Blood profile and liver histopathological analysis of broiler chickens fed dried cashew apples (Anacardium occidentale L.)

This study utilized a total of 210 one-day-old chicks to evaluate the blood profile and liver histopathology of Ross 308 broiler chickens fed diets containing Dried Cashew Apples (DCA). The chicks were allocated into seven treatment groups, each consisting of three replicates of 10 birds. The experimental design used a (2 x 3) +1 augmented factorial in a completely randomized arrangement. Diet 1 was the control (maize-soybean meal-based), while diets 2, 3, and 4 contained 10%, 20%, and 30% DCA without enzyme supplementation. Diets 5, 6, and 7 contained 10%, 20%, and 30% DCA with exogenous microbial enzyme supplementation. Data on haematological indices, serum biochemical indices, and liver histology were collected and analyzed using SPSS version 25, with means separated by Duncan’s Multiple Range Test of the same statistical package at p < 0.05. The results indicated that DCA inclusion, with or without enzymes, significantly influenced all haematological parameters except MCV, showing lower PCV, Hb concentration, and RBC counts in DCA groups compared to the control. Enzyme supplementation at 20% DCA increased PCV, while at 10% and 30% DCA, it decreased Hb concentrations and increased WBC counts at 30% DCA, with varying effects on neutrophils and lymphocytes. Serum biochemistry showed that DCA inclusion affected all parameters except AST, with the highest ALT values in birds on 10% DCA without enzymes and the lowest in the control group. Enzyme supplementation significantly influenced AST, ALT, TP, globulin, albumin, urea, and creatinine levels, particularly at 20% DCA. Liver histology showed no lesions in control birds but varying degrees of necrosis, inflammation, atrophy, and congestion in DCA-fed birds, worsening with higher DCA and enzyme levels. In conclusion, broilers can tolerate 10% DCA in their diet, but higher levels lead to physiological stress, necessitating careful consideration of DCA inclusion levels to maintain bird health and welfare.

The effects of incorporating dried cashew apple in the diet of broiler chickens on growth performance, total tract digestibility, and gut health

A total of two hundred and ten one-day-old Ross 308 broiler chicks were utilized in this study to investigate the effects of incorporating Dried Cashew Apples (DCA) in the diet of broiler chickens on growth performance, total tract digestibility, and gut health. The birds were weighed and randomly allotted to seven treatment groups with three replicates, each having 10 birds. The experimental design was a (2 X 3) + 1 augmented factorial arrangement in a completely randomized design. Diet D1 (control diet) was a maize-soybean meal-based; diets D2, D3, and D4 comprised 10, 20, and 30% DCA without exogenous enzyme supplementation, whereas diets D5, D6, and D7 included 10, 20, and 30% DCA treated with exogenous enzyme. Data were collected on growth performance, total tract digestibility, and gut morphology. Collected data were analyzed using SAS (2002) and separation of means was done using Duncan’s Multiple Range Tests of the same statistical software. The difference was set at p < 0.05. The study found that birds fed diets containing 10% DCA, with or without enzyme supplementation (D2 and D5), had a significantly higher Average Daily Gain (ADG) compared to those fed the control diet (D1). The ADG of birds fed diets D3 and D6 was similar to that of birds fed the control diet (D1). However, a significant decrease in ADG was observed as the dietary inclusion level of DCA increased to 30%, as seen in birds offered diets D4 and D7. In addition, a significant linear rise (p < 0.05) in Average Daily Feed Intake (ADFI) was seen when DCA was included in the diet. In addition, birds that were given diets containing DCA showed significantly higher nutrient digestibility (p < 0.05) compared to birds that were given the control diet (D1), except for birds that were given a diet containing 30% DCA without enzyme supplementation (D4), which showed significantly lower nutrient digestibility (p < 0.05) compared to birds that were given the control diet (D1). Ultimately, birds that were given meals containing DCA exhibited a significant enhancement in GIT development compared to birds on the control diet. Based on the findings of this study, it can be concluded that including DCA in the diet of broiler chickens at a level of up to 20% does not have any detrimental effects on their performance.

