Land-use interactions, Oil-Field infrastructure, and natural processes control hydrocarbon and arsenic concentrations in groundwater, Poso Creek Oil Field, California, USA

Like many hydrocarbon production areas in the U.S., the Poso Creek Oil Field in California includes and is adjacent to other land uses (agricultural and other developed lands) that affect the hydrology and geochemistry of the aquifer overlying and adjacent to oil development. We hypothesize that the distributions of hydrocarbons and arsenic in groundwater in such areas will be controlled by complex interactions between mixed land uses, oil-field infrastructure, and natural processes. In 2020–2021, samples of groundwater and surface water were collected and analyzed for a large suite of inorganic and organic chemicals and isotope and gas tracers to test this hypothesis. Those data are supplemented with ancillary data on historical geochemistry, hydrology, geology, and oil-field infrastructure. Hydrocarbons in groundwater (e.g., methane through pentane gases and benzene) are associated with natural processes (e.g., fault offsets or transition in sediment depositional environment) and oil-field infrastructure (e.g., fluid-migration pathways associated with uncemented annulus in oil wells or unlined pits). Arsenic concentrations >10 μg per liter (μg/L; maximum concentration 12.9 μg/L) are associated with natural processes in old, high-pH groundwater, and more recent recharge of water from natural and/or engineered recharge processes. Along the southwest margin of the oil field, pumping for drinking-water and irrigation supplies in combination with engineered groundwater recharge produce a depression in groundwater elevations where groundwater with elevated sulfate concentrations from agricultural areas and groundwater with hydrocarbons from the oil field mix to produce a zone of sulfate reduction that removes hydrocarbons and arsenic from groundwater but produces elevated sulfide (S2-) concentrations (maximum concentration 29 mg per liter, mg/L). In this study, multiple approaches were required to resolve the overlapping effects of land uses, oil-field infrastructure, and natural processes on the distributions of hydrocarbons and arsenic in groundwater. The combined use of geographic, historical, physical, chemical, isotopic, and other information to constrain processes could be a useful approach for studies in other hydrocarbon-production areas. This is particularly important where land uses affect aquifer hydrology to an extent that causes mixing of groundwaters with different chemical compositions.

Simulation of the Modes of Medium Flow Movement through a Gas Pipeline during Corrosion Tests

Elevated concentrations of corrosive carbon dioxide or hydrogen sulfide in gas and gas condensate both produced and transported through pipelines lead to serious corrosion damage to the internal surfaces of steel infrastructure facilities. The paper presents the results of studying the corrosive effect of the medium flow along the lower component of the gas pipeline, which can exhibit a dynamic, intermittent or static character. During testing, the effect of both dynamic conditions of the medium flow on the U-shaped cell and static conditions of the permanent impact of the aqueous phase on the pipeline wall during the bubble test was evaluated. Modeling of variable wetting conditions inside the gas pipeline showed that such conditions are typical and occur upon production and transportation of raw gas to the places of gas processing and purification. We have simulated dangerous operational factors that occur inside the gas pipeline: the composition of the aquatic environment, temperature, and the content of corrosive gases. When determining the resistance of steels to local forms of corrosion (pitting, wide and shallow corrosion pits), we revealed that the rate of developing local and general corrosion of steel in aggressive carbon dioxide and hydrogen sulfide conditions can reach 2 – 3 mm/year. In addition, it has been shown that the use of corrosion inhibitors for protecting the equipment and pipelines of gas facilities can effectively prevent the occurrence of internal corrosion processes. The results obtained can be used in assessing the corrosion activity of operating media and selecting the most proven corrosion inhibitors for pilot testing at gas fields.

