Proteomic insights into adventitious root formation in Larix kaempferi

The adventitious root formaton (ARF) in excised plant parts is essential for the survival of isolated plant fragments. In this study, we explored the complex mechanisms of ARF in Larix kaempferi by conducting a comprehensive proteomic analysis across three distinct stages: the induction of adventitious root primordia (C1, 0–25 d), the formation of adventitious root primordia (C2, 25–35 d), and the elongation of adventitious roots (C3, 35–45 d). We identified 1976 proteins, with 263 and 156 proteins exhibiting increased abundance in the C2/C1 and C3/C2 transitions, respectively. In contrast, a decrease in the abundance of 106 and 132 proteins suggests a significant demand for metabolic processes during the C2/C1 phase. The abundance of IAA-amino acid hydrolase and S-adenosylmethionine synthase were increased in the C2/C1 phase, underscoring the role of auxin in adventitious root induction. The decrease in abundance of photosynthesis-related proteins during the C2/C1 phase highlights the significance of initial light conditions in adventitious root induction. Moreover, variation in cell wall synthesis and metabolic proteins in the C2/C1 and C3/C2 stages suggests that cell wall metabolism is integral to adventitious root regeneration. Gene Ontology enrichment analysis revealed pathways related to protein modification enzymes, including deubiquitinases and kinases, which are crucial for modulating protein modifications to promote ARF. Furthermore, the increased abundance of antioxidant enzymes, such as peroxidases and glutathione peroxidases, indicates a potential approach for enhancing ARF by supplementing the culture medium with antioxidants. Our study provides insights into metabolic changes during ARF in L. kaempferi, offering strategies to enhance adventitious root regeneration. These findings have the potential to refine plant propagation techniques and expedite breeding processes. SignficanceThe main challenge in the asexual reproduction technology of Larix kaempferi lies in adventitious root formation (ARF). While numerous studies have concentrated on the efficiency of ARF, proteomic data are currently scarce. In this study, we collected samples from three stages of ARF in L. kaempferi and subsequently performed proteomic analysis. The data generated not only reveal changes in protein abundance but also elucidate key metabolic processes involved in ARF. These insights offer a novel perspective on addressing the challenge of adventurous root regeneration.

Identification and Expression Profile Analysis of WOX Family Genes in the Formation of Eucalyptus Adventitious Root

The WUSCHEL-related homeobox (WOX) gene family are key players in the rooting process. Eucalyptus is an important plant species of artificial forests in China. It is mainly grown through tissue culture of many excellent clonal materials, in which rooting is a key step. In the present study, by using the genome data of Eucalyptus grandis, Corymbia citriodora, E. pellita, and E. urophylla × E. grandis, the members of the eucalyptus WOX gene family were identified and analyzed by bioinformatics techniques. The eucalyptus WOX gene family members are unstable proteins, with 7 acidic proteins and 24 basic proteins, and no signal peptide region was detected. Subcellular localization prediction indicated that all these proteins are localized in the nucleus. Motif analysis showed that eucalyptus WOX genes share the same motifs. Phylogenetic tree and gene expression analyses revealed that the eucalyptus WOX genes are highly conserved during the evolution process. Moreover, the WOX protein sequences are also highly conserved within the species, with higher similarity between woody plants. The EupWOX gene showed tissue-specific expression, with EupWOX1 and EupWOX11 specifically expressed in the roots of E. urophylla × E. pellita clonal tissue culture during the late-stage rooting. This finding suggests that EupWOX1 may be a key regulatory gene induced by the root primordium and is critically related to the rooting rate. EupWOX1, EupWOX5, and EupWOX13 could be the key regulatory genes for adventitious root formation. EupWOX1, EupWOX5, and EupWOX13 could be the key regulatory genes for the elongation of adventitious roots and the growth of adventitious lateral roots. EupWOX5 and EupWOX13 could play a critical role, not only in the formation of adventitious roots and adventitious lateral roots of E. urophylla clonal tissue culture but also in the root elongation process. These results will help us understand the complexity of rooting in different lines and provide valuable information for future functional characterization of specific genes in eucalyptus clones.

Insight into regulation of adventitious root formation by arbuscular mycorrhizal fungus and exogenous auxin in tea plant (Camellia sinensis L.) cuttings.