Influence of various methods of processing soybeans on protein digestibility and reduction of nitrogen deposits in the natural environment – a review

Abstract Soybean meal (SBM) is a significant source of protein for poultry due to its high concentration of protein and amino acids profile. The primary objective of SBM processing is the reduction and deactivation of anti-nutritional factors (ANFs) to enhance nutrient digestibility. However, eliminating ANFs does not necessarily correlate with increased protein and amino acid content. Several processing methods, such as soaking, cooking, and commercial techniques, improve SBM protein digestibility. To sustain feeding qualities, soybeans must undergo heating to remove inherent chemicals that interfering with poultry digestion. However, both under and over-processing can impair meal digestibility. Extrusion heating significantly contributes to protein and starch digestibility while reducing certain ANFs. Thermomechanical and enzyme-assisted procedures reduce heat-resistant proteins more efficiently than extraction methods, producing soy protein concentrate (SPC) and fermentation. These techniques reduce ANFs in SBM, activating digestive enzymes and alleviating digestive tract pressure, leading to decreased endogenous protein deficits. Exogenous enzyme supplementation is a biotechnological approach for enhancing the nutritional content of SBM and certain other protein-rich products. However, different processing methods not only affect protein digestibility but also impact poultry production, gastrointestinal health, and the environment due to higher nitrogen excrement. Ammonia (NH3) emissions, are a significant environmental concern in chicken farming, resulting from uric acid production during protein consumption. It has a negative impact on the environment and the health of birds/humans. This review investigates the impact of several processing techniques on the digestibility of SBM protein and the reduction of N2 regarding one health concept. Future research should focus on identifying optimal processing methods maximizing protein digestibility while minimizing environmental nitrogen impacts.

Effects of enzyme supplementation on growth performance, digestibility of phosphorus, femur parameters and fecal microbiota in growing pigs fed different types of diets.

A 42-days study was conducted to evaluate the effects of different dietary types (corn-or wheat-soybean meal-based diet) and phytase (Phy) or a multi-carbohydrase and phytase complex (MCPC) supplementation on growth performance, digestibility of phosphorus (P), intestinal transporter gene expression, plasma indexes, bone parameters, and fecal microbiota in growing pigs. Seventy-two barrows (average initial body weight of 24.70 ± 0.09 kg) with a 2 × 3 factorial arrangement of treatments and main effects of diet type (corn-or wheat-soybean meal-based-diets) and enzyme supplementation (without, with Phy or with MCPC). Each group was designed with 6 replicate pens. The MCPC increased (p < 0.05) average daily gain (ADG) and final body weight (BW). A significant interaction (p = 0.01) was observed between diet type and enzyme supplementation on apparent total tract digestibility (ATTD) of P. The ATTD of P was higher (p < 0.05) in wheat soybean meal-based diets compared to corn-soybean meal-based diets. Compared with the corn-soybean meal-based diet, the relative expression of SLC34A2 and VDR genes in the ileum and SLC34A3 in jejunum of growing pigs fed the wheat-soybean meal based diet was lower (p < 0.05). The MCPC significantly reduced (p < 0.05) the relative expression of TRPV5 and CALB1 genes in the ileum and increased the expression of CALB1 in the duodenum compared to control diet. The phytase increased (p < 0.05) the relative expression of SLC34A1 gene in the duodenum in comparison to control diet and MCPC-supplemented diet. The Ca and P contents in plasma from pigs fed corn-soybean meal-based diet were higher (p < 0.05) than those from pigs fed wheat-soybean meal-based diet, and the parathyroid hormone (PTH) and calcitonin (CT) concentrations were lower (p < 0.05) than those fed wheat-soybean meal-based diet. The content of Ca and P in the femur and the bone strength of pigs in the corn-soybean meal group were significantly higher (p < 0.05) than those in the wheat-soybean meal groups. The phytase increased (p < 0.05) the Ca and P content and bone strength of the femur. Additionally, diet type and both enzymes significantly improved fecal microbial diversity and composition. Taken together, diet type and exogenous enzymes supplementation could differently influence the growth performance, utilization of phosphorus, intestinal transporter gene expression, bone mineralization and microbial diversity and composition in growing pigs.

339 An Evaluation of the Effects of Feeding an Exogenous Fibrolytic Enzyme Product on the Growth Performance and Apparent Nutrient Digestibility of Growing Bison Calves Fed in a Commercial Feed Yard

