The Effectiveness of Transcranial Magnetic Stimulation in Treating Apraxia.

Apraxia can be detected when engaging in mental motor envisioning exercises. The nonverbal skills of manufacturing, representation, strategizing, arithmetic, visual sensitivity, and motor skills are all related to apraxia. Limb apraxia also negatively affects communication gestures and linguistic skills. The impairment of brain regions related to motion patterns is the primary cause of apraxia. People with apraxia may struggle to complete a variety of tasks because they are unable to focus on various movements. Apraxia can result from injury to the premotor cortex since it has a role in the left hemisphere-dependent selection of movements. Cognitive and complicated motor system deficits are hallmarks of the corticobasal syndrome. Apraxia of the limbs and visuospatial abnormalities are typical clinical types. TMS was used to study these problems; however, no research was done on the relationship between TMS parameters and clinical types. It is possible for changes in brain activity to last a long time when repetitive TMS (rTMS) is utilized. Electromyography shows that noninvasive TMS of the motor cortex causes target muscle spasms (MEP). The human motor cortex is a part of the cerebral cortex that is involved in the organization, management, and execution of voluntary movements. TMS and other neuroimaging techniques are frequently used to identify changes in this region. Cortical motor excitability varies among different diagnoses; therefore, it is important to determine the effectiveness of TMS. Therefore, this study aims to review the causes and neurophysiological simulation of apraxia along with the principles and effects of TMS on apraxia.

Galvanic vestibular stimulation modulates EEG markers of voluntary movement in Parkinson’s disease

We recently showed that vestibular stimulation can produce a long-lasting alleviation of motor features in Parkinson’s disease. Here we investigated whether components of the motor related cortical response that are commonly compromised in Parkinson’s – the Bereitschaftspotential and mu-rhythm event-related desynchronization – are modulated by concurrent, low frequency galvanic vestibular stimulation (GVS) during repetitive limb movement amongst 17 individuals with idiopathic Parkinson’s disease. Relative to sham, GVS was favourably associated with higher amplitudes during the late and movement phases of the Bereitschaftspotential and with a more pronounced decrease in spectral power within the mu-rhythm range during finger-tapping. These data increase understanding of how GVS interacts with the preparation and execution of voluntary movement and give added impetus to explore its therapeutic effects on Parkinsonian motor features.

The Simultaneous Application of Transcranial Magnetic Stimulation and Virtual Reality to Treat Cognitive Deficits Among Stroke Patients: A Randomized Controlled Trial.

Background and objective Integrating virtual reality (VR) and transcranial magnetic stimulation (TMS) offers a promising strategy for stroke rehabilitation, as it specifically focuses on reorganizing neural connections and activating brain activity in the cortex. The main goal is to create equitable connections between the brain's two hemispheres to enhance the execution of voluntary movements by stimulating the central executive network (CEN) to strengthen both motor and cognitive abilities. This study aims to propose a therapeutic approach that can improve cognitive recovery and overall quality of life in patients after a stroke. Methods A total of 69 participants were enrolled in the study based on certain inclusion and exclusion criteria. The patients underwent pre-assessment and wererandomly allocated into three groups: Group 1 received simultaneous repetitive TMS (rTMS)and virtual reality treatment (VRT), Group 2 received rTMS combined with sham VRT, and Group 3 received sham stimulation with VRT, in a 1:1:1 ratio using opaque, sealed, and stapled envelopes (SNOSE). Post-assessment was carried out using the same measures: the National Institutes of Health Stroke Scale (NIHSS), Addenbrooke's Cognitive Test (ACE III), and Montreal Cognitive Assessment (MOCA). Statistical analysis was conducted to determine the specific outcomes. Data analysis was carried out using IBM SPSS Statistics version 29 (IBM Corp., Armonk, NY), employing student's t-test for within-group comparisons and repeated measures ANOVA for between-group comparisons. The significance level was set at 5%. Results The results demonstrated statistical significance in NIHSS scores across all treatment groups (p<0.001). Regarding cognitive outcomes, improvements were observed in memory, language, and overall cognitive performance (ACE III) within all groups (p<0.05), with significant between-group outcomes (p = 0.009, p = 0.01, p = 0.004, respectively), suggesting variations in treatment effects across cognitive domains. However, no significant differences between groups were found in terms of fluency and visuospatial skills (p = 0.49, p = 0.13), indicating no treatment effects in these domains. Conclusions Based on our findings, the combined intervention involving rTMS and VRT, compared to sham treatments, demonstrates promising outcomes in alleviating stroke severity and improving specific cognitive functions such as memory, language, and overall cognitive performance. Additionally, the combined administrationoffers a more effective therapy than when they are administered separately.