Adventitious root (AR) development, affected by various biotic and abiotic factors, is the most important procedure in tea plant (Camellia sinensis L.) cutting propagation. Establishing symbiotic relationships with most terrestrial plants, AMF (Arbuscular mycorrhizal fungus) can mediate the AR formation of several herbaceous and woody plants in previous studies. In this paper, effects of combined application of AMF and exogenous auxin on AR formation of cuttings from different tea plant varieties ('Pingyangtezao', 'Longjing 43' and 'Longjingchangye') were studied. Then we also performed RNA-Seq analysis with 'Pingyangtezao' cuttings aiming to find the possible auxin-related pathway of AM fungal regulation on AR formation. To accurately uncover the regulatory mechanism of AMF on AR formation of tea cuttings, rooting process were separated into four stages (S0, non-rooting; S1, AR protrusion; S2, AR formation and S3, AR elongation) at the same sampling time. Results showed that IBA treatment increased the mycorrhizal colonization rate, especially in 'Pingyangtezao' variety (from 37.58% to 46.29%). Both inoculating AMF and addition of IBA promoted the AR formation, and rooting of different tea plant varieties showed different dependence on auxin. AMF could alleviate the effect of auxin-related inhibitors (2,3,5-triiodobenzoic acid, L-α-(Aminooxy)-β-phenylpropionic acid and α-(phenylethyl-2-oxo)-IAA) on rooting of tea cuttings, even though the colonization of AMF was hindered at various degrees. Transcriptomic analysis showed that different numbers of differentially expressed genes (DEGs) at various rooting stages of tea cuttings with the most at S2 stage (1360 DEGs), indicating the increasing regulation by AMF with the development of AR. Similar trend was found in auxin-related DEGs, and family genes of YUC, GH, PIN, LAX, SAUR, AUX, and ABP involved in the AM fungal regulation on AR formation of tea cuttings. Additionally, AMF strongly mediated auxin transport and signal transduction pathways in tea cuttings as showed by the results of correlation analysis. Overall, interaction of AMF and exogenous auxin in promoting rooting and the preliminary mechanism of AMF regulating AR formation of tea cuttings was deciphered in this paper, which may provide a basis for further deep mechanistic research and cutting propagation of tea production.

Phytotoxicity of microplastics to the floating plant Spirodela polyrhiza (L.): Plant functional traits and metabolomics

Freshwater ecosystems are gradually becoming sinks for terrestrial microplastics (MPs), posing a potential ecological risk. Although the effects of MPs on plankton and aquatic animals in freshwater ecosystems have been given increasing attention, the toxicity of MPs to the metabolism of aquatic plants remains unclear. Here, the model aquatic plant Spirodela polyrhiza (L.) Schleid. (S. polyrhiza) was exposed to polyvinyl chloride (PVC; 0, 10, 100 and 1000 mg/L) MPs, and changes in the plant functional traits and physiological metabolism were monitored. The results showed that the high dose of PVC MPs decreased the adventitious root elongation ratio by 41.68% and leaf multiplication ratio by 61.03% of S. polyrhiza, and resulted in the decrease in anthocyanin and nitrogen contents to 63.45% and 84.21% of the control group, respectively. Moreover, the widely targeted metabolomics analysis results showed 37 differential metabolites in the low-dose treatment and 119 differential metabolites in the high-dose treatment. PVC MPs interfered with organic matter accumulation by affecting carbon metabolism, nitrogen metabolism, amino acid metabolism and lipid metabolism, and S. polyrhiza resists PVC MP stress by regulating the synthesis and metabolism of secondary metabolites. PVC MPs had concentration-related toxicological effects on plant functional traits, inhibited plant growth and reproduction, affected plant nutrient metabolism, and exhibited profound effects on the nitrogen fate of aquatic plant habitats. Overall, we systematically summarized the metabolic response mechanisms of aquatic plants to PVC MP stress, providing a new perspective for studying the effects of MPs on plant trait function and ecological risks.

Ortet Age Effect, Anatomy and Physiology of Adventitious Rooting in Tilia mandshurica Softwood Cuttings