Abstract While exogenous enzyme supplementation has been tested in ruminants as early as the 1960’s, industry adoption of such technologies has been slow. With high feed costs, technologies such as feed enzyme could potentially help producers to lower the production cost by improving feed efficiency. A 69-day growing experiment was conducted at a commercial feed yard in Western NE using bison calves [n = 228; initial body weight (BW) = 269 kg, SD = 41 kg] to determine the effects of supplementation of an exogenous fibrolytic enzyme from T. reesei on calf dry matter intake (DMI), growth performance and nutrient apparent digestibility. A completely randomized design was utilized with two treatments, control (CON) or enzyme (CX), and four pens, with calves assigned randomly to pen and pen assigned to TRT (48 to 66 calves per pen). Individual calf BW were obtained on day 0, day 35, and day 69. The DMI was measured daily by pen, with feed offered adjusted daily and fecal samples were composited and analyzed on pen-basis. Data were analyzed using JMP statistical software, with initial BW included as a covariate in the analyses. Average daily gain and DMI in calves fed the CX treatment was 7% greater (0.895 vs. 0.830 kg/d; P = 0.02), and 11% less (7.83 vs. 8.75 kg/d; P = 0.02) than in calves fed the CON treatment, respectively, resulting a 18% numeric improvement in feed efficiency for calves fed the CX treatment compared with calves fed the CON treatment. Apparent starch digestibility tended to improve (P = 0.07) by 15% in calves fed the CX treatment compared with calves fed the CON treatment, while apparent neutral detergent fiber (NDF) digestibility (as a percent of NDF) was numerically improved by 9% in calves fed the CX treatment compared with calves fed the CON treatment. Although the mode of action warrants further investigation, these data suggest that feeding an exogenous fibrolytic enzyme product may be beneficial for growing bison calves, likely in part because of greater apparent nutrient digestibility.

Effect of exogenous fibrolytic enzymes supplementation to improve voluntary intake, availability of nutrients and growth performance in weaned crossbred calves

The present study was carried out to evaluate the effect of supplementation of exogenous fibrolytic enzymes (EFE) on voluntary feed intake, availability of different nutrients and growth performance in weaned crossbred calves. Fifteen weaned healthy female Jersey crossbred calves (weight 79.73±3.46 kg; age 234.5±11.33 days) were divided equally into three groups viz., Control (T0), Treatment-1 (T1) and Treatment-2 (T2). Growth trial was conducted on these calves for 90 days. Animals in each treatment group were fed ad lib. total mixed ration (TMR) (CP 12%, TDN 65%) individually for 90 days. Calves under control (T0) group were fed ad lib. TMR (without EFE). Calves under T1 and T2 were fed ad lib. TMR supplemented with EFE cellulase and xylanase @ 8000 and 16000 IU/kg DM of TMR and with EFE cellulase and xylanase @ 12000 and 24000 IU/kg DM of TMR, respectively. Significantly greater DMI per unit body weight (kg/100 kg BW and g/kg W0.75) was recorded in enzyme supplemented groups compared to control group; however, difference between T1 and T2 was non-significant. Addition of EFE in the TMR diet of calves increased TDN and DCP intake (g/kg W0.75) in T1 and T2 than T0 . Supplementation of EFE also enhanced TDN (%), DCP (%), DNDF (%), DE (Mcal/kg of feed) and ME (Mcal/kg of feed) value of the diets under T1 and T2 than T0. Average daily weight gain (g/d/calf) was estimated greater in T1 and T2 than T0. The feed conversion efficiency was similar among three treatments. Whereas, plasma total protein and albumin concentration increased significantly due to supplementation of EFE in the diet of calves. Therefore, it may be concluded that supplementation of exogenous fibrolytic enzymes (cellulase and xylanase @ 8000 and 16000 IU/kg TMR DM) may boost growth potential of finisher Jersey crossbred calves with enhanced intake of nutrients.

Effect of exogenous fibrolytic enzymes supplementation on nutrient intake and digestibility in Black Bengal kids

The present study was carried out to evaluate the effect of supplementation of exogenous fibrolytic enzymes (EFE) to the Total Mixed Ration (TMR) at two different levels on intake, nutrient digestibility and nutrient availability in Black Bengal kids. A digestibility trial was conducted on 15 post-weaned Black Bengal kids divided into three groups. Control (T0) group was fed ad lib. TMR was prepared from concentrate mixture and green fodder @ 40: 60 on DM basis. T1 and T2 groups were supplemented with EFE cellulase and xylanase @ 8000 and 16000 IU/kg TMR DM and 12000 and 24000 IU/kg TMR DM, respectively. The study revealed significantly higher digestibility of nutrients, i.e. DM, OM, EE, NDF, ADF, hemicellulose, cellulose and total carbohydrates in both enzyme supplemented groups than the control group. However, the digestibility of CP, cellulose and hemicellulose was significantly higher only in T2 group than the control. There was no significant difference between two enzyme supplemented groups in terms of digestibility of different nutrients except for CP which was significantly higher in T2 than T1. The difference in voluntary intake of DM and OM were non-significant but the intake of CP, DCP and TDN were higher in enzyme supplemented groups. Similarly, TDN, DE and ME content of the diet were also higher in two enzyme supplemented groups than control. However, there was no significant difference between T1 and T2 in terms of nutrient availability and intake. Based on the present study, supplementation of EFE @ cellulase 8000 and xylanase 16000 IU/ kg DMwas found to be optimum for improving the nutrient digestibility and availability in Black Bengal kids, which may further improve the productive and reproductive performance of the animals.