The Role of Cerebellum and Basal Ganglia Functional Connectivity in Altered Voluntary Movement Execution in Essential Tremor.

Substantial evidence highlights the role of the cerebellum in the pathophysiology of tremor in essential tremor (ET), although its potential involvement in altered movement execution in this condition remains unclear. This study aims to explore potential correlations between the cerebellum and basal ganglia functional connectivity and voluntary movement execution abnormalities in ET, objectively assessed with kinematic techniques. A total of 20 patients diagnosed with ET and 18 healthy subjects were enrolled in this study. Tremor and repetitive finger tapping were recorded using an optoelectronic kinematic system. All participants underwent comprehensive 3T-MRI examinations, including 3D-T1 and blood-oxygen-level dependent (BOLD) sequences during resting state. Morphometric analysis was conducted on the 3D-T1 images, while a seed-based analysis was performed to investigate the resting-state functional connectivity (rsFC) of dorsal and ventral portions of the dentate nucleus and the external and internal segments of the globus pallidus. Finally, potential correlations between rsFC alterations in patients and clinical as well as kinematic scores were assessed. Finger tapping movements were slower in ET than in healthy subjects. Compared to healthy subjects, patients with ET exhibited altered FC of both dentate and globus pallidus with cerebellar, basal ganglia, and cortical areas. Interestingly, both dentate and pallidal FC exhibited positive correlations with movement velocity in patients, differently from that we observed in healthy subjects, indicating the higher the FC, the faster the finger tapping. The findings of this study indicate the possible role of both cerebellum and basal ganglia in the pathophysiology of altered voluntary movement execution in patients with ET.

Cortico-cortical connectivity is influenced by levodopa in tremor-dominant Parkinson's disease

Resting tremor is the most common presenting motor symptom in Parkinson's disease (PD). The supplementary motor area (SMA) is a main target of the basal-ganglia-thalamo-cortical circuit and has direct, facilitatory connections with the primary motor cortex (M1), which is important for the execution of voluntary movement. Dopamine potentially modulates SMA and M1 activity, and both regions have been implicated in resting tremor. This study investigated SMA-M1 connectivity in individuals with PD ON and OFF dopamine medication, and whether SMA-M1 connectivity is implicated in resting tremor. Dual-site transcranial magnetic stimulation was used to measure SMA-M1 connectivity in PD participants ON and OFF levodopa. Resting tremor was measured using electromyography and accelerometry. Stimulating SMA inhibited M1 excitability OFF levodopa, and facilitated M1 excitability ON levodopa. ON medication, SMA-M1 facilitation was significantly associated with smaller tremor than SMA-M1 inhibition. The current findings contribute to our understanding of the neural networks involved in PD which are altered by levodopa medication and provide a neurophysiological basis for the development of interventions to treat resting tremor.

