Phytochemical screening and anti-inflammatory activities of different fractions from Citrullus lanatus leaves: A comprehensive study

Inflammation, a protective immune response, can contribute to maintaining homeostasis but can also cause harm when chronic or excessive. Citrullus lanatus has been suggested to possess antioxidant and anti-inflammatory activities that may aid in preventing and treating oxidative stress and inflammation. The main objective of this study was to evaluate the levels of phytochemicals, as well as the antioxidant and anti-inflammatory activities of Citrullus lanatus leaves extract. The ethyl acetate, hexane, butanol fraction and aqueous methanol extract were subjected to various standard quantitative phytochemical analysis, while the hexane fraction and aqueous methanol extract were subjected to standard in vitro anti-inflammatory assays. Spectrophotometric and gravimetric methods were used in this study. The in vitro anti-denaturation (stabilization) assay was used for detecting a wide range of antioxidant compounds. The ethyl acetate fraction had the highest flavonoid and polyphenol levels, followed by hexane, butanol, and aqueous methanol extract, respectively. Quantitative phytochemical analysis showed the levels of flavonoids, polyphenols, tannins, alkaloids and saponins present in the different fractions of C. lanatus. The human erythrocyte membrane stabilization assay showed that extracts were able to stabilize human red blood cell membrane against hypotonic-induced hemolysis in five different samples. The extract of Citrullus lanatus was found to have the highest antioxidant activity. This study has shown that the leaves of Citrullus lanatus can be used in ethnomedicine for the prevention and treatment of ailments caused by oxidative stress and inflammation-related diseases.

Involvement of TNFα, IL-1β, COX-2 and NO in the anti-inflammatory activity of Tamarix aphylla in Wistar albino rats: an in-vivo and in-vitro study

BackgroundWith the emergence of many side effects from synthetic drugs, there is an urgent need to find a natural alternative to these products. Therefore, our primary aim was to evaluate the anti-inflammatory activity of Tamarix aphylla (TA) and investigate the potential mechanism underlying this action.MethodsInitially, to ensure the safety of the extract and for dose selection, we performed an acute oral toxicity Assay through the oral administration of graded doses up to 4 g\\kg in Wistar rats. then, we used the carrageenan-induced edema model to elucidate the anti-inflammatory activity. Using specific ELISA kits, we measured the levels of TNF-α, IL-1β, COX-2 and NO inside the inflamed paw tissue. Finally, for the in-vitro anti-inflammatory experiment, we used the erythrocyte membrane stability test.ResultsBased on the acute oral toxicity assay, T. aphylla was considered generally safe and three different doses of 100, 200, and 400 mg/kg were chosen for further experiments. Additionally, TA expressed a significant (P < 0.05) anti-inflammatory activity, showing the maximum inhibition percentage at the fifth hour of measurement at 53.47% and 70.06%, at doses of 200 and 400 mg/kg respectively, compared to 63.81% for the standard drug. Similarly, we found that TA effectively reduced the levels of TNF-α and IL-1β at all tested doses (100-200-400 mg/kg) to a greater extent than the standard drug. Moreover, at 400 mg/kg, TA was able to significantly lower the levels of COX-2 and NO inside the inflamed tissue to a level comparable (P < 0.05) with that measured inside the paw tissue of normal rats. Finally, Tamarix aphylla at 100, 200 and 400 mg/kg doses significantly (P < 0.05) inhibited the heat-induced hemolysis of RBCs membrane by 67.78, 74.82 and 82.08%, respectively, compared to 83.89% produced by Aspirin.ConclusionT. aphylla produced a significant (P < 0.05) anti-inflammatory activity compared to the standard drugs either through the reduction of pro-inflammatory mediators or the protection of the lysosomal membrane.

