Evaluation of antibacterial activity and susceptibility antibiotics of native lactic acid bacteria from tilapia (Oreochromis niloticus) in Ivory Coast

Lactic Acid Bacteria probiotics play a crucial role in improving aquaculture productivity however, their use required beforehand a rigorous selection. This study appeared as a preliminary research in LAB potential probiotics selection. The aim of study was to select LAB strains potentially probiotic isolated from gut intestine of tilapia (Oreochromis niloticus) based on antibacterial activity and susceptibility antibiotics tests. The antibacterial activity was carried by agar well diffusion method against five pathogen indicators (E. coli ATCC 25922, K. pneumoniae ATCC 43816, P. aeruginosa ATCC 27853, P. mirabilis JCM1669, and S. aureus ATCC 25913). The susceptibility antibiotic test was performed by disc diffusion method by 12 antibiotics use. Seventy-two (72) from 154 LAB isolates completed the antibacterial activity test. With 79.08% inhibition, S. aureus was the pathogen most inhibited, whereas E. coli had the least inhibition, at 61.44%. In terms of antibiotic susceptibility testing, out of the 72 LAB isolates from earlier studies, only 25 LAB isolates demonstrated resistance to ciprofloxacin, gentamycin, and oxacillin and sensitivity to nine out of the twelve antibiotics employed. The 25 LAB isolates seemed suitable candidates for more probiotic tests.

In vitro and in vivo evaluation of osteogenesis and antibacterial activity of Mg[sbnd]Ga alloys

MgGa alloys are considered highly potential biodegradable materials, owing to its good mechanical properties and appropriate corrosion resistance. However, it is still far from application due to the lack of biological evaluation. In the present study, biocompatibility, osteogenesis and antibacterial activity of extruded Mg–xGa (x = 1 and 5 wt%) alloys were investigated by in vitro cell culture experiments and in vivo implantation. The cell adhesion and proliferation of osteoblast precursor cells (MC3T3-E1) showed the excellent cytocompatibility of Mg–1Ga and poor cytocompatibility of Mg–5Ga. The osteogenic activity was evaluated and revealed that Ga3+ in the Mg–1Ga extract had the ability to enhance osteogenic differentiation through the facilitation of its early stages. In vivo studies in a rat femoral condyle model revealed that both Mg–1Ga and Mg–5Ga significantly promoted new bone formation without causing any adverse effects. Mg–5Ga exhibited a much higher corrosion rate in vivo than Mg–1Ga. Its osteogenic activity was better due to the rapid release of Mg2+ and Ga3+, but this caused premature structural integrity loss. Mg–1Ga and Mg–5Ga released Ga3+ to inhibit E. coli and S. aureus, with antibacterial rate increasing with Ga content. Our studies demonstrate that Mg–Ga alloys have the potential to be used as osteogenic and antibacterial implant materials. Statement of significanceThis study evaluates the biocompatibility, osteogenesis, and antibacterial activity of Mg–Ga alloys, which are promising biodegradable materials for medical applications. The study finds that Mg–1Ga exhibits excellent cytocompatibility and promotes osteogenic differentiation, facilitating the early stages of osteoblast precursor cell development. Fabrication of poly (1,4-phenylene ether ether sulfone) modified with MWCNTs/reduced (GO-oxSWCNTs) NCs for enhanced antimicrobial activities