Aspects of motor functioning of blind and visually impaired children - the importance of somatopedic treatment

Introduction. Given that vision significantly influences motor function, particularly in the execution of voluntary movements, this study aimed to determine whether targeted somatopedic training could enhance certain motor aspects in visually impaired children. The study focuses on the following components: coordination, balance, manipulative dexterity, and walking. Material and Methods. The sample comprised 60 children from three schools: the Primary School for Vision Protection ?Dragan Kovacevic? in Belgrade, the School for Visually Impaired Children ?Veljko Ramadanovic? in Zemun, and the Elementary School ?Djordje Krstic? in Belgrade. The experimental group included children aged 6-15 years with visual impairments, average intellectual abilities, and normal neurological and psychological status. The control group was matched by number, gender, and age, consisting of students from regular schools. We assessed motor functioning using tests for hand manipulative skills, coordination of upper and lower extremities, and body balance while walking and standing. Results. Statistically significant differences were observed between the experimental and control groups in all examined subtests: manipulative dexterity (p=0.006); coordination of upper extremities (p=0.029); coordination of upper and lower extremities (p=0.005); maintaining balance during walking (p=0.002); maintaining balance while standing (p=0.024), and walking (p=0.010). Conclusion. The results clearly indicate the importance of somatopedic treatment in improving motor functions of blind and visually impaired children.

Extended source analysis of movement related potentials (MRPs) for self-paced hand and foot movements demonstrates opposing cerebral and cerebellar laterality: a preliminary study

The cerebellum is known to have extensive reciprocal connectivity with the cerebral cortex, including with prefrontal and posterior parietal cortex, which play an important role on the planning and execution of voluntary movement. In the present article we report an exploratory non-invasive electrophysiological study of the activity of the cerebellum and cerebrum during voluntary finger and foot movements. In a sample of five healthy adult subjects, we recorded EEG and the electro-cerebellogram (ECeG) with a 10% cerebellar extension montage during voluntary left and right index finger and foot movements. EMG was recorded from finger extensors and flexors and from the tibialis anterior and soleus muscles and was used to generate triggers for movement related averaging (−2000 to +2000 ms). Source analysis was conducted over five epochs defined relative to EMG onset: whole epoch (−1000 to +1000 ms), pre-move 1000 (−1000 to 0 ms), pre-move 500 (−500 to 0 ms), post-move 500 (0 to +500 ms) and post-move 1000 (0 to +1000 ms). This yielded a total of 123 cerebral and 65 cerebellar dipole clusters from across all epochs, including the pre-movement epochs, which were then subject to statistical analysis. These demonstrated predominantly contralateral dominance for the cerebral clusters, but predominantly ipsilateral dominance for the cerebellar clusters. In addition, both cerebral and cerebellar clusters showed evidence of a somatotopic gradient, medially (X-axis) for the cerebral clusters, and medially and dorso-ventrally (Z-axis) for the cerebellar clusters. These findings support the value of recording cerebellar ECeG and demonstrate its potential to contribute to understanding cerebellar function.

The role of ipsilateral motor network in upper limb movement.

The execution of voluntary movements is primarily governed by the cerebral hemisphere contralateral to the moving limb. Previous research indicates that the ipsilateral motor network, comprising the primary motor cortex (M1), supplementary motor area (SMA), and premotor cortex (PM), plays a crucial role in the planning and execution of limb movements. However, the precise functions of this network and its interplay in different task contexts have yet to be fully understood. Twenty healthy right-handed participants (10 females, mean age 26.1 ± 4.6years) underwent functional MRI scans while performing biceps brachii representations such as bilateral, unilateral flexion, and bilateral flexion-extension. Ipsilateral motor evoked potentials (iMEPs) were obtained from the identical set of participants in a prior study using transcranial magnetic stimulation (TMS) targeting M1 while employing the same motor tasks. The voxel time series was extracted based on the region of interest (M1, SMA, ventral PM and dorsal PM). Directed functinal connectivity was derived from the extracted time series using time-resolved partial directed coherence. We found increased connectivity from left-PMv to both sides M1, as well as right-PMv to both sides SMA, in unilateral flexion compared to bilateral flexion. Connectivity from left M1 to left-PMv, and left-SMA to right-PMd, also increased in both unilateral flexion and bilateral flexion-extension compared to bilateral flexion. However, connectivity between PMv and right-M1 to left-PMd decreased during bilateral flexion-extension compared to unilateral flexion. Additionally, during bilateral flexion-extension, the connectivity from right-M1 to right-SMA had a negative relationship with the area ratio of iMEP in the dominant side. Our results provide corroborating evidence for prior research suggesting that the ipsilateral motor network is implicated in the voluntary movements and underscores its involvement in cognitive processes such as movement planning and coordination. Moreover, ipsilateral connectivity from M1 to SMA on the dominant side can modulate the degree of ipsilateral M1 activation during bilateral antagonistic contraction.

