Natural polymer coated magnetic nanoparticles for biosensing

AbstractSuperparamagnetic iron oxide nanoparticles coated with natural polymers have found many applications in the field of biosensors as magnetic carriers due to the combination of two characteristics: the possibility of controlling the particles movement by applying external magnetic fields and attaching biomolecules to the particle through a chemical bond. The aim of this work was to prepare long-term stabilized particles with acid groups available to be used as magnetic carriers of biomolecules. In this study, we present the synthesis of maghemite nanoparticles coated with alginate, a natural polymer. Particles were characterized by electron microscopy. Magnetic properties were studied by vibrating sample magnetometry, which revealed the superparamagnetic behavior of maghemite nanoparticles. Horseradish peroxidase (HRP) was chemically bonded to the maghemite-alginate particles and the enzymatic activity of HRP was determined by a colorimetric technique. The maghemite-alginate particles were immobilized onto the surface of a gold electrode by means of a magnet and the HRP activity was followed electrochemically, showing that these particles can be successfully used in amperometric sensors. Furthermore, maghemite-alginate particles were also used for the diagnosis of the foot-and-mouth disease by means of an enzyme-linked immunoassay with electrochemical detection. 3ABC protein, a non-structural protein of the virus, was linked to the maghemite-alginate particles and used to selectively detect anti-3ABC antibodies in cattle sera.

A quantitative study of the enzymatic activity of horseradish peroxidase at a planar poly(acrylic acid) brush

The enzymatic activity of horseradish peroxidase (HRP) has been quantified at a planar poly(acrylic acid) (PAA) brush. A PAA brush is known to exhibit an unusual protein binding affinity, since proteins adsorb at low ionic strength only. At elevated ionic strengths of a few 100 mM proteins desorb, and the PAA brush becomes largely protein resistant. In this study, HRP was used to catalyze the oxidation of Amplex Red to resorufin by hydrogen peroxide. The fluorescence of resorufin was recorded using a microplate reader. As compared to a bare silica surface, the enzymatic activity of HRP is higher by more than one order of magnitude at a PAA brush. This increase results from a higher degree of adsorption and a reasonable preservation of the HRP activity. Upon adsorption at a PAA brush from a 20 mU/mL (0.1 μg/mL) solution, the molecular enzymatic activity of HRP is reduced to about 11%. However, when a HRP molecule is desorbed from a PAA brush by increasing the ionic strength, a gain of the molecular enzymatic activity by only 52% can be observed. Overall, this study illustrates the potential applicability of a PAA brush as a biocompatible material coating.

Enzyme activity of horseradish peroxidase immobilized in chitosan matrices in alternated layers

The successful immobilization of enzymes such as horseradish peroxidase (HRP) in solid films is essential for applications in sensors and for fundamental studies aimed at identifying possible biotechnological devices. In this study we show that HRP can be immobilized in alternated layers with chitosan as the template material. The activity of HRP in HRP/chitosan films was preserved for several weeks, and could be detected optically upon monitoring the reaction with pyrogallol. The morphology of the film displayed stripes that disappeared after reaction with pyrogallol. Though the activity in the HRP/chitosan film was lower than in a homogeneous solution or in an LB film investigated earlier, the response was linear for a considerable period of time, which may be advantageous for sensing hydrogen peroxide.

Biocatalysis in Water-in-Ionic Liquid Microemulsions: A Case Study with Horseradish Peroxidase

In this article we report the first results on the enzymatic activity of horseradish peroxidase (HRP) microencapsulated in water-in-ionic liquid (w/IL) microemulsions using pyrogallol as the substrate. Toward this goal, the system used in this study was composed of anionic surfactant AOT (sodium bis(2-ethyl-1-hexyl)sulfosuccinate)/hydrophobic IL [C(8)mim][Tf(2)N] (1-octyl-3-methyl imidazolium bis(trifluoromethylsulfonyl)amide)/water/1-hexanol. In this system, the catalytic activity of HRP was measured as a function of substrate concentrations, W(0) (molar ratio of water to surfactant), pH, and 1-hexanol content. The curve of the activity-W(0) profile was found to be hyperbolic for the new microemulsion. The apparent Michaelis-Menten kinetic parameters (k(cat) and K(m)) were estimated and compared to those obtained from a conventional microemulsion. Apparently, it was found that HRP-catalyzed oxidation of pyrogallol by hydrogen peroxide in IL microemulsuions is much more effective than in a conventional AOT/water/isooctane microemulsion. The stability of HRP solubilized in the newly developed w/IL microemulsions was examined, and it was found that HRP retained almost 70% of its initial activity after incubation at 28 degrees C for 30 h.

