Zwitterionic Block Copolymers for the Synthesis and Stabilization of Perovskite Nanocrystals.

Traditional hot injection methods for the preparation of cesium lead halide perovskite nanocrystals (CsPbX3 PNCs, where X=Cl, Br, or I) rely on small molecule surfactants to produce PNCs with cube, plate, or rod-like morphologies. Here, we describe a new method whereby zwitterionic block copolymers are employed as macromolecular ligands in PNC synthesis, affording PNCs with excellent colloidal stability, high photoluminescence quantum yield, and in some cases distinctly non-cubic shapes. The block copolymers used in this study - composed of a poly(n-butyl methacrylate) hydrophobic block and zwitterionic methacrylate hydrophilic blocks - dissolve in useful solvents for PNC growth despite containing large mole percentages of zwitterionic groups. PNCs prepared with block copolymer ligands were found to disperse and retain their fluorescence in a range of polar organic solvents and were amenable to direct integration into optically transparent nanocomposite thin films with high PNC content.

Novel Strategy for the Synthesis of Polymer/Pigment Hybrid Latex via Sulfur-Free RAFT-Mediated Emulsion Polymerization

Reversible addition–fragmentation chain transfer (RAFT) emulsion polymerization is a powerful tool for polymer encapsulation of pigment nanohybrids. Conventional RAFT emulsion polymers, however, contain sulfur residues that contribute to undesired odor and deleterious effects on final materials. Our strategy relies on an amphiphilic macromonomer poly(methacrylic acid-co-methyl methacrylate) P(MAA-co-MMA) with an ω-unsaturated end group synthesized in situ via cobalt(II)-mediated catalytic chain transfer polymerization (CCTP) at the surface of C.I. Pigment Blue 15:3 (PB) particles. Subsequently, these macromonomers are used as the living points to mediate the in situ sulfur-free RAFT (SF-RAFT) emulsion polymerization of monomers (butyl methacrylate (BMA) and butyl acrylate (BA)). It was found that a well-controlled polymerization process was achieved by semibatch SF-RAFT emulsion polymerization on the PB surface, as evidenced by the smooth increase in the molecular weight of polymer chains as the polymerization progressed and by transmission electron microscopy (TEM) results. Due to the sealing effect, these polymer/PB hybrid particles exhibited excellent colloidal stability in the aqueous phase. More importantly, film-forming hybrid particles with a soft P(BMA-co-BA) shell were successfully prepared in this work, which suggested that SF-RAFT-mediated polymerization may offer a useful alternative approach to traditional RAFT emulsion polymers for the preparation of organic/inorganic nanohybrids.

Facile synthesis of water-dispersible poly(3-hexylthiophene) nanoparticles with high yield and excellent colloidal stability

SummaryThere has been growing interest in water-processable conjugated polymers for biocompatible devices. However, some broadly used conjugated polymers like poly(3-hexylthiophene) (P3HT) are hydrophobic and they cannot be processed in water. We herein report a facile yet highly efficient assembly method to prepare water-dispersible pyridine-containing P3HT (Py-P3HT) nanoparticles (NPs) with a high yield (>80%) and a fine size below 100 nm. It is based on the fast nanoprecipitation of Py-P3HT stabilized by hydrophilic poly(acrylic acid) (PAA). Py-P3HT can form spherical NPs at a concentration up to 0.2 mg/mL with a diameter of ∼75 nm at a very low concentration of PAA, e.g., 0.01–0.1 mg/mL, as surface ligands. Those negatively charged Py-P3HT NPs can bind with metal cations and further support the growth of noble metal NPs like Ag and Au. Our self-assembly methodology potentially opens new doors to process and directly use hydrophobic conjugated polymers in a much broader context.