Fast-Spiking Interneurons of the Premotor Cortex Contribute to Initiation and Execution of Spontaneous Actions.

Planning and execution of voluntary movement depend on the contribution of distinct classes of neurons in primary motor and premotor areas. However, timing and pattern of activation of GABAergic cells during specific motor behaviors remain only partly understood. Here, we directly compared the response properties of putative pyramidal neurons (PNs) and GABAergic fast-spiking neurons (FSNs) during spontaneous licking and forelimb movements in male mice. Recordings centered on the face/mouth motor field of the anterolateral motor cortex (ALM) revealed that FSNs fire longer than PNs and earlier for licking, but not for forelimb movements. Computational analysis revealed that FSNs carry vastly more information than PNs about the onset of movement. While PNs differently modulate their discharge during distinct motor acts, most FSNs respond with a stereotyped increase in firing rate. Accordingly, the informational redundancy was greater among FSNs than PNs. Finally, optogenetic silencing of a subset of FSNs reduced spontaneous licking movement. These data suggest that a global rise of inhibition contributes to the initiation and execution of spontaneous motor actions.SIGNIFICANCE STATEMENT Our study contributes to clarifying the causal role of fast-spiking neurons (FSNs) in driving initiation and execution of specific, spontaneous movements. Within the face/mouth motor field of mice premotor cortex, FSNs fire before pyramidal neurons (PNs) with a specific activation pattern: they reach their peak of activity earlier than PNs during the initiation of licking, but not of forelimb, movements; duration of FSNs activity is also greater and exhibits less selectivity for the movement type, as compared with that of PNs. Accordingly, FSNs appear to carry more redundant information than PNs. Optogenetic silencing of FSNs reduced spontaneous licking movement, suggesting that FSNs contribute to the initiation and execution of specific spontaneous movements, possibly by sculpting response selectivity of nearby PNs.

Subtle changes in central dopaminergic tone underlie bradykinesia in essential tremor

IntroductionIn this research, our primary objective was to explore the correlation between basal ganglia dopaminergic neurotransmission, assessed using 123I-FP-CIT (DAT-SPECT), and finger movements abnormalities in patients with essential tremor (ET) and Parkinson’s disease (PD). MethodsWe enrolled 16 patients with ET, 17 with PD, and 18 healthy controls (HC). Each participant underwent comprehensive clinical evaluations, kinematic assessments of finger tapping. ET and PD patients underwent DAT-SPECT imaging. The DAT-SPECT scans were subjected to both visual and semi-quantitative analysis using DaTQUANT®. We then investigated the correlations between the clinical, kinematic, and DAT-SPECT data, in patients. ResultsOur findings confirm that individuals with ET exhibited slower finger tapping than HC. Visual evaluation of radiotracer uptake in both striata demonstrated normal levels within the ET patient cohort, while PD patients displayed reduced uptake. However, there was notable heterogeneity in the quantification of uptake within the striata among ET patients. Additionally, we found a correlation between the amount of radiotracer uptake in the striatum and movement velocity during finger tapping in patients. Specifically, lower radioligand uptake corresponded to decreased movement velocity (ET: coef. = 0.53, p-adj = 0.03; PD: coef. = 0.59, p-adj = 0.01). ConclusionThe study's findings suggest a potential link between subtle changes in central dopaminergic tone and altered voluntary movement execution, in ET. These results provide further insights into the pathophysiology of ET. However, longitudinal studies are essential to determine whether the slight reduction in dopaminergic tone observed in ET patients represents a distinct subtype of the disease or could serve as a predictor for the clinical progression into PD.

Longitudinal study of clinical and neurophysiological features in essential tremor.