Conducting polymer based fluorescence quenching as a new approach to increase the selectivity of immunosensors

Polypyrrole (Ppy) has been shown to be a superior matrix for fluorescence detection based immunosensors: (i) the fluorescence of polypyrrole and polypyrrole modified by entrapped proteins was almost not detectable when this polymer was excited by near UV 325 nm light; (ii) polypyrrole quenched the fluorescence of such fluorescence agents as fluoresceine 5(6)-isothiocyanate, rhodamine B and enzyme–horseradish peroxidase (HRP) by almost 100% if they were deposited in the solution as a drop at the Ppy surface followed by evaporation of the solvent. According to our knowledge, this work is first application of Ppy in the design of a fluorescence-based immunosensor, where low Ppy fluorescence background and Ppy induced fluorescence quenching were exploited. These sensors were devoted to the detection of antibodies against bovine leukemia virus (BLV) protein gp51 (anti- gp51-Ab). A biological recognition system of this fluorescence immunosensor model was based on polypyrrole with entrapped BLV proteins gp51 ( gp51/Ppy). This gp51/Ppy layer was applied for the detection of anti- gp51-Ab. Secondary antibodies against anti- gp51-Ab labeled with HRP (Ab*) were applied as fluorescence-detectable labels that are able to recognize specifically and interact with the complex of gp51 proteins and anti- gp51-Ab antibodies ( gp51/anti- gp51-Ab). It was demonstrated that fluorescence of non-specifically adsorbed Ab* was almost completely quenched by the Ppy substrate. In addition, enzymatic activity of HRP was exploited as a traditional reference method for verification of the formation of the immune complex gp51/anti- gp51-Ab/Ab*.

Probing the change of enzymatic activity of horseradish peroxidase induced by membrane permeation using tryptophan fluorescence

Changes in the molecular structure of horseradish peroxidase (HRP-4C) induced by membrane ultrafiltration, as well as its impact on the enzymatic activity, were monitored using three complementary fluorescence techniques: steady-state fluorescence, fluorescence anisotropy and picosecond time-resolved fluorescence. Ultrafiltration experiments were performed using membranes of polyethersulfone (PES) with 30 and 100 kDa and membranes of regenerated cellulose (RC) with 10 and 30 kDa. The results obtained clearly shows HRP-4C structural changes during ultrafiltration, which were essentially controlled by hydrophobic and electrostatic protein–membrane interactions. The structural changes observed depend on the affinity of the membrane material to ions, such as Fe 3+ and Ca 2+, which have a substantial structural and functional relevance to this enzyme. Additionally, it was found that the impact of the ultrafiltration process on the enzymatic activity was a direct consequence of the depletion of iron from the protein structure, causing irreversible structural alterations on HRP-4C, and thus inducing severe changes in its enzymatic activity.

Encapsulation of Enzymes within Polymer Spheres To Create Optical Nanosensors for Oxidative Stress

We describe the fabrication and characterization of poly(ethylene glycol) (PEG) hydrogel spheres containing the enzyme horseradish peroxidase (HRP) for application as optical nanosensors for hydrogen peroxide. HRP was encapsulated in PEG hydrogel spheres by reverse emulsion photopolymerization, yielding spheres with a size range from 250 to 350 nm. Encapsulated HRP activity and sensitivity to hydrogen peroxide were investigated by the Amplex Red assay based on the fluorescence response as a function of H2O2. These HRP-loaded spheres were then introduced to murine macrophages with Amplex Red in the culture media. After phagocytosis, the biocompatibility of spheres was determined by live cell staining using calcein AM (5 microM). The HRP-loaded PEG hydrogel spheres were activated (i.e., fluorescent oxidized Amplex Red produced within the spheres) by oxidative stresses such as exogenous H2O2 (100 microM) and lipopolysaccharide (1 microg/mL), which induced the production of endogenous peroxide inside macrophages. The results presented here indicate that after polymerization, the enzyme activity of HRP was still maintained and that using these HRP-containing nanospheres, peroxide production could be sensed locally within cells.

