ecological and agricultural consequences of ongoing changes in UV-B radiation.

Inhibition of cutaneous heat-sensitive Ca2+ -permeable transient receptor potential vanilloid 3 channels alleviates UVB-induced skin lesions in mice.

Ultraviolet B (UVB) radiation causes skin injury by trigging excessive calcium influx and signaling cascades in the skin keratinocytes. The heat-sensitive Ca2+ -permeable transient receptor potential vanilloid 3 (TRPV3) channels robustly expressed in the keratinocytes play an important role in skin barrier formation and wound healing. Here, we report that inhibition of cutaneous TRPV3 alleviates UVB radiation-induced skin lesions. In mouse models of ear swelling and dorsal skin injury induced by a single exposure of weak UVB radiation, TRPV3 genes and proteins were upregulated in quantitative real-time PCR and Western blot assays. In accompany with TRPV3 upregulations, the expressions of proinflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were also increased. Knockout of the TRPV3 gene alleviates UVB-induced ear swelling and dorsal skin inflammation. Furthermore, topical applications of two selective TRPV3 inhibitors, osthole and verbascoside, resulted in a dose-dependent attenuation of skin inflammation and lesions. Taken together, our findings demonstrate the causative role of overactive TRPV3 channel function in the development of UVB-induced skin injury. Therefore, topical inhibition of TRPV3 may hold potential therapy or prevention of UVB radiation-induced skin injury.

Transcriptomic Analysis Reveals the Involvement of Flavonoids Synthesis Genes and Transcription Factors in Dracaena cambodiana Response to Ultraviolet-B Radiation

Ultraviolet-B (UV-B) radiation is a major abiotic stress that dragon trees are exposed to during their growth and development; however, it is also an environmental signal perceived by plants that affects the flavonoid pathway. Previous studies have demonstrated that amounts of flavonoids are contained in dragon tree resin, otherwise known as dragon’s blood. However, the traits and mechanism involved in the UV-B-mediated increase in flavonoids in dragon trees are still unknown. Here, we studied the response of Dracaena cambodiana under full solar UV-B radiation. The results showed that the contents of total flavonoids in D. cambodiana significantly increased after UV-B radiation exposure. Then, the transcriptome was used for determining the interactive mechanism of flavonoid accumulation and UV-B stress. Differential expression analyses identified 34 differentially expressed genes (DEGs) involved in flavonoid synthesis; specifically, 24 of the identified DEGs were significantly up-regulated after UV-B radiation exposure. In addition, 57 DEGs involved in Ca2+/kinase sensors, 58 DEGs involved in ROS scavenging and the plant hormone pathway, and 116 DEGs transcription factors in 5 families were further identified and analyzed. Finally, we deduced the potential mechanism of UV-B-promoting flavonoid formation to neutralize ROS damage derived from UV-B radiation in D. cambodiana based on the gene co-expression network and previous studies from other plants. Considering that wild dragon tree populations are currently highly threatened by anthropogenic and natural stressors, the interactive studies between D. cambodiana plants and UV-B radiation provide valuable information toward understanding the mechanism of dragon’s blood formation and help us reveal the evolution of D. cambodiana, with the eventual goal of aiding in the global conservation of this precious biological resource.

Long-term variability of human health-related solar ultraviolet-B radiation doses for the 1980s to the end of the 21st century.

Solar ultraviolet-B (UV-B) radiation has played a crucial role in the evolution of life on Earth, and potential changes in its levels could affect the health and functionality of humans and the ecosystems. UV exposure presents both risks and benefits to humans. However, optimal UV-B radiation exposure depends on several environmental and physiological factors and cannot be easily determined. The present document provides a review of the current state of knowledge relative to the effects of UV-B radiation on human health. A brief description of the physical mechanisms that control the levels of solar UV-B radiation at the Earth's surface is provided, with special emphasis on the role of ozone and the importance of the Montreal Protocol. A comprehensive review of studies reporting current trends in levels of surface solar UV-B radiation and projections of future levels reveals the dominant role of climatic changes in the long-term variability of UV-B radiation and its impact on the development of melanomas as well as eye disorders. The review provides strong evidence that despite the success of the Montreal Protocol and the expected ozone recovery, the future evolution of the levels of solar UV-B radiation at the Earth's surface is not certain.

