Transcriptomic and physiological analyses reveal that jasmonic acid and abscisic acid coordinately regulate cold stress response in Myriophyllum aquaticum

Myriophyllum aquaticum, renowned for its ability to purify aquaculture wastewater, faces a significant challenge due to its susceptibility to cold stress. However, the molecular mechanisms underlying M. aquaticum's response to cold stress remain poorly understood. In this study, we unraveled the molecular mechanisms governing M. aquaticum's response to cold stress through transcriptome sequencing and physiological analyses. Our investigation revealed 12,684 differentially expressed genes during cold stress. Gene ontology and KEGG analyses demonstrated the involvement of plant hormone signaling and transcriptional reprogramming related genes in M. aquaticum's response to cold stress. Further investigations highlighted the role of jasmonic acid (JA) and abscisic acid (ABA) in M. aquaticum's cold acclimation. Firstly, cold stress significantly increased endogenous levels of JA and ABA in M. aquaticum plants. Secondly, the transcript levels of several crucial JA and ABA signaling genes were prominently elevated, as shown by expression profiling and qRT-PCR analyses. Thirdly, the application of exogenous JA or ABA effectively mitigated foliar damage and improved cold tolerance in M. aquaticum plants. Furthermore, the expression of JA and ABA biosynthesis and signaling genes was significantly upregulated in JA- or ABA-pretreated plants when exposed to cold stress. Collectively, our findings provide evidence that JA and ABA contribute to the cold acclimation of M. aquaticum plants. Our research provides insights into the molecular mechanisms underlying cold acclimation of M. Aquaticum plants and presents a novel strategy to improve their cold tolerance, thereby expanding their potential application in phytoremediation efforts in high latitude areas.

Detection and Estimation of Endogenous Jasmonic Acid and Salicylic Acid in Potato Genotypes Imposed with Defense Modulators Using High Performance Liquid Chromatography

Jasmonic Acid (JA) and Salicylic Acid (SA) are pivotal signaling molecules involved in the complex network of plant defense mechanisms. Enhancing the levels of JA and SA through exogenous application or genetic manipulation has shown promising results in bolstering plant resistance to a wide range of diseases. This study investigates the effects of JA, SA and Compost Tea (CT) treatments on the endogenous JA and SA responses of four distinct potato genotypes-KH (Kufri Himalini, Solanum tuberosum), KJ (Kufri Jyothi, Solanum tuberosum), AC4 (Solanum sparsipillium) and AC6 (Solanum spegzinii). High-Performance Liquid Chromatography (HPLC) analysis was conducted to determine endogenous detectable levels of JA and SA defence phytohormones. The absorption spectra showed peak absorptions at 295 nm and 325 nm for JA (25 µg ml-1) and SA (10 µg ml-1) respectively. Retention times of JA and SA were determined to be RT-3.74 and RT-2.86 min respectively. The genotypes were subjected to various concentrations of JA and SA, and their respective impacts on endogenous JA and SA were measured. Genotype-specific responses were evident, with all genotypes displaying distinct sensitivities with respect to endogenous detectable JA in CT (KH-15.42 µg ml-1, KJ-133.67 µg ml-1, AC4-10.36 µg ml-1, AC6-7.92 µg ml-1), JA (KH-83.03 µg ml-1, KJ-184.14 µg ml-1, AC4-286.95 µg ml-1, AC6-9.2 µg ml-1) treatments while no detectable JA and SA was observed in Control, SA and JA+SA combined treatments. JA could enhance higher amount of detectable endogenous JA than CT treatment. The application of JA and SA enhancement could demonstrate success in various other crops rendering resistance against diseases in an eco-friendly approach.