Effect of Sulfide on the Processes of Transformation of Nitrogen Compounds and the Microbial Community Composition in the Anammox Bioreactor

Anammox is one of the most important processes in the global nitrogen cycle and the basis for an efficient technology of nitrogen removal from wastewater. The effect of the presence of sulfide in wastewater on the transformation of nitrogen compounds by the anammox community has been insufficiently studied. The present work dealt with the effect of sulfide on nitrogen removal efficiency and the dynamics of nitrogen species in a laboratory sequencing batch bioreactor modeling the functioning of the anammox community carrying out ammonium oxidation via nitritation and anammox and nitrite oxidation. The 16S rRNA gene profiling of the community of the anammox-activated sludge attached to the stationary carrier revealed members of the key physiological groups: ammonium oxidizers of the genus Nitrosomonas, nitrite oxidizers of the genus Nitrosospira, and anammox bacteria of the genera Candidatus Brocadia and Ca. Jettenia, as well as members of other bacterial genera. Nitrate removal was not sensitive to sulfide at concentrations up to 50 mg S/L and decreased by 17% at 100 mg/L. The threshold of sulfide sensitivity for group I nitrifiers was ~50 mg/L, while anammox bacteria were resistant to sulfide concentrations of up to 100 mg S/L in the incoming water. Group II nitrifiers were probably the most sulfide-sensitive components of the community. A drastic increase in the abundance of members of the family Hydrogenophilaceae at elevated sulfide concentrations, together with the precipitation of elemental sulfur, may indicate sulfide oxidation either by molecular oxygen or via nitrate reduction; this finding requires further investigation. This is the first report on the different effects of sulfide on the growth rate of members of the nitrifying genus Nitrosomonas, increasing/decreasing or not affecting it for different phylotypes at elevated sulfide concentrations.

Microbial survival mechanisms within serpentinizing Mariana forearc sediments.

Marine deep subsurface sediment is often a microbial environment under energy-limited conditions. However, microbial life has been found to persist and even thrive in deep subsurface environments. The Mariana forearc represents an ideal location for determining how microbial life can withstand extreme conditions including pH 10-12.5 and depleted nutrients. The International Ocean Discovery Program Expedition 366 to the Mariana Convergent Margin sampled three serpentinizing seamounts located along the Mariana forearc chain with elevated concentrations of methane, hydrogen, and sulfide. Across all three seamount summits, the most abundant transcripts were for cellular maintenance such as cell wall and membrane repair, and the most abundant metabolic pathways were the Entner-Doudoroff pathway and tricarboxylic acid cycle. At flank samples, sulfur cycling involving taurine assimilation dominated the metatranscriptomes. The in situ activity of these pathways was supported by the detection of their metabolic intermediates. All samples had transcripts from all three domains of Bacteria, Archaea, and Eukarya, dominated by Burkholderiales, Deinococcales, and Pseudomonales, as well as the fungal group Opisthokonta. All samples contained transcripts for aerobic methane oxidation (pmoABC) and denitrification (nirKS). The Mariana forearc microbial communities show activity not only consistent with basic survival mechanisms, but also coupled metabolic reactions.

Cysteine Biosynthesis in Campylobacter jejuni: Substrate Specificity of CysM and the Dualism of Sulfide.

Campylobacter jejuni is a highly successful enteric pathogen with a small, host-adapted genome (1.64 Mbp, ~1650 coding genes). As a result, C. jejuni has limited capacity in numerous metabolic pathways, including sulfur metabolism. Unable to utilise ionic sulfur, C. jejuni relies on the uptake of exogenous cysteine and its derivatives for its supply of this essential amino acid. Cysteine can also be synthesized de novo by the sole cysteine synthase, CysM. In this study, we explored the substrate specificity of purified C. jejuni CysM and define it as an O-acetyl-L-serine sulfhydrylase with an almost absolute preference for sulfide as sulfur donor. Sulfide is produced in abundance in the intestinal niche C. jejuni colonises, yet sulfide is generally viewed as highly toxic to bacteria. We conducted a series of growth experiments in sulfur-limited media and demonstrate that sulfide is an excellent sulfur source for C. jejuni at physiologically relevant concentrations, combating the view of sulfide as a purely deleterious compound to bacteria. Nonetheless, C. jejuni is indeed inhibited by elevated concentrations of sulfide and we sought to understand the targets involved. Surprisingly, we found that inactivation of the sulfide-sensitive primary terminal oxidase, the cbb3-type cytochrome c oxidase CcoNOPQ, did not explain the majority of growth inhibition by sulfide. Therefore, further work is required to reveal the cellular targets responsible for sulfide toxicity in C. jejuni.

