Propagation of Yaba virus in embryonated hen eggs

The effect of trachoma virus vaccine on the course of experimental trachoma infection in blind human volunteers.

The TRIC-Taiwan-1-1958 strain of elementary body virus isolated from a trachoma patient on Taiwan has been proven capable of reproducing trachoma by experimental inoculation of six human volunteers. Virus material derived from the seventh passage in embryonated hen eggs caused the clinical picture of trachoma in every inoculation, even at the dilution of 10(-4) of infected yolk sacs (approximately 1 EID(50)). There was a similar clinical picture with each inoculation beginning with an acute follicular conjunctivitis which progressed for 4 months and then persisted with chronic changes until 9 months when treatment was begun. The illness was generally more acute than would be expected in natural trachoma. That trachoma was reproduced was shown by the involvement of the cornea with epithelial keratitis and pannus, and by the occurrence of gelatinous follicles and eventual cicatrization of the conjunctiva. These clinical findings were supported by repeated demonstrations of typical inclusion bodies of Halberstaedter-Prowazek from conjunctival and even corneal cells, by repeated reisolation of elementary body virus in egg yolk sacs, and by the development of complement-fixing antibody with a "specific" trachoma antigen in each volunteer. Control inoculations with adenovirus type 4 and normal yolk sac showed different clinical and laboratory findings. Experimental trachoma vaccine was given to three of the volunteers to study its effect on the course of illness. An antibody response to the vaccine was demonstrated and there was a modification of disease in the volunteers receiving vaccine. While the three volunteers who received placebo each developed cross-infection of their uninoculated eye and had an acute reactivation of the bilateral disease after 1 to 2 months of antibiotic eye ointment therapy, the vaccinated volunteers remained free of infection in uninoculated eyes and showed no relapse after ointment therapy. Treatment with sulfamethoxypyridazine, a sulfa drug with prolonged action, proved to be an effective and relatively simple method of therapy for experimental trachoma.

The propagation and preservation of Eperythrozoon coccoides.

SUMMARY: The propagation of Eperythrozoon coccoides in embryonated hen eggs inoculated by the yolk sac and intravenous routes is recorded. Fourteen continuous serial passages were made in the yolk sac and sixteen passages were made after intravenous inoculation into the chick embryo. The mortality among infected embryos was low; splenic enlargement was the only lesion detected. Once the strain had become established. E. coccoides was seen in blood smears taken from some eggs after each passage. Yolk sac or blood from infected embryos proved to be infective for mice on each occasion they were examined; 10−6 dilutions of yolk sac were also infective. E. coccoides was preserved for short periods at − 30° in mouse and chick- embryo blood, but not in yolk sac. The organism was also preserved in mouse blood and chick-embryo tissues at −79°. The presence of 10% (v/v) sterile glycerol appeared to increase the stability of E. coccoides in chick-embryo tissues stored at −79°.

Blood volume and red cell concentration in the normal chick embryo.

A method for the determination of red cell mass, which employs the use of radioisotopically-labeled erythrocytes, in the embryonated hen egg is described and results of total blood volume, plasma volume, red cell mass and red cell concentration determinations during the period from 9 to 18 days of incubation presented. Considerations of the values obtained in terms of absolute quantities and relative to embryonic mass are discussed. It is suggested that expansion of the red cell mass and somatic growth are portions of a common process and may be subject to a complex of similar regulatory factors in the normal chick embryo.

SummaryMengo encephalomyelitis (ME) virus was readily propagated in the embryonated hen egg. Infected embryos showed extensive haemorrhages and were usually dead within 48 to 96 hours after inoculation. Sections showed widespread degenerative changes and haemorrhages throughout the entire embryo. The embryo was susceptible to inoculation of ME virus by the amniotic, yolk sac, and allantoic routes and onto the “dropped” chorio-allantoic membrane. After amniotic inoculation, there was a slight lag in virus multiplication, after which rapid multiplication occurred and titers of virus of 10-7 were obtained in suspensions of whole embryos by 48 hours after inoculation. Similar titers were obtained in suspensions of whole embryos and yolk sacs after inoculation by the amniotic or yolk sac routes. The embryo was less susceptible to inoculations by the allantoic route. SummaryStudies are reported in which the Lederle stock strain of canine distemper virus was adapted and maintained through 74 serial passages on the chorio-allantoic membranes of developing chick embryos. The chick embryo adapted strain retained its capacity to produce lethal infection in ferrets through the 24th egg passage level but apparently lost its ability to do so between the 24th and 28th egg passages. Ferrets inoculated with chick embryo adapted virus of passage levels higher than the 28th transfer failed to show any signs of disease, but were found immune on rechallenge with fully virulent ferret spleen virus. Cross neutralization tests carried out in chick embryos and in ferrets indicate that the chick embryo adapted strain and the parent strain of ferret spleen virus are immunologically related.