Poly (1,4-phenylene ether ether sulfone) (PEES) is a commonly used polymer in membrane technology for water treatment applications such as water purification and blood dialyzing in hemodialysis. In this study, PEES was chemically modified by nitration, yielding nitrated Poly (1,4-phenylene ether ether sulfone) (NPEES). Following that, NPEES nanocomposites (NCs) comprise multi-walled carbon nanotubes (MWCNTs), and the process involved the synthesis of reduced graphene oxide-oxidized single-walled carbon nanotubes, abbreviated as reduced (GO-oxSWCNTs). Various characterization techniques were used on the created membranes, such as Fourier-transform infrared spectroscopy (AT-FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis. All polymer nanocomposites were found to be amorphous, according to the XRD patterns. SEM scans revealed random crater-like features on the surface of NPEES, but MWCNTs and reduced (GO-oxSWCNTs) NCs were distributed evenly on the polymer surface. The primary goal of this study was to evaluate the antimicrobial activity of modified NPEES membranes against two Gram-positive bacteria, Staphylococcus aureus (S. aureus) and Bacillus subtilis (B. subtilis), two Gram-negative bacteria, Pseudomonas aeruginosa (P. aeruginosa) and Escherichia coli (E. coli), and a fungus, Candida albicans (C. albicans). All modified membranes, including NPEES, NPEES/MWCNTs NCs, and NPEES/MWCNTs/reduced (GO-oxSWCNTs) NCs, exhibited antibacterial activity against S. aureus and B. subtilis. Notably, when compared to NPEES/MWCNTs NCs and NPEES/MWCNTs/reduced (GO-oxSWCNTs) NCs, the NPEES membrane had higher antibacterial activity, generating a 12 mm inhibitory zone. Furthermore, molecular docking studies revealed a strong fit of the tested polymer nanocomposites into the DNA gyrase B active site (PDB ID: 4uro), which was consistent with the practical results of their antibacterial activity evaluation.

Synthesis, Characterization and Evaluation of Antibacterial activity of Amine-Boranes

Amine-Boranes (AB) are an elite class of reagent and belongs to the Borane-Lewis family, has found extensive use in chemical synthesis. Interesting aspect of AB reagents are to display chemo selectivity among the functional groups. The present investigation highlights on the innovative, green synthesis and characterization of the three of unique AB complexes, including triethylamine-borane (aliphatic), aniline-borane (aromatic), and (aromatic substituted) N,N-dimethylaniline-borane (DMAB) under an open air condition. The 11B NMR technique was used to characterize the synthesized AB reagents. The chemical shift values obtained were in accordance with the results of previous research studies, and it's interesting to note that there is a difference in 11B-NMR chemical shift values between the borane complexes of aromatic and aliphatic amines. The antibacterial efficacy of three AB reagents has been stated for the first instance by using disc diffusion method against four bacterial strains: Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa, and Staphylococcus aureus. When compared to other AB complexes, the DMAB reagent demonstrated significant antibacterial properties against the four different bacterial species.

Formulation of mouth rinse from natural plant\'s leaf extract and evaluation of antibacterial activity

Herbal liquid formulations are the preparation for oral health related issues like toothache, swelling, redness, bacterial or fungal infection in mouth, tooth decay, bleeding gums, weakened gums, etc. Such preparation also improves gums strength, removing bad breath and give fresh feel during good start of the day. Dentists generally use such preparations as antimicrobials before or after oral surgery to sanitize the surface of affected gums and teeth to avoid further contamination. Herbs like Tulsi, Clove, Mint, Ginger etc., are generally used either alone or in combination in herbal mouth-wash preparations which also render antibacterial activities. The other herbs like Eucalyptus and Liquorices have been reported to possess antibacterial, anti-oxidant, aromatic, anti- inflammatory and sweetening properties. In the present work, the research continues with formulation and evaluation of a herbal mouth rinse by using a combination of selected herbal extracts of Tulsi, Bael, Eucalyptus leaves and Liquorices rhizome for their antimicrobial activity against S. aureus using agar well diffusion plate technique. It was found that the formulated herbal mouth rinse or preparations gave satisfactory results in controlling the microorganism. Different physicochemical tests of prepared preparations were performed to figure out their pH, phase separation, homogeneity and scent.,