Mitigation of Acute Hydrogen Sulfide and Ammonia Emissions from Swine Manure during Three-Hour Agitation Using Pelletized Biochar

The risk of inhalation exposure to elevated concentrations of hydrogen sulfide (H2S) and ammonia (NH3) during the agitation of stored swine manure is high. Once or twice a year, farmers agitate manure before pump-out and application to fields. Agitation of the swine manure causes the short-term releases of highly toxic levels of H2S and NH3. In our previous pilot-scale studies, the biochar powder showed significant mitigation of H2S and NH3 emissions when it was surficially applied to manure immediately before agitation. However, fine biochar powder application poses hazards by itself and may not be practical to apply on a farm scale, especially when livestock and workers are present. We hypothesized that applying pelletized biochar to manure surfaces is just as effective as applying powder to protect farmers and animals from excessive exposure to H2S and NH3. This work reports on the lab-scale proof-of-the-concept trials with biochar pellets on the lab scale. The objective was to compare the biochar pellets and biochar powder on their effectiveness of mitigation on H2S and NH3 gases during 3-h-long swine manure agitation. Three scenarios were compared in (n = 3) trials: (i) control, (ii) 12.5 mm thick surficial application to manure surface of biochar powder, and (iii) an equivalent (by mass) dose of pelletized biochar applied to the manure surface. The biochar powder was bound with 35% (wt) water into ~5 × 10 mm (dia × length) pellets. The biochar powder was significantly (p < 0.05) more effective than the biochar pellets. Still, pellets reduced total H2S and NH3 emissions by ~72% and ~68%, respectively (p = 0.001), compared with ~99% by powder (p = 0.001). The maximum H2S and NH3 concentrations were reduced from 48.1 ± 4.8 ppm and 1810 ± 850 ppm to 20.8 ± 2.95 ppm and 775 ± 182 ppm by pellets, and to 22.1 ± 16.9 ppm and 40.3 ± 57 ppm by powder, respectively. These reductions are equivalent to reducing the maximum concentrations of H2S and NH3 during the 3-h manure agitation by 57% and 57% (pellets) and 54% and 98% (powder), respectively. Treated manure properties hinted at improved nitrogen retention, yet they were not significant due to high variability. We recommend scaling up and trials on the farm-scale level using biochar pellets to assess the feasibility of application to large manure surfaces and techno-economic evaluation.

Cutting out the middle clam: lucinid endosymbiotic bacteria are also associated with seagrass roots worldwide.

Seagrasses and lucinid bivalves inhabit highly reduced sediments with elevated sulphide concentrations. Lucinids house symbiotic bacteria (Ca. Thiodiazotropha) capable of oxidising sediment sulphide, and their presence in sediments has been proposed to promote seagrass growth by decreasing otherwise phytotoxic sulphide levels. However, vast and productive seagrass meadows are present in ecosystems where lucinids do not occur. Hence, we hypothesised that seagrasses themselves host these sulphur-oxidising Ca. Thiodiazotropha that could aid their survival when lucinids are absent. We analysed newly generated and publicly available 16S rRNA gene sequences from seagrass roots and sediments across 14 seagrass species and 10 countries and found that persistent and colonising seagrasses across the world harbour sulphur-oxidising Ca. Thiodiazotropha, regardless of the presence of lucinids. We used fluorescence in situ hybridisation to visually confirm the presence of Ca. Thiodiazotropha on roots of Halophila ovalis, a colonising seagrass species with wide geographical, water depth range, and sedimentary sulphide concentrations. We provide the first evidence that Ca. Thiodiazotropha are commonly present on seagrass roots, providing another mechanism for seagrasses to alleviate sulphide stress globally.