Botrytis cinerea-induced F-box protein 1 enhances disease resistance by inhibiting JAO/JOX-mediated jasmonic acid catabolism in Arabidopsis

Jasmonic acid (JA) is a crucial phytohormone that regulates plant immunity. The endogenous JA level is determined by the rates of its biosynthesis and catabolism in plants. The activation of JA biosynthesis has been well documented; however, how plants repress JA catabolism upon pathogen infection remains elusive. In this study, we identified and characterized Botrytis cinerea–induced F-box protein 1 (BFP1) in Arabidopsis. The expression of BFP1 was induced by B. cinerea in a JA signaling–dependent manner, and BFP1 protein was critical for plant defense against B. cinerea and plant response to JA. In addition, BFP1 overexpression increased plant defenses against broad-spectrum pathogens without fitness costs. Further experiments demonstrated that BFP1 interacts with and mediates the ubiquitination and degradation of jasmonic acid oxidases (JAOs, also known as jasmonate-induced oxygenases, JOXs), the enzymes that hydroxylate JA to 12OH-JA. Consistent with this, BFP1 affects the accumulation of JA and 12OH-JA during B. cinerea infection. Moreover, mutation of JAO2 complemented the phenotypes of the bfp1 mutant. Collectively, our results unveil a new mechanism used by plants to activate immune responses upon pathogen infection: suppressing JA catabolism.

Transcription factors BZR2/MYC2 modulate brassinosteroid and jasmonic acid crosstalk during pear dormancy.

Bud dormancy is an important physiological process during winter. Its release requires a certain period of chilling. In pear (Pyrus pyrifolia), the abscisic acid (ABA)-induced expression of DORMANCY-ASSOCIATED MADS-box (DAM) genes represses bud break, whereas exogenous gibberellin (GA) promotes dormancy release. However, with the exception of ABA and GA, the regulatory effects of phytohormones on dormancy remain largely uncharacterized. In this study, we confirmed brassinosteroids (BRs) and jasmonic acid (JA) contribute to pear bud dormancy release. If chilling accumulation is insufficient, both 24-epibrassinolide (EBR) and methyl jasmonic acid (MeJA) can promote pear bud break, implying that they positively regulate dormancy release. BRASSINAZOLE RESISTANT 2 (BZR2), which is a BR-responsive transcription factor, inhibited PpyDAM3 expression and accelerated pear bud break. The transient overexpression of PpyBZR2 increased endogenous GA, JA, and JA-Ile levels. In addition, the direct interaction between PpyBZR2 and MYELOCYTOMATOSIS 2 (PpyMYC2) enhanced the PpyMYC2-mediated activation of Gibberellin 20-oxidase genes PpyGA20OX1L1 and PpyGA20OX2L2 transcription, thereby increasing GA3 contents and accelerating pear bud dormancy release. Interestingly, treatment with 5 μm MeJA increased the bud break rate, while also enhancing PpyMYC2-activated PpyGA20OX expression and increasing GA3,4 contents. The 100 μm MeJA treatment decreased the PpyMYC2-mediated activation of the PpyGA20OX1L1 and PpyGA20OX2L2 promoters and suppressed the inhibitory effect of PpyBZR2 on PpyDAM3 transcription, ultimately inhibiting pear bud break. In summary, our data provide insights into the crosstalk between the BR and JA signaling pathways that regulate the BZR2/MYC2-mediated pathway in the pear dormancy release process.

Exogenous methyl jasmonate enhanced the antioxidant capacity of Malus baccata by stimulating jasmonate signalling under suboptimal low root-zone temperature

Methyl jasmonate (MeJA) has been documented to mitigate the oxidative stress caused by abiotic triggers. In this study, we investigated the mechanism through which exogenous MeJA alleviates oxidative damage in the roots of Malus baccata under a suboptimal low root-zone temperature. A suboptimal low root-zone temperature promoted increases in the activity and transcription of allene oxide synthase (AOS) and jasmonic acid carboxyl methyltransferase (JMT), elevated the levels of endogenous jasmonic acid (JA), MeJA and jasmonate isoleucine (JA-Ile), and induced the accumulation of malondialdehyde (MDA) and reactive oxygen species (ROS). The addition of exogenous MeJA further increased the JA (1 h, 12–24 h), MeJA (1–24 h), and JA-Ile (1–6 h) contents and enhanced the activities and transcription of lipoxygenase (LOX) and JMT. The upregulated expression of JASMONATE ZIM-DOMAIN (JAZ) and MdMYC2 indicated that suboptimal low root-zone temperature could activate JA signalling in roots, and exogenous MeJA could further strengthen JA signalling under low-temperature stress. Exogenous MeJA alleviated oxidative damage to stressed roots by enhancing ascorbate–glutathione (AsA–GSH) cycle activity. In addition, pre-treatment with phenidone (an inhibitor of lipoxygenase) exerted the opposite effects to those induced by exogenous MeJA treatment. These findings indicated that exogenous MeJA enhanced antioxidant systems and detoxified ROS by regulating JA signalling and thereby protected M. baccata roots from damage caused by suboptimal low root-zone temperature stress.