Essential tremor (ET) is a common and heterogeneous disorder characterized by postural/kinetic tremor of the upper limbs and other body segments and by non-motor symptoms, including cognitive and psychiatric abnormalities. Only a limited number of longitudinal studies have comprehensively and simultaneously investigated motor and non-motor symptom progression in ET. Possible soft signs that configure the ET-plus diagnosis are also under-investigated in follow-up studies. We aimed to longitudinally investigate the progression of ET manifestations by means of clinical and neurophysiological evaluation. Thirty-seven ET patients underwent evaluation at baseline (T0) and at follow-up (T1; mean interval ± SD =39.89 ± 9.83 months). The assessment included the clinical and kinematic evaluation of tremor and voluntary movement execution, as well as the investigation of cognitive and psychiatric disorders. A higher percentage of patients showed tremor in multiple body segments and rest tremor at T1 as compared to T0 (all p-values < 0.01). At T1, the kinematic analysis revealed reduced finger-tapping movement amplitude and velocity as compared to T0 (both p-values < 0.001). The prevalence of cognitive and psychiatric disorders did not change between T0 and T1. Female sex, absence of family history, and rest tremor at baseline were identified as predictive factors of worse disease progression. ET progression is characterized by the spread of tremor in multiple body segments and by the emergence of soft signs. We also identified possible predictors of disease worsening. The results contribute to a better understanding of ET classification and pathophysiology.

Quantitative analysis of the Golgi impregnated human (neo)striatal neurons: Observation of the morphological characteristics followed by an emphasis on the functional diversity of cells

BackgroundThe (neo)striatum is the major input structure of the basal nuclei, which is involved in the execution of voluntary movements, but also in controlling the processes that lead to the movement, such as motivation and cognition. The striatum provides its function through an interaction between projection neurons and interneurons. The aim of this study was to quantify the morphological properties of neurons in the precommissural putamen and precommissural caudate nucleus head and to evaluate whether there is a difference in cell morphology between different cell groups within one part and between the same cell groups within different parts of the striatum. MethodsA total of 652 neuronal images of human striatum were observed. The features of the neuronal morphology (soma size, dendritic field size, shape of neuronal image, dendritic curviness, dendritic branching complexity) were observed by determining appropriate parameters of digital images of neurons. ResultsAccording to the presence of spines on the soma and/or dendrites, neurons were qualitatively classified into 446 spiny and 206 aspiny cells. The analysis of the distribution of the dendritic field area shows that spiny and aspiny neurons from both parts of the neostriatum can be decomposed into two distributions, which means that they can be classified into subgroups. A quantitative analysis of the spiny/aspiny neurons in the human putamen or caudate nucleus head has shown that there is a statistically significant difference between them. By comparing the morphology of neurons of the same group between different parts of the human neostriatum (putamen and caudate nucleus), it was also determined that there is a statistically significant difference. ConclusionSince the morphology and function of neurons are in close correlation, it can be assumed that different groups of neurons in the human striatum might support functional diversity of the studied area.

Characterization of Parkinson's Disease using spectral features of kinetic tremor: correlation of on-line digitized handwriting and classical motor scales