In Vitro Studies on the Effects of Aqueous Extracts of Syzygium Aromaticum Linn. (Cloves) and Zingiber Officinale Roscoe (Ginger) on Erythrocyte Osmotic Fragility in Sickle Cells

Sickle cell disease is a genetically inherited blood disease that is characterized by the production of abnormal erythrocytes, shaped like a sickle. Definite cure for sickle cell disease is yet to be established, therefore, palliative methods such as antioxidant agents which reduce occurrence of intravascular haemolysis are employed. Naturally occurring antioxidants are found in Syzygium aromaticum Linn. (Clove) and Zingiber officinale (ginger) as documented by previous researches are therefore tested on the erythrocyte osmotic fragility of sickle cell red blood cells (RBC). Aqueous extracts of clove and ginger were obtained by maceration and antioxidant activity of the extracts was determined by 2, 2,-diphenyl-1-picryl-hydrazyl (DPPH) using ascorbate as standard. Blood samples from twenty five sickle cell disease subjects were collected and divided into five groups. Group one was control, groups two and three were incubated with 1mg/ml and 10mg/ml of clove extract respectively, while groups four and five were incubated with 1mg/ml and 10mg/ml of ginger extract respectively. Erythrocyte osmotic fragility was determined using graded series of hypotonic Sodium Chloride (NaCl) solutions of concentrations ranging from 0.1% to 0.9%. Analysis of Variance (ANOVA) and Turkey test was used to analyze data and test of significance was applied at p?0.05. The results revealed the presence of phenols, alkaloids, flavonoid and tannins in both clove and ginger extracts; glycosides were present in ginger but absent in clove. Antioxidant at low concentration of 50µg/ ml showed percentage inhibition of clove and ginger were 67.43% and 11.87%, respectively, and that of ascorbate was 20.15%. At high concentration of 250µg/ml, clove and ginger had percentage inhibition of 93.8% and 81.18% respectively, while ascorbate had 84.06%. Erythrocyte osmotic fragility test results showed complete haemolysis at 0.1% NaCl concentration in all groups. At 0.2% NaCl concentration, percentage haemolysis of the group treated with 10mg of clove 69.12 ± 0.05, was significantly lower (p?0.05) than the control (98.12 ± 0.05), while the group treated with 10mg ginger extract showed 98.02 ± 0.04 which is not significantly lower (p?0.05) than that of the control. At 0.5% NaCl concentration, percentage haemolysis of groups treated with 10mg clove extract and 10mg ginger extract were 8.400.18 and 20.42 ± 0.18, respectively, which were significantly lower (p?0.05) than that of the control group (45.90 ± 0.10). The groups treated with 1mg clove extract and 1mg ginger extract showed percentage haemolysis values of 38.04 ± 0.10 and 30.02 ± 0.10 respectively at 0.5% NaCl concentration which were significantly lower (p?0.05) than the control group (45.90 ± 0.10). Clove showed higher antioxidant activity than ginger. Both plants also demonstrated a capacity for maintaining erythrocyte membrane stability, with clove being better at lowering erythrocyte fragility than ginger.

Recent Progress on Genetically Modified Animal Models for Membrane Skeletal Proteins: The 4.1 and MPP Families.

The protein 4.1 and membrane palmitoylated protein (MPP) families were originally found as components in the erythrocyte membrane skeletal protein complex, which helps maintain the stability of erythrocyte membranes by linking intramembranous proteins and meshwork structures composed of actin and spectrin under the membranes. Recently, it has been recognized that cells and tissues ubiquitously use this membrane skeletal system. Various intramembranous proteins, including adhesion molecules, ion channels, and receptors, have been shown to interact with the 4.1 and MPP families, regulating cellular and tissue dynamics by binding to intracellular signal transduction proteins. In this review, we focus on our previous studies regarding genetically modified animal models, especially on 4.1G, MPP6, and MPP2, to describe their functional roles in the peripheral nervous system, the central nervous system, the testis, and bone formation. As the membrane skeletal proteins are located at sites that receive signals from outside the cell and transduce signals inside the cell, it is necessary to elucidate their molecular interrelationships, which may broaden the understanding of cell and tissue functions.

Bioactivity profiling of native and hybrid varieties of pumpkin peel (Cucurbita maxima Linn.)