Airborne Methyl Jasmonate is Metabolized to Jasmonic Acid and 12‐Hydroxyjasmonic Acid, and Induces Jasmonate Biosynthesis in Marchantia polymorpha

AbstractMethyl jasmonate (MeJA) is a volatile jasmonate compound commonly used to induce defense responses in spermatophytes. This study reports that airborne MeJA‐d3, deuterated MeJA, increases the levels of (dinor‐)12‐oxo‐phytodienoic acids [(dn‐)OPDAs] and jasmonic acid (JA) as well as JA‐d3 and 12‐hydroxyjasmonic acid‐d2 (12‐OH‐JA‐d2), MeJA‐d3 metabolites, in the model bryophyte Marchantia polymorpha. Enhancement of JA biosynthesis was substantiated by the expression of JA biosynthetic genes induced by airborne MeJA. Additionally, each of enantiomers, (+)‐MeJA and (−)‐MeJA, was observed to induce the accumulation of JA and (dn‐)OPDAs in M. polymorpha. This study demonstrates that airborne MeJA is metabolized to JA and 12‐OH‐JA, and induces JA biosynthesis in M. polymorpha. Moreover, the transient increase in endogenous JA level after airborne MeJA treatment provides concrete evidence that M. polymorpha biosynthesizes JA.

Unveiling the molecular mechanism involving anthocyanins in pineapple peel discoloration during fruit maturation

Peel color is a key factor that affects the fruit’s aesthetic and economic values. Limited knowledge is available on the regulation of pineapple peel discoloration. Here, we report that a decrease in anthocyanin biosynthesis, particularly cyanidin, is predominantly associated with the pineapple peel color change during maturation. The findings suggest that the changes in the expression of key structural genes (early and late biosynthetic genes) of the anthocyanin (cyanidin) biosynthesis pathway are responsible for peel discoloration. Based on a gene co-expression analysis and a transient expression, two transcription factors i.e., AcHOX21 and AcMYB12, were identified, whose’ downregulation leads to reduced anthocyanin accumulation with fruit maturation. The endogenous levels of jasmonic acid, gibberellic acid, and auxins are also involved in anthocyanin-content-led peel discoloration. Overall, the discovery of genes regulating anthocyanin biosynthesis in pineapple peel provides a theoretical basis for improving the fruit’s aesthetic value through genetic engineering.

Whole Genome Analysis of SLs Pathway Genes and Functional Characterization of DlSMXL6 in Longan Early Somatic Embryo Development.

Strigolactones (SLs), a new class of plant hormones, are implicated in the regulation of various biological processes. However, the related family members and functions are not identified in longan (Dimocarpus longan Lour.). In this study, 23 genes in the CCD, D27, and SMXL family were identified in the longan genome. The phylogenetic relationships, gene structure, conserved motifs, promoter elements, and transcription factor-binding site predictions were comprehensively analysed. The expression profiles indicated that these genes may play important roles in longan organ development and abiotic stress responses, especially during early somatic embryogenesis (SE). Furthermore, GR24 (synthetic SL analogue) and Tis108 (SL biosynthesis inhibitor) could affect longan early SE by regulating the levels of endogenous IAA (indole-3-acetic acid), JA (jasmonic acid), GA (gibberellin), and ABA (abscisic acid). Overexpression of SMXL6 resulted in inhibition of longan SE by regulating the synthesis of SLs, carotenoids, and IAA levels. This study establishes a foundation for further investigation of SL genes and provides novel insights into their biological functions.

IQD1 Involvement in Hormonal Signaling and General Defense Responses Against Botrytis cinerea.

