Microbial communities in the liver and brain are informative for postmortem submersion interval estimation in the late phase of decomposition: A study in mouse cadavers recovered from freshwater.

Bodies recovered from water, especially in the late phase of decomposition, pose difficulties to the investigating authorities. Various methods have been proposed for postmortem submersion interval (PMSI) estimation and drowning identification, but some limitations remain. Many recent studies have proved the value of microbiota succession in viscera for postmortem interval estimation. Nevertheless, the visceral microbiota succession and its application for PMSI estimation and drowning identification require further investigation. In the current study, mouse drowning and CO2 asphyxia models were developed, and cadavers were immersed in freshwater for 0 to 14 days. Microbial communities in the liver and brain were characterized via 16S rDNA high-throughput sequencing. Only livers and brains collected from 5 to 14 days postmortem were qualified for sequencing. There was significant variation between microbiota from liver and brain. Differences in microbiota between the cadavers of mice that had drowned and those only subjected to postmortem submersion decreased over the PMSI. Significant successions in microbial communities were observed among the different subgroups within the late phase of the PMSI in livers and brains. Eighteen taxa in the liver which were mainly related to Clostridium_sensu_stricto and Aeromonas, and 26 taxa in the brain which were mainly belonged to Clostridium_sensu_stricto, Acetobacteroides, and Limnochorda, were selected as potential biomarkers for PMSI estimation based on a random forest algorithm. The PMSI estimation models established yielded accurate prediction results with mean absolute errors ± the standard error of 1.282 ± 0.189 d for the liver and 0.989 ± 0.237 d for the brain. The present study provides novel information on visceral postmortem microbiota succession in corpses submerged in freshwater which sheds new light on PMSI estimation based on the liver and brain in forensic practice.

Estimation of Early Postmortem Interval from Long Noncoding RNA Gene Expression in the Incised Cutaneous Wound: An Experimental Study.

The assessment of alteration of postmortem RNA expression has forensic significance in estimating postmortem interval. To evaluate wound healing progression and the effect of different postmortem intervals, histopathological changes, immunohistochemical matrix metalloproteinase-9 (MMP-9) expression, and long noncoding fatty acid oxidation (lncFAO), RNA expression was assessed in the incised cutaneous wound model. A full-thickness cutaneous wound was inflicted on 75 rats. All 15 rats were sacrificed at different post-infliction intervals (0, 2, 4, 8 and 10 days), and the cutaneous wounds (n = 5) were excised at different postmortem intervals (0, 5, and 24 h after euthanasia). The maximal inflammatory healing stage was detected at day 4 post-infliction, while near complete healing, thick mature collagen deposition was detected at day 10 post-infliction. LncFAO expression was significantly over-expressed with increasing wound age. MMP-9 was detectable on injury day with continuous elevation until 8 days post-wounding, which later decreased. Although histopathological and immunohistochemical examinations within 24 h postmortem did not show any remarkable changes, lncFAO RNA expression showed a significant negative correlation with hours passed since death. The combined use of histopathological changes, immunohistochemical expression of MMP-9, and molecular expression of lncFAO could be appropriate in wound dating verification. Among these factors, lncFAO could be a reliable indicator in postmortem interval estimation.

Post-Mortem Interval and Microbiome Analysis through 16S rRNA Analysis: A Systematic Review.

The determination of the Post-Mortem Interval (PMI) is an issue that has always represented a challenge in the field of forensic science. Different innovative approaches, compared to the more traditional ones, have been tried over the years, without succeeding in being validated as successful methods for PMI estimation. In the last two decades, innovations in sequencing technologies have made it possible to generate large volumes of data, allowing all members of a bacterial community to be sequenced. The aim of this manuscript is to provide a review regarding new advances in PMI estimation through cadaveric microbiota identification using 16S rRNA sequencing, in order to correlate specific microbiome profiles obtained from different body sites to PMI. The systematic review was performed according to PRISMA guidelines. For this purpose, 800 studies were identified through database searching (Pubmed). Articles that dealt with PMI estimation in correlation with microbiome composition and contained data about species, body site of sampling, monitoring time and sequencing method were selected and ultimately a total of 25 studies were considered. The selected studies evaluated the contribution of the various body sites to determine PMI, based on microbiome sequencing, in human and animal models. The results of this systematic review highlighted that studies conducted on both animals and humans yielded results that were promising. In order to fully exploit the potential of the microbiome in the estimation of PMI, it would be desirable to identify standardized body sampling sites and specific sampling methods in order to align data obtained by different research groups.