Tilia mandshurica is recognized as one of the most valuable timber, ornamental and nectariferous tree species, and its cutting propagation is very difficult. To evaluate the influence of ortet age on the rooting of T. mandshurica cuttings and the relationship between the variations of nutrients, enzyme activities, endogenous hormones and the formation of adventitious roots, a series of softwood cuttings of T. mandshurica were collected from 2-, 7-, 15- and 20-year-old healthy ortets, respectively. The rooting percentage, mean root number and total root length were investigated after 60 days of cultivation. Phenotypic and anatomical changes and the variations of organic nutrients, enzyme activities and endogenous hormones were measured during adventitious root formation. The results showed that ortet age effect existed in T. mandshurica softwood cuttings. The adventitious rooting of 2-year-old ortets was superior to that of 7-, 15- and 20-year-old ortets. No root primordium was visible in T. mandshurica softwood cuttings before cultivation, and the root primordia were induced after planting. The adventitious roots derived from vascular cambium and callus in cuttings. Soluble sugar increased during root primordium induction and decreased in adventitious root elongation. Soluble protein decreased during root primordium induction and increased in adventitious root elongation. Polyphenol oxidase (PPO) activity increasing and peroxidase (POD) and 3-indoleacetic acid oxidase (IAAO) activities decreasing were conducive to root primordium induction. High 3-indoleacetic acid (IAA) and gibberellin (GA3) level could promote root primordium induction and low IAA and GA3 level was beneficial to adventitious root elongation. Abscisic acid (ABA) and zeatin riboside (ZR) inhibited root primordium induction and adventitious root elongation. IAA, ABA and ZR coordinated with each other in the process of root primordium induction and adventitious root formation. IAA/ABA increasing could promote root primordium induction, and IAA/ZR increasing was beneficial to root primordium induction and adventitious root elongation. The soluble sugar content, PPO activity, IAA/ABA and IAA/ZR of softwood cuttings from 2-year-old ortets were relatively high, which may be the reason for the better adventitious rooting. The present study provides a reference to build a propagation by cuttings technology of linden trees.

STUDY ON in vitro INDUCTION OF ROOTING AND CHANGES IN ENDOGENOUS HORMONE CONTENT OF Lagerstroemia indica ‘ZIJINGLING’

Different media had different effects on the induction of rooting of Lagerstroemia indica ‘Zijingling’. The aims of this study were to identify the best rooting medium, determine the changes in endogenous hormone content in aseptic seedlings of ‘Zijingling’, and then analyze its role in the rooting process, to improve the rooting rate. Using a test tube seedling of ‘Zijingling’ as the experimental material, the tissue cells were observed via paraffin sectioning, and the changes in endogenous hormone content during the rooting process were determined using high-performance liquid chromatography. The results showed that 1/2 MS Medium had the most significant effect on rooting in the basic medium. The promoting effects of different auxins on rooting decreased in magnitude in the order 3-Indolebutyric acid (IBA) > Naphthalene acetic acid (NAA)> Indole-3-aceticacid (IAA). The optimal rooting medium was 1/2 MS + 0.6 mg L-1 IBA+ 15 g L-1 sucrose + 5 g L-1 agar + 200 mg L-1 activated carbon , and the highest induction rate of adventitious roots was 92.5%. The rooting of ‘Zijingling’ is classified as a primordial type of induced rooting. Exogenous IBA content promoted an increase in endogenous IAA and Gibberellic acid (GA3) contents. High contents of IAA, GA3, and Abscisic acid (ABA) and low content of Zeatin riboside (ZR) promoted the growth of adventitious roots, whereas high contents of IAA and ZR, and low contents of GA3 and ABA were required for the induction of root primordia. High levels of IAA/ZR and low levels of IAA/ABA promoted the differentiation of root primordia. However, low levels of IAA/ZR and high levels of IAA/ABA promoted adventitious root elongation.

Auxin and Its Interaction With Ethylene Control Adventitious Root Formation and Development in Apple Rootstock.

Adventitious root (AR) formation is indispensable for vegetative asexual propagation. Indole-3-butyric acid (IBA) functioned indirectly as precursor of IAA in regulating AR formation. Ethylene affects auxin synthesis, transport, and/or signaling processes. However, the interactions between auxin and ethylene that control AR formation in apple have not been elucidated. In this study, we investigated the effects of IBA and its interaction with ethylene on AR development in apple. The results revealed that IBA stimulated the formation of root primordia, increased the number of ARs, and upregulated expression of genes (MdWOX11, MdLBD16, and MdLBD29) involved in AR formation. Comparison of different periods of IBA application indicated that IBA was necessary for root primordium formation, while long time IBA treatment obviously inhibited root elongation. RNA-seq analysis revealed that many plant hormone metabolism and signal transduction related genes were differentially expressed. IBA stimulated the production of ethylene during AR formation. Auxin inhibiting ARs elongation depended on ethylene. Together, our results suggest that the inhibitory role of auxin on AR elongation in apples is partially mediated by stimulated ethylene production.