Entrapping curcumin in the hydrophobic reservoir of rice proteins toward stable antioxidant nanoparticles

Loading hydrophobic phytochemicals by protein particles is challenged by either insufficient loading capacity or colloidal instability. In this study, hydrosoluble curcumin-loaded nanoparticles were prepared using hydrophobic rice proteins (RPs). The fabrication was facilitated by dissolving curcumin and RPs at pH 12.0 before neutralizing the basicity. The formation of nanoparticles was studied by fluorescence and infrared spectroscopy, and the microstructures were studied by transmission electron microscopy. The results showed the entrapping of curcumin inside the hydrophobic reservoir formed by the acid-induced refolding of RPs. Due to promoted burial of the hydrophobic moieties, the curcumin-loaded RP-nanoparticles displayed excellent colloidal stability during the 28-day storage. Bearing 144.41 mg/g protein of curcumin, the nanoparticles demonstrated prominent antioxidant properties, with the DPPH scavenging capacity increased by up to 88.62% compared to the free curcumin. This study harnessed hydrophobic attractions as a tool for management of the protein-phytochemical interactions, overcoming colloidal instabilities of both individual components.

Rice bran-modified wheat gluten nanoparticles effectively stabilized pickering emulsion: An interfacial antioxidant inhibiting lipid oxidation

This study investigated the fabrication of Pickering emulsion stabilized by rice bran-modified wheat gluten nanoparticle (RGNP) and evaluated the effect of rice bran-modification on lipid oxidation of emulsified soybean oil, with native wheat gluten nanoparticle (CGNP) as the control. Compared with CGNP, RGNP exhibited stronger antioxidant activities due to high content of polyphenols. RGNP were roughly spherical (mean size of about 250 nm) with balanced wettability (104.6 ± 2.3°) and had excellent colloidal thermal stability. When used at a concentration of 1.5 wt%, both CGNP and RGNP could stabilize Pickering emulsions at varying oil volume fractions (0.20–0.85). For high internal phase emulsions, nanoparticles adsorbed at an oil–water interface forming an interconnected network structure, depressing the oxidation of soybean oil. RGNP as an interfacial antioxidant further improved the Pickering emulsion’ oxidative stability. This work may expand the application of wheat gluten as an antioxidant solid particle in Pickering emulsion.

Stabilization of Lead-Reduced Metal Halide Perovskite Nanocrystals by High-Entropy Alloying.

Colloidal metal halide perovskite (MHP) nanocrystals (NCs) are an emerging class of fluorescent quantum dots (QDs) for next-generation optoelectronics. A great hurdle hindering practical applications, however, is their high lead content, where most attempts addressing the challenge in the literature compromised the material’s optical performance or colloidal stability. Here, we present a postsynthetic approach that stabilizes the lead-reduced MHP NCs through high-entropy alloying. Upon doping the NCs with multiple elements in considerably high concentrations, the resulting high-entropy perovskite (HEP) NCs remain to possess excellent colloidal stability and narrowband emission, with even higher photoluminescence (PL) quantum yields, ηPL, and shorter fluorescence lifetimes, τPL. The formation of multiple phases containing mixed interstitial and doping phases is suggested by X-ray crystallography. Importantly, the crystalline phases with higher degrees of lattice expansion and lattice contraction can be stabilized upon high-entropy alloying. We show that the lead content can be approximately reduced by up to 55% upon high-entropy alloying. The findings reported here make one big step closer to the commercialization of perovskite NCs.

Resveratrol Triggered the Quick Self-Assembly of Gallic Acid into Therapeutic Hydrogels for Healing of Bacterially Infected Wounds.

Programing self-assembly of naturally bioactive molecules has been a wide topic of great significance for biomedical uses. Despite the fact that plant-derived polyphenols with catechol or pyrogallol moieties have been widely studied to construct nanocomplexes or nanocoatings via self-polymerization, there is no report on the self-assembly of these polyphenols into therapeutic hydrogels for potential applications. Here, we reported that adding a very small amount of resveratrol (Res) into the gallic acid (GA) aqueous solution could trigger the quick self-assembly of GA to form a fibrous hydrogel within 5 min through hydrogen bonds and π-π interactions. The length of GA/Res (GR) fibrils in gels varied from 100 to 1000 microns, with a diameter of around 1 μm. Notably, these GR hydrogels showed excellent colloid stability, providing better slow release and outstanding biocompatibility. Also, in vivo experiments indicated the hydrogels had high antibacterial effects and excellent wound healing capabilities in a total skin defect model via regulating the expression of inflammatory factors (IL-6, IL-1β, and TNF-α) due to the release of therapeutic agents (GA and Res) into the matrix. Overall, our results provide a new strategy to accelerate self-assembly of GA by adding Res to form hydrogels, which is further proved as a promising therapeutic carrier for wound healing.