Distillers dried grains with soluble and enzyme inclusion in the diet effects broilers performance, intestinal health, and microbiota composition

This study tested the effect of distillers dried grains with soluble (DDGS) inclusion in a broiler diet, with or without supplementation of exogenous enzymes, on the microbiota composition, intestinal health, diet digestibility and performance. A total of 288 one-day-old chickens was assigned to 6 treatments (8 replicate of 6 birds each) according to a completely randomized design with a 3×2 factorial scheme with 3 DDGS levels (0, 7 and 14%) and 2 inclusions of exogenous enzymes (with or without a multicarbohydrase complex+phytase [MCPC]). The results exhibited that DDGS inclusion up to 14% did not impair broilers performance up to 28 d, however, DDGS-fed animals exhibited significant improvement with the MCPC supplementation. No effects of the enzymes in the ileal digestibility were found at 21 d. DDGS inclusion in the diet affected dry matter and gross energy digestibility. Broilers fed diets with MCPC were found to have less intestinal histological alteration thus better gut health. No effect of DDGS, enzyme or interaction of those were observed for intestinal permeability and in the serum inflammatory biomarker (calprotectin) at 7 and 28 d. The increase of DDGS percentage in the diet reduced the diversity of the ileal microbiota but increased the cecal microbiota diversity. The inclusion of DDGS showed positive effects on microbiota composition due to a reduction of Proteobacteria phylum in the ileum at 28d and a reduction in the presence of Enterococcaceae family in the ileum at 14 and 28d. The inclusion of MCPC complex might promote beneficial changes in the ileal and cecal microbiota due reduce of Proteobacteria, Bacillaceae and Enterobacteriaceae. The supplementation of xylanase, β-glucanase, arabinofuranosidase and phytase to a DDGS diet improves performance and intestinal health allowing the use of these subproduct in the poultry nutrition.

Influence of partial replacement of ground barley with whole barley and exogenous enzyme supplementation in pelleted diets on growth performance, nutrient utilisation and foregut development in broiler starters

A 3 × 2 factorial arrangement of treatments, evaluating three inclusion levels of whole barley (WB; 0, 141, 282 g/kg) in a barley-based diet and two levels of carbohydrase (Carb; 0 and 0.15 g/kg of feed), was used. The AMEn and standardised digestible amino acid contents of the test barley were used in formulating the basal diet. Whole barley was added to the diet at the expense of ground barley (GB; 3.0 mm sieve size). The diets contained 5.0 g/kg titanium dioxide as an inert marker. Each of the six experimental diets was pelleted and offered ad libitum to six replicate cages (eight birds/cage). Body weights and feed intake were recorded weekly throughout the 21-d trial. The AMEn was determined using the classical total excreta collection method (17–20 d). On d-21, ileal digesta were collected from the terminal ileum to determine the coefficient of apparent ileal digestibility of nutrients. Jejunal digesta were collected for the determination of intestinal digesta viscosity. There was no significant (P > 0.05) interaction between WB and Carb for any measured parameter. Compared to the birds fed GB diets, those fed WB at 141 g/kg had a higher (P < 0.05) weight gain by 36 g/bird, with no effect on the feed intake or feed efficiency. The inclusion of WB tended (P = 0.058) to enhance N digestibility from 0.741 in GB diets to 0.775 in diets with 282 g/kg WB. Irrespective of Carb supplementation, compared to the GB diet, WB inclusion at 141 and 282 g/kg improved (P < 0.001) starch digestibility (by 3.6 % and 5.7 %, respectively) and AMEn (by 1.85 % and 2.89 %, respectively). These improvements were not associated with any impact (P > 0.05) on gizzard development. Jejunal digesta viscosity was not influenced (P > 0.05) by WB inclusion. Adding Carb increased starch digestibility and energy utilisation by 2.24 % and 0.12 MJ/kg, respectively, possibly due to 0.35 cP reduction in jejunal digesta viscosity (P < 0.05) by the supplemental Carb. Overall, WB could be included in barley-based broiler starter diets up to 282 g/kg of diet with no adverse effects on performance.