Cryptogamic Packed Biofilter as Potential Adsorbent for CO2, NH3, and H2S Impurities from Biogas

The presence of elevated concentrations of carbon dioxide, hydrogen sulfide, ammonia, and trace impurities in biogas affect its caloric value as well as causes corrosion and is extremely toxic. There are various methods in existence for removal of these impurities, but most are chemically based and expensive and are limited in use. In our work, cryptogams (moss) integrated with soil and biochar packed in a filter have been employed for simultaneous removal of CO2, H2S, and NH3, from biogas. Different soil types rich in metallic oxides at different masses of 100 g, 150 g, and 200 g with a fixed mass of moss and biochar were tested in an on-site experiment to determine the removal efficiency (RE) and sorption capacity (SC). The adsorption dynamics of the filters were investigated at two flow rates, 80 ml/min and 100 ml/min, by determining removal efficiency. For the contribution of each substrate, sorption capacity and breakthrough time were determined by considering 5 g of each substrate that made up the filter. The soils with a high content of extractable cations showed excellent adsorption capacity for H2S by about 20 g S/100 g, which was higher than other adsorbents tested. It was found that integrated biofilter made up of bed arrangement of the soil, biochar, and moss plant improved the quality of biogas with SC of 11 g S and RE of 93% for H2S, 72% for NH3, and 68% for CO2.

What Triggers the Annual Cycle of Cyanobacterium Oscillatoria sp. in an Extreme Environmental Sulfide-Rich Spa?

A seasonal cycle of sulfide, nitrate, phosphate, ammonium, chlorophyll a (Chl a) and Oscillatoria sp. abundance (<100 μm), as well as the relative contribution of taxonomic phytoplanktonic groups (cyanobacteria, green algae, cryptomonads, diatoms and dinoflagellates) to total Chl a were measured by fluorometric measurements at La Hedionda sulfide-rich spa (southern Spain). Fluorometry determined that cyanobacteria Chl a concentration correlated positively with the abundance of Oscillatoria sp. Aggregates at 45–100 μm equivalent spherical diameter (ESD) and was used as an indicator of Oscillatoria sp. Abundance, including for aggregates <45 and >100μm (ESD). In addition, air temperature, radiation and precipitation were downloaded from meteorological databases. In agreement with the meteorological annual cycle observed in air temperature, radiation and precipitation, sulfide concentration at La Hedionda Spa shows an annual cycle with concentrations around 40 μM in winter and up to 200 μM in the dry summer period. Phytoplankton composition was dominated by cyanobacteria (mainly Oscillatoria sp.), but other groups were also represented (green algae, cryptomonads, diatoms and dinoflagellates), although they remained constant throughout the year (median Chl a < 0.2 μg L−1). Cyanobacteria, in contrast, showed an annual cycle with a significantly higher median in summer (Chl a = 1.6 μg L−1) than in winter (Chl a = 0.4 μg L−1). No linear relationship between nutrients and cyanobacteria concentration was observed, but an optimum curve of cyanobacteria concentration to sulfide concentration was fitted through a general additive model (GAM). The four-fold increase of cyanobacteria concentration under exposition of an elevated sulfide concentration can be due to higher growth rates at elevated sulfide concentrations reported for an Oscillatoria sp. strain isolated during the same annual cycle at La Hedionda and we suggest that the selective agent, sulfide, positively triggers Oscillatoria sp. proliferation in summer. According to our findings, the Oscillatoria sp. population of La Hedionda not only is sulfide-resistant, but requires sulfide in its optimal niche.

