Engineering Material Properties of Transcription Factor Condensates to Control Gene Expression in Mammalian Cells and Mice.

Phase separation of biomolecules into condensates is a key mechanism in the spatiotemporal organization of biochemical processes in cells. However, the impact of the material properties of biomolecular condensates on important processes, such as the control of gene expression, remains largely elusive. Here, the material properties of optogenetically induced transcription factor condensates are systematically tuned, and probed for their impact on the activation of target promoters. It is demonstrated that transcription factors in rather liquid condensates correlate with increased gene expression levels, whereas stiffer transcription factor condensates correlate with the opposite effect, reduced activation of gene expression. The broad nature of these findings is demonstrated in mammalian cells and mice, as well as by using different synthetic and natural transcription factors. These effects are observed for both transgenic and cell-endogenous promoters. The findings provide a novel materials-based layer in the control of gene expression, which opens novel opportunities in optogenetic engineering and synthetic biology.

Multiscale computational study to predict the irradiation-induced change in engineering properties of fusion reactor materials

In this study, we address the impact of irradiation conditions in a tokamak on the engineering properties of materials, leading to potential degradation of in-vessel components over their lifecycle. Our approach involves a predictive model for irradiation-induced damage, employing a multiscale computational framework. This framework integrates various simulation techniques, including Monte Carlo-based neutronics (OpenMC), dislocation dynamics (DD) using MoDELib, and finite element analysis (FEA) with Code_Aster. This integration offers a versatile solver capable of analysing tokamak components exposed to different irradiation doses and temperature conditions.To showcase the utility of this multiscale computational framework, we present a case study focused on tungsten monoblock designs. We assess the failure probabilities of these designs at different stages of their lifecycle. Neutron heating and damage energy values are obtained from OpenMC neutronics simulations. The neutron heating values serve as volumetric heat sources for the FEA thermal simulation. We calculate the displacement per atom (dpa) across the monoblock at various full power days (day 0, day 100, and day 1000) using the damage energy values. The irradiation-induced defect densities, dependent on temperature and dpa, are inputs to DD microstructural simulations performed on the representative volume element (RVE) using MoDELib. This allows us to obtain the yield stress of the material. Subsequently, the thermal fields from the FEA thermal simulation, along with the dpa and temperature-dependent yield stress from the DD simulation, are implemented for FEA mechanical simulations.To evaluate the failure probability of the monoblock designs at different stages of their lifecycle, we conduct an SDC-IC assessment, incorporating a plastic flow localization rule within the current framework. This comprehensive approach provides insights into the thermo-mechanical behaviour of in-vessel components subjected to neutron irradiation, offering a predictive capability for assessing their performance over time.

Prediction and optimization of civil engineering material properties based on artificial intelligence

In the modern research field of predicting and optimizing the performance of civil engineering materials, the incorporation of artificial intelligence (AI) has provided a new perspective. The research is based on a thorough examination of the central role of AI technology and the application of different mathematical models in the task of material performance prediction and optimization, summarizing the current state of AI technology application in the field of civil engineering material performance prediction. Against this backdrop, the research further proposes a brand-new AI-based civil engineering material performance prediction method. This study, aiming to realize the optimization of material performance prediction tasks, utilized cutting-edge machine learning techniques and mathematical models, proposing this new AI prediction framework. Detailed discussions on design, implementation, and evaluation were conducted in the forecast framework, which includes a large number of tables, mathematical functions, and references. Through rigorous study and implementation, the prediction framework exhibited admirable performance, demonstrating extensive applicability for civil engineering material performance prediction and optimization. In conclusion, this new AI-based civil engineering material performance prediction method has demonstrated encouraging results. This study provides a new horizon, illustrating the tremendous potential and value of AI for predicting the performance of civil engineering materials, thereby improving efficiency and effectiveness in the civil engineering field. Overall, this new AI prediction method provides a fresh pathway for future research, making an essential contribution to anticipating and optimizing in civil engineering field.

