Modeling excitability in cerebellar stellate cells: Temporal changes in threshold, latency and frequency of firing

Cerebellar stellate cells are inhibitory molecular interneurons that regulate the firing properties of Purkinje cells, the sole output of cerebellar cortex. Recent evidence suggests that these cells exhibit temporal increase in excitability during whole-cell patch-clamp configuration in a phenomenon termed runup. They also exhibit a non-monotonic first-spike latency profile as a function of the holding potential in response to a fixed step-current. In this study, we use modeling approaches to unravel the dynamics of runup and categorize the firing behavior of cerebellar stellate cells as either type I or type II oscillators. We then extend this analysis to investigate how the non-monotonic latency profile manifests itself during runup. We employ a previously developed, but revised, Hodgkin–Huxley type model to show that stellate cells are indeed type I oscillators possessing a saddle node on an invariant cycle (SNIC) bifurcation. The SNIC in the model acts as a “threshold” for tonic firing and produces a slow region in the phase space called the ghost of the SNIC. The model reveals that (i) the SNIC gets left-shifted during runup with respect to Iapp=Itest in the current-step protocol, and (ii) both the distance from the stable limit cycle along with the slow region produce the non-monotonic latency profile as a function of holding potential. Using the model, we elucidate how latency can be made arbitrarily large for a specific range of holding potentials close to the SNIC during pre-runup (post-runup). We also demonstrate that the model can produce transient single spikes in response to step-currents entirely below ISNIC, and that a pair of dynamic inhibitory and excitatory post-synaptic inputs can robustly evoke action potentials, provided that the magnitude of the inhibition is either low or high but not intermediate. Our results show that the topology of the SNIC is the key to explaining such behaviors.

Functional Reorganization of Local Circuit Connectivity in Superficial Spinal Dorsal Horn with Neuropathic Pain States.

The spinal dorsal horn is the first relay structure coding for pain transmission and modulation. Previous anatomical and electrophysiological studies have examined spinal dorsal horn circuit connections and network activity. Further work is required to understand spinal cord sensory information processing that underlies pathological neuropathic pain states. Our previous studies suggest that peripheral nerve injury enhances presynaptic excitatory input onto spinal superficial dorsal horn neurons, which in turn contributes to pathologic nociception. The potential changes in local postsynaptic circuits in the dorsal horn that lead to pathologically heightened behavioral responses to pain remain largely unexplored. We combined whole-cell electrophysiological recordings with laser-scanning photostimulation to test whether peripheral nerve injury in the spinal nerve ligation (SNL) mouse model of neuropathic pain leads to alterations in the functional connectivity of spinal cord circuits including lamina II excitatory interneurons. Here we show that SNL enhances excitation and decreases inhibition to lamina II excitatory interneurons along with their increased glutamate-evoked excitability. The enhanced excitatory postsynaptic input and connectivity evoked by SNL eventually return to normal levels concurrently with the resolution of the neuropathic pain states. The physiological pattern highly correlates with mouse pain behaviors following SNL, supporting a neurophysiological mechanism of central sensitization and neuropathic pain that is functionally localized to the spinal dorsal horn. Together, these data support that SNL induces functional changes in synaptic input and connectivity to lamina II excitatory interneurons that code for pain perception, and thus provide new insights into the mechanism and locus of pain hypersensitivity.

Defensive Behaviors Driven by a Hypothalamic-Ventral Midbrain Circuit.

The paraventricular hypothalamus (PVH) regulates stress, feeding behaviors and other homeostatic processes, but whether PVH also drives defensive states remains unknown. Here we showed that photostimulation of PVH neurons in mice elicited escape jumping, a typical defensive behavior. We mapped PVH outputs that densely terminate in the ventral midbrain (vMB) area, and found that activation of the PVH→vMB circuit produced profound defensive behavioral changes, including escape jumping, hiding, hyperlocomotion, and learned aversion. Electrophysiological recordings showed excitatory postsynaptic input onto vMB neurons via PVH fiber activation, and in vivo studies demonstrated that glutamate transmission from PVH→vMB was required for the evoked behavioral responses. Photostimulation of PVH→vMB fibers induced cFos expression mainly in non-dopaminergic neurons. Using a dual optogenetic-chemogenetic strategy, we further revealed that escape jumping and hiding were partially contributed by the activation of midbrain glutamatergic neurons. Taken together, our work unveils a hypothalamic-vMB circuit that encodes defensive properties, which may be implicated in stress-induced defensive responses.