This study aims to develop innovative heterocyclic nanocomposites incorporating silver nanoparticles (SNPs) for potential therapeutic applications targeting infections, gastric ulceration, inflammation, and oxidative damage. By synthesizing new derivatives of spiro-thiazolidine-carbonitrile (Py-ST-X) and incorporating them into Ag nanoparticles (AgNPs) and carbon nanotubes (CNTs), we have prepared Ag@Py-ST-X and Ag@Py-ST-X@CNT nanocomposites, respectively. The physical properties of these materials were studied using XRD, TEM, SEM, and Zeta potential techniques. In our investigation involving rats with gastric ulcers, we observed noteworthy inhibitory effects on gastric acid enzyme activity, specifically H+/K+ATPase, by Ag@Py-ST-NO2 and Ag@Py-ST-Br nanocomposites, demonstrating reductions of 25 and 34%, respectively, compared to untreated ulcers. Nanotubulation of these compounds further improved their inhibitory efficacy to 29 and 45%, respectively. Additionally, these nanoparticles showed the most potent myeloperoxidase (MPO)-inhibitory activity, demonstrating 36 and 49% inhibition, respectively, with nanotubulated versions reaching 44 and 53%. Moreover, Ag@Py-ST-NO2@CNT and Ag@Py-ST-Br@CNT nanotubes showed significant antioxidant activity, reducing thiobarbituric acid reactive substances (TBARS) by 35 and 51%, and hydrogen peroxide (H2O2) levels by 49 and 71%, respectively. These therapeutic effects were confirmed by reductions in gastric surface area (GSA) by 44% and 52%, a decrease in ulcer index (UI) from 80% to 44 and 38%, and an increase in curative index (CI) from 19 to 55 and 62% following administration of Ag@Py-ST-NO2@CNT and Ag@Py-ST-Br@CNT, respectively. Histological studies support these findings, suggesting the potential of these nanocomposites as promising candidates for treating various disorders.

Structural and computational exploration of zwitterionic and quinoidal forms in Schiff base compound

The stable NH-form of the Schiff base compound, (Z)-6-(((4-chlorobenzyl) amino) methylene)-2-hydroxycyclohexa-2,4-dienone was synthesized and characterized by FTIR, SEM, EDAX anlysis and X-ray crystallographic analysis. Structural analysis was carried out to examine the NH-form of the halogen-based Schiff base compound. Additionally, an analysis of bond lengths, based on previously reported NH-forms of other Schiff bases, was conducted to distinguish between the zwitterion and quinoid structures. Further, Hirshfeld surface analysis was performed to visualize and quantify intermolecular interactions, supported by shape index, 2D fingerprint plots, and Enrichment ratio analyses, highlighting the importance of these interactions. Energy frameworks were established to deepen the understanding of crystal packing through the computation of intermolecular interaction energies. QTAIM analysis was performed to visualize and quantify intramolecular interactions, while Density Functional Theory (DFT) studies elucidated molecular properties such as the HOMO-LUMO energy gap and its derivatives. Evaluation of antibacterial activity against S. aureus was conducted via the disc diffusion assay supplemented by molecular docking studies. This comprehensive approach provides valuable structural insights into the NH-form, as well as molecular and biological aspects of (Z)-6-(((4-chlorobenzyl) amino) methylene)-2-hydroxycyclohexa-2,4-dienone.

Evaluation of phytoactive contents and antibacterial activities of green synthesised cerium oxide nanoparticles using Melastoma sp. leaf extract as the capping agent

Simple and green methods of developing nanoparticles (NPs) have attracted the attention of researchers. Literature on utilising leaf extract to prepare cerium oxide (CeO2 NPs) is scarce. The present study synthesised leaf-mediated-CeO2 NPs to produce nanopowders of controllable sizes for further applications. The study is the first to report the optimised parameters (pH 7, 5 g/150 mL concentration of the leaf extract, and 3 h of reaction time) of procuring CeO2 NPs using Melastoma sp. leaf extract as the capping agent with excellent properties. The absorbance of the NPs suspension obtained in this study was recorded at approximately 252 nm with Ultraviolet–Visible (UV–Vis) Spectroscopy. Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM), X-ray Diffraction (XRD), and Transmission Electron Microscopy (TEM) were also utilised to characterise and confirm the CeO2 NPs prepared. The XRD spectra documented the purity of the NPs at specific diffraction patterns, while TEM revealed the spherical form of the NPs with a particle size of 16 nm. The formation of CeO2 NPs has been confirmed from the FTIR spectra procured, which exhibited a Ce–O peak at 555 nm. Phytochemical screening test and FT-IR analysis of leaf extract revealed the existence of flavonoids, terpenoids, sugars, saponins, quinones, and glycosides. The NPs suspensions of varying concentrations (control, 50, 100, 150, 200, and 250 μg/mL) were prepared and employed for evaluations against Gram-positive and -negative bacteria. Resultantly, CeO2 NPs demonstrated antibacterial activities against both bacteria types. The highest antibacterial activities were recorded against E. coli and K. pneumonia at 1.83 ± 0.137 and 1.83 ± 0.14 mm maximum inhibition zones, respectively, at 250 mg/uL of the NPs.