Numerically Simulated pH‐Induced Reactivation of Catalytic Activity of Horseradish Peroxidase

A model mechanism that accounts for the experimentally observed pH-dependent enhancement of enzymatic activity of horseradish peroxidase through the catalase-like pathway is proposed. Predictions of the model are tested against a number of experimental results to confirm that kinetic constants used in the numerical simulation are correctly chosen and that the model can be used to emulate the reaction between horseradish peroxidase and hydrogen peroxide in a wide range of conditions.

Single-Molecule Surface-Enhanced Raman and Fluorescence Correlation Spectroscopy of Horseradish Peroxidase

We report on the spectroscopy and enzymatic activity of horseradish peroxidase (HRP) coupled to Ag nanoparticles. We show that vibrational spectra of single HRP molecules can be detected by measuring surface-enhanced Raman scattering (SERS) from isolated and immobilized protein−nanoparticle aggregates. However, the nanoparticle−protein interaction results in decreased enzymatic activity, as shown by an ensemble-averaged HRP activity assay. We demonstrate that H2O2, one of the enzyme substrates in the assay, reacts with the Ag surface and that it is necessary to pretreat the Ag particles with H2O2 to retain full HRP activity in the Ag sol. However, the concomitant changes in the Ag surface adsorbate layer caused by this treatment prevent strong HRP adsorption and therefore also single-molecule SERS sensitivity. Layers of H2O2-treated Ag particles can instead be used to enhance the fluorescence signal from single functional HRP molecules.

Effects of acetonitrile on horseradish peroxidase (HRP)-anti HRP antibody interaction

The effects of the water-miscible organic solvent acetonitrile on the enzymatic activity of horseradish peroxidase (HRP) and on HRP-anti-HRP binding have been investigated. Results showed that both the catalytic activity of HRP and the binding ability of the antibody were affected on increasing the concentration of the organic solvent. The activity of HRP varied with the organic composition of the solvent, indicating that the conformation of the enzyme was affected. The binding ability of the antibody also decreased significantly with an increase of the organic composition of the solvent, and in absolute acetonitrile, the activity of the antibody is about 500 times lower than that in aqueous medium. Binding reversibility experiments indicated that the antibody was not irreversibly damaged in solutions with acetonitrile composition greater than 80% and below 40%; however, an irreversible decrease in the binding was observed in solutions with an acetonitrile composition between 40 and 80%. The reduction in the binding ability is probably due to the irreversible conformation changes in the antibody.

New method of deposition of biomolecules for bioelectronic purposes

A laser induced plasma vaporization and ionization technique is proposed for electric field assisted deposition of proteins. Experiments were carried out depositing thick layers of lysozyme on metal, creating submillimeter patterns. The enzymatic activity of horseradish peroxidase deposited by this method was tested, together with that of the enzyme laccase from a micro-organism. The applicability of this method to the construction of nanometric patterns for bioelectronic purposes is discussed.