POST-MORTEM INTERVAL ESTIMATION BASED ON LIVER AND STRIATED MUSCLE COMPUTED TOMOGRAPHY ATTENUATIONS IN RATS

OBJECTIVE: The interest on post-mortem computed tomographic studies targeting post-mortem interval (PMI) estimations is increasing. Most studies have been conducted in an uncontrolled or relatively less controlled condition. However, this study investigates the usefulness of computed tomography attenuations of the liver and striated muscles in PMI estimation under strictly controlled environmental conditions. MATERIAL AND METHODS: Post-mortem computed tomography images of 30 Wistar Albino rats were obtained in the first 30 min after death and 12, 24, 36, 48, 72, 96, 120, 144 and 168-hours following death. Repeated attenuations of both thelateral lobes of the liver and paraspinal muscle were recorded in 0.12-cm2 regions of interest. RESULTS: The attenuation values of the liver and paraspinal muscles were higher 12-hour post-mortem. A significant difference was observed between the images obtained 30 minutes post-mortem and those obtained 12 hours post-mortem in both male (p=0.01) and female rats (p=0.01). Different cut-off attenuation values for the liver and muscle tissue were determined to estimate the post-mortem interval. CONCLUSIONS: The use of attenuation values of the liver and striated muscles with the proposed model was found to be beneficial for the determination of PMI. The cut-off values of attenuation, especially for the liver, can be used in the delimitation of the post-mortem interval.

New Advances, Challenges and Opportunities in Forensic Applications of Microbiomics.

The succession of microbiota is closely associated with several essential factors, including race, sex, health condition, lifestyle, postmortem interval, etc., and it has great potential application value in forensic medicine. This paper summarizes recent studies on the forensic applications of the microbiome, including individual identification, geographical feature identification, origin identification of the tissue or body fluid, and postmortem interval estimation, and introduces the current machine learning algorithms for microbiology research based on next-generation sequencing data. In addition, the current problems facing forensic microbiomics such as the extraction and preservation of samples, construction of standardization and database, ethical review and practical applicability are discussed. Future multi-omics studies are expected to explore micro ecosystems from a comprehensive and dynamic perspective, to promote the development of forensic microbiomics application.

Visualization of Literature Information on Postmortem Interval Estimation Indexed by CNKI Database from 1990 to 2020.

To explore the development process of the postmortem interval (PMI) research in China from January 1990 to August 2020, research hotspots in different periods, authors and cooperation between institutions, and to provide a reference for the better development of PMI inference research through the visualization of the literature information of the PMI estimation research indexed in China National Knowledge Infrastructure (CNKI). The information visualization analysis software CiteSpace 5.7.R1 was used to carry out big data analysis on hotspots, high-frequency keywords, authors, institutions and other information in the research literature on PMI inference from January 1990 to August 2020 indexed in CNKI. The peak time of publication of PMI was from 2006 to 2010 with 114 articles. In keyword co-occurrence network, the effective hot words were forensic entomology, DNA content analysis and some emerging words such as artificial intelligence and big data. In the cooperation network of institutions, the high-frequency institutions were mainly the scientific research institutions. The author cooperation network showed a trend of co-aggregation and multi-cooperation. With the development of science and technology, the research on PMI estimation based on traditional methods is mature and novel strategies are emerging. Big data and artificial intelligence combined with forensic science provide new research directions on PMI estimation.

Dental DNA as an Indicator of Post-Mortem Interval (PMI): A Pilot Research.