Biogenic Synthesis of Copper Oxide Nanoparticles from Aloe vera: Antibacterial Activity, Molecular Docking, and Photocatalytic Dye Degradation.

Green synthesis methods offer a cost-effective and environmentally friendly approach to producing nanoparticles (NPs), particularly metal-based oxides. This study explores the green synthesis of copper oxide nanoparticles using Aloe vera (Aloe barbadensis Miller) leaf extract. The characterization revealed a unique sago-shaped morphology revealed by field-emission scanning electron microscopy and X-ray diffraction analysis. Distinctive metal-oxygen bonds at 521 and 601 cm-1 were confirmed by Fourier-transform infrared (FT-IR) spectroscopy. Furthermore, UV-visible spectroscopy revealed absorbance at 248 nm, suggesting electron transitions across energy bands and varying surface conduction electrons. The band gap value indicated the presence of quantum confinement effects, which were probably caused by the distinctive morphology and surface structure of the biogenic NPs. Additionally, molecular docking studies were carried out against key proteins of Salmonella typhi and Listeria monocytogenes, namely, listeriolysin O (PDB ID: 4CDB), internalin (InlA) (PDB ID: 1O6T), Salmonella effector protein (SopB) (PDB ID: 4DID), and YfdX (PDB ID: 6A07) using AutoDock 4.2. The results revealed binding energies against S. typhi and L. monocytogenes proteins, indicating potential interactions establishing the foundation for further in-depth understanding of the molecular basis underlying the observed antibacterial effects in vitro against S. typhi, Klebsiella pneumoniae, Pseudomonas aeruginosa, and L. monocytogenes. Antibacterial activity evaluation yielded impressive results, with CuO NPs displaying significant activity against S. typhi and L. monocytogenes, exhibiting zones of inhibition values of 13 ± 0.02 and 15 ± 0.04 mm, respectively. Moreover, the CuO NPs demonstrated remarkable photocatalytic efficacy, resulting in the degradation of 77% of the methylene blue dye when exposed to UV irradiation. This study highlighted the potential of green-synthesized CuO NPs derived from A. vera with their unique morphology, interesting spectroscopic properties, and promising antibacterial and photocatalytic activities.

Synthesis, molecular docking simulation, and antimicrobial activities of novel bis-heterocycles linked to piperazine and vanillin units as novel hybrid molecules via Hantzsch, Biginelli, and Michael’s reactions

Bacterial infections are a global issue, causing sickness and death, mainly in developing nations. Resistance to current medicines is a significant issue in healthcare. Overcoming the resistance problem will necessitate the development of molecules with novel modes of action that do not cross-react with existing medicines. Synthesis of new bis-heterocycles linked to the piperazine core and vanillin unit is disclosed in this regard. The target compounds were produced via the reaction of 4,4′-((piperazine-1,4-diylbis(2-oxoethane-2,1-diyl))bis(oxy))bis(3-methoxybenzaldehyde) with various reagents using the Hantzsch, Biginelli, and Michael’s reactions. All the generated compounds were tested for their antibacterial activity against a variety of bacterial strains. The antibacterial activity evaluations revealed that compound 32 has the most promising activity against Pseudomonas aeruginosa (19.5 ± 0.7 mm) compared to Ofloxacin (17 mm) with minimum inhibitory concentration (MIC) equaled to 625 µg/mL. A molecular docking investigation revealed that compound 32 had the greatest binding affinity for bacterial enoyl reductase (−36.13 ± 0.2 Kcal/mole) when compared to the co-crystallized ligand (−16.8 ± 0 Kcal/mole).

Green Synthesis, Characterization, and Evaluation of Photocatalytic and Antibacterial Activities of Co3O4-ZnO Nanocomposites Using Calpurnia aurea Leaf Extract.