Dehydration kinetics of nanoconfined water in beryl probed by high temperature single crystal synchrotron X-ray diffraction

Understanding changes in material properties through external stimuli plays a key role in validating the expected performance of materials and engineering material properties in a controlled manner. Here, we introduce a fundamental protocol to deduce dehydration reactions kinetics of water confined in nanopore channels, with the cyclosilicate beryl as the scaffold of interest, using time-resolved synchrotron X-ray diffraction (SXRD), in the temperature interval of 298–1038 K. The crystal structure of beryl determined up to 1038 K, in temperature increment as small as 10 K, indicates the presence of channel ions Na and Fe and a gradual decrease of water upon heating.

The coupled effect of aspect ratio and strut micro-deformation mode on the mechanical properties of lattice structures

Lattice structures have been integrated into various industrial applications owing to their unique compressive properties. Mechanical characterisation is usually done by testing a small specimen which is assumed representative of the utilised lattice. A specimen's aspect ratio (height to diameter/width ratio) is known to affect compressive properties in various engineering materials, yet its influence in lattices has not been investigated thoroughly. In this study, titanium lattice specimens designed with different aspect ratios (ranging from 0.5 to 3.0) and four different topologies (displaying bend and stretch-dominated micro-deformation modes) were fabricated using powder bed fusion and tested in quasi-static compression. Their compressive properties and failure modes were evaluated using acquired stress-strain curves and digital image correlation (DIC) analysis. Reducing the aspect ratio in the bend-dominated lattices increased the measured stiffness of the specimens by up to 40%. Conversely, increasing the aspect ratio of the stretch dominated lattices increased the measured stiffness of the specimens by up to 30%. For both topology types, decreasing the aspect ratio increased the measured strength of the specimens, but the effect was less than that observed for stiffness. Different responses were attributed to gradient strain accumulation and different failure patterns (densification versus shear banding) that were observed depending on the combination of aspect ratio and topology. These findings are particularly important for better predicting the mechanical behaviour of lattice-based components that have aspect ratios outside the range of conventional test specimens.

Digital Multiphase Composites via Additive Manufacturing.

Mechanical properties of traditional engineering materials are typically coupled to each other, presenting a challenge to practitioners with multi-dimensional material property requirements. In this work, continuous, independent control over multiple mechanical properties is demonstrated in composite materials realized using additive manufacturing. For the first time, composites additively manufactured from rigid plastic, soft elastomer, and liquid constituents are experimentally characterized, demonstrating materials which span four orders of magnitude in modulus and two orders of magnitude in toughness. By forming analytical mappings between relative concentrations of constituents at the microscale and resulting macroscale material properties, inverse material design is enabled; the method is showcased by printing artifacts with prescribed toughness and elasticity distributions. The properties of these composites are placed in the context of biological tissues, showing they have promise as mechanically plausible tissuemimics.

Investigating the mechanical properties of al7075 metal matrix composite with improved performance through the incorporation of fe3o4 and RHS

Heavily employed in engineering, Metal Matrix Composites are the primary focus of this research. Metal Matrix Composites have much improved mechanical properties and find extensive use across several sectors, including automotive and aerospace. The experiment entails using aluminium (Al7075) as the base material and incorporating iron oxide and RHS in weight ratios of 3%, 6%, and 9% to fabricate a strengthened Metal Matrix Composite. The stir-casting technique is used for the fabrication of these composites. The stir casting technology is selected because to its advantages, including its straightforward manufacturing process, cost efficiency, and reliable distribution of reinforced particles. The Fe3O4 reinforcing particle and RHS have a significant impact on the engineering material properties. The examination revealed enhancements in mechanical properties, including tensile, compression, and hardness strengths. The tensile strength of the 9% wt. reinforced Fe3O4 and RHS is found to be the highest at 362 MPa, while the yield strength is measured at 262 MPa, when compared to the original Al- 7075 alloy. Furthermore, the recently manufactured Al7075/Fe3O4 and RHS Metal Matrix Composite were subjected to microstructural examinations using SEM and EDS methodologies. The investigations demonstrated a uniform and homogeneous dispersion of Fe3O4and RHS reinforced particles inside the composite material.