Modeling Neural Adaptation in Auditory Cortex

Neural responses recorded from auditory cortex exhibit adaptation, a stimulus-specific decrease that occurs when the same sound is presented repeatedly. Stimulus-specific adaptation is thought to facilitate perception in noisy environments. Although adaptation is assumed to arise independently from cortex, this has been difficult to validate directly in vivo. In this study, we used a neural network model of auditory cortex with multicompartmental cell modeling to investigate cortical adaptation. We found that repetitive, non-adapted inputs to layer IV neurons in the model elicited frequency-specific decreases in simulated single neuron, population-level and local field potential (LFP) activity, consistent with stimulus-specific cortical adaptation. Simulated recordings of LFPs, generated solely by excitatory post-synaptic inputs and recorded from layers II/III in the model, showed similar waveform morphologies and stimulus probability effects as auditory evoked responses recorded from human cortex. We tested two proposed mechanisms of cortical adaptation, neural fatigue and neural sharpening, by varying the strength and type of inter- and intra-layer synaptic connections (excitatory, inhibitory). Model simulations showed that synaptic depression modeled in excitatory (AMPA) synapses was sufficient to elicit a reduction in neural firing rate, consistent with neural fatigue. However, introduction of lateral inhibition from local layer II/III interneurons resulted in a reduction in the number of responding neurons, but not their firing rates, consistent with neural sharpening. These modeling results demonstrate that adaptation can arise from multiple neural mechanisms in auditory cortex.

Hypothalamic Nuclear Glucocorticoid Receptors: Acute Stress and Rapid Actions.

Stress, be it physical or psychological, causes the activation of a neuroendocrine cascade that involves both central and peripheral mechanisms. In the simplest terms, central release of CRH from the paraventricular nucleus of thehypothalamus (PVN)andsubsequent releaseofACTH from the pituitary result in the peripheral release of glucocorticoids (cortisol in primates and corticosterone in many rodent species) from the adrenal gland (1). Glucocorticoids then act via negative feedback both centrally and peripherally through the glucocorticoid receptor (GR) to inhibit the further release of CRH from the PVN and ACTH from the pituitary (2). This process is a delicate balance that helps restore homeostasis, but perturbation of these responses can lead to neurobehavioral and physiological dysfunctions, including mood disorders and obesity (3–5). Hence, understanding the basic mechanisms that mediate this neuroendocrine loop has broad implications for both health and disease. Two companion papers in this issue by Solomon et al (6) and Nahar et al (7) have helped to further reveal the mechanisms through which glucocorticoids influence hypothalamic-pituitary-adrenal (HPA) function. Both reports use a powerful mouse model in which Sim1-cre recombinase transgenic mice are bred with homozygous floxed GR mice. Sim1 is a transcription factor specifically expressed in the PVN, supraoptic nucleus, and periventricular nucleus (8) and is responsible for the proper maturation of neurosecretory cells in the PVN (9). As Solomon et al report, theoffspring fromthis cross showsignificantly reduced GR levels in the PVN while leaving GR levels intact in the pituitary and other brain regions important in glucocorticoid-dependent negative feedback (6), thus resulting in a relatively anatomically restricted GR reduction. Using these PVN GR knockdown (KD) mice, Solomon et al (6) find sex-specific and stressor-dependent effects on HPA reactivity. In particular, they show GR KD mice respond to a brief 30-minute session of restraint stress with significantly increased and prolonged ACTH and corticosterone responses compared with their littermate controls, suggesting reduced negative feedback on the HPA axis (6). Notably, however, these effects of GR KD were observed only in males because female GR KD mice show reduced restraint-induced ACTH and no differences in stress-induced corticosterone responsiveness. In response to chronic variable stress, neither male nor female GR KD mice show any differences in HPA reactivity. Hence, the authors conclude that there are significant sex differences in the role that PVN GR plays in response to acute stress and that changes in HPA reactivity after chronic stress in male and female mice are independent of GR in the PVN (6). Complimenting this line of research, the report by Nahar et al (7) investigated rapid nongenomic actions of glucocorticoids in modulating excitatory and inhibitory transmission in the mouse hypothalamus. They show that hypothalamic magnocellular neurons respond to dexamethasone (DEX), a synthetic glucocorticoid, with a rapid suppression of excitatory postsynaptic inputs (ie, glutamatergic synapses). The authors further demonstrate that this effect of DEX is mediated by postsynaptic membrane associated receptors and G protein signaling (7), similar to their seminal work in rats (10–12). Rapid Nongenomic Glucocorticoid Actions in Male Mouse Hypothalamic Neuroendocrine Cells Are Dependent on the Nuclear Glucocorticoid Receptor.