The peel part of the two varieties (native and hybrid) of pumpkin are nutritionally enrich, so to make it economically viable it could be an important component in food and pharmaceutical industries. For this purpose, this study focus on to assess the various biological activities of two varieties of pumpkin peel. To evaluate the phytochemical potentiality of the peel of two varieties of pumpkin as a wide-ranging bioactivity profiling, in vitro anti-hyperglycemic activity, anti-inflammatory activity, anti-hemolytic activity and in vitro antioxidant activity along with total phenolic and flavonoid contents were evaluated. The peel of the native variety showed more phenolic contents than that of hybrid one and the values of the phenolic contents of the native and hybrid varieties were 74.76 ± 1.7 and 68.76 ± 0.87 mg GAEs/g respectively. In contrast, the peel of the hybrid variety showed more flavonoid content compared to native one and the values of the flavonoid contents of the native and hybrid varieties were 34.11 ± 0.13 and3 6.66 ± 1.29 mg QEs/g respectively. The two varieties exhibited concentration-dependent moderate antioxidant activity along with intermediate inhibitory activity against two enzymes. These activities were in order: native peel > hybrid peel. In the case of in vitro anti-hemolytic activity, both extracts showed intermediate erythrocyte membrane stabilization activities in order: hybrid peel > native peel. This research presented some supporting evidence for analyzing the comparative biological activities, and the findings could be helpful in enhancing a better understanding of these varieties as well as other pumpkin species. Finally, in this study, the bioactive compounds in the pumpkin peel may validate their enormous potential as a source of bio-based preservatives, an emergent component of functional food in food industries and in pharmaceutical industries as therapeutic agents that may enhance consumers’ health and promote a circular economy.

Pentaclethra eetveldeana Leaves from Four Congo-Brazzaville Regions: Antioxidant Capacity, Anti-Inflammatory Activity and Proportional Accumulation of Phytochemicals.

Oxidative stress and inflammation play a key role in the occurrence of neurodegenerative diseases. Traditionally, Pentaclethra eetveldeana leaves are used in dementia treatment. Therefore, this study aimed to evaluate the antioxidant and anti-inflammatory activities as well as the phytochemical composition of Pentaclethra eetveldeana leaves from four Congo-Brazzaville regions. The 1.2-diphenyl-1-picrylhydrazyl radical-scavenging, β-carotene bleaching and molybdenum reduction assays were used to assess the antioxidant activity. The protein denaturation and erythrocyte membrane stabilization tests were used to analyze the anti-inflammatory activity. Phytochemical screening, the quantification of polyphenols by spectrophotometry, as well as the determination of extraction yields were carried out. It was found that the extracts reduced molybdenum; furthermore, compared to ascorbic acid, they showed better antiradical activity and inhibited lipid peroxidation. Moreover, globally, the membrane-stabilizing power of the aqueous extracts was superior or comparable to diclofenac, while the same extracts were less effective for the inhibition of denaturation. All of the aqueous extracts contained polyphenols, saponins, alkaloids, anthraquinones, reducing sugar and cardiotonic glycosides. The total polyphenols, tannins and proanthocyanidins are produced proportionally from one region to another. Finally, the leaves from Brazzaville and Boundji contain flavonols, while those from Makoua and Owando contain flavones. Thus, Pentaclethra eetveldeana leaves contribute to traditional dementia treatment through their antioxidant and anti-inflammatory properties.

Looking beyond Arthrospira: Comparison of antioxidant and anti-inflammatory properties of ten cyanobacteria strains

Cyanobacteria are a widely distributed group of photosynthetic prokaryotes with potential to produce biologically active compounds. Arthrospira is the most widespread grown cyanobacteria for commercial purposes; however, many other species could be used as sources of a wide variety of bio-products. The pigment phycocyanin explains part of the therapeutic properties attributed to Arthrospira (e.g. antioxidant and anti-inflammatory). Many chronic diseases include, on its etiology, oxidative stress and abnormalities in the inflammatory response; thus, there is great interest in finding new sources of natural antioxidants and anti-inflammatory compounds. The aim of this work was to evaluate growth attributes, chemical composition and antioxidant and anti-inflammatory properties of ten cyanobacteria isolates. After 15 days of culture, biomass production ranged between 0.06 g/L (Anabaena) and 0.27 g/L (Phormidium) in 20 L volume cultures. Protein content of dry biomass ranged between 28.86 % (Nodularia) and 56.39 % (Arthrospira) while phycocyanin content ranged between 0.86 % (Nodularia) and 6.66 % (Spirulina). Aqueous extracts were evaluated regarding total phenolic content (TPC), total flavonoids content (TFC) and antioxidant activities (DPPH, ABTS and FRAP). Spirulina and Desertifilum exhibited the highest phenols and flavonoids contents; as well as the highest ABTS and FRAP antioxidant activities. Five strains were selected for measuring anti-inflammatory activity by the human erythrocyte membrane stabilization assay. All the strains evaluated exhibited a remarkable anti-inflammatory capacity, being the highest value for Desertifilum. Strains of Spirulina and Desertifilum were also tested for their ability to scavenge reactive oxygen species (ROS) on Hek293 cells; both strains showed significant intracellular antioxidant capacity and were not cytotoxic. This research demonstrated that other cyanobacterial strains, besides the popular Arthrospira, might be new sources of antioxidant and anti-inflammatory compounds for biotechnological applications in functional food, nutraceutical or pharmaceutical industries.