Dietary supplementation with xylooligosaccharides and exogenous enzyme improves milk production, energy utilization efficiency and reduces enteric methane emissions of Jersey cows

BackgroundSustainable strategies for enteric methane (CH4) mitigation of dairy cows have been extensively explored to improve production performance and alleviate environmental pressure. The present study aimed to investigate the effects of dietary xylooligosaccharides (XOS) and exogenous enzyme (EXE) supplementation on milk production, nutrient digestibility, enteric CH4 emissions, energy utilization efficiency of lactating Jersey dairy cows. Forty-eight lactating cows were randomly assigned to one of 4 treatments: (1) control diet (CON), (2) CON with 25 g/d XOS (XOS), (3) CON with 15 g/d EXE (EXE), and (4) CON with 25 g/d XOS and 15 g/d EXE (XOS + EXE). The 60-d experimental period consisted of a 14-d adaptation period and a 46-d sampling period. The enteric CO2 and CH4 emissions and O2 consumption were measured using two GreenFeed units, which were further used to determine the energy utilization efficiency of cows.ResultsCompared with CON, cows fed XOS, EXE or XOS + EXE significantly (P < 0.05) increased milk yield, true protein and fat concentration, and energy-corrected milk yield (ECM)/DM intake, which could be reflected by the significant improvement (P < 0.05) of dietary NDF and ADF digestibility. The results showed that dietary supplementation of XOS, EXE or XOS + EXE significantly (P < 0.05) reduced CH4 emission, CH4/milk yield, and CH4/ECM. Furthermore, cows fed XOS demonstrated highest (P < 0.05) metabolizable energy intake, milk energy output but lowest (P < 0.05) of CH4 energy output and CH4 energy output as a proportion of gross energy intake compared with the remaining treatments.ConclusionsDietary supplementary of XOS, EXE or combination of XOS and EXE contributed to the improvement of lactation performance, nutrient digestibility, and energy utilization efficiency, as well as reduction of enteric CH4 emissions of lactating Jersey cows. This promising mitigation method may need further research to validate its long-term effect and mode of action for dairy cows.

Microbial inulinase promotes fructan hydrolysis under simulated gastric conditions.

Fermentable oligo-, di-, monosaccharides and polyols (FODMAPs) have emerged as key contributors to digestive discomfort and intolerance to certain vegetables, fruits, and plant-based foods. Although strategies exist to minimize FODMAP consumption and exposure, exogenous enzyme supplementation targeting the fructan-type FODMAPs has been underexploited. The objective of this study was to test the hydrolytic efficacy of a food-grade, non-genetically engineered microbial inulinase preparation toward inulin-type fructans in the INFOGEST in vitro static simulation of gastrointestinal (GI) digestion. Purified inulin was shown to undergo acid-mediated hydrolysis at high gastric acidity as well as predominantly inulinase-mediated hydrolysis at lower gastric acidity. Inulinase dose-response simulations of inulin, garlic, and high-fructan meal digestion in the gastric phase suggest that as little as 50 inulinase units (INU) and up to 800 INU per serving promote fructan hydrolysis better than the control simulations without inulinase. Liquid chromatography-mass spectrometry (LC-MS) profiling of fructo-oligosaccharides (FOS) in the gastric digestas following inulinase treatment confirms the fructolytic activity of inulinase under simulated digestive conditions. Altogether, these in vitro digestion data support the use of microbial inulinase as an exogenous enzyme supplement for reducing dietary fructan-type FODMAP exposure.

Effect of phytase and xylanase enzymes on growth performance and Mucin2 gene expression in broiler chickens.