Field-dependent THz transport nonlinearities in semiconductor nano structures.

Charge transport nonlinearities in semiconductor quantum dots and nanorods are studied. Using a density matrix formalism, we retrieve the field-dependent nonlinear mobility and show the possibility of intra-pulse gain. We further demonstrate that the dynamics of master equations can be captured in an analytical formula for the field-dependent charge carrier mobility, e.g. for two-level systems. This equation extends the linear response theory based Kubo-Greenwood result to nonlinear processes at elevated field strength, easily reached in THz transport spectroscopy. With these tools we analyze the field strength, chirp, temperature and dephasing dependence of the charge carrier mobility in the model system of CdSe quantum dots and wires. Stark broadening and Rabi splitting result in strong alterations of the mobility spectra, pronounced at low temperatures. The mobility spectra are strongly temperature and pulse shape dependent in the nonlinear regime. The findings are of immediate interest e.g. for nonlinear THz generation, conversion and amplification in 6G technology and nano electronics. Our results further enable experimentalists to fit and understand measured charge transport nonlinearities with analytical expressions and to design nanosystems with engineered material properties.

Analysis of microstructure, surface morphology, and scratch resistance of high velocity oxy-fuel coated nickel based hybrid nano-composites

In recent years, high velocity oxygen fuel (HVOF) coating technology has gained significant interest in the field of surface engineering. In this study, we have investigated the microstructure, surface morphology, and scratch resistance of HVOF coated Nickel based novel hybrid nano-composites. The coatings were prepared using a high-velocity oxy-fuel (HVOF). HVOF coatings have the ability to enhance the surface properties of various materials, making them more resistant to wear, corrosion, and erosion. Thermal spraying technique, and the microstructure and surface morphology of the coatings were analyzed using scanning electron microscopy (SEM). The scratch test was performed using a TR-101, DUCOM made Scratch tester to evaluate the scratch resistance of the coatings. The results of the study revealed that the HVOF coated novel hybrid nano-composites exhibited a fine-grained microstructure with a dense and homogeneous surface morphology. The scratch resistance of the coatings was significantly improved due to the addition of nano composites. The load required to cause the first visible scratch on the coated surface was found to be significantly higher than that of the uncoated substrate. The scratch morphology of the coatings was also studied, which revealed that the scratch-induced damage was mainly due to the plastic deformation of the coating material. The results suggest that the HVOF coated novel hybrid nano composites have great potential in improving the surface properties of various engineering materials and can be used in applications where high wear resistance is required. Overall, the study demonstrates that the combination of HVOF coating technology and hybrid nano-composites can result in coatings with improved microstructure, surface morphology, and scratch resistance, which makes them ideal for use in applications wherever wear resistance is critical.

Subsurface imaging dataset acquired at the Garner Valley Downhole Array site using a dense network of three-component nodal stations

There is a growing need to characterize the engineering material properties of the shallow subsurface in three dimensions for advanced engineering analyses. However, imaging the near-surface in three dimensions at spatial resolutions required for such purposes remains in its infancy and requires further study before it can be adopted into practice. To enable and accelerate research in this area, we present a large subsurface imaging data set acquired using a dense network of three-component (3C) nodal stations acquired in 2019 at the Garner Valley Downhole Array (GVDA) site. Acquisition of this data set involved the deployment of 196 stations positioned on a 14 × 14 grid with a 5 m spacing. The array was used to acquire active-source data generated by a vibroseis truck and an instrumented sledgehammer, and passive-wavefield data containing ambient noise. The active-source acquisition included 66 vibroseis and 209 instrumented sledgehammer source locations. Multiple source impacts were recorded at each source location to enable stacking of the recorded signals. The active-source recordings are provided in terms of both raw, uncorrected units of counts and corrected engineering units of meters per second. For each source impact, the force output from the vibroseis or instrumented sledgehammer was recorded and is provided in both raw counts and engineering units of kilonewtons. The passive-wavefield data include 28 h of ambient noise recorded over two nighttime deployments. The data set is shown to be useful for active-source and passive-wavefield three-dimensional imaging and other subsurface characterization techniques, which include horizontal-to-vertical spectral ratios (HVSRs), multichannel analysis of surface waves (MASW), and microtremor array measurements (MAM).