Comparison of models to evaluate microbial sulphide generation and transport in the near field of a SF/HLW repository in Opalinus Clay

In a deep geological repository for radioactive waste in the Opalinus Clay, disposal canisters may be corroded due to sulphide produced by sulphate reducing bacteria (SRB). This paper presents two computational models, a reactive transport model (RTM) and a simplified semi-analytical model (SM), to evaluate the potential of SRB to generate elevated sulphide concentrations, to assess sulphide fluxes to the canister and, in a simplified manner, the resulting canister corrosion.Calculations performed in the context of the repository's safety assessment based on a shared conceptual model reveal that the two computational models are complementary. The reactive transport model incorporates state-of-the-art understanding of the system's geochemistry, but is currently too computationally demanding to be applied in probabilistic safety assessment sensitivity analyses. The simplified model is fast and efficient, but some of its assumptions need to be verified, and some parameter values need to be calibrated using the more complete reactive transport model. Nonetheless, given the same set of assumptions, the two models predict comparable magnitudes of sulphide fluxes to the canister and comparable canister corrosion depths.

Identification of the source of volatile sulfur compounds produced in milk during thermal processing

Volatile sulfur compounds in ultra-pasteurized (UP) milk are the major contributors to sulfur/burnt and eggy flavors, and these flavors are disliked by consumers. Previous research has established distinct differences in flavor profiles of fluid milk processed by high temperature, short time pasteurization (HTST) and UP by direct steam injection (DSI-UP) or indirect heat (IND-UP). An understanding of the contribution of the individual milk proteins to sulfur off-flavors would clarify the source of sulfur flavors in UP milks. The objective of this study was to determine the source of volatile sulfur compounds in fluid milk with a specific focus on the comparison of heat treatment effects on milks by HTST and UP. Formulated skim milks (FSM) were manufactured by blending micellar casein concentrate and serum protein isolate (SPI). Three different caseins as a percentage of true protein (FSM95, FSM80, and FSM60) were formulated to determine the source of sulfur/burnt and eggy flavors. Freshly processed micellar casein concentrate or SPI were blended to achieve a true protein content of about 3.2%. Raw skim milk served as a control. Skim milk and FSM were pasteurized at 78°C for 15 s (HTST) or 140°C for 2.3 s by IND-UP or DSI-UP. The experiment was replicated twice. Sensory properties of milks and FSM were documented by descriptive sensory analysis. Volatile sulfur compounds in milks and FSM were evaluated using solid-phase microextraction followed by gas chromatography-triple quadrupole mass spectrometry combined with a sulfur selective flame photometric detector. The FSM with higher SPI as a percent of true protein had higher sensory sulfur/burnt and eggy flavors along with elevated concentrations of hydrogen sulfide and carbon disulfide compared with skim milk or FSM with lower proportions of SPI. Sulfur compounds including dimethyl sulfide, dimethyl disulfide, dimethyl trisulfide, dimethyl sulfoxide, and methional were not associated with sulfur/burnt and eggy flavors, which suggests that these compounds may not specifically contribute to the sulfur/burnt and eggy off-flavors of UP milks. Sensory panelists found higher overall aroma impact, and cooked, sulfur/burnt, and eggy flavors for DSI-UP, followed by IND-UP and HTST. The combination of sensory and instrumental methods used in the current study effectively identified that milk serum proteins are the source of sulfur compounds in milk, and further confirmed the contribution of hydrogen sulfide and carbon disulfide to eggy and sulfur/burnt flavors, respectively.

Degassers in drinking water supply

In the Tyumen region, groundwater is widely used for household and drinking water supply, the chemical composition of which is influenced by the increased marshland and peat resistance of soils and, in some cases, the presence of oil and gas-bearing horizons. Underground water typically has a high content of dissolved organic impurities (permanganate oxidability 11.7 - 23.0 mg/dm3), elevated concentrations of free carbon dioxide (20.0 200.0 mg/dm3), hydrogen sulfide (0.20 - 1.95 mg/dm3) and methane (5.3 - 60.0 mg/dm3), dissolved forms of iron (1.14 - 14.00 mg/dm3), manganese (0.02 - 3.80 mg/dm3). Analysis of iron removal plants operation was performed. Analysis of the region's iron removal plants showed that the process of iron removal of water depends on the degree of saturation of water with air oxygen with parallel removal of dissolved gases. Pre-degassing of water with high efficiency degassers is a necessary link of the process chain even when reagent purification techniques are used. Analysis and systematization of existing degassers to remove dissolved carbon dioxide have shown that despite the high-efficiency of some degasser models, they are complex in design and require considerable capital construction and operation costs. There is a need to develop a design not complicated in design, having high degassing efficiency at low economic costs.