Andrographolide protects acute ultraviolet-B radiation-induced photodamages in the mouse skin.

This study evaluates the preventive role of andrographolide (ADP) against ultraviolet-B (UVB) radiation-induced photoaging in the mouse skin. Grouped mice were subjected to UVB irradiation at the dose of 180mJ/cm2 for ten consecutive days. The ADP (3.6mgkg/b.wt) was topically administrated 1h before each UVB-irradiation. We observed that UVB irradiation enhances lipid peroxidative thiobarbituric acid reactive substances (TBARS) and attenuated antioxidant enzyme activities in the mouse skin. Conversely, the ADP pretreatment reversed the lipid peroxidation and restored antioxidant status in the mouse skin. Histological observations clearly show the preventive effect of ADP against UVB-induced dermal tissue damage. Further, ADP slow-down the UVB-mediated expression of inflammatory proteins such as CD34, iNOS, NF-κB COX-2, IL-6, and IL-10 in the mouse skin. Moreover, western blotting studies revealed that ADP prevents UVB exposure-mediated apoptosis markers p53 overexpression in the mouse skin. Thus, ADP protects mouse skin from UVB radiation exposure through impeding inflammation, oxidative stress, and apoptosis.

Analysis of Indoor Radon Data Using Bayesian, Random Binning, and Maximum Entropy Methods.

Three statistical methods: Bayesian, randomized data binning and Maximum Entropy Method (MEM) are described and applied in the analysis of US radon data taken from the US registry. Two confounding factors—elevation of inhabited dwellings, and UVB (ultra-violet B) radiation exposure—were considered to be most correlated with the frequency of lung cancer occurrence. MEM was found to be particularly useful in extracting meaningful results from epidemiology data containing such confounding factors. In model testing, MEM proved to be more effective than the least-squares method (even via Bayesian analysis) or multi-parameter analysis, routinely applied in epidemiology. Our analysis of the available residential radon epidemiology data consistently demonstrates that the relative number of lung cancers decreases with increasing radon concentrations up to about 200 Bq/m3, also decreasing with increasing altitude at which inhabitants live. Correlation between UVB intensity and lung cancer has also been demonstrated.

Inhibition of Ultraviolet‐B Radiation Induced Photodamage by Trigonelline Through Modulation of Mitogen Activating Protein Kinases and Nuclear Factor‐κB Signaling Axis in Skin

Cutaneous photodamage is incited via exposure of ultraviolet-B (UV-B) radiation to skin, characterized by the manifestation of oxidative stress, inflammation, collagen degradation and apoptosis which translates to external aging signs such as wrinkle formation and leathery skin appearance. Meanwhile, it increases cellular susceptibility to photocarcinogenesis. Several studies have accumulated evidence regarding the usage of natural agents in reversing the clinical signs of photoaging as well as preventing photo-toxicity at molecular level. In this study, we have explored the therapeutic potential of natural agent Trigonelline (TG) against UV-B radiation mediated skin photodamage. Various parameters modulated by the exposure of UV-B radiation were investigated in human skin cells and chronic photodamage mice model (Balb/c). We found that TG alleviates UV-B radiation induced photodamage in human skin cells and Balb/c skin mice. TG treatment in UV-B irradiated skin cells abates UV-B radiation mediated phototoxicity, oxidative stress, inflammation and apoptosis. At molecular level, we observed TG treatment significantly prevents the reactive oxygen species (ROS) generation and lipid peroxidation, restores collagen synthesis and matrix metalloproteinase (MMPs) levels. The in vitro findings were replicated in the in vivo model. We found that the TG acts potentially via modulation of ROS-MAPKs-NF-κB axis. Collectively, we propose that TG acts antagonistically against UV-B mediated skin damage and has strong potential to be developed as a therapeutic and cosmetical agent against photodamage disorders.