Development of exogenous enzymes is one of the most important discoveries in animal nutrition. The supplementation of exogenous enzymes in broiler diets allows for supplying nutrient deficiencies and to decrease endogenous losses. The effects of phytase (Hostazym and Phyzyme) and xylanase (Ronozyme) enzymes were investigated on growth performance and Mucin2 gene expression in broilers. A completely randomized design was applied, including 7 treatments, 4 replicates and 25 birds per replicates. A total of 700 male Ross (308) broiler chickens were fed with similar diets supplemented by Hostazym and Phyzyme (500 and 1000FTU/kg) and Ronozyme (100 and 200EXU/kg). Weight gain (WG), feed intake (FI) and feed conversion ratio (FCR) were determined for three phases and entire rearing period. On 42 days of age, four birds per replicate were slaughtered. Total RNA was extracted from jejunum samples, and Mucin2 gene expression was measured by real-time PCR. Phytase and xylanase enzymes had a significant effect (p<0.05) on traits (WG and FCR) in grower and finisher phases and whole rearing period, but FI was not affected by enzymes (p>0.05). Carcass (74.13g) and breast (27.76g) weights by Hostazym (1000FTU/kg) were higher than other treatments (p<0.05). Weight of liver, bursa and spleen were significantly influenced by enzymes (p<0.05). Likewise, bursa and spleen weights in Hostazym (1000FTU/kg feed) and Ronozyme (200EXU/kg feed) were significantly higher than other treatments (p<0.05). Mucin2 gene expression was affected by enzymes in whole treatments. The lowest amount of Mucin2 gene expression belonged to Ronozyme (200 and 100EXU/kg), and the highest was belonging to Hostazym (1000 FTU/kg). Phytase enzymes have higher effect on broiler performance and Mucin2 gene expression compared to xylanase. High doses of Hostazym (1000FTU/kg feed) could be supplemented in broiler chicken diets to improve optimum growth and feed efficiency.

Growth performance, nutrient digestibility, antioxidant state, ileal histomorphometry, and cecal ecology of broilers fed on fermented canola meal with and without exogenous enzymes

This study was conducted to evaluate the effects of supplementation of exogenous enzymes in broiler diets that includes fermented canola meal on performance, nutrient digestibility, biochemical indication, antioxidative capacity, digestive enzyme activity, immune responses, and gut health. Five hundred 1-day-old Ross 308 broiler chicks were randomly allocated into five experimental groups (5 replicate/group), the first group: a control (CON) contained a basal diet, and the second to the fifth groups were fed diets as follows: containing 20% canola meal (CM), contains 20% fermented canola meal (FCM), contains 20% canola meal and exogenous enzymes at 0.02%/kg feed (ECM), and contains 20% fermented canola meal and exogenous enzymes at 0.02%/kg feed (EFC), respectively. At the finisher phase, the best body weight gain, feed conversion ratio, and nutrient utilization were associated with chickens fed EFC compared to other groups (P < 0.05). Total protein, albumin, alanine aminotransferase, and superoxide dismutase levels increased (P < 0.05), while cholesterol and malondialdehyde levels decreased in chickens fed on EFC. Likewise, there was a significant increase in the relative weight of the bursa of Fabricius and antibody titer against Newcastle disease, whereas the weight of abdominal fat decreased in the EFC group compared to other groups. Furthermore, there was a significant improvement in the activity of lipase and amylase enzymes (P < 0.05) in the EFC group. Fermented canola meal addition improved gut health (decreased Escherichia coli, increased Lactobacillus, and the highest values of villus height). Overall, these results confirmed that supplementing a fermented canola meal diet with exogenous enzymes improved growth performance through enhancing nutrient digestibility, immunity, antioxidant capacity, and gut health. Thus, adding enzymes to a diet containing fermented canola meal can be recommended as an alternative protein source that could be safely used to replace up to 20% soybean meal in broiler diets.

Effect of exogenous fibrolytic enzymes supplementation on voluntary intake, availability of nutrients and growth performance in Black Bengal kids (Capra hircus)

A Growth experiment was conducted on fifteen weaned Black Bengal kids divided into three groups. Kids under control (T0) group were fed ad libitum Total Mixed Ration (TMR) prepared from concentrate mixture and green fodder @ 40: 60 (on a DM basis). Kids under T1 and T2 groups were supplemented with two levels of EFE (T1 with cellulase and xylanase @ 8000 and 16,000IU/kg TMR DM and T2 with 12,000 and 24,000IU/kg TMR DM). A digestion trial was also conducted during the last phase of the growth trial. The intake of nutrients, like DM, OM, CP, TDN, and DCP (g/kg W0.75) were significantly (P < 0.001) higher in enzyme-supplemented treatments. Supplementation of EFE also increased (P < 0.001) intake of DE and ME (Mcal/ kg W0.75) in Black Bengal kids. The differences in all the blood biochemical parameters and enzymes (glucose, total protein, globulin, albumin, urea, AST, and ALT) were non-significant (P > 0.05) among the three treatment groups. Average daily body weight gain (ADG, g/d/kid) was increased significantly (P < 0.05) by 7.00 % in T1 and 7.19 % in T2 over the control group (T0). The FCE (feed conversion efficiency) was also greater (P < 0.05) in both enzyme-supplemented (T1 and T2) groups compared to the control. The results depicted no additional advantage due to the supplementation of a higher level of an enzyme mixture. Therefore, it may be concluded that the supplementation of EFE @ cellulase 8000 and xylanase 16,000IU/kg diet DM increased the intake of different nutrients and growth performance in weaned Black Bengal kids.