Nitric oxide alleviates cadmium- but not arsenic-induced damages in rice roots

Nitric oxide (NO) has signalling roles in plant stress responses. Cadmium (Cd) and arsenic (As) soil pollutants alter plant development, mainly the root-system, by increasing NO-content, triggering reactive oxygen species (ROS), and forming peroxynitrite by NO-reaction with the superoxide anion. Interactions of NO with ROS and peroxynitrite seem important for plant tolerance to heavy metal(oid)s, but the mechanisms underlying this process remain unclear. Our goal was to investigate NO-involvement in rice (Oryza sativa L.) root-system after exposure to Cd or As, to highlight possible differences in NO-behaviour between the two pollutants. To the aim, morpho-histological, chemical and epifluorescence analyses were carried out on roots of different origin in the root-system, under exposure to Cd or As, combined or not with sodium nitroprusside (SNP), a NO-donor compound. Results show that increased intracellular NO levels alleviate the root-system alterations induced by Cd, i.e., inhibition of adventitious root elongation and lateral root formation, increment in lignin deposition in the sclerenchyma/endodermis cell-walls, but, even if reducing As-induced endodermis lignification, do not recover the majority of the As-damages, i.e., enhancement of AR-elongation, reduction of LR-formation, anomalous tissue-proliferation. However, NO decreases both Cd and As uptake, without affecting the pollutants translocation-capability from roots to shoots. Moreover, NO reduces the Cd-induced, but not the As-induced, ROS levels by triggering peroxynitrite production. Altogether, results highlight a different behaviour of NO in modulating rice root-system response to the toxicity of the heavy metal Cd and the metalloid As, which depends by the NO-interaction with the specific pollutant.

Sugar enhances waterlogging-induced adventitious root formation in cucumber by promoting auxin transport and signalling.

Waterlogging is a severe environmental stress that causes severe crop productivity losses. Cucumber (Cucumis sativus L.) survives waterlogging by producing adventitious roots (ARs) that enhance gas exchange. Little is known about the role of light and sugars in the waterlogging-induced production of ARs. The role of these factors in AR production was therefore studied in cucumber seedlings grown in the absence or presence of waterlogging and different light conditions. The effect of photosynthesis was studied by removing the shoots of the seedlings and replacing them with exogenous applications of sucrose or stachyose. Shoot removal inhibited AR emergence and elongation. However, the exogenous application of sugars fully restored AR emergence and partially restored root elongation. The exogenous application of a synthetic auxin restored AR emergence but not AR elongation. Transcriptome profiling analysis was used to determine the effects of light on gene expression in the hypocotyls under these conditions. The levels of transcripts encoding proteins involved in auxin transport and signalling were higher in the light and following the exogenous application of sucrose and stachyose. These results show that the waterlogging-induced emergence of ARs is regulated by the interaction between sugars and auxin, whereas AR elongation depends only on sugars alone.

Hypoxia and the group VII ethylene response transcription factor HRE2 promote adventitious root elongation in Arabidopsis.

Soil water‐logging and flooding are common environmental stress conditions that can impair plant fitness. Roots are the first organs to be confronted with reduced oxygen tension as a result of flooding. While anatomical and morphological adaptations of roots are extensively studied, the root system architecture is only now becoming a focus of flooding research. Adventitious root (AR) formation shifts the root system higher up the plant, thereby facilitating supply with oxygen, and thus improving root and plant survival.We used Arabidopsis knockout mutants and overexpressors of ERFVII transcription factors to study their role in AR formation under hypoxic conditions and in response to ethylene.Results show that ethylene inhibits AR formation. Hypoxia mainly promotes AR elongation rather than formation mediated by ERFVII transcription factors, as indicated by reduced AR elongation in erfVII seedlings. Overexpression of HRE2 induces AR elongation to the same degree as hypoxia, while ethylene overrides HRE2‐induced AR elongation.The ERFVII transcription factors promote establishment of an AR system that is under negative control by ethylene. Inhibition of growth of the main root system and promotion of AR elongation under hypoxia strengthens the root system in upper soil layers where oxygen shortage may last for shorter time periods.