Identification of the bHLH gene family in Dracaena cambodiana reveals candidate genes involved in flavonoid biosynthesis

The basic helix-loop-helix (bHLH) transcription factors (TFs) regulate multiple biological processes by altering the expression of target genes. However, the bHLH superfamily genes in the dragon tree (Dracaena cambodiana Pierre ex Gagnep) have not been characterized, and their role in flavonoid biosynthesis remain unclear. Here, 62 bHLH genes in D. cambodiana were identified, and these genes were classified into 19 subfamilies based on a phylogenetic comparison with proteins from Arabidopsis. Transcriptomic analysis revealed that 8 DcbHLHs were up-regulated when exposed to the inducer of dragon’s blood and ultraviolet radiation b (UVB). Interaction network analysis revealed that 3 bHLHs (DcbHLH2, DcbHLH5 and DcbHLH56) are likely involved in flavonoid biosynthesis. In addition, DcbHLH5 and flavonoids biosynthetic genes were co-expressed in response to the inducer and UVB exposure. Furthermore, DcbHLH5 could bind to the promoters of flavonoid biosynthesis genes and activate their expression. This study suggests that DcbHLH proteins maybe involved in flavonoid biosynthesis.

Human umbilical cord mesenchymal stem cell-derived and dermal fibroblast-derived extracellular vesicles protect dermal fibroblasts from ultraviolet radiation-induced photoaging in vitro.

Ultraviolet B (UVB) radiation is a major cause of aging in dermal fibroblasts. Human umbilical cord mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) show antioxidant activity. In this study, the anti-aging effects of MSC-EVs on dermal fibroblast photoaging induced by UVB radiation were evaluated, and the effects of extracellular vesicles derived from dermal fibroblasts (Fb-EVs) were compared. Human umbilical cord mesenchymal stem cells and human dermal fibroblasts were cultured, and MSC-EVs and Fb-EVs were isolated and characterized. Human dermal fibroblasts were cultured in the absence or presence of different concentrations of EVs 24 hours prior to UVB radiation exposure. Cell proliferation and cell cycle were evaluated, and senescent cells and intracellular ROS were detected. The expressions of matrix metalloproteinase-1 (MMP-1), extracellular matrix protein collagen type 1 (Col-1), and antioxidant proteins such as glutathione peroxidase 1 (GPX-1), superoxide dismutase (SOD), and catalase were also analyzed. Pretreatment with MSC-EVs or Fb-EVs significantly inhibited the production of ROS induced by UVB radiation, increased dermal fibroblast proliferation, protected cells against UVB-induced cell death and cell cycle arrest, and remarkably decreased the percentage of aged cells. Pretreatment with MSC-EVs or Fb-EVs promoted the expressions of GPX-1 and Col-1 and decreased the expression of MMP-1. Both MSC-EVs and Fb-EVs protected dermal fibroblasts from UVB-induced photoaging, likely through their antioxidant activity.

Ultraviolet-B radiation exposure lowers the antioxidant capacity in the Arabidopsis thaliana pdx1.3-1 mutant and leads to glucosinolate biosynthesis alteration in both wild type and mutant

Pyridoxine (vitamin B6) and its vitamers are used by living organisms both as enzymatic cofactors and as antioxidants. We used Arabidopsis pyridoxine biosynthesis mutant pdx1.3-1 to study the involvement of the PLP-synthase main polypeptide PDX1 in plant responses to ultraviolet radiation of two different qualities, one containing primarily UV-A (315–400 nm) and the other containing both UV-A and UV-B (280–315 nm). The antioxidant capacity and the flavonoid and glucosinolate (GS) profiles were examined. As an indicator of stress, Fv/Fm of photosystem II reaction centers was used. In pdx1.3-1, UV−A + B exposure led to a significant 5% decrease in Fv/Fm on the last day (day 15), indicating mild stress at this time point. The antioxidant capacity of Col-0 wildtype increased significantly (50–73%) after 1 and 3 days of UV−A + B. Instead, in pdx1.3-1, the antioxidant capacity significantly decreased by 44–52% over the same time period, proving the importance of a full complement of functional PDX1 genes for the detoxification of reactive oxygen species. There were no significant changes in the flavonoid glycoside profile under any light condition. However, the GS profile was significantly altered, both with respect to Arabidopsis accession and exposure to UV. The difference in flavonoid and GS profiles reflects that the GS biosynthesis pathway contains at least one pyridoxine-dependent enzyme, whereas no such enzyme is used in flavonoid biosynthesis. Also, there was strong correlation between the antioxidant capacity and the content of some GS compounds. Our results show that vitamin B6 vitamers, functioning both as antioxidants and co-factors, are of importance for the physiological fitness of plants.