Reactive transport calculations to evaluate sulphide fluxes in the near-field of a SF/HLW repository in the Opalinus Clay

Radioactive waste is planned to be disposed in a deep geological repository in the Opalinus Clay (OPA) rock formation in Switzerland. Copper coating of the steel disposal canister is considered as an option to ensure complete waste containment for a period of 100’000 years. Sulphide is a potential corroding agent to copper. While background sulphide concentrations in the OPA are very low (ca. 10−11 mol/L), it cannot be ruled out that sulphide reducing bacteria (SRB) thriving at discrete locations within the engineered barrier system (EBS) of the repository could generate significantly higher dissolved sulphide concentrations. The aim of this study is to conservatively evaluate the potential for SRB to generate and sustain elevated sulphide concentrations in the repository near-field by considering specific geochemical bottle-necks, to quantify the maximum sulphide fluxes towards the canister, and to assess their potential to corrode the copper canister coating. To address these objectives reactive transport calculations are performed, and main conceptual and parametric uncertainties of the model are assessed.

H2S events in the Peruvian oxygen minimum zone facilitate enhanced dissolved Fe concentrations

Dissolved iron (DFe) concentrations in oxygen minimum zones (OMZs) of Eastern Boundary Upwelling Systems are enhanced as a result of high supply rates from anoxic sediments. However, pronounced variations in DFe concentrations in anoxic coastal waters of the Peruvian OMZ indicate that there are factors in addition to dissolved oxygen concentrations (O2) that control Fe cycling. Our study demonstrates that sediment-derived reduced Fe (Fe(II)) forms the main DFe fraction in the anoxic/euxinic water column off Peru, which is responsible for DFe accumulations of up to 200 nmol L−1. Lowest DFe values were observed in anoxic shelf waters in the presence of nitrate and nitrite. This reflects oxidation of sediment-sourced Fe(II) associated with nitrate/nitrite reduction and subsequent removal as particulate Fe(III) oxyhydroxides. Unexpectedly, the highest DFe levels were observed in waters with elevated concentrations of hydrogen sulfide (up to 4 µmol L−1) and correspondingly depleted nitrate/nitrite concentrations (<0.18 µmol L−1). Under these conditions, Fe removal was reduced through stabilization of Fe(II) as aqueous iron sulfide (FeSaqu) which comprises complexes (e.g., FeSH+) and clusters (e.g., Fe2S2|4H2O). Sulfidic events on the Peruvian shelf consequently enhance Fe availability, and may increase in frequency in future due to projected expansion and intensification of OMZs.

The Evolution of Sulfide in Shallow Aquatic Ecosystem Sediments: An Analysis of the Roles of Sulfate, Organic Carbon, and Iron and Feedback Constraints Using Structural Equation Modeling

AbstractThe generation of elevated concentrations of sulfide in sediment pore waters that are toxic to rooted macrophytes is problematic in both marine and freshwaters. In marine waters, biogeochemical conditions that lead to toxic levels of sulfide generally relate to factors that affect oxygen dynamics or the sediment iron concentration. In freshwaters, increases in surface water sulfate have been implicated in decline of Zizania palustris (wild rice), which is important in wetlands across the Great Lakes region of North America. We developed a structural equation (SE) model to elucidate key variables that govern the evolution of sulfide in pore waters in shallow aquatic habitats that are potentially capable of supporting wild rice. The conceptual basis for the model is the hypothesis that dissimilatory sulfate reduction is limited by the availability of both sulfate and total organic carbon (TOC) in the sediment. The conceptual model also assumes that pore water sulfide concentrations are constrained by the availability of pore water iron and that sediment iron supports the supply of dissolved iron to the pore water. A key result from the SE model is that variations in three external variables (sulfate, sediment TOC, and sediment iron) contribute nearly equally to the observed variations in pore water sulfide. As a result, management efforts to mitigate against the toxic effects of pore water sulfide on macrophytes such as wild rice should approach defining a protective sulfate threshold as an exercise tailored to the geochemistry of each site that quantitatively considers the effects of ambient concentrations of sediment Fe and TOC.