Single-step acid-catalyzed synthesis of luminescent colloidal organosilica nanobeads

We present a single-step, room-temperature synthesis of fluorescent organosilica nanobeads (FOS NBs). The FOS NBs were synthesized under aqueous conditions using (3-aminopropyl)triethoxysilane (APTES) as the silicon source in the presence of l-ascorbic acid (L-AA). In the APTES/L-AA/water ternary phase, the hydrolysis and condensation reaction of APTES occurred under acidic conditions to form spherical FOS NBs with an average diameter of 426.8 nm. FOS NBs exhibit excellent colloidal stability in aqueous media. The formation of FOS NBs was complete within a 10 min reaction time, which indicates potential for large-scale mass-production synthesis of luminescent colloidal NBs. The FOS NBs exhibited blue photoluminescence (PL) under UV excitation in the absence of an additional high temperature calcination process or with the incorporation of any fluorophores. This phenomenon is attributed to the presence of carbon-containing defects, which act as luminescent centers formed by the reaction between amino groups in the APTES and l-ascorbic acid reductant. Finally, the results of a cytotoxicity test and cellular uptake experiments revealed that the FOS NBs showed potential as optical contrast agents for bioimaging.Graphical

Boehmite nanoparticles for potential enhancement of tribological performance of lubricants

A set of nanolubricants was prepared by varying concentrations of Boehmite nanoparticles (0.25,0.5,0.75,1 wt%) in synthetic base oil (PAO-6) with a fixed concentration of dispersant (PIBSA). This is the first study exploring Boehmite mineral for its antiwear and extreme pressure performance. Nanolubricants showed excellent colloidal stability for over 30 days without any sedimentation and improved viscosity index by 4.35% at 1 wt% concentration. Four ball tribometer was used for exploring the tribological performance of Boehmite nanolubricants. A significant 24% wear reduction over base oil was observed for optimum NPs concentration of 0.5 wt% beyond that performance deteriorated. On the other hand, increased NPs concentration enhanced load-bearing capacity for EP test and the highest enhancement of 40% was observed for 0.75 and 1 wt% concentrations. Based on surface characterization techniques, we confirm dehydration of Boehmite NPs into Aluminium oxide (Al2O3) under tribostress. We further confirm regenerative, sacrificial protective tribofilm composed of tribosintered Al2O3 along with oxide film on the rubbing surfaces.

Precursor Heterogeneity Driven Mo-Te Nanoparticle Structural Diversification for Cancer Photo-Theranostics.

Chemical reactions between homogeneous precursors are typically used to synthesize monodisperse nanoparticles with well-controlled size and morphology. It is difficult to predict the evolved nanostructures when using two heterogeneous precursors. In this study, three types of Mo-Te nanoparticles shaped like leaves, spindles, and rice grains (denoted respectively as nanoleaf, nanospindle, and nanorice) were obtained from dextrose-mediated proton-coupled electron transfer reaction between the solid polyoxomolybdate (POM) and the ionic tellurite anion as precursors. All produced nanoparticles had excellent optical absorption in the ultraviolet(UV)-visible(Vis)-near-infrared(NIR) regions, with only slight deviations among them. After confirming nanoparticles' photothermal conversion and photocatalytic activity at multiple wavelengths, the Mo-Te nanorice was tested as a potential agent for cancer treatment due to its minimum toxicity, excellent colloidal stability, and intrinsic anticancer effect. Excellent treatment efficacy and clearance were confirmed in vitro and in vivo. Due to their photoacoustic imaging capability, the injection of pristine nanoparticles could also realize phototheranostics without using additional drugs, probes, or photosensitizers.

Enhanced Anticancer Performance of Eco-Friendly-Prepared Mo-ZnO/RGO Nanocomposites: Role of Oxidative Stress and Apoptosis.