Cement-stabilized large-sized steel slag base course based on vibration compaction method: Mechanical properties, shrinkage properties, and numerical simulation

The excellent mechanical properties and wear resistance of steel slag contribute to the properties of road engineering materials. The hot-stuffing aged slag was chosen as raw material for this study. Cement-stabilized large-sized steel slag (CSLS) mixtures containing different structural types, cement contents, and curing time were prepared by vibration compaction method. Afterward, the properties of CSLS mixtures such as unconfined compressive strength (UCS), indirect tensile strength (ITS), compressive resilience modulus (CRM), dry shrinkage and temperature shrinkage were investigated. Ultimately, the Mises stress and deflection value of CSLS road structure with different construction methods and construction seasons were explored by ABAQUS.

Neuromorphic sensorimotor loop embodied by monolithically integrated, low-voltage, soft e-skin

Artificial skin that simultaneously mimics sensory feedback and mechanical properties of natural skin holds substantial promise for next-generation robotic and medical devices. However, achieving such a biomimetic system that can seamlessly integrate with the human body remains a challenge. Through rational design and engineering of material properties, device structures, and system architectures, we realized a monolithic soft prosthetic electronic skin (e-skin). It is capable of multimodal perception, neuromorphic pulse-train signal generation, and closed-loop actuation. With a trilayer, high-permittivity elastomeric dielectric, we achieved a low subthreshold swing comparable to that of polycrystalline silicon transistors, a low operation voltage, low power consumption, and medium-scale circuit integration complexity for stretchable organic devices. Our e-skin mimics the biological sensorimotor loop, whereby a solid-state synaptic transistor elicits stronger actuation when a stimulus of increasing pressure is applied.

Engineering Properties and Environmental Impact of Soil Mixing with Steel Slag Applied in Subgrade

The purpose of this study was to evaluate the feasibility of the large-scale application of steel slag (SL) in subgrade. Subgrade materials with three kinds of SL proportions were first prepared. Then, a compaction test, liquid-plastic limit combined-measurement test, and a California bearing ratio (CBR) test were applied to determine the best proportion between SL and intact soil (S), i.e., SL/S. Subsequently, static and dynamic tests and a volume stability test were carried out for soil mixed with SL at the optimum proportion (SSL). In addition, a composition analysis of infiltration fluid and a permeability test of SSL were performed. The test results showed that compared to S, the physical properties of SSL were significantly improved, especially the liquid-plastic limit, as well as the soil water stability. The optimum proportion of SL was determined as 50% of soil by mass. At the optimum proportion, SSL had the highest CBR value of 60%, which had both economic and engineering compaction performance, leading to a large-scale utilization rate of SL. The static and dynamic characteristics showed that the addition of SL would greatly improve the shear strength and dynamic modulus of soil, mainly expressed as the increase of internal friction angle. The volume stability of SSL could also meet the requirements of the Chinese specification. After adding 2% cement, the strength and stability of SSL was further improved. In addition, the environmental impact test proved that the infiltration liquid did not pollute surface water nor underground secondary water. Although the permeability coefficient of SSL with the optimum proportion of 50% was higher than that of pure soil, it still belonged to the normal value of clay and silty clay, and good impermeability would ensure the controllability of potential trace elements. Based on the test results of mechanical properties and environmental impact, SSL proved to have the potential for green road material engineering properties. This study proposes a reliable and practical method to promote the utilization of steel slag.