Teeth have proven to be a reliable source of DNA for forensic analysis as the pulp is rich in cells and protected from damaging factors and contamination by dental hard tissues. The pilot study aims to evaluate the feasibility of Next-Generation sequencing analysis on dental pulp to detect genetic mutations in DNA caused by post-mortem cell necrosis. We used a 56-gene oncopanel kit on a sample of 17 teeth extracted from living patients. Time of the tooth avulsion was assumed as death of the individual and Post-mortem Interval (PMI) was the time elapse since the DNA extraction and analysis. Days and Accumulated Degree Days (ADD) were assumed as measures of PMI that ranged between 0 to 34 days. Only 38 of the 56 considered genes proved to be affected by mutations (101), thus being of forensic interest. More specifically, 14 mutations occurred only in a specific range of PMIs/ADD; 67 were detected (alone or as clusters of the same gene) at specific PMI/ADD; 22 occurred at every PMI/ADD, except for some specific intervals. Since dental pulp was not targeted by any oncological diseases and all teeth were intact, vital, and from patients with unremarkable medical history, it could be assumed that mutations were due to post-mortem DNA changes induced by pulp death and the increasing time elapse since death. This pilot study found encouraging results in the application of NGS analysis on dental DNA, especially for PMIs of several days for which the traditional tools for PMI estimation have limitations. Further research on a larger sample of PMI and validation research on a larger sample of PMI and validation of the results are indeed necessary.

Effects of Tissue Type and Temperature on Selected Life-History Traits of the Flesh Fly, Sarcophaga crassipalpis (Macquart, 1839) (Diptera: Sarcophagidae).

The flesh fly, Sarcophaga crassipalpis Macquart 1839 (Diptera: Sarcophagidae), colonizes estimation of the minimum post-mortem interval (min-PMI). This study examined the effects of different types of tissues and temperature on the development of S. crassipalpis. To the best of our knowledge, the present study is the first to investigate the effects of life-history information of S. crassipalpis. Larvae were reared on three tissue types, chicken heart, bovine minced meat, and bovine tongue, at five temperatures, 15, 20, 25, 30, and 32°C. Pupal and larval development time, adult weight, and pupal and larval survival differed significantly for tissue type and temperature. Temperature had a significant effect on pupal weight but tissue type did not significantly affect pupal weight. The duration of the larval and pupal stages (except of chicken heart, 30°C) decreased with increasing temperature, except for 32°C, in all tissue types. Larval survival was lowest at 32°C in bovine minced meat, bovine tongue, and chicken heart, and at 32°C, all pupae failed to reach the adult stage in minced meat. The results of this study highlight the necessity of considering larval diet and temperature in forensic investigations.

The impact of maceration on the ‘Osteo-ome’; a pilot investigation

The bone proteome, i.e., the ‘osteo-ome’, is a rich source of information for forensic studies. There have been advances in the study of biomolecule biomarkers for age-at-death (AAD) and post-mortem interval (PMI) estimations, by looking at changes in protein abundance and post-translational modifications (PTMs) at the peptide level. However, the extent to which other post-mortem factors alter the proteome, including ‘maceration’ procedures adopted in human taphonomy facilities (HTFs) to clean bones for osteological collections, is poorly understood. This pilot study aimed to characterise the impact of these ‘cleaning’ methods for de-fleshing skeletons on bone biomolecules, and therefore, what further impact this may have on putative biomarkers in future investigations. Three specific maceration procedures, varying in submersion time (one week or two days) and water temperature (55 °C or 87 °C) were conducted on six bovid tibiae from three individual bovines; the proteome of fresh and macerated bones of each individual was compared. The maceration at 87 °C for two days had the greatest proteomic impact, decreasing protein relative abundances and inducing specific PTMs. Overall, these results suggest that routinely-employed maceration procedures are harsh, variable and potentially threaten the viability of discovering new forensic biomarkers in macerated skeletal remains. SignificanceFor the first time, the application of bone proteomics in understanding maceration procedures was conducted to help address the risks for experimental confounding associated with this post-mortem cleaning technique. This pilot study demonstrates that recent advances in biomarker discovery for post-mortem interval and age-at-death estimation using bone proteomics has potential for confounding by differing and destructive bone-cleaning methods.