ZnO nanoparticles (NPs) have attracted great attention in cancer therapy because of their novel and tailorable physicochemical features. Pure ZnO NPs, molybdenum (Mo)-doped ZnO NPs, and Mo-ZnO/reduced graphene oxide nanocomposites (Mo-ZnO/RGO NCs) were prepared using a facile, inexpensive, and eco-friendly approach using date palm (Phoenix dactylifera L.) fruit extract. Anticancer efficacy of green synthesized NPs/NCs was examined in two different cancer cells. The potential mechanism of the anticancer activity of green synthesized NPs/NCs was explored through oxidative stress and apoptosis. The syntheses of pure ZnO NPs, Mo-ZnO NPs, and Mo-ZnO/RGO NCs were confirmed by X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS), and photoluminescence (PL). Dynamic light scattering (DLS) study indicated the excellent colloidal stability of green prepared samples. Mo-ZnO/RGO NCs exhibited threefold higher anticancer activity in human colon (HCT116) and breast (MCF7) cancer cells as compared to pure ZnO NPs. The anticancer activity of Mo-ZnO/RGO NCs was mediated through reactive oxygen species, p53, and the caspase-3 pathway. Moreover, cytocompatibility of Mo-ZnO/RGO NCs in human normal colon epithelial (NCM460) and normal breast epithelial cells (MCF10A) was much better than those of pure ZnO NPs. Altogether, green stabilized Mo-ZnO/RGO NCs exhibited enhanced anticancer performance and improved cytocompatibility because of green mediated good synergism between ZnO, Mo, and RGO. This study suggested the high nutritional value fruit-based facile preparation of ZnO-based nanocomposites for cancer therapy.

Magnetite nanoparticles synthesized using grape fruit extract: synthesis, morphology, hyperthermia application and catalytic activity in hydrogen peroxide decomposition

The paper presents a simple one-step "green" approach to the synthesis of magnetite nanoparticles. The magnetite nanoparticles were synthesized using fruit extract obtained from grape peels and grape pulp. The formation of magnetite nanoparticles was confirmed by X-ray diffraction analysis (XRD), infrared spectroscopy (IR), Mössbauer spectroscopy, scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS). The crystallites sizes of magnetite nanoparticles are 7 and 14 nm for Fe3O4-peel and Fe3O4-pulp samples, respectively. Scanning electron microscopy images show that the samples consist of agglomerated nanoparticles. The synthesized magnetite nanoparticles showed good prospects for their use in magnetic hyperthermia. SAR values ​​of 0.488 W/g and 1.330 W/g for Fe3O4-peel and Fe3O4-pulp samples, respectively. The synthesized magnetic nanoparticles show excellent colloidal stability in the aqueous solutions. The maximum hyperthermia temperatures are 42.28 °C and 42.48 °C for Fe3O4-peel and Fe3O4-pulp samples, respectively. Studies of the catalytic activity of magnetite were performed in decomposition of hydrogen peroxide in a batch mode. The degrees of H2O2 decomposition are 67.5% and 65.25% for the Fe3O4-peel and Fe3O4-pulp samples respectively. The high catalytic activity of the synthesized samples makes them promising candidates for the decomposition of hydrogen peroxide in wastewater disinfection.

Anchoring Group-Mediated Radiolabeling of Inorganic Nanoparticles─A Universal Method for Constructing Nuclear Medicine Imaging Nanoprobes.