The Effect of Ultraviolet-B Radiation Exposure on Hibiscus cannabinus Linn with its Phytochemical and Pharmacological Responses

Aim: Exposure of UV-B Radiation on Hibiscus cannabinus Linn and to evaluate both Phytochemical and Pharmacological responses. Objective: UV-B can increase the nutrients contents from plants, plants become tastier and increases the yield and UV-will make plants resistant to fungal infections and stimulate the production of phytomarkers. Material and Methods: Hibiscus cannabinus Linn seeds were soaked with 50% H2O2 for 12 hours and then inoculated with the Azospirillum. UV-B radiation cabinet was provided by UV lamps which are above 15 cm from control maintained. Normal daylight exposed control group Seedlings irradiated for 2 hour per day (12.30 pm to 2.30 pm) for 20 days. Results and Discussion: 20 days treatment increases total phenolic contents and sugars content were decreased in uncovered leaf tissue. Protein content was at first diminished but expanded on the 20 day of UV-B treatment. Also, perform pharmacological studies no toxic elements are observed 20 days treatment and pharmacological activity by using anticancer activity by using human colon HT-29 cancer cell line. UV light exposed plants shows good anticancer activity when compared with non UV exposed Plant.

Cooler temperatures slow the repair of DNA damage in tadpoles exposed to ultraviolet radiation: Implications for amphibian declines at high altitude.

Ultraviolet B radiation (UVBR) damages the DNA of exposed cells, causing dimers to form between adjacent pyrimidine nucleotides. These dimers block DNA replication, causing mutations and apoptosis. Most organisms utilize biochemical or biophysical DNA repair strategies to restore DNA structure; however, as with most biological reactions, these processes are likely to be thermally sensitive. Tadpoles exposed to elevated UVBR at low environmental temperatures have significantly higher rates of mortality and developmental deformities compared with tadpoles exposed to the same levels of UVBR at higher environmental temperatures. We hypothesized that low environmental temperatures impair the primary enzymatic (photolyase) DNA repair pathway in amphibians, leading to the accumulation of DNA damage. To test this hypothesis, we compared DNA repair rates and photolyase gene expression patterns in Limnodynastes peronii. Tadpoles were acutely exposed to UVBR for 1hr at either 20 or 30°C, and we measured DNA damage and photolyase expression levels at intervals following this exposure. Temperature had a significant effect on the rate of DNA repair, with repair at 30°C occurring twice as fast as repair at 20°C. Photolyase gene expression (6-4 PP and CPD) was significantly upregulated by UVBR exposure, with expression levels increasing within 6hr of UVBR exposure. CPD expression levels were not significantly affected by temperature, but 6-4 PP expression was significantly higher in tadpoles in the 30°C treatment within 12hr of UVBR exposure. These data support the hypothesis that DNA repair rates are thermally sensitive in tadpoles and may explain why enigmatic amphibian declines are higher in montane regions where UVBR levels are naturally elevated and environmental temperatures are lower.