Advances in artificial intelligence-based microbiome for PMI estimation.

Postmortem interval (PMI) estimation has always been a major challenge in forensic science. Conventional methods for predicting PMI are based on postmortem phenomena, metabolite or biochemical changes, and insect succession. Because postmortem microbial succession follows a certain temporal regularity, the microbiome has been shown to be a potentially effective tool for PMI estimation in the last decade. Recently, artificial intelligence (AI) technologies shed new lights on forensic medicine through analyzing big data, establishing prediction models, assisting in decision-making, etc. With the application of next-generation sequencing (NGS) and AI techniques, it is possible for forensic practitioners to improve the dataset of microbial communities and obtain detailed information on the inventory of specific ecosystems, quantifications of community diversity, descriptions of their ecological function, and even their application in legal medicine. This review describes the postmortem succession of the microbiome in cadavers and their surroundings, and summarizes the application, advantages, problems, and future strategies of AI-based microbiome analysis for PMI estimation.

Not by the Book: Observations of Delayed Oviposition and Re-Colonization of Human Remains by Blow Flies.

We demonstrate one case of an exaggerated delay in blow fly colonization and two cases of blow fly re-colonization after cessation of the consumption phase on human remains at the Anthropology Research Facility (University of Tennessee). We recommend interdisciplinary collaboration to better interpret evidence encountered from scenarios comparable to these examples.Postmortem interval estimations can be complicated by the inter-individual variation present in human decomposition. Forensic entomologists may especially face challenges interpreting arthropod evidence in scenarios that are not “by the book”, or that vary in unexpected ways. Therefore, it is important to report instances where blow fly colonization does not align with expected soft tissue decomposition as blow fly larvae are often used to produce a time of colonization (TOC) estimation to infer a minimum PMI. We followed the decomposition and blow fly activity of three human donors at the Anthropology Research Facility (University of Tennessee). Delayed oviposition occurred on one donor 115 d post-placement, whereas two donors experienced blow fly re-colonization after cessation of the consumption phase, one 22 d and one more than 200 d after blow fly larvae were last observed. A null hypothesis model tested whether the entomological TOC and anthropological total body score (TBS) estimations encompassed the time of placement (TOP) for each donor. While the null hypothesis was rejected for all TOC estimations, it could not be rejected for the TBS estimations. We discuss how the non-linear nature of human decomposition can pose challenges to interpreting blow fly evidence and suggest that forensic entomology practitioners should recognize these limitations in both research endeavors and applied casework.

Carrion flies (Insecta: Diptera) found on human cadavers in Chiba prefecture, Honshu, Japan, with the first record of Fannia prisca from a human corpse.

Forensic entomology (FE) involves the collection and analysis of necrophagous insects and arthropods for postmortem interval estimation based on their growth and succession. In Japan, research on FE is limited, and the predominant species of necrophagous insects and their distribution have not been clarified. The present study aimed to clarify the actual situation of insects, targeting Diptera collected from human cadavers, in Chiba Prefecture. We targeted the cases for which specimens could be collected from forensic autopsies conducted at the Legal Medicine Department of Chiba University Graduate School of Medicine or from human bodies handled by the Chiba Police in 2019. We specifically chose cases from which adult insects could be bred. Fifty cases were applicable, and 47 cases were successful in rearing adult flies. Sixteen species of Diptera were identified: six species of Calliphoridae, four species of Sarcophagidae, and a few species of Fanniidae, Muscidae, Phoridae, Piophilidae, and Stratiomyidae. The most frequently observed species were Lucilia sericata (20 cases) and Chrysomya pinguis (18 cases). For the first time, globally, we recorded the presence of Fannia prisca in a human cadaver. In conclusion, this is the first report to reveal the presence of carrion flies in human death cases in Chiba Prefecture, Japan. Research in FE is important in the context of the Japanese natural and social environment since this can aid forensic investigations conducted by authorities.