Nuclear medicine imaging has aroused great interest in the design and synthesis of versatile radioactive nanoprobes, while most of the methods developed for radiolabeling nanoprobes are difficult to satisfy the criteria of clinical translation, including easy operation, mild labeling conditions, high efficiency, and high radiolabeling stability. Herein, we demonstrated the universality of a simple but efficient radiolabeling method recently developed for constructing nuclear imaging nanoprobes, that is, ligand anchoring group-mediated radiolabeling (LAGMERAL). In this method, a diphosphonate-polyethylene glycol (DP-PEG) decorating on the surface of inorganic nanoparticles plays an essential role. In principle, owing to the strong binding affinity to a great variety of metal ions, it can not only endow the underlying nanoparticles containing metal ions including some main group metal ions, transition metal ions, and lanthanide metal ions with excellent colloidal stability and biocompatibility but also enable efficient radiolabeling through the diphosphonate group. Based on this assumption, inorganic nanoparticles such as Fe3O4 nanoparticles, NaGdF4:Yb,Tm nanoparticles, and Cu2-xS nanoparticles, as representatives of functional inorganic nanoparticles suitable for different imaging modalities including magnetic resonance imaging (MRI), upconversion luminescence imaging (UCL), and photoacoustic imaging (PAI), respectively, were chosen to be radiolabeled with different kinds of radionuclides such as SPECT nuclides (e.g., 99mTc), PET nuclides (e.g., 68Ga), and therapeutic SPECT nuclides (e.g., 177Lu) to demonstrate the reliability of the LAGMERAL approach. The experimental results showed that the obtained nanoprobes exhibited high radiolabeling stability, and the whole radiolabeling process had negligible impacts on the physical and chemical properties of the initial nanoparticles. Through passive targeting SPECT/MRI of glioma tumor, active targeting SPECT/UCL of colorectal cancer, and SPECT/PAI of lymphatic metastasis, the outstanding potentials of the resulting radioactive nanoprobes for sensitive tumor diagnosis were demonstrated, manifesting the feasibility and efficiency of LAGMERAL.

Preparation and cellular uptake behaviors of uniform fiber-like micelles with length controllability and high colloidal stability in aqueous media

Fragmentation/disassembly of fiber-like micelles generated by living crystalline-driven self-assembly (CDSA) is usually encountered in aqueous media, which hinders the applications of micelles. Herein, we report the generation of uniform fiber-like micelles consisting of a π-conjugated oligo(p-phenylenevinylene) core and a cross-linking silica shell with grafted poly(ethylene glycol) (PEG) chains by the combination of living CDSA, silica chemistry and surface grafting-onto strategy. Owing to the presence of crosslinking silica shell and the outmost PEG chains, the resulting micelles exhibit excellent dispersity and colloidal stability in PBS buffer, BSA aqueous solution and upon heating at 80 °C for 2 h without micellar fragmentation/disassembly. The micelles also show negligible cytotoxicity toward both HeLa cervical cancer and HEK239T human embryonic kidney cell lines. Interestingly, micelles with Ln of 156 nm show the “stealth” property with no significant uptake by HeLa cells, whereas some certain amounts of micelles with Ln of 535 nm can penetrate into HeLa cells, showing length-dependent cellular uptake behaviors. These results provide a route to prepare uniform, colloidally stable fiber-like nanostructures with tunable length and functions derived for biomedical applications.

Facile synthesis of superparamagnetic nickel-doped iron oxide nanoparticles as high-performance T1 contrast agents for magnetic resonance imaging.

Small-sized iron oxide nanoparticles (IONPs) are an excellent alternative to clinical gadolinium-based contrast agents (GBCAs) in T1-weighted magnetic resonance imaging (MRI) due to their biosafety. However, their relaxation efficiency and contrast enhancement are not significant compared with those of GBCAs, and the nanomaterials face stability problems in vivo and difficulties in large-scale synthesis. Here, we designed and synthesized a series of 7 nm monodispersed biocompatible and superparamagnetic nickel-doped iron oxide nanoparticles (NiIO NPs) as safe contrast agents (CAs) for ultra-sensitive T1-weighted MR imaging. The nickel doping strategy transforms the iron oxide nanoparticles from traditional T2 CAs to T1 CAs. The Ni0.31Fe2.69O4 NPs exhibited a high longitudinal relaxivity (r1) of 9.1 mM-1 s-1, which is much higher than that of IONPs and 2.4 times that of clinical T1 CAs (Magnevist, 3.8 mM-1 s-1). NiIO NPs enable high-resolution T1-weighted and T2-weighted dual-mode MR imaging in vivo with long circulation time. These results, in hand with the excellent colloidal stability and superior biosafety, demonstrate the potential use of NiIO NPs for accurate diagnosis in deep-tissue T1-T2 dual-mode MR imaging.

Ultra-high SERS detection of consumable coloring agents using plasmonic gold nanostars with high aspect-ratio spikes.