Postnatal Effects of the Ethanol on the Encephalon Mass and Neocortex Architecture in Wistar Rat Offspring Submitted to Acute Prenatal Injection [E12] and in vitro Ethanol Effects on Wistar Rat and Human Erythrocytes

Aims: To verify the ethanol effects of the acute injections in pregnant Wistar rats (E12) on the offspring in P8. Additionally, measures of the brain and corporal masses and the stability of erythrocytes were performed in rats and humans.&#x0D; Study Design: Histological analysis and a spectrophotometer were used to count neural cells and osmotic fragility in erythrocytes.&#x0D; Place and Duration of Study: The immunohistochemistry study was partially performed at the Federal University of Uberlândia, and at the University of Toyama-Japan, the experiments on the brain, body masses, and stability of erythrocyte membranes in humans were performed from 2012 to 2014.&#x0D; Methodology: Eighteen pregnant female rats (180-230g) were housed in cages at 22 ± 0.4°C on a 12 h light/dark cycle with free access to food and water. On the 11th day of pregnancy (E12), 12 rats received three intraperitoneal (i.p.) injections of a 20% ethanol solution (3 g of ethanol/kg of body weight) at 8 hours intervals.&#x0D; Results: The number of neural cells was lower in rats treated with ethanol than to controls to each lobe and total count in all lobes (P = 0.001). Significant alterations of neocortical tissue in ethanol-treated were observed, as the decrease of brain mass (P = 0.05) in linear regression. The stability of erythrocyte membranes was verified with D50 equal to 0.46 g·dL-1 (± 0.05).&#x0D; Conclusion: According to this work, the ethanol injection in the rats showed marked brain tissue destruction relative to the control, and ethanol’s effects on erythrocytes indicated that membrane destruction could be one of the causes of brain cell disruption in neural migration.

Correlation between sphingomyelin and the membrane stability of mammalian erythrocytes.

Lipid compositions of mammalian erythrocyte membranes are different among species. Therefore, the information on hemolysis from mammalian erythrocytes is useful to understand membrane properties of human erythrocytes. In this work, pressure-induced hemolysis and hypotonic one were examined using erythrocytes of human, sheep, cow, cat, dog, pig, horse, rat, and mouse. Pressure-induced hemolysis was suppressed by membrane sphingomyelin, whereas hypotonic hemolysis decreased upon increment of cell diameter. Mass spectra of erythrocyte membrane lipids demonstrated that sphingomyelin with an acyl chain 24:1 was associated with the suppression of pressure-induced hemolysis. In cow erythrocytes, pressure-induced hemolysis was greatly suppressed and the detachment of cytoskeletal proteins from the membrane under hypotonic conditions was also inhibited. Taken together, these results suggest that sphingomyelin with 24:1 fatty acid plays an important role in the stability of the erythrocyte membrane, perhaps via cholesterol.

Correlations between health indicators in women who survived breast cancer

The increase in breast cancer survivors (BCS) has established unprecedented demands on health systems worldwide. In this context, new knowledge is needed to subsidize strategies to handle this population. The objective of this study was to combine the correlations between anthropometric, hematological, biochemical, biophysicochemical, muscle strength, and age variables in BCS. Twenty-two volunteers (51.7 ± 9.2 years old) with a history of mastectomy and lymphadenectomy participated in the study. Among the results, the correlations of the body mass index with lipid profile, strength, and body composition draw special attention. The associations of lipid profile with erythrocyte membrane stability, body composition, and leukocyte count, the associations of the red cell distribution width (RDW) with body mass index, lipid profile, and strength, and the positive correlation between age and leukocyte count also deserve highlights. Such findings allow concluding that investigations that deal singly with health indicators represent a severe limitation in the study of the public in question, and it is necessary to avoid them. The complex network of interactions observed between the various parameters studied reinforces the importance of a global biological analysis of attractive cost-benefit and great practical potential. Teams with medical, nutrition, physical therapy, and physical education professionals, among other areas, can use this knowledge to improve BCS's interdisciplinary approaches in short, medium, and long-term management.