Corticosteroids act classically via cognate nuclear receptors to regulate gene transcription; however, increasing evidence supports rapid, nontranscriptional corticosteroid actions via activation of membrane receptors. Using whole-cell patch clamp recordings in hypothalamic slices from male mouse genetic models, we tested for nongenomic glucocorticoid actions at glutamate and gamma aminobutyric acid (GABA) synapses in hypothalamic neuroendocrine cells, and for their dependence on the nuclear glucocorticoid receptor (GR). In enhanced green fluorescent protein-expressing CRH neurons of the paraventricular nucleus (PVN) and in magnocellular neurons of the PVN and supraoptic nucleus (SON), dexamethasone activated postsynaptic membrane-associated receptors and G protein signaling to elicit a rapid suppression of excitatory postsynaptic inputs, which was blocked by genetic deletion of type I cannabinoid receptors and a type I cannabinoid receptor antagonist. In magnocellular neurons, dexamethasone also elicited a rapid nitric oxide-dependent increase in inhibitory postsynaptic inputs. These data indicate a rapid, synapse-specific glucocorticoid-induced retrograde endocannabinoid signaling at glutamate synapses and nitric oxide signaling at GABA synapses. Unexpectedly, the rapid glucocorticoid effects on both excitatory and inhibitory synaptic transmission were lost with conditional deletion of GR in the PVN and SON in slices from a single minded-1-cre-directed conditional GR knockout mouse. Thus, the nongenomic glucocorticoid actions at glutamate and GABA synapses on PVN and SON neuroendocrine cells are dependent on the nuclear GR. The nuclear GR, therefore, is responsible for transducing the rapid steroid response at the membrane, or is either a critical component in the signaling cascade or regulates a critical component of the signaling cascade of a distinct membrane GR.

Dysfunction of Cortical Dendritic Integration in Neuropathic Pain Reversed by Serotoninergic Neuromodulation

Neuropathic pain is caused by long-term modifications of neuronal function in the peripheral nervous system, the spinal cord, and supraspinal areas. Although functional changes in the forebrain are thought to contribute to the development of persistent pain, their significance and precise subcellular nature remain unexplored. Using somatic and dendritic whole-cell patch-clamp recordings from neurons in the anterior cingulate cortex, we discovered that sciatic nerve injury caused an activity-dependent dysfunction of hyperpolarization-activated cyclic nucleotide-regulated (HCN) channels in the dendrites of layer 5 pyramidal neurons resulting in enhanced integration of excitatory postsynaptic inputs and increased neuronal firing. Specific activation of the serotonin receptor type 7 (5-HT7R) alleviated the lesion-induced pathology by increasing HCN channel function, restoring normal dendritic integration, and reducing mechanical pain hypersensitivity in nerve-injured animals in vivo. Thus, serotoninergic neuromodulation at the forebrain level can reverse the dendritic dysfunction induced by neuropathic pain and may represent a potential therapeutical target.