The vesicomyid bivalve habitat at cold seeps supports heterogeneous and dynamic macrofaunal assemblages

The high biodiversity found at cold seeps, despite elevated concentrations of methane and hydrogen sulfide, is attributed to multiple sources of habitat heterogeneity. In addition to geological and geochemical processes, biogenic habitats formed by large symbiont-bearing taxa, such as bivalves and siboglinid tubeworms, or by microbial mats drive the biodiversity of small-sized fauna. However, because these habitat-forming species also depend on geochemical gradients, the respective influence of abiotic and biotic factors in structuring associated macrofaunal communities is often unresolved. The giant pockmark Regab located at 3200m depth on the Congo margin is characterized by different fluid-flow regimes, providing a mosaic of the most common biogenic habitats encountered at seeps: microbial mats, mussel beds, and vesicomyid clam beds; the latter being distributed along a gradient of environmental conditions from the center to the periphery of the pockmark. Here, we examined the structure of macrofaunal communities in biogenic habitats formed in soft sediment to (1) determine the influence of the habitats on the associated macrofaunal communities (inter-habitat comparison), (2) describe how macrofaunal communities vary among vesicomyid clam beds (intra-habitat comparison) and (3) assess the inter-annual variation in vesicomyid beds based on repeated sampling at a three-year interval. The highest densities were found in the microbial mat communities in intermediate fluid-flow areas, but they had low diversity — also observed in the sediment close to mussel beds. In contrast, vesicomyid beds harbored the highest diversity. The vesicomyid beds did not show a homogeneous macrofaunal community across sampled areas; instead, density and composition of macrofauna varied according to the location of the beds inside the pockmark. The clam bed sampled in the most active, central part of the pockmark resembled bacterial mat communities by the presence of highly sulfide-tolerant species living at the sediment surface, along with vesicomyid juveniles. This similarity suggests a gradual change in community composition from mats to clam beds. Inter-annual comparisons of the different clam beds highlighted that the most active central site had a more variable community than its peripheral counterparts. Finally, a rapid shift in community structure, particularly in polychaete families, in experimentally reduced oxygen concentrations in the central part of Regab, suggests that high beta-diversity communities can withstand intense variation in geochemical conditions. These community dynamics are likely related to the diversity and to the plasticity of the vesicomyids themselves, because they can cope with high spatial and temporal environmental variability at a very local scale.

Kinetics of sulfate reduction and sulfide precipitation rates in sediments of a bar-built estuary (Pescadero, California)

The bar-built Pescadero Estuary in Northern California is a major fish rearing habitat, though recently threatened by near-annual fish kill events, which occur when the estuary transitions from closed to open state. The direct and indirect effects of hydrogen sulfide are suspected to play a role in these mortalities, but the spatial variability of hydrogen sulfide production and its link to fish kills remains poorly understood. Using flow-through reactors containing intact littoral sediment slices, we measured potential sulfate reduction rates, kinetic parameters of microbial sulfate reduction (Rmax, the maximum sulfate reduction rate, and Km, the half-saturation constant for sulfate), potential sulfide precipitation rates, and potential hydrogen sulfide export rates to water at four sites in the closed and open states. At all sites, the Michaelis–Menten kinetic rate equation adequately describes the utilization of sulfate by the complex resident microbial communities. We estimate that 94–96% of hydrogen sulfide produced through sulfate reduction precipitates in the sediment and that only 4–6% is exported to water, suggesting that elevated sulfide concentrations in water, which would affect fish through toxicity and oxygen consumption, cannot be responsible for fish deaths. However, the indirect effects of sulfide precipitates, which chemically deplete, contaminate, and acidify the water column during sediment re-suspension and re-oxidation in the transition from closed to open state, can be implicated in fish mortalities at Pescadero Estuary.