This study describes a process by which serine proteases that contain an S-1 arginine subsite and active site histidine may be inactivated and subsequently quantitated using a combination of peptidyl chloromethylketone chemistry and immune recognition technology. Active site labeling and inactivation of proteases is attained by modification of the active site histidine with a peptidyl chloromethylketone. In the specific illustrations demonstrated, we used the compound biotinyl-epsilon-aminocaproyl-phenylalanylprolylarginyl chloromethylketone. This reagent reacts quantitatively and specifically with the active site histidine of a wide variety of proteases that are elaborated in the coagulation and fibrinolytic system. The inactivated enzyme(s) may be quantitated by combinations of antiprotein antibodies and avidin binding technology using the biotin moiety on the peptide inhibitor. We have demonstrated the capability of capture of inactivated enzyme products directly on to solid-phase avidin with subsequent quantitation of bound protein using specific antibodies. In the converse system we have captured specific proteases using antiprotein antibodies in the solid phase and have quantitated bound enzyme by using avidin. Subsequent detection and quantitation has been achieved using the enzymatic activity of horseradish peroxidase conjugated either to the antibody or to avidin. Both types of assays are feasible, with avidin capture being the preferred mode when enzyme is evaluated in the presence of excess zymogen, as would be common in the evaluation of most blood-clotting enzymes. Assays are illustrated for tissue plasminogen activator, plasmin, thrombin, factor Xa, and activated protein C, which can measure protease concentrations as low as 50 pmol/L. Specific applications of the assays are provided in studies of the activation of prothrombin by the prothrombinase complex and of factor X with Russell's viper venom factor X activator. These assays measure the mass of active site present in the reaction mixture and are relatively independent of subspecies of enzyme or the environment in which the activity is generated. These assay systems provide powerful tools for elucidating product-precursor relationships in multienzyme feedback reactions involving zymogen activation.

Enzymatic detection of native and derivatized horseradish peroxidase in sodium dodecyl sulfate polyacrylamide gels

We developed procedures for the restoration of peroxidatic activity in native horseradish peroxidase (HRP) and HRP conjugated to wheat germ agglutinin (WGA-HRP) following electrophoresis in SDS-polyacrylamide gels (SDS-PAGE). After extraction of SDS with isopropanol from gels containing HRP and WGA-HRP, the peroxidatic activity in these probes could be demonstrated by tetramethylbenzidine (TMB) chemistry. This procedure also showed HRP enzyme activity in electrophoresed tissue homogenates containing HRP. Both free HRP as well as WGA-HRP preparations contain several molecular weight species that display peroxidatic activity. These findings are important for cell biological studies utilizing these substances as molecular probes. The procedures described here should be useful for the analysis of the enzymatically active molecular forms of these frequently used markers in vitro and in vivo.

Fixation variables in horseradish peroxidase neurohistochemistry. I. The effect of fixation time and perfusion procedures upon enzyme activity.

In a series of neurohistochemical experiments the effect of aldehyde fixation upon the detection of horseradish peroxidase (HRP) was examined. These experiments demonstrated that: a) Increments in fixation of as little as 1 hr significantly decreased the number of labeled neurons; 12-hr fixation abolished HRP activity in many neuronal populations and significantly reduced the apparent size of the injection site. b) This negative fixation effect was greatest where the HRP concentration was low (e.g. in small, lightly labeled neurons) but was still evident in areas of high concentration (e.g. large, heavily labeled neurons). c) This effect was also most prominent when a less sensitive diaminobenzidine histochemical procedure was employed but was still apparent with a more sensitive benzidine dihydrochloride procedure. d) Immersion of the brain in fixative after perfusion produced a greater attenuation of HRP activity in more superficial areas. e) Immersion of the brain in buffer to terminate fixation produced a prolonged and unpredictable gradient of fixation. f) Excess, unbound fixative inhibited the histochemical reaction per se and had to be removed from the tissue but prolonged washing did not resurrect enzyme activity which was lost by fixation. To obviate these problems and optimize HRP enzyme activity a new perfusion-fixation procedure was developed. It entails 30 min fixation by perfusion which is terminated by a subsequent 30 min perfusion with cold sucrose-fuller to wash out unbound fixative. This allows the tissue to be processed immediately, produces a uniform and morphologically adequate fixation, and minimizes the negative effects of fixation on HRP enzyme activity.