Effects of Barley- and Oat-Based Diets on Some Gut Parameters and Microbiota Composition of the Small Intestine and Ceca of Broiler Chicken

Barley and oats can be alternatives of corn and wheat in poultry nutrition and used at higher inclusion rates. Both cereals contain hulls, a structural fiber source, that can be beneficial for the gizzard function of birds. They also contain high amounts of β-glucans, of which about 60–70% is water soluble. Soluble β-glucans increase gut viscosity, impair digestion, and modify gut microbiota. The aim of this trial was to evaluate the effects of feeding oats and barley at high inclusion rates and with exogenous glucanase on some relevant gut parameters and the microbiota composition of jejunum content (JC), jejunum mucosa (JM), and cecal content (CC). A total of 360 male, Ross 308 broiler chickens were allocated randomly into three treatment groups of 5 replicate pens with 24 chickens. Beside a corn and soybean meal-based control diet (C), a barley (B)- and oat (O)-based treatment was used. In all feeding phases, barley was fed at 40, while oats at 20% inclusion rate. At day 40, 10 birds per treatment were slaughtered and gut viscosity, the cecal short chain fatty acid (SCFA) concentration, and the microbiota composition of the different gut parts determined. In spite of the glucanase enzyme addition, the barley-based diet significantly increased the viscosity of the ileal content and this was also the diversity of the bacteriota in the small intestine. On the other hand, this treatment decreased the microbial diversity in the ceca and resulted in lower SCFA contents. Barley increased the abundance of the phyla Bacteroidetes and decreased that of Firmicutes and some of them genera in the JC and CC. Oats had only a slight effect on the measured parameters. The results highlight the importance of also taking into account the soluble fiber fractions of the feedstuffs in diet formulation and to adapt the exogenous enzyme supplementation of to the actual soluble fiber contents.

Impact of mashing protocol on the formation of fermentable sugars from millet in gluten-free brewing

The production of fermentable sugars (FS) in gluten-free (GF) brewing is hindered by the high starch gelatinization temperatures of GF malts and lower diastatic power compared to barley malt. Our previous work has demonstrated that starch gelatinization was the primary hurdle, and when decoupled from a single mash phase, high concentrations of FS could be produced. However, more research was required to improve the applicability of GF brewing. In this study, millet was used as a model GF malt demonstrating that despite the low α-amylase and β-amylase activities compared to barley malt ∼ 90 % of the FS (∼110 g/L) could be produced within 40 min. Limitations to enzyme extraction and separation due to coarse milling and lautering initially limited FS by ∼ 30 g/L, requiring additional processing or exogenous enzyme supplements that improved fermentable sugar generation by ∼ 20 g/L. Overall, millet is a promising brewing ingredient, provided appropriate mashing procedures are implemented.

Arazyme in combination with dietary carbohydrolases influences odor emission and gut microbiome in growing-finishing pigs

This study evaluated the effects of supplementing feed with arazyme and dietary carbohydrolases derived from invertebrate gut-associated symbionts on the noxious gas emissions, gut microbiota, and host-microbiome interactions of pigs. Here, 270 and 260 growing pigs were assigned to control and treatment groups, respectively. The tested feed additives contained a mixture of arazyme (2,500,000 Unit/kg) and synergetic enzymes, xylanase (200,000 Unit/kg) and mannanase (200,000 Unit/kg), derived from insect gut-associated symbionts in a 7.5:1:1 ratio. The control group was fed a basal diet and the treatment group was fed the basal diet supplemented with 0.1 % enzyme mixture (v/v) for 2 months. Odorous gases were monitored in ventilated air from tested houses. Fecal samples were collected from steel plate under the cage at the completion of the experiment to determine chemical composition, odor emissions, and bacterial communities. There was a significant decrease in the concentration of NH3 (22.5 vs. 11.2 ppm; P < 0.05), H2S (7.35 vs. 3.74 ppm; P < 0.05), trimethylamine (TMA) (0.066 vs. 0.001 ppm; P < 0.05), and p-cresol (0.004 ppm vs. 0 ppm; P < 0.05) at 56 d in treatment group compared with the control group. Moreover, fecal analysis results showed that exogenous enzyme supplementation caused a reduction in VFAs and indole content with approximately >60 % and 72.7 %, respectively. The result of gas emission analysis showed that NH3 (9.9 vs. 5.3 ppm; P < 0.05) and H2S (5.8 vs. 4.1 ppm; P < 0.05) were significantly reduced in the treatment group compared to the control group. The gut microbiota of the treatment group differed significantly from that of the control group, and the treatment group altered predicted metabolic pathways, including sulfur and nitrogen related metabolism, urea degradation. The results demonstrated that supplementing feed with arazyme with dietary carbohydrolases effectively controls noxious gas emissions and improves health and meat quality of pigs.