The green synthesis of transition metal oxide nanocomposites using plant extracts is a new and effective method that avoids the involvement of hazardous chemicals. Nondegradable organic pollutants and antibiotic drug resistance have become serious public health issues worldwide. Hence, the main objective of this study is to synthesize Co3O4-ZnO nanocomposites using Calpurnia aurea leaf extract and evaluate its photocatalytic and antibacterial activities. The green synthesized particles were characterized using UV-vis spectra, Fourier transform infrared spectroscopy, X-ray diffraction techniques, and scanning electron microscopy combined with energy-dispersive X-ray studies. The synthesized particles were found to be crystalline in nature with average crystallite sizes of 23.82, 14.79, 15.99, 16.46, and 21.73 nm. Scanning electron microscopy shows the spherical morphology of Co3O4-ZnO NCs, and energy-dispersive X-ray analysis confirms the formation of highly pure ZnO NPs and Co3O4-ZnO NCs. The photocatalytic activity was performed under natural sunlight using malachite green as an organic dye pollutant. The green synthesized ZnO NPs, Co3O4 NPs, 1:4, 1:3, and 1:2 Co3O4-ZnO NCs showed high degradation efficiency after 60 min of irradiation. The synthetic material showed good potential against Staphylococcus aureus and Escherichia coli, with the highest growth inhibition recorded by 1:2 Co3O4-ZnO NCs. The kinetics study of the photocatalytic degradation was confirmed as pseudo first order, and the possible mechanisms for both photocatalytic and antibacterial activities were clearly determined.

Evaluation of antibacterial, cytotoxicity, and apoptosis activity of novel chromene-sulfonamide hybrids synthesized under solvent-free conditions and 3D-QSAR modeling studies

In this study, eleven novel chromene sulfonamide hybrids were synthesized by a convenient method in accordance with green chemistry. At first, chromene derivatives (1–9a) were prepared through the multi-component reaction between aryl aldehydes, malononitrile, and 3-aminophenol. Then, synthesized chromenes were reacted with appropriate sulfonyl chlorides by grinding method to give the corresponding chromene sulfonamide hybrids (1–11b). Synthesized hybrids were obtained in good to high yield and characterized by IR, 1HNMR, 13CNMR, CHN and melting point techniques. In addition, the broth microdilution assay was used to determine the minimal inhibitory concentration of newly synthesized chromene-sulfonamide hybrids. The MTT test was used to determine the cytotoxicity and apoptotic activity of the newly synthesized compounds against fibroblast L929 cells. The 3D‑QSAR analysis confirmed the experimental assays, demonstrating that our predictive model is useful for developing new antibacterial inhibitors. Consequently, molecular docking studies were performed to validate the findings of the 3D-QSAR analysis, confirming the potential binding interactions of the synthesized chromene-sulfonamide hybrids with the target enzymes. Molecular docking studies were employed to support the 3D-QSAR predictions, providing insights into the binding interactions between the newly synthesized chromene-sulfonamide hybrids and their target bacterial enzymes, thereby reinforcing the potential efficacy of these compounds as antibacterial agents. Also, some of the experimental outcomes supported or conflicted with the pharmacokinetic prediction (especially about compound carcinogenicity). The performance of ADMET predictor results was assessed. The work presented here proposes a computationally driven strategy for designing and discovering a new sulfonamide scaffold for bacterial inhibition.

Construction of self‐healing gallium (III)‐cross‐linked konjac glucomannan/polyacrylamide hydrogels for efficiently killing bacteria and accelerating wound healing

AbstractGallium ions have a special action mechanism that interferes bacterial iron metabolism, thereby possessing great potential for the treatment of bacterial infections. In this work, gallium ions‐cross‐linked konjac glucomannan/polyacrylamide (KGM/PAM/Ga3+) hydrogels are successfully constructed with satisfactory swelling ability, appropriate mechanical properties and adjustable degradation performance. Ga ions as a kind of cross‐linking agent can enhance the stability, and more importantly, improve the bactericidal ability of hydrogel, thus forming novel functional KGM/PAM/Ga3+ hydrogels for treating wounds. After the evaluation of antibacterial activity and biocompatibility, KGM/PAM/Ga3+ hydrogels can effectively inhibit the proliferation of bacteria through the gradual and sustainable release of Ga ions from the hydrogel with negligible cytotoxicity and good blood compatibility. Furthermore, they possess exceptional self‐healing behavior, and can be readily adapted to varying degrees of bending, which matches skin wound dressing application. Finally, the results of wound healing performance and histological evaluation demonstrate that KGM/PAM/Ga3+ hydrogels can efficiently accelerate infected wound healing and facilitate the regeneration of skin tissues.