Retrograde labeling of central nervous pathways with tritiated or Evans blue-labeled bovine serum albumin

In an attempt to develop a technique for the retrograde tracing of neuronal pathways which does not depend on enzyme activity of horseradish peroxidase (HRP), we have examined the retrograde transport of bovine serum albumin (BSA) by neurons of the central nervous system. Following injections of either tritiated or Evans blue-labeled BSA into the hippocampal formation of the rat, labeled neurons were found in the ipsilateral entorhinal area in a pattern which was highly reminiscent of that obtained with HRP. The retrograde transport of BSA provides a method for the retrograde tracing of central nervous pathways which is completely independent of the HRP method, making it possible to use both substances for studies of collateral innervation, and can be used with any fixative, or no fixative at all.

Circulation and turnover of synaptic vesicle membrane in cultured fetal mammalian spinal cord neurons.

Intact neurons in cultures of fetal rodent spinal cord explants show stimulation-dependent uptake of horseradish peroxidase (HRP) into many small vesicles and occasional tubules and multivesicular bodies (MVB) at presynaptic terminals. Presynaptic terminals were allowed to take up HRP during 1 h of strychnine-enhanced stimulation of synaptic transmitter release and then "chased" in tracer-free medium either with strychnine or with 10 mM Mg++ which depresses transmitter release. Tracer-containing vesicles are lost from terminals under both chase conditions; the loss is more rapid (4-8 h) with strychnine than with 10 mM Mg++ (8-16 h). There is a parallel decrease in the numbers of labeled MVB's at terminals. Loss of tracer with 10 mM Mg++ does not appear to be due to the membrane rearrangements (exocytosis coupled to endocytosis) that presumably lead to initial tracer uptake; terminals exposed to HRP and Mg++ for up to 16 h show little tracer uptake into vesicles. Nor is the decrease likely to the due to loss of HRP enzyme activity; HRP is very stable in solution. During the chases there is a striking accumulation of HRP in perikarya that is far more extensive in cultures initially exposed to tracer with strychnine than 10 mM Mg++ regardless of chase conditions. Much of the tracer ends up in large dense bodies. These findings suggest that synaptic vesicle membrane turnover involves retrograde axonal transport of membrane to neuronal perikarya for further processing, including lysosomal degradation. The more rapid (4-8 h) loss of tracer-containing vesicles with strychnine may reflect vesicle membrane reutilization for exocytosis.

The effect of methanol on granulocyte and horseradish peroxidase quantitatively studied in a film model system.

Using a model system of polyacrylamide films into which peroxidase had been incorporated, the effect of methanol on histochemically demonstrated enzyme activity could be investigated in quantitative terms. After staining with 3,3′-diaminobenzidine in the presence of hydrogen peroxide it appeared that methanol exerted a marked inhibiting effect on granulocyte peroxidase activity, but did not influence the enzymatic activity of horseradish peroxidase.

The interaction of soluble horseradish peroxidase with mouse peritoneal macrophages in vitro.

The in vitro interaction of soluble horseradish peroxidase (HRP) with homogeneous mono layers of mouse macrophages has been studied using sensitive biochemical and cytochemical techniques. The compartmentalization of HRP in extracellular and intracellular sites has been quantitatively evaluated. A significant fraction is bound to a serum-derived layer, which coats the surface of culture vessels and may be removed by appropriate washes. Macrophages interiorize HRP as a solute in pinocytic vesicles without appreciable binding of the glycoprotein to the plasma membrane. Uptake is directly proportional to the concentration of HRP in the culture medium. 1 x 10(6) cells ingest 0.0025% of the administered load per hr over a wide range of concentrations. Cytochemically, all demonstrable HRP is sequestered within the endocytic vesicles and secondary lysosomes of the vacuolar apparatus. After uptake, the enzymatic activity of HRP is inactivated exponentially with a half-life of 7-9 hr, until enzyme is no longer detectable. When macrophages have pinocytosed trace-labeled HRP-(125)I, cell-associated isotope disappears with a t (1/2) of 20-30 hr and they release monoiodotyrosine-(125)I into the culture medium. We were unable to obtain evidence that significant amounts of HRP (>2%) can be exocytosed after uptake, can exist intact on the cell surface, or can be digested extracellularly. It is difficult to reconcile these observations with several of the postulated mechanisms whereby macrophages are thought to play a prominent role in the induction of an immune response.