New Trends in Immunohistochemical Methods to Estimate the Time since Death: A Review.

The identification of a reliable and accurate post-mortem interval (PMI) is a major challenge in the field of forensic sciences and criminal investigation. Several laboratory techniques have recently been developed that offer a better contribution to the estimation of PMI, in addition to the traditional physical or physico-chemical (body cooling, lividity, radiocarbon dating, rigor mortis), chemical (autolysis), microbiological (putrefaction), entomological, as well as botanical parameters. Molecular biology (degradation pattern of macromolecules such as proteins, DNA, RNA), biochemical analysis of biological fluids (such as blood, cerebrospinal fluid, and vitreous humor), and immunohistochemistry are some of the most recent technological innovations. A systematic review of the literature was performed with the aim of presenting an up-to-date overview on the correlation between the immunohistochemical (IHC) expression of specific antigenic markers at different PMIs. The systematic review was performed according to PRISMA guidelines. Scopus and PubMed were used as search engines from January 1, 1998 to March 1, 2022 to evaluate the effectiveness of immunohistochemistry in estimating PMI. The following keywords were used: (immunohistochemical) OR (immunohistochemistry) AND (time since death) OR (post-mortem interval) OR (PMI). A total of 6571 articles were collected. Ultimately, 16 studies were included in this review. The results of this systematic review highlighted that IHC techniques, in association with traditional methods, add, in Bayesian terms, additional information to define a more accurate time of death and PMI. However, current IHC results are numerically limited and more data and studies are desirable in the near future.

Impact of using Sarcophaga (Liopygia) argyrostoma (Robineau-Desvoidy, 1830) as a toxicological sample in detecting clonazepam for forensic investigation

BackgroundSarcophagidae along with Calliphoridae (superfamily Oestroidea) were known as important colonizers of cadavers and could be used to estimate the postmortem interval (PMI), through tracking the flies colonizing pattern and/or monitoring their growth rates on cadavers. Many previous researches discussed the impact of toxins in decomposing corpses, on the developmental stages of insects, which would affect the accuracy of PMI estimation. Clonazepam belongs to the benzodiazepines, and it is one of the most routinely used drugs to control humans’ seizures. The current study aimed to investigate the effect of clonazepam on the developmental stages of Sarcophaga argyrostoma, one of the most widespread Sarcophagidae in Giza Governorate. Also, we investigate the ability of these developmental stages to detect the drug.ResultsThe current study used different concentrations of clonazepam (25, 50, and 100 mg/ml) as an in vitro application of the flesh fly S. argyrostoma. The drug has affected significantly the morphological measurements (weight, length, and width) of the different developmental stages, especially in the highest concentration (100 mg/ml). Quantitative and qualitative analyses were applied, by using high-performance liquid chromatography-mass spectrometry (HPLC–MS) and protein carbonyls amount respectively, to detect clonazepam in the second, early third, and late third larval instars and prepupa of S. argyrostoma. The results showed that the relationship between the concentration of the drug and its detection in the same developing instar is interdependent, using HPLC–MS. However, the drug faded from instar to the following one and transformed to its metabolite form. Measuring the protein carbonyls amount (OD/mg protein/min) revealed an elevation in the macromolecules damage, compared to the control groups, in almost all treated groups.ConclusionsThe current data suggested that clonazepam has oxidative damage in S. argyrostoma. While HPLC–MS was efficient in measuring the concentration of the drug in the insect, protein carbonyls analysis was a time- and cost-saving method and could be used to detect the drug in insects qualitatively.Graphical abstract

Drowning versus postmortem submersion: Biochemical and histopathological examination and estimation of postmortem intervals (an experimental study)

Estimation of the postmortem interval (PMI) is a very essential task for forensic experts especially in criminal cases. drowning is still the most difficult tasks for forensic pathologists to be diagnosed and differentiated from a body disposed of in water following death especially after long post-mortem period. The objective of this study was to estimate the PMI in drowning in comparison to postmortem submersion using mRNA expression of AQP1 in addition to histological and immunohistochemical examination of caspase-3 in the adrenal and thyroid glands of adult albino rats. Adult albino rats were divided into 2 groups; group A (drowning) and group B (post-mortem submersion). Thyroid and adrenal glands were examined at different PMI. The results revealed significant differences at AQP1 levels between drowning and postmortem submersion at different PMI. Histological changes and caspase-3 expression immunologically in both glands were helpful.