IQ Domain 1 (IQD1) is a novel Arabidopsis thaliana calmodulin-binding protein, which was found to be a positive regulator of glucosinolate (GS) accumulation and plant defense responses against insects. We demonstrate here that the IQD1 overexpressing line (IQD1OXP) was also more resistant also to the necrotrophic fungus Botrytis cinerea, whereas an IQD1 knockout line (iqd1-1) was much more sensitive. Furthermore, we showed that IQD1 is up-regulated by jasmonic acid (JA) and downregulated by salicylic acid (SA). A comparison of whole transcriptome expression between iqd1-1 and wild type plants revealed a substantial downregulation of genes involved in plant defense and hormone regulation. Further examination revealed a marked reduction of SA and increases in the levels of ethylene, JA and abscisic acid response genes in the iqd1-1 line. Moreover, quantification of SA, JA, and abscisic acids in IQD1OXP and iqd1-1 lines relative to the wild type, showed a significant reduction in endogenous JA levels in the knockout line, simultaneously with increased SA levels. Relations between IQD1OXP and mutants defective in plant-hormone response indicated that IQD1 cannot rescue the absence of NPR1 or impaired SA accumulation in the NahG line. IQD1 cannot rescue ein2 or eto1 mutations connected to the ethylene pathway involved in both defense responses against B. cinerea and in regulating GS accumulation. Furthermore, IQD1cannot rescue the aos, coi1 or jar1mutations, all involved in the defense response against B. cinerea and it depends on JAR1 to control indole glucosinolate accumulation. We also found that in the B. cinerea, which infected the iqd1-1 mutant, the most abundant upregulated group of proteins is involved in the degradation of complex carbohydrates, as correlated with the sensitivity of this mutant. In summary, our results suggest that IQD1 is an important A. thaliana defensive protein against B. cinerea that is integrated into several important pathways, such as those involved in plant defense and hormone responses.

Investigating the Mechanism of Unilateral Cross Incompatibility in Longan (Dimocarpus longan Lour.) Cultivars (Yiduo × Shixia).

Longan (Dimocarpus longan Lour.) is an important subtropical fruit tree in China. Nearly 90% of longan fruit imports from Thailand are from the cultivar Yiduo. However, we have observed that there exists a unilateral cross incompatibility (UCI) when Yiduo is used as a female parent and Shixia (a famous Chinese cultivar) as a male parent. Here, we performed a comparative transcriptome analysis coupled with microscopy of pistils from two reciprocal pollination combinations [Shixia♂ × Yiduo♀(SY) and Yiduo♀ × Shixia♂(YS)] 4, 8, 12, and 24 h after pollination. We also explored endogenous jasmonic acid (JA) and jasmonyl isoleucine (JA-Ile) levels in pistils of the crosses. The microscopic observations showed that the UCI was sporophytic. The endogenous JA and JA-Ile levels were higher in YS than in SY at the studied time points. We found 7,251 differentially expressed genes from the transcriptome analysis. Our results highlighted that genes associated with JA biosynthesis and signaling, pollen tube growth, cell wall modification, starch and sucrose biosynthesis, and protein processing in endoplasmic reticulum pathways were differentially regulated between SY and YS. We discussed transcriptomic changes in the above-mentioned pathways regarding the observed microscopic and/or endogenous hormone levels. This is the first report on the elaboration of transcriptomic changes in longan reciprocal pollination combination showing UCI. The results presented here will enable the longan breeding community to better understand the mechanisms of UCI.

Key Genes in the JAZ Signaling Pathway Are Up-Regulated Faster and More Abundantly in Caterpillar-Resistant Maize

Jasmonic acid (JA) and its derivatives, collectively known as jasmonates (JAs), are important signaling hormones for plant responses against chewing herbivores. In JA signaling networks, jasmonate ZIM-domain (JAZ) proteins are transcriptional repressors that regulate JA-modulated downstream herbivore defenses. JAZ repressors are widely presented in land plants, however, there is only limited information about the regulation/function of JAZ proteins in maize. In this study, we performed a comprehensive expression analysis of ZmJAZ genes with other selected genes in the jasmonate pathway in response to feeding by fall armyworm (Spodoptera frugiperda, FAW), mechanical wounding, and exogenous hormone treatments in two maize genotypes differing in FAW resistance. Results showed that transcript levels of JAZ genes and several key genes in JA-signaling and biosynthesis pathways were rapidly and abundantly expressed in both genotypes in response to these various treatments. However, there were key differences between the two genotypes in the expression of ZmJAZ1 and ZmCOI1a, these two genes were expressed significantly rapidly and abundantly in the resistant line which was tightly regulated by endogenous JA level upon feeding. For instance, transcript levels of ZmJAZ1 increase dramatically within 30min of FAW-fed Mp708 but not Tx601, correlating with the JA accumulation. The results also demonstrated that wounding or JA treatment alone was not as effective as FAW feeding; this suggests that insect-derived factors are required for optimal defense responses.