Selection by seed weight improves traits related to seedling establishment in Panicum coloratum L. var. makarikariense

AbstractSelection for heavy seed was implemented to overcome establishment problems in Panicum coloratum var. makarikariense. A collection covering the genetic diversity of the species available in Argentina was used as a base population (BP), and two cycles of recurrent phenotypic selection to increase seed weight were performed. Seedlings, obtained from seeds from each cycle and the BP, were further evaluated in the greenhouse. An increase of 17% in mean seed weight was obtained after two cycles of recurrent selection. No differences in germination or emergence were detected among the selected materials and the BP. However, seedlings from selected materials were taller and showed higher leaf elongation rate, adventitious roots elongation rate and total growth rate than that from BP. This research also revealed that in P. coloratum var. makarikariense, more vigorous seedlings can be selected from genotypes that produce heavier seeds.

Molecular mechanism of adventitious root formation in rice

Adventitious roots account for the majority of the rice root system and play an irreplaceable role in rice growth and development. Rice adventitious roots are formed by division of the innermost ground meristem cells in the central cylinder, and some lateral roots are observable in the adventitious root system. Multiple hormones have been implicated in the regulation of root development. Auxin is involved in the initiation of adventitious roots, whereas cytokinin inhibits adventitious root initiation, but promotes adventitious root elongation. Other phytohormones such as nitric oxide, ethylene, brassinosteroid, jasmonic acid and gibberellin may be also involved in regulating adventitious root initiation and development. Additionally, more than 600 root development related quantitative trait loci (QTLs) have been located by QTL analysis of root traits.

Growth and Anatomical Parameters of Adventitious Roots Formed on Mung Bean Hypocotyls Are Correlated with Galactoglucomannan Oligosaccharides Structure

The effect of galactoglucomannan oligosaccharides (GGMOs) compared with chemically modified oligosaccharides, GGMOs-g (with reduced number of D-galactose side chains) and GGMOs-r (with reduced reducing ends) on mung bean (Vigna radiata (L.) Wilczek) adventitious roots formation, elongation, and anatomical structure have been studied. All types of oligosaccharides influenced adventitious root formation in the same way: stimulation in the absence of exogenous auxin and inhibition in the presence of exogenous auxin. Both reactions are probably related with the presence/content of endogenous auxin in plant cuttings. However, the adventitious root length was inhibited by GGMOs both in the absence as well as in the presence of auxin (IBA or NAA), while GGMOs-g inhibition was significantly weaker compared with GGMOs. GGMOs-r were without significant difference on both processes, compared with GGMOs. GGMOs affected not only the adventitious root length but also their anatomy in dependence on the combination with certain type of auxin. The oligosaccharides influenced cortical cells division, which was reflected in the cortex area and in the root diameter. All processes followed were dependent on oligosaccharides chemical structure. The results suggest also that GGM-derived oligosaccharides may play an important role in adventitious roots elongation but not in their formation.

Genotypic Variation in Responses of Cassava (Manihot esculenta Crantz) to Drought and Rewatering: Root System Development

Soil moisture condition is a major factor that affects root system development and thus, crop production. This study aimed to evaluate genotypic variations of cassava in root system structures and their responses to different soil moisture conditions by examining various root traits including production and elongation of adventitious roots and their laterals. Four pot experiments were conducted and different genotypes of various backgrounds were grown under well-watered, droughted, droughted to rewatered conditions. One field experiment was also conducted with selected genotypes till maturity. Results showed that substantial genotypic variations exist in root system structure, and the effects of the soil moistures were significant for most of the root traits. The principal component analysis (PCA) showed that the lateral root development mainly accounted for the variations in root system structure regardless of soil moisture conditions. The PCA on the differences between droughted and well-watered control, and droughted-rewatered and the control further indicated that the branching ability of adventitious roots was mainly responsible for the root system responses to drought as well as rewatering. Genotypic ranking in root system responses to drought was almost consistent among the pots and field experiments. Genotypicvariations in rooting depth were relatively small while those in horizontal spread were apparent in the field experiment.The ability to maintain adventitious root elongation under drought, resulting in relatively large horizontal spread of root system and to recover sharply from drought by lateral root branching may be related to good growth and yield performance under field.

Adventitious Root Development and Ginsenoside Production in Panax ginseng, Panax quinquefolium and Panax japonicum

This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of <TEX>$NH_4NO_3$</TEX> from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with <TEX>$NH_4NO_3$</TEX>. Two-step culture protocol: <TEX>$NH_4NO_3$</TEX>-free medium for first two weeks of culture, followed by <TEX>$NH_4NO_3$</TEX> containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.