Comparative analysis of fetal pig decomposition processes in burials of variable depths and wrapping.

This research examined the effects that the variables of burial depth and presence of plastic wrapping had on the decomposition rate of fetal pig (Sus scrofa) remains in a New England environment. The decomposition of 56 fetal pigs was observed in four independent variable groups: 20 cm depth unwrapped, 20 cm wrapped, 60 cm unwrapped, and 60 cm wrapped, with exhumation at months 1, 2, 3, 6, 9, 12, and 18. The authors hypothesized that the rate of decay would be slower for wrapped remains and/or for remains at a greater burial depth. Analysis of these remains consisted of preburial and postburial mass, adipocere coverage, skeletal exposure, and decomposition quantified as Total Body Score (TBS). The difference between preburial and postburial mass was reported as a loss percentage to account for varying preburial masses. Wrapping was a significant influencer of mass loss percentage, with p=0.0298 but not for the TBS, with p=0.17565. Burial depth did not have a significant effect on either mass loss percentage or TBS, with p=0.1956 and 0.08969, respectively. This study suggests that wrapping has a greater influence on decomposition patterns than burial depth in this environment, particularly the mass loss percentage. It is suggested that there are limitations with the use of TBS in Postmortem Interval (PMI) estimation, such as variable burial conditions and body characteristics.

Bacterial Succession in Microbial Biofilm as a Potential Indicator for Postmortem Submersion Interval Estimation.

Bacteria acts as the main decomposer during the process of biodegradation by microbial communities in the ecosystem. Numerous studies have revealed the bacterial succession patterns during carcass decomposition in the terrestrial setting. The machine learning algorithm-generated models based on such temporal succession patterns have been developed for the postmortem interval (PMI) estimation. However, the bacterial succession that occurs on decomposing carcasses in the aquatic environment is poorly understood. In the forensic practice, the postmortem submersion interval (PMSI), which approximately equals to the PMI in most of the common drowning cases, has long been problematic to determine. In the present study, bacterial successions in the epinecrotic biofilm samples collected from the decomposing swine cadavers submerged in water were analyzed by sequencing the variable region 4 (V4) of 16S rDNA. The succession patterns between the repeated experimental settings were repeatable. Using the machine learning algorithm for establishing random forest (RF) models, the microbial community succession patterns in the epinecrotic biofilm samples taken during the 56-day winter trial and 21-day summer trial were determined to be used as the PMSI predictors with the mean absolute error (MAE) of 17.87 ± 2.48 ADD (≈1.3 day) and 20.59 ± 4.89 ADD (≈0.7 day), respectively. Significant differences were observed between the seasons and between the substrates. The data presented in this research suggested that the influences of the environmental factors and the aquatic bacterioplankton on succession patterns of the biofilm bacteria were of great significance. The related mechanisms of such influence need to be further studied and clarified in depth to consider epinecrotic biofilm as a reliable predictor in the forensic investigations.

Entomological identification of the post-mortem colonization of wolf cadavers in different decomposition stages