Disclosure of salicylic acid and jasmonic acid-responsive genes provides a molecular tool for deciphering stress responses in soybean

Hormones orchestrate the physiology of organisms. Measuring the activity of defense hormone-responsive genes can help understanding immune signaling and facilitate breeding for plant health. However, different from model species like Arabidopsis, genes that respond to defense hormones salicylic acid (SA) and jasmonic acid (JA) have not been disclosed in the soybean crop. We performed global transcriptome analyses to fill this knowledge gap. Upon exogenous application, endogenous levels of SA and JA increased in leaves. SA predominantly activated genes linked to systemic acquired resistance and defense signaling whereas JA mainly activated wound response-associated genes. In general, SA-responsive genes were activated earlier than those responding to JA. Consistent with the paradigm of biotrophic pathogens predominantly activating SA responses, free SA and here identified most robust SA marker genes GmNIMIN1, GmNIMIN1.2 and GmWRK40 were induced upon inoculation with Phakopsora pachyrhizi, whereas JA marker genes did not respond to infection with the biotrophic fungus. Spodoptera exigua larvae caused a strong accumulation of JA-Ile and JA-specific mRNA transcripts of GmBPI1, GmKTI1 and GmAAT whereas neither free SA nor SA-marker gene transcripts accumulated upon insect feeding. Our study provides molecular tools for monitoring the dynamic accumulation of SA and JA, e.g. in a given stress condition.

Application of jasmonic acid at the stage of visible brown necrotic spots in Magnaporthe oryzae infection as a novel and environment-friendly control strategy for rice blast disease.

Rice blast disease is one of the most common rice diseases worldwide. It is essential to improve disease resistance through environment-friendly methods, while maintaining yield and quality parameters. In this study, jasmonic acid (JA), a plant hormone with anti-fungal activity, was obtained, at both low (100 μmol/L) and high (400 μmol/L) concentrations in rice leaves, before, during, and after infection, respectively. JA could inhibit germination and appressorium formation of rice blast spores in a dose-dependent manner. A total of 400-μmol/L JA treatment significantly enhanced cell viability and endogenous JA level in rice leaves. Furthermore, rice leaves inoculated with Magnaporthe oryzae and sprayed with JA 72 h post-inoculation showed the maximum symptom relief and the highest endogenous JA production among all treatment approaches. The expressions of defense-related genes, OsPR10a and OsAOS2, were highly up-regulated in response to JA, whereas OsEDS1 was down-regulated. Hence, we revealed that exogenous JA could activate JA signaling to effectively control the symptoms of rice blast.

Abscisic acid in preservation of Taraxacum pieninicum in the form of synthetic seeds in slow growth conditions

Taraxacum pieninicum Pawł. is listed as critically endangered species, for which currently applied protection methods are insufficient. The aim of this study was to investigate the possibility of T. pieninicum storage in the form of synthetic seeds under slow-growth conditions in combination with ABA treatment, as one of the ex situ protection methods of this species. The obtained results indicated that darkness was much more favorable condition for synseed storage and did not generate additional stress during cold exposure in contrast to the light conditions. The preculture of shoot tips on the medium supplemented with ABA led to a decrease in the shoots proliferation rate and inhibition of their growth. ABA clearly inhibited growth of the encapsulated shoot tips also during cold storage. Biochemical parameters showed that ABA effectively reduced the negative effect of the cold stress, what was found on the basis of analyzes of H2O2 and TBARS levels in the stored material. Moreover, synseeds stored under light conditions and treated with ABA exhibited decreased level of endogenous jasmonic acid what indicated interaction between those two phytohormones at a low temperature. The study also demonstrated that in vitro culture, cold storage and ABA treatment had no effect on the flowering process of this species after acclimatization to ex vitro conditions.