Effect of glycine on synaptic transmission at the third order giant synapse of the squids Alloteuthis subulata and Loligo vulgaris

Intracellular microelectrode recordings were made from presynaptic and postsynaptic regions of the third order giant synapses of the squids Alloteuthis subulata and Loligo vulgaris. Synaptically generated postsynaptic action potential trains, and excitatory postsynaptic potentials (EPSPs) were reversibly decreased by glycine, β-alanine or taurine while presynaptic action potentials (APs) were unaltered. Glycine was effective in the presence of strychnine (30–50 μM), NMDA (500 μM), AP-5 (50 μM), CPP (100 μM), or MK 801 (which also had no effect on normal synaptic transmission). The glycine effect was reduced reversibly by d-tubocurarine (100 μM) and blocked by reducing extracellular chloride by 50% with propionate. Excitatory postsynaptic currents (EPSCs) were decreased by glycine addition without altering resting membrane conductance. We postulate that glycine or a glycine like substance provides an excitatory postsynaptic input during synaptic stimulation. Bath addition of glycine desensitises these receptors and decreases the amplitude of the EPSPs and EPSCs. Modulation of this synaptic input may provide an effective mechanism to suppress or potentiate synaptic transmission in the squid giant synapse.

Neural spike generator entrainment by regularly spaced synaptic input

It has been known for 30 years that the output of a repetitively firing neuron or pacemaker can be synchronized (locked) to regularly spaced inhibitory or excitatory postsynaptic input potentials. Conditions for stable locking have been determined mathematically, demonstrated in computer simulation, and locking has been observed in vivo. We have developed a neural spike generator circuit model which exhibits stable locking to externally derived simulated inhibitory or excitatory post-synaptic inputs. Conditions for stable 1 : 1 lock, in which pacemaker output frequency matches that of the periodic input, are derived. These take the form of expressions for stable delay and convergence factor which incorporate known or measurable parameters of the circuit model. The expressions have been evaluated and shown to compare satisfactorily with experimental observations of locking by our circuit model.

Action of caffeine on pyloric motorneurons in the crustacean stomatogastric ganglion

1. 1. Effects of caffeine on pyloric motorneurons in the stomatogastric ganglion of the crab, Cancer pagurus and of the crayfish, Astacus leptodactylus were investigated. 2. 2. Caffeine increases the pyloric cycle rate or initiates cyclic pyloric discharge activity in non-rhythmically active preparations. 3. 3. The action of caffeine on bursting pacemaker neurons (PD (pyloric dilator)-neurons) is time and dose dependent. Caffeine depolarizes the membrane potential and induces or potentiates burst discharge. The burst frequency, the amplitude of the pacemaker potential and the duration of the action potentials is increased. These effects are reversible after removal of caffeine. 4. 4. Caffeine decreases the threshold for initiation of bursting activity elicited by excitatory postsynaptic input (EPSPs) and also decreases the threshold for initiation of action potentials by depolarizing current injection. 5. 5. After blocking the activity of the Na-pump by ouabain, caffeine has an additional effect on the membrane permeability and increases the permeability ratio p Na/p K. 6. 6. Bursting discharge activity or slow membrane oscillations are maintained as long as caffeine is in the bath solution. In Ca-free solution containing caffeine the membrane potential readily depolarizes to a steady level. 7. 7. Intracellular injection of caffeine elicits spontaneous hyperpolarizations of the membrane potential with a reversal potential close to the K-equilibrium potential. This suggests the activation of a K-conductance by internal caffeine—possibly by liberation of internal calcium. 8. 8. The results suggests that caffeine either increases the Na-resting condunctance and/or decreases the K-resting conductance and the voltage activated K-conductance. The Ca-activated K-conductance appears to be increased by the action of caffeine and further supports the notion that this current component is a major ingredient for pacemaker activity in these cells. It is concluded that the alternations of membrane permeability could provide a basis for understanding the excitatory action of caffeine at pyloric motorneurons. Caffeine may be a useful tool for initiation of fixed neural programs.