Effect of auxins on Karwinskia humboldtiana root cultures

Effects of auxins (IAA, IBA and NAA) on K. humboldtiana root culture cultivated in 16-h photoperiod or in dark have been observed. Light affected positively the production of biomass when cultivated on medium supplemented with NAA in 10 and 25 μ mol −1 concentrations. In the presence of IAA and IBA these values were significantly lower. The growth dynamics of root cultures depended on the auxin used. The best adventitious roots elongation and lateral roots induction on media supplemented with IBA has been ascertained. Morphological and anatomical differences in dependence on auxin used were observed. NAA supported the formation of huge callus-like mass besides mostly very short roots, especially under the light. Similarly IAA induced short roots, and IBA seems to be the most effective substance for the root elongation in this model system. NAA induced roots with larger diameter under the light compared with the other two auxins used. The reason is in the different anatomical structure of roots which was characterized by higher number of cell layers and large intercellulars in the cortex. The shape of cortical cells in the presence of IBA depended on the light conditions. Isodiametric cortical cells were present in roots cultivated in 16-h photoperiod, irregularly-shaped cells in the dark. The effect of light conditions was the smallest in the case of roots grown on IAA enriched media.

Practical Factors Controlling in vitro Adventitious Root Formation from Walnut Shoot Microcuttings

Various factors were found to influence the in vitro induction and elongation of adventitious roots from walnut shoot microcuttings. Diverse walnut genotypes (Juglans regia, J. nigra × J. regia hybrids) and selected elite J. regia clones were micropropagated throughout the establishment of in vitro shoot-tip cultures. New evidence is presented here that demonstrates the importance of the genotype and juvenility of the plant material on the in vitro rooting ability. Selection of the best adapted genotypes to multiplication and rooting, and rejuvenation of mature clones through repetitive subcultures or micrografting were examined. Adult J. regia clones were rejuvenated through subsequent subcultures and their rooting was consequently improved. The same results were not accomplished by micrografting on juvenile shoots. A differential response to auxin type and concentration was observed for Juglans regia or J. nigra × J. regia clones. A short prerooting culture in multiplication medium, lowering the sucrose concentration in the root elongation medium and increasing the atmospheric carbon dioxide during the root elongation phase affected the number of shoots forming roots as well as the quality of plantlets and roots.

An Hsp90 Inhibitor, Geldanamycin, as a Brassinosteroid Antagonist: Evidence from Salt‐Exposed Roots of Vigna radiata

Abstract: Treatment of non‐stressed mung bean cuttings with 24‐epibrassinolide (BR) inhibited elongation of adventitious roots, but it stimulated root elongation in plants exposed to 70 mM NaCl. These findings confirm that brassinosteroids are involved in control of division and elongation of root cells. We found that both positive and negative effects are counteracted by the presence of geldanamycin in the root medium. Because of the high specificity of geldanamycin for the chaperone Hsp90, this suggests that, as in animal cells, Hsp90 is involved in expression or transduction of the brassinosteroid signals in plants. Moreover, a detailed analysis of root ranking showed an additional geldanamycin‐insensitive mode of action of BR, especially observed under salt stress conditions. These two modes differently affected elongation of the adventitious roots, depended on the phases of differentiation and on the presence of an environmental constraint.

Effect of Wild-Type and Mutant Plant Growth-Promoting Rhizobacteria on the Rooting of Mung Bean Cuttings.

Mung bean cuttings were dipped in solutions of wild type and mutant forms of the plant growth-promoting rhizobacterium Pseudomonas putida GR12-2 and then incubated for several days until roots formed. The bacteria P. putida GR12-2 and P. putida GR12-2/aux1 mutant do not produce detectable levels of the enzyme 1-aminocyclopropane-1-carboxylate (ACC) deaminase, whereas P. putida GR12-2/acd36 is an ACC deaminase minus mutant. All bacteria produce the phytohormone indole-3-acetic acid (IAA), and P. putida GR12-2/aux1 overproduces it. Treatment of cuttings with the above-mentioned bacteria affected the rates of ethylene production in the cuttings in a way that can be explained by the combined effects of the activity of ACC deaminase localized in the bacteria and bacterial produced IAA. P. putida GR12-2 and P. putida GR12-2/acd36-treated cuttings had a significantly higher number of roots compared with cuttings rooted in water. In addition, the wild type influenced the development of longer roots. P. putida GR12-2/aux1 stimulated the highest rates of ethylene production but did not influence the number of roots. These results are consistent with the notion that ethylene is involved in the initiation and elongation of adventitious roots in mung bean cuttings.