The effect of the Satureja montana ethanol extract on the morphological changes of erythrocytes

The present study investigated the antioxidant activity of ethanol extracts of the leaves of Satureja montana and their influence on the membrane stability of erythrocytes ex vivo. The ethanol extracts showed a very potent antioxidant activity of EC50 = 0.055mg/ml. Rat blood samples were treated with 96% ethanol extracts in different concentrations of 100 ?g/ml, 200 ?g/ml, and 300 ?g/ml, and morphological analyses were carried out. The results showed significant differences in the shape of the erythrocytes incubated with higher extract concentrations. Considerable morphological changes were observed at a concentration of 200 ?g/ml which was characterised by the highest percentage of stomatocytes, while the highest percentage of echinocyte formation was observed at a concentration of 300 ?g/ml. The results of this investigation indicated that the ethanol extracts of S. montana exhibited a possible protective effect on the membrane stability of erythrocytes.

SYNTHESIS AND PRECLINICAL STUDY OF ANTIOXIDANT ACTIVITY OF [1,2,4]TRIAZOLO[1,5-A]PYRIDINE DERIVATIVES

Purpose. Synthesis and study of antioxidant properties of [1,2,4]triazolo[1,5-a]pyridine derivatives and their potential toxicity using preclinical diagnostic methods. Methodology. Derivatives of [1,2,4]triazolo[1,5-a]pyridine were synthesized on the basis of 2-aminopyridines and ethyl isothiacyanatocarbonate, with subsequent cyclization of intermediate adducts. The resulting 2-amino[1,2,4]triazolo[1,5-a]pyridines were reacted with isoamyl nitrite in the presence of CuHal2, which led to the synthesis of volatile 2-halogeno[1,2,4]triazolo[1,5- a]pyridine. The toxicity of the obtained compounds was studied by the degree of hemolysis of erythrocytes in a hypotonic NaCl solution. The intensity of free-radical oxidation of lipids was assessed spectrophotometrically by the formation of TBА-active products. The antioxidant activity of the substances was determined by inhibiting the oxidation of adrenaline under conditions of artificial oxidative stress. Scientific novelty. For the first time, the antioxidant activity of the [1,2,4]triazolo[1,5-a]pyridine derivatives described in the paper was investigated and their toxicity was studied according to the degree of hemolysis of erythrocytes in a hypotonic NaCl solution. Conclusions. The in vitro cytotoxicity study model showed that compounds 5a and 5b, which contain bromine atoms in the 2, 5, and 7 positions of the pyridinotriazole heterosystem, exert the greatest influence on the stability of erythrocyte membranes. The study of antioxidant activity showed that the highest values of antioxidant activity are characteristic of compounds of the 4a-d series, namely compounds 4b and 4c, which are characterized by the presence of an amino group in the 2nd position and the presence of a bromine atom as a substitute in the 1st and 5th positions of the pyridine fragment of the heterosystem. The dynamics of antioxidant activity on the model of inhibition of adrenaline oxidation confirms the opinion about the lowest toxicity and the highest ability to inhibit the formation of reactive oxygen species of compounds 4b and 4c (40.16 and 38.82% relative to ionol). So, with the use of preclinical diagnostic methods, we proved the presence of physicochemical activity in a number of investigated compounds. 2-amino-[1,2,4]triazolo[1,5a]pyridine derivatives with a bromine atom as a substitute in the pyridine fragment of the molecule should be considered as potential antioxidants in the future, and the studied condensed system can be considered a promising heterocyclic framework for creating potential antioxidant agents.

In vitro effects of 2-methyl-3-propylbutane-1,4-diol purified from Alstonia boonei on erythrocyte membrane stabilization and mitochondrial membrane permeabilization.