Tremor is one of the motor impairments of Parkinson’s disease (PD). To date, kinetic tremor in PD is barely examined and there is lack of information on the relation between its severity and scales related to specific motor deficits [1]. In this study, we aimed at investigating the correlation between handwriting-related kinetic tremor and motor score measures by motor part of Unified Parkinson's Disease Rating Scale (UPDRS-III) [2], using a digitizing tablet. In this preliminary study, eight PD patients (7 M/1 F; age 74.5±8 years) draw an accurate Archimedes’ Spiral (AS) and fast, overlapped Circles (C) for a duration of 15 seconds. All patients underwent motor deficit assessment using UPDRS-III. Power Spectral Density of both velocity and acceleration profiles in their horizontal, vertical, and curvilinear components was estimated by using Welch’s method, with a Hamming window on intervals of 5 s and a 50% overlap. To analyze the power distribution related to different movement-associated phenomena, the ratio between two frequency bands (BME/BT) and the BT bandwidth (BW) were calculated for each subject. BME is the band of voluntary Movement Execution required by the task, ranging from 0.2 to 4 Hz, and BT is the band associated with involuntary Tremor, ranging from 4.0 to 12 Hz [3]. Normalized Jerk, a classic kinematic feature representing handwriting fluidity, was also estimated. The correlation between the parameters and UPDRS-III scores were assessed using Spearman’s rank correlation coefficients. All the evaluations were conducted in the pharmacological on state of PD patients. A positive correlation was found between Jerk and UPDRS-III scores (Table 1). On the contrary BME /B T correlates negatively with motor scale scores in horizontal and vertical velocities (Vx, Vy) for both tasks, vertical and curvilinear acceleration (Ay, Ac) for AS task, horizontal and vertical acceleration (Ax, Ay) for C task. Only Ay of C task shows correlation with motor scores. The results highlight that the severity of motor deficits in PD patients, as assessed by a widely employed motor scale, correlates with the outcomes of spectral and kinematic analysis of handwriting that indicate a loss of fluency, an increased power at BT level and a thinning of the spectral peak of B T . This suggests that handwriting assessment of parkinsonian dysgraphia can be used to implement clinical evaluation and represents a non-invasive, low-cost method for the identification of objective and reproducible biomarkers of kinetic tremor.

Altered Dynamic Information Flow through the Cortico-Basal Ganglia Pathways Mediates Parkinson's Disease Symptoms.

Parkinson’s disease (PD) is a progressive neurodegenerative disorder caused by dopamine deficiency. To elucidate network-level changes through the cortico-basal ganglia pathways in PD, we recorded neuronal activity in PD monkeys treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine. We applied electrical stimulation to the motor cortices and examined responses in the internal (GPi) and external (GPe) segments of the globus pallidus, the output and relay nuclei of the basal ganglia, respectively. In the normal state, cortical stimulation induced a triphasic response composed of early excitation, inhibition, and late excitation in the GPi and GPe. In the PD state, cortically evoked inhibition in the GPi mediated by the cortico-striato-GPi “direct” pathway was largely diminished, whereas late excitation in the GPe mediated by the cortico-striato-GPe-subthalamo (STN)-GPe pathway was elongated. l-DOPA treatment ameliorated PD signs, particularly akinesia/bradykinesia, and normalized cortically evoked responses in both the GPi and GPe. STN blockade by muscimol injection ameliorated the motor deficit and unmasked cortically evoked inhibition in the GPi. These results suggest that information flow through the direct pathway responsible for the initiation of movements is largely reduced in PD and fails to release movements, resulting in akinesia/bradykinesia. Restoration of the information flow through the direct pathway recovers execution of voluntary movements.

Examining the neural antecedents of tics in Tourette syndrome using electroencephalography.

Tourette syndrome (TS) is a neurodevelopmental disorder characterized by the occurrence of motor and vocal tics. TS is associated with cortical-striatal-thalamic-cortical circuit dysfunction and hyper-excitability of cortical limbic and motor regions that lead to the occurrence of tics. Importantly, individuals with TS often report that their tics are preceded by premonitory sensory/urge phenomena (PU) that are described as uncomfortable bodily sensations that precede the execution of a tic and are experienced as an urge for motor discharge. While tics are most often referred to as involuntary movements, it has been argued by some that tics should be viewed as voluntary movements that are executed in response to the presence of PU. To investigate this issue further, we conducted a study using electroencephalography (EEG). We recorded movement-related EEG (mu- and beta-band oscillations) during (1) the immediate period leading up to the execution of voluntary movements by a group of individuals with TS and a group of matched healthy control participants, and (2) the immediate period leading up to the execution of a tic in a group of individuals with TS. We demonstrate that movement-related mu and beta oscillations are not reliably observed prior to tics in individuals with TS. We interpret this effect as reflecting the greater involvement of a network of brain areas, including the insular and cingulate cortices, the basal ganglia and the cerebellum, in the generation of tics in TS. We also show that beta-band desynchronization does occur when individuals with TS initiate voluntary movements, but, in contrast to healthy controls, desynchronization of mu-band oscillations is not observed during the execution of voluntary movements for individuals with TS. We interpret this finding as reflecting a dysfunction of physiological inhibition in TS, thereby contributing to an impaired ability to suppress neuronal populations that may compete with movement preparation processes.