The arbuscular mycorrhizal mycelium from barley differentially influences various defense parameters in the non-host sugar beet under co-cultivation.

The interactions between arbuscular mycorrhizal fungi (AMF) and non-host species are poorly studied. Particularly scarce is information on members of the Amaranthaceae/Chenopodiaceae family. Sugar beet (Beta vulgaris) plants were co-cultivated with a host species (Hordeum vulgare) in the presence (+AMF) or absence of Rhizophagus intraradices to explore the hypothesis that the presence of an active, pre-established AMF mycelium induces defense responses in the non-host species. Biomass of sugar beet did not respond to the +AMF treatment, while its root exudation of organic acids and phenolic acids was drastically decreased upon co-cultivation with +AMF barley. The most conspicuous effect was observed on a wide range of potential defense parameters being differentially influenced by the +AMF treatment in this non-host species. Antioxidant defense enzymes were activated and the level of endogenous jasmonic acid was elevated accompanied by nitric oxide accumulation and lignin deposition in the roots after long-term +AMF treatment. In contrast, significant reductions in the levels of endogenous salicylic acid and tissue concentration and exudation of phenolic acids indicated that AM fungus hyphae in the substrate did not induce a hypersensitive-type response in the sugar beet roots and downregulated certain chemical defenses. Our results imply that the fitness of this non-host species is not reduced when grown in the presence of an AMF mycelium because of balanced defense costs. Further studies should address the question of whether or not such modulation of defense pattern influences the pest resistance of sugar beet plants under field conditions.

The expression characteristics of methyl jasmonate biosynthesis–related genes in Cymbidium faberi and influence of heterologous expression of CfJMT in Petunia hybrida

Cymbidium faberi Rolfe (Orchidaceae) is an herbaceous plant native to China, where it has a long history of cultivation owing to its beautiful flower pattern and floral fragrance. Previously, we conducted a transcriptome analysis of the flower and vegetative buds to elucidate the mechanisms of flower development in C. faberi. In the present study, we found nine secondary metabolic pathways through the KEGG pathway database that were related to the biosynthesis of methyl jasmonate (MeJA) and other volatile organic compounds. qRT-PCR was performed to analyze the expression levels of four key genes in the MeJA pathway. Among these, CfJMT (jasmonic acid carboxyl methyltransferase) had higher transcript levels in sepals, petals and labella than in other tissues. CfJMT was cloned from the petals of full-bloom flowers of C. faberi. The predicted CfJMT protein sequence contains conserved jasmonic acid methyl transferase-7 domains, indicating that it belongs to the SABATH protein family. The CfJMT coding sequence driven by the CaMV35S promoter was successfully transformed into Petunia hybrida through an Agrobacterium-mediated method. Although MeJA could not be detected in either wild-type or transgenic petunia plants, the leaves of the transgenic plants were smaller than those of wild-type plants and pollen development was abnormal. These results indicate that heterologous expression of CfJMT may change the levels of endogenous jasmonic acid and other hormones, but that the content of MeJA is not increased significantly by transformation with CfJMT alone. Thus, other related genes and regulation factors may play important roles in this process.

The transcription factor VaNAC17 from grapevine (Vitis amurensis) enhances drought tolerance by modulating jasmonic acid biosynthesis in transgenic Arabidopsis.

Expression of VaNAC17 improved drought tolerance in transgenic Arabidopsis by upregulating stress-responsive genes, modulating JA biosynthesis, and enhancing ROS scavenging. Water deficit severely affects the growth and development of plants such as grapevine (Vitis spp.). Members of the NAC (NAM, ATAF1/2, and CUC2) transcription factor (TF) family participate in drought-stress-induced signal transduction in plants, but little is known about the roles of NAC genes in drought tolerance in grapevine. Here, we explored the role of VaNAC17 in Vitis amurensis, a cold-hardy, drought-tolerant species of grapevine. VaNAC17 was strongly induced in grapevine by drought, exogenous abscisic acid (ABA), and methyl jasmonate (MeJA). A transient expression assay in yeast indicated that VaNAC17 functions as a transcriptional activator. Notably, heterologous expression of VaNAC17 in Arabidopsis thaliana enhanced drought tolerance. VaNAC17-expressing Arabidopsis plants showed decreased reactive oxygen species (ROS) accumulation compared to wild-type plants under drought conditions. RNA-seq analysis indicated that VaNAC17 expression increased the transcription of downstream stress-responsive genes after 5days of drought treatment, especially genes involved in jasmonic acid (JA) biosynthesis (such as LOX3, AOC1 and OPR3) and signaling (such as MYC2, JAZ1, VSP1 and CORI3) pathways. Endogenous JA levels increased in VaNAC17-OE plants under drought stress. Taken together, these results indicate that VaNAC17 plays a positive role in drought tolerance by modulating endogenous JA biosynthesis and ROS scavenging.