A recent review on the ethnomedicinal, chemical, pharmacological, and toxicological properties of Alstonia boonei revealed the plant's potential in the treatment and management of a range of diseases. However, most of these pharmacological effects are only traceable to the crude form of the plant extract and not specific natural products. Phytochemical investigation of the methanol fraction of the methanol extract of the stem-bark of Alstonia boonei led to the isolation and identification of 2-methyl-3-propylbutane-1,4-diol. The structures were elucidated by the application of 1D-, and 2D-NMR spectroscopic analyses and by comparison with literature data. In this study, the membrane stabilizing activity, mitochondrial membrane permeability transition pore opening, cytochrome c release, mitochondrial ATPase activity, and prevention of mitochondrial lipid peroxidation activity of 2-methyl-3-propylbutane-1,4-diol (MPBD) isolated from A. boonei were determined. The results showed that MPBD significantly (p < .05) prevented peroxidation of mitochondrial membrane lipids and hemolysis using both the heat-induced and hypotonic solution-induced membrane stabilization assays. On the contrary, the compound caused large amplitude swelling of rat liver mitochondria in the absence of calcium, significant (p < .05) cytochrome c release and enhancement of mitochondrial ATPase activity in vitro. Our findings suggest that MPBD showed characteristic biological properties useful in modulating cell death.

IN VITRO AND IN VIVO SCREENING OF ANTI-INFLAMMATORY ACTIVITY OF METHANOLIC AND AQUEOUS EXTRACTS OF ANOGEISSUS LATIFOLIA LEAVES

Objective: To evaluate and compare anti-oxidant and anti-inflammatory activity of methanolic and aqueous extract of Anogeissus latifolia leaves Methods: The in vitro antioxidant activity was investigated using nitric oxide radical inhibition activity assay, hydroxyl radical scavenging activity assay, DPPH free radical scavenging assay, and reducing power assay. The in vitro anti-inflammatory activity was investigated using erythrocyte membrane stabilization, inhibition of protein denaturation, and proteinase inhibitory activity the in vivo anti-inflammatory activity was investigated using carrageenan-induced rat paw edema. The biochemical parameters were evaluated in the blood, which included the determination of serum glutamic oxaloacetic transaminase and serum glutamic pyruvic transaminase values and in the liver, which includes the estimation of lipid peroxidation, reduced glutathione, and superoxide dismutase. Results: The methanolic extract caused a significant dose depended on the reduction of inflammation when compared with the aqueous extract of Anogeissus latifolia. The anti-inflammatory activity of all groups was found to be comparable to the standard indomethacin group. The maximum percent inhibition in paw edema was found in methanolic extract of Anogeisuss latifolia at a dose of 500 mg/kg was 53.33%, with significant anti-inflammatory activity p&lt;0.001. Conclusion: The leaf extract of Anogeissus latifolia possesses anti-oxidant and anti-inflammatory activity. The therapeutic effect of Anogeisuss latifolia extracts will encourage its use in the treatment of inflammation.

Wearable Antennas for Sensor Networks and IoT Applications: Evaluation of SAR and Biological Effects.

In recent years, there has been a rapid development in the wearable industry. The growing number of wearables has led to the demand for new lightweight, flexible wearable antennas. In order to be applicable in IoT wearable devices, the antennas must meet certain electrical, mechanical, manufacturing, and safety requirements (e.g., specific absorption rate (SAR) below worldwide limits). However, the assessment of SAR does not provide information on the mechanisms of interaction between low-intensity electromagnetic fields emitted by wearable antennas and the human body. In this paper, we presented a detailed investigation of the SAR induced in erythrocyte suspensions from a fully textile wearable antenna at realistic (net input power 6.3 mW) and conservative (net input power 450 mW) conditions at 2.41 GHz, as well as results from in vitro experiments on the stability of human erythrocyte membranes at both exposure conditions. The detailed investigation showed that the 1 g average SARs were 0.5758 W/kg and 41.13 W/kg, respectively. Results from the in vitro experiments demonstrated that the short-term (20 min) irradiation of erythrocyte membranes in the reactive near-field of the wearable antenna at 6.3 mW input power had a stabilizing effect. Long-term exposure (120 min) had a destabilizing effect on the erythrocyte membrane.