Human-driven biodiversity loss is progressively becoming a problem with dramatic consequences for the conservation of vital ecosystems. The increasing number of illegal killings of the grey wolf (Canis lupus, Linnaeus, 1758), a threatened species, displays the need for investigation and prosecution of such offences. Forensic entomology makes use of the knowledge about necrophagous insects to estimate a minimum time-since-death interval of the deceased person or animal, which can give important information on a possible perpetrator. The cadaver fauna along five decomposition stages of wolves in Germany was investigated in the period 2014–2021. The insects from 70 wolf cadavers, originating from all over Germany, were provided by the Leibniz Institute for Zoo and Wildlife Research Berlin. The accumulated degree day (ADD) model was applied for the post-mortem interval estimation on wolf cadavers for the first time. A total of 20 coleopteran species and 14 different dipteran species were discovered and identified. Almost 99 % of all insect specimens were from the order of Diptera, and beetles (Coleoptera) accounted for only 1 % of the cadaver fauna. The blowflies (Calliphoridae) are of particular importance for forensic issues, accounting for about 66 % of all families. Carrion beetles (Silphidae) were found as the second most abundant family (about 21 %). In addition, combining all cases, a steadily increasing insect species richness S was detected from early decay to advanced decay (fresh S = 8; bloated S = 12; active decay S = 21; advanced decay S = 34). In the following remains stage, the species number decreased again (S = 24). However, no significant difference in the number of species was found between the stages of decay when the cases were considered individually. The temporal pattern of insect appearance was found to be congruent with those of previous studies. Furthermore, a time of death was determined for each case and compared to the pathologist's estimates. This study provides insights into the arthropod fauna of wolf remains for the first time, applies the ADD-Model for post-mortem interval estimation, and discusses the suitability of forensic entomology for wildlife death investigations.

Post-Mortem Interval of Human Skeletal Remains Estimated with Handheld NIR Spectrometry.

Based on a deep-learning technique for post-mortem interval approach (ranging from 1 day up to 2000 years) an estimation of post-mortem interval with an accuracy of almost 100% in bones is possible.Estimating the post-mortem interval (PMI) of human skeletal remains is a critical issue of forensic analysis, with important limitations such as sample preparation and practicability. In this work, NIR spectroscopy (NIRONE® Sensor X; Spectral Engines, 61449, Germany) was applied to estimate the PMI of 104 human bone samples between 1 day and 2000 years. Reflectance data were repeatedly collected from eight independent spectrometers between 1950 and 1550 nm with a spectral resolution of 14 nm and a step size of 2 nm, each from the external and internal bone. An Artificial Neural Network was used to analyze the 66,560 distinct diagnostic spectra, and clearly distinguished between forensic and archaeological bone material: the classification accuracies for PMIs of 0–2 weeks, 2 weeks–6 months, 6 months–1 year, 1 year–10 years, and >100 years were 0.90, 0.94, 0.94, 0.93, and 1.00, respectively. PMI of archaeological bones could be determined with an accuracy of 100%, demonstrating the adequate predictive performance of the model. Applying a handheld NIR spectrometer to estimate the PMI of human skeletal remains is rapid and extends the repertoire of forensic analyses as a distinct, novel approach.

Regression Models for Postmortem Interval Estimation by using Accumulated Degree Hours and Potassium in Vitreous and Blood

The biochemical changes of corpse are using forthe postmortem interval (PMI) estimation, but decomposition isvaried due to many factors. Ambient temperature is believed to be the most important factor which is related tobiochemical change in the human corpse by using accumulated degree hours (ADH). The aim of this researchwas to develop exponential formula to estimate the PMI based on a potassium and ADH as the parameters. Thesecondary data analysis is conducted on 3 articles published in Thailand. The correlation data were analyzed bypotassium, ADH, and time of death in each experiment. The temperature data was provided by the ThailandMeteorology Department (TMD). These results suggested that this developed method can use for PMI estimation.The additional step, namely to calculate the ADH from the finding date back until the predicted day of death fromcase reported and TMD. The regression analysis had a high reliability. The potassium level significantly estimatesthe PMI (R2 = 0.9183, 0.8296 and 0.7142 for pig vitreous, corpse vitreous and corpse blood, respectively). Whenusing the ADH in pig vitreous, the R2 increased to 0.9931. The developed potassium regression models are apractical method to measure PMI. The PMI can be estimated using this method, but caution is advised in case witha long PMI. It can be used to estimate the mortality time of the fresh-early decomposition stage, up to 48 hours.Due to the longer mortality, there may be a gradual rate of change and samples cannot be collected.