Solution-based SERS detection by using a portable Raman instrument has emerged as an important tool due to its simplicity, and flexibility for rapid and on-site screening of analyte molecules. However, this method has several shortcomings, including poor sensitivity especially for weak-affinity analyte molecules, where there is no close contact between the plasmonic metal surface and analyte molecule. Examples of weak-affinity molecules include pigment molecules that are commonly used as a consumable coloring agent, such as allura red (AR), and sunset yellow (SY). As high consumption of colorant agents has been shown to cause adverse effects on human health, there is a strong need to develop a simple and practical sensing system with high sensitivity for these agents. Here we present a novel, highly sensitive solution-based SERS detection method for AR, and SY by using CTAC capped gold nanostars (GNS) having different aspect ratios (GNS-2, GNS-4, and GNS-5) without utilizing any aggregating agents which can enhance SERS signal however it reduces batch to batch reproducibility. The influence of the aspect ratio of GNS on SERS properties was investigated. We have achieved a limit of detection (LOD) of AR and SY as low as 0.5 and 1 ppb, respectively by using GNS-5 with the advantages of minimal sample preparation by just mixing the analyte solution into a well plate containing GNS solution. In addition, excellent colloidal stability and reproducibility have further enhanced the applicability in real-world samples. Overall, our results evidence that the solution-based SERS detection platform using high aspect-ratio GNS can be applied for practical application to detect pigment molecules in real samples with satisfactory results.

Targeted Theranostic 111In/Lu-Nanotexaphyrin for SPECT Imaging and Photodynamic Therapy.

Theranostic nanoparticles aim to integrate diagnostic imaging and therapy to facilitate image-guided treatment protocols. Herein, we present a theranostic nanotexaphyrin for prostate-specific membrane antigen (PSMA)-targeted radionuclide imaging and focal photodynamic therapy (PDT) accomplished through the chelation of metal isotopes (In, Lu). To realize nanotexaphyrin's theranostic properties, we developed a rapid and robust 111In/Lu-nanotexaphyrin radiolabeling method using a microfluidic system that achieved a high radiochemical yield (>90%). The optimized metalated nanotexaphyrin displayed excellent chemical, photo, and colloidal stabilities, potent singlet oxygen generation, and favorable plasma circulation half-life in vivo (t1/2 = 6.6 h). Biodistribution, including tumor accumulation, was characterized by NIR fluorescence, SPECT/CT imaging, and γ counting. Inclusion of the PSMA-targeting ligand enabled the preferential accumulation of 111In/Lu-nanotexaphyrin in PSMA-positive (PSMA+) prostate tumors (3.0 ± 0.3%ID/g) at 48 h with tumor vs prostate in a 2.7:1 ratio. In combination with light irradiation, the PSMA-targeting nanotexaphyrin showed a potent PDT effect and successfully inhibited PSMA+ tumor growth in a subcutaneous xenograft model. To the best of our knowledge, this study is the first demonstration of the inherent metal chelation-driven theranostic capabilities of texaphyrin nanoparticles, which, in combination with PSMA targeting, enabled prostate cancer imaging and therapy.

Pulmonary Delivery of Reactive Oxygen Species/Glutathione-Responsive Paclitaxel Dimeric Nanoparticles Improved Therapeutic Indices against Metastatic Lung Cancer.

Chemotherapeutics often failed to elicit optimal antitumor responses against lung cancer due to their limited exposure and accumulation in tumors. To achieve an effective therapeutic outcome of paclitaxel (PTX) against metastatic lung cancer with attenuated systemic and local toxicities, pulmonary delivery of redox-responsive PTX dimeric nanoparticles (NPs) was introduced. PTX dimers conjugated through variable lengths of diacid linkers containing disulfide bonds (-SS-) (i.e., α-PTX-SS-PTX, β-PTX-SS-PTX, and γ-PTX-SS-PTX) were initially synthesized and were subsequently self-assembled into uniform nanosized particles in the presence of vitamin E TPGS with high drug loading capacity (DE > 97%). Among various redox-sensitive scaffolds, β-PTX-SS-PTX NPs exhibited an optimal reactive oxygen species/glutathione-responsive drug release behavior, causing a lower local toxicity profile of PTX in the lungs. The scaffolds also demonstrated excellent colloidal stability, cellular uptake efficiency, and discriminating cytotoxicity between cancer and healthy cells. Further, they depicted an improved lung retention as compared to the control nanovesicles (β-PTX-CC-PTX) devoid of the redox-sensitive disulfide motif. In the B16F10 melanoma metastatic lung cancer mouse model, intratracheally delivered β-PTX-SS-PTX NPs exhibited a stronger anticancer potential with reduced systemic toxicity as compared to Taxol intravenous injection containing an equivalent PTX dose. The PTX dimeric NPs could also dramatically reduce the local toxicity relative to Taxol following their pulmonary delivery. Near-Infrared Fluorescent Micelles from Poly(norbornene) Brush Triblock Copolymers for Nanotheranostics.