An evaluation of sustained attention with frontal electroencephalogram

Background: The activity in various regions of the brain can be assessed through changes in spectral and temporal aspects of EEG, in reference to particular and diverse physiological states. The band powers calculated through frequency domain decline through the baseline and execution of voluntary movements. The deterioration in perceptual efficiency resulting from prolonged focused attention and conditions. The present study investigated the feasibility of using Clock task as a means to evaluate the sustained attention through frontal EEG. Method: The Clock task, a paradigm for the assessment of sustained attention was designed. The participants have to monitor and react when the hand of the clock stops slightly longer than usual. The sample comprised of 6 participants who complete Clock task. Frontal EEG signals were recorded. Results: The left inferior frontal gyrus are associated with vigilance in beta frequency bands when attention is directed towards task. The performance in clock task, showed high brain activity when the subject was engaged in maintaining vigilance. The exploration of beta activity strengthens the Sensory feedback through visual modality during static motor control. Conclusion: The ventral pathway of the visual information processing in the described cognitive task may play a significant role to regulate attentional processes. The study concluded the left prefrontal and temporal regions plays a significant role in the modulation of sustained attention. The area can be potentially used to monitor changes both at physiological and psychological level during alertness.

Pre-movement changes in sensorimotor beta oscillations predict motor adaptation drive

Beta frequency oscillations in scalp electroencephalography (EEG) recordings over the primary motor cortex have been associated with the preparation and execution of voluntary movements. Here, we test whether changes in beta frequency are related to the preparation of adapted movements in human, and whether such effects generalise to other species (cat). Eleven healthy adult humans performed a joystick visuomotor adaptation task. Beta (15–25 Hz) scalp EEG signals recorded over the motor cortex during a pre-movement preparatory phase were, on average, significantly reduced in amplitude during early adaptation trials compared to baseline, late adaptation, or aftereffect trials. The changes in beta were not related to measurements of reaction time or reach duration. We also recorded local field potential (LFP) activity within the primary motor cortex of three cats during a prism visuomotor adaptation task. Analysis of these signals revealed similar reductions in motor cortical LFP beta frequencies during early adaptation. This effect was present when controlling for any influence of the reaction time and reach duration. Overall, the results are consistent with a reduction in pre-movement beta oscillations predicting an increase in adaptive drive in upcoming task performance when motor errors are largest in magnitude and the rate of adaptation is greatest.

Beta Rebound as an Index of Temporal Integration of Somatosensory and Motor Signals.

Modulation of cortical beta rhythm (15–30 Hz) is present during preparation for and execution of voluntary movements as well as during somatosensory stimulation. A rebound in beta synchronization is observed after the end of voluntary movements as well as after somatosensory stimulation and is believed to describe the return to baseline of sensorimotor networks. However, the contribution of efferent and afferent signals to the beta rebound remains poorly understood. Here, we applied electrical median nerve stimulation (MNS) to the right side followed by transcranial magnetic stimulation (TMS) on the left primary motor cortex after either 15 or 25 ms. Because the afferent volley reaches the somatosensory cortex after about 20 ms, TMS on the motor cortex was either anticipating or following the cortical arrival of the peripheral stimulus. We show modulations in different beta sub-bands and in both hemispheres, following a pattern of greater resynchronization when motor signals are paired with a peripheral one. The beta rebound in the left hemisphere (stimulated) is modulated in its lower frequency range when TMS precedes the cortical arrival of the afferent volley. In the right hemisphere (unstimulated), instead, the increase is limited to higher beta frequencies when TMS is delivered after the arrival of the afferent signal. In general, we demonstrate that the temporal integration of afferent and efferent signals plays a key role in the genesis of the beta rebound and that these signals may be carried in parallel by different beta sub-bands.