Rapeseed meal processing and dietary enzymes modulate excreta inositol phosphate profile, nutrient availability, and production performance of broiler chickens

This study aimed to assess the effect of rapeseed meal (RSM) processing method, where solvent extraction occurred under standard industry conditions (ST) or cold-pressed hexane extraction was employed (MT), and exogenous enzyme supplementation (phytase [PHY] and xylanase [XYL]) alone or in combination on key nutritional factors of broiler chickens. A randomized control experiment was performed using 144 male Ross 308 broilers in a 2 × 2 × 3 factorial arrangement. Three diets including a nutritionally complete wheat-based basal diet (BD), a diet containing 200 g/kg of RSM extracted under ST and another diet containing 200 g/kg of RSM extracted under MT were produced. Each diet was then split into 4 parts and was fed as is, or supplemented with PHY at 1,500 FTU/kg or XYL at 16,000 BXU/kg, alone or in combination, resulting in 12 diets in total. Response criteria: feed intake (FI), weight gain (WG), and feed conversion ratio (FCR), from 7 to 21 d age, AMEn, retention coefficients for dry matter (DMR), nitrogen (NR), fat (FR), and the profile of inositol phosphate esters (IP2-6) and myo-inositol (MI) in excreta. Diets containing MT had higher AMEn compared to ST diets (P < 0.05). There was RSM by PHY interaction for FI, as only birds fed MT diet responded to PHY supplementation with reduced FI and FCR (P < 0.001). Feeding XYL reduced overall FI and FCR (P < 0.05). Feeding PHY reduced IP6 and increased MI in excreta (P < 0.001). Feeding XYL and PHY in combination reduced MI in excreta compared to PHY only (P = 0.05). Compared to BD, birds fed RSM diets had an increased IP6 (P < 0.05) and MI concentration in excreta (P < 0.01). This may be due to IP ester differences in RSM and BD.

Xylanase Supplementation in Wheat-Based Diets of Laying Hens Affects the Egg Yolk Color, Carotenoid and Fatty Acid Profiles.

Wheat is rich in non-starch polysaccharides (NSP) and their degradation in poultry diets is promoted by exogenous carbohydrases. The objective here was to evaluate the effect of adding an intrinsically thermostable xylanase on wheat-based diets for laying hens in yolk color, carotenoid and fatty acid profiles of eggs. A total of 128 laying hens were used for 12 weeks. They were randomly allocated to four dietary treatments with different levels of xylanase: T1: control (no xylanase), T2: 30,000 U/g, T3: 45,000 U/g and T4: 90,000 U/g, with 32 birds, 16 replicates per treatment (2 birds/replicate). At the end of the experimental period, egg yolk color index, redness (a*) and yellowness (b*) of egg yolks were found significantly higher in all the enzyme supplemented diet groups (T2, T3, T4) compared with the control (T1). Canthaxanthin levels were significantly higher in T3 than T1 (p < 0.05). Total n-3, n-6 and total polyunsaturated fatty acids (FAs) were significantly higher in T4 compared with the control (p < 0.01), while the reverse trend was evidenced for monounsaturated FAs. Additionally, total n-3 FAs were higher in the T2 than T1 (p < 0.005). Overall, the results showed that exogenous xylanase enzyme supplementation in wheat-based diets for laying hens contribute to maintaining egg yolk color. Overall, exogenous xylanase enzyme supplemented at all levels in wheat-based laying hens’ diets improved egg yolk color compared to the control diet. The enzyme supplemented at the higher level (90,000 U/g) improved polyunsaturated and reduced monounsaturated egg yolk fatty acid content.