Epicatechin exerts dual action to shield sickling and hydroxyurea-induced myelosuppression: Implication in sickle cell anemia management

Hydroxyurea (HU) is the key drug to treat Sickle cell anemia (SCA). However, its treatment is associated with the liability of myelosuppression. The present study aimed to investigate the potential of epicatechin as a supplementation therapy for the symptomatic management of SCA under HU therapy. A panel of experiments were performed at first to observe epicatechin's effect on sickling and hemolytic behaviour using SCA patient's blood (ex vivo). Thereafter, the effect of HU in the presence or absence of epicatechin was investigated on cytokine inhibition in rat splenocytes (ex vivo) as well as alterations in hematological parameters and kidney function tests in rats (in vivo). Then, any effect of epicatechin on pharmacokinetic modulation of HU in rats was elucidated along with the underlying mechanism using a battery of in vitro and in vivo models. Epicatechin exhibited potent action on anti-sickling, polymerization inhibition, and erythrocyte membrane stability. It did not show any inherent hemolytic activity and reduced TNF-α level during concomitant administration with HU. Based on hematological changes in rats, epicatechin treatment aided to the beneficial effect of HU and prevented the treatment-linked disadvantageous effects of HU like neutropenia. The plasma exposure of HU was significantly augmented in rats upon simultaneous oral administration of epicatechin with HU. Down-regulation of Oatp1b2 and catalase possibly contributed to the pharmacokinetic interaction of HU. Epicatechin is found to be a promising candidate and should be explored at a reduced dose level of HU towards offsetting the dose-dependent myelosuppressive effect of HU under the frame of supplementation therapy in SCA.

Inhibition of Plasmodium berghei growth by alkaloid extract of Phyllanthus amarus in mice increased haem level and stabilized erythrocyte membrane

Objectives: The Plasmodium parasite is known for its ability to destabilize the red cell membrane causing the release of free haem which is converted to haemozoin as haem is toxic to parasite. In combination with other factors, accumulation of haemozoin is associated with anaemia which manifest in severe malaria. Compounds directed at preventing erythrocyte attack and increasing haem concentrations could serve as new sources of antimalarials. Therefore, this study assessed the ability of Phyllanthus (P.) amarus alkaloid extract, a phytochemical, already reported to possess antimalarial activity, in reducing erythrocyte deformity and preventing parasite multiplication by increasing haem concentrations. Methods: The study involved measurement of parasite clearance, changes in haematological indices and degree of erythrocyte membrane stabilization (using heat-induced and hypotonic-solution induced destabilization assays) in Plasmodium (P.) berghei infected mice treated with alkaloid extract of P. amarus using documented methods. Results: Infection of experimental mice with Plasmodium berghei, induced malaria with evidence of anaemia (reduced PCV and Hb), reduction in haematological indices (RBC, WBC, platelets, lymphocytes, and eosinophil), increased serum haemozoin and haem concentration and destabilization of erythrocyte membrane.

STUDY OF ASPIRIN EFFECT ON STABILITY OF MEMBRANES OF RAT BLOOD ERYTHROCYTES BY ACIDIC HEMOLYSIS METHOD

In this work the effect of aspirin on stability of rat blood erythrocyte membranes was studied, using acidic hemolysis method. Hemolysis rate of erythrocytes was shown to depend on preservation time of rat erythrocyte suspension. With increasing of preservation time the original value of optic density of rat erythrocyte suspension decreases and hemolysis time reduces. It was also shown that in the presence of aspirin the membrane stability of low-stable erythrocytes increases, moreover, hemolysis duration enhances, while aspirin leads to decreasing of membrane stability of both high-stable and enhanced-stable erythrocytes of rats.

A desirability approach for Antidiabetic and Antiinflammatory efficacy of Ficus subincisa bark

Background: This study aimed to examine the anti-inflammatory and antidiabetic effects of the green solvent extract from F.subincisa Buch-Ham.ExSm.This paper also explains how bio-based products have been highly safe and alternative to the use of organic solvent. Methods: The serial extraction was carried out with increasing polarity using a magnetic stirrer: d-limonene, Isopropyl alcohol, Ethyl lactate, and Hydroalcohol.The concentrated and dried extracts were subjected to analysis the antidiabetic activity and anti-inflammatory activity were assessed by employing standard in vitro techniques. Result: In vitro anti-inflammatory assays were performed using heat-induced bovine serum albumin (BSA) denaturation and erythrocyte membrane stabilization assays.α-amylase and α-glucosidase were used to evaluate the anti-diabetic activity of F.subincisa bark in vitro. Diclofenac sodium, Acarbose, and aspirin were used as reference drugs. The in vitro method showed significant anti-inflammatory and anti-diabetic properties of different bio-based extracts tested. Conclusions: Findings from this study showed that F.subincisa exhibited substantial anti-inflammatory and antidiabetic actions in vitro models.