Exploring the link between ultraviolet B radiation and immune function in amphibians: implications for emerging infectious diseases

Amphibian populations the world over are under threat of extinction, with as many as 40% of assessed species listed as threatened under IUCN Red List criteria (a significantly higher proportion than other vertebrate group). Amongst the key threats to amphibian species is the emergence of novel infectious diseases, which have been implicated in the catastrophic amphibian population declines and extinctions seen in many parts of the world. The recent emergence of these diseases coincides with increased ambient levels of ultraviolet B radiation (UVBR) due to anthropogenic thinning of the Earth's protective ozone layer, raising questions about potential interactions between UVBR exposure and disease in amphibians. While reasonably well documented in other vertebrate groups (particularly mammals), the immunosuppressive capacity of UVBR and the potential for it to influence disease outcomes has been largely overlooked in amphibians. Herein, we review the evidence for UVBR-associated immune system disruption in amphibians and identify a number of direct and indirect pathways through which UVBR may influence immune function and disease susceptibility in amphibians. By exploring the physiological mechanisms through which UVBR may affect host immune function, we demonstrate how ambient UVBR could increase amphibian susceptibility to disease. We conclude by discussing the potential implications of elevated UVBR for inter and intraspecific differences in disease dynamics and discuss how future research in this field may be directed to improve our understanding of the role that UVBR plays in amphibian immune function.

The impact of ultraviolet B (UV-B) radiation in combination with different temperatures in the early life stage of zebrafish (Danio rerio).

Ultraviolet B (UV-B) radiation is an environmental stressor with detrimental effects on many aquatic organisms including fish. In addition, UV-B exposure combined with other environmental factors could have even more negative effects. The purpose of this study was to investigate the effect of UV-B radiation exposure on zebrafish embryos/larvae in terms of survival, developmental toxicity and the mRNA levels of the genes related to oxidative stress and innate immune response at different temperatures (24 °C, 28 °C and 30 °C). Zebrafish embryos were exposed to 3.3 W m-2 UV-B radiation and/or 24 °C, 28 °C (for the control) and 30 °C temperatures between 4 and 96 h post-fertilization. The mortality, hatching rate, malformations and heartbeat rate were evaluated. The results demonstrated that UV-B exposure or different temperatures (24 °C and 30 °C) induced developmental toxicity, including delayed hatching, increased the occurrence of malformations, and reduced the heartbeat rate and survival. The combined exposure to UV-B and different temperatures (24 °C and 30 °C) resulted in greater adverse effects on embryonic development. Furthermore, RT-PCR results showed that the mRNA levels of superoxide dismutase 1 (sod1), catalase 1 (cat1), heat shock protein 70 (hsp70), interleukin-1 beta (il-1β) and tumor necrosis factor alpha (tnfα) genes were significantly up-regulated in all of the treatment groups. These results revealed that the interaction between UV-B and temperature impaired the development of zebrafish embryos and disrupted their metabolism.

Developmental Phase-Specific Mortality After Ultraviolet-B Radiation Exposure in the Two-Spotted Spider Mite.

Exposure to ambient ultraviolet-B (UVB) radiation generates DNA lesions, such as cyclobutane pyrimidine dimers and 6-4 pyrimidine-pyrimidine photoproducts in Tetranychus urticae Koch (Acari: Tetranychidae). Larvae appeared normal and healthy after UVB irradiation. Conversely, many mites were trapped in their old epidermis or experienced retarded development and shrunk, thus failing to molt from protochrysalises to protonymphs and died. This suggested that DNA lesions per se were not causing lethality in mites unless damaged genes were expressed. UVB-induced DNA lesions may have interfered with DNA replication and gene expression during the physiological changes of morphogenesis in the chrysalis stage. Comprehensive gene expression analysis by RNA sequencing revealed that gene expression involving epidermal tissue (characteristically cuticular protein genes) and myosin heavy chain muscle-like genes were downregulated in protochrysalises irradiated with UVB at the larval stage. We conclude that the success of protochrysalis molting is determined by whether the DNA lesions of genes, particularly those connected with morphogenesis, are repaired before expression at the protochrysalis stage.