Colonization of endophyte Acremonium sp. D212 in Panax notoginseng and rice mediated by auxin and jasmonic acid.

Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms underlying colonization of Acremonium spp. remain unclear. In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng, enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin biosynthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid (IAA) and jasmonic acid (JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. notoginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate (MeJA) (2-15 μmol/L) and 1-naphthalenacetic acid (NAA) (10-20 μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and miR393b-overexpressing lines, and the colonization was rescued by MeJA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. D212 in host plants.

Potential of jasmonic acid (JA) in accelerating postharvest yellowing of broccoli by promoting its chlorophyll degradation

Chlorophyll degradation is the main reason for postharvest yellowing of broccoli. To uncover the role of jasmonic acid (JA) on the degradation of chlorophyll, broccoli flowers were treated with exogenous methyl jasmonate (MeJA) and diethyldithiocarbamic acid (DIECA). We found a surge of endogenous JA content with the yellowing process, and a significant correlation between JA and chlorophyll content. MeJA treatments led to increased endogenous JA, increased allene oxide cyclase (AOC) activity, and enhanced expression of JA synthesis genes. MeJA caused a stronger reduction in the maximum quantum yield (Fv/Fm), fluorescence decline ratio (Rfd), and total chlorophyll content, advanced the peak of pheide a oxygenase (PAO) activity, and up-regulated the expression of chlorophyll degradation genes. The DIECA treatment resulted in lower endogenous levels of JA, and AOC and 12-oxo-phytodienoic acid reductase (OPR) activity. This study revealed that the potential role of JA on broccoli yellowing is to promote the chlorophyll degradation.

Microbial Volatile Organic Compounds Produced by Bacillus amyloliquefaciens GB03 Ameliorate the Effects of Salt Stress in Mentha piperita Principally Through Acetoin Emission

We investigated the effects of microbial volatile organic compounds (mVOC) emitted by Bacillus amyloliquefaciens GB03 on Mentha piperita growing under different levels of NaCl stress, by evaluating their growth-promoting potential and ability to increase salt tolerance effects. Plants were exposed to bacterial VOCs without having any physical contact with the rhizobacteria. The VOCs emitted by the rhizobacteria (mVOCs) were analyzed using SPME fibers. An increase in the level of salt concentration led to a decrease in plant growth. However, these negative effects of salinity were inhibited in the plants exposed to mVOCs. Plants grown in a saline media and exposed to GB03 VOCs had significantly better morphological characteristics and higher total chlorophyll content compared to controls. The level of endogenous jasmonic acid (JA), salicylic acid, and abscisic acid increased in salt-stressed plants compared to controls. The level of JA did not show any change in plants grown in a saline media either exposed to mVOCs or not. In contrast, the amount of salicylic acid increased remarkably in salt-stressed plants exposed to mVOCs compared to controls (salt-stressed plants not exposed to mVOCs), but the levels of abscisic acid decreased in salt-stressed plants exposed to mVOCs. The chromatographic analyses of the mVOCs produced by salt-stressed GB03 bacteria were similar, regardless of the concentration of salt in the media where the bacteria were grown, although it was observed that the relative percentage of acetoin increased with salt concentration. After determining that acetoin was the main VOCs compound, we exposed plants to acetoin, which demonstrated that acetoin caused similar effects on plants grown under salt stress conditions as those exposed to GB03 mVOCs. Based on these results, the use of mVOCs from PGPR is suggested as a useful technological innovation to facilitate the growth of M. piperita in salt-stressed environments.