Experimental modal analysis of fly ash-based geopolymer concrete specimens via modal circles, mode indication functions, and mode shape animations

The polymerization process of geopolymer concrete is entirely different from the hydration process of ordinary Portland cement concrete. Therefore, it is necessary to re-examine this new material's engineering properties from the ground up. To date, researchers have investigated many engineering properties, but in the whole literature, the available information about the modal characteristics is minimal. Additionally, since the evaluations in the literature are made only on the A ratio (sodium silicate to sodium hydroxide ratio), no significant relationship has been reported between damping and activators. So, this study is conducted to draw attention to this gap in the literature and examine the effect of fundamental geopolymer concrete component ratios on modal characteristics. For these purposes, large-scale heat-cured low-calcium fly ash-based geopolymer concrete mix variations are produced and investigated by performing experimental modal analysis via the advanced modal circle method and mode indication functions. It is found that individually analyzing the total amount of water from the activators and the pure chemical dosages provides more meaningful results than the traditional A ratio. According to the analysis from this perspective, increasing the dosage of pure sodium silicate in the mixture generally increases the damping; on the other hand, it decreases the resonance frequency and dynamic elasticity modulus. Obtained findings of this study reveal some of the methods and benefits of appropriately designing this sustainable material against dynamic loads.

Nonlocal Probing of Amplitude Mode Dynamics in Charge-Density-Wave Phase of EuTe 4

Amplitude mode is collective excitation emerging from frozen lattice distortions below the charge-density-wave (CDW) transition temperature T CDW and relates to the order parameter. Generally, the amplitude mode is non-polar (symmetry-even) and does not interact with incoming infrared photons. However, if the amplitude mode is polar (symmetry-odd), it can potentially couple with incoming photons, thus forming a coupled phonon–polariton quasiparticle that can travel with light-like speed beyond the optically excited region. Here, we present the amplitude mode dynamics far beyond the optically excited depth of ∼150 nm in the CDW phase of ∼10-μm-thick single-crystal EuTe 4 using time-resolved x-ray diffraction. The observed oscillations of the CDW peak, triggered by photoexcitation, occur at the amplitude mode frequency ω AM . However, the underdamped oscillations and their propagation beyond the optically excited depth are at odds with the observation of the overdamped nature of the amplitude mode measured using meV-resolution inelastic x-ray scattering and polarized Raman scattering. The ω AM is found to decrease with increasing fluence owing to a rise in the sample temperature, which is independently confirmed using polarized Raman scattering and ab-initio molecular dynamics simulations. We rationalize the above observations by explicitly calculating two coupled quasiparticles—phonon–polariton and exciton–polariton. Our data and simulations cannot conclusively confirm or rule out the one but point toward the likely origin from propagating phonon–polariton. The observed non-local behavior of amplitude mode thus provides an opportunity to engineer material properties at a substantially faster time scale with optical pulses.

Segregation behaviors of nonmetallic impurities on grain boundary of austenitic steel

Grain boundary (GB) always takes a critical role in the mechanical properties of most engineering materials, and austenitic steel (γ-Fe) is no exception. However, the microscopic mechanism of impurity segregation on γ-Fe GB is still unclear. In this work, we explored the stabilities of eight γ-Fe GBs by density functional theory (DFT) and chose the Σ5 (310) GB as a representative. Here, the separation energy, GB energy, strengthening energy, and theoretical tensile stress of Σ5 (310) GB doped by eight common nonmetallic elements (H, B, C, N, O, Si, P, and S) were analyzed to evaluate the effect of impurities segregation comprehensively. The results show that except for H and Si, impurities of B, C, N, O, P and S all can stabilize Σ5 (310) GB. However, only the B element can strengthen Σ5 (310) GB. Finally, the calculated electron transfer of doped Σ5 (310) GB unveils the superiority of the boron element and the segregation effect of other impurities. This work provides a systematical insight into the effect of impurity doping on Σ5 (310) GB, which will be a useful reference for γ-Fe processing research.