Effect of UVB radiation exposure in the expression of genes and proteins related to apoptosis in freshwater prawn embryos

Our previous studies showed that embryos of the freshwater prawn Macrobrachium olfersii exposed to ultraviolet B (UVB) radiation exhibited DNA damage, excessive ROS production, mitochondrial dysfunction and increased hsp70 expression, which are able, independently or together, to induce apoptosis. Thus, we attempted to elucidate some key apoptosis-related genes (ARG) and apoptosis-related proteins (ARP) and their expression during different stages of embryonic development, as well as to characterize the chronology of ARG expression and ARP contents after UVB radiation insult. We demonstrate that p53, Bax and Caspase3 genes are active in the embryonic cells at early embryonic developmental stages, and that the Bcl2 gene is active from the mid-embryonic stage. After UVB radiation exposure, we found an increase in ARP such as p53 and Bak after 3h of exposure. Moreover, an increase in ARG transcript levels for p53, Bax, Bcl2 and Caspase3 was observed at 6h after UVB exposure. Then, after 12h of UVB radiation exposure, an increase in Caspase3 gene expression and protein was observed, concomitantly with an increased number of apoptotic cells. Our data reveal that ARG and ARP are developmentally regulated in embryonic cells of M. olfersii and that UVB radiation causes apoptosis after 12h of exposure. Overall, we demonstrate that embryonic cells of M. olfersii are able to active the cell machinery against environmental changes, such as increased incidence of UVB radiation in aquatic ecosystems.

Cell damage caused by ultraviolet B radiation in the desert cyanobacterium Phormidium tenue and its recovery process

Phormidium tenue, a cyanobacterium that grows in the topsoil of biological soil crusts (BSCs), has the highest recovery rate among desert crust cyanobacteria after exposure to ultraviolet B (UV-B) radiation. However, the mechanism underlying its recovery process is unclear. To address this issue, we measured chlorophyll a fluorescence, generation of reactive oxygen species (ROS), lipid peroxidation, and repair of DNA breakage in P. tenue following exposure to UV-B. We found that UV-B radiation at all doses tested reduced photosynthesis and induced cell damage in P. tenue. However, P. tenue responded to UV-B radiation by rapidly reducing photosynthetic activity, which protects the cell by leaking less ROS. Antioxidant enzymes, DNA damage repair systems, and UV absorbing pigments were then induced to mitigate the damage caused by UV-B radiation. The addition of exogenous antioxidant chemicals ascorbate and N-acetylcysteine also mitigated the harmful effects caused by UV-B radiation and enhanced the recovery process. These chemicals could aid in the resistance of P. tenue to the exposure of intense UV-B radiation in desertified areas when inoculated onto the sand surface to form artificial algal crusts.

Photoprotective effect of coumarin and 3-hydroxycoumarin in sea urchin gametes and embryonic cells

Ultraviolet radiation B (UVB) represents 5% of all solar UV radiation and chronic exposure can induce harmful biological responses, including skin cancer. Prospection of new drugs with photoprotective properties and less toxic effects is constant and natural products have been the main options in this field. Coumarins are a group of natural phenolic compounds that shows several pharmacological activities. The aim of present work was to investigate the effect of coumarin and six derivatives in sea urchin gametes and zygotes exposed to UVB. Embryonic development assay was used to monitor UVB embryotoxicity. Firstly, we demonstrated that coumarin inhibited first embryonic cell division from 5μM (EC50=52.9μM) and its derivatives showed an embryotoxic effect ten times higher. Then, gametes or zygotes were treated with coumarin compounds before or after UVB exposure (UVB doses ranged from 0.056 to 0.9kJm−2). Pretreatment of gametes or zygotes with coumarin or 3-hydroxycoumarin (1μM, both) decreased UVB embryotoxic effect. Protective effect of the compounds was observed only when cells were treated previous to UVB exposure. Coumarin derivatives 4-hydroxycoumarin, 6-hydroxycoumarin, 7-hydroxycoumarin, 6,7-dihydroxycoumarin and 6-methoxy-7-hydroxycoumarin did not exhibit photoprotective activity. Our data provides evidences that coumarin and 3-hydroxycoumarin can be a promising class of photoprotective drugs.