This contribution describes the design and synthesis of multifunctional micelles based on amphiphilic brush block copolymers (BBCPs) for imaging and selective drug delivery of natural anticancer compounds. Well-defined BBCPs were synthesized via one-pot multi-step sequential grafting-through ring-opening metathesis polymerization (ROMP) of norbornene-based macroinitiators. The norbornenes employed contain a poly(ethylene glycol) methyl ether chain, an alkyl bromide chain, and/or a near-infrared (NIR) fluorescent cyanine dye. After block copolymerization, post-polymerization transformations using bromide-azide substitution, followed by the strain-promoted azide-alkyne cycloaddition (SPAAC) allowed for the functionalization of the BBCPs with the piplartine (PPT) moiety, a natural product with well-documented cytotoxicity against cancer cell lines, via an ester linker between the drug and the polymer side chain. The amphiphilic BBCPs self-assembled in aqueous media into nano-sized spherical micelles with neutral surface charges, as confirmed by dynamic light scattering analysis and transmission electron microscopy. During self-assembly, paclitaxel (PTX) could be effectively encapsulated into the hydrophobic core to form stable PTX-loaded micelles with high loading capacities and encapsulation efficiencies. The NIR fluorescent dye-containing micelles exhibited remarkable photophysical properties, excellent colloidal stability under physiological conditions, and a pH-induced disassembly under slightly acidic conditions, allowing for the release of the drug in a controlled manner. The in vitro studies demonstrated that the micelles without the drug (blank micelles) are biocompatible at concentrations of up to 1 mg mL-1 and present a high cellular internalization capacity toward MCF-7 cancer cells. The drug-functionalized micelles showed in vitro cytotoxicity comparable to free PPT and PTX against MCF-7 and PC3 cancer cells, confirming efficient drug release into the tumor environment upon cellular internalization. Furthermore, the drug-functionalized micelles exhibited higher selectivity than the pristine drugs and preferential cellular uptake in human cancer cell lines (MCF-7 and PC3) when compared to the normal breast cell line (MCF10A). This study provides an efficient strategy for the development of versatile polymeric nanosystems for drug delivery and image-guided diagnostics. Notably, the easy functionalization of BBCP side chains via SPAAC opens up the possibility for the preparation of a library of multifunctional systems containing other drugs or functionalities, such as target groups for recognition.

Experimental study on properties of hybrid stable & surfactant-free nanofluids GNPs/CNCs (Graphene nanoplatelets/cellulose nanocrystal) in water/ethylene glycol mixture for heat transfer application

The heat transfer capacity of any thermal cooling system depends on two factors, i.e., the selection of the coolant and the geometrical pattern of the approach. This article summarizes nanofluids' preparation ranging from 0.01 % to 0.2 % using Graphene nanoplatelets & CNC dispersed in a base fluid. The combination of water with ethylene glycol (EG) is a form of customary heat transmit liquids regularly utilized in numerous energy practices to maintain the water's decent cooling (or heating) capability; thus, 60:40 ratio of EG: W mixture used as the base fluid for thermo-physical properties enhancements. These nanofluids prepared are not used with surfactants as it results in generating bubbles and contaminating the heat transfer channels, influencing the overall performance. XRD &FESEMtechniques were used to analyze the surface. The investigated nanofluids remained stable, with no substantial sedimentation for 30 days. The results of GNPs/CNC nanofluids at 0.1% volume concentration has proper stability showing excellent colloidal stability in the base fluid of EG: W at a ratio of 60:40. The present hybrid nanofluid has the ability to switch the traditional heat transfer fluids leading to efficient & compact thermal structures.