Using field and laboratory experiments, we examined the effects of decomposing Ulva on Spartina alterniflora growth, soil biogeochemistry and nitrogen dynamics. High levels of Ulva exposure resulted in reductions in above- and belowground biomass, while lower levels of Ulva exposure resulted in reductions in only belowground biomass. Porewater ammonium in soil that contained decomposing Ulva quickly attained potentially toxic levels. In addition, amending soil with Ulva led to elevated porewater concentrations of sulfide and trithiane, an organosulfur compound and potential biocide. Use of a 15N tracer documented plant uptake of Ulva-derived nitrogen, but higher nitrogen availability did not stimulate growth. Our findings support the hypothesis that decaying Ulva mats may create hotspots of adverse physiochemical conditions in salt marshes. However, because our Ulva additions were higher than typically found in coastal marshes, additional field and laboratory studies are needed to establish more firmly whether similarly adverse responses are observed under natural conditions. Colonization of plant substrates at hydrothermal vents and cold seeps in the northeast Atlantic and Mediterranean and occurrence of symbiont-related bacteria.

Reducing conditions with elevated sulfide and methane concentrations in ecosystems such as hydrothermal vents, cold seeps or organic falls, are suitable for chemosynthetic primary production. Understanding processes driving bacterial diversity, colonization and dispersal is of prime importance for deep-sea microbial ecology. This study provides a detailed characterization of bacterial assemblages colonizing plant-derived substrates using a standardized approach over a geographic area spanning the North-East Atlantic and Mediterranean. Wood and alfalfa substrates in colonization devices were deployed for different periods at 8 deep-sea chemosynthesis-based sites in four distinct geographic areas. Pyrosequencing of a fragment of the 16S rRNA-encoding gene was used to describe bacterial communities. Colonization occurred within the first 14 days. The diversity was higher in samples deployed for more than 289 days. After 289 days, no relation was observed between community richness and deployment duration, suggesting that diversity may have reached saturation sometime in between. Communities in long-term deployments were different, and their composition was mainly influenced by the geographical location where devices were deployed. Numerous sequences related to horizontally-transmitted chemosynthetic symbionts of metazoans were identified. Their potential status as free-living forms of these symbionts was evaluated based on sequence similarity with demonstrated symbionts. Results suggest that some free-living forms of metazoan symbionts or their close relatives, such as Epsilonproteobacteria associated with the shrimp Rimicaris exoculata, are efficient colonizers of plant substrates at vents and seeps.

Organic enrichment at salmon farms in the Bay of Fundy, Canada: DEPOMOD predictions versus observed sediment sulfide concentrations

A model for predicting benthic impacts of fish farms (DEPOMOD) was used to pre- dict organic carbon deposition rates at 6 salmon farms in the southwestern New Brunswick (SWNB) area of the Bay of Fundy, Canada. Model predictions of the seafloor area with elevated deposition rates were compared to the areas of seafloor with elevated observed sulfide concentra- tions. DEPOMOD predictions with resuspension appeared to overestimate the rate of resuspen- sion of waste particles where current speeds were moderate to high; therefore, model runs with- out resuspension were used for comparisons. There were no consistent relationships between current speeds and the predicted (without resuspension) area with elevated deposition rates and the areas with elevated sulfide concentrations. There was a positive relationship between the areas with elevated deposition rates and the areas with elevated sulfide concentrations at 3 sites, with a better fit when the DEPOMOD runs used average daily feeding rates during 1 mo periods including the date of sediment sampling (compared to average feeding rates during 3 mo feeding periods ending near the date of sampling). Because the predicted area with elevated deposition rates (without resuspension) was strongly correlated with the feeding rate, it is important that the appropriate feeding rate be used in model runs. At sediment sampling stations where predicted deposition rates were low, sulfide concentrations were usually low; however, at sampling stations where predicted deposition rates were elevated, sulfide concentrations showed high variability. Implications for the use of DEPOMOD for management of the salmon aquaculture industry in SWNB are discussed.