A study on the applicability of scoria gravel an alternative base course material through blending with marble waste aggregate

This study aims to evaluate the applicability of Scoria gravel as an alternative base course material in flexible pavements through blending with marble waste aggregate (MWA) by modifying the physical and mechanical engineering properties of scoria. Non-Probabilistic sampling techniques and experimental methods were used. To achieve the objectives of the study, the laboratory tests were passed in three steps. First, the Engineering properties of materials were independently tested; the result proves the marginality of scoria gravel. Second, scoria gravel was mechanically stabilized by 20% percentages by weight increments of MWA. The obtained engineering properties test results at 20:80 Scoria to MWA mix ratios are 2.56%, 21.38%, 18.59%, 19.27%, 17.45%, 13.77%, Non-Plastic, 1.21%, and 73.4%, for Specific Gravity, Aggregate Crushing Value, Aggregate Impact Value, Loss Angeles Abrasion, Flakiness Index, Elongation Index, Atterberg's limit, Water Absorption, and California Bearing Ratio (CBR) respectively. These test results fulfilled the ERA standard specification for GB2 and GB3 base course materials. However, the CBR test results showed a failure to meet the standard spécification. Thus, 20:80 Scoria to MWA percentage by weight ratio was selected as a control mixture. So, Crushed Stone Aggregate was added at 5% percentage by weight to improve the CBR of scoria gravel. Therefore, the CBR value of 82.13% attained the ERA standard Specification limit for base course materials at 15:60:25 percentages by weight ratio of scoria gravel, MWA, and CSA respectively. Finally, Based on this study it was recommended to use scoria gravel as an alternative base course construction material, when it was found abundantly near construction vicinity.

Indentation of pore-spanning lipid membranes: Spring-stiffening or -softening responses and apparent stiffness prediction

Atomic force microscopy (AFM)-based nanoindentation has emerged as a key nanomechanical mapping method to probe mechanical properties of two-dimensional engineering and biological materials. In comparison with extensive analytical studies on the indentation of freestanding solid thin films, surprisingly little attention has been drawn on analytical predictions of mechanical responses of freestanding lipid membranes to indentation in the framework of Helfrich membrane theory. Here we perform systematic theoretical studies with formulated analysis on indentation of nanopore-spanning lipid membranes. Two types of lipid membranes are considered. One is the membrane subject to uniform area stretching and the membrane is found to exhibit a spring-stiffening behavior during indentation, while the other is the membrane of constant tension and a spring-softening behavior at intermediate indentation is exhibited. The smaller the membrane tension is, the clearer the spring-softening feature is. For both types of membranes, analytical solutions of load–depth relationship at small indentation and apparent membrane stiffnesses are obtained at small and large tension. For the stretched lipid membranes, a cubic nonlinearity arising from membrane stretching is specified. Roles of membrane bending rigidity, (pre-)tension, or area compression modulus, and indenter size in membrane indentation are elucidated. Moreover, pressure of contact between the membrane and indenter and a line load of contact acting along the contact edge are obtained based on the shell theory. Our results provide fundamental insights on nonlinear mechanical behaviors of the lipid membrane indentation.

Fungi in Mycelium-Based Composites: Usage and Recommendations.

Mycelium-Based Composites (MBCs) are innovative engineering materials made from lignocellulosic by-products bonded with fungal mycelium. While some performance characteristics of MBCs are inferior to those of currently used engineering materials, these composites nevertheless prove to be superior in ecological aspects. Improving the properties of MBCs may be achieved using an adequate substrate type, fungus species, and manufacturing technology. This article presents scientifically verified guiding principles for choosing a fungus species to obtain the desired effect. This aim was realized based on analyses of scientific articles concerning MBCs, mycological literature, and patent documents. Based on these analyses, over 70 fungi species used to manufacture MBC have been identified and the most commonly used combinations of fungi species-substrate-manufacturing technology are presented. The main result of this review was to demonstrate the characteristics of the fungi considered optimal in terms of the resulting engineering material properties. Thus, a list of the 11 main fungus characteristics that increase the effectiveness in the engineering material formation include: rapid hyphae growth, high virulence, dimitic or trimitic hyphal system, white rot decay type, high versatility in nutrition, high tolerance to a substrate, environmental parameters, susceptibility to readily controlled factors, easy to deactivate, saprophytic, non-mycotoxic, and capability to biosynthesize natural active substances. An additional analysis result is a list of the names of fungus species, the types of substrates used, the applications of the material produced, and the main findings reported in the scientific literature.
